Multimedia fugacity models are effective tools for studying the environmental behaviour and occurrence of contaminants of emerging concern (CECs) and assessing associated risks, especially when experimental data is limited. These models describe processes controlling chemical partitioning, transport, and reactions in environmental media using mathematical statements based on the concept of fugacity. To aid in identifying and prioritizing CECs for future local monitoring, we present here the application of a level III multimedia fugacity model assuming non-equilibrium between compartments and steady-state conditions. This model estimated predicted environmental concentrations (PECs), persistence, distribution, and transport of ten plant protection products (PPPs) in the Venice Lagoon, a complex coastal environment under high anthropogenic pressure. The model was evaluated through uncertainty and sensitivity analysis using the Monte Carlo approach and by comparing PECs with PPP concentrations measured during four sampling campaigns. Results showed good agreement with field data, with the highest concentrations in water and sediments estimated for glyphosate, followed by imidacloprid, metaflumizone, and triallate. The model indicated accumulation of all investigated PPPs in sediments. For most chemicals, advection outflow and degradation in the water column were the main removal mechanisms, while volatilization was significant only for oxadiazon and triallate. Sensitivity and uncertainty analysis revealed that degradation rates, organic carbon/water partitioning coefficients (KOC), and parameters describing air-water interactions had the strongest influence on the model's results, followed by inputs accounting for sediment sinking and resuspension. The lack of data on PPP degradation in brackish waters accounted for most of the uncertainty in model results. This work shows how a relatively simple multimedia model can offer new insights into the environmental behaviour of PPPs in a complex transitional waterbody such as the Venice lagoon, providing useful data for the identification of the CECs to be prioritised in future local monitoring efforts.

BACKGROUND: The reuse of dredged sediments in ports and lagoons is a big issue as it should not affect the quality and the equilibrium of ecosystems. In the lagoon of Venice, sediment management is of crucial importance as sediments are often utilized to built-up structures necessary to limit erosion. However, the impact of sediment reuse on organisms inhabiting this delicate area is poorly known. The Manila clam is a filter-feeding species of high economic and ecological value for the Venice lagoon experiencing a drastic decline in the last decades. In order to define the molecular mechanisms behind sediment toxicity, we exposed clams to sediments sampled from different sites within one of the Venice lagoon navigable canals close to the industrial area. Moreover, we investigated the impacts of dredged sediments on clam's microbial communities. RESULTS: Concentrations of the trace elements and organic chemicals showed increasing concentrations from the city of Venice to sites close to the industrial area of Porto Marghera, where PCDD/Fs and PCBs concentrations were up to 120 times higher than the southern lagoon. While bioaccumulation of organic contaminants of industrial origin reflected sediments' chemical concentrations, metal bioaccumulation was not consistent with metal concentrations measured in sediments probably due to the activation of ABC transporters. At the transcriptional level, we found a persistent activation of the mTORC1 signalling pathway, which is central in the coordination of cellular responses to chemical stress. Microbiota characterization showed the over-representation of potential opportunistic pathogens following exposure to the most contaminated sediments, leading to host immune response activation. Despite the limited acquisition of new microbial species from sediments, the latter play an important role in shaping Manila clam microbial communities. CONCLUSIONS: Sediment management in the Venice lagoon will increase in the next years to maintain and create new canals as well as to allow the operation of the new mobile gates at the three Venice lagoon inlets. Our data reveal important transcriptional and microbial changes of Manila clams after exposure to sediments, therefore reuse of dredged sediments represents a potential risk for the conservation of this species and possibly for other organisms inhabiting the Venice lagoon.

Butyltin (TBT, DBT, and MBT) effects on molluscs, especially endocrine disruption in bivalves and gastropods, have been widely investigated. Imposex, the superimposition of male characters onto female gonochoristic Caenogastropods, is the most studied biological effect of TBT. TBT compounds are among the priority hazardous substances within Directives 2000/60/EC (WFD) and 2008/105/EC. The Environmental Quality Standards (EQSs) set by the WFD for TBT are quite difficult to quantify by means of chemical analysis, without the use of expensive and high performance methods. Assuming that EQSs set for TBT were derived from evidence of imposex development at very low concentrations, this specific biomarker could be used as an indirect measure of assessing levels of bioavailable BTs. Therefore, this study aims to validate the use of imposex development as an investigative tool to monitor the bioavailable fraction of BTs within the WFD, by comparing imposex levels and BT concentrations in Nassarius nitidus from the Venice Lagoon. BT concentrations and imposex levels in N. nitidus, collected in 2013, had decreased when compared to previous studies in the same area. Both VDSI and RPLI correlated positively with BT body burden in females, confirming that imposex is a valid tool to monitor bioavailable BTs. However, TBT is still a matter of concern in the Venice Lagoon, as TBT concentrations were still higher than its degradation products suggesting recent fresh TBT inputs in the studied area. To propose imposex levels as an indicator of the impact of BTs within the WFD, classification class boundaries and Ecological Quality Ratios were introduced. As a preliminary attempt, imposex levels were also compared to the OSPAR Commission EcoQOs which linked imposex levels in Nassarius reticulatus with TBT concentrations in water. Based on this comparison the degree of imposex development in the Venice Lagoon suggested that TBT concentrations in water should be over the EQS-AA concentration and, at one site, also over the EQS-MAC. From all the results obtained in this work, it appears that imposex evaluation in N. reticulatus could give information about the ecological status regarding BT compounds under the WFD and also be used to monitor their effects and support chemical analyses until more sensitive methods become available.

The aim of this study was to compare imposex and butyltin compounds (BTs) data, collected before and after the organotin ban in 2008, in order to assess temporal and spatial variation of the phenomenon, the decline of BT contamination, and the effects on Hexaplex trunculus population in the coastal area of the northern Adriatic Sea, close to the Venice Lagoon. Both in marine and in lagoon sites, the results obtained in 2013-2015 showed a significant decline in the incidence of imposex in respect to those from the 2002 survey. In 2002, lagoon samples exhibited Relative Penis Size Index (RPSI) higher than marine samples, whereas no differences were detected in the recent survey, when all RPSI values were below 0.6%. Vas Deference Sequence Index (VDSI) mean values were over 4 before the ban introduction and below this value after that, indicating more critical conditions for gastropod population in 2002 rather than in 2013-15. Percentage of sterile females was up to 69% in 2002, whilst in the more recent survey no sterile female was found. Range of BT concentrations in gastropods decreased from 252 to 579 to 16-31ng∑BT/g d.w. BT body burdens varied according to a gender dependant pattern, with higher concentrations observed in females than in males. A first attempt to propose a classification based on BT impact on H. trunculus, according to the Water Framework Directive, revealed that most sites were in Bad ecological status before the ban and attained a Poor/Moderate status after that.

The study focused on selected trace elements (As, Cd, Cr, Hg, Ni, Pb) monitored in surface waters of the Venice Lagoon catchment basin (North East Italy) over the period 2000-2015. The monitoring was undertaken to verify the achievement of the quality objectives set by the European and national legislations. The available results have been analyzed to evaluate the chemical status of water bodies. The limit of quantification (LOQ) of the applied analytic techniques appears critical for the adequate water monitoring; for some parameters, the percentage of not visible values due to non-satisfactory LOQ was higher in the beginning of the period; the subsequent improvement of LOQ allowed assessing the respect of environmental quality standards (EQSs). The study analyzes time trends in single stations and the differences between detected concentrations in the considered stations. Moreover, maximum concentrations and water flows have been considered to understand the potential correlation. Cumulated frequency curves for the most critical parameters have been built to identify situation of potential overtaking of the EQSs in force. The most polluted sampling stations of the drainage basin for the six trace elements were found in Cuori and Fiumazzo rivers. Although LOQs changed over time, the recorded trends show a quality improvement and a good compliance with respect to EQSs set by European legislation, while considering EQSs set by local special legislation, the objectives are not yet satisfied. Arsenic is ubiquitous; thus, it can be supposed to be originated as a background environmental concentration, while nickel appears of industrial origin according to its point and local presence.

Microplastics (MPs) and nanoplastics (NPs) both represent significant concerns in environmental sciences. This paper aims to develop a convenient and efficient methodology for the detection and measurement of MPs and nanoparticles from surface seawater and to apply it to the water samples collected from the UNESCO site of Venice and its lagoon, more precisely in the Venice-Lido Port Inlet, Grand Canal under Rialto Bridge, and Saint Marc basin. In this study, MPs were analyzed through optical microscopy for their relative abundance and characterized based on their color, shape, and size classes, while the concentration and the mean of nanoparticles were estimated via the Nanoparticle Tracking Analysis technique. Bulk seawater sampling, combined with filtration through a cascade of stainless-steel sieves and subsequent digestion, facilitates the detection of MPs of relatively small sizes (size classes distribution: >1 mm, 1000-250 µm, 250-125 µm, 125-90 µm, and 90-32 µm), similar to the size of MPs ingested by marine invertebrates and fishes. A protocol for minimizing interference from non-plastic nanoparticles through evaporation, digestion, and filtration processes was proposed to enrich the sample for NPs. The findings contribute to the understanding of the extent and characteristics of MPs and nanoparticle pollution in the Venice Lagoon seawater, highlighting the potential environmental risks associated with these pollutants and the need for coordinated approaches to mitigate them. Aquaculture productivity in coastal lagoons is endangered by a complex interplay of anthropogenic and environmental factors, amplified by the effects of climate change in these sensitive areas. To reach a more comprehensive assessment of farming sites quality, a quantitative Weight of Evidence approach (QWoE) is applied for the first time to data collected at four Manila clam (R. philippinarum) farming sites in the Venice lagoon (Italy). This included sediment quality, chemical bioaccumulation, and biological responses. Results revealed a greater hazard for sites closer to the open sea. In these areas, the combination of sediment characteristics and a higher frequency of salinity and temperature stress could explain the alterations measured at a transcriptional and biomarker level. The findings demonstrate that a QWoE approach that integrates multiple sources of evidence should also include physicochemical conditions in order to better understand the impacts of human activities and other stressors on clam aquaculture productivity.

MOSE is a system of mobile gates engineered to temporarily isolate the Venice Lagoon from the Adriatic Sea and to protect the city from flooding during extreme high tides. Within the framework of the Venezia2021 program, we conducted two enclosure experiments in July 2019 (over 48 h) and October 2020 (over 28 h) by means of 18 mesocosms, in order to simulate the structural alterations that microphytobenthos (MPB) assemblages might encounter when the MOSE system is operational. The reduced hydrodynamics inside the mesocosms favored the deposition of organic matter and the sinking of cells from the water column towards the sediment. Consequently, MPB abundances increased over the course of both experiments and significant changes in the taxonomic composition of the community were recorded. Species richness increased in summer while it slightly decreased in autumn, this latter due to the increase in relative abundances of taxa favored by high organic loads and fine grain size. By coupling classical taxonomy with 18S rRNA gene metabarcoding we were able to obtain a comprehensive view of the whole community potential, highlighting the complementarity of these two approaches in ecological studies. Changes in the structure of MPB could affect sediment biostabilization, water turbidity and lagoon primary production.

Extreme sea levels (ESLs) affect coastal ecosystems worldwide. Protection and adaptation strategies rely on the characterization of the extreme's occurrence probability in the future. However, knowledge on the occurrence rate and duration of ESLs is also needed to properly characterize the associated future risk. In this paper, we focus on the Venice lagoon, where a system of storm surge barriers can disconnect the lagoon from the sea to mitigate ESLs. Using long-term (96 years) sea level records, we model occurrence rate and duration of ESLs while accounting explicitly for seasonality and mean sea level rise. While historically ESLs occurred in the winter season, we project a significant increase (up to 10-fold with a mean sea level increase of +100 cm) of the occurrence rate also in the summer season, when disconnections from the sea can have profound impacts on the lagoon's ecosystem. We also predict an increase in ESLs durations up to 200 h, leading to longer disconnections of the lagoon from the sea in the future. Therefore, several adaptation strategies will be needed to limit the adverse effects of storm surge barriers on the lagoon ecosystem.

In recent years recurrent bivalve mass mortalities considerably increased around the world, causing the collapse of natural and farmed populations. Venice Lagoon has historically represented one of the major production areas of the Manila clam Ruditapes philippinarum in Europe. However, in the last 20 years a 75 % decrease in the annual production has been experienced. While climate change and anthropogenic interventions may have played a key role in natural and farmed stocks reductions, no studies investigated at multiple levels the environmental stressors affecting farmed Manila clam to date. In this work we carried out a long-term monitoring campaign on Manila clam reared in four farming sites located at different distances from the southern Venice Lagoon inlet, integrating (meta)genomic approaches (i.e. RNA-seq; microbiota characterization), biometric measurements and chemical-physical parameters. Our study allowed to characterize the molecular mechanisms adopted by this species to cope with the different environmental conditions characterizing farming sites and to propose hypotheses to explain mortality events observed in recent years. Among the most important findings, the disruption of clam's immune response, the spread of Vibrio spp., and the up-regulation of molecular pathways involved in xenobiotic metabolism suggested major environmental stressors affecting clams farmed in sites placed close to Chioggia's inlet, where highest mortality was also observed. Overall, our study provides knowledge-based tools for managing Manila clam farming on-growing areas. In addition, the collected data is a snapshot of the time immediately before the commissioning of MoSE, a system of mobile barriers aimed at protecting Venice from high tides, and will represent a baseline for future studies on the effects of MoSE on clams farming and more in general on the ecology of the Venice Lagoon.

This study investigated the occurrence of selected metals and metalloids (As, Cd, Cr, Cu, Hg, Ni, Pb, Zn) in radio-dated sediment cores from a coastal lagoon (Lagoon of Venice, Italy) by critically reviewing and grouping available data. Pre-industrial concentrations (pICs, estimated for the period before the early 1900s) for the Venice lagoon are identified according to the 2σ statistical procedure (2σ-pICs), i.e., the upper bound values of the dataset distribution. Results show the following 2σ-pICs (µg/g, d.w.): As 15,9; Cd 0,6; Cr 38,0; Cu 18,1; Hg 1,1; Ni 32,9; Pb 29,4; Zn 94,5. Most of the estimated 2σ-pICs are comparable to previously assessed background values. In the case of Hg, on the contrary, 2σ-pIC is remarkably higher than background values, reflecting a significant anthropogenic contribution also in the pre-industrial period. The results of this work may support the evaluation of the temporal evolution of metal concentrations in sediments of the Venice lagoon. Results are compared with background concentrations (BC) observed in previous studies conducted in the lagoon and in other areas of the Adriatic Sea, as well as with benchmarks set in Italy for sediment assessment.

Lagoon water, suspended particulate matter, and sediment samples from seven sites at Lagoon of Venice were collected from 2019 to 2021 in order to study the presence of the herbicide glyphosate (N-(phosphonomethyl)glycine), among the most widely used agricultural chemicals worldwide, but its occurrence in lagoon water environment has not been deeply investigated. The sites were selected considering a supposed diversity of inputs and of pollution levels. An analytical method based on ion chromatography coupled with tandem mass spectrometry was optimized and validated for lagoon water, marine particulate matter, and sediment samples. Maximum concentrations of glyphosate were 260 and 7 ng L-1 for lagoon water and suspended particulate matter, respectively, and 15 ng g-1 for sediment, with some spatial and temporal fluctuations. Our results demonstrate that glyphosate content in the Venice Lagoon mainly depends on external forcing from river inlets and agricultural lagoon activities.

Amyl salicylate (AS) is a fragrance massively used as a personal care product and following the discharged in wastewaters may end up in the aquatic environment representing a potential threat for the ecosystem and living organisms. AS was recently detected in water of the Venice Lagoon, a vulnerable area continuously subjected to the income of anthropogenic chemicals. The lagoon is a relevant area for mollusc farming, including the Mediterranean mussels (Mytilus galloprovincialis) having an important economic and ecological role. Despite high levels of AS occurred in water of the Lagoon of Venice, no studies investigated the possible consequences of AS exposures on species inhabiting this ecosystem to date. For the first time, we applied a multidisciplinary approach to investigate the potential effects of the fragrance AS on Mediterranean mussels. To reach such a goal, bioaccumulation, cellular, biochemical, and molecular analyses (RNA-seq and microbiota characterization) were measured in mussels treated for 7 and 14 days with different AS Venice lagoon environmental levels (0.1 and 0.5 µg L-1). Despite chemical investigations suggested low AS bioaccumulation capability, cellular and molecular analyses highlighted the disruption of several key cellular processes after the prolonged exposures to the high AS concentration. Among them, potential immunotoxicity and changes in transcriptional regulation of pathways involved in energy metabolism, stress response, apoptosis and cell death regulations have been observed. Conversely, exposure to the low AS concentration demonstrated weak transcriptional changes and transient increased representation of opportunistic pathogens, as Arcobacter genus and Vibrio aestuarianus. Summarizing, this study provides the first overview on the effects of AS on one of the most widely farmed mollusk species.

Transitional coastal zones are subject to high degrees of temporal fluctuation in environmental conditions, with these patterns varying in space. Gaining an in depth understanding of how sessile organisms cope with and respond to such environmental changes at multiple scales is needed to i) advance fundamental knowledge, ii) predict how organisms may react to stressors and iii) support the management of halieutic resources in transitional coastal areas. We addressed this question using mussels (Mytilus galloprovincialis) as model system. Valve-gaping sensor were deployed at multiple sites within the southern Venice Lagoon over a period of 6 months, to investigate the existence of periodicity in valve-gaping and its relationship with environmental variables, such as temperature and chlorophyll-a. Gaping behaviour was found to have periodic rhythms, of ~12 h and ~ 24 h, which were most pronounced in the inner part of lagoon part and were strongest during summer months. In autumn, the dual periodicity became weaker and mostly the 12 h remained. Gaping was closely linked with tide, but the relationship in terms of phasing varied upon location. Surprisingly, no clear direct relationships were found with chlorophyll-a, but food delivery may be mediated by tide itself. The results highlight the heterogeneity of behaviour and the endogenic nature of circadian rhythms in space and time. These findings have important implications for management of transitional areas where tidal alteration may have impacts on key behaviours, and emphasize the importance of characterizing their rhythms before using these as stress indicator. Moreover, the described tidal relationships should be included in growth models of bivalves in these systems.

The paper reports the results obtained after 4 years of aquatic angiosperm transplants in areas of the Venice Lagoon (North Adriatic Sea, Mediterranean) where meadows almost disappeared due to eutrophication, pollution and overexploitation of clam resources. The project LIFE12 NAT/IT/000331-SeResto, funded by the European Union, allowed to recolonize the Habitat 1150* (coastal lagoons) in the northernmost part of the lagoon, by extensive manual transplants of small sods or single rhizomes of Zostera marina, Zostera noltei, Ruppia cirrhosa and, in some stations also of Cymodocea nodosa. Over the 4 years of the project more than 75,000 rhizomes were transplanted in 35 stations with the support of local stakeholders (fishermen, hunters and sport clubs). Plants took root in 32 stations forming extensive meadows on a surface of approx. 10 km2 even if some failures were recorded in areas affected by outflows of freshwater rich in nutrients and suspended particulate matter. The rapid recovery of the ecological status of the involved areas was the result of this meadow restoration, which was in compliance with Water Framework Directive (WFD 2000/60/EC) objectives. Moreover, the monitoring of environmental parameters in the water column and in surface sediments allowed to identify the best conditions for successful transplants. Small, widespread interventions and the participation of local stakeholders in the environmental recovery, make this action economically cheap and easily transposable in other similar environments.

In this study, recent and aged inputs of five classes of organic contaminants (i.e. PCBs, OCPs, PCDD/Fs, PAHs, and n-Alkanes) were evaluated in eight deep sediment cores of the Venice Lagoon, collected along the path of a new waterway whose excavation is under evaluation by local authorities, to assess the environmental quality status of the area. Diagnostic indices were calculated for identifying pollutant distribution patterns and their major emission sources, whose relative contribution was quantified by a Positive Matrix Factorization source apportionment model. Sedimentary depth profiles highlighted higher contamination in the top layer, mainly related to ship traffic combustion and vehicular/industrial emissions from the mainland. Nevertheless, a significant level of pollution has been detected also in the deeper layers, probably due to the transport of particulate matter through the aquifers underlying the lagoon seabed. The results underlined the threat posed by the possible resuspension of pollutants in the water column during contaminated sediment dredging.

To evaluate the hazard assessment of dredging and disposal of sediments, an in-depth chemical and toxicological characterization of sediments was carried out in Venice Lagoon. The bioaccessible and mobile concentration of trace elements in superficial and deep sediments (in the Holocene-Pleistocene limit) can deeply affect the quality of the aquatic ecosystem. Geospeciation and total concentration of trace elements in sediment cores were assessed and several toxicological tests were carried out. The indices of contamination (e.g., geochemical accumulation index, Igeo, pollution load index, PLI) and of speciation (individual contamination factor, ICF and global contamination factor, GCF) were evaluated. Factor Analysis (FA) was performed to explore possible significant correlations between toxicity data and sediment chemistry. Results underlined that the total concentration of trace elements cannot adequately assess the hazard, while bioavailability, mobility, and toxicity of trace elements allow a thorough evaluation of the environmental threats. The integration of results from chemical characterization ex ante and toxicity assays will provide for a better sustainable handling and management of sediments.

Developing reliable tools to trace food origin represents a major goal for producers and control authorities. Here, we test the hypothesis whether NGS-generated data could provide a reliable tool to ensure seafood traceability. As a test case, we used the Manila clam Ruditapes philippinarum, a bivalve mollusk of high commercial interest with worldwide distribution, collected in the Venice lagoon sites subjected to prohibition of clam harvesting because of chemical contamination as well as in authorized clam harvesting areas. The results obtained demonstrated that the geographic origin of Manila clam may be more accurately determined basing on microbiome data than single nucleotide polymorphisms. In particular, combining microbiome data with machine-learning techniques, we provide the experimental evidence that it is possible to trace the clam place of origin at high spatial resolution. Considering its low cost and portability, NGS-analysis of microbiome data might represent a cost-effective, high-resolution tool for reliable food traceability.

This data article includes the dataset of wind setup in the Venice lagoon computed by means of a 2-D hydrodynamic model. The capability of the model to reproduce the hydrodynamic regime of the lagoon has been extensively investigated, with particular attention to the calibration of the wind shear stress at the water surface, in order to precisely characterize the contribute of wind setup on the water level estimation inside the lagoon. We analyze the wind setup induced considering all the reliable wind speeds (with step of 1 m/s) and wind directions (with step of 30°) potentially blowing over the Venice lagoon, comparing the results obtained considering the present not-regulated configuration of the lagoon (pre-Mo.S.E. scenario) to the regulated configuration (post-Mo.S.E. scenario), which refers to the hydrodynamic regime when the Mo.S.E. movable barriers will be operational. The analysis shows that the wind setup significantly increases when the gates at the three inlets of the Venice lagoon are regulated, up to exceeding four times the pre-Mo.S.E. scenario. We deem this result is of paramount importance for the management of the Mo.S.E. barriers and for the definition of their operating strategy aiming at preventing the flooding at all the urban settlements of the lagoon.

The representation of the temporal dynamics of ecosystem services (ES) is a crucial research frontier in the field of ES modeling. In fact, most current ES models focus on static ES assessments, that need to be repeated with different inputs per time step to explore potential changes in ES. Here, we present a new approach for the dynamic modeling of multiple ES, based on the Petri Net modeling framework. The key features are: (i) multiple ES are modeled together as a single network, using a social-ecological systems (SES) perspective; (ii) the model accounts for the interactions occurring among ES, by distinguishing between the ES whose provision is mediated by some type of human input, which can produce some side-effects on the system, and those that are generated directly through ecosystem functions and do not generate side-effects; (iii) the model can reproduce the effects of changing drivers on the elements of the SES. These features allow to use the model to explore how ES can evolve over time under different "what-if" scenarios. The importance of considering the ES interactions is tested, showing that failing to include them in the model remarkably affects the results. Due to its complexity, the model should be used as an exploratory tool, focusing on the analysis of the general trends of multiple ES provision, rather than on the generation of quantitative projections. A first conceptual application to the Venice lagoon, Italy, is presented, in which the trends of 13 different ES are simulated. This application shows the potential of the model in exploring the development produced by climate change and socio-economic pressures, and the effects of a set of possible management actions. This modeling approach can contribute to generate new perspectives on the dynamic modeling of multiple ES and on the integrated management of SES.
