RESUMO

Acidity is a key factor controlling fruit flavor and quality. In a previous study, combined transcriptome and methylation analyses identified a P3A-type ATPase from apple (Malus domestica), MdMa11, which regulates vacuolar pH when expressed in Nicotiana benthamiana leaves. In this study, the role of MdMa11 in controlling fruit acidity was verified in apple calli, fruits, and plantlets. In addition, we isolated an AP2 domain-containing transcription factor, designated MdESE3, based on yeast one-hybrid (Y1H) screening using the MdMa11 promoter as bait. A subcellular localization assay indicated that MdESE3 localized to the nucleus. Analyses of transgenic apple calli, fruits, and plantlets, as well as tomatoes, demonstrated that MdESE3 enhances fruit acidity and organic acid accumulation. Meanwhile, chromatin immunoprecipitation quantitative PCR (ChIP-qPCR), luciferase (LUC) transactivation assays, and GUS reporter assays indicated that MdESE3 could bind to the ethylene-responsive element (ERE; 5'-TTTAAAAT-3') upstream of the MdMa11 transcription start site, thereby activating its expression. Furthermore, MdtDT, MdDTC2, and MdMDH12 expression increased in apple fruits and plantlets overexpressing MdESE3 and decreased in apple fruits and plantlets where MdESE3 was silenced. The ERE was found in MdtDT and MdMDH12 promoters, but not in the MdDTC2 promoter. The Y1H, LUC transactivation assays, and GUS reporter assays indicated that MdESE3 could bind to the MdtDT and MdMDH12 promoters and activate their expression. Our findings provide valuable functional validation of MdESE3 and its role in the transcriptional regulation of MdMa11, MdtDT, and MdMDH12 and malic acid accumulation in apple.

The mismatch negativity and the P3a of the event-related EEG potential reflect the electrocortical response to a deviant stimulus in a series of stimuli. Although both components have been investigated in various paradigms, these paradigms usually incorporate many repetitions of the same deviant, thus leaving open whether both components vary as a function of the deviant's position in a series of deviant stimuli-i.e. whether they are subject to qualitative/quantitative habituation from one instantiation of a deviant to the next. This is so because the detection of mismatch negativity/P3a in the event-related EEG potential requires an averaging over dozens or hundreds of stimuli, i.e. over many instantiations of the deviant per participant. The present study addresses this research gap. We used a two-tone oddball paradigm implementing only a small number of (deviant) stimuli per participant, but applying it to a large number of participants (n > 230). Our data show that the mismatch negativity amplitude exhibits no decrease as a function of the deviant's position in a series of (standard and) deviant stimuli. Importantly, only after the very first deviant stimulus, a distinct P3a could be detected, indicative of an orienting reaction and an attention shift, and thus documenting a dissociation of mismatch negativity and P3a.

The influence of effort expenditure on the subjective value in feedback involving material reward has been the focus of previous research. However, little is known about the impact of effort expenditure on subjective value evaluations when feedback involves reward that is produced in the context of social interaction (e.g. self-other agreement). Moreover, how effort expenditure influences confidence (second-order subjective value) in feedback evaluations remains unclear. Using electroencephalography, this study aimed to address these questions. Event-related potentials showed that, after exerting high effort, participants exhibited increased reward positivity difference in response to self-other (dis)agreement feedback. After exerting low effort, participants reported high confidence, and the self-other disagreement feedback evoked a larger P3a. Time-frequency analysis showed that the high-effort task evoked increased frontal midline theta power. In the low (vs. high)-effort task, the frontal midline delta power for self-other disagreement feedback was enhanced. These findings suggest that, at the early feedback evaluation stage, after exerting high effort, individuals exhibit an increased sensitivity of subjective value evaluation in response to self-other agreement feedback. At the later feedback evaluation stage, after completing the low-effort task, the self-other disagreement feedback violates the individuals'high confidence and leads to a metacognitive mismatch.

BACKGROUND: Deficits in event-related potential (ERP) including duration mismatch negativity (MMN) and P3a have been demonstrated widely in chronic schizophrenia (SZ) but inconsistent findings were reported in first-episode patients. Psychotropic medications and diagnosis might contribute to different findings on MMN/P3a ERP in first-episode patients. The present study examined MMN and P3a in first episode drug naïve SZ and bipolar disorder (BPD) patients and explored the relationships among ERPs, neurocognition and global functioning. METHODS: Twenty SZ, 24 BPD and 49 age and sex-matched healthy controls were enrolled in this study. Data of clinical symptoms [Positive and Negative Symptoms Scale (PANSS), Young Manic Rating Scale (YMRS), Hamilton Depression Rating Scale (HAMD)], neurocognition [Wechsler Adult Intelligence Scale (WAIS), Cattell's Culture Fair Intelligence Test (CCFT), Delay Matching to Sample (DMS), Rapid Visual Information Processing (RVP)], and functioning [Functioning Assessment Short Test (FAST)] were collected. P3a and MMN were elicited using a passive auditory oddball paradigm. RESULTS: Significant MMN and P3a deficits and impaired neurocognition were found in both SZ and BPD patients. In SZ, MMN was significantly correlated with FAST (r = 0.48) and CCFT (r = -0.31). In BPD, MMN was significantly correlated with DMS (r = -0.54). For P3a, RVP and FAST scores were significant predictors in SZ, whereas RVP, WAIS and FAST were significant predictors in BPD. CONCLUSIONS: The present study found deficits in MMN, P3a, neurocognition in drug naïve SZ and BPD patients. These deficits appeared to link with levels of higher-order cognition and functioning.

Here, we investigated the central auditory processing and attentional control associated with both recovery and prolongation of occupational burnout. We recorded the event-related brain potentials N1, P2, mismatch negativity (MMN) and P3a to nine changes in speech sounds and to three rarely presented emotional (happy, angry and sad) utterances from individuals with burnout (N = 16) and their matched controls (N = 12). After the 5 years follow-up, one control had acquired burnout, half (N = 8) of the burnout group had recovered, and the other half (prolonged burnout) still had burnout. The processing of acoustical changes in speech sounds was mainly intact. Prolongation of the burnout was associated with a decrease in MMN amplitude and an increase in P3a amplitude for the happy stimulus. The results suggest that, in the absence of interventions, burnout is a persistent condition, associated with alterations of attentional control, that may be amplified with the prolongation of the condition.

The human auditory system excels at detecting patterns needed for processing speech and music. According to predictive coding, the brain predicts incoming sounds, compares predictions to sensory input and generates a prediction error whenever a mismatch between the prediction and sensory input occurs. Predictive coding can be indexed in electroencephalography (EEG) with the mismatch negativity (MMN) and P3a, two components of event-related potentials (ERP) that are elicited by infrequent deviant sounds (e.g., differing in pitch, duration and loudness) in a stream of frequent sounds. If these components reflect prediction error, they should also be elicited by omitting an expected sound, but few studies have examined this. We compared ERPs elicited by infrequent randomly occurring omissions (unexpected silences) in tone sequences presented at two tones per second to ERPs elicited by frequent, regularly occurring omissions (expected silences) within a sequence of tones presented at one tone per second. We found that unexpected silences elicited significant MMN and P3a, although the magnitude of these components was quite small and variable. These results provide evidence for hierarchical predictive coding, indicating that the brain predicts silences and sounds.

Attention operates through top-down and bottom-up processes, and a balance between these processes is crucial for daily tasks. Imperilling such balance could explain ageing-associated attentional problems such as exacerbated distractibility. In this study, we aimed to characterize this enhanced distractibility by investigating the impact of ageing upon event-related components associated with top-down and bottom-up attentional processes. MEG and EEG data were acquired from 14 older and 14 younger healthy adults while performing a task that conjointly evaluates top-down and bottom-up attention. Event-related components were analysed on sensor and source levels. In comparison with the younger group, the older mainly displayed (1) reduced target anticipation processes (reduced CMV), (2) increased early target processing (larger P50 but smaller N1) and (3) increased processing of early distracting sounds (larger N1 but reduced P3a), followed by a (4) prolonged reorientation towards the main task (larger RON). Taken together, our results suggest that the enhanced distractibility in ageing could stem from top-down deficits, in particular from reduced inhibitory and reorientation processes.

BACKGROUND: The P3a response is thought to reflect involuntary orienting to an unexpected stimulus and has been connected with set-shifting and inhibition in some studies. In our exploratory study, we investigated if the amplitude and the latency of the P3a response were associated with the performance in a modified flanker task measuring inhibition and set-shifting in 10-year-old children (N = 42). Children participated in electroencephalography (EEG) measurement with an auditory multifeature paradigm including standard, deviating, and novel sounds. In addition, they performed a separate flanker task requiring inhibition and set-shifting skills. RESULTS: The P3a latencies for deviant sounds were associated with the reaction time reflecting inhibition: the shorter the response latencies were, the faster the reaction time was. The P3a latencies for novel sounds were not linked to the reaction times reflecting either inhibition or set-shifting. In addition, the magnitude of the P3a response was not associated with the performance in the flanker task. CONCLUSIONS: Our results suggest that P3a response latency and reaction speed reflecting inhibitory skills are based on shared neural mechanism. Thus, the present study brings new insight to the field investigating the associations between behavior and its neural indices.

INTRODUCTION: The present study compared the mismatch negativity (MMN) and P3a waveforms among early-phase psychosis (EPP; n = 13) individuals and healthy controls (n = 30) to contribute to the research on these waveforms as potential biomarkers for schizophrenia. METHODS: MMN and P3a were elicited with a novelty paradigm using complex stimuli with electrophysiological technology. RESULTS: No significant group differences of amplitude were observed with either waveform. Increased asociality and blunted affect were associated with a reduction in both MMN and P3a waveforms indicating a relationship between these negative symptoms and cognitive deficits. Good social and occupational functioning correlated with improved MMN and P3a waveforms in the EPP group. CONCLUSIONS: This study suggests that MMN and P3a may be more appropriately used as an indicator of illness progression and symptomology rather than a biomarker in the early phase of the illness.

Previous work suggests that musical training in childhood is associated with enhanced executive functions. However, it is unknown whether this advantage extends to selective attention-another central aspect of executive control. We recorded a well-established event-related potential (ERP) marker of distraction, the P3a, during an audio-visual task to investigate the maturation of selective attention in musically trained children and adolescents aged 10-17 years and a control group of untrained peers. The task required categorization of visual stimuli, while a sequence of standard sounds and distracting novel sounds were presented in the background. The music group outperformed the control group in the categorization task and the younger children in the music group showed a smaller P3a to the distracting novel sounds than their peers in the control group. Also, a negative response elicited by the novel sounds in the N1/MMN time range (~150-200 ms) was smaller in the music group. These results indicate that the music group was less easily distracted by the task-irrelevant sound stimulation and gated the neural processing of the novel sounds more efficiently than the control group. Furthermore, we replicated our previous finding that, relative to the control group, the musically trained children and adolescents performed faster in standardized tests for inhibition and set shifting. These results provide novel converging behavioral and electrophysiological evidence from a cross-modal paradigm for accelerated maturation of selective attention in musically trained children and adolescents and corroborate the association between musical training and enhanced inhibition and set shifting.

BACKGROUND: Individuals with attention-deficit hyperactivity disorder (ADHD) often display over-response to stimuli that are irrelevant to the ongoing task, and their attentional abilities disproportionately worsen in the presence of competing stimuli. Auditory event-related potentials (ERPs) such as mismatch negativity (MMN) and P3a using the passive oddball paradigm have been studied in children and adolescents with ADHD. Still, there is no such data for adults with ADHD. This study aimed to compare the MMN and P3a and their clinical and neurocognitive correlations between drug-naive adults with ADHD and control adults. METHODS: We recruited 52 adults with ADHD (26.5 ± 6.2 years), and 62 age-matched controls (25.6 ± 5.6 years). They received the psychiatric interviews, auditory ERP, the Conners' continuous performance test (CCPT), and the Cambridge gambling test (CGT). They also completed the questionnaires about ADHD symptoms and real-world executive functions. MMN and P3a were assessed during a passive duration-deviant auditory oddball paradigm from the midline electrodes Cz. RESULTS: Adults with ADHD demonstrated smaller Cz MMN amplitude, more severe ADHD symptoms, poorer attention profiles (CCPT), and a wide range of executive dysfunctions than controls. As for the correlates, Cz peak amplitude of MMN correlated with inattention symptoms, executive dysfunctions, attentional vigilance (CCPT), and decision-making (CGT) in ADHD adults but only with decision-making in controls. CONCLUSIONS: Our findings that smaller amplitude of MMN and its differential associated pattern with inattention, real-world executive dysfunction, and decision-making, in drug-naive adults with ADHD from adult controls, provide evidence to support the potential electrophysiological biomarker for adult ADHD.

The neural processing of incoming stimuli can be analysed from the electroencephalogram (EEG) through event-related potentials (ERPs). The P3 component is largely investigated as it represents an important psychophysiological marker of psychiatric disorders. This is composed by several subcomponents, such as P3a and P3b, reflecting distinct but interrelated sensory and cognitive processes of incoming stimuli. Due to the low EEG signal-to-noise-ratio, ERPs emerge only after an averaging procedure across trials and subjects. Thus, this canonical ERP analysis lacks in the ability to highlight EEG neural signatures at the level of single-subject and single-trial. In this study, a deep learning-based workflow is investigated to enhance EEG neural signatures related to P3 subcomponents already at single-subject and at single-trial level. This was based on the combination of a convolutional neural network (CNN) with an explanation technique (ET). The CNN was trained using two different strategies to produce saliency representations enhancing signatures shared across subjects or more specific for each subject and trial. Cross-subject saliency representations matched the signatures already emerging from ERPs, i.e., P3a and P3b-related activity within 350-400 ms (frontal sites) and 400-650 ms (parietal sites) post-stimulus, validating the CNN+ET respect to canonical ERP analysis. Single-subject and single-trial saliency representations enhanced P3 signatures already at the single-trial scale, while EEG-derived representations at single-subject and single-trial level provided no or only mildly evident signatures. Empowering the analysis of P3 modulations at single-subject and at single-trial level, CNN+ET could be useful to provide insights about neural processes linking sensory stimulation, cognition and behaviour.

Studies with non-human primates have suggested an excitatory influence of the thalamus on the cerebral cortex, with the centromedian-parafascicular complex (CM-Pf) being particularly involved in processes of sensory event-driven attention and arousal. To define the involvement of the human CM-Pf in bottom-up and top-down auditory attention, we simultaneously recorded cortical EEG activity and intracranial local field potentials (LFPs) via electrodes implanted for deep brain stimulation for the treatment of neuropathic pain. The patients (N â€‹= â€‹6) performed an auditory three-class oddball paradigm with frequent standard stimuli and two types of infrequent deviant stimuli (target and distractor). We found a parietal P3b to targets and a central P3a to distractors at the scalp level. Subcortical recordings in the CM-Pf revealed enhanced activation to targets compared to standards. Interarea-correlation analyses showed that activation in the CM-Pf predicted the generation of longer latency P3b scalp potentials specifically in the target condition. Our results provide first direct human evidence for a functional temporal relationship between target-related activation in the CM-Pf and an enhanced cortical target response. These results corroborate the hypothetical model of a cortico-basal ganglia loop system that switches from top-down to bottom-up mode in response to salient, task-relevant external events that are not predictable.

PD is a progressive and complex neurological disorder with heterogeneous symptomatology. PD is characterized by classical motor features of parkinsonism and nonmotor symptoms and involves extensive regions of the nervous system, various neurotransmitters, and protein aggregates. Extensive evidence supports auditory dysfunction as an additional nonmotor feature of PD. Studies indicate a broad range of auditory impairments in PD, from the peripheral hearing system to the auditory brainstem and cortical areas. For instance, research demonstrates a higher occurrence of hearing loss in early-onset PD and evidence of abnormal auditory evoked potentials, event-related potentials, and habituation to novel stimuli. Electrophysiological data, such as auditory P3a, also is suggested as a sensitive measure of illness duration and severity. Improvement in auditory responses following dopaminergic therapies also indicates the presence of similar neurotransmitters (i.e., glutamate and dopamine) in the auditory system and basal ganglia. Nonetheless, hearing impairments in PD have received little attention in clinical practice so far. This review summarizes evidence of peripheral and central auditory impairments in PD and provides conclusions and directions for future empirical and clinical research. © 2020 International Parkinson and Movement Disorder Society.

Musical expertise promotes both the perception and the processing of music. The aim of the present study was to analyze if musicians compared to non-musicians already have auditory processing advantages at the neural level. 50 musicians and 50 non-musicians worked on a task to determine the individual auditory difference threshold (individual JND threshold). A passive oddball paradigm followed while the EEG activity was recorded. Frequent standard sounds (528 hertz [Hz]) and rare deviant sounds (individual JND threshold, 535 Hz, and 558 Hz) were presented in the oddball paradigm. The mismatch negativity (MMN) and the P3a were used as indicators of auditory discrimination skills for frequency differences. Musicians had significantly smaller individual JND thresholds than non-musicians, but musicians were not faster than non-musicians. Musicians and non-musicians showed both the MMN and the P3a at the 535 Hz and 558 Hz condition. In the individual JND threshold condition, non-musicians, whose individual JND threshold was at 539.8 Hz (and therefore even above the deviant sound of 535 Hz), predictably showed the MMN and the P3a. Musicians, whose individual JND threshold was at 531.1 Hz (and thus close to the standard sound of 528 Hz), showed no MMN and P3a-although they were behaviorally able to differentiate frequencies individually within their JND threshold range. This may indicate a key role of attention in triggering the MMN during the detection of frequency differences in the individual JND threshold range (see Tervaniemi et al. in Exp Brain 161:1-10, 2005).

The P3a observed after novel events is an event-related potential comprising an early fronto-central phase and a late fronto-parietal phase. It has classically been considered to reflect the attention processing of distracting stimuli. However, novel sounds can lead to behavioral facilitation as much as behavioral distraction. This illustrates the duality of the orienting response which includes both an attentional and an arousal component. Using a paradigm with visual or auditory targets to detect and irrelevant unexpected distracting sounds to ignore, we showed that the facilitation effect by distracting sounds is independent of the target modality and endures more than 1500 ms. These results confirm that the behavioral facilitation observed after distracting sounds is related to an increase in unspecific phasic arousal on top of the attentional capture. Moreover, the amplitude of the early phase of the P3a to distracting sounds positively correlated with subjective arousal ratings, contrary to other event-related potentials. We propose that the fronto-central early phase of the P3a would index the arousing properties of distracting sounds and would be linked to the arousal component of the orienting response. Finally, we discuss the relevance of the P3a as a marker of distraction.

With the greying population, it is increasingly necessary to establish robust and individualized markers of cognitive decline. This requires the combination of well-established neural mechanisms, and the development of increasingly sensitive methodologies. The P300 event-related potential (ERP) has been one of the most heavily investigated neural markers of attention and cognition, and studies have reliably shown that changes in the amplitude and latency of the P300 ERP index the process of aging. However, it is still not clear whether either the P3a or P3b sub-components additionally index levels of cognitive impairment. Here, we used a traditional visual three-stimulus oddball paradigm to investigate both the P3a and P3b ERP components in sixteen young and thirty-four healthy elderly individuals with varying degrees of cognitive ability. EEG data extraction was enhanced through the use of a novel signal processing method called Functional Source Separation (FSS) that increases signal-to-noise ratio by using a weighted sum of all electrodes rather than relying on a single, or a small sub-set, of EEG channels. Whilst clear differences in both the P3a and P3b ERPs were seen between young and elderly groups, only P3b amplitude differentiated older people with low memory performance relative to IQ from those with consistent memory and IQ. A machine learning analysis showed that P3b amplitude (derived from FSS analysis) could accurately categorise high and low performing elderly individuals (78% accuracy). A comparison of Bayes Factors found that differences in cognitive decline within the elderly group were 87 times more likely to be detected using FSS compared to the best performing single electrode (Cz). In conclusion, we propose that P3b amplitude could be a sensitive marker of early, age-independent, episodic memory dysfunction within a healthy older population. In addition, we advocate for the use of more advanced signal processing methods, such as FSS, for detecting subtle neural changes in clinical populations.

BACKGROUND: Cognitive deficits are already present in early stages of schizophrenia. P3a and P3b event-related potentials (ERPs) are believed to underlie the processes of attention and working memory (WM), yet limited research has been performed on the associations between these parameters. Therefore, we explored possible associations between P3a/b amplitudes and cognition in a large cohort of antipsychotic-naïve, first-episode schizophrenia (AN-FES) patients and healthy controls (HC). METHODS: Seventy-three AN-FES patients and 93 age- and gender-matched HC were assessed for their P3a/b amplitude with an auditory oddball paradigm. In addition, subjects performed several subtests from the Cambridge Neuropsychological Test Automated Battery (CANTAB). RESULTS: AN-FES patients had significantly reduced P3a/b amplitudes, as well as significantly lower scores on all cognitive tests compared with HC. Total group correlations revealed positive associations between P3b amplitude and WM and sustained attention and negative associations with all reaction time measures. These associations appeared mainly driven by AN-FES patients, where we found a similar pattern. No significant associations were found between P3b amplitude and cognitive measures in our HC. P3a amplitude did not correlate significantly with any cognitive measures in either group, nor when combined. CONCLUSIONS: Our results provide further evidence for P3a/b amplitude deficits and cognitive deficits in AN-FES patients, which are neither due to antipsychotics nor to disease progress. Furthermore, our data showed significant, yet weak associations between P3b and cognition. Therefore, our data do not supply evidence for deficient P3a/b amplitudes as direct underlying factors for cognitive deficits in schizophrenia.

Sleep loss has a major effect on cognitive tasks that are dependent on the maintenance of active sustained attention. This study examines the effects of sleep deprivation on automatic information processing, more specifically, its effect on processes leading to involuntary auditory attention capture by task-irrelevant auditory events. Two experiments were run. In the first, 13 participants were totally sleep-deprived (TSD); in the second, 16 participants were partially sleep-deprived (PSD), sleeping only four hours. Event-related potentials were recorded while participants discriminated the duration of equiprobable short and long auditory tones. At rare times, a small change to the pitch of these stimuli occurred. This deviant was however irrelevant to the duration detection task. As expected, TSD had a significant effect on the attention-dependent duration detection task; performance was worse and the P3b, associated with ease of detection, was attenuated. PSD had a similar, but reduced effect. Critically, the small pitch deviant resulted in less behavioural distraction following TSD compared to normal sleep.Consistent with this, the P3a, associated with the attention capture process, was significantly reduced following both TSD and PSD. Processes related to the passive switching of attention to potentially critical, but unattended, stimulus events are moderated by sleep deprivation.

The purpose of this study was to investigate the occurrence of an endogenously-evoked no-go N2b. Previous literature focused on the N2b being evoked by exogenous auditory stimuli. In this study, no-go stimuli were the absence of a gap in a 1000-ms noise burst (i.e., no-gap trials). ERPs were measured from 35 participants while performing a gap-detection task and passively listening to the same stimuli. Participants were asked to press a button when they heard a gap in the noise burst (go trials) and to withhold their button press when they did not perceive a gap in the noise burst (no-go trials). The current study's gap-detection task had predictable timing (gaps always occurred at 500 ms after noise burst onset) and high probability of gaps occurring (10:1); therefore, participants built up an expectancy that gaps would occur on most trials at 500 ms. For no-gap trials, this meant that a participant's expectancy was violated and thus a N2b-P3a response was generated. We found that all participants had N2b-P3a responses to no-gap trials. Overall, this study demonstrated that the no-go N2b-P3a response can be evoked by an endogenous signal in the form of the omission of an expected gap in noise.
