RESUMO

Understanding the complex elements affecting signal resolution in cytometry is key for quality experimental design and data. In this study, we incorporate autofluorescence as a contributing factor to our understanding of resolution in cytometry and corroborate its impact in fluorescence signal detection through mathematical predictions supported by empirical evidence. Our findings illustrate the critical importance of autofluorescence extraction via full spectrum unmixing in unmasking dim signals and delineating the expression and subset distribution of low abundance markers in discovery projects. We apply our findings to the precise definition of the tissue and cellular distribution of a weakly expressed fluorescent protein that reports on a low-abundance immunological gene. Exploiting the full spectrum coverage enabled by Aurora 5L, we describe a novel approach to the isolation of pure cell subset-specific autofluorescence profiles based on high dimensionality reduction algorithms. This method can also be used to unveil differences in the autofluorescent fingerprints of tissues in homeostasis and after immunological challenges.

BACKGROUND: This study presents a novel photoelectrochemical (PEC) conversion method for ion-selective electrodes (ISEs) based on CdS semiconductor film. The motivation stems from the need to enhance the sensitivity and precision of ISEs for various analytical applications. RESULTS: We synthesized CdS film on FTO conductive glass via a hydrothermal method and utilized this electrode as the working electrode. Under visible light irradiation, CdS generated photocurrent that is proportional to its applied voltage within a large potential window of ∼0.80 V. Ascorbic acid (AA) effectively inhibited electron-hole complexation, enhancing photocurrent stability. Potential modulation from ISEs acting as the reference electrode further regulated photocurrent generation, demonstrating excellent sensitivity and linearity for a wide range of ion concentrations. The method was validated by detecting serum calcium levels, showing agreement with traditional ISEs potentiometry and ICP-OES methods. SIGNIFICANCE: This photoelectrochemical conversion strategy offers a promising approach for sensitive and accurate ion detection, with potential applications in clinical diagnostics and environmental monitoring.

OBJECTIVES: The diet and lifestyle affect our life. Inadequate nutrition can cause various diseases including cardiovascular diseases. The aim of this study was to show the correlations between the fruit and vegetable diet and high signal resolution pulse wave parameters. DESIGN: This was an observational study. SETTINGS: The study was done during two-weeks rehabilitation treatment. PARTICIPANTS: In this study 154 people using the fruit and vegetable diet have been examined. MEASUREMENTS: The participants were monitored using a new diagnostic method high signal resolution pulseoximetry (HSR-PW). They were examined two times: before starting the diet and after two weeks of using it. The high signal resolution pulse wave and its characteristic parameters have been compared. RESULTS: Analyzing the research results at the beginning and after two weeks of using this diet, the improvement of selected parameters has been noticed. With the improvement in the pulse wave was observed weight loss, improved blood counts (e.g. cholesterol, triglycerides) as well as decreased blood pressure particularly in people with treated hypertension. CONCLUSIONS: The study shows that applied fruit and vegetable diet influenced favorably the people using it and contributed to the improvement of the HSR-PW parameters which are the source of information about the state of the cardiovascular system.
