National dairy production is characterized by significant heterogeneity in production techniques and herd and farmer profiles. Our objective was to characterize 18 dairy farms under family farming in Senador Guiomard, Acre State, from January to December 2019. Data were gathered through 255-question semi-structured forms applied to farmers in monthly visits. The data were then stored in Excel® spreadsheets and subjected to descriptive statistics. A joint analysis showed the following characteristics: farmers with low schooling level (89%); farmers of advanced age (61%); non-adoption of good dairy practices (100%); inadequate milk transport (72%); adequate brucellosis vaccination (100%); non-specialized herds (40%); pasture production system (100%); low productivity per area (873 L/ha/year) and per cow (4.0 liters/cow/day); low number of lactating cows (53%); and gross milk income (29,466.65 R$/year) representing 69% of the total gross income. The region studied has positive factors such as market guarantees, proximity to large market centers, reasonable land prices, suitable climate, ease of credit, rural education, and technical assistance. Overall, the farms are managed under a traditional production system, with low use of technology and unsatisfactory zootechnical indexes. However, the region has favorable conditions for dairy farming development.(AU)

Toxoplasma gondii is an intracellular zoonotic protozoan parasite usually infects human and animal worldwide. This study aimed to analyze the sero-prevalence of T. gondii in blood of lactating animals and human living in close proximity and also to detect Toxoplasma DNA in unpasteurized milk of the studied animals. A total of 233 blood and milk samples were collected from lactating animals, and 735 blood samples were taken from humans in District Upper Dir, Khyber Pakhtunkhwa, Pakistan. The blood samples were analyzed through ELISA while the milk samples were analyzed by PCR for the presence of T. gondii DNA. A standard questionnaire was introduced to collect the data from the participants. In animals, the reported sero-prevalence was 32.18% for IgM, 17.16% for IgG, and 6.4% for both IgM and IgG. The reported positivity for T. gondii DNA in milk was 14.44%, 34.8%, 20%, and 26% in sheep, goats, cows, and buffaloes, respectively. In the human blood samples, 9.8% were found positive for IgM and 11.2% for IgG while none of the samples was found positive for both IgM and IgG. Overall sero-prevalence reported in females was significantly higher than the male (p< 0.0001) were the significant risk factors associated with T. gondii infections in animals. In conclusion, T. gondii infection is prevalent in lactating animals and humans using their raw milk in the study area. It is suggested that raw milk should be considered as a vehicle for the transmission of T. gondii to humans. Proper pasteurization of milk is very useful in limiting the transmission of infection. Awareness and control programs should be implemented to prevent the infection.

The aim of this study was to verify the efficiency and ovulation time after the administration of different inducers for synchronization of ovulation in beef cows. One hundred and eight non-lactating cows were distributed into the control group (CG; untreated; n=28), estradiol benzoate (EB) group (EBG; n=28); 17 beta-estradiol (17ßE) group (17ßEG; n=28), and deslorelin (DES) group (DESG; n=24). On day minus 11 (D-11) of the protocol, the CG underwent application of cloprostenol and ultrasound examination (US); on D0, progesterone (P4) was inserted plus EB; on D7, cloprostenol was applied; on D9, P4 was removed and cloprostenol plus 400 IU of equine chorionic gonadotropin (eCG) was injected. The EBG was subjected to treatment identical to that of the CG, except on D10, when the cows received EB. The 17ßE was subjected to the same protocol used in the CG except for the administration of 17ßE on D10. And, the DESG was subjected to the same treatment as the CG, except on D10, when the group received DES acetate. Twelve hours after the administration of EB, 17ßE and DES, ovarian US were performed every 6 hours. The preovulatory follicle (POF) diameters measured before ovulation were 19.5; 14.7; 18.7 and 19.8 mm respectively for CG, EBG, 17ßEG and DESG; and the time intervals between inducer application and ovulation were 20.2; 18.9; 21.0 and 22.5 hours respectively. In conclusion, all ovulation inducers were efficient in promoting ovulation; the inducers caused ovulation between 18.9 and 22.5 hours; EB promoted ovulation in a shorter time (P<0.05); 17ßE and DES showed greater variation in application/ovulation time between groups.

This study was conducted to determine the nutrient status, changes in the hematological and selected blood metabolites profile of mid-lactating dairy cows during wet and dry seasons under Indonesia's tropical environment. Forty mid-lactating FH cows were selected from the Livestock Farm Complex (Dairy Community) in Yogyakarta, with an average body weight of 421.77±28.60 kg and 1st to 3rd lactation. They were housed in a semi-open housing system with concrete flooring covered with rubber mats and provided with feeders and drinkers. They were fed twice daily, and drinking water was given ad libitum. Measurements were made on the intakes of feed (and nutrients). The variables measured were nutrient intakes, blood hematology, and metabolites profile and measured over two seasons, namely wet and dry seasons. The results of the measurements made were compared using independent t-test analysis. The result indicated that intake of CP, RLYM, RNEUT, NEUT, RBC, HGB, MCV, MCHC, MPV, albumin, urea nitrogen, and total protein was differed significantly (P < 0.05) between wet and dry seasons, respectively. It was concluded that the nutrient status of mid-lactating dairy cows in the wet season was higher than during the dry season, especially the intake of CP. In the wet season, it has a positive effect on the basic hematological parameters.

Background: The intensive reproductive management in the dairy farms requires inclusion of a large group of replacement heifers in the breeding program for a shorter period. In this aspect, a creation of effective estrus synchronization protocols with timed artificial insemination (TAI) by sexed semen and optimization of the current ones have a crucial role for obtaining high pregnancy rate. These protocols are beneficial, because they led to reduced interval to first AI, lack of need for estrus detection, and allow obtaining a large group of female calves for on time. Because of limited fertilizing potential of sexed spermatozoa this type of semen is applied mainly for heifers, as fertility is higher compared to lactating cows. The main objective of the present study was to investigate the effect of ovarian status on the reproductive performance in dairy heifers subjected to estrus synchronization and timed artificial insemination with sexed semen. Materials, Methods & Results: Forty-eight Holstein healthy heifers separated in 2 groups were subjected to PGF2α-GnRH pre-treatment and Ovsynch or PRID-5-day estrus synchronization protocol and timed artificial insemination (TAI) with sex-sorted semen, staring 6 days after end of the hormonal pre-treatment. The ovarian status (presence of follicles with or not a corpus luteum) of the heifers at the begin of the treatment, on day of TAI and pregnancy rate in different groups were determined and compared. Additionally, the ovarian status at the begin of the treatment, ovarian status and size of preovulatory follicle (PF) on day of TAI and total values for both groups according to reproductive performance (pregnant or non-pregnant) were also analyzed. On day of TAI the animals with PF and a lack of corpus luteum (CL) in both groups were more that those with PF and CL (39.3% and 30% vs. 60.7 % and 70%), with significant (P < 0.05) difference in PRID-5-day group. The pregnancy rate tended to be higher in PRID-5 day than Ovsynch treatment (65% vs. 35.7%). A higher percentage (100% and 67.9%) of the pregnant animals in both treatments had not CL on day of TAI, and the size of the PF (1.58 ± 0.12 cm and 1.64 ± 0.13 cm) was increased (P < 0.05). Similar effects of the ovarian status on reproductive performance were obtained after a comparison of the total values between pregnant and non-pregnant animals. The percentage of heifers with observed PF without CL on day of TAI was significantly greater (P < 0.05) in pregnant compared to non-pregnant group (91.3% vs. 40%). The opposite dependence was determined for the parameter presence of follicles and corpus luteum (8.7% vs. 60%; P < 0.05). Furthermore, the size of the PF measured immediately before TAI was increased in animals became pregnant (1.60 ± 0.12 cm vs. 1.34 ± 0.17 cm; P < 0.05). Discussion: The analysis of the obtained results showed that the ovarian status on day of TAI affects the reproductive performance in dairy heifers subjected to estrus synchronization and timed artificial insemination with sexed semen. Hormonal pre-treatment with onset of PRID-5-day protocol 6 days later and TAI with sex-sorted semen ensure acceptable pregnancy rate. The greater preovulatory follicle and a lack of corpus luteum before insemination provide significantly (P < 0.05) more pregnant animals, compared to the cases when CL is presented. Ultrasound determination of the ovarian function before insemination can be used in selection of heifers for TAI with sex-sorted semen.

The objective of this study was to evaluate the efficacy of the antibiotic therapy associated with vaccination on the microbiological cure rate of subclinical mastitis caused by Staphylococcus aureus in lactating dairy cows. A total of five herds, from which 72 cows (120 mammary quarters - MQ) were diagnosed with S. aureus subclinical mastitis, were included in this study. Cows were randomly allocated to one of three treatment groups: a) Control (no treatment); b) ATB (antibiotic therapy); and c) ATB+VAC (antibiotic therapy plus vaccination against S. aureus). Intramammary treatment consisted of twice-daily infusion of ampicillin 75mg + cloxacillin 200mg, for 5 days. Parenteral treatment was done by injection of a single dose (7.5mg/kg) of enrofloxacin, on the first day of the treatment protocol. Vaccinated cows received three doses of a commercial vaccine 14 days before treatment (d-14), on the first day of treatment protocol (d1), and 14 days after the treatment protocol (d+14). Non-treated cows had a lower cure rate (0.06) than cows treated with ATB (0.84) and ATB+VAC (0.85). No difference in cure rate was observed between cows treated with ATB and ATB+VAC. On the other hand, vaccinated cows had lower somatic cell count (SCC) after 28 days of the treatment protocols (4.76 log10) than non-treated cows (5.37 log10). In conclusion, treatment with intramammary ampicillin and cloxacillin, associated with intramuscular enrofloxacin presented a high cure rate for SCM caused by S. aureus during lactation. The use of vaccination against S. aureus in association with antibiotic therapy did not increase the cure rate of MQ during lactation, but it was effective in reducing the SCC when compared to non-treated MQ. Although to ensure that the decrease of the SCC in ATB+VAC group was associated with the vaccination, the study should have included an additional group of only vaccinated cows, without antimicrobial therapy, with was not done in the present study, and therefore is one of the limitations of the experimental protocol used.

Infrared thermography is a non-invasive diagnostic tool for early detection of subclinical mastitis in pasture-raised dairy cows. The study was conducted on eight dairy farms, in Sena Madureira, Acre, in the Western Amazon, during the rainy season. One hundred and thirteen lactating crossbred cows were monitored, with measurements performed once day before milking, from 02:00 to 06:00 in the morning. The California Mastitis Test (CMT) was subsequently applied. Data processing was performed in the R programming language and evaluated by linear models. Our results show that cow udder surface temperature by infrared thermography showed significant differences (p<0.05) between the months of the year. Cow udder temperatures of thermographic images were significantly different (p<0.05) from CMT results for animals with subclinical mastitis. CMT results showed that 45 cows (39.8%) in January, 52 cows (46%) in February and 57 cows (50.4%) in March had subclinical mastitis. Infrared thermography is a technique that can be used for the early detection of subclinical mastitis in dairy herds in a grazing system, as it detected an increase in the udder surface temperature of the same cows that were positive for CMT.

To evaluate the bromatological composition and ruminal degradability of dry matter (DM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF) of Xaraés palisade grass (Urochloa brizantha ‘Xaraes syn Brachiaria brizantha) under grazing in integrated crop, livestock (ICL), and forest (ICLF) systems, we conducted an in situ degradability trial in randomized blocks with three non-lactating 3/4 Gyr × 1/4 Holstein cows, provided with ruminal cannula. The management of Xaraés palisade grass was similar in both systems, differing only regarding shading in the ICLF system provided by eucalyptus trees (average 65% crown cover). Grass samples were incubated for 0, 3, 6, 9, 12, 24, 36, 48, 72, and 96 hours. Considering the passage rate 2% h-1, the Xaraés palisade grass of ICL system had greater NDF effective degradability in relation to ICLF (46.38% vs 44.98%). However, the palisade grass CP potential degradability was greater in the ICLF than in the ICL system (68.92% vs. 65.40%). The presence of trees in the pasture has effect on nutritional traits of the Xaraés palisade grass, increasing its protein content and degradability and reducing its fiber degradability.(AU)

This study aimed to evaluate the ingestive behavior and body surface temperature of F1 Holstein × Zebu cows that were non-lactating in the final third of gestation and managed in two pastoral environments. Forty F1 Holstein × Zebu cows divided into two pastoral environments were evaluated: signal grass (Urochloa decumbens cv. Basilisk) deferred at the beginning of regrowth and pasture of xaraés grass (Urochloa brizantha cv. Xaraés) in vegetative growth in four periods during the day (morning, afternoon, night and dawn), following the completely randomized design in a factorial scheme. Each cow is the experimental unit. The total grazing time was 32.22% higher in the signal grass than xaraés grass (averageof 479.50 minutes). There was an interaction between the pastoral environments and the times of day on the black globe temperature and humidity index (BGHI). In the pasture of signal grass, the BGHI was 19.07% higher in the morning and afternoon periods than the average (68.95) verified during the night and dawn. F1 Holstein × Zebu cows grazing time in the final third of gestation is influenced by the pasture condition and not by the climate since they feed in a high BGHI environment.(AU)

F1 Holstein × Zebu cows grazing time in the final third of gestation is influenced by the pasture condition and not by the climate since they feed in a high BGHI environment.

The objective of this study was to evaluate productive, nutritional, and economic traits in lactating cows on pasture fed diets with different cation-anion balances (DCAB). Ten lactating ¾ Holstein × » Dairy Gyr cows in the middle third of lactation, at an average age of 70 ± 4.6 months and an average body weight of 400 ± 55.2 kg, were distributed into five treatments in a 5 × 5 Latin square experimental design with two simultaneous squares. Treatments consisted of diets with DCAB of +237, +258, +294, +347, or +419 mEq dry matter (DM). No effects of intake were observed. There was no significant effect of DCAB on milk yield. The milk protein content was not influenced by the DCAB. Body condition score was not significantly affected by the DCAB. The apparent digestibilities of dry matter and nutrients (crude protein, neutral detergent fiber, ether extract, non-fibrous carbohydrates, and total digestible nutrients) were not affected by the DCAB. Marginal rate of return did not show significant differences. Under good conditions of pasture, forage availability, and quality associated with the lactation phases of the cows, all diets were efficient in milk production, dry matter intake, and digestibility. However, in economic terms, the most attractive DCAB was +237 mEq kg-1 DM.(AU)

Summer heat stress in northwest Mexico compromises the physiological thermoregulation capacity and productive performance of lactating Holstein cows, and supplementation of minerals appears to reduce the adverse impact of heat stress in cattle. The objective herein was to evaluate the effects of an injectable mineral supplement containing phosphorus, selenium, potassium, magnesium, and copper on physiological responses, milk production, and milk composition of Holstein cows exposed to heat stress. Sixteen cows were blocked by parity and assigned to one of two treatments (n = 8) using a randomized complete block design: 1) control cows and 2) mineral-treated cows. All cows were exposed to environmental heat stress conditions (i.e., temperature-humidity index = 79.4 ± 4.3 units). No study variable was affected (P ≥ 0.20) by the treatment x sampling day interaction. While the mineral supplement did not affect any physiological variable in the afternoon, this treatment decreased breaths per min (P = 0.01) and most body surface temperatures (P ≤ 0.06; head, shoulder, leg, right-flank, and udder) in the morning. There was no effect (P = 0.37) of the mineral supplementation on milk yield but increased (P ≤ 0.03) the percentages of solids non-fat, protein, lactose, and density in the milk. In conclusion, Holstein cows' physiological thermoregulation and milk composition experiencing summer heat stress were improved by applying an injectable mineral supplement.

This study aimed to evaluate the effect of replacing dietary crude protein with increasing levels of urea on the digestive, productive and economic parameters of crossbred midlactation dairy cows under grazing conditions (Urochloa brizantha cv. Xaraés). Five crossbred cows (Holstein x Zebu) were distributed in a Latin square design (5 x 5) at five experimental periods of 21 days each. Replacement levels of dietary crude protein by urea were 0%, 7%, 14%, 21% and 28%. The intakes of dry matter, crude protein, neutral detergent fiber, and total digestible nutrients, as well as the coefficient of digestibility, were not affected by increasing levels of urea (p>0.05), except for the intake of non-fibrous carbohydrates that increased linearly with supplemental urea levels (p0.05) for cows fed by increasing levels of urea. The financial indicators (gross margin, net margin, profitability, and rate of return) were positive, exceeding operating expenses and the total operating cost. The partial replacement of dietary crude protein by urea in mid-lactation dairy cows under grazing conditions does not interfere with the productive parameters. The results of this study demonstrate that replacing 28% of dietary crude protein by urea could be done in order to reduce feeding costs without negative impact on the performance of lactating cows grazing tropical grass pastures.

This study was performed to evaluate the effects of supplementation of lactating Nelore cows with calcium salts of long-chain fatty acids (Ca-LCFA) of soybean oil on in vitro oocytes competency, cleavage rate, embryo production rate, pregnancy rate, and sex ratio of transferred embryos. Fifteen multiparous Nelore cows grazed on a Tifton 85 pasture were allotted to two treatments: (1) supplement without fat inclusion (CONT, n = 6) and (2) supplement containing 47.2 g Ca-LCFA /kg (to provide an intake of 0.2 kg Ca-LCFA/day) based on dry matter (n = 9). Animals were subjected to 2.53 ± 1.35 rounds of consecutive ovum pick up at intervals of 27.65 ± 21.72 days. Oocytes were morphologically evaluated, selected, maturated, fertilized, and cultivated for embryo production in vitro and then transferred into recipients who had previously undergone an embryo transfer protocol at a fixed time. The number of aspirated oocytes, the proportion of viable oocytes, cleavage rate, embryo percentage, pregnancy rate, and sex ratio were not affected when Nelore cows were supplemented with Ca-LCFA of soybean oil compared with non-supplemented Nelore cows. In conclusion, our results suggest that feeding a supplement containing 47.2 g Ca-LCFA/kg (to provide an intake of 0.2 kg Ca-LCFA/day) to Nelore cows does not affect in vitro embryo production in lactating Nelore cows.
