Climate change has caused major changes in abiotic factors, with water stress as the greatest threat to agricultural production. The measures aimed at alleviating the problems caused by this limiting production factor have occurred through the adoption of sustainable strategies, especially microbial biotechnology, which uses the interactions between the microorganism and the plant, ensuring productive quality and inducing plant resistance to stresses biotic and abiotic. The objective of the present work was to evaluate the biological nitrogen fixation and the development of bean seedlings, with co-inoculation of two types of inoculants, which were subjected to water stress by different pot capacities. The experiment was conducted in a greenhouse, at Universidade Paranaense - UNIPAR, from April to June 2019. The experimental design was completely randomized (DIC), with 5 replications, 16 treatments and 80 experimental units. The cultivar used was SCS Riqueza. The parameters evaluated were pot capacity (25%, 50%, 75% and 90%); small, large and total nodules, shoot and root length, dry and fresh weight, total carbon and nitrogen. The evaluation of the morphological parameters of the bean seedlings indicated that the co- inoculation technique promoted beneficial effects for the dry mass parameters of shoot, nodule and root. The analysis of the percentage of carbon and nitrogen in the tissues of the seedlings provided an increase in the concentration of these elements in treatments that involved co-inoculation (Azospirillum brasilensis and Rhizobium tropici) with pot capacities of 25 and 75% (CV), demonstrating that the association of microorganisms is beneficial in the limiting water situation.(AU)


A mudança climática tem causado grandes mudanças nos fatores abióticos, sendo o estresse hídrico a maior ameaça à produção agrícola. As medidas destinadas a aliviar os problemas causados por este fator limitante de produção ocorreram através da adoção de estratégias sustentáveis, especialmente a biotecnologia microbiana, que utiliza as interações entre o microorganismo e a planta, garantindo a qualidade produtiva e induzindo a resistência da planta ao estresse biótico e abiótico. O objetivo do presente trabalho foi avaliar a fixação biológica de nitrogênio e o desenvolvimento de mudas de feijão, com co-inoculação de dois tipos de inoculantes, que foram submetidos ao estresse hídrico por diferentes capacidades de vaso. A experiência foi realizada em uma estufa, na Universidade Paranaense - UNIPAR, de abril a junho de 2019. O projeto experimental foi completamente randomizado (DIC), com 5 réplicas, 16 tratamentos e 80 unidades experimentais. A cultivar utilizada foi a SCS Riqueza. Os parâmetros avaliados foram a capacidade do vaso (25%, 50%, 75% e 90%); nódulos pequenos, grandes e totais, comprimento do rebento e da raiz, peso seco e fresco, carbono total e nitrogênio. A avaliação dos parâmetros morfológicos das mudas de feijão indicou que a técnica de co-inoculação promoveu efeitos benéficos para os parâmetros de massa seca do turião, nódulo e raiz. A análise da porcentagem de carbono e nitrogênio nos tecidos das mudas proporcionou um aumento na concentração destes elementos nos tratamentos que envolveram a co-inoculação (Azospirillum brasilensis e Rhizobium tropici) com capacidades de vaso de 25 e 75% (CV), demonstrando que a associação de microorganismos é benéfica na situação limite da água.(AU)


El cambio climático ha provocado importantes cambios en los factores abióticos, siendo el estrés hídrico la mayor amenaza para la producción agrícola. Las medidas encaminadas a paliar los problemas causados por este factor limitante de la producción se han producido mediante la adopción de estrategias sostenibles, especialmente la biotecnología microbiana, que utiliza las interacciones entre el microorganismo y la planta, asegurando la calidad productiva e induciendo la resistencia de la planta a los estreses bióticos y abióticos. El objetivo del presente trabajo fue evaluar la fijación biológica de nitrógeno y el desarrollo de plántulas de frijol, con la co-inoculación de dos tipos de inoculantes, que fueron sometidos a estrés hídrico por diferentes capacidades de maceta. El experimento se realizó en un invernadero, en la Universidade Paranaense - UNIPAR, de abril a junio de 2019. El diseño experimental fue completamente al azar (DIC), con 5 repeticiones, 16 tratamientos y 80 unidades experimentales. El cultivar utilizado fue SCS Riqueza. Los parámetros evaluados fueron capacidad de maceta (25%, 50%, 75% y 90%); nódulos pequeños, grandes y totales, longitud de brotes y raíces, peso seco y fresco, carbono y nitrógeno total. La evaluación de los parámetros morfológicos de las plántulas de frijol indicó que la técnica de coinoculación promovió efectos beneficiosos para los parámetros de masa seca de brotes, nódulos y raíces. El análisis del porcentaje de carbono y nitrógeno en los tejidos de las plántulas proporcionó un aumento en la concentración de estos elementos en los tratamientos que involucraron la coinoculación (Azospirillum brasilensis y Rhizobium tropici) con capacidades de maceta de 25 y 75% (CV), demostrando que la asociación de microorganismos es beneficiosa en la situación de agua limitante.(AU)

The objective of this study was to evaluate the agronomic efficiency of rhizobia strains already approved or in the selection stage for cowpea, in the Recôncavo da Bahia region, Brazil. Two field experiments were performed in the municipalities of Cruz das Almas and Maragogipe, Bahia, Brazil. A randomized block design was used with seven nitrogen sources and four replicates. Nitrogen sources consisted of the strains UFRB FA51B1, UFRB BA72C2-1, UFLA 03-164T, UFLA 03-84, INPA 03-11B, and two controls without inoculation, one with mineral nitrogen and another without. All the strains nodulated cowpea. The efficiency of the strains was determined by the number of nodules, nodule dry matter, total dry matter, grain yield, nitrogen accumulation in shoots and grain, and relative efficiency. The strain UFLA 03-164T can be recommended for biomass production, green manure, and promotion of grain yields in both soils. In Maragogipe, the UFLA 03-84 and INPA 03-11B strains can be recommended for biomass production, green manure, and promotion of grain yields. The UFLA 03-164T strain showed great potential to promote and grain yield in the two municipalities studied. The strain INPA 03-11B can be recommended for Maragogipe soil. UFLA 03-84 can also be recommended to increase grain yield in Cruz das Almas.

Glutamine synthetase (GS), encoded by glnA, catalyzes the conversion of L-glutamate and ammonium to L-glutamine. This ATP hydrolysis driven process is the main nitrogen assimilation pathway in the nitrogen-fixing bacterium Azospirillum brasilense. The A. brasilense strain HM053 has poor GS activity and leaks ammonium into the medium under nitrogen fixing conditions. In this work, the glnA genes of the wild type and HM053 strains were cloned into pET28a, sequenced and overexpressed in E. coli. The GS enzyme was purified by affinity chromatography and characterized. The GS of HM053 strain carries a P347L substitution, which results in low enzyme activity and rendered the enzyme insensitive to adenylylation by the adenilyltransferase GlnE.


A glutamina sintetase (GS), codificada por glnA, catalisa a conversão de L-glutamato e amônio em L-glutamina. Este processo dependente da hidrólise de ATP é a principal via de assimilação de nitrogênio na bactéria fixadora de nitrogênio Azospirillum brasilense. A estirpe HM053 de A. brasilense possui baixa atividade GS e excreta amônio no meio sob condições de fixação de nitrogênio. Neste trabalho, os genes glnA das estirpes do tipo selvagem e HM053 foram clonados em pET28a, sequenciados e superexpressos em E. coli. A enzima GS foi purificada por cromatografia de afinidade e caracterizada. A GS da estirpe HM053 possui uma substituição P347L que resulta em baixa atividade enzimática e torna a enzima insensível à adenililação pela adenililtransferase GlnE.

The objective of this work was to verify the influence of light environments combined with rhizobia inoculation on cowpea growth and productivity. A completely random design was used in a 4x4 factorial scheme, with four light environments, four nitrogen sources and eight replicates in split plot parcels. Light environments were set by means of photo-conversion and thermo-conversion nettings (Aluminet®, red net and black net) and control treatment without shading (full sun). Nitrogen sources were constituted by the strains INPA 03-11B - SEMIA 6462 (Bradyrhizobium elkanni) and UFLA 03-84 - SEMIA 6461 (Bradyrhizobium viridifuturi), and two control treatments: with 70 kg ha-1 of mineral nitrogen and without N. Plant height, indices of chlorophyll a, b and total chlorophill, the number of leaves, number of nodules, dry matter of nodules, dry matter of the aerial portion, dry matter of roots and total dry matter, relative efficiency, gathering of nitrogen in the aerial portion, number, length and matter of pods per plant and dry matter of 100 grains, were evaluated. . There was interaction between light conditions and nitrogen sources for the number of nodules. Individual effect was observed in all other variables. Strain INPA 03-11B was able to promote higher nodulation in cowpea plants in light environments under full sun and Aluminet and the strain UFLA 03-84 only under full sun conditions. However, the efficiency of diazotrophic bacteria to promote vegetative growth, nitrogen gathering and production was not influenced by different light environments. Thus, full sun cultivation is recommended, independently of the nitrogen source used.

Azotobacter vinelandii is a gram-negative soil bacterium that produces two biopolymers of biotechnological interest, alginate and poly(3-hydroxybutyrate), and it has been widely studied because of its capability to fix nitrogen even in the presence of oxygen. This bacterium is characterized by its high respiration rates, which are almost 10-fold higher than those of Escherichia coli and are a disadvantage for fermentation processes. On the other hand, several works have demonstrated that adequate control of the oxygen supply in A. vinelandii cultivations determines the yields and physicochemical characteristics of alginate and poly(3-hydroxybutyrate). Here, we summarize a review of the characteristics of A. vinelandii related to its respiration systems, as well as some of the most important findings on the oxygen consumption rates as a function of the cultivation parameters and biopolymer production.

The effect of management (ecological and conventional) on functional groups of microorganisms of soil in agroecosystems with different resilience scores reported to climate variability in Anolaima, Colombia was evaluated. Were found clustering associated with management and cellulolytic bacteria and fungi abundances. No differences found in diversity of phosphate solubilizing or nitrogen-fixing microorganisms, related to management. The diversity of microbial functional groups was affected by the climatic condition of sampling season. Management was relevant in relationships between resilience scores to climate variability and cellulolytic microorganisms; in ecological agroecosystems, biodiversity knowledge, agroecological main structure, and the participation of farmers in organizations were important.

Brazil nut is a very important nontimber forest product in the Amazon region. Propagation of this tree still represents a challenge due to slow and uneven seed germination. In this context, plant growth-promoting bacteria can facilitate the process of propagation. The aims of this study were to isolate and characterize endophytic bacteria from the roots of Brazil nut trees in native terra firme forest and cultivation areas in northern Brazil, and to identify mechanisms by which bacteria act in plant growth promotion. Overall, 90 bacterial isolates were obtained from the roots of Brazil nut trees in monoculture, agroforestry and native forest areas by using different semisolid media. The isolates were characterized by sequencing the 16S rRNA gene. Plant growth-promoting characteristics were evaluated by the presence of the nifH gene, aluminum phosphate solubilization and the production of indole compounds. The isolates were affiliated with 18 genera belonging to 5 different classes (α-Proteobacteria, ß-Proteobacteria, γ-Proteobacteria, Bacilli and Actinobacteria). The genus Bacillus was predominant in the forest and monoculture areas. Fourteen isolates presented the nifH gene. Most of the bacteria were able to solubilize aluminum phosphate and synthetize indole compounds. The results indicated high diversity of endophytic bacteria present among the roots of Brazil nut trees, mainly in the agroforestry area, which could be related to soil attributes. Among the 90 isolates, the 22 that presented the best results regarding plant growth promotion traits were good candidates for testing in seedling production of Brazil nut trees. (AU)

ABSTRACT To mitigate the deleterious effects of abiotic stress, the use of plant growth-promoting bacteria along with diazotrophic bacteria has been increasing. The objectives of this study were to investigate the key enzymes related to nitrogen and carbon metabolism in the biological nitrogen fixation process and to elucidate the activities of these enzymes by the synergistic interaction between Bradyrhizobium and plant growth-promoting bacteria in the absence and presence of salt stress. Cowpea plants were cultivated under axenic conditions, inoculated with Bradyrhizobium and co-inoculated with Bradyrhizobium sp. and Actinomadura sp., Bradyrhizobium sp. and Bacillus sp., Bradyrhizobium sp. and Paenibacillus graminis, and Bradyrhizobium sp. and Streptomycessp.; the plants were also maintained in the absence (control) and presence of salt stress (50 mmolL-1 NaCl). Salinity reduced the amino acids, free ammonia, ureides, proteins and total nitrogen content in nodules and increased the levels of sucrose and soluble sugars. The co-inoculations responded differently to the activity of glutamine synthetase enzymes under salt stress, as well as glutamate synthase, glutamate dehydrogenase aminating, and acid invertase in the control and salt stress. Considering the development conditions of this experiment, co-inoculation with Bradyrhizobium sp. and Bacillus sp. in cowpea provided better symbiotic performance, mitigating the deleterious effects of salt stress.

The adoption of cover crops in commercial plantations has been shown to be an alternative to promote greater soil protection, enhancement in its structure and aggregation, optimization of accumulation and nutrient cycling to obtain greater production of biomass and higher nitrogen fixation. To determine the accumulation of micronutrients of Pueraria phaseoloides L., according to age, established as a soil cover in industrial oil palm crops, an experiment was carried out at Companhia Agroindustrial (CRAI) in the municipality of Tailândia, State of Pará, in an Oxisol. The experimental design was a randomized block with four replicates. The results allow to conclude that the accumulation of nutrients, except for Mn and Cu in the live Pueraria phaseoloides L. plant cover reduced over the years and those of B, Mn, Zn, and Cu were greater in the dead Pueraria phaseoloides L. plant cover and for Cl it was in the live Pueraria phaseoloides L. plant cover. The decreasing order of total micronutrient extraction was the following: Cl> Mn> B> Zn> Cu.


A adoção de plantas de cobertura em plantios comerciais tem se mostrado uma alternativa para promover ao solo maior proteção, melhorias em sua estrutura e agregação e otimização de acúmulo e ciclagem de nutrientes visando maior produção de biomassa e maior fixação de N2. Com o objetivo de determinar o acúmulo de micronutrientes puerária (Pueraria phaseoloides L.), em função da idade, estabelecida como cobertura de solo em plantações industriais de dendezeiros, instalou-se experimento na Companhia Real Agroindustrial (CRAI) no município de Tailândia, Estado do Pará, em Latossolo Amarelo. O delineamento experimental foi o de blocos ao acaso com quatro repetições. Os resultados permitem concluir que: o acúmulo de nutrientes com exceção do Mn e Cu na cobertura verde de Pueraria phaseoloides L., reduziu com o decorrer dos anos; os de B, Mn, Zn e Cu foram maiores na cobertura morta, enquanto que o de Cl na cobertura verde. A ordem decrescente da extração total de micronutrientes foi: Cl > Mn > B > Zn > Cu.

RESUMEN Las legumbres juegan un rol fundamental en la agricultura moderna; su inclusión en los sistemas agrícolas permite hacer un uso más sostenible de los recursos, ya que mejoran la fertilidad de los suelos, reducen la dependencia a los fertilizantes químicos al fijar el nitrógeno del aire, favorecen la rotación de cultivos tradicionales, principalmente cereales, y mejoran el tenor proteico de los cultivos que le suceden en la rotación. En este artículo se entregan aspectos relacionados con la situación agrícola actual de las legumbres en Chile, con datos estadísticos en cuanto a la evolución de la superficie sembrada, rendimiento, importaciones y requerimiento interno. A su vez, se dan a conocer las principales ventajas agronómicas de este grupo de cultivos y los problemas de manejo más importantes que han llevado, a este rubro, a tener la situación actual de desmedro en términos de producción en el país. Finalmente, se presentan una serie de desafíos factibles de llevar a cabo y que podrían revitalizar a este grupo de cultivos tan importantes desde un punto de vista ambiental como alimenticio.


ABSTRACT Legumes play a fundamental role in modern agriculture; their inclusion in the farming system allows a sustainable use of resources. This is due to the fact that legumes improve soil fertility by fixing atmospheric nitrogen, thus reducing the need for chemical fertilizer. Likewise, they improve crop rotation, mainly in cereal crops, increasing the protein content of successive crops. In this article, we describe aspects of the current legume situation in Chile with statistical data relative to the trends of sowing area, yield, imports and internal demand. This article also presents the agronomical advantages of using legumes and the main problems associated with crop management that has deteriorated production in this country. Finally, we present possible solutions to reinvigorate production of this crop, which is important from both an environmental and nutrition perspective.

El uso indiscriminado de fertilizantes químicos ha contribuido al deterioro de las propiedades biológicas, físicas y químicas del suelo, lo que derivó en la pérdida de su capacidad productiva. Por esta razón, se ha planteado como alternativa tecnológica el uso de biofertilizantes. El objetivo de esta investigación fue desarrollar un sistema de fermentación líquida y de enquistamiento adecuado para la multiplicación de Azotobacter chroococcum cepa AC1, una bacteria utilizada en la formulación de un biofertilizante actualmente producido por CORPOICA, Colombia. Se emplearon diseños estadísticos secuenciales para determinar las condiciones del sistema de fermentación. Se evaluaron la interacción entre la agitación, la aireación y el pH sobre la biomasa viable obtenida de AC1 (UFC/ml), que se tomó como variable de respuesta. Además, se evaluó la capacidad de enquistamiento de esta bacteria empleando 2 agentes de enquistamiento, AE01 y AE02. La actividad potencial promotora del crecimiento vegetal fue evaluada por medio del ensayo de ARA (fijación biológica de nitrógeno), la técnica de azul de fosfomolibdeno (solubilización de fosfato) y la reacción colorimétrica empleando el reactivo de Salkowski (producción de compuestos indólicos). Se evidenciaron efectos significativos (p <0,05) sobre la producción de biomasa de los 3 factores evaluados (pH, aireación y agitación) individualmente, de una interacción dual y en la interacción tripartita, teniendo un efecto positivo sobre la variable de respuesta la aireación y agitación. La adición de los inductores de enquistamiento AE01 y AE02 demostró la capacidad de la cepa AC1 para formar quistes en condiciones de estrés. Asimismo, las condiciones de fermentación y el enquistamiento no afectaron las actividades biológicas evaluadas.


The indiscriminate use of chemical fertilizers has contributed to the deterioration of the biological, physical and chemical properties of the soil, resulting in the loss of its productive capacity. For this reason, the use of biofertilizers has emerged as a technological alternative. The objective of this research was to develop a suitable liquid fermentation system and encystment for the multiplication of Azotobacter chroococcum AC1 strain, a bacterium employed in a biofertilizer formulation produced at present by CARPOICA, Colombia. Sequential statistical designs were used to determine the conditions in the fermentation system. The interaction between agitation, aeration and pH was evaluated on the viable biomass (CFU/ml) of AC1. In addition, the encystment ability of the strain was evaluated using two encystment agents and the potential plant growth-promoting rhizobacteria (PGPR) activity was assessed by different techniques, such as nitrogen fixation by ARA, phosphate solubilization by the phospho-molybdenum-blue reaction and indolic compound production by colorimetric reaction using the Salkowski reagent. Results showed significant effects (p <0.05) on the viable biomass in the three conditions (pH, aeration and agitation) tested individually, in one dual interaction and one tripartite interaction, were demonstrated to have a positive effect on the response variable aeration and agitation. The addition of the two encystment agents evaluated, AE01 and AE02, demonstrated the ability of AC1 to form cysts under stress conditions. Likewise, fermentation and encystment conditions did not affect the biological activities tested.

The objective of this study is to verify the population density and the dynamics of tillering in the Marandu palisade grass sward subjected to nitrogen (N) fertilization strategies, characterized by the N supply via urea or bacterial inoculant (Azospirillum brasilense). The treatments comprised of four nitrogen fertilization strategies: (A) Without fertilization, (B) 80 kg N/ha, (C) inoculant (A. brasilense), and (D) 80 kg N/ha + inoculant, distributed in a randomized complete block design, with three replications. The nitrogen supply strategies were evaluated during six periods: October, November, and December (2012) as well as January, March, and April (2013). The nitrogen dose or inoculant had no effect on the tiller appearance rate (TAR), tiller mortality rate (TMR), tiller survival rate (TSR), or tiller population density (TPD). However, these variables were influenced by the season. The TAR and TSR were higher at the beginning of the experimental period (October) and lower towards the end of the period (March-April), whereas, TMR and TPD exhibited the opposite behavior, with lower values in October and higher from January onward. Neither the nitrogen nor the inoculant influenced the population dynamics of the tillers in Marandu palisade grass.(AU)


Objetivou-se com este estudo verificar a densidade populacional e a dinâmica do perfilhamento em dosséis de capim Marandu submetidos a estratégias de adubação nitrogenada, caracterizadas pelo fornecimento de N via ureia ou inoculante (Azospirillum brasilense). Os tratamentos foram quatro estratégias de adubação nitrogenada: sem fertilização, 80kg N/ha, inoculante (A. brasilense) e 80kg N/ha + inoculante, distribuídos em um delineamento de blocos completos ao acaso, com três repetições por tratamento. As estratégias de aporte nitrogenado foram avaliadas em seis épocas: outubro, novembro e dezembro de 2012; e janeiro, março e abril de 2013. Não foi verificada influência da dose de nitrogênio ou do inoculante sobre a taxa de aparecimento (TAP), a taxa de mortalidade (TMP), a taxa de sobrevivência (TSP) e a densidade populacional de perfilhos (DPP). No entanto, essas variáveis foram influenciadas pela época do ano. A TAP e a TSP apresentaram maiores valores no início do período experimental (outubro) e menor valor ao final do período (março a abril). A TMP e a DPP expressaram respostas opostas, com menores valores em outubro e maiores a partir de janeiro. Não houve influência do nitrogênio e do inoculante sobre a dinâmica populacional de perfilhos em capim Marandu.(AU)

To maximize the contribution of biological nitrogen fixation in common bean, it is necessary to use bacterial strains that are more adapted, competitive, and efficient in the symbiotic process. In this regard, the aim of this study was to evaluate the agronomic efficiency (AE) of three bacterial strains isolated from acid soils with high Al content from the Amazon region in an Argissolo Vermelho Distrófico típico soil (Typic Rhodustults - USDA Classification) from the municipality of Formiga, MG, Brazil. We compared their AE to that of the reference strain CIAT 899T and of two controls without inoculation (one without and another with 80 kg ha-1 of N-urea). The results indicated that inoculation with the strains UFLA 02-100 and UFLA 02-127 provides grain yield equivalent to inoculation with the reference strain and to the control with mineral N. Thus, both have potential for recommendation as inoculants for common bean.


Para maximizar a contribuição da fixação biológica de nitrogênio no feijoeiro-comum é necessária a utilização de estirpes de bactérias mais adaptadas, competitivas e eficientes no processo simbiótico. Nesse sentido, objetivou-se avaliar, em um Argissolo Vermelho Distrófico típico do município de Formiga-MG, a eficiência agronômica (EA) de três estirpes isoladas de solos ácidos e com alto teor de Al da Amazônia e comparar suas EA à da estirpe referência CIAT 899T e à de dois controles sem inoculação (um sem e outro com 80 kg ha-1 of N-ureia). Os resultados indicaram que a inoculação com as estirpes UFLA 02-100 e UFLA 02-127 propicia rendimento de grãos equivalente ao da estirpe referência e ao do controle com N mineral e que por isso, ambas têm potencial para ser recomendadas como inoculantes para o feijoeiro-comum.

Abstract Presence of the relatively new sulfonylurea herbicide monosulfuron-ester at 0.03-300 nmol/L affected the growth of two non-target nitrogen-fixing cyanobacteria (Anabaena flos-aquae and Anabaena azotica) and substantially inhibited in vitro Acetolactate synthase activity, with IC50 of 3.3 and 101.3 nmol/L for A. flos-aquae and A. azotica, respectively. Presenting in 30-300 nmol/L, it inhibited protein synthesis of the cyanobacteria with less amino acids produced as its concentration increased. Our findings support the view that monosulfuron-ester toxicity in both nitrogen-fixing cyanobacteria is due to its interference with protein metabolism via inhibition of branch-chain amino acid biosynthesis, and particularly Acetolactate synthase activity.

Abstract Aneurinibacillus aneurinilyticus strain CKMV1 was isolated from rhizosphere of Valeriana jatamansi and possessed multiple plant growth promoting traits like production of phosphate solubilization (260 mg/L), nitrogen fixation (202.91 nmol ethylene mL-1 h-1), indole-3-acetic acid (IAA) (8.1 µg/mL), siderophores (61.60%), HCN (hydrogen cyanide) production and antifungal activity. We investigated the ability of isolate CKMV1 to solubilize insoluble P via mechanism of organic acid production. High-performance liquid chromatography (HPLC) study showed that isolate CKMV1 produced mainly gluconic (1.34%) and oxalic acids. However, genetic evidences for nitrogen fixation and phosphate solubilization by organic acid production have been reported first time for A. aneurinilyticus strain CKMV1. A unique combination of glucose dehydrogenase (gdh) gene and pyrroloquinoline quinone synthase (pqq) gene, a cofactor of gdh involved in phosphate solubilization has been elucidated. Nitrogenase (nif H) gene for nitrogen fixation was reported from A. aneurinilyticus. It was notable that isolate CKMV1 exhibited highest antifungal against Sclerotium rolfsii (93.58%) followed by Fusarium oxysporum (64.3%), Dematophora necatrix (52.71%), Rhizoctonia solani (91.58%), Alternaria sp. (71.08%) and Phytophthora sp. (71.37%). Remarkable increase was observed in seed germination (27.07%), shoot length (42.33%), root length (52.6%), shoot dry weight (62.01%) and root dry weight (45.7%) along with NPK (0.74, 0.36, 1.82%) content of tomato under net house condition. Isolate CKMV1 possessed traits related to plant growth promotion, therefore, could be a potential candidate for the development of biofertiliser or biocontrol agent and this is the first study to include the Aneurinibacillus as PGPR.

Abstract The family Leguminosae comprises approximately 20,000 species that mostly form symbioses with arbuscular mycorrhizal fungi (AMF) and nitrogen-fixing bacteria (NFB). This study is aimed at investigating and confirming the dependence on nodulation and biological nitrogen fixation in the specie Piptadenia gonoacantha (Mart.) Macbr., which belongs to the Piptadenia group. Two consecutive experiments were performed in a greenhouse. The experiments were fully randomized with six replicates and a factorial scheme. For the treatments, the two AMF species and three NFB strains were combined to nodulate P. gonoacantha in addition to the control treatments. The results indicate this species’ capacity for nodulation without the AMF; however, the AMF + NFB combinations yielded a considerable gain in P. gonoacantha shoot weight compared with the treatments that only included inoculating with bacteria or AMF. The results also confirm that the treatment effects among the AMF + NFB combinations produced different shoot dry weight/root dry weight ratios. We conclude that AMF is not necessary for nodulation and that this dependence improves species development because plant growth increases upon co-inoculation.

A great number of species and individuals of scandent legumes establishing symbiosis with nitrogen fixing bacteria occurs in the Amazon Forest. These symbiosis probably play an important role in contributing to nitrogen incorporation in this ecossystem. The objectives of this study were to evaluate the growth of eight species of scandent legumes in five nursery substrates; to compare nodulation with rhizobia strains introduced or native to these substrates; and to characterize phenotypically and genetically these rhizobia. The experiment was carried out in a completely randomized design with five replications. Five to seven months after seedling emergency, according to the legume species, growth and nodulation parameters were determined. Rhizobia identification of strains was carried out by 16S rRNA gene partial sequencing. The survival of seedlings after the transplanting varied from 93 to 98%, in Ultisol (Argissolo in Brazilian classification), collected in primary forest, and fertilized with all nutrients, except nitrogen (ULTfert); and in a clay and sand mixture, in a ratio 3:2 (CONV), respectively. Species with height superior to 30 cm, in general, grew better in substrates with higher fertility: ULTfert and Humic Gley soil (HG). Seven out of the eight species were able to nodulate. The percentage of nodulation per substrate was: SAND, washed sand with mixed inoculum of 100 rhizobia strains plus fertilization (100), HG (80), CONV (100), ULT, A-horizon of red-yellow Ultisol collected in the Ducke Forest Reserve (Manaus) (44), and ULTfert (55%). Bradyrhizobium spp. were isolated from nodules of all species and substrates. Burkolderia fungorum was isolated from Dalbergia inundata. For Dalbergia riedelli and Dalbergia inundata, this is the first report on the identification of symbiotic strains. Um grande número de espécies e indivíduos de leguminosas escandentes estabelecendo simbiose com bactérias fixadoras de nitrogênio ocorrem na floresta Amazônica. Estas simbioses provavelmente desempenham importante papel, contribuindo para incorporação de nitrogênio neste ecossistema. Os objetivos deste trabalho foram avaliar o crescimento de oito espécies de leguminosas escandentes em cinco substratos no viveiro; comparar a nodulação com as estirpes de rizóbio introduzidas ou nativas destes substratos; e caracterizar esses rizóbios fenotípica e geneticamente. O experimento foi conduzido em delineamento inteiramente casualizado com cinco repetições. Cinco a sete meses após a emergência das mudas parâmetros de crescimento e nodulação foram determinados. A identificação das estirpes de rizóbios foi feita pelo sequenciamento parcial do gene 16S rRNA. A sobrevivência das mudas após o transplante variou de 93 a 98%, em um Argissolo (Ultisol de acordo com a classificação da USDA), coletado em floresta primária e adubado com todos os nutrientes, exceto nitrogênio (ULTfert), e na mistura argila e areia, na proporção 3:2 (CONV), respectivamente. Espécies que apresentaram altura superior a 30 cm, no geral, cresceram melhor nos substratos de maior fertilidade: ULTfert e solo Glei Húmico (GH). Das oito espécies estudadas, sete nodularam. A porcentagem de nodulação por substrato foi: AREIA, areia lavada com inóculo misto de 100 estirpes de rizóbio mais adubação (100), GH (80), CONV (100), ULT, horizonte A de Argissolo vermelho-amarelo coletado na Reserva Florestal Ducke (Manaus) (44) e ULTfert (55%). Bradyrhizobium spp. foram isolados de nódulos de todas as espécies e substratos. Burkolderia fungorum foi isolada de Dalbergia inundata. Este é o primeiro relato de identificação de estirpes simbióticas para Dalbergia riedelli e Dalbergia inundata. The strain BR 3262 was isolated from nodule of cowpea (Vigna unguiculata L. Walp) growing in soil of the Atlantic Forest area in Brazil and it is reported as an efficient nitrogen fixing bacterium associated to cowpea. Firstly, this strain was assigned as Bradyrhizobium elkanii, however, recently a more detailed genetic and molecular characterization has indicated it could be a Bradyrhizobium pachyrhizi species. We report here the draft genome sequence of B. pachyrhizi strain BR 3262, an elite bacterium used as inoculant for cowpea. The whole genome with 116 scaffolds, 8,965,178 bp and 63.8% of C+G content for BR 3262 was obtained using Illumina MiSeq sequencing technology. Annotation was added by the RAST prokaryotic genome annotation service and shown 8369 coding sequences, 52 RNAs genes, classified in 504 subsystems.

The strain BR 3267 is a nitrogen-fixing symbiotic bacteria isolated from soil of semi-arid area of Brazilian Northeast using cowpea as the trap plant. This strain is used as commercial inoculant for cowpea and presents high efficient in nitrogen fixation as consequence of its adaptation potential to semi-arid conditions. We report here the draft genome sequence of Bradyrhizobium sp. strain BR 3267, an elite bacterium used as inoculant for cowpea. Whole genome sequencing of BR 3267 using Illumina MiSeq sequencing technology has 55 scaffolds with a total genome size of 7,904,309 bp and C+G 63%. Annotation was added by the RAST prokaryotic genome annotation service and has shown 7314 coding sequences and 52 RNA genes.

O objetivo do trabalho foi avaliar as características produtivas e nutricionais do capim- xaraés inoculado com bactérias diazotróficas associativas, em primeiro cultivo em Latossolo Vermelho do Cerrado. O experimento foi realizado em casa de vegetação. O delineamento experimental foi inteiramente ao acaso, constituído por fatorial 6x3: três estirpes de bactérias diazotróficas associativas (MTAz8, MTH2 e Y2), uma combinação das estirpes MTAz8 e MTH2, adubação nitrogenada e testemunha absoluta, e três cortes (30, 60 e 90 dias), em cinco repetições. A inoculação foi feita por meio da inserção de uma alíquota de 5mL de caldo bacteriano contendo 108 células mL-1 no solo próximo ao sistema radicular. As variáveis avaliadas foram: massa seca da parte aérea, porcentagem de proteína bruta, acúmulo e concentração de nitrogênio na parte aérea e determinação do número de bactérias diazotróficas do solo. No tratamento adubado com nitrogênio, a produção de massa seca aumentou ao longo dos cortes. Entre as estirpes de bactérias associativas, a Y2 apresentou maior acúmulo de nitrogênio e maior população de bactérias no meio de cultura LGI. As características nutricionais do capim-xaraés decaíram à medida que foram realizados os cortes.(AU)


The aim of this study was to evaluate yield and nutritional characteristics of Xaraés grass inoculated with diazotrophic associative bacteria in the first cultivation in the Cerrado Oxisol. The experiment was conducted in the greenhouse. The experimental design was completely randomized factorial 6x3 consisting of: three strains of diazotrophic associative bacteria (MTAz8, MTH2 and Y2), a combination of strains MTAz8 and MTH2, nitrogen fertilization and absolute control, and three cuts (30, 60 and 90 days) in five replicates. The inoculation was performed by inserting an aliquot of 5ml of the bacterial broth containing 10 8 cells ml-1 in the soil near the root zone of each plant. The variables evaluated were: dry weight of shoot, crude protein, accumulation and nitrogen concentration in the shoot and determining the number of nitrogen-fixing bacteria in the soil. The fertilized soil dry matter production increased over the cuts. Among the strains Y2 showed higher accumulation of nitrogen and largest population of bacteria in the middle of the LGI culture. The nutritional characteristics of Xaraés grass declined as the cuts were made.(AU)
