RESUMO

Pest management of emerging pests can be challenging because very little fundamental knowledge is available to inform management strategies. One such pest, the red-headed flea beetle Systena frontalis (Fabricius) (Coleoptera: Chrysomelidae), is increasingly being identified as a pest of concern in cranberries Vaccinium macrocarpon Aiton (Ericales: Ericaceae). To improve our understanding of this pest and to develop more targeted management programs, we conducted field and laboratory studies to characterize the development, seasonal emergence patterns, and density-dependent plant injury. We found that significantly more flea beetle eggs hatched when exposed to sustained cold treatment between 0 and 5°C for 15 wk than at warmer temperatures, and for shorter and longer cold-period durations. The adults emerged sporadically over the summer, were patchily distributed, fed on both fruit and foliage, and preferentially fed on new plant growth. Using soil cores, we found eggs and larvae located relatively deep (>30 cm) in the soil. These patterns indicate that S. frontalis likely overwinters as eggs, and that targeting the larval stage may be the most effective management approach. Despite the cryptic nature of the larvae, continuing to improve our understanding of this life stage will be critical to optimizing control strategies.

Populations of Schizotetranychus brevisetosus Ehara (Acari: Tetranychidae), which live on the evergreen oak (Quercus glauca), survive the coldest months as either adult females or winter eggs. Adult females comprise the majority of the population in early November and oviposit from late November to early March. Most winter eggs hatch by late March, and adults of the next generation emerge in April. This species is considered an egg-diapausing species, but the environmental cues that regulate female reproductive arrest and resumption are mostly unknown. We investigated the photoperiodic responses of autumn reproductive arrest in 10 populations collected from different elevations in Shikoku, Japan. All populations showed long-day responses to critical daylength (CDL) around 12.2 h light (12.2L) at 20 °C, though there was no linear relationship between CDL and altitude. This result explains the steep decline in the proportion of summer eggs in November. Nonreproductive females developed under 10L at 20 °C commenced oviposition 14.3-20.6 days after transferring to 15L. This long pre-oviposition period explains the reduction in eggs before winter reproduction and suggests shallow adult diapause. Eggs thus obtained hatched in 12.9-15.3 days, similarly to summer eggs. Therefore, egg diapause in S. brevisetosus is much shallower than in species on deciduous hosts, which lay their winter eggs in early autumn to hatch at leaf flush in spring. The reproductive arrest and short hatching period may be an adaptation allowing egg-laying in midwinter, when predation pressure is low.

Little research has been done on egg diapause and the embryonic development of water mites. The aim of this study was to check the impact of temperature and periods of light on hatching of larvae of Eylais extendens. Three batches of eggs which were spawned on 30 July were placed at one of three temperatures (4, 10 and 20 °C) and two periods of light (7 and 14 h per day). Egg hatching (both, percentage of hatched larvae and rate of hatching) was found to differ between 4 versus 10 °C and between 4 versus 20 °C, but not between 10 versus 20 °C. The periods of light had no influence on hatching. This synchronization of hatching, enabling the eggs to emerge from diapause in the spring, could be considered an evolutionary adaptation aimed at postponing hatching of late-spawned eggs until a time allowing for completion of the full development cycle, including the parasitic larval stage.

Bombyxin (BBX) is an insulin-like peptide exists in the silkworm Bombyx mori. Our previous studies on the effects of inhibiting BBX-B8 expression found that BBX-B8 is important for the development of organ, reproduction and trehalose metabolism in the silkworms. In this paper, we investigated the expression profile of the BBX-B8 gene and effect of BBX-B8 overexpression on the development, body weight, silk protein synthesis and egg diapause of B. mori to further understand BBX-B8 functions. BBX-B8 gene expression could be detected in the brains, midguts, anterior silkglands, ovaries, testes, fat bodies, hemolymph, malpighian tubules and embryos by RT-PCR, however it was mainly expressed in the brain. Western blots showed that the change in BBX-B8 expression was not obvious in the brain of 1- to 4-day-old larvae of fifth instar silkworms, but expression increased substantially at 5- to 6-day-old larvae of fifth instar silkworms. Transgenic silkworms overexpressing BBX-B8 were obtained by introducing non-transposon transgenic vector pIZT-B8 containing a BBX-B8 gene driven by Orgyia pseudotsugata nucleopolyhedrovirus IE2 promoter into the genome. Development duration of the transgenic silkworms was delayed by 2.5-3.5 days. Cocoon shell weight of transgenic silkworms was reduced by 4.79 % in females and 7.44 % in males, pupal weight of transgenic silkworms was reduced 6.75 % in females and 13.83 % in males compared to non-transgenic silkworms, and 5.56-14.29 % of transgenic moths laid nondiapausing eggs. All results indicated that BBX-B8 plays an important role in the development, silk protein synthesis and egg diapause of silkworm.

Diapause is an important biological characteristic for many insect species to adapt to adverse environmental conditions and maintain the continuity of the race. Compared with the traditional hydrochloric acid or/and cold storage treatment methods, the artificial corona incubation technology of silkworm (Bombyx mori) eggs has many advantages including, the absence of pollution, easy operation and safety. However, this technology has not yet been applied in sericulture. In this study, we developed a novel artificial corona instrument to successfully disrupt the diapause of newly laid and refrigerated eggs from various Chinese and Japanese lineage silkworm strains. Subsequently, we invented a very early corona treatment (VECT) strategy to prevent the diapause of newly laid silkworm eggs within 4 h of oviposition. The hatching rates of the larvae were more than 95% in all diapause silkworm strains, which was comparable to the effect of the traditional HCl treatment strategy. In addition, we developed a combination strategy of VECT and pre-blastoderm microinjection and successfully created transgenic silkworms in various diapause strains. The results of the current study can aid in improving the corona artificial incubation technology and promote its application in sericulture.

Diapause induction in multivoltine insects is an ecophysiological event that is generally triggered by seasonal cues such as photoperiod and temperature. The rice leaf bug, Trigonotylus caelestialium (Kirkaldy), feeds on various Poaceae grasses and produces several generations a year. Previous studies have shown that adults produce diapause and nondiapause eggs under short-day and long-day conditions, respectively. However, there is a distinct mid-summer peak in diapause incidence before an autumnal increase in diapause incidence in the field, which cannot be explained by the laboratory results. The present study was performed to examine the environmental factors affecting the diapause incidence in mid-summer and the adaptive significance of this phenomenon. Seasonal trends in diapause incidence differed significantly among three sites located 150-400 m apart from each other and with different host plants. The suitability of host plants differs depending on species and seasonally. Therefore, the microhabitat difference in diapause trend is believed to be due to the difference in host plants. When field-collected female adults laying diapause eggs in late June were fed a seasonally deteriorating host (the orange foxtail, Alopecurus aequalis Sobol. [Poales: Poaceae]), they kept laying diapause eggs, whereas when fed a suitable host (the wheat, Triticum aestivum L. [Poales: Poaceae]) for 5 d, they changed oviposition mode to lay nondiapause eggs. These results indicate that host-plant suitability affects the oviposition mode of T. caelestialium. Diapause-egg oviposition in mid-summer in T. caelestialium has adaptive significance as a bet-hedging strategy against unpredictable dietary conditions.

The phenological response is among the most important traits affecting a species' sensitivity to climate. In insects, strongly seasonal environments often select for a univoltine life cycle such that one seasonal extreme is avoided as an inactive stage. Through understanding the underlying mechanisms for univoltinism, and the consequences of its failure, we can better predict insect responses to climate change. Here we combine empirical data and simulation studies to investigate the consequences of an unusual diapause mechanism in a parthenogenetic matchstick grasshopper, Warramaba virgo, from arid southern Australia. Our field body temperature measurements indicate that this species is a thermoconformer and our laboratory studies of the thermal response of feeding rate imply strong constraints on winter activity. However, the species exhibits no obligate winter diapause, and eggs can develop in 1 month under constant temperatures approximating the mean soil temperature at the time of oviposition (summer). We show that diurnal temperature cycles exceeding a peak of 36 °C inhibit egg development in summer, and that this is sufficient to prevent autumnal hatching of eggs. Development is also strongly retarded below 24 °C. Microclimate-driven simulation studies of egg development show that these thermal responses provide robust maintenance of a univoltine life cycle, thereby resulting in survival of heat stress as an egg (due to limited developmental state) and avoidance of cold stress as a nymph and adult (due to overwintering in the soil as an egg).

Mode of inheritance of hatch traits in Lymantria dispar L. was determined by crossing populations nearly fixed for the phenotypic extremes. The nondiapausing phenotype was inherited via a single recessive gene and the phenotype with reduced low temperature exposure requirements before hatch was inherited via a single dominant gene. There was no evidence for sex-linkage or cytoplasmic effects with either gene. Eggs from 43 geographic populations were evaluated for hatch characteristics after being held for 60 d at 5°C followed by incubation at 25°C. There was considerable variation both within and among the populations in the proportion able to hatch, time to first hatch, and average time to hatch. Egg masses with reduced requirement for low temperatures before the eggs were ready to hatch were present in all subspecies of L. dispar and the phenotype was not fixed in most populations. The populations clustered into three distinct groups, and climatic variables were found to be rough predictors of those groups. Variation in hatch phenotypes between populations is likely an adaptation to local climate and within a population provides a bet-hedging strategy to ensure that at least some hatch synchronizes with host leaf-out. Continued vigilance to prevent movement of populations both within and between countries is warranted, because some of the alleles that confer nondiapause or reduced low temperature requirements before egg hatch are not present in all populations and their introduction would increase variation in egg hatch within a population.

Galeruca daurica (Joannis) is a new pest on the grasslands of Inner Mongolia, China. It is univoltine and overwinters in the egg stage. Larvae and adults feed on the foliage of Allium plants. To assess the requirements to terminate egg diapause and subsequent effects on post-diapause development rate, eggs were held at different temperature regimes. Exposure to low temperatures was required to terminate egg diapause. Prolonged exposure (2 mo vs 1 mo) to 5°C and outside ambient conditions (mean temperature: 10.5°C; range: -7.1-21.6°C) enhanced the termination of egg diapause. Prolonged exposure also reduced the time to egg hatch; e.g., eggs held for 2 mo versus 1 mo at 5°C developed more quickly when subsequently placed at warmer temperatures. Egg hatch was observed at 17, 21, 25, and 29°C, but not at 15°C. Regression analysis identified 16.2°C as the minimum temperature for post-diapause development. The temperature requirement to complete embryonic development (from diapause termination to egg hatch) was calculated to be 103.1 to 140.9 degree-days.
