RESUMO

The seminal characteristics of Moenkhausia oligolepis are described. Three males were induced with a single dose of carp pituitary. Semen was collected 6 h after induction, and diluted in dibasic sodium phosphate extender solution. For motility analysis, 1 µl of diluted semen was added to 10 µl of distilled water to achieve gamete activation. The average duration of total motility was 76.67 s; while the average sperm motility rate at intervals of 15 s was 95.3, 85.3, 59.6, 31.7, 13.0, 4.6 and 1.2%. To determine sperm concentration in samples, 0.5 µl of semen was diluted with 500 µl of glutaraldehyde. An aliquot of 10 µl of this dilution was utilized for cell counting. An average count of 4.97 × 109 ± 3.46 sperm/ml was obtained. Morphological analyses were performed using eosin-nigrosine dye; 20.33% of the sperm were observed to be dead. Live sperm, comprising the other 79.67%, had an average length of approximately 30 µm, with a head diameter of 4.488 ± 0.7 µm; and a flagella plus mid-piece length of 26.071 ± 12.4 µm. Of those sperm, 69% had a normal morphology, while 31% had primary and secondary abnormalities. The observed abnormality rate did not have a detrimental effect on artificial fertilization potential for the species. The description of the seminal characteristics of a species is one of the most important sets of information required for artificial reproduction of fish in captivity. It also contributes significantly to the total biological knowledge of the studied species.

Improper activation and swelling of in vitro produced eggs of European eel, Anguilla anguilla, has been shown to negatively affect embryonic development and hatching. We investigated this phenomenon by examining the effects of salinity and sea salt type on egg dimensions, cell cleavage patterns and egg buoyancy. Egg diameter after activation, using natural seawater adjusted to different salinities, varied among female eels, but no consistent pattern emerged. Activation salinities between 30-40 practical salinity unit (psu) produced higher quality eggs and generally larger egg diameters. Chorion diameters reached maximal values of 1642 ± 8 µm at 35 psu. A positive relationship was found between egg neutral buoyancy and activation salinity. Nine salt types were investigated as activation and incubation media. Five of these types induced a substantial perivitelline space (PVS), leading to large egg sizes, while the remaining four salt types resulted in smaller eggs. All salt types except NaCl treatments led to high fertilization rates and had no effect on fertilization success as well as egg neutral buoyancies at 7 h post-fertilization. The study points to the importance of considering ionic composition of the media when rearing fish eggs and further studies are encouraged.

The mangrove oyster Crassostrea rhizophorae is identified as a potentially valuable species for tropical aquaculture, however, information on the physiological mechanisms of reproduction under laboratory conditions for this species is limited. This study investigated the effects of salinity at different concentrations (15, 20, 25, 30, 35, and 40 g/L) on the induction of germinal vesicle breakdown (GVBD) of oocytes obtained through stripping, the release of polar bodies (PB1 and PB2), and the larval development of the mangrove oyster. The results revealed a relationship between salinity and the percentage of GVBD, with the most effective range being 30-40 g/L within the hydration time frame between 70 and 120 min. The release of 50 % of PB1 was detected within this salinity range, while for the release of 50 % of PB2, the saline treatments of 35 and 40 g/L showed the best results. Overall, the salinity range of 30-40 g/L is suggested as the most suitable of polyploidy induction methodologies through the retention of PB1 or PB2. Regarding larval hatching, while salinities between 25 and 40 g/L presented similar percentages, at 15 g/L no hatching was observed. This study demonstrated that salinity is a key factor in early pre- and post-fertilization stages for the successful reproduction of mangrove oyster in hatcheries and that the percentages of oocyte maturation and artificial fertilization can be optimized by adjusting salinity.

The aim was to evaluate the effect of a post-thaw dilution of Rhamdia quelen sperm in 1.1% NaCl (325 mOsm kg-1; pH 7.6; 24 °C) solution on the quality and reproductive capacity. Sperm from eight males were cryopreservation in nitrogen vapor at - 170 °C for 18 h in 0.25 mL straws in a freezing medium containing 5% fructose, 5% Powdered milk, and 10% methanol. The samples were thawed and post-thaw diluted (1:20) in NaCl solution or not (control). The higher spermatozoa velocities were observed in the post-thaw diluted samples (curvilinear (VCL) - 69 ± 11 µm s-1; average path (VAP) - 45 ± 8 µm s-1; straight-line (VSL) - 43 ± 8 µm s-1) compared to the control (VCL - 47 ± 10 µm s-1; VAP - 31 ± 6 µm s-1; VSL - 30 ± 6 µm s-1). Greater straightness (STR), progression (PROG), and beat cross frequency (BCF) were observed in the post-thaw diluted samples (STR - 96 ± 7%; PROG - 666 ± 128 µm; BCF - 42 ± 2 Hz) than in control (STR - 95 ± 5%; PROG - 463 ± 92 µm; BCF - 40 ± 2 Hz). The strongly curled tail was the only morphology change that differ between the post-thaw diluted (5 ± 2%) and control (2 ± 1%). Membrane integrity, mitochondrial activity, and normal larvae rate were not different between treatments. Fertilization and hatching were higher in the post-thaw diluted sperm (93 ± 3%; 82 ± 9%) when compared to control samples (65 ± 13%; 55 ± 17%). Were used oocytes from one female, limiting these results. The post-thaw dilution improved the sperm kinetics and reproductive parameters. Thus, this methodology can be included in the sperm cryopreservation protocol for R. quelen.

Cultured Senegalese sole (Solea senegalensis) breeders fail to spawn fertilized eggs. The implantation of large-scale in vitro fertilization protocols, to solve this problem, has been frustrated by low production of poor quality sperm. Cultured females were induced to ovulate with a 5 µg kg-1 single injection of gonadotropin-releasing hormone agonist (GnRHa) and viable eggs (82.6 ± 9.2% fertilization) were stripped 41:57 ± 1:46 h after the injection. Sperm was collected from cultured males, diluted in modified Leibovitz and used fresh to fertilize the eggs. Males were not treated with hormones. A nonlinear regression, an exponential rise to a maximum (R = 0.93, p < 0.0001) described the number of motile spermatozoa required to fertilize a viable egg and 1617 motile spermatozoa were sufficient to fertilize 99 ± 12% (±95% CI) of viable eggs. Similar, spermatozoa egg-1 ratios of 592 ± 611 motile spermatozoa egg-1 were used in large-scale in vitro fertilizations (190 512 ± 38 471 eggs). The sperm from a single male (145 ± 50 µl or 8.0 ± 6.8 × 108 spermatozoa) was used to fertilize the eggs. The mean hatching rate was 70 ± 14% to provide 131 540 ± 34 448 larvae per fertilization. The viability of unfertilized eggs stored at room temperature decreased gradually, and the sooner eggs were fertilized after stripping, the higher the viability of the eggs. The collection of sperm directly into a syringe containing modified Leibovitz significantly increased the percentage of motile spermatozoa (33.4 ± 12.2%) compared with other collection methods. The spz egg-1 ratios for Senegalese sole were at the lower end of ratios required for fish. Senegalese sole have a pair-spawning reproductive behaviour characterized by gamete fertilization in close proximity with no sperm competition. The provision of a large-scale in vitro fertilization protocol (200 µl of sperm per 100 ml of eggs) will enable the industry to operate sustainably and implement breeding programmes to improve production.

Although paedomorphosis is widespread across salamander families, only two species have ever been documented to exhibit paedomorphosis in Hynobiidae. One of these two exceptional species is Hynobius retardatus in which paedomorphosis was first reported in 1924, in specimens from Lake Kuttara in Hokkaido. This population became extinct after the last observation in 1932; since then, no paedomorphs of this species have been reported anywhere. Here, we report the rediscovery of paedomorphs of this species. Three paedomorph-like male salamanders were collected from a pond in the south Hokkaido in December 2020 and April 2021; in size, these specimens were similar to metamorphosed adults but they still displayed larval features such as external gills and a well-developed caudal fin. An artificial fertilization experiment demonstrated that they were sexually compatible with metamorphosed females, thus, confirming them to be paedomorphs. Future efforts to find additional paedomorphs in this and other populations are required to assess the prevalence of paedomorphosis in H. retardatus and to improve understanding of the ecology and evolution of paedomorphisis in Urodela.

BACKGROUND: Little is known about the life history of oarfish of the genus Regalecus, although it is a famous deep-sea fish and an apparent origin of sea serpent legends. We successfully performed artificial insemination using a recently dead pair of sexually mature individuals. We report for the first time development from fertilized eggs to early larvae in the Lampridiformes. RESULTS: Eggs required 18 days of development from fertilization to hatching under 20.5-22.5 °C conditions. Oarfish larvae had similar morphological features as other lampridiform larvae hatched in the ocean. Larvae typically faced downward and swam using pectoral fins; they frequently opened their mouths. This mouth-opening behavior and swimming ability were both consistent with osteological development. The larvae did not eat and died four days after hatching. CONCLUSIONS: This is the first successful instance of artificial insemination and hatching in the oarfish, as well as the first reliable morphological and behavioral description of lampridiform larvae.

For externally fertilizing fishes, interactions between male and female gametes have been shown to have remarkable impacts on sperm performance. Ovarian fluid (OF) and its ability to alter the swimming behavior of fish sperm makes it a determining factor of fertility. With the expansion of channel catfish (Ictalurus punctatus) ♀ × blue catfish (Ictalurus furcatus) ♂ hybrid aquaculture, it is essential to understand the impacts during fertilization and the magnitude such gametic interactions have on sperm performance and subsequent male fertility potential. This study was conducted to address the following: 1) activate blue catfish sperm with/without channel catfish OF to determine impacts on sperm performance and 2) assess if sperm behave differently when activated in the OF from individual females. Sperm (n = 4 males) were activated without OF (control) and with diluted OF from unique females (n = 6), creating 24 experimental crosses. Sperm motility (%), velocity (VCL), and longevity were analyzed using computer assisted sperm analyses software. With OF incorporated in the activation media, sperm velocity was significantly higher than the control at 10, 20, and 30 s post-activation. OF did not have an impact on motility for any females at 10 s and 20 s post-activation but became significantly higher than the control at 30 s. In all cases, OF treatments greatly increased longevity. Male × female interactions were highly significant, such that motility, velocity, and longevity were dependent on specific male-female pairs. This information shows that OF should be incorporated in aquatic media to simulate natural spawning conditions and accurately assess the fluid mechanics of sperm propulsion for each male. Additionally, there are mechanisms that drive gamete interactions that need to be explored further, which may improve selection of male-female pairs for in-vitro fertilization. On a broad scale, our results also help to shed light on the complexities of fertilization and fish reproduction overall, which may have implications for recruitment variability and recovery strategies of threatened and/or endangered freshwater species.

Due to some biological characteristics of the brown-marbled grouper, there are challenges in aquaculture enterprises focused on production of this fish. With the aim of facilitating captive breeding in brown-marbled grouper, effects on gamete post-activation, post-stripping egg storage and sperm to egg ratio on fertilization success were investigated. Results indicated eggs were fertile for 4 min after placement in seawater, thereafter, there was a reduction in fertilization. Initial curvilinear velocity (VCL) and total motility of fresh sperm was 129.3 ±â€¯3.7 µm/s and 92.9 ±â€¯2.4 %, both of which decreased as time elapsed post-activation, however, there was no significant difference in fertilization until 6 min post-activation of sperm. Compared with fertilization and hatching rates in eggs used immediately after collection (95.0 ±â€¯2.8 % and 90.1 ±â€¯4.2 %), in vitro storage of eggs had no detectable effect on fertilized egg and hatching rates until 60 min post-stripping (92.0 ±â€¯2.8 % and 89.9 ±â€¯2.7 %), indicating that immediate use of brown-marbled grouper eggs for fertilization is not necessary in aquaculture hatcheries. With regard to optimizing the sperm to egg ratio, there was acceptable fertilization with ratios ranging from 103 to 107, whereas there was a marked decrease in fertilization rate when there were lesser sperm concentrations. When the combined results from this study are considered, there could be improvements of artificial fertilization efficiency in the brown-marbled grouper with utilization of these findings in aquaculture enterprises focused on production of this fish.

Males compete pre- and post-mating to fertilize the maximum number of eggs. In polyandry, sperm competition occurs when sperm from two or more males compete to fertilize eggs from a female. Here we review how sperm competition from hatchery origin fish can cause loss of genetic variability in fish populations kept in captivity and in wild populations. In fish hatchery practices, sperm competition occurs in mass spawners that release gametes in tanks, and in artificial fertilizations when pooled semen is used. In mass spawnings sperm competition is difficult to tease apart from pre-mating competition and other post-mating selective mechanisms, whereas, studies focused on the use of pooled semen in different fish species have shown a clear relationship between sperm motility parameters and precedence in fertilization. In both situations, sperm competition will result in a loss of genetic variability that accumulates over generations, but hatchery protocols can be adjusted to mitigate it. Another source of concern regarding sperm competition for hatchery produced fish is the spatial and temporal overlap in spawning with wild individuals, either via aquaculture escapees or purposeful stocking programs. This may result in sperm competition between hatchery origin and wild males and impact natural populations. Our review suggests that in order to give every adult selected as broodstock an equal opportunity to produce offspring in captivity, mass spawning and the use of pooled semen should be limited.

Production of quality seeds is a critical need, and cryopreservation serves as an important biotechnological tool in achieving that goal. In the present study, the effects of different extenders and dilution ratios on the spermatological and fertilization parameters of cryopreserved milt were evaluated in common carp brooders. Milt was collected from Cyprinus carpio brooders and diluted and cryopreserved at three dilution ratios such as 1:40, 1:80, and 1:120, using Freshwater Fish Saline (FWFS) and Modified Fish Ringer as extenders and dimethyl sulfoxide as cryoprotectant (90:10). Motility duration, fertilization, and hatching rates were studied after different treatments. Rapid freezing of straws was done for 20 minutes at a freezing rate of -78°C/min. It can be concluded that C. carpio milt can be cryopreserved with FWFS at a dilution ratio of 1:40 to obtain the highest post-thaw motility duration (60.33 ± 1.52 seconds), fertilization rate (87.6% ± 1.52%), and hatching rate (57.1% ± 4.16%). The difference in the values between extenders, dilution ratios, and cryopreserved and fresh milt were statistically significant (p < 0.05) when analyzed using ANOVA.

The development of a new protocol for egg fertilization may increase embryo survival and benefit the aquaculture process. In the present study, a new technique of partially adding sperm to activated eggs in the artificial fertilization of burbot (Lota lota), ide (Leuciscus idus) and asp (Aspius aspius) eggs was evaluated. If the same volume of sperm was divided into two or three parts and added to eggs in 30-60s intervals, it significantly improved embryo survival at the eyed-egg-stage of development. In the present study, the periodic addition of spermatozoa to eggs affected fertilization (ide and asp) and embryo survival rates (ide, asp and burbot) and might be successfully applied under hatchery conditions.

The objective of this study was to assess the influence of temperature and time on the storage of fresh Steindachneridion parahybae oocytes. Two experiments were carried out: (1) the fertilization rates of oocytes exposed to temperatures of 5, 15, 28 (room temperature) and 35°C were assessed 15min (control), 115, 235 and 355min after release; (2) the fertilization and hatching rates, as well as the percentage of normal larvae of oocytes exposed to 14, 17 or 20°C, 20min (control) were assessed 50, 80 and 110min after stripping. In the first experiment, the highest fertilization rates (P<0.05) were obtained in the control treatment (15min, 28°C), with 74.34±5.48% oocytes showing loss of viability over time. In the second experiment, there was a reduction (P<0.05) in the fertilization rates at the temperatures and times tested. The artificial fertilization of S. parahybae oocytes is recommended immediately after collection, and if storage is necessary, it should be conducted at temperatures between 17 and 20°C.

As técnicas de fertilização artificial fazem parte da vida moderna. Porém, a que transformações subjetivas nos remetem esses avanços tecnológicos? A autora investiga as fantasias sobre o enigma das origens, examinando o caso clínico de um menino nascido de uma gestação de proveta e que chegou para tratamento. Parte da idéia de que o real vivido ingressa no aparelho psíquico empapado pelo imaginário, indo enlaçar-se com as inscrições históricas singulares ali existentes para assim produzir efeitos subjetivos; nesse sentido, este trabalho reflete também a respeito das novas formas de concepção, da produção de subjetividade e da constituição psíquica.


Artificial fertilization techniques are part of modern life. However, to what subjective transformations such technological progresses take us? This paper’s purpose is to examine the fantasies about the riddle of origins setting out from a clinical case of a boy who is the outcome of a assisted reproduction pregnancy that has come for treatment. The idea that the reality experienced enters the psychic apparatus filled by the imaginary is present becoming intertwined with existent singular historical inscriptions so as to produce subjective effects; in such a way that this paper also tends to reflect upon new ways of conception, the production of subjectivity and psychic constitution.


Las técnicas de fertilización artificial forman parte de la vida moderna. Más, a que transformaciones subjetivas nos remiten estos progresos tecnológicos? Este trabajo se propone examinar las fantasías sobre el enigma de los orígenes a partir de un caso clínico de un niño fruto de una gestación de probeta que vino a tratarse. Se parte de la idea de que lo real vivido ingresa en el aparato psíquico empapado por lo imaginario, mezclándose con las inscripciones históricas singulares allí existentes para así producir efectos subjetivos, de modo que este trabajo también trata de pensar a respecto de las nuevas formas de concepción, la producción de subjetividad y la constitución psíquica.
