Premna integrifoliaL. (Lamiaceae) is widely used in herbal formulation "Dashmoolarishta" which is useful in postnatal care. Ethyl acetate extract obtained from the leaves was evaluated for phenolic content and its antioxidant activity. Acute and subacute toxicity of the extract was studied in mice of both sexes to get an idea about LD50 value and assessed its safety profile before its application as a protective agent against different toxicities induced by xenobiotics. Phenol enriched extract (phenol content is 63.10 ± 1.26 mg/g of gallic acid equivalent and flavonoid content 75.33 ± 0.23 mg/g of rutin equivalent) showed good antioxidant activity. In acute toxicity studies it was observed that single different doses (300-5000 mg/kg b.wt.) of extract did not show any mortality of mice. Thus the LD50 of the extract was determined, and it was higher than 5000 mg/kg. There was no major change in behavioral and general appearance of mice. External morphology of liver, kidneys, lungs, spleen and heart did not show any effect of treatment. In subacute toxicity no statistically significant change in body weight, relative organ weight, food intake and water uptake, hematological, biochemical parameters were reported after comparison with control. Extract did not show significant effect in the level of antioxidant enzymes in the liver of mice of treated groups. No histopathological changes were observed in liver and kidney tissues. Thus, extract did not show any sign of toxic effects, when administered orally to male and female mice at dose level up to 1000 mg/kg. So, it can be utilized as protective agent against toxicity produced by different xenobiotics.

BACKGROUND: Premna integrifolia Linn. is a medicinal plant of an Ayurvedic importance and proved to have an anti-inflammatory, anti-diabetic, anti-microbial and hypo-lipidemic activity. Glutathione (GSH) redox status is an important parameter to assess the antioxidant activity of any neutraceuticals. OBJECTIVE: In order to assess the anti-oxidant potential of hydro alcoholic extract (HAE) of P. integrifolia, this study was aimed to evaluate the GSH redox status in high fat diet induced experimental atherosclerosis. MATERIALS AND METHODS: The present study comprises sixty Wistar rats and they were divided into six groups: the first group served as control, the second group was fed with high fat diet and the third, fourth and fifth groups were fed with high fat diet along with various concentrations of HAE of 200, 400 and 500 g/kg.b.wt respectively and the sixth group was administered high fat diet along with 10 mg/kg b.wt of atorvastatin for 30 days. GSH-dependent enzymes like GSH-peroxidase (GPx), GSH-reductase (GR) and glucose 6-phosphate dehydrogenase (G6PD) were estimated in hemolysate, kidney, heart and liver of experimental rats. RESULTS: Analysis of GSH levels showed a significant decrease in hemolysate, heart and kidney (p < 0.05) and liver (p < 0.01) in high fat-fed rats when compared to control. Activities of GPx, GR and G6PD in hemolysate and heart (p < 0.001), liver and kidney (p < 0.05) in high fat-fed rats when compared to control. Dose-dependent increase was observed in rats treated with various concentrations of HAE. CONCLUSION: The HAE of root bark of P. integrifolia is proved to have a protective role on antioxidant defense in high fat diet induced atherosclerosis model. As a whole P. integrifolia increases the GSH content in a dose-dependent manner and in turn altered the redox cycle.

The present study evaluated the hepatoprotective role of ethanol extract of P. integrifolia leaves (EEPL) on aflatoxin B1 (AFB1)-induced toxicity in mice. Mice were administered with AFB1 (0.1 mg/kg b. wt., orally) for 90 days, EEPL (400 and 600 mg/kg b. wt., orally) and silymarin (100 mg/kg b. wt., orally) in combination with AFB1. The study shows the protective effect of EEPL by the restoration of altered hematological indices and liver marker enzymes. Restoration of lipid peroxidation and glutathione content, along with activities of antioxidant enzymes, suggest amelioration of oxidative stress in AFB1-intoxicated mice. In addition, EEPL attenuated apoptosis and histopathological alterations in liver tissue. In conclusion, the current study suggests that EEPL protect mice liver against AFB1 toxicity by inhibiting oxidative stress and apoptosis. The protective activity of EEPL may be due to the enrichment of flavonoids (neohesperidin, apigenin-7-O-glucoside, catechin hydrate, cyanidin chloride, quercetin-3-galactoside, diosmin, genistein, malvin chloride, 4-hydroxy-3-methoxycinnamic acid, kaempferol-3-O-alpha-L-arabinoside, myricitrin, poncirin, vitexin and tiliroside) in the extract as identified by UPLC-QTOF-MS/MS.

The present study was designed to evaluate the ameliorative effect of ethyl acetate extract of Premna integrifolia L. (EAEPI) leaves in cyclophosphamide (CP)-induced hepatic injury in mice. Mice were intoxicated with CP (200 mg/kg b. wt., i.p.) for 5 weeks or EAEPI (400 and 600 mg/kg b. wt., orally) in combination with CP. The results demonstrated that EAEPI exerts protective effect against CP induced hepatotoxicity, as evident from restoration of altered haematological parameters and alleviations of liver marker enzymes in serum. EAEPI also attenuated oxidative stress and antioxidant markers as evident from reversal of lipid peroxidation, glutathione levels as well as activities of catalase and superoxide dismutase enzymes. Moreover, EAEPI attenuated apoptosis and histopathological liver tissue damage in CP-intoxicated mice. In conclusion, EAEPI could protect mice liver against cyclophosphamide toxicity by inhibiting oxidative stress and apoptosis.The protective activity of EAEPI may be due to presence of polyphenolic compounds as identified by UHPLC-Q-TOF-MS/MS.

Premna integrifolia (Agnimantha brihat) is a traditional medicinal plant with a prominent place in Ayurveda, Siddha and Unani systems of medicine. In this study we have evaluated the anti-inflammatory and immunomodulatory properties of the Premna integrifolia root extracts employing cyclooxygenase-1 (COX-1), cyclooxygenase-2 (COX-2), and 5-lipoxygenase (5-LOX) enzyme-based assays, lymphocyte proliferation assay, pro-and anti-inflammatory cytokines measurement. Petroleum ether extract (PEE) of Premna integrifolia showed potent inhibition of COX-2 and 5-LOX with IC50 values of 6.15 µg/mL and 11.33 µg/mL respectively. In in vitro studies on RAW 264.7 cell line, PEE showed inhibition in the formation of nitric oxide (NO), pro-inflammatory cytokines (IL-1ß, IL-6), prostaglandin E2 (PGE2) production, induction of anti-inflammatory cytokine (IL-2) and down-regulation of expression of COX-2, 5-LOX, TNF-α, IL-1ß and iNOS. PEE also significantly reduced carrageenan-induced paw edema in mouse model of inflammation. Further, attempts in isolating the active principle(s) involved in these anti-inflammatory effects of PEE by separation on RP-HPLC resulted in the isolation of four active peaks, H1, H2, H3 and H5, inhibiting COX-1, COX-2 and 5-LOX, out of which H3 was identified as 6- hydroxy salvinolone (6-HS). Present findings reveal that PEE of roots of Premna integrifolia exhibits potent anti-inflammatory and immunomodulatory activities, which could form a potential source for development of anti-inflammatory drugs. 6-HS, a COX-2/5-LOX dual inhibitor along with other lead molecules isolated from PEE of Premna integrifolia may form lead molecules for the development of COX-LOX dual inhibitors.

Two new tetrahydrofuran lignans, taungtangyiols A (1) and B (2), and eight known furofuran lignans (3-10), were isolated from the chloroform extract of Premna integrifolia wood collected in Myanmar. Their structures were elucidated by extensive spectroscopic techniques. The X-ray crystal structure of 1 clearly indicated its relative configuration. Taungtangyiols A (1) and B (2) inhibited the deposition of melanin in B16F10 mouse melanoma cells, with IC50 values of 50.7 and 40.9 µM, respectively, without notable cytotoxicity. An SAR study demonstrated that the furofuran and dioxymethylene moieties of the lignans play a vital role in inhibiting melanogenesis.

Premna integrifolia Linn. (Verbenaceae) is an important constituent of the formulation of ten roots of herbs known as Dasamula and is widely used for treating various ailments in the Indian system of medicine. Aim of this review is to provide comprehensive information on the pharmacological activities of various parts of P. integrifolia. All the relevant universally accepted electronic databases were searched with respect to the terms "Agnimantha", "Headache tree", "Premna integrifolia", "Premna obtusifolia", "Premna serratifolia" including Indian classical texts, pharmacopoeias, Ayurvedic books, journals, etc., for information without specific timeline. Complete information of the plant has been collected manually since the year 1964 and has been arranged chronologically. The collected data reflects that many ethno-medicinal claims have been confirmed through the modern in-vitro and in-vivo pharmacological studies using different extracts and their isolates of P. integrifolia. The isolation of active constituents, their biological actions, clinical safety and validation of traditional uses of P. integrifolia could provide leads for further scientific research. The information collected here will be useful to set-up research protocols for modern drugs and Ayurvedic formulation development.

Premna integrifolia Linn. (Verbenaceae) is an important woody, medicinal plant and has been prominent place in Ayurveda, Siddha and Unani system of medicines. Objective of the present review is to avail the comprehensive information on ecological biodiversity, traditional uses and phytochemistry of P. integrifolia. Information of the plant was searched using various electronic databases in reference to the terms Premna integrifolia, ecological biodiversity, traditional uses and phytoconstituents of P. integrifolia along with Ayurvedic books, Indian classical texts, pharmacopoeias, journals, etc. There is an inherent difference within the three Ayurvedic Formulary of India (AFIs) published with regard to the botanical sources of Agnimantha. Complete data of the plant has been collected manually since from the years 1947-2015 and was arranged accordingly. Available data have reports that roots of P. integrifolia are widely used for the preparation of Ayurvedic formulations like Dasamulakvatha, Arista, Curna and Chayawanprashavleh for the treatment of a variety of afflictions. It has also reported to have p-methoxy cinnamic acid, linalool, linoleic acid, ß-sitosterol and flavone luteolin, iridoid glycoside, premnine, ganiarine and ganikarine, premnazole, aphelandrine, pentacyclic terpene betulin, caryophellen, premnenol, premna spirodiene, clerodendrin-A, etc., phytoconstituents in its various parts. There is need to validate its traditional uses, isolation and confirmation of reported phytoconstituents, biological and clinical efficacy by modern analytical and biological techniques which could be recommendation for further scientific research.

OBJECTIVE: Premna integrifolia Linn (Family: Verbenaceae) synonym of Premna serratifolia has tremendous medicinal value. Preliminary pharmacological studies were performed on the methanolic extract of Premna integrifolia (MEPI) bark to investigate neuropharmacological, analgesic, and anti-inflammatory activities. MATERIALS AND METHODS: Neuropharmacology study was done by open field and hole cross test whereas acetic acid writhing test and formalin induced pain was done for analgesic activity of MEPI. Carrageenan induced inflammatory model was considered for anti-inflammatory activity evaluation. RESULTS: A statistically significant (p0.05) decrease in locomotor activity was observed at all doses in the open-field and hole-cross tests. The extract significantly (p0.05) and dose dependently reduced the writhing reflex in the acetic acid-induced writhing test as well as licking response in the formalin induced inflammatory pain. At 200 mg/kg body weight dose, MEPI showed 71.16% inhibition in carrageenan induced anti-inflammatory activity. CONCLUSION: The finding of this study suggests that MEPI will provide scientific support for the use of this species in traditional medicine.

AIM OF THE STUDY: Aim of the present study was to evaluate the anti-obesity activity of chloroform:methanol extract of P. integrifolia (CMPI) in mice fed with cafeteria diet. MATERIALS AND METHODS: Female Swiss Albino mice were divided into six groups, which received normal and cafeteria diet, standard drug simvastatin (10 mg/kg) and CMPI (50, 100 and 200 mg/kg) daily for 40 days. Parameters such as body weight, body mass index (BMI), Lee index of obesity (LIO), food consumption, locomotor behavior, serum glucose, triglyceride, total cholesterol, high density lipoprotein (HDL), low density lipoprotein (LDL), very low density lipoprotein (VLDL), atherogenic index, organ weight and organ fat pad weight were studied for evaluating the anti-obesity activity of P. integrifolia. High performance liquid chromatography (HPLC) fingerprint profile of chloroform-methanol extract was also studied using quercetin as the reference standard. RESULTS: There was a significant increase in body weight, BMI, LIO, food consumption, organ weight (liver and small intestine), organ fat pad weight (mesenteric and peri-renal fat pad) and in the levels of serum glucose, triglyceride, total cholesterol, LDL and VLDL with a significant decrease in locomotor behavior (ambulation, rearing, grooming) and HDL level in cafeteria diet group. Animals treated with CMPI showed dose dependent activity. P. integrifolia (200 mg/kg) supplementation attenuated all the above alterations, which indicates the anti-obesity activity. HPLC fingerprint profile of CMPI showed two peaks in the solvent system of 50 mm potassium diphosphate (pH-3 with ortho phosphoric acid): Methanol (30:70 v/v) at 360 nm. CONCLUSION: Present findings suggest that, CMPI possessed anti-obesity activity that substantiated its ethno-medicinal use in the treatment of obesity.

ABSTRACT This study was designed to examine the chemical composition and in vitro antioxidant activity of the hydrodistillated essential oil and various extracts obtained from Premna integrifolia Linn. GC-MS analysis of the essential oil was resulted in determination 29 different compounds, representing 95.73% of total oil. Antioxidant activities of the essential oil and organic extracts of chloroform, ethyl acetate and methanol were determined by three different test systems namely DPPH (2,2-diphenyl-1-picrylhydrazyl), superoxide and nitric oxide radical scavenging assays. The essential oil and methanol extract showed potent antioxidant activity among all the tested samples. Furthermore, the amount of total phenolic compounds was determined and its content in methanol extract was the highest as compared to other samples. The results indicate that the essential oil and extracts of Premna integrifolia could serve as an important bio-resource of antioxidants for using in the pharmaceutical industries.
