RESUMO

Punta Lara is located in the Río de la Plata estuary near industrial areas contaminated mainly by organic pollutants. In this work, the responses and status of hepatic biomarkers were studied in juvenile carp (Cyprinus carpio) by means of a 21-day field exposure in cages and collection of juvenile native fish (Leporinus obtusidens) at Punta Lara. The analyzed hepatic biomarkers were: enzymatic activity of glutathione-S-transferase (GST), catalase (CAT) and superoxide dismutase (SOD), lipid peroxidation level using the thiobarbituric acid reaction (TBARS), and CYP1A protein expression, condition factor (CF) and liver somatic (LSI) index. Taking into account oxidative stress responses, SOD activity was increased in both species, while CAT was increased in C. carpio and decreased in L. obtusidens; TBARS levels indicated that oxidative damage was possibly exerted only in L. obtusidens. Biotransformation responses mediated by CYP1A were observed in both species, while GST activity was induced mainly in carps. Considering morphometric indices, CF and LSI were significantly increased in carps while CF decreased in native species. The anthropogenic pollution detected in this study in Punta Lara was associated with differences in biomarkers on both fish species, although a different pattern of response was observed.

Attractants are a powerful tool for pest management. The lack of specific attractants for the South American fruit fly, Anastrepha fraterculus, a complex of cryptic species of great economic importance in South America, makes it difficult to monitor the pest in the field. The γ-lactone male sex and aggregation pheromones of several Anastrepha species, naturally released in a 7:3 epianastrephin to anastrephin ratio, and a structurally related naturally occurring γ-lactone ((±)-trans-tetrahydroactinidiolide) with gem-dimethyl groups (dimethyl) at C(4), were evaluated as potential attractants of this species. Different age and mating conditions of A. fraterculus males and females were evaluated during electroantennography (EAG) and field cage experiments in which polymeric lures were deployed to contain 100 mg of attractant. Epianastrephin and dimethyl were EAG+ for all fly conditions, with epianastrephin eliciting the highest response for both sexes and immature flies showing greater responsiveness than mature flies. In the field cage experiments, immature flies were only attracted to leks; virgin females were attracted to leks, dimethyl, and both epianastrephin-anastrephin formulations (95 and 70 wt.% epianastrephin); mature-mated males were attracted to leks, dimethyl and 70 wt.% epianastrephin; and mature-mated females were only attracted to leks. Our bioassays showed a promising performance of the analog dimethyl since it elicited the same response as epianastrephin, requires fewer steps to synthesize, and contains one less chiral center than the natural pheromones. The attraction to leks was recorded for all mating conditions and ages of flies and suggests that air-borne volatiles of calling males contain cues that could act as sensory traps. The addition of any of these compounds in the synthetic attractants may result in a greater attraction and thus deserves further evaluation. Dose-response experiments will provide additional information to move a step forward and validate the results obtained in open-field conditions.

In two previous papers, we calculated the dielectrophoresis (DEP) force and corresponding trajectories of high- and low-conductance 200-µm 2D spheres in a square 1 × 1-mm chamber with plane-versus-pointed, plane-versus-plane and pointed-versus-pointed electrode configurations by applying the law of maximum entropy production (LMEP) to the system. Here, we complete these considerations for configurations with four-pointed electrodes centered on the chamber edges. The four electrodes were operated in either object-shift mode (two adjacent electrodes opposite the other two adjacent electrodes), DEP mode (one electrode versus the other three electrodes), or field-cage mode (two electrodes on opposite edges versus the two electrodes on the other two opposite edges). As in previous work, we have assumed DC properties for the object and the external media for simplicity. Nevertheless, every possible polarization ratio of the two media can be modeled this way. The trajectories of the spherical centers and the corresponding DEP forces were calculated from the gradients of the system's total energy dissipation, described by numerically-derived conductance fields. In each of the three drive modes, very high attractive and repulsive forces were found in front of pointed electrodes for the high and low-conductance spheres, respectively. The conductance fields predict bifurcation points, watersheds, and trajectories with multiple endpoints. The high and low-conductance spheres usually follow similar trajectories, albeit with reversed orientations. In DEP drive mode, the four-point electrode chamber provides a similar area for DEP measurements as the classical plane-versus-pointed electrode chamber.

BACKGROUND: Pyrethroids are among the most applied adulticides worldwide to control mosquito vectors for prevention of arboviral diseases transmission. However, pesticide resistance development in a mosquito population could lead to decreased control efficacy. While most studies investigate the resistant genotype (i.e. kdr, CYP450, etc.) as explanatory variables, few field efficacy studies have measured pesticide quantities deposited at different distances from the sprayer in association with observed mosquito mortality. The current study determined field delivered amounts of an applied ULV permethrin/PBO formulation (31% permethrin + 66% piperonyl butoxide) by GC/MS and estimated practical resistance ratios using caged mosquito females. RESULTS: For field samples, the extraction method recovered 78 ± 3.92-108 ± 8.97% of the permethrin/PBO formulation when utilizing the peaks of PBO from GC/MS to estimate the concentrations of adulticide deposited near the mosquito cages. The field bioassay showed that the spatial distribution of permethrin/PBO formulation was heterogeneous among three pseudo-replicates within the same distance. Within the quantifiable permethrin/PBO range of 15.7-51.4 ng/cm2 , field-collected mosquito mortalities started at 64% and linearly increased reaching 100% only in two areas, while all Sebring susceptible mosquitoes died. The field LC95 resistance ratio (RR) of F0 Cx. quinquefasciatus ranged from 2.65-3.51, falling within the 95% CI of RR95 estimated by laboratory vial assays. Tests with and without PBO indicated P450's enzymes contributed to field resistance. CONCLUSION: Results showed the suitability of the collection and quantification method to estimate the field resistance ratio at the applied pesticide rate. Pesticide quantification would also allow the association of the known frequencies of resistance mechanisms (e.g. kdr, CYP450) with field mortalities to estimate the resistance level conferred by such mechanisms. © 2023 The Authors. Life-history studies are often conducted in a laboratory environment where it is easy to assay individual animals. However, factors such as temperature, photoperiod, and nutrition vary greatly between laboratory and field environments, making it difficult to compare results. Consequently, there is a need to study individual life histories in the field, but this is currently difficult in systems such as Daphnia where it is not possible to mark and track individual animals. Here, we present a proof of principle study showing that field cages are a reliable method for collecting individual-level life-history data in Daphnia magna. As a first step, we compared the life history of paired animals reared outside and inside cages to test the hypothesis that cages allow free flow of algal food resources. We then used a seminatural mesocosm setting to compare the performance of individual field cages versus glass jars refilled with mesocosm water each day. We found that cages did not inhibit food flow and that differences in life histories between three clones detected in the jar assays were also detectable using the much less labor-intensive field cages. We conclude that field cages are a feasible approach for collecting individual-level life-history data in systems such as Daphnia where individual animals cannot be marked and tracked.

BACKGROUND: Aedes aegypti and Aedes albopictus are two globally invasive vectors with similar ecological niches. Encounters between them can result in either competitive exclusion or stable co-existence, but it is unclear what drives these variable outcomes. Larval competition in favor of Ae. albopictus is a main hypothesis for the competitive exclusion of Ae. aegypti observed in some regions. However, the role of oviposition preference in determining the degree of competitive larval interactions in the field is not well understood. In this study, we used a combination of mark-release-recapture methods with ovitraps in the open-field and a semi-field cage to test whether gravid Ae. albopictus seek oviposition sites in response to the presence, species, and density of either conspecific or heterospecific Ae. aegypti larvae in the aquatic habitat. We conducted our study in Medellín, Colombia, where Ae. aegypti is a long-term resident and Ae. albopictus is a recent invader. RESULTS: In the open-field and semi-field cage experiments, gravid Ae. albopictus showed strong preference for ovitraps with larvae over those without. They consistently preferred ovitraps with higher density of conspecific (Ae. albopictus) larvae and low density of heterospecific (Ae. aegypti) larvae over traps with no larvae or high density of heterospecific (Ae. aegypti) larvae. In the semi-field cage experiment, traps with low density of Ae. albopictus were not preferred more or less than any other trap, but in the open-field experiment they were preferred over traps without larvae. CONCLUSIONS: We demonstrate, through open-field and semi-field cage experiments, that Ae. albopictus are more attracted to oviposition sites with larvae and that the combination of species and density of larvae influence attraction. This demonstrated preference could increase interspecific larval competition as Ae. albopictus actively seek containers with conspecific and heterospecific larvae. Any resulting competition with Ae. aegypti may favor one species over the other and alter the distribution or abundance of both. Because these species vary in vectorial capacity and insecticide resistance, effects of interspecific competition could ultimately impact arbovirus transmission rates and the success of vector control efforts .

BACKGROUND: The mirid bug Apolygus lucorum (Meyer-Dür) is a principal pest of cotton that also causes great damage to many other crops in China. A study was conducted to assess the mortality of A. lucorum from generalist arthropod predators using both molecular methods and a field-cage trial. The species-specific primer pair for the detection of A. lucorum tissues in predators was designed according to the sequences of the cytochrome oxidase subunit I (COI) gene. RESULTS: A total of 2096 generalist predators that consisted of ladybeetles, lacewings and spiders were collected, and A. lucorum remains were detected using the designed primers. Only 1.6% of these predators contained A. lucorum DNA, with the highest positive proportion (6.1%) for Harmonia axyridis larvae. In the field-cage experiment, the daily predation rates of second-instar A. lucorum nymphs by H. axyridis adults and larvae were 4.7 and 5.2% respectively. CONCLUSIONS: The overall low positive proportion of generalist predators with A. lucorum DNA detected using the molecular method, combined with the low predation rate in the field-cage experiment, indicated that the primary generalist predators likely had a limited role in the suppression of A. lucorum in the field. © 2016 Society of Chemical Industry.

The ground adulticiding program in densely populated Harris County has been employed on the basis of virus-positive mosquitoes almost exclusively using the 'rotation of chemicals best practices' stratagem. To evaluate its effectiveness, 15 comparative field cage testing events were conducted from 2011-2015 using seven wild population samples from repeatedly collected locations of Culex quinquefasciatus Say and a laboratory susceptible Sebring strain colony reared to adults. A 3 × 3 plot design was employed for exposure to ultra-low volume applications of malathion and synergized permethrin. No significant differences were found in mortality rate among testing dates, year, row placement, or relative humidity. Mortality was significantly different between adulticides (p<0.001) with mean mortality rates for malathion 96.42% (±7.95%) and permethrin 92.38% (±14.04). There was a significant temperature difference for permethrin (p<0.001) but none for malathion (p=0.644). Mosquito population mortality was statistically different by study operational area (p<0.011) and chemical (p<0.001). Susceptible colony adults used as positive controls downwind strongly aided determination of efficacy and resistance to each adulticide, providing evidence of individual application coverage, though comparative analysis was done with overall mortalities by normal methodology.

The hemlock woolly adelgid, Adelges tsugae Annand, is an invasive pest of eastern (Tsuga canadensis (L.) Carrière) and Carolina hemlock (Tsuga caroliniana Engelmann) forests in the eastern United States. Scymnus (Pullus) coniferarum Crotch (Coleoptera: Coccinellidae) is a lady beetle that preys on A. tsugae in the western United States, where A. tsugae infestations on western hemlocks are not lethal. It is thought that S. coniferarum could be an important predator that helps keep A. tsugae populations from reaching damaging levels in this region. This study assesses the potential of this predator as a biological control agent for A. tsugae in the eastern United States. S. coniferarum predation, reproductive potential, and survival were evaluated in field-cages on adelgid-infested T. canadensis at two sites in southwestern Virginia. Sampling was conducted between December 2012 and June 2014 to evaluate the impact of S. coniferarum on both generations of A. tsuage (sistens and progrediens). Adult S. coniferarum fed on both generations and all life stages of A. tsugae during both field trials at rates comparable to other adelgid-specific predators. Evidence of S. coniferarum oviposition was minimal, and may be attributed to low temperatures and prey availability. S. coniferarum mortality was greatest when exposed to winter temperatures at the higher elevation site in 2013, and least throughout the 2014 spring sample period. S. coniferarum demonstrated a high predation rate on A. tsugae and survived for extended periods of time at sites in southwest Virginia, indicating that this species could be an effective predator of hemlock woolly adelgid in similar climates.
