BACKGROUND: Atmospheric levels of polycyclic aromatic hydrocarbons (PAHs) have been monitored in many companies since 1940. Because of the use of respiratory protective equipment (RPE) and cutaneous absorption, the measurement of urinary 1-hydroxypyrene (1-OHP), metabolite of pyrene (Pyr), and, more recently, 3-hydroxybenzo[a]pyrene (3-OHBaP), metabolite of benzo[a]pyrene (BaP), has been carried out to assess PAH exposure and estimate health risks. OBJECTIVES: This study aimed to investigate the agreement between 523 air and biological levels recorded in the Exporisq-HAP database by taking into account the effectiveness of RPE. METHODS: The agreement/consistency between 523 air and biological exposure levels was assessed by estimating and comparing the probability of exceeding French limit values (LVs) for both BaP and 3-OHBaP and ACGIH LV for 1-OHP, respectively. PAH airborne levels (wPAHs) were weighted by an assigned protection factor (APF) depending on the type of mask worn by workers, while urinary 1-OHP concentrations were adjusted with the wBaP/wPyr ratio of each industrial sector (wadj1-OHP). RESULTS: Within occupational groups, there was an overall agreement between airborne PAH levels and urinary biomarker concentrations. A clear dichotomy was found between "petroleum-derived" and "coal-derived" groups, with much higher exposures in the latest group despite the use of RPEs by two-thirds of the workers. The type of RPE varied from one plant to another, which underlines the importance of taking into account their effectiveness. The analysis of urinary 3-OHBaP was not relevant for low PAH exposure levels. In addition, this biomarker underdiagnosed the exceedance of LV relative to BaP levels for 6% of "coal-derived" groups. CONCLUSIONS: The use of urinary wadj1-OHP seemed to be more protective to assess the exceedance of LVs than those of urinary 3-OHBaP and air wBaP, but adjustment of the 1-OHP concentration by the BaP/Pyr ratio requires air sampling due to highly variable ratios observed in the studied occupational groups.

Previous studies found associations between impairments of immune functions and exposure to polycyclic aromatic hydrocarbons (PAHs) in ambient air pollution in the U. S. and China. However, the results remain inconclusive due to the limitations of these studies. In this study, we aimed to examine the direction and magnitude of immune changes related to PAH exposure from household air pollution among rural women living in Gansu, China. Healthy village women (n = 34) were recruited and enrolled in the study. Questionnaires were administered. Blood and urine samples were collected and analyzed during non-heating (September 2017, "summer") and heating (January 2018, "winter") seasons. Urinary 1-hydroxypyrene (1-OHP) was quantified as the biomarker of PAH exposure. To evaluate Treg cell related immune functions, we examined immunoglobulin E (IgE), percent of T-regulatory (Treg) cells, and gene expression of following: forkhead box transcription factor 3 (Foxp3), transforming growth factor-ß (TGF-ß), interleukin 10 (IL-10), and interleukin 35 (IL-35), composed of interleukin-12 alpha (IL-12α) and Epstein-Barr-virus-induced gene 3 (EBi3). Urinary 8-hydroxy-2-deoxyguanosine (8-OHdG) was measured to evaluate oxidative DNA damage. The results showed that the concentration of 1-OHP increased from 0.90 to 17.4 µmol mol-Cr -1 from summer to winter (p < 0.001). Meanwhile, average percent of Treg cells decreased from 5.01% to 1.15% (p < 0.001); IgE and mRNA expressions of Foxp3, TGF-ß, IL-10, IL-12α and EBi3 all significantly decreased (p < 0.001); Urinary 8-OHdG increased from 12.7 to 30.3 ng mg-Cr -1 (p < 0.001). The changes in 8-OHdG, Foxp3 and TGF-ß were significantly associated with the increase of 1-OHP. The results suggested that we observed a substantial increase of PAH exposure in winter, which was significantly associated with the repression on Treg cell function and oxidative DNA damage. Exposure to PAHs in household air pollution possibly induced immune impairments among rural women in northwest China.

PURPOSE: Polycyclic aromatic hydrocarbons (PAHs) are contaminants with carcinogenic effects but little is known about their presence in environments surrounding oil drilling operations and spills or exposure levels in nearby communities. The objective of this study was to characterize PAH levels in people living near oil drilling operations in relation to fish consumption, occupation, source of water and other socio-demographic characteristics. METHODS: This pilot study examined PAH exposure by measuring 1-hydroxypyrene (1-OHP) in urine samples using high-performance liquid chromatography and fluorescence detection from 75 women and men in the Ecuadorian and Peruvian Amazon living near oil drilling operations and who answered a questionnaire collecting socio-demographic, occupational and dietary information. Data were analyzed using multiple linear regression models. RESULTS: The mean value of 1-OHP was 0.40 µmol/mol creatinine, 95% CI 0.32-0.46 µmol/mol creatinine. Women who used water from a surface source (for washing clothes or bathing) had almost twice the amount of 1-OHP in their urine (mean 1-OHP = 0.41 µmol/mol creatinine, 95% CI 0.28-0.54 µmol/mol creatinine, n = 23) as women who used water from either a well, a spring or rain (mean 1-OHP = 0.22 µmol/mol creatinine, 95% CI 0.11-0.34 µmol/mol creatinine, n = 6). Men who reported eating a bottom-dwelling species as their most commonly consumed fish (mean 1-OHP = 0.50 µmol/mol creatinine, 95% CI 0.36-0.64 µmol/mol creatinine, n = 31) had twice as much 1-OHP in their urine as men who reported a pelagic fish (mean 1-OHP = 0.25 µmol/mol creatinine, 95% CI 0.15-0.35 µmol/mol creatinine, n = 15), signaling either oral (fish consumption) or dermal (while standing in water fishing benthic species) exposure. CONCLUSIONS: More contact with surface water and benthic fish may result in higher levels of 1-OHP in human urine among the study population. Reducing the amount of oil and wastes entering the waterways in Andean Amazonia would be one way to reduce exposure.

Human biomonitoring (HBM) is an appreciated tool used to evaluate human exposure to environmental, occupational or lifestyle chemicals. Therefore, the aim of this study was to evaluate the exposure levels for environmental chemicals in urine and blood samples of children from San Luis Potosí, Mexico (SLP). This study identifies environmental chemicals of concern such as: arsenic (45.0 ± 15.0 µg/g creatinine), lead (5.40 ± 2.80 µg/dL), t,t-muconic acid (266 ± 220 µg/g creatinine), 1-hydroxypyrene (0.25 ± 0.15 µmol/mol creatinine), PBDEs (28.0 ± 15.0 ng/g lipid), and PCBs (33.0 ± 16.0 ng/g lipid). On the other hand, low mercury (1.25 ± 1.00 µg/L), hippuric acid (0.38 ± 0.15 µg/g creatinine) and total DDT (130 ± 35 ng/g lipid) exposure levels were found. This preliminary study showed the tool's utility, as the general findings revealed chemicals of concern. Moreover, this screening exhibited the need for HBM in the general population of SLP.

BACKGROUND AND AIM: Polycyclic aromatic hydrocarbons (PAHs) are the products of incomplete combustion or pyrolysis of various organic materials. Their ubiquity in the environment leads to measurable levels of exposure. However, the exposure varies strongly between different regions in Europe. Some PAHs with four or more rings are suspected to be human carcinogens. Therefore, the occupational and/or environmental exposure to PAHs may cause a significant health risk. The aim of the study was to evaluate current levels of PAH exposure in defined groups of workers. METHODS: The industrial sites selected in this survey involved PAHs originating from coal tar pitch, carbon black, bitumen, and rubber fumes. Based on the historical data, the sites were expected to exhibit quantifiable levels of exposure to PAHs. The total study population consisted of 139 persons: 108 workers (85 males and 23 females) workers were occupationally exposed in aluminium production, the production of graphite electrodes, road construction, or the rubber forming industry and 31 control individuals in two groups. RESULTS: The highest concentrations ­ 8-hour time-weighted average (TWA) levels (sum of 16 components according to the EPA list), as expected, were found in the aluminium production plant (55.15 µg.m−3) and production of graphite electrodes (54.25 µg.m−3). The lowest concentrations were found in personal air samples of road construction workers (1.93 µg.m−3). The concentrations of 1-hydroxypyrene as a pyrene metabolite (1-OHP) in the urine of the exposed group of workers were found in levels 0.74 µmol.mol−1 creatinine before the exposure and 2.27 µmol.mol−1 creatinine after the exposure (arithmetic mean values). 1-OHP concentrations in post-shift urine samples were highly correlated with the total airborne PAHs concentrations and pyrene concentrations in air. The correlation coefficients (rS) between 1-OHP concentration and pyrene or total PAHs in air were 0.710 and 0.752 (p < 0.05). This statistical analysis confirmed the effect of the occupational exposure to PAHs and pyrene on body burden in workers. However, a modifying effect of gender, smoking habits, dietary intake, genetically modified metabolism, and the use of medication on the toxicokinetics of pyrene was not determined as significant. CONCLUSION: Based on the results a strong correlation between the concentration of 1-OHP in urine as an exposure biomarker and the concentration of pyrene or PAH was affirmed.

This article reviewed the published studies on the environmental exposure to polycyclic aromatic hydrocarbons (PAHs) among children and assessed the urinary 1-hydroxypyrene (1-OHP) level as a biomarker of exposure to PAHs. The current knowledge of the potential health effects of increased 1-OHP in children was reviewed. Additionally, the influence of genetic polymorphism on the urinary 1-OHP level was discussed in this review. The assembled data showed that children who are attending schools or living close to industrial and polluted urban areas might have greater exposure to higher concentrations of PAHs with a higher level of urinary 1-OHP when compared to those children living in rural areas. Urinary 1-OHP may be a reliable biomarker for determining the genotoxic effects, oxidative stress and inflammation caused by exposure to PAHs. Strong research evidence indicated that the total body burden of PAHs should be evaluated by biomonitoring of 1-OHP in line with other urinary PAHs metabolites (with 2-3 rings) to evaluate recent total exposure to PAHs. Overall, the study suggests implementing a mitigation plan to combat air pollution to provide a cleaner environment for children.

Background: Serum carcinoembryonic antigen (CEA) is a biomarker commonly used to detect colorectal cancer. CEA levels are affected by many factors, including cardiometabolic diseases, such as cardiovascular diseases (CVDs) and diabetes. Cardiometabolic diseases and cancer share a similar pathological inflammatory pathway, which correlates with an unhealthy lifestyle. Hence, establishing an adequate CEA cut-off value might be a valuable reference for developing precision healthcare programs for cardiometabolic disease prevention. This study aimed to investigate the association between cardiometabolic risks and serum CEA and the underlying factors. Methods: A community-based, cross-sectional study was conducted between March and December 2021 on the western coast of Taiwan. Lifestyle data were assessed using a structured questionnaire. The cardiometabolic biomarkers, serum CEA, urine malondialdehyde, and 1-hydroxypyrene were quantified by the central laboratory of the collaborating hospital. Chi-square and binary multivariable logistic regression implemented in R version 4.0.2 were used to identify factors defining the risk of high serum CEA levels. Results: A total of 6,295 adult residents without cancer-related diseases completed the study. The mean age was 48.6 (SD = 16.4) years, 56% were female, 32% had metabolic syndrome, and 23% and 10% had CVDs and diabetes, respectively. Multivariate logistic regression showed that age ≥ 65 years, male sex, alcohol consumption, smoking, infrequent use of dental floss, fewer remaining teeth, CVDs, diabetes, and oxidative stress were significantly associated with serum CEA ≥ 3 ng/mL. The discriminatory performance of the area under the receiver operating characteristic curve was 0.75 (0.73-0.76), showing that this model was suitable for distinguishing high CEA levels. Conclusion: Our findings highlight the importance of understanding cardiometabolic diseases, unhealthy lifestyles, and oxidative stress, which contribute to high serum CEA. This study demonstrates that CEA, a well-known tumor marker, can help the early detection and prevention of cardiometabolic diseases via personalized lifestyle modification.

Cardiovascular diseases (CVDs) are the leading cause of morbidity and mortality worldwide. Environmental and genetic factors are recognized as risk determinants in the onset and development of CVDs. However, the interaction between both factors on CVDs risk is not still completely clarified. Therefore, the objective of this study was to evaluate the effect of the interaction between methylenetetrahydrofolate reductase (MTHFR) C677T polymorphism and polycyclic aromatic hydrocarbon (PAH) exposure (gene-environment interaction) on cardiovascular risk biomarkers in Mexican women. A cross-sectional study was completed with the participation of 390 healthy women. For all enrolled women, anthropometric measurements, serum biochemical analyses, atherogenic indexes, and serum concentrations of biomolecules used as CVD risk biomarkers were obtained. 1-Hydroxypyrene (1-OHP) was measured in urine, as an exposure biomarker of PAHs. The mean urinary level of 1-OHP in the assessed population was 1.23 ± 1.40 µmol/mol creatinine. The allelic frequency (MTHFR C677T polymorphism) identified in the registered individuals was 68.0% for the mutant allele (T-allele). Significant positive associations were detected between urinary 1-OHP levels and serum asymmetric dimethylarginine (ADMA) concentrations (p < 0.05) and atherogenic index of plasma (AIP) values (p < 0.05). Also, women with the TT genotype of the MTHFR C677T enzyme have the highest serum ADMA levels (p < 0.05) and AIP values (p < 0.05) compared to women grouped as CC genotype and CT genotype. Besides, the findings in this study suggest an interaction between environmental (PAHs exposure) and genetic (MTHFR C677T polymorphism) factors on cardiovascular risk markers (ADMA and AIP). According to the usefulness of AIP and ADMA, an increased cardiovascular risk is notable in highly exposed individuals to PAHs with the polymorphic genotype (TT) of the MTHFR enzyme. Therefore, intervention programs in the target communities are required to diminish the cardiovascular risk of the assessed individuals.

Polycyclic aromatic hydrocarbons (PAHs) are a group of chemical compounds which interest to human biological monitoring researches because of their potential carcinogenic, mutagenic, and teratogenic properties. However, the exposure of female beauticians to these contaminants is not well-reported. For biomonitoring of potential exposure of female cosmeticians to PAHs in beauty salons, urine samples were taken from cosmetologist women (n = 50.00) and housewives (n = 35.00) as the exposure group (EG) and control group (CG), respectively. Next, unmetabolized PAHs levels as well as the concentration of - 1-hydroxypyrene (1-OHP) were analyzed in these specimens. In addition, since benzene has some common source with PAHs, in this study t, t'-Muconic acid (TTMA) level was also determined as the metabolite and indicator of exposure to benzene. The results indicated a high detection frequency of the target compounds (PAHs, 1-OHP and TTMA) in the urine specimens of beauticians. The results also showed that there is a significant difference between the concentration of these pollutants in the urine specimens of the exposure and control groups. The median concentration of ΣPAHs, 1-OHP, and TTMA in the before exposure (BE) specimens collected from the exposure group were 337.42 ng/L, 593.92 ng/L, and 247.90 µg/L, respectively. However, a higher concentration of these contaminants was observed in the after exposure specimens with a median concentration of 423.29 ng/L, 745.03 ng/L, and 310.97 µg/L, respectively. In terms of contribution of PAHs compounds in total toxic equivalents, DahA, BaP, and IndP with 59.03, 28.73, and 2.86 % had the largest share. In this study, it was also observed that some kidney damage biomarkers as well as some oxidative stress injury biomarkers are positively and significantly correlated with the urinary values of ∑PAHs. Thus, it can be concluded that high health risks threaten the female beauticians regarding kidney injury and DNA oxidative stress.

BACKGROUND: The association between the biomarkers of environmental exposure, oxidative stress, and health-related behaviors in community residents living in an endemic viral hepatitis area and near petrochemical industrial complexes remains unclear. From a health promotion perspective, healthcare providers must know what to do for residents concerned about their health and living environment, especially for individual-level and modifiable risk factors. Therefore, we aimed to explore the factors associated with urinary 1-hydroxypyrene (1-OHP) and malondialdehyde (MDA). METHODS: A community-based, cross-sectional study was conducted between July 2018 and February 2019 in western coastal Yunlin County, Taiwan. All participants lived within a 10 km radius of a large petrochemical complex and did not work in the factory. This study was conducted with the local hospital through annual community health screening. Biological samples were collected and biomarkers determined and quantified in the central laboratory of the collaborating hospital. RESULTS: A total of 6335 adult residents completed the study. The mean age was 47.7 (SD = 16) years. Out of the total population, 56.4% were female, 30.1% had metabolic syndrome (MetS), and 16.8% and 14.3% had hepatitis B virus antigen (HBsAg) and hepatitis C virus antibody (anti-HCV) positivity, respectively. The median 1-OHP and MDA level was 0.11 and 0.9 µg/g creatinine with an interquartile range of 0.07-0.18, and 0.4-1.5, respectively. The MDA levels correlated with specific diseases. The multivariable ordinal logistic regression model revealed that female sex, smoking, betel nut use, HBsAg, and anti-HCV positivity were associated with higher 1-OHP levels. In men, MetS was associated with higher 1-OHP levels and regular exercise with lower 1-OHP levels. High MDA levels were associated with smoking, betel nut users, HBsAg, and anti-HCV positivity. CONCLUSIONS: The findings highlight the importance of initiating individualized health promotion programs for residents near petrochemical factories, especially for adults with substance-use and cardiometabolic risk factors. Furthermore, it is crucial to provide further treatment to patients with viral hepatitis.

The study aimed to evaluate the potential effects of the chronic exposure to chemical agents from air pollution on phenotypic and genotypic expressions of peripheral biomarkers and tumor-related proteins in mononuclear cells. This study evaluates 85 taxi drivers (outdoor workers) and 55 non-occupationally exposed persons (NOE) to air pollution (indoor workers). The biomarkers were urinary 1-hydroxypyrene (1-OHP), for organic agents, and blood As and Ni, for inorganic agents. Oxidative stress biomarkers; protein expression of ICAM-1 (CD54), ß2-integrin, L-selectin (CD62-L), and MCP1; gene expression of ICAM-1, p53 and CD26 were performed. Urinary 1-OHP and blood As and Ni levels were increased in taxi drivers and were associated with inflammatory and oxidative stress biomarkers. These exposure biomarkers were also associated with each other, suggesting a common source of exposure. The gene expression of p53, CD26 and ICAM-1 were decreased in taxi drivers and were strongly associated between them, indicating a commom regulation point. The antioxidant non-protein thiols and lycopene were negatively associated with inflammatory biomarkers, maybe regulating the immune-response. We demonstrated, for the first time, that in occupational exposure to air pollution chemicals, oxidative and inflammatory processes are involved in the immune-regulatory process, and indirectly contribute to suppressing the p53 and CD26 expressions, increasing the risk of cancer development. On the other hand, antioxidants could contribute to improving the immune-regulation, but more studies are needed.

We studied cancer patients for possible PAH exposure, using urinary concentration of 1-hydroxypyrene (1-OHP) as a biomarker of internal dose of polycyclic aromatic hydrocarbons (PAHs). The subjects included in this study belonged to various socio-demographic backgrounds, and were diagnosed with cancer (i.e. lung, head and neck or digestive tract cancer). In general, we observed high concentration of urinary 1-OHP among digestive tract cancer patients, compared with the controls (CN) (mean 1.06, median 1.03 and mean 0.62, median 0.63 µmol/mol-Cr in digestive tract cancer patients and controls respectively). The concentrations of urinary 1-OHP were higher than the background level of PAHs; therefore, these groups could have been exposed to PAHs. Highest urinary 1-OHP concentration was observed in digestive tract cancer patients (median 1.25 µmol/mol-Cr) with GSTM-1 genotype. The results of PCA were consistent with qualitative and quantitative data analysis. The contribution of urinary 1-OHP eigenvector revealed a relatively high PAH-exposure among cancer patients compared with CN, while diet and age were influential parameters among cancer patients, which could have a strong link in cancer etiology in the selected exposure groups.

Recently, in developing countries, polycyclic aromatic hydrocarbons (PAHs) have been considered contaminants of grave concern for women and children. Therefore, the aim of this study was twofold: (1) evaluate exposure assessment to PAHs using urinary 1-hydroxypyrene (1-OHP) as an exposure biomarker and (2) perform a health risk assessment in women from four different high risk scenarios in Mexico. From 2012 to 2013, in a cross-sectional study, we evaluated a total of 184 healthy women from the following scenarios: (A) indoor biomass combustion site (n = 50); (B) brick manufacturing site using different materials such as fuel sources (n = 70); (C) industrial site (n = 44); and (D) high vehicular traffic site (n = 20). 1-hydroxypyrene (1-OHP) was quantified using a high-performance liquid chromatography (HPLC) technique. Afterward, a probabilistic health risk assessment was performed (Monte Carlo analysis). Mean urinary 1-OHP levels found were 0.92 ± 0.92; 0.91 ± 0.83; 0.22 ± 0.19; and 0.14 ± 0.17 µg/L for scenario A, B, C, and D, respectively. Then, based on the measured urinary 1-OHP levels, the estimated median daily intake doses of pyrene were calculated: 659, 623, 162, and 77.4 ng/kg/day for the women participating in the study living in areas A, B, C, and D, respectively, and finally, the hazard quotient (HQ) was calculated (22 ± 21, 21 ± 20, 5.5 ± 5.5, and 2.6 ± 3.5; for areas A, B, C, and D, respectively), high health risk was noted for the women living in the studied communities. The data shown in this study (exposure levels to PAHs and health risk assessment) made it reasonable to conclude that the exposure levels found have a significant potential for generating adverse effects on human health in the studied scenarios.

Polycyclic aromatic hydrocarbons (PAHs) are a group of ubiquitous environmental contaminants raising worldwide concerns due to their carcinogenic effects. In this study, 1-hydroxypyrene (1-OHP, the most widely used biomarker of internal dose of PAHs exposure) in undiluted human urine samples (10 mL) was selectively enriched by polypyrrole-coated Fe3O4 magnetite nanocomposites (termed as Fe3O4@Ppy, 1 mg) and then directly eluted by the electrospraying solvent (acetone/benzene/acetic acid (v/v/v, 90/10/1); 100 uL) biased with -3.5 kV to produce the deprotonated 1-OHP anions for mass spectrometric analysis. The method established here significantly improved the current performance for detection of urinary 1-OHP, providing the speed for a single sample analysis within 4 min, the limits of detection (LOD) of 0.0001 µg L(-1), the linear response range of 0.001-5.000 µg L(-1) (R(2) = 0.9994), recovery rates of 90.6-96.1%, and relative standard deviation (RSD, n = 6) values between 2.9% and 8.0%. Human samples including raw human urine collected from 10 healthy volunteers (5 smokers and 5 nonsmokers) and 7 lung cancer patients have been successfully analyzed, showing that magnetic solid-phase extraction (MSPE) coupled with internal extractive electrospray ionization mass spectrometry (iEESI-MS) is an alternative strategy for high throughput quantitative detection of urinary 1-OHP for health risk assessment of PAHs exposure.

To explore whether the alteration of lncRNA expression is correlated with polycyclic aromatic hydrocarbons (PAHs) exposure and DNA damage, we examined PAHs external and internal exposure, DNA damage and lncRNAs (HOTAIR, MALAT1, TUG1 and GAS5) expression in peripheral blood lymphocytes (PBLCs) of 150 male coke oven workers and 60 non-PAHs exposure workers. We found the expression of HOTAIR, MALAT1, and TUG1 were enhanced in PBLCs of coke oven workers and positively correlated with the levels of external PAHs exposure (adjusted Ptrend < 0.001 for HOTAIR and MALAT1, adjusted Ptrend = 0.006 for TUG1). However, only HOTAIR and MALAT1 were significantly associated with the level of internal PAHs exposure (urinary 1-hydroxypyrene) with adjusted ß = 0.298, P = 0.024 for HOTAIR and ß = 0.090, P = 0.034 for MALAT1. In addition, the degree of DNA damage was positively associated with MALAT1 and HOTAIR expression in PBLCs of all subjects (adjusted ß = 0.024, P = 0.002 for HOTAIR and ß = 0.007, P = 0.003 for MALAT1). Moreover, we revealed that the global histone 3 lysine 27 trimethylation (H3K27me3) modification was positively associated with the degree of genetic damage (ß = 0.061, P < 0.001) and the increase of HOTAIR expression (ß = 0.385, P = 0.018). Taken together, our findings suggest that altered HOTAIR and MALAT1 expression might be involved in response to PAHs-induced DNA damage.

Pollution from road traffic is not only a major source of exposure to polycyclic aromatic hydrocarbons (PAHs) but also a growing problem in the city of Lahore (Pakistan). In this study, we evaluated exposure to traffic-related PAHs, among subjects including traffic police officers (TPs), rickshaw drivers (RKs) shopkeepers working near main roads (SKs) and a control group (CN) for comparative analyses. We monitored the 1-hydroxypyrene (1-OHP) as biomarkers of exposure to PAHs and its probable association with catalase (CAT), glutathione (GSH) glutathione peroxidase (GSHPx) activity as biomarkers of oxidative stress in selected cohorts from the city Lahore. Results showed that median 1-OHP concentration was significantly higher in TPs than CN (med 1.21 vs. 0.51 µmol mol-C(-1) respectively, P=0.046), followed by RKs (0.68 µmol mol-C(-1), P=0.19 vs. CN). Furthermore, GSH, GSHPx, and CAT activities were also higher in exposed subjects than CN, which indicated that they experienced oxidative stress. Similar, but less severe observations were recorded in SKs. Observation of self-reported health status showed that, on the basis of daily time spent in the middle of heavy traffic, TPs and RKs most frequently suffered from adverse head and respiratory symptoms. The study shows that increasing traffic pollution can be associated with important health risk factor not only for the workers in transport industry but also for the public. Finally, the issue of traffic pollution in Lahore city needs to be addressed on priority.

BACKGROUND: Vehicular traffic is a major source of outdoor air pollution in urban areas, and studies have shown that air pollution is worse during hours of commuting to and from work and school. However, it is unclear to what extent different commuting behaviors are a source of air pollution compared to non-commuters, and if air pollution exposure actually differs by the mode of commuting. This study aimed to examine the relationships between commuting behaviors and air pollution exposure levels measured by urinary 1-OHP (1-hydroxypyrene), a biomarker of exposure to polycyclic aromatic hydrocarbons (PAHs). METHODS: A cross-sectional study of 174 volunteers living in Montreal, 92 females and 82 males, aged 20 to 53 years was conducted in 2011. Each participant completed a questionnaire regarding demographic factors, commuting behaviors, home and workplace addresses, and potential sources of PAH exposure, and provided a complete first morning void urine sample for 1-OHP analysis. Multivariable general linear regression models were used to examine the relationships between different types of commuting and urinary 1-OHP levels. RESULTS: Compared to non-commuters, commuters traveling by foot or bicycle and by car or truck had a significantly higher urinary 1-OHP concentration in urine (p=0.01 for foot or bicycle vs. non-commuters; p=0.02 for car or truck vs. non-commuters); those traveling with public transportation and combinations of two or more types of modes tended to have an increased 1-OHP level in urine (p=0.06 for public transportation vs. non-commuters; p=0.05 for commuters with combinations of two or more types of modes vs. non-commuters). No significant difference in urinary 1-OHP variation was found by mode of commuting. CONCLUSION: This preliminary study suggests that despite the mode of commuting, all types of commuting during rush hours increase exposure to air pollution as measured by a sensitive PAH metabolite biomarker, and mode of commuting did not explain exposure variation.

OBJECTIVES: 1-hydroxypyrene is an important biomarker of exposure to polycyclic aromatic hydrocarbons (PAHs), which appears in the urine of exposed human subjects. In developing countries, where advanced instruments are not available, the importance of this biomarker demands convenient and sensitive methods for determination purposes. This study aimed at developing a methodology to quantify 1-hydroxypyrene (a biomarker of PAHs exposure) based on the UV-visible detector in the reverse phase high pressure liquid chromatography (HPLC). MATERIAL AND METHODS: A 20 µl injection of sample was used for manual injection into the HPLC Shimadzu, equipped with the SPD-20 A UV-visible detector, the LC-20AT pump and the DGU-20A5 degasser. The C-18 column was used for the purpose of the analysis. RESULTS: The method showed a good linearity (the range: R2 = 0.979-0.989), and high detectability up to the nmol level. The average retention was 6.37, with the accuracy of 2%, and the percentage of recovery remained 108%. The overall performance of this method was comparable (in terms of detection sensitivity) and relatively better than previously reported studies using the HPLC system equipped with the UV-detector. CONCLUSIONS: This method is suitable and reliable for the detection/quantification of the 1-OHP in human urine samples, using the UV-detector, however, it is less sensitive as compared to the results of a florescence detector.

This paper reports the connections between red blood cells abnormality risk of petrochemical workers and their exposure to airborne polycyclic aromatic hydrocarbons (PAHs). Urinary 1-hydroxypyrene (1-OHP), as the biomarker of PAHs exposure, was adopted to assess the exposure risk of the petrochemical workers to PAHs in Xigu, the west suburb of Lanzhou where petrochemical industries are located. Fifty-three workers, sub-grouped to 36 petrochemical workers and 17 office workers, participated in this investigation. Logistic regression model and spearman correlation analysis were performed to estimate the associations between PAHs exposure levels and red blood cells abnormality risk of petrochemical workers. Strong associations between some red cell indices (MCH, MCHC, RDW) and 1-OHP concentration were found. Results also show that the red blood cells abnormality risk increased with increasing PAHs exposure level. Compared with office workers, risk level of red blood cells abnormality in petrochemical workers was higher by 41.7 % (OR, 1.417; 95 % CI: 0.368-5.456) than that in office workers. This result was verified by the tissue-to-human blood partition coefficient for pyrene and 1-OHP. The quantitative assessments of the potential health risk through inhalation exposure to PAHs were conducted using the Incremental Lifetime Cancer Risk (ILCR) model. It was found the ILCR from inhalation exposure to PAHs for the petrochemical workers ranged from 10(-5) to 10(-4) with 95 % probability, indicating that petrochemical plant workers were under a high potential cancer risk level.

INTRODUCTION: The International Agency for Research on Cancer classifies specific polycyclic aromatic hydrocarbons (PAHs) as probable carcinogens. This study compares two PAH biomarkers and their relationship with geographic information system (GIS) based traffic density (a proxy of PAH exposure), and explores the determinants of the PAH biomarkers. METHODS: A cross-sectional study was conducted in Montreal with 200 volunteers (107 females and 93 males) ages 20 to 53 years. Data were collected by questionnaire, urine samples were used for biomarker analysis, and innovative GIS-based time- and distance-weighted traffic densities (TDWTD) were calculated for all locations of participants during the 48 h prior to urine collection. RESULTS: Detection rates of the two biomarkers were greater than 95%. Female participants had higher 1-OHP and 1-OHPG levels than males, and no relationship was detected between TDWTD in 48 h and the two PAH biomarkers. Biomarker levels were related to smoking more than one pack of cigarettes in the previous 48 h, and among non-smokers, barbecued meat consumption increased the level of urinary 1-OHP (exp ß: 1.45, 95% CI: 1.07 to 1.98). CONCLUSIONS: Both 1-OHP and 1-OHPG can be used to assess the relatively low PAH levels to which the general population is exposed. With the exception of smoking, the impact of PAH exposure factors on the biomarkers is relatively small in this study population.
