RESUMO

Mutations in the nuclear structural protein lamin A produce rare, tissue-specific diseases called laminopathies. The introduction of a human Emery-Dreifuss muscular dystrophy (EDMD)-inducing mutation into the C. elegans lamin (LMN-Y59C), recapitulates many muscular dystrophy phenotypes, and correlates with hyper-sequestration of a heterochromatic array at the nuclear periphery in muscle cells. Using muscle-specific emerin Dam-ID in worms, we monitored the effects of the mutation on endogenous chromatin. An increased contact with the nuclear periphery along chromosome arms, and an enhanced release of chromosomal centers, coincided with the disease phenotypes of reduced locomotion and compromised sarcomere integrity. The coupling of the LMN-Y59C mutation with the ablation of CEC-4, a chromodomain protein that anchors H3K9-methylated chromatin at the nuclear envelope (NE), suppressed the muscle-associated disease phenotypes. Deletion of cec-4 also rescued LMN-Y59C-linked alterations in chromatin organization and some changes in transcription. Sequences that changed position in the LMN-Y59C mutant, are enriched for E2F (EFL-2)-binding sites, consistent with previous studies suggesting that altered Rb-E2F interaction with lamin A may contribute to muscle dysfunction. In summary, we were able to counteract the dominant muscle-specific defects provoked by LMNA mutation by the ablation of a lamin-associated H3K9me anchor, suggesting a novel therapeutic pathway for EDMD.

The loss of nitrogen in soil damages the environment. Clarifying the mechanism of ammonium nitrogen (NH4+-N) transport in soil and increasing the fixation of NH4+-N after N application are effective methods for improving N use efficiency. However, the main factors are not easily identified because of the complicated transport and retardation factors in different soils. This study employed machine learning (ML) to identify the main influencing factors that contribute to the retardation factor (Rf) of NH4+-N in soil. First, NH4+-N transport in the soil was investigated using column experiments and a transport model. The Rf (1.29 - 17.42) was calculated and used as a proxy for the efficacy of NH4+-N transport. Second, the physicochemical parameters of the soil were determined and screened using lasso and ridge regressions as inputs for the ML model. Third, six machine learning models were evaluated: Adaptive Boosting, Extreme Gradient Boosting (XGB), Random Forest, Gradient Boosting Regression, Multilayer Perceptron, and Support Vector Regression. The optimal ML model of the XGB model with a low mean absolute error (0.81), mean squared error (0.50), and high test r2 (0.97) was obtained by random sampling and five-fold cross-validation. Finally, SHapely Additive exPlanations, entropy-based feature importance, and permutation characteristic importance were used for global interpretation. The cation exchange capacity (CEC), total organic carbon (TOC), and Kaolin had the greatest effects on NH4+-N transport in the soil. The accumulated local effect offered a fundamental insight: When CEC > 6 cmol+ kg-1, and TOC > 40 g kg-1, the maximum resistance to NH4+-N transport within the soil was observed. This study provides a novel approach for predicting the impact of the soil environment on NH4+-N transport and guiding the establishment of an early-warning system of nutrient loss.

This paper aims to improve the steering performance of the Ackermann personal mobility scooter based on a new meta-heuristic optimization algorithm named Differential Harris Hawks Optimization (DHHO) and the modeling of the steering encoder. The steering response in the Ackermann mechanism is crucial for automated driving systems (ADS), especially in localization and path-planning phases. Various methods presented in the literature are used to control the steering, and meta-heuristic optimization algorithms have achieved prominent results. Harris Hawks optimization (HHO) algorithm is a recent algorithm that outperforms state-of-the-art algorithms in various optimization applications. However, it has yet to be applied to the steering control application. The research in this paper was conducted in three stages. First, practical experiments were performed on the steering encoder sensor that measures the steering angle of the Landlex mobility scooter, and supervised learning was applied to model the results obtained for the steering control. Second, the DHHO algorithm is proposed by introducing mutation between hawks in the exploration phase instead of the Hawks perch technique, improving population diversity and reducing premature convergence. The simulation results on CEC2021 benchmark functions showed that the DHHO algorithm outperforms the HHO, PSO, BAS, and CMAES algorithms. The mean error of the DHHO is improved with a confidence level of 99.8047% and 91.6016% in the 10-dimension and 20-dimension problems, respectively, compared with the original HHO. Third, DHHO is implemented for interactive real-time PID tuning to control the steering of the Ackermann scooter. The practical transient response results showed that the settling time is improved by 89.31% compared to the original response with no overshoot and steady-state error, proving the superior performance of the DHHO algorithm compared to the traditional control methods.

The complex work of addressing intergenerational adversities, like violence, abuse, and neglect through perinatal psychotherapy, is understudied. Especially noticeable is the paucity of studies giving voice to the therapists. This study explored therapeutic processes through the perspectives of seven Norwegian therapists. A qualitative approach was chosen with individual interviews and a follow-up focus group. Data were analyzed using reflexive thematic analysis. We identified one overarching theme: To maintain a reflective therapeutic capacity, and two main themes with associated subthemes: 1) Alliance work when a caring system comes to therapy to fight generations of adversities and 2) The complex therapeutic work of addressing generational adversities in perinatal psychotherapy. Findings from the present study indicate that maintaining a reflective stance is essential yet challenging when addressing intergenerational adversities, requiring a holding environment for the therapists. The primary vehicle of change was perceived as a safe enough therapeutic alliance to explore new ways of being together, contrasting earlier experiences. A key question raised was how to give caregivers enough time to trust the therapist without compromising the safety and development of the child. The essence of the therapeutic work was to reduce risk factors and increase protective factors through multiple ports of entry.


El complejo trabajo de abordar el tema de adversidades intergeneracionales, como violencia, abuso y negligencia a través de la sicoterapia perinatal, no se ha estudiado lo suficiente. Especialmente perceptible es la escasez de estudios que les den voz a los terapeutas. Este estudio exploró los procesos terapéuticos a través de perspectivas de siete terapeutas noruegos. Se escogió un acercamiento cualitativo con entrevistas individuales y un grupo de enfoque como seguimiento. Se analizaron los datos usando un análisis temático reflexivo. Identificamos un tema global: Mantener una capacidad terapéutica reflexiva, así como dos temas principales con subtemas asociados: 1) Trabajo en alianza cuando un sistema de cuidados llega a la terapia, para combatir generaciones de adversidades y 2) El complejo trabajo terapéutico de abordar el tema de adversidades generacionales en la sicoterapia perinatal. Los resultados del presente estudio indicaron que mantener una postura de reflexión es esencial, aunque desafiante, cuando se aborda el tema de adversidades generacionales, lo cual requiere un entorno o espacio favorable para los terapeutas. La percepción es que el vehículo primario para el cambio es una suficientemente segura alianza terapéutica para explorar nuevas maneras de vivir juntos, contrastando las experiencias anteriores. Una pregunta clave que se planteó fue cómo darles a los cuidadores suficiente tiempo para que le tengan confianza al terapeuta sin comprometer la seguridad y el desarrollo del niño. La esencia del trabajo terapéutico fue reducir los factores de riesgo e incrementar los factores de protección por medio de múltiples puertas de entrada.


Le travail complexe qui consiste à s'attaquer aux adversités intergénérationnelles comme la violence, la maltraitance et la négligence au travers de la psychothérapie périnatale est sous-étudié. On constate surtout la rareté d'études donnant la parole aux thérapeutes. Cette étude a exploré les processus thérapeutiques au travers des perspectives de sept thérapeutes norvégiens. Une approche qualitative a été choisie avec des entretiens individuels et un groupe d'étude de suivi. Des données ont été analysées en utilisant une analyse réflective thématique. Nous avons identifié un thème général: maintenir une capacité thérapeutique réflective et deux thèmes principaux avec des sous-thèmes liés: 1) le travail d'alliance quand un système attentif en arrive à la thérapie pour contrecarrer des générations d'adversités et 2) le travail thérapeutique complexe qui consiste à s'attaquer aux adversités intergénérationnelles en psychothérapie périnatale. Les résultats de cette étude ont indiqué que le maintien d'une position de réflexion est essentiel mais pose également un défi lorsqu'on on s'attaque aux adversités intergénérationnelles, exigeant un environnement thérapeutique soutenant pour les thérapeutes. Le principal véhicule de changement a été perçu comme une alliance thérapeutique étant assez sûre pour l'exploration de nouvelles façons d'être ensemble, en contraste avec des expériences précédentes. Une question clé qui a été soulevée était de comment donner aux personnes prenant soin des enfants assez de temps pour avoir confiance au thérapeute sans compromettre la sécurité et le développement de l'enfant. L'essence du travail thérapeutique a consisté à réduire les facteurs de risque et à accroître les facteurs protecteurs au travers de plusieurs ports d'entrée.

BACKGROUND: Probiotics have gained attention for their potential maintaining gut and immune homeostasis. They have been found to confer protection against pathogen colonization, possess immunomodulatory effects, enhance gut barrier functionality, and mitigate inflammation. However, a thorough understanding of the unique mechanisms of effects triggered by individual strains is necessary to optimize their therapeutic efficacy. Probiogenomics, involving high-throughput techniques, can help identify uncharacterized strains and aid in the rational selection of new probiotics. This study evaluates the potential of the Escherichia coli CEC15 strain as a probiotic through in silico, in vitro, and in vivo analyses, comparing it to the well-known probiotic reference E. coli Nissle 1917. Genomic analysis was conducted to identify traits with potential beneficial activity and to assess the safety of each strain (genomic islands, bacteriocin production, antibiotic resistance, production of proteins involved in host homeostasis, and proteins with adhesive properties). In vitro studies assessed survival in gastrointestinal simulated conditions and adhesion to cultured human intestinal cells. Safety was evaluated in BALB/c mice, monitoring the impact of E. coli consumption on clinical signs, intestinal architecture, intestinal permeability, and fecal microbiota. Additionally, the protective effects of both strains were assessed in a murine model of 5-FU-induced mucositis. RESULTS: CEC15 mitigates inflammation, reinforces intestinal barrier, and modulates intestinal microbiota. In silico analysis revealed fewer pathogenicity-related traits in CEC15, when compared to Nissle 1917, with fewer toxin-associated genes and no gene suggesting the production of colibactin (a genotoxic agent). Most predicted antibiotic-resistance genes were neither associated with actual resistance, nor with transposable elements. The genome of CEC15 strain encodes proteins related to stress tolerance and to adhesion, in line with its better survival during digestion and higher adhesion to intestinal cells, when compared to Nissle 1917. Moreover, CEC15 exhibited beneficial effects on mice and their intestinal microbiota, both in healthy animals and against 5FU-induced intestinal mucositis. CONCLUSIONS: These findings suggest that the CEC15 strain holds promise as a probiotic, as it could modulate the intestinal microbiota, providing immunomodulatory and anti-inflammatory effects, and reinforcing the intestinal barrier. These findings may have implications for the treatment of gastrointestinal disorders, particularly some forms of diarrhea.

Understanding the drivers of ecosystem stability has been a key focus of modern ecology as the impacts of the Anthropocene become more prevalent and extreme. Marine protected areas (MPAs) are tools used globally to promote biodiversity and mediate anthropogenic impacts. However, assessing the stability of natural ecosystems and responses to management actions is inherently challenging due to the complex dynamics of communities with many interdependent taxa. Using a 12-year time series of subtidal community structure in an MPA network in the Channel Islands (United States), we estimated species interaction strength (competition and predation), prey species synchrony, and temporal stability in trophic networks, as well as temporal variation in sea surface temperature to explore the causal drivers of temporal stability at community and metacommunity scales. At the community scale, only trophic networks in MPAs at Santa Rosa Island showed greater temporal stability than reference sites, likely driven by reduced prey synchrony. Across islands, competition was sometimes greater and predation always greater in MPAs compared with reference sites. Increases in interaction strength resulted in lower temporal stability of trophic networks. Although MPAs reduced prey synchrony at the metacommunity scale, reductions were insufficient to stabilize trophic networks. In contrast, temporal variation in sea surface temperature had strong positive direct effects on stability at the regional scale and indirect effects at the local scale through reductions in species interaction strength. Although MPAs can be effective management strategies for protecting certain species or locations, our findings for this MPA network suggest that temperature variation has a stronger influence on metacommunity temporal stability by mediating species interactions and promoting a mosaic of spatiotemporal variation in community structure of trophic networks. By capturing the full spectrum of environmental variation in network planning, MPAs will have the greatest capacity to promote ecosystem stability in response to climate change.


Efectos de la protección y variación de la temperatura sobre la estabilidad temporal en una red de reservas marinas Resumen El conocimiento sobre las causas de la estabilidad ambiental ha sido un enfoque importante de la ecología moderna conforme el impacto del Antropoceno se vuelve más prevaleciente y extremo. Las áreas marinas protegidas (AMP) son herramientas que se usan en todo el mundo para promover la biodiversidad y mediar el impacto antropogénico. Sin embargo, analizar la estabilidad de los ecosistemas naturales y la respuesta a las acciones de manejo es complicado debido a las dinámicas complejas entre las comunidades y varios taxones interdependientes. Entre las islas, la competencia a veces fue mayor y la depredación siempre fue mayor en las AMP comparadas con los sitios de referencia. Los incrementos en la fuerza de interacción causaron una menor estabilidad temporal en las redes tróficas. Aunque las AMP redujeron la sincronía de presas a nivel meta-comunitario, las reducciones no fueron suficientes para estabilizar las redes tróficas. Invasive species can dramatically alter ecosystems, but eradication is difficult, and suppression is expensive once they are established. Uncertainties in the potential for expansion and impacts by an invader can lead to delayed and inadequate suppression, allowing for establishment. Metapopulation viability models can aid in planning strategies to improve responses to invaders and lessen invasive species' impacts, which may be particularly important under climate change. We used a spatially explicit metapopulation viability model to explore suppression strategies for ecologically damaging invasive brown trout (Salmo trutta), established in the Colorado River and a tributary in Grand Canyon National Park. Our goals were to estimate the effectiveness of strategies targeting different life stages and subpopulations within a metapopulation; quantify the effectiveness of a rapid response to a new invasion relative to delaying action until establishment; and estimate whether future hydrology and temperature regimes related to climate change and reservoir management affect metapopulation viability and alter the optimal management response. Our models included scenarios targeting different life stages with spatially varying intensities of electrofishing, redd destruction, incentivized angler harvest, piscicides, and a weir. Quasi-extinction (QE) was obtainable only with metapopulation-wide suppression targeting multiple life stages. Brown trout population growth rates were most sensitive to changes in age 0 and large adult mortality. The duration of suppression needed to reach QE for a large established subpopulation was 12 years compared with 4 with a rapid response to a new invasion. Isolated subpopulations were vulnerable to suppression; however, connected tributary subpopulations enhanced metapopulation persistence by serving as climate refuges. Water shortages driving changes in reservoir storage and subsequent warming would cause brown trout declines, but metapopulation QE was achieved only through refocusing and increasing suppression. Our modeling approach improves understanding of invasive brown trout metapopulation dynamics, which could lead to more focused and effective invasive species suppression strategies and, ultimately, maintenance of populations of endemic fishes.

La cuasi extinción (CE) sólo se obtuvo con una supresión a nivel meta poblacional enfocada en múltiples etapas de vida. Las tasas de crecimiento poblacional de la trucha fueron más sensibles a los cambios en edad cero y una gran mortalidad adulta. La duración de la supresión requerida para llegar a la CE para una subpoblación grande establecida fue de doce años en comparación con los cuatro de una respuesta rápida a una nueva invasión. Las subpoblaciones aisladas fueron vulnerables a la supresión; sin embargo, las subpoblaciones conectadas por medio de tributarios incrementaron la persistencia meta poblacional al fungir como refugios climáticos. Ladybirds (Coleoptera: Coccinellidae) provide services that are critical to food production, and they fulfill an ecological role as a food source for predators. The richness, abundance, and distribution of ladybirds, however, are compromised by many anthropogenic threats. Meanwhile, a lack of knowledge of the conservation status of most species and the factors driving their population dynamics hinders the development and implementation of conservation strategies for ladybirds. We conducted a review of the literature on the ecology, diversity, and conservation of ladybirds to identify their key ecological threats. Ladybird populations are most affected by climate factors, landscape composition, and biological invasions. We suggest mitigating actions for ladybird conservation and recovery. Short-term actions include citizen science programs and education, protective measures for habitat recovery and threatened species, prevention of the introduction of non-native species, and the maintenance and restoration of natural areas and landscape heterogeneity. Mid-term actions involve the analysis of data from monitoring programs and insect collections to disentangle the effect of different threats to ladybird populations, understand habitat use by taxa on which there is limited knowledge, and quantify temporal trends of abundance, diversity, and biomass along a management-intensity gradient. Long-term actions include the development of a worldwide monitoring program based on standardized sampling to fill data gaps, increase explanatory power, streamline analyses, and facilitate global collaborations.


Las catarinas (Coleoptera: Coccinellidae) proporcionan servicios que son críticos para la producción de alimento, y juegan un papel ecológico como fuente de alimento para depredadores. Sin embargo, la riqueza, abundancia y distribución de catarinas están en peligro debido a muchas amenazas antropogénicas. La carencia de conocimiento sobre el estatus de conservación de la mayoría de las especies y los factores que inciden en su dinámica poblacional dificulta el desarrollo e implementación de estrategias de conservación para las catarinas. Acciones a mediano plazo implican el análisis de datos obtenidos de programas de monitoreo y colecciones de insectos para desenmarañar el efecto de las diferentes amenazas a las poblaciones de catarinas, comprender el uso del hábitat por taxa de los que se tiene conocimiento limitado y cuantifica las tendencias temporales de la abundancia, diversidad y biomasa a lo largo de un gradiente de intensidad de manejo. Acciones a largo plazo incluyen el desarrollo de un programa de monitoreo a nivel mundial basado en muestreos estandarizados para subsanar la falta de datos, incrementar el poder explicativo, optimizar los análisis y facilitar colaboraciones globales.

Cholesterol efflux capacity (CEC) is of interest given its potential relationship with several important clinical conditions including Alzheimer's disease. The inactivation of the APOE locus in mouse models supports the idea that it is involved in determining the CEC. With that in mind, we examine the impact of the plasma metabolome profile and the APOE genotype on the CEC in cognitively healthy elderly subjects. The study subjects were 144 unrelated healthy individuals. The plasma CEC was determined by exposing cultured mouse macrophages treated with BODIPY-cholesterol to human plasma. The metabolome profile was determined using NMR techniques. Multiple regression was performed to identify the most important predictors of CEC, as well as the NMR features most strongly associated with the APOE genotype. Plasma 3-hydroxybutyrate was the variable most strongly correlated with the CEC (r = 0.365; p = 7.3 × 10-6). Male sex was associated with a stronger CEC (r = -0.326, p = 6.8 × 10-5). Most of the NMR particles associated with the CEC did not correlate with the APOE genotype. The NMR metabolomics results confirmed the APOE genotype to have a huge effect on the concentration of plasma lipoprotein particles as well as those of other molecules including omega-3 fatty acids. In conclusion, the CEC of human plasma was associated with ketone body concentration, sex, and (to a lesser extent) the other features of the plasma lipoprotein profile. The APOE genotype exerted only a weak effect on the CEC via the modulation of the lipoprotein profile. The APOE locus was associated with omega-3 fatty acid levels independent of the plasma cholesterol level.

RESUMO

The heuristic optimization algorithm is a popular optimization method for solving optimization problems. A novel meta-heuristic algorithm was proposed in this paper, which is called the Willow Catkin Optimization (WCO) algorithm. It mainly consists of two processes: spreading seeds and aggregating seeds. In the first process, WCO tries to make the seeds explore the solution space to find the local optimal solutions. In the second process, it works to develop each optimal local solution and find the optimal global solution. In the experimental section, the performance of WCO is tested with 30 test functions from CEC 2017. WCO was applied in the Time Difference of Arrival and Frequency Difference of Arrival (TDOA-FDOA) co-localization problem of moving nodes in Wireless Sensor Networks (WSNs). Experimental results show the performance and applicability of the WCO algorithm.

Global optimization problems have been a research topic of great interest in various engineering applications among which neural network algorithm (NNA) is one of the most widely used methods. However, it is inevitable for neural network algorithms to plunge into poor local optima and convergence when tackling complex optimization problems. To overcome these problems, an improved neural network algorithm with quasi-oppositional-based and chaotic sine-cosine learning strategies is proposed, that speeds up convergence and avoids trapping in a local optimum. Firstly, quasi-oppositional-based learning facilitated the exploration and exploitation of the search space by the improved algorithm. Meanwhile, a new logistic chaotic sine-cosine learning strategy by integrating the logistic chaotic mapping and sine-cosine strategy enhances the ability that jumps out of the local optimum. Moreover, a dynamic tuning factor of piecewise linear chaotic mapping is utilized for the adjustment of the exploration space to improve the convergence performance. Finally, the validity and applicability of the proposed improved algorithm are evaluated by the challenging CEC 2017 function and three engineering optimization problems. The experimental comparative results of average, standard deviation, and Wilcoxon rank-sum tests reveal that the presented algorithm has excellent global optimality and convergence speed for most functions and engineering problems.

This paper proposes an enhanced version of Equilibrium Optimizer (EO) called (EEO) for solving global optimization and the optimal power flow (OPF) problems. The proposed EEO algorithm includes a new performance reinforcement strategy with the Lévy Flight mechanism. The algorithm addresses the shortcomings of the original Equilibrium Optimizer (EO) and aims to provide better solutions (than those provided by EO) to global optimization problems, especially OPF problems. The proposed EEO efficiency was confirmed by comparing its results on the ten functions of the CEC'20 test suite, to those of other algorithms, including high-performance algorithms, i.e., CMA-ES, IMODE, AGSK and LSHADE_cnEpSin. Moreover, the statistical significance of these results was validated by the Wilcoxon's rank-sum test. After that, the proposed EEO was applied to solve the the OPF problem. The OPF is formulated as a nonlinear optimization problem with conflicting objectives and subjected to both equality and inequality constraints. The performance of this technique is deliberated and evaluated on the standard IEEE 30-bus test system for different objectives. The obtained results of the proposed EEO algorithm is compared to the original EO algorithm and those obtained using other techniques mentioned in the literature. These Simulation results revealed that the proposed algorithm provides better optimized solutions than 20 published methods and results as well as the original EO algorithm. The EEO superiority was demonstrated through six different cases, that involved the minimization of different objectives: fuel cost, fuel cost with valve-point loading effect, emission, total active power losses, voltage deviation, and voltage instability. Also, the comparison results indicate that EEO algorithm can provide a robust, high-quality feasible solutions for different OPF problems.

Breast cancer is the second leading cause of cancer death in women after lung cancer. The first-line treatment of metastatic breast cancer in premenopausal women relies on tamoxifen. The development of tamoxifen resistance is not fully understood. In this study, capillary electrophoresis with capacitively coupled contactless conductivity detector was developed to monitor the changes in lactate and pyruvate levels in supernatant media of three models of developed MCF-7 tamoxifen-resistant cells and correlate these metabolites changes with lactate dehydrogenase genes expression and glucose consumption. The electrophoretic separation was achieved under reversed electroosmotic flow conditions. The linear ranges were 0.15-5 and 0.01-1 mM with a correlation coefficient of 0.9966 and 0.9971 and the limits of detection were 0.01 and 0.02 µM for lactate and pyruvate, respectively. Inter- and intrarun accuracy were in the range of 96.88-105.94% with precision (CV, %) of ≤7.35%. The method was completely validated and the results were in agreement with those obtained using the lactate and glucose assay kits. The results revealed a significant increase in both lactate and pyruvate production in the three tamoxifen-resistant MCF-7 cells models compared to control cells. This increase was correlated with the increase of lactate dehydrogenase genes expression and the increase of glucose consumption.

Aim: To present the clinicopathologic findings of the second case of androgen-secreting adult granulosa cell tumor (AGCT) in a woman with polycystic ovary syndrome (PCOS) and discuss in the light of the literature. Methods: Description of a case and discussion of the literature. Results: A patient with oligomenorrhea, amenorrhea and hirsutism who was diagnosed as PCOS and treated by oral contraceptive for three years, then left ovarian solid and liquid mass was found and pathologically confirmed to be androgen-secreting AGCT after left oophorectomy. She got regular menstrual cycle and gave birth naturally, but clinical features of PCOS reappeared after breastfeeding. Conclusion: Androgen-secreting AGCT and PCOS have similar clinical features of hyperandrogenism, it is difficult to diagnose androgen-secreting AGCT when both diseases occur in the same patient. If the size of cystic mass in androgen-secreting AGCT is too small to differentiate from PCOM on imaging, pathological examination after surgery may be the only way to find the disease.

To broaden our understanding of a split alliance in family therapy, we investigated the frequencies and correlates of sessions in which therapists, youth, and caregivers reported markedly different perceptions of the alliance. The sample consisted of 156 Spanish families who received Alliance Empowerment Family Therapy (Escudero, Adolescentes y familias en conflicto, 2013) for child maltreatment. Family members and therapists rated the alliance on the SOFTA-s (Friedlander et al., Journal of Counseling Psychology, 2006, 53, 214) after sessions 3, 6, and 9; family members rated their perceptions of treatment progress before sessions 4, 7, and 10. A cluster analysis differentiated sessions with a split adult-youth alliance (27.7%) from a split family-therapist alliance (44.1%), and a balanced alliance (similar ratings across the three perspectives; 28.2%). Client-rated treatment progress was differentially associated with the type of alliance split and the average alliance rating, whereas better posttreatment outcomes (child functioning and family goal attainment) were associated with fewer sessions having either type of split alliance.


Con el fin de ampliar nuestros conocimientos de una alianza dividida en la terapia familiar, investigamos las frecuencias y las correlaciones de las sesiones en las cuales los terapeutas, los jóvenes y los cuidadores informaron percepciones marcadamente diferentes de la alianza. La muestra estuvo formada por 156 familias españolas que recibieron terapia familiar de empoderamiento y alianza (Escudero, 2013) para el maltrato infantil. Los miembros de la familia y los terapeutas calificaron la alianza en el SOATIF-s (Friedlander et al., 2006) después de las sesiones 3, 6 y 9; los familiares calificaron sus percepciones del avance del tratamiento antes de las sesiones 4, 7 y 10. Un análisis de agrupamiento diferenció las sesiones con una alianza dividida entre los adultos y los jóvenes (27.7 %) de una alianza dividida entre la familia y el terapeuta (44.1 %) y una alianza equilibrada (calificaciones similares entre las tres perspectivas; 28.2 %). El avance del tratamiento calificado por el paciente estuvo asociado diferencialmente con el tipo de alianza dividida y la calificación promedio de la alianza, mientras que los mejores resultados posteriores al tratamiento (el funcionamiento del niño y el logro de objetivos familiares) estuvieron asociados con menos sesiones que tenían cualquiera de los tipos de alianza dividida.

In psychotherapy research, adherence refers to the extent to which therapists deliver a treatment as intended. This study examined whether therapist adherence to two different manualized treatments was associated with improved client outcomes and whether the association was moderated by therapeutic alliance. The study sample included 320 video recordings of therapy sessions from 118 cases in a randomized controlled trial (RCT) comparing attachment-based family therapy (ABFT) with family-enhanced nondirective supportive therapy (FE-NST). Recordings were selected from early, middle, and late stages of treatment. The adherence measure consisted of 24 items representing essential therapist interventions from both treatments. Trained raters coded tapes from both therapies. Adolescent self-report of alliance was measured at session 4. Adherence to ABFT was associated with a significant increase in family cohesion at mid-treatment but not at posttreatment. Adherence to FE-NST was significantly associated with an increase in suicide ideation posttreatment. Using therapeutic alliance as a moderator, adherence to ABFT was significantly associated with a reduction in suicide ideation, family conflict, and higher client satisfaction posttreatment. Alliance did not positively affect the association of FE-NST adherence to outcomes. Findings suggest that adherence to ABFT interventions may be better linked to treatment outcomes when adolescents feel a strong alliance with their therapist. Implications for future research and therapist training are explored.


En la investigación sobre psicoterapia, la adhesión se refiere al grado en el cual los terapeutas brindan un tratamiento según lo previsto. Este estudio analizó si la adhesión del terapeuta a dos tratamientos estandarizados diferentes estuvo asociada con mejores resultados en los pacientes y si la asociación estuvo moderada por la alianza terapéutica. La muestra del estudio incluyó 320 videograbaciones de sesiones de terapia de 118 casos en un ensayo aleatorizado controlado donde se comparó la terapia familiar basada en el apego (TFBA) con la terapia de apoyo no directiva optimizada por la familia (TAND-OF). Se eligieron grabaciones de las etapas iniciales, intermedias y finales del tratamiento. El instrumento de medición de la adhesión consistió en 24 ítems que representaban intervenciones esenciales del terapeuta de ambos tratamientos. Un grupo de calificadores capacitados codificaron las grabaciones de ambas terapias. El autoinforme de alianza de los adolescentes se midió en la cuarta sesión. La adhesión a la TFBA estuvo asociada con un aumento considerable de la cohesión familiar en la mitad del tratamiento, pero no después del tratamiento. La adhesión a la TAND-OF estuvo asociada considerablemente con un aumento de la ideación suicida después del tratamiento. Utilizando la alianza terapéutica como moderadora, la adhesión a la TFBA estuvo asociada considerablemente con una reducción de la ideación suicida, el conflicto familiar y una mayor satisfacción del paciente después del tratamiento. La alianza no afectó positivamente la asociación de la adhesión a la TAND-OF con los resultados. Los resultados sugieren que la adhesión a las intervenciones de TFBA puede asociarse mejor con los resultados del tratamiento cuando los adolescentes sienten una alianza fuerte con su terapeuta. Se analizan las implicancias para futuras investigaciones y para la capacitación de los terapeutas.

Numerical optimization has been a popular research topic within various engineering applications, where differential evolution (DE) is one of the most extensively applied methods. However, it is difficult to choose appropriate control parameters and to avoid falling into local optimum and poor convergence when handling complex numerical optimization problems. To handle these problems, an improved DE (BROMLDE) with the Bernstein operator and refracted oppositional-mutual learning (ROML) is proposed, which can reduce parameter selection, converge faster, and avoid trapping in local optimum. Firstly, a new ROML strategy integrates mutual learning (ML) and refractive oppositional learning (ROL), achieving stochastic switching between ROL and ML during the population initialization and generation jumping period to balance exploration and exploitation. Meanwhile, a dynamic adjustment factor is constructed to improve the ability of the algorithm to jump out of the local optimum. Secondly, a Bernstein operator, which has no parameters setting and intrinsic parameters tuning phase, is introduced to improve convergence performance. Finally, the performance of BROMLDE is evaluated by 10 bound-constrained benchmark functions from CEC 2019 and CEC 2020, respectively. Two engineering optimization problems are utilized simultaneously. The comparative experimental results show that BROMLDE has higher global optimization capability and convergence speed on most functions and engineering problems.

HDL-mediated cholesterol efflux capacity (CEC) may protect against cardiovascular disease. However, CEC assays are not standardized, hampering their application in large cohorts and comparison between studies. To improve standardization, we systematically investigated technical differences between existing protocols that influence assay performance that have not been previously addressed. CEC was measured in 96-well plates using J774A.1 macrophages labeled with BODIPY-cholesterol and incubated for 4 h with 2% apolipoprotein B-depleted human serum. The time zero method, which calculates CEC using control wells, and the per-well method, which calculates CEC based on the actual content of BODIPY-cholesterol in each well, were compared in 506 samples. We showed that the per-well method had a considerably lower sample rejection rate (4.74% vs. 13.44%) and intra-assay (4.48% vs. 5.28%) and interassay coefficients of variation (two controls: 7.85%, 9.86% vs. 13.58%, 15.29%) compared with the time zero method. Correction for plate-to-plate differences using four controls on each plate also improved assay performance of both methods. In addition, we observed that the lysis reagent used had a significant effect. Compared with cholic acid, lysis with sodium hydroxide results in higher (P = 0.0082) and Triton X-100 in lower (P = 0.0028) CEC values. Furthermore, large cell seeding errors (30% variation) greatly biased CEC for both referencing methods (P < 0.0001) as measured by a resazurin assay. In conclusion, lysis reagents, cell numbers, and assay setup greatly impact the quality and reliability of CEC quantification and should be considered when this method is newly established in a laboratory.

Enantioseparation of chiral products has become increasingly important in a large diversity of academic and industrial applications. The separation of chiral compounds is inherently challenging and thus requires a suitable analytical technique that can achieve high resolution and sensitivity. In this context, CE has shown remarkable results so far. Chiral CE offers an orthogonal enantioselectivity and is typically considered less costly than chromatographic techniques, since only minute amounts of chiral selectors are needed. Several CE approaches have been developed for chiral analysis, including chiral EKC and chiral CEC. Enantioseparations by EKC benefit from the wide variety of possible pseudostationary phases that can be employed. Chiral CEC, on the other hand, combines chromatographic separation principles with the bulk fluid movement of CE, benefitting from reduced band broadening as compared to pressure-driven systems. Although UV detection is conventionally used for these approaches, MS can also be considered. CE-MS represents a promising alternative due to the increased sensitivity and selectivity, enabling the chiral analysis of complex samples. The potential contamination of the MS ion source in EKC-MS can be overcome using partial-filling and counter-migration techniques. However, chiral analysis using monolithic and open-tubular CEC-MS awaits additional method validation and a dedicated commercial interface. Further efforts in chiral CE are expected toward the improvement of existing techniques, the development of novel pseudostationary phases, and establishing the use of chiral ionic liquids, molecular imprinted polymers, and metal-organic frameworks. These developments will certainly foster the adoption of CE(-MS) as a well-established technique in routine chiral analysis.

The aim of the present study is to determine four anionic alkyl sulfate (AS) surfactants with different alkyl chains, namely, C8, C10, C12, and C14, in wastewater by CE with capacitively coupled contactless conductivity detection (CE-C4 D). The conditions effective for the separation of the four AS surfactants were systematically optimized and found to be in a Tris-His (50 mM/20 mM) BGE solution at a pH of 8.95, using a separation voltage of +15 kV, hydrodynamic injection by siphoning using a 20 cm injection height and an injection time of 20 s. The LODs for C8, C10, C12, and C14 were 2.58, 2.30, 2.08, and 3.16 mg/L, respectively. The conditions used to achieve the simultaneous adsorption and preconcentration of the AS surfactants using Al2 O3 beads were pH of 3 and 0.1 mM NaCl. The adsorption efficiencies were found to be 45.6, 50.8, 81.7, and 99.9%, while the desorption efficiencies reached 66.1, 70.4, 83.9, and 100.0% for C8, C10, C12, and C14, respectively. The concentrations of the AS surfactants in wastewater samples were quantified by CE-C4 D after preconcentration by simultaneous adsorption using Al2 O3 beads. The results obtained from the proposed method were consistent with those obtained by HPLC-MS/MS, with a deviation of less than 15%. Our results indicate that the CE-C4 D performed after preconcentration by an adsorption technique using Al2 O3 beads is a new, inexpensive, and suitable method for quantifying AS surfactants in wastewater samples.
