RESUMO

Tropospheric ozone [O3] is a secondary air pollutant formed from the photochemical oxidation of volatile organic compounds in the presence of nitrogen oxides, and it is one of the most damaging air pollutants to crops. O3 entry into the plant generates reactive oxygen species leading to cellular damage and oxidative stress, leading to decreased primary production and yield. Increased O3 exposure has also been shown to have secondary impacts on plants by altering the incidence and response to plant pathogens. We used the Capsicum annum (pepper)-Xanthomonas perforans pathosystem to investigate the impact of elevated O3 (eO3) on plants with and without exposure to Xanthomonas, using a disease-susceptible and disease-resistant pepper cultivar. Gas exchange measurements revealed decreases in diurnal photosynthetic rate (A') and stomatal conductance ( g s ' ), and maximum rate of electron transport (Jmax) in the disease-resistant cultivar, but no decrease in the disease-susceptible cultivar in eO3, regardless of Xanthomonas presence. Maximum rates of carboxylation (Vc,max), midday A and gs rates at the middle canopy, and decreases in aboveground biomass were negatively affected by eO3 in both cultivars. We also observed a decrease in stomatal sluggishness as measured through the Ball-Berry-Woodrow model in all treatments in the disease-resistant cultivar. We hypothesize that the mechanism conferring disease resistance to Xanthomonas in pepper also renders the plant less tolerant to eO3 stress through changes in stomatal responsiveness. Findings from this study help expand our understanding of the trade-off of disease resistance with abiotic stresses imposed by future climate change.

BACKGROUND: Auxin transcription factor (ARF) is an important transcription factor that transmits auxin signals and is involved in plant growth and development as well as stress response. However, genome-wide identification and responses to abiotic and pathogen stresses of the ARF gene family in Cucurbita pepo L, especially pathogen stresses, have not been reported. RESULTS: Finally, 33 ARF genes (CpARF01 to CpARF33) were identified in C.pepo from the Cucurbitaceae genome database using bioinformatics methods. The putative protein contains 438 to 1071 amino acids, the isoelectric point is 4.99 to 8.54, and the molecular weight is 47759.36 to 117813.27 Da, the instability index ranged from 40.74 to 68.94, and the liposoluble index ranged from 62.56 to 76.18. The 33 genes were mainly localized in the nucleus and cytoplasm, and distributed on 16 chromosomes unevenly. Phylogenetic analysis showed that 33 CpARF proteins were divided into 6 groups. According to the amino acid sequence of CpARF proteins, 10 motifs were identified, and 1,3,6,8,10 motifs were highly conserved in most of the CpARF proteins. At the same time, it was found that genes in the same subfamily have similar gene structures. Cis-elements and protein interaction networks predicted that CpARF may be involved in abiotic factors related to the stress response. QRT-PCR analysis showed that most of the CpARF genes were upregulated under NaCl, PEG, and pathogen treatment compared to the control. Subcellular localization showed that CpARF22 was localized in the nucleus. The transgenic Arabidopsis thaliana lines with the CpARF22 gene enhanced their tolerance to salt and drought stress. CONCLUSION: In this study, we systematically analyzed the CpARF gene family and its expression patterns under drought, salt, and pathogen stress, which improved our understanding of the ARF protein of zucchini, and laid a solid foundation for functional analysis of the CpARF gene.

BACKGROUND: COVID-19 has triggered a global public health crisis, and had an impact on economies, societies, and politics around the world. Based on the pathogen prevalence hypothesis suggested that residents of areas with higher infection rates are more likely to be collectivists as compared with those of areas with lower infection rates. Many researchers had studied the direct link between infectious diseases and individualism/collectivism (infectious diseases→ cultural values), but no one has focused on the specific psychological factors between them: (infectious diseases→ cognition of the pandemic→ cultural values). To test and develop the pathogen prevalence hypothesis, we introduced pandemic mental cognition and conducted an empirical study on social media (Chinese Sina Weibo), hoping to explore the psychological reasons behind in cultural value changes in the context of a pandemic. METHODS: We downloaded all posts from active Sina Weibo users in Dalian during the pandemic period (January 2020 to May 2022) and used dictionary-based approaches to calculate frequency of words from two domains (pandemic mental cognition and collectivism/individualism), respectively. Then we used the multiple log-linear regression analysis method to establish the relationship between pandemic mental cognition and collectivism/individualism. RESULTS: Among three dimensions of pandemic mental cognition, only the sense of uncertainty had a significant positive relationship with collectivism, and also had a marginal significant positive relationship with individualism. There was a significant positive correlation between the first-order lag term AR(1) and individualism, which means the individualism tendency was mainly affected by its previous level. CONCLUSIONS: The study found that more collectivist regions are associated with a higher pathogen burden, and recognized the sense of uncertainty as its underlying cause. Results of this study validated and further developed the pathogen stress hypothesis in the context of the COVID-19 pandemic.

The global COVID-19 pandemic has severely impacted the passenger flow. Facing the same pandemic, various regions differ in the resilience of population mobility due to differences in the regional cultural. This study uses mobile big data to quantifies regional mobility resilience of 358 cities in China. Study results reveal the differences in regional mobility resilience of cities through spatial autocorrelation analysis, and verify the effects of regional culture on mobility resilience using a panel logit regression model based on pathogen-stress theory. Spatial heterogeneity and autocorrelation in the regional mobility resilience of Chinese cities are identified through spatial analysis, which are manifested by various hot spots over time. Moreover, the panel regression results indicate that the COVID-19 pandemic has a significant negative effect on regional mobility resilience; and that the negative effect of COVID-19 on regional mobility resilience is amplified in the cities with high degrees of dialect diversity, while it is weakened in the cities with high degrees of cultural tightness (which have strict norms and punishments for deviance). This study provides theoretical implications for mobility resilience in the context of COVID-19 and advances the pathogen-stress theory. Study findings also provide practical recommendations for regions to enhance regional mobility resilience under the challenges of future public health crisis events.

Drawing from the terror management theory (TMT) and evolutionary perspectives of Life-History Strategy, a between-subject online experiment examined the interaction effects of pre-existing death anxiety, fear-inducing media content (coronavirus threat vs. gun violence threat vs. low threat mental disorder), and intrasexual competition for mates on online dating intentions and social distancing intentions during the COVID-19 pandemic. Results indicate the interaction effects of participants' pre-experimental death anxiety and different types of fear-inducing media content on perceived fear and intention to use online dating websites/apps as well as the interaction effects of pre-experimental intrasexual competitiveness and fear-inducing media content on social distancing intention in the context of online dating. Theoretical contributions to the terror management literature and practical implications for the online dating industry are discussed.

Infection following injury is one of the major threats which causes huge economic burden in wound care management all over the world. Injury often results with poor healing when coupled by following infection. In contrast to this, we observed enhanced survival of wound infected worms compared to wounded worms in Caenorhabditis elegans wound model while infecting with Staphylococcus aureus. Hence, the study was intended to identify the mechanism for the enhanced survival of wound infected worms through LCMS/MS based high throughput proteomic analysis. Bioinformatics analyses of the identified protein players indicated differential enrichment of several pathways including MAPK signaling, oxidative phosphorylation and phosphatidylinositol signaling. Inhibition of oxidative phosphorylation and phosphatidylinositol signaling through chemical treatment showed complete reversal of the enhanced survival during wound infection nevertheless mutant of MAPK pathway did not reverse the same. Consequently, it was delineated that oxidative phosphorylation and phosphatidylinositol signaling are crucial for the survival. In this regard, elevated calcium signals and ROS including O- and H2O2 were observed in wounded and wound infected worms. Consequently, it was insinuated that presence of pathogen stress could have incited survival in wound infected worms with the aid of elevated ROS and calcium signals.

KEY MESSAGE: Recent updates in JA biosynthesis, signaling pathways and the crosstalk between JA and others phytohormones in relation with plant responses to different stresses. In plants, the roles of phytohormone jasmonic acid (JA), amino acid conjugate (e.g., JA-Ile) and their derivative emerged in last decades as crucial signaling compounds implicated in stress defense and development in plants. JA has raised a great interest, and the number of researches on JA has increased rapidly highlighting the importance of this phytohormone in plant life. First, JA was considered as a stress hormone implicated in plant response to biotic stress (pathogens and herbivores) which confers resistance to biotrophic and hemibiotrophic pathogens contrarily to salicylic acid (SA) which is implicated in plant response to necrotrophic pathogens. JA is also implicated in plant responses to abiotic stress (such as soil salinity, wounding and UV). Moreover, some researchers have recently revealed that JA controls several physiological processes like root growth, growth of reproductive organs and, finally, plant senescence. JA is also involved in the biosynthesis of various metabolites (e.g., phytoalexins and terpenoids). In plants, JA signaling pathways are well studied in few plants essentially Arabidopsis thaliana, Nicotiana benthamiana, and Oryza sativa L. confirming the crucial role of this hormone in plants. In this review, we highlight the last foundlings about JA biosynthesis, JA signaling pathways and its implication in plant maturation and response to environmental constraints.

The pathogen stress hypothesis posits that pathogen-related threats influence regional and individual differences in collectivism since behavioral practices associated with collectivism limit the spread of infectious diseases. In support of the hypothesis, previous research demonstrates the association between individualism/collectivism and pathogen stress based on historical records or experimental manipulation. However, it is still unclear whether individuals would indeed value collectivism during the outbreak of infectious diseases. Thus, we investigated the concurrent effects of pathogen-related stress on the endorsement of individualism/collectivism by examining 9322 Koreans for 14 weeks of the COVID-19 pandemic. The results revealed that the level of collectivism among respondents were higher after than before the COVID-19 outbreak. Moreover, the average level of collectivism on a given day showed a significant association with the number of confirmed COVID-19 cases on the same day during the outbreak. Interestingly, individualism did not significantly change for the same period.

Myeloperoxidases (MPOs) are heme-linked oxidative stress-generating enzymes found abundantly in azurophilic granules of polymorphonuclear neutrophils. Mature MPOs act as potent antimicrobial agents by producing hypohalous acids using hydrogen peroxide and halide ions as substrates. These acids can readily oxidize reactive groups of biomolecules on invading microbes. In this study, we identified and characterized a homolog of MPO from rock bream (Oplegnathus fasciatus), designated as RbMPO. We analyzed the RbMPO gene for its basal expression level in physiologically important tissues and for transcriptional changes under different pathogenic stress conditions. The complete coding sequence of RbMPO consisted of 2652 nucleotides encoding an 884 amino acid sequence with a predicted molecular mass of 99.7 kDa. Our in silico analysis confirmed the typical MPO domain arrangement in RbMPO, including the propeptide, large chain and heavy chain, along with the heme peroxidase signature. Intriguingly, a C1q domain was also identified in the C-terminal region of the derived amino acid sequence. Most of the known functionally important residues of MPOs are found to be well conserved in RbMPO, showing a close evolutionary relationship with other teleostan MPOs, particularly with that of mandarin fish. RbMPO exhibited a ubiquitous basal expression in physiologically relevant tissues, with particularly high expression levels in blood cells. Basal transcript levels of RbMPO in gill and spleen tissues were found to change upon different pathogen or pathogen-derived mitogen stimulation, with detectable inductive responses. Together, these data suggest the potential involvement of RbMPO in the innate immune response in rock bream.

Social trust is derived from the interaction of environmental and social factors, which has important significance for the sustainable development of society and social governance. In particular, in the post-pandemic era, tourist activity will receive special attention in terms of its role in the development of the public's social trust. On the basis of the sample of big data, this research takes China as an example to study the influences of different geographical and environmental elements on individuals' social trust as well as the common role played by the tourist activity. The research showed that the geographical environment and tourism activities have interacting effects on public social trust. This influencing mechanism is specifically manifested as the rice-growing ratio and tourist reception level can have interacting effects on the social trust of the residents in a tourist destination; pathogen stress and tourist supply level can exert interacting effects on the social trust of the residents in an area from which tourists originate; and economic development and tourist reception level can have interacting effects on the social trust of the residents in a tourist destination. By doing so, this research provides theoretical support and practical suggestions for the recovery of the public's social trust from the perspective of tourism geography in the post-pandemic era.

What environmental factors are associated with individual differences in political ideology, and do such associations change over time? We examine whether reductions in pathogen prevalence in U.S. states over the past 60 years are associated with reduced associations between parasite stress and conservatism. We report a positive association between infection levels and conservative ideology in the United States during the 1960s and 1970s. However, this correlation reduces from the 1980s onwards. These results suggest that the ecological influence of infectious diseases may be larger for older people who grew up (or whose parents grew up) during earlier time periods. We test this hypothesis by analyzing the political affiliation of 45,000 Facebook users, and find a positive association between self-reported political affiliation and regional pathogen stress for older (>40 years) but not younger individuals. It is concluded that the influence of environmental pathogen stress on ideology may have reduced over time.

The potato plant is one of the main crops in the world. However, relatively little is known about key virulence factors of major fungal and bacterial diseases in potatoes, biocontrol measures to improve activity and stability, and the core driving forces in the control process. Here, we focus on analyzing the mechanisms by which genes, proteins, or (and) metabolites of potato pathogens as key virulence factors. Then, the single strain biocontrol agents, synthetic microbial communities, microbial microcapsule strategies were introduced, and the latter two strategies can improve stability and activity in biocontrol. Meanwhile, summarized the defense mechanisms of biocontrol and their specific issues in practical applications. Furthermore, explore how potato crop management, soil management, and climate effects, as crucial driving forces affect potato biocontrol in the system. Dynamic and systematic research, excavation of biocontrol strain resources, find the causes of regional disease resistance and exploration of biocontrol mechanism will provide promising solutions for biotic stress faced by potato in the future.

Rust infection results in stress volatile emissions, but due to the complexity of host-pathogen interaction and variations in innate defense and capacity to induce defense, biochemical responses can vary among host species. Fungal-dependent modifications in volatile emissions have been well documented in numerous host species, but how emission responses vary among host species is poorly understood. Our recent experiments demonstrated that the obligate biotrophic crown rust fungus (P. coronata) differently activated primary and secondary metabolic pathways in its primary host Avena sativa and alternate host Rhamnus frangula. In A. sativa, emissions of methyl jasmonate, short-chained lipoxygenase products, long-chained saturated fatty acid derivatives, mono- and sesquiterpenes, carotenoid breakdown products, and benzenoids were initially elicited in an infection severity-dependent manner, but the emissions decreased under severe infection and photosynthesis was almost completely inhibited. In R. frangula, infection resulted in low-level induction of stress volatile emissions, but surprisingly, in enhanced constitutive isoprene emissions, and even severely-infected leaves maintained a certain photosynthesis rate. Thus, the same pathogen elicited a much stronger response in the primary than in the alternate host. We argue that future work should focus on resolving mechanisms of different fungal tolerance and resilience among primary and secondary hosts.

The emotion of disgust is suggested to be an adaptation that evolved to keep us away from sources of infection. Therefore, individuals from populations with greater pathogen stress should have a greater disgust sensitivity. However, current evidence for a positive relationship between disgust sensitivity and the intensity of infectious diseases in the environment is limited. We tested whether disgust and contamination sensitivity changed in response to the COVID-19 pandemic. Disgust was assessed in 984 women in 2017 (before pandemic) and 633 women in 2020 (during pandemic) by a set of photographs depicting sources of infection and Pathogen and Moral of Three-Domain Disgust Scale. Further, contamination sensitivity among participants in two waves was measured by Contamination Obsessions and Washing Compulsions Subscale of Padua Inventory. State anxiety was measured with the Polish adaptation of the State-Trait Anxiety Inventory (STAI) only during the second wave of data collection. Women from the COVID-19 pandemic group assessed the photographs depicting sources of infection as more disgusting, scoring higher on Padua Inventory, but lower on Moral Disgust Domain as compared to women from before the pandemic. In addition, anxiety levels during pandemic positively correlated with scores from Pathogen Disgust Domain, Padua Inventory, and the ratings of the photographs. The participants of the study scored higher in state anxiety than the norms determined for the Polish population. Summarizing, we present evidence for differences in individual levels of disgust sensitivity in relation to pathogen stress, supporting the idea that disgust evolved to serve as protection from pathogens.

The severity of the environment has been found to have played a selective pressure in the development of human behavior and psychology, and the historical prevalence of pathogens relate to cultural differences in group-oriented psychological mechanisms, such as collectivism and conformity to the in-group. However, previous studies have also proposed that the effectiveness of institutions, rather than pathogen stress, can account for regional variation in group-oriented psychological mechanisms. Moreover, previous studies using nations as units of analysis may have suffered from a problem of statistical non-independence, namely, Galton's problem. The present study tested whether or not regional variation in pathogen stress, rather than government effectiveness, affected collectivism and conformity to social norms by adjusting the effect of global regions using hierarchical Bayesian estimation. We found that the overall effect of pathogen stress remained significant in only one out of the four indices of the regional variability of conformity, and the effects of the government effectiveness also disappeared. Instead, we found that significant effects of both pathogen stress and government effectiveness in specific regions of the world, but these effects were not stable across the measurements. These results indicate that both the effects of pathogen stress and government effectiveness need further reevaluation.

Chickpea (Cicer arietinum); the second largest legume grown worldwide is prone to drought and various pathogen infections. These drought and pathogen stresses often occur concurrently in the field conditions. However, the molecular events in response to that are largely unknown. The present study examines the transcriptome dynamics in chickpea plants exposed to a combination of water-deficit stress and Ralstonia solanacearum infection. R. solanacearum is a potential wilt disease causing pathogen in chickpea. Drought stressed chickpea plants were infected with this pathogen and the plants were allowed to experience progressive drought with 2 and 4 days of R. solanacearum infection called short duration stress (SD stresses) and long duration stress (LD stresses), respectively. Our study showed that R. solanacearum multiplication decreased under SD-combined stress compared to SD-pathogen but there was no significant change in LD-combined stress compared to LD-pathogen. The microarray analysis during these conditions showed that 821 and 1039 differentially expressed genes (DEGs) were unique to SD- and LD-combined stresses, respectively, when compared with individual stress conditions. Three and fifteen genes were common among all the SD-stress treatments and LD-stress treatments, respectively. Genes involved in secondary cell wall biosynthesis, alkaloid biosynthesis, defense related proteins, and osmo-protectants were up-regulated during combined stress. The expression of genes involved in lignin and cellulose biosynthesis were specifically up-regulated in SD-combined, LD-combined, and LD-pathogen stress. A close transcriptomic association of LD-pathogen stress with SD-combined stress was observed in this study which indicates that R. solanacearum infection also exerts drought stress along with pathogen stress thus mimics combined stress effect. Furthermore the expression profiling of candidate genes using real-time quantitative PCR validated the microarray data. The study showed that down-regulation of defense-related genes during LD-combined stress resulted in an increased bacterial multiplication as compared to SD-combined stress. Overall, our study highlights a sub-set of DEGs uniquely expressed in response to combined stress, which serve as potential candidates for further functional characterization to delineate the molecular response of the plant to concurrent drought-pathogen stress.
