Appearance of plaques on a bacterial lawn is a sign of successive rounds of bacteriophage infection. Yet, mechanisms evolved by bacteria to limit plaque spread have been hardly explored. Here, we investigated the dynamics of plaque development by lytic phages infecting the bacterium Bacillus subtilis. We report that plaque expansion is followed by a constriction phase owing to bacterial growth into the plaque zone. This phenomenon exposed an adaptive process, herein termed "phage tolerance response", elicited by non-infected bacteria upon sensing infection of their neighbors. The temporary phage tolerance is executed by the stress-response RNA polymerase sigma factor σX (SigX). Artificial expression of SigX prior to phage attack largely eliminates infection. SigX tolerance is primarily conferred by activation of the dlt operon, encoding enzymes that catalyze D-alanylation of cell wall teichoic acid polymers, the major attachment sites for phages infecting Gram-positive bacteria. D-alanylation impedes phage binding and hence infection, thus enabling the uninfected bacteria to form a protective shield opposing phage spread.

Brassinosteroids (BRs) are a class of steroid phytohormones that control various aspects of plant growth and development. Several transcriptional factors (TFs) have been suggested to play roles in BR signaling. However, their possible relationship remains largely unknown. Here, we identified a rice mutant dwarf and low-tillering 2 (dlt2) with altered plant architecture, increased grain width, and reduced BR sensitivity. DLT2 encodes a GIBBERELLIN INSENSITIVE (GAI)-REPRESSOR OF GAI (RGA)-SCARECROW (GRAS) TF that is mainly localized in the nucleus and has weak transcriptional activity. Our further genetic and biochemical analyses indicate that DLT2 interacts with two BR-signaling-related TFs, DLT and BRASSINAZOLE-RESISTANT 1, and probably modulates their transcriptional activity. These findings imply that DLT2 is implicated in a potentially transcriptional complex that mediates BR signaling and rice development and suggests that DLT2 could be a potential target for improving rice architecture and grain morphology. This work also sheds light on the role of rice GRAS members in regulating numerous developmental processes.

Blockchain's potential to revolutionize supply chain and logistics with transparency and equitable stakeholder engagement is significant. However, challenges like scalability, privacy, and interoperability persist. This study explores the scarcity of real-world blockchain implementations in supply chain and logistics since we have not witnessed many real-world deployments of blockchain-based solutions in the field. Puzzled by this, we integrate technology, user experience, and operational efficiency to illuminate the complex landscape of blockchain integration. We present blockchain-based solutions in three use cases, comparing them with alternative designs and analyzing them in terms of technical, economic, and operational aspects. Insights from a tailored questionnaire of 50 questions addressed to practitioners and experts offer crucial perspectives on blockchain adoption. One of the key findings from our work shows that half of the companies interviewed agree that they will miss the potential for competitive advantage if they do not invest in blockchain technology, and 61% of the companies surveyed claimed that their customers ask for more transparency in supply chain-related transactions. However, only one-third of the companies were aware of the main features of blockchain technology, which shows a lack of knowledge among the companies that may lead to a weaker blockchain adaption in supply chain use cases. Our readers should note that our study is specifically contextualized in a Netherlands-funded national project. We hope that researchers as well as stakeholders in supply chain and logistics can benefit from the insights of our work.

BACKGROUND: The incidence of postoperative sore throat (POST) after tracheal intubation using double-lumen endobronchial tubes (DLTs) is higher in patients with prior severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection than in the general population. This prospective trial was conducted to determine whether thermal softening of DLTs could decrease the incidence of POST or other airway injuries in patients with prior SARS-CoV-2 infection. METHODS: A total of 120 patients with prior SARS-CoV-2 infection undergoing thoracoscopic surgery were randomly assigned to two groups (n = 60 each). In the thermal softening group, the distal portion of the DLT was placed in thermostatic saline (50 °C) for 10 min before endotracheal intubation. In the control group, the distal portion of the DLT was placed in room temperature saline for 10 min before endotracheal intubation. The incidence and severity of POST and hoarseness were assessed at 1, 6 and 24 h postoperatively. The primary outcomes were the incidence and severity of POST at 6 h postoperatively. The secondary outcomes were the incidence and severity of hoarseness, vocal cord and tracheal injuries, and hemodynamic changes in patients at intubation. RESULTS: The incidence of POST at 6 h postoperatively was greater in the control group than in the thermal softening group [41 (68%) vs. 22 (37%), P = 0.001]. The overall incidence of POST at 24 h postoperatively was greater in the control group than in the thermal softening group [46 (76%) vs. 24 (40%), P < 0.001]. The overall incidence of tracheal injuries was also greater in the control group than in the thermal softening group (P = 0.016). Vocal cord injuries occurred more frequently in the control group than in the thermal softening group (P = 0.006). CONCLUSION: Thermal softening of DLTs before intubation can reduce the incidence of POST and airway injuries in patients with prior SARS-CoV-2 infection undergoing DLT insertion. With the electric power grid experiencing a rapid shift to the smart grid paradigm over a deregulated energy market, Internet of Things (IoT)-based solutions are gaining prominence, and innovative peer-to-peer (P2P) energy trading at a micro level is being deployed. Such advancement, however, leaves traditional security models vulnerable and paves the path for blockchain, a distributed ledger technology (DLT), with its decentralized, open, and transparency characteristics as a viable alternative. However, due to deregulation in energy trading markets, most of the prototype resilience regarding cybersecurity attack, performance and scalability of transaction broadcasting, and its direct impact on overall performances and attacks are required to be supported, which becomes a performance bottleneck with existing blockchain solutions such as Hyperledger, Ethereum, and so on. In this paper, we design a novel permissioned Corda framework for P2P energy trading peers that not only mitigates a new class of cyberattacks, i.e., delay trading (or discard), but also disseminates the transactions in a optimized propagation time, resulting in a fair transaction distribution. Sharing transactions in a permissioned R3 Corda blockchain framework is handled by the Advanced Message Queuing Protocol (AMQP) and transport layer security (TLS). The unique contribution of this paper lies in the use of an optimized CPU and JVM heap memory scenario analysis with P2P metric in addition to a far more realistic multihosted testbed for the performance analysis. The average latencies measured are 22 ms and 51 ms for sending and receiving messages. We compare the throughput by varying different types of flow such as energy request, request + pay, transfer, multiple notary, sender, receiver, and single notary. In the proposed framework, request is an energy asset that is based on payment state and contract in the P2P energy trading module, so in request flow, only one node with no notary appears on the vault of the node.Energy request + pay flow interaction deals with two nodes, such as producer and consumer, to deal with request and transfer of asset ownership with the help of a notary. Request + repeated pay flow request, on node A and repeatedly transfers a fraction of energy asset state to another node, B, through a notary.

The Internet of Things (IoT) paradigm aims to enhance human society and living standards with the vast deployment of smart and autonomous devices, which requires seamless collaboration. The number of connected devices increases daily, introducing identity management requirements for edge IoT devices. Due to IoT devices' heterogeneity and resource-constrained configuration, traditional identity management systems are not feasible. As a result, identity management for IoT devices is still an open issue. Distributed Ledger Technology (DLT) and blockchain-based security solutions are becoming popular in different application domains. This paper presents a novel DLT-based distributed identity management architecture for edge IoT devices. The model can be adapted with any IoT solution for secure and trustworthy communication between devices. We have comprehensively reviewed popular consensus mechanisms used in DLT implementations and their connection to IoT research, specifically identity management for Edge IoT devices. Our proposed location-based identity management model is generic, distributed, and decentralized. The proposed model is verified using the Scyther formal verification tool for security performance measurement. SPIN model checker is employed for different state verification of our proposed model. The open-source simulation tool FobSim is used for fog and edge/user layer DTL deployment performance analysis. The results and discussion section represents how our proposed decentralized identity management solution should enhance user data privacy and secure and trustworthy communication in IoT.

Due to the enormous amounts of data being generated between users, Intelligent Transportation Systems (ITS) are complex Cyber-Physical Systems that necessitate a reliable and safe infrastructure. Internet of Vehicles (IoV) is the term that describes the interconnection for every single node, device, sensor, and actuator that are Internet enabled, whether attached or unattached to vehicles. A single smart vehicle will generate a huge amount of data. Concurrently, it needs an instant response to avoid accidents since vehicles are fast-moving objects. In this work, we explore Distributed Ledger Technology (DLT) and collect data about consensus algorithms and their applicability to be used in the IoV as the backbone of ITS. Multiple distributed ledger networks are currently in operation. Some are used in finance or supply chains, and others are used for general decentralized applications. Despite the secure and decentralized nature of the blockchain, each of these networks has trade-offs and compromises. Based on the analysis of consensus algorithms, a conclusion has been made to design one that fits the requirements of ITS-IOV. FlexiChain 3.0 is proposed in this work to serve as a Layer0 network for different stakeholders in the IoV. A time analysis has been conducted and shows a capacity of 2.3 transactions per second, which is an acceptable speed to be used in IoV. Moreover, a security analysis was conducted as well and shows high security and high independence of the node number in terms of security level per the number of participants.

Objective: The aim of this study is to compare the effect of bronchial blockers (BB) and double-lumen tubes (DLT) on patients' postoperative recovery after lung resection. Method: 4,636 patients undergoing lung resection and receiving either BB or DLT intubation were reviewed and matched using the propensity score matching method. The primary outcome was the surgical duration. The secondary outcomes included diagnostic results of postoperative chest X-ray, postoperative oxygenation index, incidence of hypercapnia, hypoxemia and sore throat, chest tube duration, incidence of ICU admission, length of hospital stay and incidence of the 30-day readmission. Results: After matching, 401 patients receiving BB were matched to 3,439 patients receiving DLT. There was no statistical difference on the surgical duration between the two groups (P>0.05). However, compared with the DLT group, patients in the BB group showed more infiltrate especially at the surgery side (14.96% versus 9.07%, P<0.001) based on the chest X-ray, together with higher incidence of ICU admission (5.23% versus 2.61%, P<0.05). Additionally, no statistical differences were found between the two groups about chest tube duration, oxygenation index, incidence of hypercapnia, hypoxemia and sore throat, duration of surgery, hospital stays and 30-day readmission (P>0.05). Conclusions: Compared with the DLT, patients receiving BB technique tend to have increased pulmonary infiltrate (especially the surgery side) and higher incidence of ICU admission at the early post-operative stage, which may have an influence on the patients' recovery.

Lactococcus lactis, a lactic acid bacterium, has been widely used in the fermented dairy products. The acid tolerance of L. lactis is of great importance to food fermentation and probiotic applications. As the first barrier of bacteria, the cell wall has a protective effect on strains under many stress conditions, whereas the regulatory mechanism has rarely been reported. Here, based on the transcription analysis of 9 cell wall or membrane-related genes of L. lactis F44 under acid stress, the transcription levels of DACB, DLTD, YLBA, HRTA, WP_080613266.1 (1610), and ERFK genes were significantly increased. We constructed 9 overexpressing strains with the cell wall or membrane-related genes, respectively. It was demonstrated that the survival rates under acid stress of DACB, DLTD, and ERFK were significantly higher than that of wild-type F44. To investigate the regulatory mechanism, a DNA pull-down assay was used to identify the transcriptional regulators of these 3 genes. It was discovered that the 2-component system (TCS) transcriptional regulator TCSR7 bound to the upstream region of DLTD involved in the teichoic acid (TA) alanylation. The combination was confirmed through an electrophoretic mobility shift assay in vitro. Reverse-transcription quantitative PCR results indicated that TCSR7 upregulated the expression of DLTD gene. In addition, the transcription level of TCSR7 increased approximately 1.8-fold (log2 fold change) under acidic conditions. In summary, this study found that TCSR7 was induced by acid stress to upregulate the transcription level of the DLT operon genes, which might increase the positive charge on the cell membrane surface to increase the acid tolerance of the strain. This study lays the foundation for the regulatory mechanism of TA alanylation under acid stress.

Internet-of-Things (IoT) and sensor technologies have enabled the collection of data in a distributed fashion for analysis and evidence-based decision making. However, security concerns regarding the source, confidentiality and integrity of the data arise. The most common method of protecting data transmission in sensor systems is Transport Layer Security (TLS) or its datagram counterpart (DTLS) today, but exist an alternative option based on Distributed Ledger Technology (DLT) that promise strong security, ease of use and potential for large scale integration of heterogeneous sensor systems. A DLT such as the IOTA Tangle offers great potential to improve sensor data exchange. This paper presents L2Sec, a cryptographic protocol which is able to secure data exchanged over the IOTA Tangle. This protocol is suitable for implementation on constrained devices, such as common IoT devices, leading to greater scalability. The first experimental results evidence the effectiveness of the approach and advocate for the integration of an hardware secure element to improve the overall security of the protocol. The L2Sec source code is released as open source repository on GitHub.

Smart devices have become an essential part of the architectures such as the Internet of Things (IoT), Cyber-Physical Systems (CPSs), and Internet of Everything (IoE). In contrast, these architectures constitute a system to realize the concept of smart cities and, ultimately, a smart planet. The adoption of these smart devices expands to different cyber-physical systems in smart city architecture, i.e., smart houses, smart healthcare, smart transportation, smart grid, smart agriculture, etc. The edge of the network connects these smart devices (sensors, aggregators, and actuators) that can operate in the physical environment and collects the data, which is further used to make an informed decision through actuation. Here, the security of these devices is immensely important, specifically from an authentication standpoint, as in the case of unauthenticated/malicious assets, the whole infrastructure would be at stake. We provide an updated review of authentication mechanisms by categorizing centralized and distributed architectures. We discuss the security issues regarding the authentication of these IoT-enabled smart devices. We evaluate and analyze the study of the proposed literature schemes that pose authentication challenges in terms of computational costs, communication overheads, and models applied to attain robustness. Hence, lightweight solutions in managing, maintaining, processing, and storing authentication data of IoT-enabled assets are an urgent need. From an integration perspective, cloud computing has provided strong support. In contrast, decentralized ledger technology, i.e., blockchain, light-weight cryptosystems, and Artificial Intelligence (AI)-based solutions, are the areas with much more to explore. Finally, we discuss the future research challenges, which will eventually help address the ambiguities for improvement.

With the application of four optical CMOS sensors, it was possible to capture the trajectory of an endoscopic tool during an in vitro surgical experiment on a cardiovascular preparation. This was due to the possibility of obtaining a path when a reflective marker was attached. In the work, APAS (Ariel Performance Analysis System) software and DLT (direct linear transformation) algorithm were used. This made it possible to acquire kinematic inputs to the computational model of dynamics, which enabled, regardless of the type of surgical robot structure, derivation of the analogous motion of an endoscopic effector due to the mathematical transformation of the trajectory to joints coordinates. Experiments were carried out with the participation of a practiced cardiac surgeon employing classic endoscopic instruments and robot surgical systems. The results indicated by the experiment showed that the inverse task of kinematics of position for the surgical robot with RCM (remote center of motion) structure was solved. The achieved results from the experiment were used as inputs for deriving a numerical dynamics model of surgical robot during transient states that was obtained by applying the finite element method and by driving dynamics moments acquired through the block diagrams method using a steering system with DC (direct current) motor and PID (proportional-integral-derivative) controller. The results section illustrates the course of kinematic values of endoscopic tools which were employed to apply numerical models as inputs, the course of the driving torque of the model of the surgical robot that enabled the selection of the drive system and the strength values, stresses and displacements according to von Mises hypothesis in its structure during the analysis of transient states that made it possible to establish the strength safety of the surgical robot. For the conducted experiments, the accuracy was ±2 [mm]. In the paper, the employment of optical CMOS sensors in surgical robotics and endoscopy is discussed. The paper concludes that the usage of optical sensors for determining inputs for numerical models of dynamics of surgical robots provides the basis for setting the course of physical quantities that appear in their real object structure, in manners close to reality.

Brassinosteroids (BRs) are a class of phytohormones that modulate several important agronomic traits in rice (Oryza sativa). GSK2 is one of the critical suppressors of BR signalling and targets transcription factors such as OsBZR1 and DLT to regulate BR responses. Here, we identified OFP3 (OVATE FAMILY PROTEIN 3) as an interactor of both GSK2 and DLT by yeast-two-hybrid screening and demonstrated that OFP3 plays a distinctly negative role in BR responses. While knockout of OFP3 promoted rice seedling growth, overexpression of OFP3 led to strong BR insensitivity, which resulted in reduced plant height, leaf angle, and grain size. Interestingly, both BR biosynthetic and signalling genes had decreased expression in the overexpression plants. OFP3 overexpression also enhanced the phenotypes of BR-deficient mutants, but largely suppressed those of BR-enhanced plants. Moreover, treatment with either BR or bikinin, a GSK3-like kinase inhibitor, induced OFP3 depletion, whereas GSK2 or brassinazole, a BR synthesis inhibitor, promoted OFP3 accumulation. Furthermore, OFP3 exhibited transcription repressor activity and was able to interact with itself as well as additional BR-related components, including OFP1, OSH1, OSH15, OsBZR1, and GF14c. Importantly, GSK2 can phosphorylate OFP3 and enhance these interactions. We propose that OFP3, as a suppressor of both BR synthesis and signalling but stabilized by GSK2, incorporates into a transcription factor complex to facilitate BR signalling control, which is critical for the proper development of various tissues.

A potential rise in interest in the Internet of Things in the upcoming years is expected in the fields of healthcare, supply chain, logistics, industries, smart cities, smart homes, cyber physical systems, etc. This paper discloses the fusion of the Internet of Things (IoT) with the so-called "distributed ledger technology" (DLT). IoT sensors like temperature sensors, motion sensors, GPS or connected devices convey the activity of the environment. Sensor information acquired by such IoT devices are then stored in a blockchain. Data on a blockchain remains immutable however its scalability still remains a challenging issue and thus represents a hindrance for its mass adoption in the IoT. Here a communication system based on IOTA and DLT is discussed with a systematic architecture for IoT devices and a future machine-to-machine (M2M) economy. The data communication between IoT devices is analyzed using multiple use cases such as sending DHT-11 sensor data to the IOTA tangle. The value communication is analyzed using a novel "micro-payment enabled over the top" (MP-OTT) streaming platform that is based on the "pay-as-you-go" and "consumption based" models to showcase IOTA value transactions. In this paper, we propose an enhancement to the classical "masked authenticated message" (MAM) communication protocol and two architectures called dual signature masked authenticated message (DSMAM) and index-based address value transaction (IBAVT). Further, we provided an empirical analysis and discussion of the proposed techniques. The implemented solution provides better address management with secured sharing and communication of IoT data, complete access control over the ownership of data and high scalability in terms of number of transactions that can be handled.

IOTA is a distributed ledger technology (DLT) platform proposed for the internet of things (IoT) systems in order to tackle the limitations of Blockchain in terms of latency, scalability, and transaction cost. The main concepts used in IOTA to reach this objective are a directed acyclic graph (DAG) based ledger, called Tangle, used instead of the chain of blocks, and a new validation mechanism that, instead of relying on the miners as it is the case in Blockchain, relies on participating nodes that cooperate to validate the new transactions. Due to the different IoT capabilities, IOTA classifies these devices into full and light nodes. The light nodes are nodes with low computing resources which seek full nodes' help to validate and attach its transaction to the Tangle. The light nodes are manually connected to the full nodes by using the full node IP address or the IOTA client load balancer. This task distribution method overcharges the active full nodes and, thus, reduces the platform's performance. In this paper, we introduce an efficient mechanism to distribute the tasks fairly among full nodes and hence achieve load balancing. To do so, we consider the task allocation between the nodes by introducing an enhanced resource allocation scheme based on the weight least connection algorithm (WLC). To assess its performance, we investigate and test different implementation scenarios. The results show an improved balancing of data traffic among full nodes based on their weights and number of active connections.

Background The effectiveness of reintroducing oxaliplatin for metastatic colorectal cancer (mCRC) refractory to both oxaliplatin and irinotecan was previously reported in a phase II study (RE-OPEN). We conducted a phase I study to determine the maximum tolerated dose of oxaliplatin plus trifluridine/tipiracil (FTD/TPI) in patients with refractory mCRC. Patients and Methods Three dosages of intravenous oxaliplatin (50, 65 and 85 mg/m2) on days 1 and 15 and a fixed dose of FTD/TPI 35 mg/m2 twice daily (bid) on days 1-5 and 15-19 every 4 weeks were investigated in patients with refractory mCRC using a 3 + 3 design. Eligible patients had received prior oxaliplatin-based treatment that achieved a response or stable disease followed by confirmed disease progression at least 6 months before entering the study. Results Twelve patients were enrolled in the study. Three of six patients in the oxaliplatin 85 mg/m2 cohort had dose-limiting toxicities (DLTs) with treatment delays during the second cycle at ≥8 days due to grade ≥ 2 neutropenia or grade 2 AST/ALT increased. No DLTs were observed in the other cohorts. Grade ≥ 3 AEs were neutropenia (n = 3), thrombocytopenia (n = 1), anorexia (n = 1), and nausea (n = 1). There was no evidence of allergic reaction to oxaliplatin or severe peripheral sensory neuropathy. Conclusions A combination of FTD/TPI 35 mg/m2 bid on days 1-5 and 15-19 and oxaliplatin 85 mg/m2 on days 1 and 15 every 4 weeks could be a suitable regimen for the recommended dose of FTD/TPI plus oxaliplatin in patients with refractory mCRC.

Camera calibration is a crucial step for computer vision in many applications. For example, adequate calibration is required in infrared thermography inside gas turbines for blade temperature measurements, for associating each pixel with the corresponding point on the blade 3D model. The blade has to be used as the calibration frame, but it is always only partially visible, and thus, there are few control points. We propose and test a method that exploits the anisotropic uncertainty of the control points and improves the calibration in conditions where the number of control points is limited. Assuming a bivariate Gaussian 2D distribution of the position error of each control point, we set uncertainty areas of control points' position, which are ellipses (with specific axis lengths and rotations) within which the control points are supposed to be. We use these ellipses to set a weight matrix to be used in a weighted Direct Linear Transformation (wDLT). We present the mathematical formalism for this modified calibration algorithm, and we apply it to calibrate a camera from a picture of a well known object in different situations, comparing its performance to the standard DLT method, showing that the wDLT algorithm provides a more robust and precise solution. We finally discuss the quantitative improvements of the algorithm by varying the modules of random deviations in control points' positions and with partial occlusion of the object.

Modifications to the Gram-positive bacterial cell wall play important roles in antibiotic resistance and pathogenesis, but the pathway for the d-alanylation of teichoic acids (DLT pathway), a ubiquitous modification, is poorly understood. The d-alanylation machinery includes two membrane proteins of unclear function, DltB and DltD, which are somehow involved in transfer of d-alanine from a carrier protein inside the cell to teichoic acids on the cell surface. Here, we probed the role of DltD in the human pathogen Staphylococcus aureus using both cell-based and biochemical assays. We first exploited a known synthetic lethal interaction to establish the essentiality of each gene in the DLT pathway for d-alanylation of lipoteichoic acid (LTA) and confirmed this by directly detecting radiolabeled d-Ala-LTA both in cells and in vesicles prepared from mutant strains of S. aureus We developed a partial reconstitution of the pathway by using cell-derived vesicles containing DltB, but no other components of the d-alanylation pathway, and showed that d-alanylation of previously formed lipoteichoic acid in the DltB vesicles requires the presence of purified and reconstituted DltA, DltC, and DltD, but not of the LTA synthase LtaS. Finally, based on the activity of DltD mutants in cells and in our reconstituted system, we determined that Ser-70 and His-361 are essential for d-alanylation activity, and we propose that DltD uses a catalytic dyad to transfer d-alanine to LTA. In summary, we have developed a suite of assays for investigating the bacterial DLT pathway and uncovered a role for DltD in LTA d-alanylation.

Industry 4.0 has paved the way for a world where smart factories will automate and upgrade many processes through the use of some of the latest emerging technologies. One of such technologies is Unmanned Aerial Vehicles (UAVs), which have evolved a great deal in the last years in terms of technology (e.g., control units, sensors, UAV frames) and have significantlyr educed their cost. UAVs can help industry in automatable and tedious tasks, like the ones performed on a regular basis for determining the inventory and for preserving item traceability. In such tasks, especially when it comes from untrusted third parties, it is essential to determine whether the collected information is valid or true. Likewise, ensuring data trustworthiness is a key issue in order to leverage Big Data analytics to supply chain efficiency and effectiveness. In such a case, blockchain, another Industry 4.0 technology that has become very popular in other fields like finance, has the potential to provide a higher level of transparency, security, trust and efficiency in the supply chain and enable the use of smart contracts. Thus, in this paper, we present the design and evaluation of a UAV-based system aimed at automating inventory tasks and keeping the traceability of industrial items attached to Radio-Frequency IDentification (RFID) tags. To confront current shortcomings, such a system is developed under a versatile, modular and scalable architecture aimed to reinforce cyber security and decentralization while fostering external audits and big data analytics. Therefore, the system uses a blockchain and a distributed ledger to store certain inventory data collected by UAVs, validate them, ensure their trustworthiness and make them available to the interested parties. In order to show the performance of the proposed system, different tests were performed in a real industrial warehouse, concluding that the system is able to obtain the inventory data really fast in comparison to traditional manual tasks, while being also able to estimate the position of the items when hovering over them thanks to their tag's signal strength. In addition, the performance of the proposed blockchain-based architecture was evaluated in different scenarios.

The three-component apsXRS system senses and responds to cationic antimicrobial peptides (CAMPs), which induces the expression of the dlt operon and the genes mprF and vrafG, modifying the surface net charge in Staphylococcus epidermidis, resulting in the repulsion of CAMPs. The apsXRS system has been only studied in the S. epidermidis 1457 strain, and there are no studies of prevalence and level of expression of apsXRS in commensal and clinical isolates. From 60 isolates, those selected from commensal healthy skin (n = 20), commensal healthy conjunctive (n = 10), and clinical ocular infection (n = 30) presented the apsX, apsR, and apsS genes in their genomes. Constitutive expression of apsX, apsR, and apsS genes was determined by RT-qPCR in all isolates. It was found that expression of apsX, apsR, and apsS was 3.3-5.9-fold higher in commensal isolates stimulated with LL-37 (15 µg/mL) than in clinical isolates. Similarly, expression of the dlt operon and the genes mprF, and vraFG was 8-10-fold higher in commensal isolates than in clinical. However, LL-37 did not increase the addition of lysine in the phospholipids of the cytoplasmic membrane in any of the isolates. Mutations in the apsS loop region, apsR, and their promoter sequence were not found. These results demonstrated that apsXRS system is essential in all isolates for its constitutive expression; however, LL-37 caused an increase of apsXRS expression in commensal isolates, suggesting that S. epidermidis isolates do not respond in the same way to the presence of LL-37.
