Resumo

Background: The nonsteroidal anti-inflammatory drugs (NSAIDs) exert their analgesic effect through peripheral inhibition of prostaglandin synthesis and a variety of other peripheral and central mechanisms. However, NSAIDs are associated with some adverse effects, mainly related to the gastrointestinal, renal, and hepatic systems, highlighting the need for research to develop safer drugs. Therefore, the aim of this study was to evaluate the efficacy of preoperative oral administration of carprofen or grapiprant in female cats submitted to elective ovariohysterectomy on the quality of perioperative analgesia and the need for hypnotic and analgesic drugs. Materials, Methods & Results: Thirty-three adult female cats were selected, without defined breed and healthy based on physical examination, routine laboratory analyses (complete blood count, total protein, Heinz body investigation and serum quantification of alanine transaminase [ALT], aspartate transaminase [AST], gamma glutamyl transpeptidase [GGT], alkaline phosphatase [ALP], urea, frutosamine, and glucose) and negative tests for feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV). After 3 days of adaptation, they were submitted to ovariohysterectomy by celiotomy and randomly allocated into 2 groups according to the preoperative drug used: GCAR [carprofen - 4 mg/kg, VO, 2 h before surgery; n = 11] and GGRA (grapiprant - 2 mg/kg IV, 2 h before surgery; n = 21]. The cats were pre-medicated with acepromazine 0.05 mg/ kg IV and later submitted to general anesthesia with propofol intravenously. Anesthesia was maintained with isoflurane in 100% oxygen. After anesthetic induction, a continuous infusion of remifentanil at a rate of 10 µg/kg/h was initiated. During the transanesthetic period, the parameters of heart rate; respiratory rate; systolic, mean, and diastolic arterial pressure using the oscillometric method; electrocardiogram; rectal temperature; partial pressure of CO2 at the end of expiration: and partial saturation of O2 in hemoglobin were continuously monitored. The evaluation of nociception was based on the changes in the aforementioned physiological parameters. The rate of remifentanil used did not change over time with the use of carprofen. However, animals that received grapiprant required a lower remifentanil dose at 20, 25, and 30 min during the procedure. The female cats that received carprofen showed an increase in mean heart rate at 30 min compared to that at 20 and 25 min. In the Grapiprant group, the heart rate at 35 min was higher only than that observed at 25 min. Discussion: The remifentanil rate did not differ between the groups, even between the times for GCAR. However, the remifentanil rate was lower from 20 min of the procedure for GGRA. This decrease may be related to a decrease in the need for anesthetics and analgesics by decreasing temperature, which causes decreases in metabolism and surgical stimulation. The increase in systolic, mean, diastolic, and heart rate arterial pressure parameters observed in both treatments after 15 min of anesthesia is related to the nociceptive stimulus resulting from traction and ligation of the ovarian pedicles and maneuvers for exteriorization of the uterus. These are considered the moments of greater surgical stimulus during ovariohysterectomy, evidenced by the greater release of cortisol and increase in physiological parameters. The results of this study show that the administration of carprofen or grapiprant was clinically similar when used preemptively for perioperative analgesia in cats submitted to elective ovariohysterectomy.

Although propolis has been reported for having anti-inflammatory activities, its effects on complement system has not been much studied. This research was conducted to find out the effects of Indonesian propolis on the expression levels of C3, C1r/s, Bf, MBL, and C6 in zebrafish larvae which were induced by lipopolysaccharide (LPS). Counting of macrophages migrating to yolk sac and liver histology were carried out. Larvae were divided into four groups: CON (cultured in E3 medium only), LPS (cultured in a medium containing 0.5 μg/L LPS), LPSIBU (cultured in a medium containing LPS, and then treated with 100 μg/L ibuprofen for 24 hours), and LPSPRO (cultured in a medium containing LPS, and then immersed in 14,000 μg/L propolis for 24 hours) groups. The results showed that complement gene expression in larvae from the LPSIBU and LPSPRO groups were generally lower than in larvae from the LPS group. The number of macrophage migrations to the yolk in the LPSPRO group was also lower than in the LPS group. Histological structure of liver in all groups were considered normal. This study shows that Indonesian propolis has the potential to be used as an alternative to the substitution of NSAIDs.


Embora a própolis tenha sido relatada por ter atividade anti-inflamatória, seus efeitos no sistema complemento, uma parte do sistema imunológico inato, não foram muito estudados. Esta pesquisa foi conduzida para descobrir os efeitos da própolis da Indonésia nos níveis de expressão de C3, C1r/s, Bf, MBL e C6 em larvas de peixe-zebra induzidas por lipopolissacarídeo (LPS). Foram realizadas contagens de macrófagos que migram para o saco vitelino e histologia do fígado. As larvas foram divididas em quatro grupos: CON (cultivadas apenas em meio E3), LPS (cultivadas em meio contendo 0,5 μg/L de LPS), LPSIBU (cultivadas em meio contendo LPS e, em seguida, tratadas com 100 μg/L de ibuprofeno por 24 horas) e LPSPRO (cultivado em meio contendo LPS, e então imerso em própolis 14,000 μg/L por 24 horas). Os resultados mostraram que a expressão do gene do complemento em larvas dos grupos LPSIBU e LPSPRO foi geralmente menor que em larvas do grupo LPS. O número de migrações de macrófagos para a gema no grupo LPSPRO também foi menor que no grupo LPS. A estrutura histológica do fígado em todos os grupos foi considerada normal. Este estudo mostra que a própolis indonésia tem potencial para ser utilizada como alternativa na substituição dos AINEs (anti-inflamatórios não esteroides).

Abstract Excessive intake of non-steroidal anti-inflammatory drugs such as, diclofenac sodium (DS) may lead to toxicity in the rats. In this work, we aimed to examine the protective impact of lentil extract (LE) and folic acid (FA) on the hematological markers, the kidney tissue oxidative stress and the renal function against diclofenac sodium (DS) in male albino rats. The rats (120-150 g) were divided into four equal groups randomly, the first group kept as the untreated control. The second group was administrated with DS (11.6 mg/kg b.wt. orally once/day). The third group was received DS+FA (11.6 mg/kg b.wt.+76.9 microgram/kg b.wt.) orally once/day. The fourth group was treated with DS+LE (11.6 mg/kg b.wt.+500 mg/kg b.wt.) orally once/day. After four weeks, the results revealed that DS produced a significant decrease in the values of red blood cells (RBCs), hemoglobin concentration (Hb), hematocrit (HCT) and white blood cells (WBCs). On the other hand, there was a significant increase in the platelets count. Also, DS induced a renal deterioration; this was evidenced by the significant increase in the serum levels of urea, creatinine, uric acid, Na, Ca, Mg as well as the nitric oxide (NO) level in the kidney tissue. Also, there were a significant reduction in the serum levels of potassium (K) and reduced glutathione (GSH) in the kidney homogenates. Moreover, the findings in the rats treated by DS+LE or DS+FA showed a potential protection on the hematological markers, oxidative stress in the kidney tissue and the renal function disturbed by DS. LE and FA could play a potent role for the prevention the adverse hematological, the kidney tissue oxidative stress and the renal dysfunction caused by DS via their anti-oxidative and bioactive phytochemicals.


Resumo A ingestão excessiva de anti-inflamatórios não esteroidais, como o diclofenaco de sódio (DS), pode causar toxicidade em ratos. Neste trabalho, objetivamos examinar o impacto protetor do extrato de lentilha (LE) e ácido fólico (AF) em marcadores hematológicos, no estresse oxidativo do tecido renal e na função renal contra o diclofenaco de sódio (DS) em ratos albinos machos. Os ratos (120-150 g) foram divididos em quatro grupos iguais aleatoriamente, sendo o primeiro grupo mantido como controle não tratado. O segundo grupo foi administrado com DS (11,6 mg / kg de peso corporal por via oral uma vez / dia). O terceiro grupo recebeu DS + FA (76,9 mg / kg de peso corporal por via oral uma vez / dia). O quarto grupo foi tratado com DS + LE (500 mg / kg de peso corporal por via oral uma vez / dia). Após quatro semanas, os resultados revelaram que o DS produziu uma diminuição significativa nos valores de glóbulos vermelhos (RBCs), concentração de hemoglobina (Hb), hematócrito (HCT) e glóbulos brancos (WBCs). Por outro lado, houve um aumento significativo na contagem de plaquetas. Além disso, o DS induziu uma deterioração renal; isso foi evidenciado pelo aumento significativo dos níveis séricos de ureia, creatinina, ácido úrico, Na, Ca, Mg e também do nível de óxido nítrico no tecido renal. Além disso, houve uma redução significativa nos níveis séricos de potássio (K) e glutationa reduzida (GSH) nos homogenatos renais. Além disso, os achados nos ratos tratados com DS + LE ou DS + FA mostraram uma proteção potencial sobre os marcadores hematológicos, estresse oxidativo no tecido renal e função renal perturbada pelo DS. LE e AF podem desempenhar um papel potente na prevenção do estresse hematológico adverso, do estresse oxidativo do tecido renal e da disfunção renal causada pelo DS por meio de seus fitoquímicos antioxidantes e bioativos.

Postpartum dysgalactia syndrome (PPDS) is a common disorder affecting sows in intensive production systems. In most cases, hypogalactia is not clearly identified and assumes a subclinical aspect. Therefore, the present study aimed to evaluate the effect of a nonsteroidal anti-inflammatory drug (NSAID) based on tolfenamic acid as a prophylactic treatment for PPDS and the performance of suckling piglets. Gilts (n = 319) were randomly divided into two groups: a tolfenamic acid group (n = 157) and a control (n = 162). The tolfenamic acid group received a single intramuscular injection (1 ml/20 kg of 4% tolfenamic acid) after farrowing, whereas the control group received no treatment. The occurrence of PPDS was confirmed. All piglets (n = 4,466) were weighed at 1, 4, and 18 days of age. All litters were evaluated for weight gain, the occurrence of diarrhea, and mortality between 4 and 18 days of age. PPDS variables were analyzed using logistic regression. Piglet weights were analyzed based on covariance while considering the effects of initial weight and the presence of diarrhea. Tolfenamic acid had no significant effect on the incidence of PPDS. The tolfenamic acid group had a 0.41% lower piglet mortality rate until 18 days of age. Tolfenamic acid administered prophylactically to gilts after farrowing reduced piglet mortality during lactation and promoted weight gain.


A síndrome da disgalactia pós-parto (PPDS) é uma doença comum e importante que afeta as matrizes suínas em sistemas intensivos de produção. Na maioria dos casos, a hipogalactia não é claramente identificada, assumindo um aspecto subclínico. Portanto, o presente estudo teve como objetivo avaliar o efeito de um medicamento anti-inflamatório não esteroide (AINE) baseado em ácido tolfenâmico como tratamento profiláctico para PPDS e o desempenho de leitões em aleitamento. As leitoas (n = 319) foram divididas aleatoriamente em dois grupos de tratamento: um grupo de ácido tolfenâmico (n = 157) e um grupo de controle (n = 162). O grupo tratado recebeu uma única injeção intramuscular (1 ml/20 kg de 4% de ácido tolfenâmico) após o parto, enquanto que o grupo de controle não recebeu nenhum tratamento. A ocorrência de PPDS foi então verificada. Todos os leitões (n = 4466) foram pesados com 1, 4, e 18 dias de idade. Todas as leitegadas foram avaliadas quanto ao ganho de peso, ocorrência de diarreia e mortalidade entre os 4 e 18 dias de idade. As variáveis PPDS foram analisadas através de regressão logística. Os pesos de leitões foram analisados com base na covariância, considerando os efeitos do peso inicial e a presença de diarreia. Não houve efeito significativo do ácido tolfenâmico sobre a ocorrência de PPDS. O grupo do ácido tolfenâmico teve menos 0,41% de mortalidade dos leitões até aos 18 dias de idade. O ácido tolfenâmico administrado profilaticamente nas leitoas após o parto reduziu a mortalidade dos leitões durante a lactação e aumentou o ganho de peso nos leitões.

Resumo

Apesar dos avanços na medicina equina, certas condições de claudicação ainda representam um desafio significativo para os clínicos veterinários, especialmente quando se trata de afecções distais, como feridas perfurantes no casco do equino. Essas lesões podem afetar estruturas vitais, incluindo a articulação interfalângica distal, o osso navicular, a bolsa do osso navicular, o tendão flexor digital profundo, a bainha sinovial e a terceira falange. O presente estudo tem como objetivo esclarecer que um diagnóstico mais rápido e preciso das estruturas afetadas e do grau de lesões resultantes leva a um tratamento mais eficaz. Além disso, foi demonstrado que o tratamento convencional com antibióticos e anti-inflamatórios não esteroides sistêmicos -AINEs,em combinação com terapias alternativas, como campo magnético pulsado, laser classe III e Ozonioterapia, apresentou um efeito sinérgico com resultados excelentes.(AU)


Despite advances in equine medicine, certain claudication conditions still pose a significant challenge to veterinary clinicians, especially when it comes to distal affections such as puncture wounds on the equine hoof. These injuries can affect vital structures, including the distal interphalangeal joint, navicular bone, navicular bone pouch, deep digital flexor tendon, synovial sheath, and third phalanx. The present study aims to clarify that a more rapid and accurate diagnosis of the affected structures and the resulting degree of injury leads to more effective treatment. Furthermore, it was shown that conventional treatment with antibiotics and systemic non-steroidal anti-inflammatory drugs -NSAIDs, in combination with alternative therapies, such as pulsed magnetic field, class III laser and ozone therapy, presented a synergistic effect with excellent results.(AU)


A pesar de los avances en medicina equina, ciertas afecciones claudicantes siguen planteando un reto importante a los clínicos veterinarios, especialmente cuando se trata de afecciones distales como las heridas perforantes en el casco equino. Estas lesiones pueden afectar a estructuras vitales, como la articulación interfalángica distal, el hueso navicular, la bolsa del hueso navicular, el tendón flexor digital profundo, la vaina sinovial y la tercera falange. El presente estudio pretende aclarar que un diagnóstico más rápido y preciso de las estructuras afectadas y del grado de lesión resultante conduce a un tratamiento más eficaz. Además, se demostró que el tratamiento convencional con antibióticos y antiinflamatorios no esteroideos sistémicos -AINE, en combinación con terapias alternativas, como el campo magnético pulsado, el láser de clase III y la ozonoterapia, presentaban un efecto sinérgico con excelentes resultados.(AU)

Background: Bite injuries are commonly found in small animal clinics, especially in male dogs, due to their habits of fighting and territorial disputes. In general, the lesions are treated with the use of compresses and bandages, however, it is extremely important to carefully evaluate the site, in order to carry out the appropriate treatment. In cases of tissue loss, an increasingly used practice is reconstructive surgery, which launches alternatives for treatment through flaps and skin grafts, promoting healing and satisfactory aesthetic repair. This report describes the case of a dog with a facial bite, treated with reconstructive surgery. Case: A 5-year-old male mixed-breed dog, with a body mass of 5 kg, was treated and submitted to a physical examination, due to the owner's complaint that he had been attacked by another dog, causing a bite on the face. On physical examination, the animal showed normal vital parameters, except for mild hyperthermia due to rectal temperature (39.5°C). On the face, it was possible to observe a lacerative lesion in the temporomandibular and masseteric regions, with loss of skin and muscle, in addition to having an aspect of infection and necrosis. To describe the hematological profile, a blood count was performed, showing no changes. In the biochemical tests, the present alteration was a slight increase in Alkaline Phosphatase (393.8 IU/L). Surgical treatment was indicated through facial reconstruction with a skin graft. After performing the antisepsis, the lesion was debrided, the edges were regularized and the injured tissue was curetted. For reconstruction of the defect, we used a flap obtained through a perpendicular incision at the base of the ear, in a caudal direction, and this flap was pulled and transposed over the wound. Flap synthesis was performed with 3-0 nylon thread. After surgery, a compressive bandage was performed, which was removed after 24 h. It was indicated to avoid exertion in the area and to return in 10 days for the evaluation of the surgery and removal of the stitches. Discussion: Traumatic injuries in animals are being increasingly reported, with male and non-neutered dogs being the most prevalent patients. This report is in line with other studies, with the patient's profile being a male dog, with an average age, in line with the literature. In general, in certain types of wounds, the affected animals can develop sepsis and even progress to death, however, in this case the patient did not show changes in vital parameters and in hematological tests. Reconstructive surgeries are performed with the aim of correcting skin defects and knowledge of the available techniques is important. It is also necessary to carry out a good planning, in order to avoid complications in the trans and postoperative period. In the present case, we chose to use a flap obtained through an incision perpendicular to the base of the ear, transposing it over the wound, in addition to relieving tissue tension through skin divulsion, increasing its elastic potential. The use of bandages is considered extremely important for the success of the surgical procedure, as it provides an efficient barrier against infections, trauma and adhesions. However, we emphasize the need to use ointments, gauzes and bandages concomitantly, in order to have the desired success in the postoperative period. In addition to proper surgical techniques, pain management and infection control are important for patient recovery. It is important to use non-steroidal anti-inflammatory drugs and opioids to obtain analgesia, in addition to antibiotic therapy before and after surgery. The reported case demonstrated that lesions in dogs with skin loss can be reconstructed using skin flaps, with an excellent recovery and good prognosis, as was the case with the patient in this report.

Além disso, o DS induziu uma deterioração renal; isso foi evidenciado pelo aumento significativo dos níveis séricos de ureia, creatinina, ácido úrico, Na, Ca, Mg e também do nível de óxido nítrico no tecido renal. Além disso, houve uma redução significativa nos níveis séricos de potássio (K) e glutationa reduzida (GSH) nos homogenatos renais. Além disso, os achados nos ratos tratados com DS + LE ou DS + FA mostraram uma proteção potencial sobre os marcadores hematológicos, estresse oxidativo no tecido renal e função renal perturbada pelo DS. LE e AF podem desempenhar um papel potente na prevenção do estresse hematológico adverso, do estresse oxidativo do tecido renal e da disfunção renal causada pelo DS por meio de seus fitoquímicos antioxidantes e bioativos.

Background: Squamous cell carcinoma (SCC) is a malignant neoplasm associated with prolonged exposure to ultraviolet light and lack of skin pigment in white-coated animals. The disease diagnosis is based on the animal's history, clinical signs and histopathology of the samples collected. The therapeutic alternatives are diverse. Among them, one of the most used is the metronomic chemotherapy which is based on the use of cytostatics drugs, applied in low and frequent doses for a longer period of time. One of the mechanisms of action of this type of therapy is the decrease in angiogenesis. Non-steroidal anti-inflammatory drugs (NSAIDs) are simultaneously used to maximize the angiogenic effect. The objective of this study is to report the results of a treatment accomplised with metronomic chemotherapy (cyclophosphamide with meloxicam) in a domestic feline which presented a squamous cell tumor. Case: A 7-year-old female cat, mixed breed, not neutered, weighing 3.5 kg, was assisted at the Veterinary Specialties Teaching Clinic (CEEV) of the Technical University of Machala (UTMACH), presenting bilateral ulcerative lesion in the nasal plane for 5 months. Chronic exposure to sunlight is referred to in the patient's anamnesis. Aspiration cytology performed in the patient revealed an inflammatory process with a predominance of neutrophils and macrophages, and showed absence of neoplastic cells. A treatment based on antibiotics and corticosteroids was adopted. As there were no favorable results, an excisional biopsy procedure was chosen to collect the affected tissues. The result of the histopathological analysis was a level 2 squamous cell carcinoma (SCC). The initial treatment consisted of surgical removal of the tumor and later the use of metronomic chemotherapy based on Cyclophosphamide, Meloxicam, DHA (docosahexaenoic acid) and Sucralfate for 5 months was adopted. At the beginning of the second phase of treatment, the patient developed a macula that progressively diminished. Currently, the patient does not have any recurrence of the diagnosed neoplasm. Discussion: The diagnosis of squamous cell carcinoma was based on clinical and histopathological findings. The macroscopic lesions were crusted ulcers that do not heal in the nasal plane and auricular pavilion, as reported in the literature. SCC is characterized by the infiltration of squamous cells in the dermal layer of the skin, forming epithelial islands with keratin pearls. The histopathological findings of this case present the same characteristics. The climatic conditions of Ecuador, high temperatures with greater incidence of sunlight, hypopigmented areas of the animal's body and chronic exposure to solar radiation are risk factors present in this case that foster the development of this malignant neoplasm. There are multiple treatments to be introduced in this sort of pathology. In this case, surgical removal of the tumor was adopted, followed by metronomic chemotherapy based on cyclophosphamide at 10 mg/m2 and meloxicam at a 0.05 mg/kg per day for a period of time of five months, obtaining favorable results and absence of recurrence. Despite metronomic chemotherapy being easy to administer, with fewer side effects, which includes a better acceptance of care takers who are reluctant to administer conventional chemotherapy, further studies are still necessary in what concerns certain variables such as age, carcinoma level, appropriate dosages and drugs according to each neoplasm.

Excessive intake of non-steroidal anti-inflammatory drugs such as, diclofenac sodium (DS) may lead to toxicity in the rats. In this work, we aimed to examine the protective impact of lentil extract (LE) and folic acid (FA) on the hematological markers, the kidney tissue oxidative stress and the renal function against diclofenac sodium (DS) in male albino rats. The rats (120-150 g) were divided into four equal groups randomly, the first group kept as the untreated control. The second group was administrated with DS (11.6 mg/kg b.wt. orally once/day). The third group was received DS+FA (11.6 mg/kg b.wt.+76.9 microgram/kg b.wt.) orally once/day. The fourth group was treated with DS+LE (11.6 mg/kg b.wt.+500 mg/kg b.wt.) orally once/day. After four weeks, the results revealed that DS produced a significant decrease in the values of red blood cells (RBCs), hemoglobin concentration (Hb), hematocrit (HCT) and white blood cells (WBCs). On the other hand, there was a significant increase in the platelets count. Also, DS induced a renal deterioration; this was evidenced by the significant increase in the serum levels of urea, creatinine, uric acid, Na, Ca, Mg as well as the nitric oxide (NO) level in the kidney tissue. Also, there were a significant reduction in the serum levels of potassium (K) and reduced glutathione (GSH) in the kidney homogenates. Moreover, the findings in the rats treated by DS+LE or DS+FA showed a potential protection on the hematological markers, oxidative stress in the kidney tissue and the renal function disturbed by DS. LE and FA could play a potent role for the prevention the adverse hematological, the kidney tissue oxidative stress and the renal dysfunction caused by DS via their anti-oxidative and bioactive phytochemicals.

Resumo

Lornoxicam é um anti-inflamatório não esteroidal (AINE) de uso exclusivamente humano, pois não existem doses estabelecidas nem indicações para o uso veterinário. Contudo, casos de intoxicação acidental em cães têm sido descritos. Neste estudo relata-se um caso de intoxicação acidental por lornoxicam em um cão atendido num hospital veterinário universitário. Trata-se de um cão, macho, da raça Shih Tzu, de aproximadamente 7 meses de idade, que havia ingerido 6 comprimidos de lornoxicam (Xepa®). Foram realizados exames de imagem e laboratoriais, sendo constatada gastrite medicamentosa e, posteriormente, quadro suspeito de peritonite. O animal foi tratado com antieméticos, protetores gástricos e antimicrobianos.(AU)

However, cases of accidental poisoning in dogs have been described. This work aims to report a case of accidental poisoning by lornoxicam in a dog treated at an university veterinary hospital. A male Shih Tzu dog, approximately 7 months old, who had ingested six lornoxicam tablets (Xepa®). Imaging and laboratory tests were performed, and drug gastritis was found and, later, a suspected condition of peritonitis. The animal was treated with antiemetics, gastric protectors and antimicrobials.(AU)

Resumo

O presente trabalho avaliou os efeitos analgésicos do carprofeno associado ou não à condroitina com glicosamina no tratamento de 26 cães com osteoartrite, distribuídos nos grupos: GT ­ carprofeno (Carproflan®); GTP ­ carprofeno e glicosaminoglicanos (Carproflan® e Procart®); e GC ­ somente analgésico (dipirona). A evolução dos pacientes foi classificada na escala de claudicação no dia do primeiro atendimento e aos 7, 14 e 21 dias. No GT, 60% dos animais apresentou melhora clínica a partir do 7º dia, 30% não alterou seu escore de claudicação e 10% apresentou êmese a partir do 7º dia, com suspensão da medicação. No GTP, 60% dos animais apresentou melhora clínica a partir do 7º dia, 30% não alterou seu escore de claudicação, 5% apresentou êmese a partir do 7º dia e 5% a partir do 14º dia, com suspensão da medicação. No GC, nenhum animal apresentou melhora clínica. Conclui-se que os animais submetidos ao tratamento com carprofeno apresentaram melhora na claudicação, principalmente quando associado aos glicosaminoglicanos.(AU)


The analgesic effects of carprofen associated or not with chondroitin with glucosamine were evaluated in the treatment of 26 dogs with osteoarthritis, distributed into groups: TG ­ carprofen (Carproflan®); PTG ­ carprofen and glycosaminoglycans (Carproflan® and Procart®); and CG ­ only analgesic (dipyrone). The evolution of patients was classified on the lameness scale on the first day of care and at 7, 14 and 21 days. In the TG, 60% of the animals showed clinical improvement from the 7th day, 30% did not change their lameness score and 10% had emesis from the 7th day, with discontinuation of the medication. In the PTG, 60% of the animals showed clinical improvement from the 7th day, 30% did not change their lameness score, 5% had emesis from the 7th day and 5% from the 14th day, with discontinuation of the medication. In the CG, no animal showed clinical improvement. It was concluded that animals undergoing treatment with carprofen showed improvement in lameness, especially when associated with glycosaminoglycans.(AU)

Background: Wounds that occur with tissue necrosis and that result from the application of medications through the most diverse accesses are described as drug skin medical embolism or Nicholas syndrome in human medicine, with wide description. In veterinary medicine, this subject has not yet been described extensively and specifically in veterinary medicine, especially regarding to wounds that occurred after the application of non-intravenous medications in horses, even though these lesions are recurrent in the clinical routine. This report aims to describe a case of skin necrosis in a horse, due to phenylbutazone infection. Case: A 7 year-old Mangalarga Marchador horse, weighing 400 kg, was admitted to the Veterinary Hospital for Large Animals of the Universiade Federal Rural do Rio de Janeiro (UFRRJ), with a history of phenylbutazone injection to the left side of the neck. The animal had an extensive wound on the neck and face on the left side and was characterized by the presence of cold and devitalized skin, with a hardened and parched appearance and that easily detached. During the anamnesis, a single administration of 10 mL of a non-steroidal anti-inflammatory drug based on phenylbutazone was reported intramuscularly for about 10 days to control the pain resulting from the claudication present for 14 days. The medication was administered in the region of the lateral border of the neck, on the left side. After drug administration, the animal presented an increase in volume at the application site. After 24 h, the lesion spread from the inoculation region, extending to the head and chest of the animal. During debridement, it was found that the lesion did not reach the underlying muscle tissue. In addition to the wound, the animal had upper eyelid palsy, lower lip, and auricular ptosis. Treatment with surgical debridement of devitalized tissue, topical application of ozonated sunflower oil, ketanserin, and a free skin graft was instituted. During hospitalization, the animal had a corneal ulcer in the left eye with an unfavorable prognosis due to paralysis of the upper eyelid, with enucleation of the affected eyeball. The animal was under veterinary care for 180 days and was discharged when his wound was already in an advanced stage of healing. Discussion: The history of the application of phenylbutazone intramuscularly and the location and characteristics of the lesion presented by the patient in the present report suggest that this animal presented aseptic tissue necrosis resulting from the administration of non-steroidal anti-inflammatory drugs, phenylbutazone. Although aseptic tissue necrosis, better known as Nicolau's syndrome or drug embolism cutis, is widely characterized and described in this species, there are studies in the literature that reproduce the syndrome in pigs and rabbits. Phenylbutazone was able to cause arterial damage, mainly in the tunica intima of the artery in which the medication was administered, with perivascular inflammatory infiltrate and subsequent skin necrosis at the site of administration. In addition to the skin lesion, the animal started to show signs compatible with the left facial nerve lesion, evidenced by the immobility of the upper eyelid and labial and ear ptosis. This resulted in corneal ulceration and subsequent enucleation. The animal also developed chewing difficulty in the first months of hospitalization. This dysfunction may be due to a lesion of the mandibular nerve, responsible for innervating the masticatory muscles and the oral mucosa. However, the animal showed improvement in this aspect, no longer showing this condition after 90 days of hospitalization. The treatment used was successful in healing the wound.

O aumento da população pet no Brasil causou maior proximidade da convivência de gatos com o ser humano e a falta de conhecimento do proprietário quanto às particulares felinas pode favorecer a ocorrência de toxicoses. Assim, o presente estudo apresenta as principais causas de intoxicações em gatos registrados no Hospital Veterinário (Hovet) da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo (FMVZ-USP), no período de janeiro de 2010 a dezembro de 2021. Os resultados mostraram que a frequência de ocorrência anual de casos de intoxicações em gatos variou de 0,71% (5/704) a 2,68% (9/336), e os principais agentes responsáveis por intoxicações foram os praguicidas anticolinesterásicos, como carbamatos e organofosforados (50% - 48/96), e os medicamentos, em particular, os anti-inflamatórios não esteroidais (31,25% - 30/96). Houve casos em que não foi identificada a substância envolvida (7,29% - 7/96), o que dificulta a abordagem terapêutica. Em casos de urgência, devem ser inicialmente instituídas as medidas destinadas à manutenção das funções vitais do animal, e, posteriormente, de suporte ao animal. Nestes casos, é importante a realização do diagnóstico diferencial das possíveis causas de intoxicação de acordo com os sinais e histórico clínico, bem como a colheita de amostras para análise toxicológica.(AU)


The increase in the pet population in Brazil with the greater proximity of the coexistence of cats with humans and the owner's lack of knowledge about feline particulars can favor the occurrence of toxicosis. Thus, the present study performs a retrospective survey of poisoning in cats registered by the Daily Care Records of cats received at the Veterinary Hospital (Hovet) of the School of Veterinary Medicine and Animal Science at the University of São Paulo (FMVZ-USP), from January 2010 to December 2021. The results showed that the annual occurrence of cases of poisoning in cats ranged from 0.71% (5/704) to 2.68% (9/336), and the main agents responsible for poisoning were anticholinesterase pesticides, such as carbamates and organophosphates ( 50% - 48/96), and medications, in particular, non-steroidal anti-inflammatory drugs (31,25% - 30/96). There were also cases in which it was not possible to identify the substance involved (7.29% - 7/96), which makes the therapeutic approach difficult. In these cases, it is important to make a differential diagnosis to potential cases of intoxication according to clinic signs and clinic history, in addition to collecting samples for toxicologic analysis. In urgent cases, measures must be used to maintain the animal's vital functions and, later, supportive measures for the animal.(AU)

A pitiose é uma infecção invasiva ulcerativa piogranulomatosa causada pelo oomiceto Pythium insidiosum, parasita de plantas aquáticas em águas estagnadas. Apesar de não serem raros os relatos de pitiose nas espécies domésticas, a espécie equina é a mais afetada. A enfermidade pode ser adquirida através da colonização de lesões traumáticas e do folículo piloso. Em casos dessa infecção, pode-se encontrar hifas recobertas por células necróticas, formando massas branco-amareladas semelhantes a corais, denominadas de kunkers. As lesões são localizadas prioritariamente nas extremidades distais dos membros e na porção ventral da parede tóraco-abdominal. O diagnóstico da pitiose está relacionado a um prognóstico reservado dependendo do grau de comprometimento anatômico que a enfermidade se encontra. Diante do exposto, este trabalho teve como objetivo relatar um caso de pitiose cutânea em um equino, fêmea da raça Mangalarga Machador. O animal apresentava lesões ulcerativas no membro torácico esquerdo e no membro pélvico esquerdo. O tratamento escolhido foi o cirúrgico e imunoterápico, com intuito de remover todo o tecido lesionado e, posteriormente, coletar material para realização do exame histopatológico. O equino foi submetido a um protocolo pós-operatório com a terapia antitetânica por via intramuscular. A antibioticoterapia sistêmica foi realizada com penicilina benzatina, anti-inflamatório esteroidal, dexametasona foi realizada uma vez ao dia, durante cinco dias. Posteriormente, optou-se pelo uso do anti-inflamatório não esteroidal maxicam uma vez ao dia, durante cinco dias. O animal recebeu quatro doses do imunoterápico PITIUM-VAC por via subcutânea, com intervalo de 14 dias entre uma e outra aplicação, apresentando uma resposta satisfatória ao tratamento.


Pythiosis is an invasive ulcerative pyogranulomatous infection caused by the Oomycete Pythium insidiosum, a parasite of aquatic plants in standing water. Although reports of pythiosis in domestic species are not uncommon, equine species are the most affected. The disease can be acquired through the colonization of traumatic lesions and hair follicles. Hyphae covered by necrotic cells, forming yellowish-white coral-like masses called "kunkers" can be found. The lesions are located primarily on the distal extremities of the limbs and on the ventral portion of the thoracoabdominal wall. The diagnosis of pythiosis is linked to a poor prognosis, depending on the degree of anatomical involvement of the disease. Thus, this work aimed to report a case of cutaneous pythiosis in a female Mangalarga Machador equine. The animal presented ulcerative lesions on the left thoracic limb and the left pelvic limb. The chosen treatment was surgery and immunotherapy to remove all the injured tissue and, later, collect the material for histopathological examination. The horse was submitted to a post-operative protocol with intramuscular anti-tetanus therapy. Systemic antibiotic therapy with benzathine penicillin, steroidal anti-inflammatory, dexamethasone was performed once a day for five days. Subsequently, it was decided to use the non-steroidal anti-inflammatory maxicam once a day for five days. The animal received four doses of the immunotherapy PITIUM-VAC subcutaneously, with an interval of 14 days between applications, presenting a satisfactory response to the treatment.

After analgesic administration, we evaluated general activity in the Open-Field and anxiety-like behavior in the Elevated Plus Maze of vasectomized mice. We divided C57BL/6J male mice into eight groups: saline, three non-operated control groups treated with 10 mg/kg meloxicam, 20 mg/kg tramadol, or both intraperitoneally, and four vasectomized mice groups treated with the same analgesic protocol as the control groups. One group of vasectomized mice received both treatments and an additional 10 mg/kg lidocaine at the incision site. We conducted the vasectomy via scrotal approach under isoflurane inhalation anesthesia and performed behavioral tests after full anesthesia recovery. Mice treated with meloxicam demonstrated low ambulation, spontaneous activity, and rearing frequency. Mice treated with tramadol showed spontaneous behavior compared with the saline control. Due to behavior changes demonstrated by meloxicam controls, we were unable to identify whether meloxicam provided adequate analgesia. Vasectomized mice treated with tramadol showed general activity behavior similar to their control but displayed significantly less rearing, suggesting that they were under potential signs of pain or discomfort. In conclusion, the Open Field test and the Elevated Plus Maze can usefully pre-evaluate analgesic protocols to identify possible interference caused by adverse drug effects. For future directions, an appropriate regimen of meloxicam and tramadol for enhancing mice welfare post vasectomy should be better investigated.

Background: Enostosis like lesions are characterized by areas of intramedullary sclerosis affecting the long bones andtheir presence in any cases may be not associated with lameness. It has a migratory characteristic and, therefore there isthe occurrence of lameness at different sites from the initial lesion. Its etiology is speculative and has been attributed tointraosseous increased pressure, of Havers’ canals compression, stimulation of unmyelinated fibers and circulating plateletaggregates. Diagnosis is made through nuclear scintigraphy and associated with radiographic examination. This paperaims to report a clinical case on the use of scintigraphy for the reference diagnosis of enostosis-type injury and treatmentthrough surgical bone decompression.Case: A 2-year-old thoroughbred mare, weighing 483 kg, with a history of acute lameness of the left pelvic limb associatedwith the no previous signs of trauma and no noteworthy changes in radiographic and ultrasound images, was referred to theHorse Center Veterinary Clinic. In the examination of the locomotor system, the animal presented a 2/5 degree lamenessin a straight line, with accentuated exacerbation of the same after flexion of the left femoro-tibio-patellar joint. In addition,presented a reduction in the caudal phase of the stride and croup asymmetry associated with mild myopenia. The findingsof the scintigraphic exam characterized by intense focal area of hyperconcentration of medullary radiopharmaceutical inthe proximal third of the right third metatarsal, and multiple areas of hyperconcentration in the aspect proximal to thedistal third of the left tibia. In the radiographic images, multifocal radiopaque regions that coincided with the areas ofradiopharmaceutical hyperconcentration were observed. ABSTRACT Purpose: The present study aimed at testing a new formulation of mesalazine linked to chondroitin sulfate and its components alone in the treatment of actinic proctitis in rats. Methods: Forty-seven female Wistar rats were submitted to pelvic radiation and divided into eight groups: control A, mesalazine A, chondroitin A, and conjugate A, gavage of the according substance two weeks after irradiation and sacrifice three weeks after oral treatment; control C, mesalazine C, chondroitin C, and conjugate C, sacrifice six weeks after oral treatment. The rectum was submitted to histological characterization for each of the findings: inflammatory infiltrate, epithelial degeneration, mucosal necrosis, and fibrosis. Results: The inflammatory infiltrate was more intense in chondroitin A, mesalazine A, and conjugate C. The collagen deposition was less intense in chondroitin A, and mesalazine A, and more intense in control C. Conclusions: Mesalazine and chondroitin alone were efficacious in inducing a delayed inflammatory response, hence reducing the late fibrosis. The conjugate was able to induce an ever more delayed inflammatory response.

Resumo

Dipyrone (metamizole) is well-known for its powerful effect with central and peripheral activity. This meta-analysis involved articles published between 1973 and 2021, revealing that Brazil is the country which most published scientific articles relating the use of dipyrone in dogs, and this drug is widely recommended as an analgesic to control pain in cases of postoperative and cancer. Dipyrone is one of the favorite drugs used in small animal clinic in Brazil, and 12 commercial brands are available to use in dogs at doses among 25 to 50mg/kg for oral, intravenous and intramuscular administration. The effects of dipyrone may be potentiated when used in combination with other analgesic agents such as tramadol. In several studies, the occurrence of vomiting has been observed as an adverse effect, especially when the drug is used during surgical procedures, but metamizole has presented a low potential to cause gastric ulceration. The meta-analysis study of the use of dipyrone in dogs shows the clinical importance of this drug in Brazil, being an effective and safe medication, as long as it is used in the indicated dose of 25 mg/kg.

Esta meta-nálise envolveu estudos publicados entre os anos de 1973 a 2021, revelando que o Brasil é o país que mais publicou artigos científicos envolvendo o uso de dipirona em cães, sendo este fármaco amplamente recomendado como analgésico para controlar a dor em casos de câncer e dor pós-operatória. É um dos medicamentos preferidos da clínica médica de pequenos animais no Brasil. 12 marcas comerciais estão disponíveis para uso em cães em doses que variam de 25 a 50g para administração oral, intravenosa e intramuscular. Os efeitos da dipirona podem ser potencializados quando usada em combinação com outros analgésicos, como o tramadol. Em vários estudos, a ocorrência de vômito tem sido observada como efeito adverso, principalmente quando o medicamento é usado durante procedimentos cirúrgicos, mas tem baixo potencial para causar ulceração gástrica. O estudo de meta-análise do uso de dipirona em cães evidencia a importância do uso clínico deste fármaco no Brasil, sendo um medicamento eficaz e seguro para cães, desde que utilizada na dose indicada de 25 mg / kg.
