RESUMO

Various separation technologies are used to deal with the enormous amounts of animal waste that large livestock operations generate. When the recycled waste stream is land applied, it is essential to lower the pathogen load to safeguard the health of livestock and humans. We investigated whether cationic polymers, used as a flocculent in the solid/liquid separation process, could reduce the pathogen indicator load in the animal waste stream. The effects of low charge density cationic polyacrylamide (CPAM) and high charge density cationic polydicyandiamide (PDCD) were investigated. Results demonstrated that CPAM was more effective than PDCD for manure coagulation and flocculation, while PDCD was more effective than CPAM in reducing the pathogen indicator loads. However, their combined use, CPAM followed by PDCD, resulted in both improved solids separation and pathogen indicator reduction.

One practice for handling farm dairy effluent (DE) comprises recycling them to the soil with the challenge of balancing the tradeoff associated with environmental pollution through nutrient and microorganism loading. This study investigated seasonal bacterial community composition, diversity, abundance, and pathogenic indicators in untreated (Raw) and lagoon-stabilized (Lagoon) DE. The correlation between bacterial profiles and DE physicochemical characteristics was also analyzed. Pathogen-indicator bacteria were studied by enumerating viable counts and the bacterial community structure by 16S rRNA gene sequence analysis. Lagoon storage effectively reduced total solids (64%), suspended solids (77%), organic carbon (40%), and total nitrogen (82%), along with total coliforms, Escherichia coli, and enterococci. However, this efficiency was compromised in winter. Lagoon and Raw sample bacterial communities presented different compositions, with several environmental variables correlating to microbial community differences. Lagoon-treated DE exhibited the most diverse bacterial community, dominated by Firmicutes (40%), Proteobacteria (30%), and Bacteroidota (7.6%), whereas raw DE was mainly composed of Firmicutes (76%). Regardless of the season, dominant genera included Trichococcus, Romboutsia, Corynebacterium, and Paeniclostridium. Overall, the study emphasizes the importance of lagoon treatment for DE stabilization, showcasing its role in altering bacterial community composition and mitigating environmental risks associated with pathogens and nutrients, particularly in summer.

In recent years, composting has increasingly been promoted as a reliable method for sanitizing Faecal Sludge (FS) from onsite sanitation systems, particularly where there are opportunities to use the recovered nutrients in agriculture. However, there remain gaps in our understanding of the fate of infectious faecal pathogens during composting, particularly in tropical climates. This study investigated the influence of different locally available bulking agents on the inactivation efficiency of composting by tracking the fate of four key indicator organisms (E. coli, Salmonella spp., Enterococci spp., and viable helminth eggs). Dewatered FS was mixed with different bulking agents - i.e. Sawdust (SD), Coffee husks (CH) and Brewery waste (BW). Compost piles of FS:SD, FS:CH, and FS:BW in a volumetric ratio of 1:2 were set-up in duplicate (3 m3 each), composted on a pilot scale and monitored weekly for the survival of pathogen indicators for a period of 15 weeks. The study findings suggest that the different bulking agents have a statistically significant (p < 0.05) effect on the temperature evolution and survival of pathogen indicators in compost. CH was the most suitable bulking agent for composting with FS as piles containing CH exhibited higher pathogen inactivation efficiency and shorter inactivation periods of 6 weeks compared to 8 weeks for SD and BW piles. Time-temperature was the most important factor responsible for pathogen inactivation. However, other mechanisms such as indigenous microbial and toxic by-products such as NH4+-N also played an important role in the inactivation of pathogens. The results suggest that co-composting of FS with a sawdust, coffee husk or brewery waste for 8 weeks with thermophilic temperatures of about 48-60 °C sustained in the composting piles for more than 38 days, using 7 days turning frequency, is sufficient to ensure complete sanitization of FS before reuse in agriculture.

Rotavirus A (RVA), bovine torovirus (BToV), bovine enterovirus (BEV) and bovine coronavirus (BCV) at a bovine farm in Ibaraki prefecture were monitored by one-step multiplex reverse transcription polymerase chain reaction (RT-PCR), with the aim of confirming the reduction of "viral pathogen indicators". A total of 960 bovine fecal samples were collected from calves less than 2 month-old within the period from October 2016 to October 2018 every 2 months at the bovine farm. In each sampling, 40 samples were taken from calves 3 week-old or less, and 40 samples from calves over 3 week-old, in principle. At the end of September 2017, the farm introduced improvement of hygiene protocols on boots by exchanging boots and appropriate usage of a footbath at the entrance of calf sheds. In the comparison of the virus detection by RT-PCR, prevalence of all 4 viruses was significantly reduced (P<0.01) in calves 3 week-old or less after the improvement. The mortality of calves less than 2 month-old was also significantly reduced after the improvement of hygiene protocols. These data suggest that the proper control of boots at calf sheds is important, perhaps even vital, for rearing hygiene measures at bovine farms so as to attain substantial decrease in the prevalence of pathogens.

Reducing pathogenic risks in surface waters impacted by leaking or overflowing sewage requires the ability to detect human excreta in raw sewage, discriminate human excreta from other types of animal excreta, and differentiate between treated wastewater and raw sewage. We used the relative concentrations of a degradable, human-specific pharmaceutical and a persistent artificial sweetener to indicate the presence of human excreta, its degree of environmental degradation, and the amount of dilution by freshwater sources. Samples were collected and analyzed for acetaminophen and sucralose between 2016 and 2018 from wastewater treatment plants (WWTPs) and streams in metropolitan Lexington, Kentucky (USA). Both co-analytes were consistently present in raw sewage, with acetaminophen in higher concentrations than sucralose. The presence of acetaminophen was related primarily to untreated human excreta, with concentrations rapidly decreasing upon treatment to nearly undetectable levels in WWTP effluents and streams. Sucralose in surface waters was related to inputs of both raw sewage and WWTP effluents. The ratio of acetaminophen to sucralose concentrations in raw sewage and spiked river waters exhibited linear decay kinetics with respect to time, with larger decay constants observed at higher temperatures. This co-analyte indicator approach was evaluated at a local site previously suspected of receiving raw sewage. The presence and ratios of the co-analytes indicated the presence of domestic sewage that was not fully treated.

Solar disinfection (SODIS) of urban wastewater can be a suitable technology for improving the microbiological quality of reclaimed water as a complement to other extensive and environmentally friendly technologies such as microalgae biotreatment. The objective of this work is to evaluate the feasibility of incorporating the SODIS technology at the end of a pilot scale urban wastewater treatment plant (WWTP) where the processes are based on microalgae biotechnology and comprising three Upflow Anaerobic Sludge Blanket (UASB, 20m3 each one) reactor, six High Rate Algal Ponds (HRAP, 32m2 each one), and a Dissolved Air Flotation (DAF, 1m3) unit. E. coli concentration was monitored at the effluent of the different units (UASB, HRAP, DAF) of the pilot WWTP. The efficiency of the SODIS process was studied for the inactivation of three of the commonly employed indicator microorganisms (Escherichia coli, Enterococcus spp. and Clostridium perfringens) using a compound parabolic collector (CPC) for five months under various conditions of irradiance and temperature. E. coli and Enterococcus spp. were more effectively disinfected by the SODIS unit (2.9 and 2.5 logarithms of reduction on average, respectively) than by the HRAP (2 and 1.1) or the DAF (0.9 and 0.1). On the contrary, the DAF technology achieved better reduction rates of C. perfringens (1.7) than the SODIS (0.9) and the HRAP (0.1). No regrowth of any microorganisms was detected during dark storage after the SODIS treatment. Incorporating a SODIS unit after the non-conventional WWTP processes substantially increases the possibilities for reuse of the treated water after receiving a cumulative UV radiation dose of 25W·h/m2 (50min of normalized time of solar illumination). The surface requirement of the SODIS equipment would be 3.5 times smaller than the HRAP's surface.

Electro-dewatering (ED) increases biosolids dryness from 10-15 to 30-50%, which helps wastewater treatment facilities control disposal costs. Previous work showed that high temperatures due to Joule heating during ED inactivate total coliforms to meet USEPA Class A biosolids requirements. This allows biosolids land application if the requirements are still met after the storage period between production and application. In this study, we examined bacterial regrowth and odour emissions during the storage of ED biosolids. No regrowth of total coliforms was observed in ED biosolids over 7d under aerobic or anaerobic incubations. To mimic on-site contamination during storage or transport, ED samples were seeded with untreated sludge. Total coliform counts decreased to detection limits after 4d in inoculated samples. Olfactometric analysis of ED biosolids odours showed that odour concentrations were lower compared to the untreated and heat-treated control biosolids. Furthermore, under anaerobic conditions, odorous reduced sulphur compounds (methanethiol, dimethyl sulphide and dimethyl disulphide) were produced by untreated and heat-treated biosolids, but were not detected in the headspaces above ED samples. The data demonstrate that ED provides advantages not only as a dewatering technique, but also for producing biosolids with lower microbial counts and odour levels.
