Resumo

We assessed the extent to which CO2 levels altered different hatching and chick parameters. In Experiments 1 and 2, a total of 16,184 eggs from Cobb 500 breeders were incubated in single stage incubators under three different conditions: (a) standard ventilated incubator (CON, Exp.1 and 2); (b) increasing CO2 levels during the first 10 days of incubation until 0.7% (V7000, Exp. 1) and (c) until 0.8% (V8000, Exp. 2). High levels of CO2 improved hatchability, possibly due to lower embryo mortality from ED18 to ED21. Internal and external pipping in experiment V8000 started later than in CON; nevertheless, the hatch still occurred before in V8000 as a result of the shorter durations of external pipping and hatch. In Experiment 3, a total of 12,138 eggs from Cobb 500 were incubated in single stage incubators under three different conditions: (a) standard ventilated incubator (CON); (b) increasing CO2 levels until 1.0% with ventilation (V10000); and (c) increasing CO2 levels until 1.0% without ventilation (NV10000). Hypercapnic conditions led to better hatchability and lower embryo mortality from ED18 to ED21. Internal pipping started earlier in NV10000, but only V10000 differed from CON in terms of the average time for hatch. Hypercapnic groups also showed shorter durations of external pipping and hatch when compared to CON. Post-hatch analysis revealed no differences among incubation conditions in terms of body weight gain, feed conversion ratio, mortality by sudden death syndrome, and production factor. Nevertheless, V10000 showed a lower mortality by ascites and a better viability when compared to CON, while NV10000 presented a higher mortality by other causes. Altogether, our findings indicate that in addition to not being detrimental to embryo survival, high CO2 levels reduce embryonic mortality at 18-21 days of incubation and increase hatchability.(AU)

This study focuses on incubation parameters, egg morphometrics, and body mass development, hatching, and behavioral adaptations to heat stress within a colony of freshwater-breeding Black Skimmers (Rynchops niger) located in the private nature reserve of Serviço Social do Comércio (SESC) in the northern Pantanal, Mato Grosso, Brazil. Temperatures of nest, eggs, and surface substratum, as well as the development of embryos, were surveyed using thermal imaging, a method allowing digital recording from a distance and in a fraction of the time of traditional measuring techniques. The mean egg dimensions (n = 71) were 4.48 (± 0.13) × 3.27 (± 0.07) cm; the mean mass at hatching was 24.3 (± 1.9) g, with a significant decrease over incubation time. The mean surface temperature of eggs varied from 30.9℃ to 39.7℃, while the sand surface temperature was 20℃ at 06:00 h, rising to 47.7℃ at 11:00 h. There was a significant increase (7%) in egg surface temperature throughout incubation. Incubation-bout durations (n = 2108) were correlated with the microclimatic conditions of the substratum, becoming shorter with increasing sand-surface temperature around midday. Egg hatching lasted one day, and siblings hatched no more than 24 h apart. The mean body mass on Day 1 after hatching was 16.8 (± 1.6) g (n = 6). Three days after hatching, chicks moved to new sand depressions provided by parents near the original nest, where they remained motionless or tried to hide under riparian vegetation. The single chick that fledged had a growth rate of K = 0.117 and a t10₋90 value of 37.3 days. On Day 7, dorsal pintail feathers and primaries appeared, which were open on Day 15. After 14 days, the chick was able to regulate its body temperature, and no more feeding by parental birds during the daytime was observed. On Day 21, the immature plumage was fully developed. Fledging was completed on Day 27. Our study demonstrates that thermal imaging is a useful method of surveying egg and embryo development in the Black Skimmer, reducing nest disturbance and observation efforts.(AU)

The objective of this study was to determine the effects that breeder age has on digestive and immune system development; the transfer of immunoglobulins to egg yolk, yolk sac, and neonate chicks; and the immune response of chicks up to 35 days old. Three ages (32, 42, and 52 weeks) of Hubbard breeders were studied with ages as treatments. A total of 425 eggs were weighed for each of the three treatments and incubated. After hatching, a total of 300 1-day-old chicks were used in each treatment. We studied the development of the gastrointestinal tract and immune system of progeny and IgY transfer from breeder to progeny. Chicks from 52-week-old breeders had greater gastrointestinal tract growth up to seven days of life and greater body weight at 14 days. Older breeders (52 weeks) had higher amounts of IgY in serum and egg yolk. Chicks from the youngest breeders (32-weeks-old) had a better immune response at two weeks post-vaccination. It can be concluded that the older breeders have a greater capacity to immunize progeny up to 14 days. Strategies can be developed to increase IgY in the serum of young breeders and, consequently, increase the innate immunity of the newly-hatched chicks.(AU)

Yolk proteins undergo digestion either inside the egg yolk or in the surrounding yolk sac membrane (YSM) before being consumed by the developing avian embryo. However, the mechanisms underlying the digestion of yolk proteins during embryogenesis are largely unexplored in the pigeon Columba livia domestica. To better understand these mechanisms, the present study examined the classes of activated proteases in the egg yolk and the gene expression patterns of cathepsin B (CTSB) and cathepsin D (CTSD), which encode for lysosomal cysteine and aspartic proteases, respectively, in the YSM. We investigated the activated proteases by applying different types of protease inhibitors to yolk samples taken from incubation day 16. Then, we detected the mRNA levels of CTSB and CTSD in the YSM at incubation days 6, 8, 10, and 12-17. Both cysteine and aspartic proteases appeared to be activated in the egg yolk. Moreover, CTSB expression increased progressively and reached the maximum value on day 13; however, it decreased significantly on days 14 and 15 and further reduced toward hatching (day 17). In contrast, CTSD expression was weak and fluctuated insignificantly during development. Our results suggest that the degradation of yolk proteins at late developmental stages largely occurs in the egg yolk itself, probably by the activated cysteine and aspartic proteases. Furthermore, cathepsin B in the YSM seems to have a primary role in protein digestion, but this role decreases toward hatching.(AU)

Resumo

Niedenzuella (Tetrapterys) multiglandulosa, a vine plant found in Brazil, has been correlated to outbreaks of poisoning in cattle and buffaloes, generating economic losses related to the death due to heart failure, miscarriage, abortion, stillbirth, and neonatal mortality. The aim of this study was to examine the embryotoxic potential of the aqueous plant extract on in vitro bovine embryos. In vitro study was performed in five replicates of bovine embryo culture assigned in two groups: control, in vitro embryo culture medium without the aqueous plant extract; treated group, with addition of 2.7mg/mL of aqueous plant extract (10%) to the embryo culture on the sixth day of culture. Cleavage rate was evaluated at day 2 of the cell culture. Viability, hatchability and underdevelopment of blastocysts on the seventh, eighth, and ninth days (D7, D8, and D9, respectively) of culture were assessed under stereoscopic microscope. On day 7, blastocysts were submitted to TUNEL assay to determine apoptotic index. In vitro exposure of bovine embryos to of N. multiglandulosa resulted in reduced embryo development and survival, evaluated by dark cytoplasm indicating poor morphology and poor quality with marked reduction of hatchability. We observed a significant reduction of blastocyst production/number of cleaved embryos (60.6% vs 41.5%); reduction of blastocysts production/total number of matured bovine oocytes (35.1% vs 21.3%); and embryonic hatching rates (38.0% vs 10.0%). However, no effects were observed on the apoptotic rate. In conclusion, aqueous extract of N. multiglandulosa leaves reduces bovine embryo viability in vitro, suggesting possible detrimental effects on embryo development.(AU)


Niedenzuella (Tetrapterys) multiglandulosa, uma videira encontrada no Brasil, tem sido correlacionada a surtos de intoxicações em bovinos e búfalos, gerando perdas econômicas relacionadas à morte por insuficiência cardíaca, aborto, natimorto e mortalidade neonatal. O objetivo deste estudo foi examinar o potencial embriotóxico do extrato vegetal aquoso em embriões bovinos in vitro. O estudo in vitro foi realizado em cinco repetições de cultura de embriões bovinos distribuídos em dois grupos: controle, meio de cultura de embriões in vitro sem o extrato aquoso da planta; grupo tratado, com adição de 2,7mg / mL de extrato vegetal aquoso (10%) à cultura do embrião no sexto dia de cultivo. A taxa de clivagem foi avaliada no dia 2 da cultura de células. Viabilidade, eclodibilidade e subdesenvolvimento de blastocistos no sétimo, oitavo e nono dia (D7, D8 e D9, respectivamente) de cultura foram avaliados em microscópio estereoscópico. No dia 7, os blastocistos foram submetidos ao ensaio TUNEL para determinar o índice apoptótico. Observamos redução significativa da produção de blastocisto / número de embriões clivados (60,6% vs 41,5%); redução da produção de blastocistos / número total de oócitos bovinos maturados (35,1% vs 21,3%); e taxas de eclosão embrionária (38,0% vs 10,0%). No entanto, nenhum efeito foi observado na taxa de apoptose. Em conclusão, o extrato aquoso das folhas de N. multiglandulosa reduz a viabilidade do embrião bovino in vitro, sugerindo possíveis efeitos prejudiciais no desenvolvimento embrionário.(AU)

This paper tested a semi-natural reproduction and embryo development of Mylossoma duriventre embryo until hatching. Six females with bulging and softness coelomic cavity and eighteen males that release sperm under light belly compression were selected. Two doses of luteinizing hormone (0.0125 mg/kg and 0.125 mg/kg) were apllied for females performing a semi-natural reproduction.After fertilization the embryos were transferred to an incubator and were evaluated every 15-min intervals during the first hours of development and at one-hour intervals until hatching. The embryos are megalecites with meroblastic division, being visualized six different stages of development: fertilization, cleavage, blastula, gastrula, segmentation and pharyngula. In gastrulation (120-min post-fertilization), there is formation of "tail bud" and appearance of somites during segmentation (510- min post-fertilization), after the blastopore closure. The hatching occurred with 20 hours of development, at a temperature of 24°C. It was possible to reproduce the pacu peva in captivity and the embryonic development showed some specificity when compared to other species. The results of the reproduction and embryogenesis are needed because these are the critical phases that limit the survival of the larvae in fish farming.


Este estudo descreve a reprodução seminatural e o desenvolvimento morfológico de embriões de Mylossoma duriventre até a eclosão. Foram selecionadas seis fêmeas com protuberância e maciez de cavidade celomática e dezoito machos que espermiaram por compressão da cavidade celomática. Foram aplicadas duas doses de hormônio luteinizante (0.0125 mg/kg and 0.125 mg/kg) para fêmeas caracterizando a reprodução semi-natural. Depois da fertilização os embriões foram transferidos para a incubadora e foram avaliados em intervalos de 15 min durante a primeira hora de desenvolvimento e intervalos de uma hora até a eclosão. Os embriões são megalécitos com divisão meroblástica, sendo visualizado seis diferentes estágios de desenvolvimento: fertilização, mórula, blástula, gástrula, segmentação e faringula. Na gastrulação (120min pós fertilização), ocorre a formação do “botão da cauda” e aparecimento dos somitos durante a segmentação (510 min pós fertilização), depois o fechamento de blastóporo. A eclosão ocorreu com 20 horas de desenvolvimento à temperatura de 24ºC. É possível reproduzir pacu-peva em cativeiro e seu desenvolvimento embrionário apresenta especificidades quando comparado com outras espécies. Os resultados em relação à reprodução e a embriogênese são necessários pois, é a fase crítica que limita a sobrevivência das larvas em sistemas produtivos e para o entendimento da biologia da espécie.

Objectives of the present study were to assess the hatchery based interventions and their impact on hatching traits and subsequent performance of broilers. In total, 3000 eggs of Hubbard broiler breeders were equally divided into Small (50-55 g), Medium (56-61 g) and Large (62-70 g) eggs. The eggs from each category were distributed into 20 trays each containing 50 eggs. Each tray was considered as a replicate. At the end of the 18th day of incubation, the eggs of each weight category were divided into four different patio strategies i.e. P0, P3, P5 and P7 where feed and water were provided to the chicks for 0, 3, 5 and 7 days in the hatcher followed by conventional rearing. Data were collected for hatch window, hatching traits, growth, physical asymmetry, welfare and blood profile in broilers. Statistical analysis showed that the smaller eggs had significantly (P 0.05) shorter hatch window compared to larger and medium eggs. Hatching traits were significantly (p 0.05) better in medium eggs but the embryonic mortalities did not differ (p>0.05). The post-hatch performance was significantly (p 0.05) better in P3 birds from medium eggs. Physical asymmetry and scores of feather and gait were not affected by both treatments (p>0.05). Blood profile was also significantly (p 0.05) better in P3 birds from medium-size eggs. It was recommended that the eggs of 56-61g should be selected for better hatchability and the chicks should be provided with feed and water within the hatcher for at-least three days for optimum performance on rearing facility.(AU)

Resumo

Bisphenol A (BPA) is a monomer used in the production of polycarbonate, a polymer commonly found in plastics, epoxy resins and thermal papers. The presence of BPA in food, water, air and dust has been of great concern in recent years not only due to environmental and ecological issues but also because of its supposed risk to public health related to its mutagenic and carcinogenic potential. In this study we evaluated the toxicity of bisphenol A in zebrafish embryos (Danio rerio) and determined the 50% lethal concentration (LC50) of this chemical. BPA was used at concentrations ranging from 1 M to 100 M in E3 medium/0.5% dimethylsulfoxide (DMSO) from previously prepared stock solutions in 100% DMSO. Controls included embryos exposed only to E3 medium or supplemented with 0.5% DMSO. Camptothecin (CPT), a known inhibitor of cell proliferation was used as positive control at a concentration of 0.001 M in E3 medium/0.5% DMSO. Adults zebrafish were placed for breeding a day before the experimental set up, then, viable embryos were collected and selected for use. Experiments were carried out in triplicates, according to specifications from Organization for Economic Cooperation and Development (OECD). One embryo/well (25 embryos per concentration) was distributed in 96 well microplates in presence or absence of the chemicals. The plates were kept in BOD incubators with a controlled temperature of 28.5 ºC and with photoperiod of 14 h light:10 h dark. After 24h, 48h, 72h and 96h exposure, the exposed embryos were evaluated according to the following parameters: mortality, coagulation, rate of heartbeat, hatching and presence of morphological abnormalities. Photography was obtained by photomicroscopy. Apoptosis was evaluated by DNA ladder assay. DNA was extracted by phenol:chloroform method and analyzed by 2% agarose gel electrophoresis. DNA fragments were visualized after ethidium bromide staining in ultraviolet transilluminator. The LC50 determined for BPA was 70 M after 24 hours, 72 M after 48 hours, 47 M after 72 hours and 31 M after 96 hours exposure. BPA induced morphological and physiological alterations such as yolk sac and pericardial edema, hatching delay or inhibition, spine deformation, decreasing in heartbeat rate and mortality. In conclusion, this study demonstrated that BPA induced marked malformations in zebrafish embryos at concentrations above 25 M corroborating the current concerns related to the widespread presence of BPA in the air, food and water used by humans as well as in the bodily fluids and tissues.(AU)


Bisfenol A (BPA) é um monômero utilizado na produção de policarbonato, um polímero comumente encontrado em plásticos, resinas epóxi e papéis térmicos. A presença de BPA em alimentos, água, ar e poeira tem sido motivo de grande preocupação nos últimos anos, não só devido a questões ambientais e ecológicas, mas também ao suposto risco para a saúde pública relacionado ao seu potencial mutagênico e carcinogênico. Neste estudo avaliamos a toxicidade do bisfenol A em embriões de peixe-zebra (Danio rerio) e determinamos a concentração letal 50% (LC50) deste composto químico. O BPA foi usado na faixa de concentração entre 1 M e 100M em meio E3/0,5% de dimetilsulfóxido (DMSO), preparado a partir de soluções estoques em 100% DMSO. Os controles negativos incluíram embriões expostos apenas ao meio E3 ou suplementado com 0,5% DMSO. Camptotecina (CPT), um conhecido inibidor da proliferação celular, foi usado como controle positivo a uma concentração de 0,001 M em meio E3/0,5% DMSO. Peixes-zebra adultos foram colocados para reprodução um dia antes da montagem experimental, em seguida, embriões viáveis foram coletados e selecionados para uso. Os experimentos foram realizados em triplicata, de acordo com as especificações da Organização para Cooperação e Desenvolvimento Econômico (OCDE). Um embrião/ poço (25 embriões por concentração) foi distribuído em microplacas de 96 poços na presença ou ausência dos compostos químicos. As placas foram mantidas em incubadoras BOD com temperatura controlada de 28,5 ºC e com fotoperíodo de 14h claro:10h escuro. Após 24h, 48h, 72h e 96h, os embriões expostos foram avaliados de acordo com os seguintes parâmetros: mortalidade, presença de coagulação, taxa do batimento cardíaco, eclosão e presença de anormalidades morfológicas. Fotografias foram obtidas por fotomicroscopia. A apoptose foi avaliada pelo ensaio de DNA ladder. O DNA foi extraído pelo método fenol:clorofórmio e analisado por eletroforese em gel de agarose a 2%. Fragmentos de DNA foram visualizadas após coloração com brometo de etídio em um transiluminador ultravioleta. A LC50 determinada para o BPA foi 70 M após 24 horas, 72 M após 48 horas, 47 M após 72 horas e 31 M após exposição por 96 horas. O BPA induziu alterações morfológicas e fisiológicas como edema de saco vitelino e edema pericárdico, atraso no tempo ou inibição da eclosão, deformação da coluna vertebral, diminuição da taxa de batimentos cardíacos e mortalidade. Em conclusão, este estudo demonstrou que o BPA induziu grande número de malformações em embriões de peixe-zebra em concentrações acima de 25 M, corroborando as preocupações atuais relacionadas a presença generalizada do BPA no ar, alimento e água usados pelos seres humanos bem como nos fluidos e tecidos corporais.(AU)

Resumo

Birds develop thermoregulatory control during the last days of incubation. Different temperature conditioning programs have been proposed to prepare broiler chicks for post-hatch life. This study aimed to investigate the effects of short-term temperature (STT) stimulation and breeder age on hatching performance, embryo development, yolk absorption, and post-hatch performance of male and female broilers. A 2 × 2 factorial completely randomized design was used, with two breeder ages (30 and 60 wk), two temperature incubation programs (control and STT), and 2,520 eggs per treatment. Eggs were distributed in two large-scale commercial incubators with a capacity of 120,960 eggs. The control group was subjected to a standard single-stage incubation program (37.2-37.4 °C), whereas the STT group was subjected to a temperature increment of 1 °C for 4 h on embryonic days 16, 17, 18, and 19. Embryos were analyzed at 16 and 19 days of incubation, and chicks at hatch. At 19 days of incubation, STT conditioning resulted in lower yolk sac weight in embryos from 60-wk-old breeders and higher relative weight of the gastrointestinal tract in embryos from 30-wk-old breeders. At hatch, males had lower residual yolk weight, females had greater length, and chicks from 60-wk-old breeders subjected to STT had higher body weight and relative weight of the gastrointestinal tract. Eggs from 30-wk-old breeders showed higher fertility and hatchability and lower infertility, mortality, and second-grade chick percentages. Regarding performance, it was found that males had higher body weight from 7 to 42 days of age and higher whole leg yields. Females, on the other hand, had higher fat yields. Breeder age and sex influenced gastrointestinal tract development, carcass yield, and performance. A 1 °C increase in incubation temperature for 4 h from days 16 to 19 of incubation affected yolk absorption and digestive tract development in chicks from 60-wk-old breeders without, however, influencing performance results.(AU)


As aves desenvolvem o controle do sistema termorregulatório durante os últimos dias de incubação, dessa forma, diferentes programas temperatura de incubação têm sido descritos para preparar os pintos de corte para a vida pós-eclosão. Objetivou-se nesse estudo, estabelecer os efeitos de alterações curtas na temperatura de incubação e da idade da matriz no desempenho de eclosão, desenvolvimento embrionário, absorção da gema e desempenho pós-eclosão de frangos de corte machos e fêmeas. O delineamento experimental foi inteiramente casualizado em fatorial 2 x 2 com 2 idades de matriz (30 e 60 semanas) e 2 programas de incubação de temperatura, controle (CT) e grupo de treinamento de temperatura de curto prazo (STT) e 2.520 ovos em cada tratamento. Os ovos foram distribuídos em 2 incubadoras comerciais de grande escala com capacidade para 120.960 ovos. O grupo CT seguiu um programa de incubação de estágio único padrão (37,2-37,4°C) e o STT teve um incremento em + 1 ° C por 4 hs nos dias 16, 17, 18 e 19 de incubação. Os embriões foram analisados aos 16 e 19 dias e pintinhos na eclosão. Aos 19 dias de incubação, embriões de matrizes com 60 semanas apresentaram menor peso do saco vitelino e embriões de matrizes com 30 semanas apresentaram maior porcentagem de trato gastrointestinal quando em STT. Na eclosão, os machos apresentaram menor vitelo residual, e as fêmeas apresentaram maior comprimento, pintinhos mais pesados e maior trato gastrointestinal quando originárias de matrizes de 60 semanas de idade e expostos ao STT. Na eclosão, os ovos com 30 semanas de idade apresentaram maior porcentagem de fertilidade e eclodibilidade e menor infertilidade, mortalidade e pintos de secunda categoria. No desempenho, os machos apresentaram maior peso corporal do 7° ao 42° dia e maiores valores de porcentagem de pernas e as fêmeas maiores valores de porcentagem de gordura. A idade das matrizes e o sexo influenciaram o desenvolvimento visceral, o rendimento de carcaça e o desempenho dos frangos de corte. O aumento de 1°C durante 4 horas entre os dias 16 e 19 de incubação afetou a absorção da gema e o trato digestivo em pintos de matrizes com 60 semanas de idade, sem refletir nos resultados de desempenho.(AU)

Bisphenol A (BPA) is a monomer used in the production of polycarbonate, a polymer commonly found in plastics, epoxy resins and thermal papers. The presence of BPA in food, water, air and dust has been of great concern in recent years not only due to environmental and ecological issues but also because of its supposed risk to public health related to its mutagenic and carcinogenic potential. In this study we evaluated the toxicity of bisphenol A in zebrafish embryos (Danio rerio) and determined the 50% lethal concentration (LC50) of this chemical. BPA was used at concentrations ranging from 1 µM to 100 µM in E3 medium/0.5% dimethylsulfoxide (DMSO) from previously prepared stock solutions in 100% DMSO. Controls included embryos exposed only to E3 medium or supplemented with 0.5% DMSO. Camptothecin (CPT), a known inhibitor of cell proliferation was used as positive control at a concentration of 0.001 µM in E3 medium/0.5% DMSO. Adults zebrafish were placed for breeding a day before the experimental set up, then, viable embryos were collected and selected for use. Experiments were carried out in triplicates, according to specifications from Organization for Economic Cooperation and Development (OECD). One embryo/well (25 embryos per concentration) was distributed in 96 well microplates in presence or absence of the chemicals. The plates were kept in BOD incubators with a controlled temperature of 28.5 ºC and with photoperiod of 14 h light:10 h dark. After 24h, 48h, 72h and 96h exposure, the exposed embryos were evaluated according to the following parameters: mortality, coagulation, rate of heartbeat, hatching and presence of morphological abnormalities. Photography was obtained by photomicroscopy. Apoptosis was evaluated by DNA ladder assay. DNA was extracted by phenol:chloroform method and analyzed by 2% agarose gel electrophoresis. DNA fragments were visualized after ethidium bromide staining in ultraviolet transilluminator. The LC50 determined for BPA was 70 µM after 24 hours, 72 µM after 48 hours, 47 µM after 72 hours and 31 µM after 96 hours exposure. BPA induced morphological and physiological alterations such as yolk sac and pericardial edema, hatching delay or inhibition, spine deformation, decreasing in heartbeat rate and mortality. In conclusion, this study demonstrated that BPA induced marked malformations in zebrafish embryos at concentrations above 25 µM corroborating the current concerns related to the widespread presence of BPA in the air, food and water used by humans as well as in the bodily fluids and tissues.


Bisfenol A (BPA) é um monômero utilizado na produção de policarbonato, um polímero comumente encontrado em plásticos, resinas epóxi e papéis térmicos. A presença de BPA em alimentos, água, ar e poeira tem sido motivo de grande preocupação nos últimos anos, não só devido a questões ambientais e ecológicas, mas também ao suposto risco para a saúde pública relacionado ao seu potencial mutagênico e carcinogênico. Neste estudo avaliamos a toxicidade do bisfenol A em embriões de peixe-zebra (Danio rerio) e determinamos a concentração letal 50% (LC50) deste composto químico. O BPA foi usado na faixa de concentração entre 1 µM e 100µM em meio E3/0,5% de dimetilsulfóxido (DMSO), preparado a partir de soluções estoques em 100% DMSO. Os controles negativos incluíram embriões expostos apenas ao meio E3 ou suplementado com 0,5% DMSO. Camptotecina (CPT), um conhecido inibidor da proliferação celular, foi usado como controle positivo a uma concentração de 0,001 µM em meio E3/0,5% DMSO. Peixes-zebra adultos foram colocados para reprodução um dia antes da montagem experimental, em seguida, embriões viáveis foram coletados e selecionados para uso. Os experimentos foram realizados em triplicata, de acordo com as especificações da Organização para Cooperação e Desenvolvimento Econômico (OCDE). Um embrião/ poço (25 embriões por concentração) foi distribuído em microplacas de 96 poços na presença ou ausência dos compostos químicos. As placas foram mantidas em incubadoras BOD com temperatura controlada de 28,5 ºC e com fotoperíodo de 14h claro:10h escuro. Após 24h, 48h, 72h e 96h, os embriões expostos foram avaliados de acordo com os seguintes parâmetros: mortalidade, presença de coagulação, taxa do batimento cardíaco, eclosão e presença de anormalidades morfológicas. Fotografias foram obtidas por fotomicroscopia. A apoptose foi avaliada pelo ensaio de DNA ladder. O DNA foi extraído pelo método fenol:clorofórmio e analisado por eletroforese em gel de agarose a 2%. Fragmentos de DNA foram visualizadas após coloração com brometo de etídio em um transiluminador ultravioleta. A LC50 determinada para o BPA foi 70 µM após 24 horas, 72 µM após 48 horas, 47 µM após 72 horas e 31 µM após exposição por 96 horas. O BPA induziu alterações morfológicas e fisiológicas como edema de saco vitelino e edema pericárdico, atraso no tempo ou inibição da eclosão, deformação da coluna vertebral, diminuição da taxa de batimentos cardíacos e mortalidade. Em conclusão, este estudo demonstrou que o BPA induziu grande número de malformações em embriões de peixe-zebra em concentrações acima de 25 µM, corroborando as preocupações atuais relacionadas a presença generalizada do BPA no ar, alimento e água usados pelos seres humanos bem como nos fluidos e tecidos corporais.

The potential of silicon (Si) to control nematodes is often reported in association with the resistance induction mechanism. The objective of this work was to evaluate the hypothesis of resistance induction in plants against nematodes through the application of Si. In vitro experiments were carried out to evaluate the effect of Si and potassium chloride (KCl) on the hatching and mortality of Meloidogyne paranaensis juveniles. For the purpose of a greenhouse experiment, tomato plants were used as a model for bifurcated roots, using the respective control treatments, KCl and distilled water. Ten days after treatment, 2,000 eggs of M. paranaensis were inoculated. At 55 days post-inoculation, the reproduction factor and number of nematodes per gram of root were evaluated. Additionally, the tissue nutrient concentration and gas exchange variables were analyzed. Silicon reduces nematode viability, when in direct contact with the nematode in in vitro experiments and in bifurcated plants. When plants were treated with Si on one side of the root, no nematode control was observed on the opposite corresponding side. Results suggested that Si has a positive effect on the reduction of M. paranaensis population, but probably induced resistance to nematode only through direct action.

This study was designed to determine adequate levels for sodium butyrate inclusion in pre-hatching and pre-starter feed in order to minimize the negative effects of post-hatch delayed placement on broiler chicks. Newly-hatched chicks were allotted in a completely randomized design, with five treatments, each comprising five replicates of ten birds each. Five dietetic levels of sodium butyrate (control, 0.050, 0.075, 0.100 and 0.200%) were used in the pre-starter feed offered to the chicks in the transporting box and during the pre-starter phase. Performance, yolk sac retraction, plasma glucose concentration, weight and histomorphometry of the small intestine were evaluated after 24 hours of feed access and at 7 days of age. A metabolic trial was performed when the chicks were seven to ten days of age. Supplementation of 0.1% sodium butyrate increased the development of broiler chicks' intestinal villi at seven days of age but was not able to improve performance in the pre-starter phase. Supplementation with sodium butyrate in a diet offered in the transportation box does not improve broiler performance in the pre-starter phase. Supplementation of up to 0.16% is recommended, in order to improve the metabolizability of the ether extract for broilers at seven days of age.(AU)


Objetivou-se, com este estudo, encontrar níveis adequados de inclusão de butirato de sódio em ração pós-eclosão e pré-inicial, buscando minimizar os efeitos negativos do jejum de pintos de corte. Os animais foram distribuídos em delineamento inteiramente ao acaso, com cinco tratamentos e cinco repetições de 10 aves cada. Foram utilizadas cinco suplementações de butirato de sódio (controle; 0,05; 0,075; 0,100 e 0,200%) na ração pré-inicial, fornecidas aos pintos na caixa de transporte e durante a fase pré-inicial. Após 24 horas de acesso ao alimento e aos sete dias de idade, foram avaliados desempenho, retração do saco vitelino, concentração de glicose plasmática, peso e histomorfometria do intestino delgado. De sete a 10 dias de idade, realizou-se ensaio de metabolizabilidade. A suplementação de 0,10% de butirato de sódio aumentou o desenvolvimento das vilosidades intestinais dos pintos de corte aos sete dias de idade, mas não foi capaz de melhorar o desempenho na fase pré-inicial. A suplementação com butirato de sódio em dieta oferecida na caixa de transporte não melhora o desempenho dos frangos na fase pré-inicial. Recomenda-se suplementação de até 0,16%, a fim de melhorar a metabolizabilidade do extrato etéreo para frangos de corte aos sete dias de idade.(AU)

The hatchery is one of the most important segments of the poultry chain, and generates an abundance of data, which, when analyzed, allow for identifying critical points of the process . The aim of this study was to evaluate the applicability of the data mining technique to databases of egg incubation of broiler breeders and laying hen breeders. The study uses a database recording egg incubation from broiler breeders housed in pens with shavings used for litters in natural mating, as well as laying hen breeders housed in cages using an artificial insemination mating system. The data mining technique (DM) was applied to analyses in a classification task, using the type of breeder and house system for delineating classes. The database was analyzed in three different ways: original database, attribute selection, and expert analysis. Models were selected on the basis of model precision and class accuracy. The data mining technique allowed for the classification of hatchery fertile eggs from different genetic groups, as well as hatching rates and the percentage of fertile eggs (the attributes with the greatest classification power). Broiler breeders showed higher fertility (> 95 %), but higher embryonic mortality between the third and seventh day post-hatching (> 0.5 %) when compared to laying hen breeders eggs. In conclusion, applying data mining to the hatchery process, selection of attributes and strategies based on the experience of experts can improve model performance.

The main objective of this study was to evaluate the effect of placement time on the performance of broiler chickens and the development of their gastrointestinal tract. Two methodologies for measuring broiler performance were compared, one considering day of pulling as the first day, the other considering day of placement as the first day. A total of 1,056 one-day old male Cobb® 500 broiler chicks were subjected to treatments of different placement times after pulling from the hatchery: 3, 24, 48, and 72 h. The studied traits were: feed intake, body weight, feed conversion, viability, and gastrointestinal tract development. When day of pulling was considered the first day, feed intake and body weight at 39 days decreased as placement time increased. However, when day of placement was considered the first day, fasting up to 72 h did not have any negative effect on broiler performance at 39 days post-placement. Placement time did not affect yolk sac utilization or liver weight. At nine days post-placement, weights of gizzard + proventriculus, pancreas, and small intestine increased with increasing placement time. At seven days of age, there was no effect of placement time on villus height or crypt depth. It is possible to place broiler chicks up to 72 h post-hatching with no negative effects when day of placement is considered the first day for evaluating broiler performance.(AU)

During the development of bird embryos, thenutrients are stored within the egg, but carbohydrate supplementation isunsatisfactory upon hatching. Poor carbohydrate supplementation candamage the embryo development and negatively affect poultry weight. Inovo feeding is a promising technique to improve the quality of newly hatchedchicks as it uses systems for feeding the embryos by inoculating nutrientsinto the egg amniotic fluid. However, further studies are needed in order toimprove this technique and create effective implementations and protocolsthat can be widely used in the industry. This study aims to both review theliterature on the use of in-ovo feeding and understand the perspectives ofits use in the broiler production chain.

Nas aves, os nutrientes utilizados durante o desenvolvimento embrionário estão contidos no ovo, sendo escassa a reserva de carboidratos logo após a eclosão do pintainho. Essa restrição pode causar prejuízos ao desenvolvimento embrionário, impactando negativamente no peso das aves. Diante disso, a nutrição in ovo mostra-se como uma técnica promissora, que visa prover nutrientes ao embrião por meio da inoculação de soluções com efeitos benéficos ao seu desenvolvimento. Contudo, são necessários mais estudos no intuito de aprimorar a técnica e criação de protocolos efetivos que possam ser amplamente utilizados pela indústria. Nesse contexto, o objetivo desta revisão de literatura foi abordar e depreender as perspectivas da utilização da técnica nutrição in ovo na cadeia produtiva de frangos de corte.
