RESUMO

The nanozyme-based strategy is currently one of the frontiers in the detection of toxic heavy metal ions. However, the utilization of noble metal free nanozymes to construct an economically and environmentally sustainable methodology remains largely unknown. Here, chitosan-functionalized molybdenum(IV) selenide nanosheets (CS-MoSe2 NS), greenly synthesized by an ionic liquid-assisted grinding method, were exploited for the colorimetric sensing of mercury ions (Hg2+). The sensing principle was based on the activating effect of Hg2+ on CS-MoSe2 NS nanozyme activities, triggered by the in situ reduction of chitosan-captured Hg2+ ions on a MoSe2 NS surface. Using 3,3',5,5'-tetramethylbenzidine (TMB) as a colorimetric indicator, the concentrations of activator-like Hg2+ ions could be quantitatively and selectively monitored, reaching a limit of detection of 3.5 nM with the ultraviolet-visible spectrophotometer. In addition, the integration system of CS-MoSe2 NS with a smartphone achieved a portable detection limit as low as 8.4 nM Hg2+ within 15 min and showed high specificity and anti-interfering ability over other ions and great practicability in real water and serum samples. The eco-friendly properties of such sensing system were also confirmed. This work emphasizes the rational portable assembly of biocompatible nanozymes like CS-MoSe2 NS for the field detection of Hg2+ in food, biological, and environmental samples.

IMPORTANCE: Ceftriaxone-based antimicrobial therapies for gonorrhea are threatened by waning ceftriaxone susceptibility levels and the global dissemination of the high-level ceftriaxone-resistant gonococcal FC428 clone. Combination therapy can be an effective strategy to restrain the development of ceftriaxone resistance, and for that purpose, it is important to find an alternative antimicrobial to replace azithromycin, which has recently been removed in some countries from the recommended ceftriaxone plus azithromycin dual-antimicrobial therapy. Ideally, the second antimicrobial should display synergistic activity with ceftriaxone. We hypothesized that bacitracin might display synergistic activity with ceftriaxone because of their distinct mechanisms targeting bacterial cell wall synthesis. In this study, we showed that bacitracin indeed displays synergistic activity with ceftriaxone against Neisseria gonorrhoeae. Importantly, strains associated with the FC428 clone appeared to be particularly susceptible to the bacitracin plus ceftriaxone combination, which might therefore be an interesting dual therapy for further in vivo testing.

Green and white asparagus are quite different crops but can be harvested from the same plant. They have distinct morphological differences due to their mode of cultivation and they are characterised by having contrasting appearance and flavour. Significant chemical differences are therefore expected. Spears from three varieties of both green and white forms, harvested in two consecutive seasons were analysed using headspace GC-MS and LC-MS with an untargeted metabolomic workflow. Mainly C5 and C8 alcohols and aldehydes, and phenolic compounds were more abundant in green spears, whereas benzenoids, monoterpenes, unsaturated aldehydes and steroidal saponins were more abundant in white ones. Previously reported key asparagus volatiles and non-volatiles were detected at similar or not significantly different levels in the two asparagus types. Spatial metabolomics revealed also that many volatiles with known positive aroma attributes were significantly more abundant in the upper parts of the spears and showed a decreasing trend towards the base. These findings provide valuable insights into the metabolome of raw asparagus, the contrasts between green and white spears as well as the different chemical distributions along the stem.
