RESUMO

Aggregation of erythrocytes in the blood of rabbits was shown to depend on concentration of proteoglycans in the blood plasma. In experimental sepsis (Str. haemolyticus) there occurred simultaneous increase of concentration of proteoglycans in plasma and increased aggregation of erythrocytes in the blood. In pregnancy, the concentration of proteoglycans in the blood was constant at a low level throughout the period of pregnancy.

The paper is concerned with the results of fractionated thermal-radiation action on Brown-Pearce tumor in rabbits, sarcoma-37 (C-37) and Lewis carcinoma (LLC) in outbred and hybrid F1 (C57B1/6 x CBA) mice. Rabbit tumors were subjected to hyperthermia on UHF Plot units 4 h after irradiation, and in mice with a water-bath directly before irradiation. The frequency of Brown-Pearce tumor regression was increased as a result of hyperthermia in 4 h. The comparison of the effectiveness of fractionated radiation or thermal-radiation action on murine tumors (2 of 3 fractions for 4 days) indicated different type changes of the value of the coefficient of thermal enhancement of hyperthermia. For C-37 it was decreased with an increase in the number of fractions, for LLC it was increased, probably as a result of reoxygenation and high radiosensitivity of C-37.

RESUMO

Rabbits with the Brown-Pierce carcinoma have been studied in chronic experiments for dynamics of the permanent potential reflecting bioelectrochemical processes in different structures of the hypothalamic and limbic system: medial preoptic area, ventromedial nucleus, posterior hypothalamic area, basal and lateral amygdalas, hippocampus, nucleus coeruleus as well as in central nucleus of the suture. The space-temporal pattern of neurodynamic changes in the subcortical brain structures has been found. These changes correlated with developmental characteristics of the Brown-Pierce carcinoma.

Specific activity of glycerol-3-phosphate dehydrogenase, estimated in direct and reverse reactions, was decreased, as compared with normal state, without any alterations in the ratio of activities, in muscle homogenates of rabbits with Brown-Pearce carcinoma. The enzymatic activity was decreased by an order of magnitude in homogenates of the tumor as compared with muscle homogenates of the tumor-bearing animals simultaneously with relative stimulation of the reverse reaction. Specific activity of the crystalline enzyme from muscles of tumor-bearing animals was decreased as compared with normal tissues but the ratio of activities was 2-fold increased due to the reverse reaction. In the carcinoma activity of one of the three enzyme fractions, obtained after isoelectrofocusing, was similar to the control values; in the second fraction the ratio of activities was decreased due to inhibition of the direct reaction, the third fraction was inactive. Amino acid composition of crystalline glycerol-3-phosphate dehydrogenase, isolated from tumor-bearing animals, was different in content of 5 amino acids from the enzyme amino acid composition of controls, thus suggesting the structural differences of the proteins from these fractions.

Lymphopoiesis stimulating drugs were injected into the blood of rabbits bearing Brown-Pearce transplantable tumor. It was found that treatment with mannitol targeted on osmoregulation mechanisms and terrilytin--on blood and lymph coagulation intensified the lymphogenic migration of tumor cells. When used in an experimental series involving endolymphic transplantation of tumor cells, the said drugs promoted tumor cell passage from lymph to blood, which resulted in an increased number of metastasis-bearing animals. It was also found that the promoting effect may be counteracted by preliminary endolymphic injection of a fat-soluble antitumor preparation. Lymphopoiesis and lymph flow stimulation in conjunction with thoracic duct draining may be used for removal of tumor cells from the lymphatics.

Experiments on rabbits with implanted liver tumours have shown that iso-dense tumours, which are not visible on CT, may be demonstrated by dynamic CT following intravenous contrast injection. They may appear as hypodense areas during a period of maximal organ perfusion, if the tumours have not taken up the contrast. The ability to recognise tumours depends largely on intravascular and, only to a smaller extent, on interstitial contrast enhancement.

RESUMO

The neural formations of intact rabbit major arteries and nerves of the same arteries in rabbits with the II and IV stages of the malignant Brown--Pearce tumor have been studied. Periarterial neural fasciculi, fibers and terminal branches have been revealed. According to ultrastructure of their cytoplasmic matrix, they can be divided into adrenergic, cholinergic or sensitive. When the malignant tumor is growing, the arterial neural formations undergo certain distrophic-destructive changes; they are especially pronounced in the membranous structures of the neural elements. Their cytoplasmic and basal membranes are partly or completely destroyed. In mitochondria the matrix is altered, some of them are vacuolized. Neurotubules and vesicles change their form and size and are partly destroyed. Membranous structures of lemmocytes are altered. The electron microscopic investigation adds something new to our data on ultrastructural changes in the arterial neural formations under growth of the malignant tumor previously obtained by means of neurohistological and neurohistochemical methods.

RESUMO

The efficacy of the active specific immunotherapy and immune prophylaxis against the Brown Pearce carcinoma in rabbits was tested. The tumor cells taken as antigens were either untreated or treated with Mitomycin C combined or not with vibrio-cholerae-neuraminidase. The immune prophylaxis using tumor cells prepared this way proved to be very effective. In contrast, immunotherapy of rabbits with tumors was not effective, even when immunization was started directly after transplantation. Possible mechanisms were analyzed in-vitro and in-vivo assays. The cellular reactivity of sensitized spleen cells was positive, the humoral reaction was weaker. This kind of reaction could only be demonstrated with tumor cells pretreated with vibrio-cholerae-neuraminidase. The findings shall only be discussed with respect to the possible causes of ineffective immunotherapy.

Morphological observation was performed to see the effect of bronchial arterial infusion therapy (BAI) of mitomycin C (MMC) and also of the ischemia to the transplanted bronchial carcinoma. Single shot of 2mg/kg of MMC showed destructive changes on the tumor. Single shot of 3mg/kg and repeated administration of MMC brought relatively severe intra-arterial inflammatory changes such as intimal edema, thickening and proliferation which suggested ischemic effect on the tumor due to poor perfusion from stenosis or obstruction of the vessels. On the other hand a simple ligation of the artery also brought about more than moderate destructive changes in the tumor. Therefore, mechanisms of the effect of the BAI of MMC to the lung tumor might involve the secondary effect from ischemia in addition to the effect of MMC itself.
