RESUMO

Molecular methods were used to determine the generic placement of two species of Entomophthorales known only from resting spores. Historically, these species would belong in the form-genus Tarichium, but this classification provides no information about phylogenetic relationships. Using DNA from resting spores, Zoophthora independentia, infecting Tipula (Lunatipula) submaculata in New York State, is now described as a new species and Tarichium porteri, described in 1942, which infects Tipula (Triplicitipula) colei in Tennessee, is transferred to the genus Zoophthora. We have shown that use of molecular methods can assist with determination of the phylogenetic relations of specimens within the form-genus Tarichium for an already described species and a new species for which only resting spores are available.

RESUMO

The subgenus Vestiplex of the genus Tipula is one of the most species-rich groups in the family Tipulidae. Here, we present the first mitochondrial genome of the subgenus Vestiplex. The nearly complete mitochondrial genome of Tipula aestiva Savchenko, 1960 (Genbank accession number: OM287601) was 16083 bp in length and consisted of 13 PCGs, 22 tRNA and two rRNA genes. The phylogenetic the tree of family Tipulidae was reconstructed based on 13 PCGs sequences using the maximum likelihood method. The result strongly supported the monophyly of the family Tipulidae. The subgenus Vestiplex is indicated as the sister group of subgenera Pterelachisus and Formotipula.

The caucasica species group in the subgenus Lunatipula is redefined and now consists of five species native to the Caucasus. Tipula (L.) eleniya sp. nov. is described as new to science, and variations in the male terminalia in two populations are noted. Two subspecies (quadridentataquadridentata and quadridentatapaupera) are elevated to species rank. Detailed photo's complement the descriptions of all five species (caucasica, eleniya, paupera, quadridentata, talyshensis), and data on ecology and distribution patterns are included as well as identification keys to males and females. Tipula caucasica is recorded from the West Caucasus and Tipula quadridentata is recorded from Dagestan (Russia) for the first time. Parallel evolution is traced in the male terminalia of the new species and in several non caucasica species group of Palaearctic Lunatipula.

The genus Tipula Linnaeus, 1758 is a large group of crane flies with more than 2400 known species from 41 subgenera. In this study, we report the first complete mitochondrial (mt) genome sequence of the subgenus Tipula (Yamatotipula), which is a circular molecule of 15,668 bp with an AT content of 77.2%. The mt genome contains 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and a long non-coding region. Three conserved overlapping regions, 8 bp between tRNATrp and tRNACys , 7 bp between ATP8 and ATP6, and 7 bp between ND4 and ND4L, are found. Phylogenetic analysis reveals that the Tipulomorpha includes the family Trichoceridae and the Trichoceridae is sister-group to the remaining Tipulomorpha.

Species of Tipula (Sivatipula) biprocessa sp. n. from Guangxi, China is described and illustrated as new in the subgenus Tipula (Sivatipula) Alexander, 1964. Tipula (Sivatipula) parvauricula Alexander, 1941 is redescribed and illustrated based on additional morphological characters. Semen pump of this subgenus is discussed. A key to all described species in this group is compiled.

All available type material of Tipula stackelbergi Alexander, Tipula usuriensis Alexander and Tipula subpruinosa Mannheims were examined. Tipula (Yamatotipula) stackelbergistat. rev. is elevated from a subspecies of Tipula (Yamatotipula) pruinosa Wiedemann to a valid species. Two new synonyms are proposed: Tipula usuriensissyn. n. proved to be a junior synonym of. Tipula (Yamatotipula) pruinosa and Tipula subpruinosasyn. n. a junior synonym of Tipula (Yamatotipula) freyana Lackschewitz. Tipula (Yamatotipula) stackelbergi is redescribed, male and female terminalia of Tipula (Yamatotipula) pruinosa are illustrated and discussed. Female terminalia of Tipula (Yamatotipula) freyana are described and illustrated for the first time. A key to both sexes of Tipula (Yamatotipula) stackelbergi and Tipula (Yamatotipula) pruinosa, and a key to females of Tipula (Yamatotipula) chonsaniana, Tipula (Yamatotipula) freyana and Tipula (Yamatotipula) moesta are provided. Subspecies are not uncommon among crane flies, but their ranges and traits are poorly known. An interdisciplinary approach (genetics, ecology, taxonomy) is suggested if subspecific ranks are to be used in tipuloid systematics.
