RESUMO

The red-eared slider (Trachemys scripta elegans) is renowned for its remarkable adaptations, yet much of its complex biology remains unknown. In this pioneering study, we utilized a combination of gross anatomy, scanning electron microscopy (SEM), light microscopy, and immunofluorescence techniques to examine the tongue's omnivorous adaptation in this species. This research bridges a critical knowledge gap, enhancing our understanding of this intriguing reptile. Gross examination revealed a unique arrowhead-shaped tongue with a median lingual fissure and puzzle-piece-shaped tongue papillae. SEM unveiled rectangular filiform, conical, and fungiform papillae, with taste pores predominantly on the dorsal surface and mucous cells on the lateral surface of the papillae. Histologically, the tongue's apex featured short rectangular filiform and fungiform papillae, while the body exhibited varying filiform shapes and multiple taste buds on fungiform papillae. The tongue's root contained lymphatic tissue with numerous lymphocytes surrounding the central crypt, alongside lingual skeletal musculature, blood and lymph vessels, and Raffin corpuscles in the submucosa. The lingual striated muscle bundles had different orientations, and the lingual hyaline cartilage displayed a bluish coloration of the ground substance, along with a characteristic isogenous group of chondrocytes. Our research represents the first comprehensive application of immunofluorescence techniques to investigate the cellular intricacies of the red-eared slider's tongue by employing seven distinct antibodies, revealing a wide array of compelling and significant findings. Vimentin revealed the presence of taste bud cells, while synaptophysin provided insights into taste bud and nerve bundle characteristics. CD34 and PDGFRα illuminated lingual stromal cells, and SOX9 and PDGFRα shed light on chondrocytes within the tongue's cartilage. CD20 mapped B-cell lymphocyte distribution in the lingual tonsil, while alpha smooth actin (α-SMA) exposed the intricate myofibroblast and smooth muscle network surrounding the lingual blood vessels and salivary glands. In conclusion, our comprehensive study advances our knowledge of the red-eared slider's tongue anatomy and physiology, addressing a significant research gap. These findings not only contribute to the field of turtle biology but also deepen our appreciation for the species' remarkable adaptations in their specific ecological niches.

Micromorphological and phytochemical studies play a major role in quality control and standardization of traditional or herbal medications. In the present research, micromorphological assessment of Heliotropium rarifloum stocks was performed through light and scanning electron microscopies (LM & SEM). The anatomy of leaves, stem and root showed salient histological features. Both surfaces of the leaves had setose glandular trichomes measuring 20-38 × 6-15 µm. The lower epidermis had comparatively a maximum anomocytic stomata (16-35) and stomatal index (12-33). The mature pollen grains were small (74 µm) and spheroidal shaped, with psilate exine (2 µm) sculpturing. Vein termination and vein islet number of the upper epidermis were 5-20 and 5-15, respectively. The palisade ratio of the leaf lamina for the upper and lower epidermis was 2-10 and 2-8. LM and SEM of the powdered samples displayed crystals, phloem fibers, xylem, vessels, sieve tube elements, companion cells, and tracheids. Extractive values determination, fluorescence, and phytochemical analysis were employed for quality control according to the World Health Organization (WHO) guidelines. Phytochemical screening revealed various secondary metabolites. It is suggested that H. rariflorum might be a reliable source of nutrients and secondary metabolites and might be more medically effective. The current findings confirm its standardization and validation.

In the present study, pentastomids belonging to the order Cephalobaenida were isolated from the lungs of Berber skinks Eumeces schneideri (Famiy: Scincidae), which were morphologically described by light and scanning electron microscopy and taxonomically justified by 18s rDNA molecular analyses of the parasites. Seventeen host specimens were collected from well-vegetated wadis at high altitudes, Jizan, Saudi Arabia as new type locality; twelve specimens (70.59%) were infected. All of the recovered parasites were adults, possessed small broadly triangular cephalothorax flattened on the ventral surface and merged smoothly with a uniformly thick and squat abdomen and terminated in a pair of divergent lobes. The results obtained indicated that the parasites belong to the sharp-tipped posterior-hook Raillietiella spp. distinguished from other raillietiedids of the same group some important characteristic features including annulus number, shape and dimensions of the buccal cadre, copulatory spicules, and anterior and posterior hooks. The anterior hook of the female specimens (n=5) had a blade length (AB) of 135±5 (110-146) µm and shank length (BC) 158±5 (150-169) µm while the posterior hook was much larger with AB measuring 221±5 (200-236) µm and BC 286±6 (280-289) µm. For the male specimens (n=5), the anterior hook had an AB of 73±3 (72-75) µm and a BC 102±5 (100-103) µm. The posterior hook was much larger with AB 190.6±5 (190-191) µm and BC 221±5 (280-289) µm. The morphological characterization of the recovered parasites was closely similar to R. aegypti previously isolated from the same host. Sequence alignment by the maximum likelihood analysis for the data obtained from the 18S rDNA analysis of the parasites exhibits identities ranging between 88-95% with pentastomid genera recovered from the GenBank. The phylogenetic tree supported the inclusion of the parasites within the monophyletic Pentastomida clade with maximum identity to the raillietiellid species. The recovered sequences from the present study were deposited in GenBank under Accession number MK970649.1. The present molecular analysis was the first to confirm the taxonomic position of R. aegypti isolated from the host examined.

Parasitic gastroenteritis (PGE) caused by infection of the gut with parasitic nematodes is one of the most important diseases of livestock animals from both financial and welfare perspectives. Parascaris equorum and Habronema microstoma are of the most endemic nematodes of the world which are currently the major cause of PGE of the domestic horses in Egypt. The present investigation introduced the first morphological description of these nematodes recovered from the domestic horse, Equus ferus caballus (Equidae), in Egypt by light and scanning electron microscopy. Seven P. equorum (fifth stage) and 18 adults of H. microstoma were recovered from the gastrointestinal tracts of four young domestic horses collected during the year of 2015. Microscopic examination of the isolated fifth stage P. equorum revealed that it possessed a long body with a broad anterior end equipped by large shamrock-like lips with deep transverse groove on medial surface set off from the rest of the body by a deep post-labial constriction giving the body a shouldered appearance. The total body length was 12-15 (14 ± 2) cm for males and 13-18 (16 ± 2) cm for females. Lips were three in number in the form of one dorsal and two sub-ventral surrounding the central stoma. The isolated adult worms of H. microstoma were whitish in color narrowed slightly at the anterior end. Single lateral ala in the cephalic region in both sexes was observed. The buccal vestibule was markedly thickened and equipped by two tridentate teeth. The adult worms had two bilobed lateral lips surrounding the mouth with four sub-median cephalic papillae and two amphids. The males were 14.5-18.0 (17.2 ± 0.3) mm long and 1.23-1.57 (1.42 ± 0.3) mm wide. The posterior end was spirally coiled and had wide caudal alae. The spicules were unequal. The females were 13.5-21.0 (16.2 ± 0.3) mm long and 1.55-1.75 (1.69 ± 0.3) mm wide. The anal pore had a thin upper rim and was located 177.0 µm from the posterior end.

Diatoms inhabit different aquatic and non-aquatic environments, including soils. The naviculoid genus Luticola is widespread in various habitats and accounts for 264 species that are only based on morphological and morphometric characteristics. These parameters can greatly vary during the life cycle, making the species very similar to each other and complicating their unambiguous identification. During a study on soil algal diversity in the Russian Far East (Jewish Autonomous Region), we isolated a strain of naviculoid diatom and examined it using an integrative approach (phylogenetic, morphological, ultrastructural data, and life cycle). Phylogenetic analyses, based on chloroplast rbcL gene data, showed affinity of the new strain with the genus Luticola. Our alga shares morphological features typical of the genus members but differs from them by having valves with a larger width and hook-shaped external proximal raphe ends deflected to the side opposite the stigma. It was revealed that the strain reproduces via two types of sexual reproduction-isogamy and cis-anisogamy. Based on these phenotypic traits, we described the new isolate as Luticola tenera sp. nov.

Prevention of fungal diseases caused by Fusarium species, including F. culmorum, and thus the accumulation of mycotoxins in wheat ears, is a constant challenge focused on the development of new, effective crop management solutions. One of the currently most ecologically attractive approaches is biological control using natural antagonistic microorganisms. With this in mind, the antagonistic potential of thirty-three Clonostachys and Trichoderma strains was assessed in this work. Screening tests were carried out in in vitro cultures, and the observed potential of selected Trichoderma and Clonostachys strains was verified in field and semi-field experiments with two forms of wheat: winter cv. Legenda and spring cv. Bombona. Three strains, namely C. rosea AN291, T. atroviride AN240 and T. viride AN430 were reported to be most effective in inhibiting the growth of F. culmorum KF846 and the synthesis of DON, 3AcDON and ZEN under both laboratory and semi-controlled field conditions. Observations of the contact zones of the tested fungi in dual cultures exposed their mycoparasitic abilities against KF846. In addition, studies on liquid cultures have demonstrated the ability of these strains to eliminate F. culmorum toxins. Meanwhile, the strains of T. atroviride AN35 and T. cremeum AN392 used as soil inoculants in the field experiment showed a different effect on the content of toxins in ears (grains and chaffs), while improved wheat yield parameters, mainly grain health in both wheat cultivars. It is concluded that the selected Trichoderma and Clonostachys strains have a high potential to reduce the adverse effects of F. culmorum ear infection; therefore, they can be further considered in the context of potential biocontrol factors and as wheat crop improvers.

In Egypt, domestic rabbits are commercially used for meat production. Adversely, gastrointestinal helminthiasis largely delays such concern. Passalurus ambiguus is a common intestinal nematode that infects rabbits causing intestinal disorders and death in severe infection. Therefore, domestic rabbits collected from several locations (poultry slaughter shops, rabbits farms, markets and public owners) in Qena, Upper Egypt. Passalurus ambiguus worms were detected in 90 out of 200 rabbits (45%). Grossly, they appeared as small white nodules in the appendix of the rabbits examined. Adult worms were described morphologically using light and scanning electron microscopy. Adult male measured 4.622 mm (2.838-7.172 mm) in length and 0.278 mm (0.139-0.558 mm) in width. Adult female measured 5.622 mm (2.347-9.532 mm) in length and 0.314 mm (0.185-0.381 mm) in width. SEM showed a transverse cuticular striations on the body of both males and females, four papillae on the dorsal and ventral surface and the mouth was triangular and surrounded by three teeth. Phylogenetic data proved the occurrence of the intestinal nematode, Passalurus ambiguus. Microscopically, heavy worms infestated the appendiceal layers causing appendicitis manifested by granuloma with hyperplasia in the epithelial lining inducedobstruction in the lumen. It could be concluded that morphological features, molecular phylogenetic data, and histopathological findings clearly identified the present species as Passalurus ambiguus Rudolphi, 1819.

Investigation of alternative energy sources is need of current time due to growing power crisis and associated environmental issues. Biodiesel is considered as sustainable power source and promising alternative to fossil fuels. Therefore, our current investigation aimed to identify micromorphological characters of 10 novel nonedible oil-yielding seeds through scanning electron microscopy. It was revealed from light microscopic study that there is variation in seed size from 3 to 15 mm in length and 2 to 11 mm in width. Likewise, a huge variation in color was observed such as light green, greenish yellow, blackish brown, and various shades of brown. Presence and absence of Hilum was observed, and compression of seeds varied from depressed, lateral, and dorsoventral. Seed's shape differs from ovate, clavate, triangular ovate, cuneiform, ovoid, and elliptical shape. Seed oil content fall in range of 18-58% (wt/wt). Free fatty acid content of the seeds varies from 0.3 to 3.1 mg KOH/g. Ultrastructure of seeds exhibited huge variation in shape, size, periclinal wall, anticlinal wall, and surface ornamentation. Nonedible seeds varied in wall structure from angular, wavy, dentate entire, irregular, puzzled, elongated, even, and polygonal. The periclinal wall arrangements show alteration from flat, looped, raised, depressed, lofty, even, pentagonal, polygonal, and undulate seed margins. Outcomes of this investigation recommended that scanning electron microscopy could act as a helpful tool in disclosing the hidden micromorphological characters among nonedible oil-yielding seeds and subsequently helping in correct, authentic seed identification and classification as potential feedstock for biodiesel.

Rapid advances in the field of pathogen detection have opened new opportunities and better understanding for their management approaches. Aim of this study was to elucidate histopathological observations of different tissues affected by Macrophomina phaseolina and to observe the defense responses of plant growth promoting rhizobacteria (PGPR) in mungbean plants. Sections of the stem and root were prepared and stained with ferric chloride, Lugol's iodine and Wiesner's reagent and were then observed under multiple microscopic techniques. Results revealed that both pathogen and PGPR produce responses on the plant that include colonization of xylem vessels by hyphae and sclerotia, hypertrophy and hyperplasia of the cells, destruction of xylem fibers and amyloplasts in parenchymatous cells; and production of gels by the plant were observed. There was a significant increase in lignin and phenolic compounds deposition in stem and root sections of PGPR treated and non-treated mungbean plants. Whereas the soil amended with PGPR showed very less to no starch production. Moreover, production of gels and gums were also observed in both stem and root sections. Compared to light microscopy, scanning electron microscope provided greater depth of focus and resolution of the pathogen attack on plant tissues, associated bacteria. As a whole, the data demonstrated that inoculation of PGPR can be an effective strategy to stimulate plant growth and they could significantly activate disease resistance against M. phaseolina.

The present study was undertaken to gather information on selected species of Echinochloa based on their morphological and palyonological examination through light and scanning electron microscopy. As the Echinochloa species are most problematic grass weeds and exhibit high interspecific and intraspecific variability so due to their great morphological diversity it is difficult to recognize them. A lot of variation was observed in morphological features of studied taxa. In the present study, E. colona had more or less spreading dense racemes whereas in E. crus-galli racemes were linear to ovate. Upper glume surface was pubescent in E. colona, hispid in E. crus-galli and spinose in E. walteri. E. colona was comprised of five-nerved glumes whereas in E. crus-galli glumes were three-nerved. Although pollen basic characters in family Poaceae remains consistent but great variation was observed in pollen of studied taxa. The pollen size was quite variable among the species of genus Echinochloa. In polar view, the size of pollen grains ranged from 27.5 (20-35) µm to 40 (30-50) µm. E. crus-galli was appeared to be the smallest in size whereas E. colona was the largest.). P/E ratio ranged from 0.84 to 1.15 µm among the species. The highest value of pollen fertility was found in E. colona as 93.33% and lowest was in E. crus-galli as 87.50%. The morphological key of studied taxa is also given to study variations. In conclusion, it was observed that microscopy of morphological and palynological assessment can play a vital role in identification of taxonomically problematic taxa.

Live cell fluorescence imaging is the method of choice to visualize dynamic cellular processes in time and space, such as adhesion to and invasion of polarized epithelial cells by Salmonella enterica sv. Typhimurium. Scanning electron microscopy provides highest resolution of surface structures of infected cells, providing ultrastructure of the apical side of host cells and infecting Salmonella. Combining both methods toward correlative light and scanning electron microscopy (CLSEM) enables new insights in adhesion and invasion mechanisms regarding dynamics over time, and high spatial resolution with precise time lines. To correlate fast live cell imaging of polarized monolayer cells with scanning electron microscopy, we developed a robust method by using gold mesh grids as convenient CLSEM carriers for standard microscopes. By this, we were able to unravel the morphology of the apical structures of monolayers of polarized epithelial cells at distinct time points during Salmonella infection.

The late Miocene is marked by pronounced environmental changes and the appearance of strong temperature and precipitation seasonality. Although environmental heterogeneity is to be expected during this time, it is challenging to reconstruct palaeoenvironments using plant fossils. We investigated leaves and dispersed spores/pollen from 6.4 to 6 Ma strata in the intermontane Florina-Ptolemais-Servia Basin (FPS) of northwestern Greece. To assess how well plant fossils reflect the actual vegetation of the FPS, we assigned fossil taxa to biomes providing a measure for environmental heterogeneity. Additionally, the palynological assemblage was compared with pollen spectra from modern lake sediments to assess biases in spore/pollen representation in the pollen record. We found a close match of the Vegora assemblage with modern Fagus-Abies forests of Turkey. Using taxonomic affinities of leaf fossils, we further established close similarities of the Vegora assemblage with modern laurophyllous oak forests of Afghanistan. Finally, using information from sedimentary environment and taphonomy, we distinguished local and distantly growing vegetation types. We then subjected the plant assemblage of Vegora to different methods of climate reconstruction and discussed their potentials and limitations. Leaf and spore/pollen records allow accurate reconstructions of palaeoenvironments in the FPS, whereas extra-regional vegetation from coastal lowlands is probably not captured.

Palyno-anatomical study of monocots taxa using Light and Scanning Electron Microscopy (SEM) was first time conducted with a view to evaluating their taxonomic significance. Studied plants were collected from different eco-climatic zones of Pakistan ranges from tropical, sub-tropical, and moist habitats. The aim of this study is to use palyno-anatomical features for the correct identification, systematic comparison, and investigation to elucidate the taxonomic significance of these features, which are useful to taxonomists for identifying monocot taxa. A signification variation was observed in quantitative and qualitative characters by using the standard protocol of light microscopy (LM) and SEM. Epidermal cell length varied from maximum in Allium griffthianum (480 ± 35.9) µm at the adaxial surface to minimum in Canna indica (33.6 ± 8.53) µm on abaxial surface. Maximum exine thickness was observed in Canna indica (4.46) µm and minimum in Allium grifthianum (0.8) µm. Variation was observed in shape and exine ornamentation of the pollen, shape of the epidermal cell, number, size, and type of stomata, guard cell shape, and anticlinal wall pattern. Based on these palyno-anatomical features a taxonomic key was developed, which help in the discrimination of studied taxa. In conclusion, LM and SEM pollen and epidermal morphology is explanatory, significant, and can be of special interest for the plant taxonomist in the correct identification of monocots taxa.

We report for the first time the presence of cluster crystals of calcium oxalate within the glandular trichomes and oil bodies in the mesophyll for Baccharis species. Moreover, the comparative leaf anatomy and micro-morphology of six species of Baccharis, namely B. illinita, B. microdonta, B. pauciflosculosa, B. punctulata, B. reticularioides, and B. sphenophylla is investigated by light and scanning electron microscopy. The studied species exhibited differences in their leaf anatomical features such as the morphology of the cuticle, type and occurrence of the stomata, presence or absence of glandular trichomes, shape of the flagelliform trichomes, and the arrangement of the mesophyll tissues. These differences can be helpful in the species identification and classification and could represent informative characters for the reconstruction of the evolution of the genus.

Resumen En el presente estudio se aisló un pentastómido perteneciente al orden Cephalobaenida del pulmón de un eslizón bereber (Eumeces schneideri, Familia: Scincidae). Se efectuó su descripción morfológica basada en observación por microscopía óptica y de bar rido y se justificó su ubicación taxonómica mediante análisis molecular del gen 18S del ADNr. Se recolectaron 17 especímenes del citado huésped en valles ubicados a elevadas altitudes, en la región de Jizan (Arabia Saudí); 12 de ellos (70,59%) estaban infectados. Todos los parásitos recuperados eran adultos y poseían un pequeño cefalotórax triangular, aplanado en la super ficie ventral, que se fusionaba con un abdomen abultado y terminado en un par de lóbulos divergentes. Los resultados indicaron que este parásito pertenece a Raillietiella spp., que agrupa especies con gancho posterior puntiagudo; estas se distinguen de otros miembros de la Familia Raillietiella por algunos rasgos característicos, como el número de anillos y la forma y dimensiones del cuadro bucal, las espículas copulatorias y los ganchos anterior y posterior. La caracterización morfológica demostró que el parásito recuperado era muy similar a R. aegypti, previamente aislada del mismo huésped. El alineamiento de secuencias mediante el método de probabilidad máxima basado en el análisis del gen 18s del ADNr detectó identidades del 88-95% con los géneros de pentastómidos disponibles en GenBank. Dentro del árbol filogenético se pudo incluir este parásito dentro del clado monofilético pentastómido con máxima identidad con las especies de Raillietiella. Las secuencias obtenidas fueron depositadas en GenBank, con número de acceso MK970649.1. El presente análisis molecular confirma por primera vez la posición taxonómica de Raillietiella aegypti, anteriormente aislado del mismo huésped.

Abstract Ocotea porosa (Nees & Mart.) Barroso, commonly known as "imbuia", "canela-imbuia" or "imbuia-amarela" in Brazil, is a tree of the Southern Atlantic Forest. The present study investigates the anatomy of leaf and stem, volatile oil chemistry, as well as cytotoxicity and insecticidal activities of the essential oil of O. porosa. Species identification was achieved by anatomy features, mainly due to paracytic and anomocytic stomata; non-glandular trichomes; biconvex midrib and petiole with a collateral open arc vascular bundle; presence of a sclerenchymatous layer, starch grains and crystal sand in the stem; and the presence of phenolic compounds in the epidermis, phloem and xylem of the midrib, petiole and stem. The main volatile components of the essential oil were α-pinene (19.71%), β-pinene (13.86%) and bicyclogermacrene (24.62%). Cytotoxicity against human cancer cell (MCF-7), mouse cancer cell (B16F10) and mouse non-tumoral cell (McCoy) was observed as well as insecticidal activity of the essential oil against susceptible 'Ft. Dix' bed bugs (Cimex lectularius L.) by topical application.

Abstract The present work investigates the leaf and stem anatomy, chemical composition and insecticidal activities (against Cimex lectularius Linnaeus, 1758) of the volatile oils of Schinus molle L., Anacardiaceae, a Brazilian native traditional medicinal plant. Noteworthy micro-morphological features that can help in the identification and quality control of the species include the presence of isobilateral and amphistomatic leaves, anomocytic and cyclocytic stomata, capitate glandular and conical non-glandular trichomes, large secretory ducts in the midrib, presence of druses and prismatic crystals, and the petiole vascular system comprising of five vascular bundles arranged in U-shape and an additional dorsal bundle. The major components of the volatile oil include β-pinene (14.7%), α-pinene (14.1%), limonene (9.4%) and muurolol (11.8%). Insecticidal activities of the volatile oil against bed bugs were investigated for the first time; strong toxicity by fumigation with the volatile oil of S. molle was observed and reported herein.

ABSTRACT Piper caldense C. DC., Piperaceae, commonly known as "pimenta-d'água", "pimenta-darda" or "paguarandy" in Brazil, is a shrub that grows mainly in humid and shaded habitats. The present study investigates the anatomy of the leaves and stems of P. caldense by light and scanning electron microscopy in order to provide supporting data for correct identification of the species. The leaves are hypostomatic, have a 2-layered hypodermis, and posses pearl glands. The midrib shows a 'U'-shaped stele comprised of about ten collateral vascular bundles. The main anatomical marker of the stem is the presence of a continuous sclerenchymatous sheath in the pith. Two forms of calcium oxalate crystals, namely crystal sand and raphides, are observed in this species.

ABSTRACT The present work provides a comparative account of the morpho-anatomy of six species of Eucalyptus, namely E. badjensis Beuzev. & Welch, E. benthamii Maiden & Cambage, E. dunnii Maiden, E. grandis W.Hill, E. globulus Labill. and E. saligna Sm., Myrtaceae. Leaf samples of these six species were investigated by light and scanning electron microscopy. The observed microscopic features that can be useful in the identification and quality control of the studied species include the morphology of epicuticular waxes, presence of prismatic crystals on the leaf surface, leaf midrib shape and arrangement of its vascular system, and the presence or absence of the sclerenchymatous fiber caps in the vascular bundle.
