Resumo

Emergências obstétricas são comuns na rotina clínica de cães e gatos. Dentre as emergências obstétricas uma das mais importantes é a distocia. De modo geral, a distocia ocorre quando há falha no momento do parto ou na expulsão do feto, sendo que a causa mais comum de distocia nas duas espécies é a inércia uterina primária. Outra emergência obstétrica de ocorrência frequente em cadelas e gatas é a piometra. Esta doença é uma infecção que ocorre no útero, causada por alterações hormonais na fase do diestro dos animais. As duas enfermidades (distocia e piometra) podem ter o envolvimento de anticoncepcionais, tanto nas cadelas como nas gatas. Em relação ao tratamento, na maioria das vezes, as distocias em cadelas e gatas são abordadas por meio da realização de cesarianas. Em piometra, a terapêutica mais efetiva é a realização de ovariohisterectomia. Neste estudo, 43 cadelas e 20 gatas com emergências obstétricas foram monitoradas durante 90 dias. Entre as cadelas, 29 (67,44%; 29/43) apresentaram piometra e 13 (30,23%; 13/43) distocia. Além disso, uma cadela apresentou prolapso vaginal. Entre as gatas, 13 apresentaram distocia (65%; 13/20) e sete (35%; 7/20) piometra. Algumas dessas emergências obstétricas foram relacionadas ao uso de anticoncepcional. Assim, os clínicos veterinários devem desencorajar esta prática e incentivar a castração das fêmeas.


Obstetric emergencies are common in the clinical routine of dogs and cats. One of the most important obstetric emergencies is dystocia. In general, dystocia occurs when there is a failure at the parturition time or expulsion of the fetus, and the most common cause of dystocia in both species is primary uterine inertia. Another frequently occurring obstetric emergency in bitches and cats is piometra. This disease is an infection that occurs in the uterus caused by hormonal changes in the diestrus phase of animals. In both diseases (dystocia and pyometra), contraceptives can be involved in female dogs and cats. Regarding the treatment, most of the time, dystocias in bitches and cats are addressed through cesarean sections. In pyometra, the most effective therapy is to perform ovariohysterectomy. In the present study, 43 female dogs and 20 female cats with emergency obstetric were monitored during 90 days. Among bitches, 29 (67.44%; 29/43) had pyometra, and 13 (30.23%; 13/43) showed dystocia. In addition, one female dog presented vaginal prolapse. Among the cats, 13 had dystocia (65%; 13/20) and seven (35%; 7/20) pyometra. Some of these obstetric emergencies were related to contraceptive use. Thus, veterinary clinicians must discourage this practice and encourage the neutering of females.

Background: Pyometra is a bacterial and hormone-induced reproductive disease that occurs in the post-estrus luteal phase in intact queens. Pyometra is more common in the diestrus period due to the high progesterone concentration (in queens that mated, spontaneously ovulated, or were induced to ovulate). However, it can also be seen due to the use of exogenous hormones such as progesterone for the suppression of estrus. More research is needed in cases of pyometra in queens, as well as in bitches. Because, considering that the pathogenesis and characteristics of feline pyometra is similar to bitches, studies on pyometra-affected bitches are taken as reference in studies and applications on queens. From this point of view, the aims of this study were to reveal the changes in complete blood count, blood gas, and serum biochemistry parameters in feline pyometra cases and to determine the correlation between the mentioned parameters. Materials, Methods & Results: In the study, a total of 25 female cats of different breeds were used, between the ages of 6 months and 7 years, 15 were diagnosed with pyometra, and 10 healthy. Anamnesis, clinical findings, and ultrasonographic examinations were used in the diagnosis of pyometra. Abdominal ultrasonography was performed on queens brought to the clinic with complaints such as anorexia, polydipsia, polyuria, abdominal tension, and fever. The control group (n =10) consisted of queens that were introduced to the clinic and were reproductively healthy. Before any treatment in queens with pyometra and the control group, 1 mL blood samples were taken from v. cephalica to evaluate complete blood count, blood gases and serum biochemistry parameters. In complete WBC, Lym, Mon, Gra, RBC, Hb, HCT, MCV, MCH and PLT parameters and, blood gas parameters such as pH, pCO2 , pO2 , sO2 , Na, K, Cl, lactate, glucose, HCO3 , and BE were also evaluated in taken blood samples. Biochemical parameters BUN, creatinine, ALT, AST, ALP, amylase, T.BIL, D.BIL, P, CHOL, TG, LDH, TP, CPK, ALP, Ca, GGT were measured in serum samples. After examination and laboratory analysis, ovariohysterectomy was performed on queens as a treatment. Granulocyte, WBC, HCT and MCH levels of the pyometra group were higher (P < 0.05) and Lym levels were lower (P < 0.05) compared to the control group. According to these results, pH, HCO3 , and BE were lower (P < 0.05) in queens with pyometra than those in the control group, while Na and lactate parameters were higher (P < 0.05). According to the results of biochemical analysis, it was determined that BUN, creatinine levels, GGT, and LDH enzyme activities were found to be higher in the pyometra group compared to the control group, while the Ca level was found to be low (P < 0.05). A positive correlation was observed between BUN and creatinine and LDH, WBC, granulocyte, HCT, and lactate, and a negative correlation between lymphocytes, pH, and BE in the correlation analysis performed on queens with pyometra and control group. However, a positive correlation was observed between creatinine and LDH and HCT, and a negative correlation between lymphocyte, pH and BE. Discussion: There is not enough information about pyometra in queens. As a result, it was determined that there were significant changes in complete blood count, blood gases and serum biochemical parameters in queens with pyometra in this study. These changes were generally thought to be related to dehydration and sepsis or endotoxemia. In addition, it was evaluated that prerenal azotemia occurring in pyometra affected queens may cause renal dysfunction. For this reason, it is thought that the results obtained in the presented study may contribute to the diagnosis, treatment, and prognosis of pyometra cases in queens.

The reproductive biotechnologies directly assist in the development of different methods of reproductive evaluation. The objective of this study was to develop a new method to determine the estrous cycle phase in equine females through vaginal cytology throughout the seasons, as well as to establish an estimate of inflammatory cells present in the vagina. Six mares were evaluated at different ages without a defined breed and reproductive activity and were subjected to ultrasound evaluation of the reproductive system and cytological analysis of the vaginal region. In the summer, there was a predominance of keratinized epithelial cells in the estrus and intermediate in the diestrus. In the autumn, there was a predominance of keratinized cells in estrus and parabasal cells in estrus and diestrus. In anestrus (winter), a greater number of parabasal cells was identified in relation to the other cell types. In estrus (spring), there was a predominance of parabasal and intermediate cells. Conversely, in diestrus, parabasals were found in greater numbers. The evaluation of inflammatory cells of the vaginal epithelium of mares showed greater activity in the summer; however, there are no reference values for healthy animals in the literature, and it is necessary to conduct studies on the subject. It is concluded that there is an influence of cyclicity on the vaginal epithelium of the equine species, varying according to the season. Additionally, vaginal cytological evaluation is an important complementary tool in the diagnosis of vaginitis in mares, requiring further research on the subject.(AU)


As biotecnologias reprodutivas auxiliam diretamente no desenvolvimento de diferentes métodos de avaliação repro-dutiva. O objetivo deste estudo foi desenvolver um novo método para determinar a fase do ciclo estral em fêmeas equinas através da citologia vaginal ao longo das estações, bem como estabelecer uma estimativa das células inflamatórias presentes na vagina. Seis éguas foram avaliadas em diferentes idades, sem raça definida e em atividade reprodutiva, e foram submetidas à avaliação ultrassonográfica do aparelho reprodutor e análise citológica da região vaginal. No verão, houve predomínio de células epiteliais queratinizadas no estro e intermediárias no diestro. No outono, houve predomínio de células queratinizadas no estro e células parabasais no estro e diestro. No anestro (inverno), foi identificado maior número de células parabasais em relação aos demais tipos celulares. No estro (primavera), houve predomínio de células parabasais e intermediárias. Por outro lado, no diestro, os parabasais foram encontrados em maior número. A avaliação das células inflamatórias do epitélio vaginal de éguas mostrou maior atividade no verão; entretanto, não há valores de referência para animais saudáveis na literatura, sendo necessário a rea-lização de estudos sobre o assunto. Conclui-se que há influência da ciclicidade no epitélio vaginal da espécie equina, variando de acordo com a estação do ano. Além disso, a avaliação citológica vaginal é uma importante ferramenta complementar no diagnóstico da vaginite em éguas, necessitando de mais pesquisas sobre o assunto.(AU)

As cadelas possuem características especiais da fisiologia da reprodução que as diferenciam das outras espécies domésticas. Elas são monoéstricas não sazonais com ovulação espontânea. A puberdade ocorre com variação entre 6 e 24 meses de idade conforme raça, tamanho individual e predisposições individuais. O interestro ocorre com intervalos de 5 a 12 meses sendo em média com 6 meses. O ciclo reprodutivo das cadelas é composto de 4 fases distintas: proestro (duração de 5 a 20 dias), estro (duração de 5 a 15 dias), diestro (duração de 50 a 80 dias) e anestro (duração de 80 a 240 dias). O diagnóstico e acompanhamento das fases do ciclo estral é feito com observação de comportamento reprodutivo, sintomas e sinais anatômicos e fisiológicos, citologia vaginal, dosagem do nível sérico de progesterona, vaginoscopia e utrassonografia dos órgãos do sistema reprodutivo, em especial dos ovários em cada fase do ciclo estral.(AU)


Female dogs have special features of reproduction physiology that differentiate them from other domestic species. Theu are non-seasonal monoestrus with spontaneous ovulation. Puberty occurs with a variation between 6 and 24 months of age according to breed, individual size, and individual predispositions. The inter-estrus occurs at intervals of 5 to 12 months, with an average of 6 months. The reproductive cycle of bitches is composed of 4 distinct phases: proestrus (duration from 5 to 20 days), estrus (duration from 5 to 15 days), diestrus (duration from 50 to 80 days) and anestrus (duration from 80 to 240 days). The diagnosis and monitoring of the phases of the estrous cycle is made with observation of reproductive behavior, symptoms and anatomical and physiological signs, vaginal cytology, measurement of serum progesterone levels, vaginoscopy and ultrasound of the organs of the reproductive system, especially the ovaries in each phase of the estrus cycle.(AU)

Abstract The aim of this research was to evaluate the use of pyridoxine hydrochloride and its associated side effects in the treatment of pseudopregnancy in female dogs. A total of 40 female dogs, with no defined breed, in non-gestational diestrus, with clinical complaint of milk production were selected. The female dogs were divided into four experimental groups of 10 animals each, treated orally for 20 days with 10mg/kg/day (G1) and 50mg/kg/day (G2) of pyridoxine hydrochloride (vitamin B6), 5μg/kg/day of cabergoline (G3), and with a placebo, in the case of the control group (G4). The effects of the treatments on milk production were investigated, as well as possible systemic side effects, macroscopic uterine and ovarian alterations, and uterine histology. During the investigated period, G2 and G3 were equally efficient (P>0.05) in lactation suppression, differing (P>0.05) from the other groups. There were no systemic side effects or uterine changes associated with administration of the studied drug. Vitamin B6 (50mg/kg) has shown to be a safe and economically viable alternative for lactation interruption in female dogs with pseudopregnancy.

Background: Pyometra is defined as chronic purulent inflammation of the uterus that causes changes in hematologicaland biochemical parameters. The disease is characterized with bacterial infection and pus accumulation in the uterus.Transabdominal B-mode ultrasonography provides easy and certain diagnosis of this disease. The hemodynamic changesin pyometra are evaluated by Doppler ultrasonography. The aim of the study is to determine the changes in hematologicalparameters and Doppler indices in bitches with pyometra, diestrus healthy bitches and evaluate the relationship betweenhematological parameters and hemodynamic indices within the both groups.Materials, Methods & Results: A total number of 27 bitches were enrolled in the study. The healthy diestrus bitches (groupH; n = 7) aged 6.2 ± 1.14 years, weighted 14.57 ± 3.75 kg. The bitches with pyometra (group PYO; n = 20) aged 9.1 ± 0.62years and weighted 17.65 ± 2.60 kg. Before all bitches had ovariohisterectomy, hematological analyses were performed.Transabdominal ultrasonography (USG) was performed with a 6.6 MHz convex transducer. The widest cross-sectionaluterine diameter (UD), wall thickness of uterine horns (UWT) and presence of luminal content were evaluated. Diameterof the uterine artery (DUA) was measured on a mapped color image using the USG software program. The examinationwas carried out with pulsed-wave Doppler USG to characterize the waveform of the uterine artery (UA). Anechogenic areasin uterine lumen, increase in UD and UWT were observed in group PYO. All cases in group PYO had luminal content inboth uterine horns ranging from 1.2 to 5.6 cm. The DUAs were measured in group H and in group PYO as 1.75 ± 0.03mm, 1.94 ± 0.08 mm; respectively (P < 0.05). Diferentemente de outras espécies domésticas, a maturação in vitro (MIV) de oócitos caninos apresenta sucesso limitado. O objetivo deste estudo foi avaliar o efeito do HEPES na maturação in vitro de oócitos caninos obtidos de cadelas em diestro e anestro. Os ovários foram coletados, isolados assepticamente e transportados refrigerados a uma temperatura de 4 ºC. Os complexos cumulus-oócito (CCOs), provenientes das duas fases do ciclo estral, foram submetidos a dois tratamentos: meio TCM-199 com adição de 25 mM de HEPES (GT) e meio sem suplementação (GC). Depois de 72 horas de maturação, os CCOs foram desnudados, fixados e corados com HOESCHT 33342 para avaliação da maturação nuclear. Os oócitos obtidos da fase de anestro e diestro do GT demonstraram, em relação ao grupo GC, maior frequência de oócitos nos estágios de M-II (p<0,01). Comparando-se os diferentes status reprodutivos, observou-se que os oócitos obtidos da fase de diestro apresentaram índices maiores de QVG e M-II. Nossos resultados demonstraram que o HEPES preserva a viabilidade e morfologia oocitária, indispensáveis para a aquisição da competência meiótica, potencializando as taxas de M-II, e que os oócitos obtidos da fase de diestro estão mais aptos a completarem a maturação oocitária.


Unlike other domestic animals, in vitro maturation (IVM) of canine oocytes still has limited success. The aim of this study was to evaluate the effect of HEPES on the in vitro maturation of dog oocytes in anestrus and diestrus. Ovaries were collected, aseptically isolated and transported refrigerated at a temperature of 4ºC. Cumulus-oocyte complexe (COCs) from the two phases of estrous cycle were subjected to two treatments: medium TCM-199 with addition of 25 mM HEPES (TG) and medium without supplementation (CG). After 72 h of maturation, COCs were denuded, fixed and stained with Hoechst 33342 to assess nuclear maturation. The oocytes obtained from the anestrus and diestrus phases of the TG showed a higher frequency of oocytes in metaphase II (M-II) (p <0.01) stage.Comparing the different reproductive status, it was observed that the oocytes obtained from the diestrus phase presented higher rates of GVBD (germinal vesicle breakdown) and M-II. Our results showed that HEPES preserves the viability and oocyte morphology essential for the acquisition of meiotic competence, increasing the M-II rates and that the oocytes obtained from the diestrus phase are better able to complete oocyte maturation.

Resumo

O objetivo deste estudo foi obter o perfil eletroforético das proteínas séricas em éguas cíclicas e verificar as diferenças entre as fases folicular e luteal do ciclo estral nesta espécie. Foram utilizadas 18 éguas, totalizando 36 amostras de soro, sendo duas de cada égua. As amostras foram colhidas no estro e no diestro. As proteínas séricas totais foram obtidas pelo método do Biureto, a partir da utilização de Kits comerciais (LABTEST) e, as diferentes subfrações proteicas, por eletroforese em gel de poliacrilamida (SDS-PAGE). Background: Induction of ovulation is a key procedure for horse assisted reproduction technologies, such as for artificial insemination (AI) and embryo transfer. The application of hCG remains as the primary ovulation-inducing agent for horse assisted reproduction, but alternatives are in demand to avoid its adverse effects, such as loss of ovulation-inducing efficiency over multiple applications by hCC-antibody production. Despite reports on alternative ovulation-inducing agents, pair-wise comparisons of such agents under similar conditions have been limited. Under such scenario, the work was aimed to determine the efficiency of both hCG and Buserelin at inducing ovulation in Mangalarga Marchador mares raised in the Northeast of Brazil under an AI program.Materials, Methods & Results: Mares were initially selected based on their reproductive performance, the absence of clinical-reproductive alterations and adequate body condition score. Mares in diestrus were randomly distributed in three experimental conditions, received 5 mg of Dinoprost and were monitored daily for estrus detection. After estrus detection, ovaries were monitored by ultrasonography, in 12-h intervals, until the follicle reached 35 mm. At this time-point, ovulation was induced with 0.042 mg of Buserelin endovenously, with 3,000 IU hCG by an intramuscular shot, and control mares received 2 mL of saline solution, also by an intramuscular shot. Both hCG and Buserelin displayed similar efficiencies (P > 0.05) for induction of ovulation and that both agents were effective (P 0.05) from those found in the Control. All ovulations occurred within a 72-h period after treatment.[...]

O antígeno CA 15-3 é uma proteína presente no soro utilizado no acompanhamento de mulheres com câncer de mama, essencialmente na detecção de metástases. Os objetivos deste estudo foram avaliar a efetividade e a viabilidade da utilização do marcador tumoral CA 15-3 em cadelas, comparando-se os valores do marcador entre cadelas sem e com neoplasia mamária, avaliando-se alterações nos valores do marcador após a mastectomia, e suas correlações entre o tipo histológico. Foi realizada a quantificação sérica do marcador tumoral CA 15-3 (teste de eletroquimioluminescência), em vinte cadelas hígidas (grupo controle) e vinte cadelas com neoplasia mamária (grupo teste). Os animais com neoplasia tiveram a dosagem do marcador realizada antes e 10 dias após a mastectomia. Ainda, foi realiza a citologia vaginal no momento da mastectomia e foram estabelecidos três grupos de acordo com a fase estral de cada cadela, Diestro, Proestro e Anestro. As massas tumorais foram encaminhadas para exame histopatológico. A avaliação dos dados de citologia vaginal entre os grupos Diestro, Proestro e Anestro pelo teste de ANOVA não demonstrou diferença estatística significativa entre os valores encontrados. E na análise para a comparação dos valores do marcador tumoral com os tipos histológicos de neoplasias, divididas em dois grupos, benignas e malignas, utilizando o teste de Mann-Whitney Rank Sum Test, o teste não demonstrou diferença estatística significativa visto que p>0,05. Os valores encontrados do marcador no grupo controle foram uma média de 0,19+0,39 U/mL, no grupo pré-mastectomia 1,56+0,39 U/mL e pós-mastectomia 0,66+0,27 U/mL. Em análise estatística com a comparação de grupo pré e pós-mastectomia, e do grupo controle com o grupo pré e pós-mastectomia observou-se significância com p< 0,005. Assim, observou-se diferença nos valores do marcador antes e depois da remoção cirúrgica da neoplasia, sugerindo seu possível uso como controle de crescimento tumoral pós-mastectomia individual. Porém há muita variação dos resultados nos diferentes métodos existentes, e não há ainda um padrão dos valores de referência para cada método, sendo necessários mais estudos sobre o uso dos marcadores.(AU)

A citologia uterina consiste em método prático e eficiente para o diagnóstico de afecções uterinas na égua, contribuindo para a conduta terapêutica e o prognóstico quanto à fertilidade das fêmeas. Assim, o objetivo deste trabalho foi comparar diferentes técnicas de coleta de material para citologia uterina em éguas, a fim de se verificarem possíveis diferenças qualitativas e quantitativas e a praticidade na execução das metodologias. Para tanto, 12 éguas multíparas foram submetidas ao exame citológico uterino, durante o estro e o diestro, por meio de escova ginecológica e lavado uterino de baixo volume. Com esse ensaio, observou-se que não houve diferença significativa (P>0,05) no resultado da citologia uterina entre as diferentes técnicas avaliadas. No entanto, quando comparadas as fases do ciclo estral, houve uma diferença significativa (P<0,05) quanto ao percentual médio de polimorfonucleares neutrófilos, sendo de 8,07±2,80 no estro e de 1,55±0,28 no diestro. Conclui-se, desse modo, que as técnicas estudadas apresentaram resultados igualmente eficazes para detecção de neutrófilos ao exame citológico do útero em ambas as fases avaliadas do ciclo estral, apesar de, na fase de estro, a detecção de maior percentual de neutrófilos possibilitar um diagnóstico e um prognóstico mais precoces de endometrite e, portanto, uma conduta terapêutica mais adequada.(AU)


The endometrial cytology consists in an efficient and practical method for the diagnosis of uterine disorders in mares, contributing for therapeutic conduct and prognostic evaluation regarding the females' fertility. Therefore, the aim of this study was to compare different techniques of material sampling for endometrial cytology in mares, in order to verify possible qualitative and quantitative differences in the practicality of implementation of this methodology. Twelve multiparous mares underwent an endometrial cytological exam, during estrus and diestrus, by means of a cytobrush and a low-volume uterine flush. With this test, no statistical difference was observed (P>0.05) regarding the result of endometrial cytology between the evaluated techniques. However, in comparison to the estrous cycle's phases, a significant difference was verified (P<0.05) on the average percentage of polymorphonuclear neutrophils: 8.07±2.80 in estrus and 1.55±0.28 in diestrus. Thus, we conclude that the studied techniques presented equally effective results for the detection of neutrophils in the cytological exam for both evaluated stages of the estrous cycle, whereas in estrus phase, the detection of a higher percentage of neutrophils allows a more precocious diagnostic and prognostic of endometritis and, therefore, a more adequate therapeutic approach.(AU)

Resumo

A citologia uterina consiste em método prático e eficiente para o diagnóstico de afecções uterinas na égua, contribuindo para a conduta terapêutica e o prognóstico quanto à fertilidade das fêmeas. Assim, o objetivo deste trabalho foi comparar diferentes técnicas de coleta de material para citologia uterina em éguas, a fim de se verificarem possíveis diferenças qualitativas e quantitativas e a praticidade na execução das metodologias. Para tanto, 12 éguas multíparas foram submetidas ao exame citológico uterino, durante o estro e o diestro, por meio de escova ginecológica e lavado uterino de baixo volume. Com esse ensaio, observou-se que não houve diferença significativa (P>0,05) no resultado da citologia uterina entre as diferentes técnicas avaliadas. Resumo

The abortifacient efficacy of prostaglandin F2alpha products (PGF) involves induction of luteolysis,stimulation of uterine contraction and cervical dilation. PGF will induce luteolysis and depress progesterone (P4)concentrations to nearly non-detectable levels very easily after day 25 or 30 following just a 4-5 day course oftreatment. PGF can be used also earlier in pregnancy provided that the onset of cytological diestrus is timedprecisely and treatment course is longer than 5 days. With late abortion, the treatment must be continued untilverification of efficacy by ultrasound as partial abortion of litters can occur. Other uses of PGF includeshortening of diestrus in bitches, emptying the uterine content and improving semen quality in dogs. Antiprolactinagents include dopamine agonists (bromocriptine or cabergoline), and serotonin antagonists(metergoline). Anti-prolactin drugs can be used to control pseudopregnancy, induce abortion and the resumptionof estrus. Antiemetic (metoclopramide and domperidone) and antipsychotic drugs (chlorpromazine andsulpiride) have prolactin release properties which are widely documented in human medicine; their clinicalefficacy in the treatment of agalactia or hypogalactia is anecdotally reported also in bitches and queens.Aglepristone saturates P4 receptors causing abortion when administered at the dose of 10 mg/kg twice 24 hoursapart. The early administration of aglepristone at 0 to 25 days after mating is approximately 99% effective inprevention of pregnancy; however, care should be taken when using it without having assayed serum P4 or afterday 35 of pregnancy. Aglepristone can be used for several other purposes among which induction of parturitionand treatment of feline mammary hypertrophy. The clinical use of other hormonal drugs such as estrogens andsympatho-mimetics for conditions such as urinary incontinence and retrograde ejaculation is briefly discussed.(AU)
