RESUMO

Biogenic nanosilver (bNAg) has emerged as a potentially less toxic alternative to synthetic nanosilver. However, no studies have evaluated its effects on a fish species from the Neotropical region. Thus, our aim was to evaluate the effects of a bNAg on the Neotropical fish Prochilodus lineatus. For this purpose, after 24 h of exposure to 100 µg L-1 of bNAg, blood samples were collected to evaluate hematological, genotoxic, and plasma parameters. Gills and liver were sampled to evaluate biomarkers of oxidative stress and brain samples were used to assess neurotoxicity. The fish presented hyperglycemia, an increased number of erythrocytes, a reduction in antioxidant enzyme activity in both tissues evaluated, and a decrease in lipid peroxidation in the gills. We concluded that P. lineatus is a sensitive species to this nanoparticle, since it presented alterations in several biomarkers after an acute exposure.

The long-term covariation (2002-2017) of lipids, polychlorinated biphenyls (PCBs), and sewage tracers was studied in the detritivorous fish (Prochilodus lineatus) and settling detritus from the Rio de la Plata. Fatty fish from polluted Buenos Aires area (BA) exhibited a significant decrease of muscle lipids (71 ± 12 to 29 ± 8.6% dry weight; p < 0.0001), triglycerides (94 to 85%, p < 0.001), and 18 carbon fatty acids (18C-FA: 59 ± 4.8 to 48 ± 1.4%; p < 0.01), reflecting a reduction of lipid accumulation, largely triglycerides enriched in 18C-FA, with a concomitant ∼20-times decline of PCBs (∼20 to >1 µg g-1 dw). The 2017 individuals of the BA series converged with leaner and more pristine northern fish (N), which showed no significant temporal variation (20 ± 10% lipids, 67 ± 8.7% triglycerides, 41 ± 8.1% 18C-FA, and 0.22 ± 0.42 µg g-1 dw PCB). In contrast, the fecal sterol tracer coprostanol remained abnormally higher in BA fish muscle with no significant temporal trend (120 ± 102 vs 6.6 ± 10 µg g-1 dw or 4.4 ± 2.8 vs 0.63 ± 1.2% sterols at N). The same pattern was observed in BA settling detritus, i.e., a temporal decrease of PCBs with high, stable coprostanol concentrations denoting sustained sewage inputs, while northern detritus was enriched in plant sterols. This long-term covariation of lipids and PCBs in fish muscle from polluted BA converging with more pristine and homogeneous northern specimens while maintaining a sewage-derived diet provides rare field evidence of the declining effect of PCBs controlling the temporal variation of muscular lipids in fish.

The flavonoid metal-insecticide [Mg(hesp)2(phen)], denominated MgHP, has high potential for controlling agricultural pests. If applied in large scale, it may reach aquatic ecosystems and be harmful to the biota. This study evaluated the effects of MgHP in the gills of the Neotropical fish, Prochilodus lineatus by determining the activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione s-transferase (GST), and the levels of glutathione (GSH) and lipid peroxidation (LPO) after 24 and 96 h exposure to 0, 1, 10, 100 and 1000 µg L-1. The histopathological changes with emphases to mitochondria-rich cells (MRC) were evaluated as well. After 24 h exposure the enzyme activities and the GSH and LPO levels were unchanged however, after 96 h exposure to high MgHP concentration (1000 µg L-1), the GST activity and GSH levels increased. Oxidative stress measured as LPO levels did not occur after MgHP exposure in both periods. Gill tissue alterations increased after MgHP exposure to 10, 100 and 1000 µg L-1. Cellular atrophy, pillar cells changes, filament epithelium hyperplasia and hypertrophy, lamellar epithelium hyperplasia were the most frequent histopathology. MRC in the filament epithelium decreased after exposure to 24 h and increased after 96 h indicating possible transitory osmo-ionic disruption. P. lineatus exhibited high tolerance to MgHP. The increased GST activity and GSH levels after 96 h exposure suggested possible MgHP accumulation and concentration- and time-dependent response. Histopathology in the gills of exposed fish occurred at high MgHP concentrations. These results suggested that the MgHP into water, at high concentrations, affect the gills by changing GST activity, GSH levels and histology being useful biomarkers for MgHP water contamination.

The aim of the present study was to perform comparative histological analyses of the ontogenetic development of two fish species endemic to the São Francisco River in Brazil: Prochilodus argenteus and Lophiosilurus alexandri. Histological analyses were performed every 24 h from the moment of hatching until 14 days post-hatching (dph) for the observation of larval development and until 39 dph for the observation of gonadal development. Whole larvae were fixed in Bouin's solution and the histological slides were stained with haematoxylin-eosin. Lophiosilurus alexandri larvae had a larger body size compared with P. argenteus larvae since hatching. Lophiosilurus alexandri larvae had mouth opening and pigmentation of the eyes upon hatching, whereas these events were observed at 1 dph in P. argenteus larvae. The visualisation and the inflation of the swim bladder occurred at 1 and 3 dph, respectively, in the P. argenteus, whereas these events occurred at 2 and 8 dph, respectively, in L. alexandri. Yolk granules were absorbed at 4 dph in P. argenteus and the 10 dph in L. alexandri. At 7 dph, the digestive tube was more differentiated in L. alexandri than P. argenteus and at 14 dph, the digestive system of both species had features of their eating habits: broad stomach and short intestine in L. alexandri, typical of carnivorous habits; stomach with a mechanical function and long intestine in P. argenteus, typical of detritivorous habits. The epithelial lining tissue, formed by a single layer of cells in the newly hatched larvae (0 dph), differentiated throughout the study, exhibiting scales in P. argenteus and numerous club cells in the middle epithelial region of L. alexandri at 39 dph. Undifferentiated gonads with somatic cells and primordial germ cells were observed at 39 dph, with caudal-cranial migration since 1 dph in both species. The anatomic changes during the ontogeny of P. argenteus and L. alexandri larvae are directly associated with the evolutionary history of each species, which explains their feeding habits, behaviour and distribution in the environment: Prochilodus argenteus is detritivorous and actively swims in the water column, whereas L. alexandri is carnivorous and inhabits bottom regions. At 39 dph neither species exhibited sexual differentiation.

Increasing pollution levels have turned our attention to assessing lethal and sublethal effects of toxic agents using the most informative techniques possible. We must seek non-invasive or non-lethal sampling methods that represent an attractive alternative to traditional techniques of environmental assessment in fish. Detergents are amongst the most common contaminants of water bodies, and LAS (Linear Alkylbenzene Sulfonate) is one of the most used anionic surfactant on the market. Our study analyzed morphological alterations (histological and histochemical) of the scale epithelium of Prochilodus lineatus under exposure to two concentrations of LAS, 3.6mg/L and 0.36mg/L, for a period of 30 days and evaluated at 14, 21 and 30 days. In order to establish morphological analysis of the scale epithelium as a new non-lethal environmental assessment tool that is reliable and comparable to classic methods, the relative sensibility of this technique was compared to a commonly used method of environmental assessment in fish, the estimation of the effects of pollutants upon branchial morphology. Two experiments were carried out, testing animals in tanks, and in individual aquariums. Results of analyses on gill tissue show that exposure to 3.6mg/L of surfactant caused severe damage, including hyperplasia, hypertrophy and fusion at 14 days, with aneurisms at 21 and 30 days; while exposure to 0.36mg/L had lighter effects on the organ, mainly lower incidence of fusion and hyperplasia. Aditionally, scale morphology was altered severely in response to 3.6mg/L of LAS, consistently showing increased mucous and club cell production. Epithelial thickness was the most variable parameter measured. Scale epithelium sensibility has the potential to be a reliable environmental marker for fish species since it has the advantage of being less invasive when compared to traditional methods. However, more studies are required to increase the robustness of the technique before it can be generally applied.

In this study, diet-tissue discrimination factors and turnover rates were determined from the somatic tissues of a detritivorous fish Prochilodus lineatus. The carbon (Δ(13) C) and nitrogen (Δ(15) N) diet-tissue discrimination factors varied for all feed rations with a range of Δ(13) C values between -1·9 and 3·6‰ and Δ(15) N between 3·3 and 5·7‰. Carbon turnover rate in the blood was 23·1 days for the C3 ration and 34·7 days for the C4 ration, in the liver was 9·9 days under the C3 ration and nitrogen turnover rate was the same (23·1 days) in the liver for both C4 and C3 -C4 rations, and 13·9 days in the muscle for C3 -C4 ration.

Twenty polymorphic microsatellite markers were developed for the Neotropical fish Leporinus obtusidens using a next generation sequencing approach and tested in two other characifomes species, Schizodon platae and Prochilodus lineatus. Microsatellite loci alleles in L. obtusidens ranged between 2 and 20 alleles per locus (mean = 5·7), with expected heterozygosity values ranging from 0·097 to 0·956 (mean = 0·578) and observed heterozygosity values ranging from 0·000 to 0·800 (mean = 0·400) in a sample of 20 specimens from the lower Paraná River (Argentina). Most of these markers will be a valuable tool for captive breeding and stocking programmes, as well as for analyses of population connectivity and genetic structure in this broadly distributed Neotropical migratory fish.

Background: Semaprochilodus insignis is a migratory fish of commercial and subsistence importance to communities in the Amazon. Despite the high intensity of exploitation, recent studies have not been carried out to assess the genetic status of its stocks. Methods: This study is the first to estimate genetic diversity and to test the existence of spatial and temporal structuring of S. insignis through sequencing of the mtDNA control region (n = 241) and eight microsatellite loci (n = 180) of individuals sampled at 11 sites distributed in the Brazilian Amazon basin. Results: Results for both markers were congruent, revealing a homogeneous genetic diversity in all the sampled locations, in addition to the absence of spatial and temporal genetic structure, indicating that the species forms a large panmictic population in the Brazilian Amazon. Discussion: Although overfishing does not yet appear to have affected the levels of genetic variability of S. insignis, signals of reduction of the effective population size and a bottleneck provide an early alert to the effects of overfishing. Thus, the ever-decreasing populations may threaten S. insignis in the future. Therefore, it is hoped that the results of this study may contribute to the elaboration of management plans or any other measures that aim at the management and conservation of this species of great importance for the Amazon basin.

The Aquidauana River is one of the most important rivers in the Pantanal region, Brazil. However, its waters have been contaminated by nearby anthropogenic activities, threatening native fish species. In this study, our objectives were: 1) to determine the concentrations of Al, As, Cd, Cu, Fe, Mn, Ni, Pb, and Zn in water and sediment samples from the Aquidauana River and to assess the risks posed to aquatic biota; 2) to quantify the concentration of these elements in muscle and liver tissue samples from four native fish species; 3) to evaluate the potential bioaccumulation of inorganic elements in the muscles and liver; and 4) to investigate genotoxicity biomarkers and their association with the inorganic element concentrations present in the muscle tissue. Water and fish samples were collected in November 2020. The concentrations of Al, As, Cd, Cu, Fe, and Pb in the water samples were in disagreement with the Brazilian legislation and presented risks to the aquatic biota. In terms of mixtures of inorganic elements, there was a great increase in the risk to biota. The As concentration did not meet the Brazilian standard for sediments in the sample collected at sampling site 6. The concentrations of Cd and Pb in the muscle tissue of Hypostomus regani, Prochilodus lineatus, Brycon hilarii, and Mylossoma duriventre exceeded the Brazilian standards for human consumption. H. regani showed greater genotoxic damage, and the higher the Al and Fe concentrations in the muscle tissue, the higher the frequencies of lobulated nuclei and nuclear invaginations. Together, our results demonstrate the negative impacts on native fish species from the Aquidauana River contamination and indicate risks to Pantanal biodiversity.

Scales of Prochilodus magdalenae, a Colombian endemic fish species, were used to obtain chitosan for application as an antibacterial agent integrated into starch-based films. Analysis of its composition during the demineralization and deproteinization process indicated that minerals and protein were both removed successfully. At this point, mild conditions for the deacetylation process were employed, namely, 2, 4, and 6 wt.% NaOH at room temperature for 16 h. Chitosan processed under 2 wt.% NaOH had low molecular weight, with the lowest value of 107.18 ± 24.99 kDa, which was closely related to its antibacterial activity. Finally, this chitosan was integrated into a banana starch-based film, and its antibacterial activity was assayed in Escherichia coli and Staphylococcus aureus cultures, with positive results in the former culture, especially due to the low-molecular-weight characteristic of chitosan.

Fipronil is a broad-use insecticide with severe toxicity to fish. Biomarkers responses and bioaccumulation were evaluated on Prochilodus lineatus after exposure to environmentally relevant concentrations of fipronil (0.5 µg L-1, 9 µg L-1, and 100 µg L-1) in a prolonged flow-through assay and ex vivo gills short-term exposition. Lipid peroxidation (LPO), oxidatively modified proteins (PO), the activity of superoxide dismutase (SOD), the content of reduced glutathione (GSH), antioxidant capacity against peroxyles (ACAP), and acetylcholinesterase (AChE) were evaluated. Besides, levels of fipronil and metabolites were analyzed by GC-ECD. At the end of the flow-through assay, fipronil, Fp. sulfone and Fp. desulfinyl were detected in fish, being liver the target organ. Fipronil prolonged exposition promoted oxidative damage in lipids and proteins, alterations in the defense system and low-antioxidant capacity in organs of P. lineatus. The brain AChE activity was affected after prolonged exposition. Ex vivo gills exposition to fipronil promoted changes in antioxidant capacity and damage to lipids, providing a fast and suitable test to assess the pesticide exposure in fish. The results revealed that fipronil at environmental concentrations would be an inducer of oxidative stress in this fish, becoming a vulnerable species to the effects of fipronil in aquatic environments.

B or supernumerary chromosomes are dispensable elements that are widely present in numerous eukaryotes. Due to their non-recombining nature, there is an evident tendency for repetitive DNA accumulation in these elements. Thus, satellite DNA plays an important role in the evolution and diversification of B chromosomes and can provide clues regarding their origin. The characiform Prochilodus lineatus was one of the first discovered fish species bearing B chromosomes, with all populations analyzed so far showing one to nine micro-B chromosomes and exhibiting at least three morphological variants (Ba, Bsm, and Bm). To date, a single satellite DNA is known to be located on the B chromosomes of this species, but no information regarding the differentiation of the proposed B-types is available. Here, we characterized the satellitome of P. lineatus and mapped 35 satellite DNAs against the chromosomes of P. lineatus, of which six were equally located on all B-types and this indicates a similar genomic content. In addition, we describe, for the first time, an entire population without B chromosomes.

Complete mitochondrial sequences can be rapidly obtained and are widely available, providing a great source of species information and allowing for the discovery of new specific molecular markers. However, for some taxonomic groups, traditional approaches for species delimitation are impaired by the low genetic distance values. In these cases, other species-level markers are used. For Prochilodus, which includes important neotropical fish species, species-level delimitation usually results in poor phylogenetic resolution when using mitochondrial COI/cytB genes as barcoding markers because of low genetic variability and low species-level resolution. Thus, in this study, we developed an approach to design and validate new barcoding markers with high species-level resolution obtained from the D-loop region, using Prochilodus spp. as a model. For the new barcoding marker validation, the amplicon region was used to infer the phylogenetic relationships of Prochilodus spp. through three distinct methods: Bayesian inference (BI), Neighbor-Joining method (NJ), and Maximum Likelihood method (ML). The phylogenetic relationships of Prochilodus spp. revealed high resolution at species-level, nonoverlapping clades, and high branch support. The genetic distance results allied to two different clustering methods (Bayesian Poisson tree processes and automatic barcode gap discovery) revealed the existence of a barcoding gap, thus, validating the use of the barcoding markers designed in this study. The approach proposed here may, therefore, be expanded to other taxa to access and validate new barcoding markers with higher resolution at the species level.

Acid proteases from sábalo stomach mucosa were recovered using salting-out procedure. This single step produced an enzyme extract purified 1.8-fold over the crude extract with a recovery of 45.1% of its initial proteolytic activity. Sábalo proteases exhibited the highest activity at 45 °C-pH 2.0, showed pH stability between 2.0 and 5.0 and retained more than 70% of its activity after incubation at pH 7.0 for 2 h. Fish extract was unstable at temperatures greater than 45 °C. Its activity was inhibited by pepstatin A but not by PMSF, while EDTA and SDS showed partial inhibitory effects. Presence of CaCl2 and MgCl2 increased the proteolytic activity, while increasing concentrations of NaCl strongly decreased it. In addition, compared to the acid extraction method, the use of sábalo enzymatic extract increased 1.7 times the yield of collagen extraction.

Morphological and cytochemical studies of peripheral blood cells of fish have improved the understanding of their functions and cell types. The present study performed the Morphological and cytochemical analysis of the peripheral blood of Prochilodus lineatus, Characiform native to South America, which has been gaining space in local aquaculture and as a species introduced in Asia. Our analysis provided information on the morphological and cytochemical characteristics of the leukocytes, for the formulation of hypothesis about their role in the immune system of the species. It was found that Prochilodus lineatus has morphological and cytochemical features in common with other fish species, mainly of the Characiformes order. However, we detected the presence of heterophils and PAS positive granulocytes simultaneously with neutrophils. We also found that heterophils and PAS positive granulocytes are very similar, both morphologically and cytochemically.(AU)


O estudo da morfologia e da citoquímica das células do sangue periférico dos peixes tem sido eficaz para o entendimento de suas funções e dos tipos celulares. Este estudo realizou a análise morfológica e citoquímica do sangue periférico de Prochilodus lineatus, caracídeo nativo da América do Sul que vem ganhando espaço na aquicultura local e como espécie introduzida na Ásia. Essa análise forneceu informações sobre a morfologia e as características citoquímicas dos leucócitos, visando a hipóteses sobre suas funções. Verificou-se que estas são semelhantes em vários aspectos a outras espécies, principalmente da ordem Characiformes. No entanto, neste estudo detectou-se a presença dos heterofilos e da célula granulocítica especial, simultaneamente à presença dos neutrófilos. Ainda, foi verificado que os heterofilos e a célula granulocítica especial são muito semelhantes morfológica e citoquimicamente.(AU)

Although biotelemetry studies bring useful information, tagging is a highly invasive procedure. For this reason, we assess if intracoelomic tagging alters survivorship, weight gain and/or reproductive capacity of a neotropical migratory fish. In August 2016, 60 fish were equally and randomly distributed among 3 treatments: Control (anesthesia), sham surgery (anesthesia and surgery), and true surgery (anesthesia, surgery and tagging). Surveys for weight and tag expulsion were performed periodically from surgery through spawning (November/2016), with a final survey occurring in May/2017. Mortality was higher in true surgery (25% died) and for males. Twenty percent expelled tags, and initial weight loss followed by gain were verified for all treatments. Females of all treatments spawned, and fecundity and fertility were not different. Then, future studies should consider the losses due to death and tag expulsion when defining the sample number. We also encourage further investigations about differences in mortality between sex and weight variation, as well as any other factor that leads to increase in mortality. This is the first study that investigated tagging effects in reproduction of neotropical fishes, and since the methods and tags used are not species-specific, we infer that similar effects could occur in other species.(AU)


Embora estudos de biotelemetria gerem informações úteis, processos de marcação são invasivos. Por isso, avaliamos se a marcação intracelomática altera sobrevivência, ganho de peso e/ou capacidade reprodutiva de um peixe migrador neotropical. Em agosto de 2016, 60 peixes foram distribuídos igualmente e aleatoriamente entre os tratamentos controle (anestesia), cirurgia-falsa (anestesia e cirurgia) e cirurgia-verdadeira (anestesia, cirurgia e marcação). Vistorias para pesagem e observação de expulsão de marcas foram realizadas periodicamente da cirurgia até a desova (novembro/2016), com uma vistoria final em maio/2017. A mortalidade foi maior nos peixes marcados (25% morreram) e entre machos. Vinte por cento expulsaram as marcas e, para todos os tratamentos, foi observado perda de peso inicial com posterior recuperação. Fêmeas de todos os tratamentos desovaram; fecundidade e fertilidade não foram diferentes. Novos estudos devem considerar as perdas de marcas por mortalidade e rejeição na definição do número amostral. Também recomendamos estudos que investiguem a mortalidade diferencial entre sexos e variação de peso, bem como fatores causadores de mortalidade. Este é o primeiro estudo que investigou efeitos da marcação na reprodução de peixes neotropicais e, uma vez que métodos e marcas utilizadas não são espécie-específicos, inferimos que efeitos similares podem ocorrer em outras espécies.(AU)

In fish ovaries, postovulatory follicles (POFs) are key biomarkers of breeding and provide an interesting model for studying the relationship between autophagy and apoptosis. In this study, we investigated the immunohistochemical expression of autophagic and apoptotic proteins to improve the knowledge on the mechanisms regulating ovarian remodeling after spawning. Females from three neotropical fish species kept in captivity were submitted to hormonal induction. After ova stripping, ovarian sections were sampled daily until 5 days postspawning (dps). Similar events of POF regression were detected by histology, terminal transferase-mediated dUTP nick-end labeling (TUNEL), and electron microscopy in the three species: follicular cells hypertrophy, progressive disintegration of the basement membrane, gradual closing of the follicular lumen, theca thickening, and formation of large autophagic vacuoles preceding apoptosis of the follicular cells. Autophagic and apoptotic proteins were assessed by immunohistochemistry. Morphometric analysis of the immunolabeling revealed a more intense reaction for bcl-2 and beclin-1 (BECN1) in POFs at 0 to 1 dps and for bax at 2 to 3 dps (P < 0.001), the later period being the peak of apoptosis of the follicular cells. The immunostaining for cathepsin-D was more elevated until 2 to 3 dps and decreased significantly at 4 to 5 dps, when the POFs were in late stage of regression. Double labeling for BECN1 and caspase-3 indicated a shift in the relationship between autophagy and apoptosis at 2 to 3 dps, a critical period in determining the fate of follicular cells in POFs. Together, these results indicate that the bcl-2 family, BECN1, and cathepsin-D can be involved in the regulation of ovarian remodeling in teleost fish.

Abstract: We investigated spatial and temporal variations in the distribution of fish eggs and larvae in the Guareí River, a free-flowing tributary located in the upper section of the Jurumirim Reservoir, Paranapanema River, Brazil. Fish eggs and larvae were sampled ~ fortnightly from November 2017 to March 2018 (Cycle 1), and from November 2018 to March 2019 (Cycle 2), at three sites distributed along the entire course of the Guareí River. We collected 859 fish eggs and 1,340 larvae, comprising 19 taxa. We found reproductive activity of several fish species in the Guareí River, including long-distance migrants. However, there was no spatial differences in the assemblage structure and densities of fish eggs and larvae along the channel of Guareí River. We recorded temporal differences in larvae density only in Cycle 2. The consistent capture of fish eggs and larvae in two reproductive cycles is strong evidence that this river is a spawning site for a variety of fish species. Thus, we recommend maintaining its natural flow regime.


Resumo: Investigamos as variações espaciais e temporais na distribuição de ovos e larvas de peixes no rio Guareí, um afluente de fluxo livre localizado na parte superior do Reservatório de Jurumirim, bacia do rio Paranapanema, Brasil. Ovos e larvas de peixes foram amostrados ~ quinzenalmente de novembro de 2017 a março de 2018 (Ciclo 1), e de novembro de 2018 a março de 2019 (Ciclo 2), em três locais distribuídos ao longo de todo o curso do rio Guareí. Foram coletados 859 ovos de peixes e 1.340 larvas, compreendendo 19 táxons. Nós encontramos atividade reprodutiva de várias espécies no rio Guareí, incluindo espécies migradoras de longa distância. Contudo, não houve diferenças espaciais na estrutura das assembleias e nas densidades de ovos e larvas e ao longo do canal do rio Guareí. Diferenças temporais foram significativas apenas para densidade de larvas no Ciclo 2. A captura consistente de ovos e larvas de peixes em dois ciclos reprodutivos é uma forte evidência de que este rio é um local para a reprodução de várias espécies de peixes. Assim, recomendamos a manutenção do seu regime de fluxo natural.

RESUMEN Los Sistemas de Agro Acuicultura Integrada (SAAI) han sido estudiados como opción para la producción de pescado en comunidades con recursos limitados. Así mismo, el uso de perifiton se ha convertido recientemente en una alternativa viable por el aporte de alimento natural de bajo costo. El objetivo de este estudio fue evaluar el efecto del perifiton sobre el desempeño productivo del policultivo Piaractus sp. (cachama híbrida) y Prochilodus magdalenae (bocachico) en SAAI en la comunidad Indígena de Jimaín, Colombia. Se sembraron 1,5 alevinos de cachama híbrida y 1,5 bocachicos/m2, con peso promedio de 0,49 ± 0,14 g y 1,83 ±1,61 g respectivamente, en seis estaques en tierra de 90 m2 tres de los cuales contaron con varas de madera (3 varas/m2) para la fijación de perifiton. La duración del cultivo fue de 135 días. Los tratamientos (CS: con sustrato y SS: sin sustratos) recibieron una dieta suplementaria dos veces al día, calculada como porcentaje de la biomasa con ajuste quincenal. Se realizaron biometrías quincenales. Los datos productivos por especie y para el policultivo se evaluaron mediante ANOVA con un modelo lineal general (p < 0,05). Los datos de peso (g) y longitud estándar (cm) por especie se evaluaron mediante un modelo mixto de medidas repetidas. No se encontraron efectos del sustrato sobre los parámetros productivos analizados tanto para cada especie, como para el policultivo. Se presentó interacción significativa para el peso del bocachico el día 90 (CS: 74,7 y SS: 47,1). Según los resultados obtenidos, la utilización de perifiton y el manejo en policultivo de especies nativas requiere más estudios.


ABSTRACT Integrated agri-aquaculture systems (IAAS) have been studied as an option for fish production in communities with limited resources. Likewise, the use of periphyton has recently become a viable alternative for its contribution of low-cost natural food. The effect of periphyton use on the productive yield of a hybrid cachama and bocachico polyculture in IAAS, was studied in the Jimaín Indigenous community, Colombia. For this experiment, 1.5 hybrid cachama and 1.5 bocachico fingerlings/m2 were stocked with average weights of 0.49 ± 0.14 g and 1.83 ± 1.61 g, respectively, in six 90 m2 land ponds; three of these ponds had wooden dowels (three dowels/m2) for periphyton fixation. The growth evaluation lasted 135 days. The treatments (WS: with substrate and WOS: without substrates) received a supplementary diet, adjusted as a percentage of the biomass with biweekly adjustment, twice a day. Biometrics were performed every two weeks. The productive data by species and for the polyculture were evaluated using an ANOVA with a general linear model (p <0.05). Weight (g) and standard length (cm) data by species were evaluated using a mixed model of repeated measures. No substrate effects were found on the productive parameters analyzed for each species as well as for the polyculture. A significant interaction was observed for bocachico weight on day 90 (WS: 74.7 and WOS: 47.1). According to the obtained results, optimizing the use of periphyton and the management of native species polyculture requires further studies.

Early development from the egg fertilization to complete resorption of the yolk-sac is a critical period in the life cycle of teleost fish. Knowledge of this process provides essential parameters for aquaculture and identification of spawning sites in the wild. In the present study, a comparative morphological analysis of the oocyte surface as well as early development was performed in four commercially valuable species from the São Francisco River: Brycon orthotaenia, Leporinus obtusidens, Prochilodus argenteus, and Salminus franciscanus. Stripped oocytes, embryo, and yolk-sac larvae were analyzed by scanning electron microscopy (SEM) and histology. A set of 10 lectins was used for investigation of lectin-binding pattern in oocytes. In the four species, the outer layer of the zona radiata reacted to most lectins, indicating complex polysaccharides at the oocyte surface while no reactivity was detected in the inner zona radiata and yolk globules. Typical structural arrangements were recognized at the micropylar region by SEM. The four species showed nonadhesive eggs, short embryonic period (18-20 h at 24 ± 1°C), and poorly developed larvae at hatching. At 24 h posthatching (hph), larvae of the four species had neuromasts on the body surface. Rudimentary cement glands for larval attachment were identified on the cephalic region at 24 and 48 hph in B. orthotaenia and S. franciscanus, and following they were in regression. The time for whole yolk resorption varied among species from 48 to 120 hph, occurring earlier in S. franciscanus, followed by B. orthotaenia, P. argenteus, and L. obtusidens. The formation of the digestive tract and the mouth opening indicated initiation of exogenous feeding 24 h before complete resorption of the yolk. Together, our data indicate similarities in the early development among species that may be related to the life cycle strategies and phylogeny.
