RESUMO

Although stem cell therapy is a promising alternative for treatment of degenerative diseases, there are just few reports on the use of stem cells therapy in horse's reproductive system. This study aims to evaluate the effect of intratesticular injection of bone marrow mesenchymal stromal/stem cells (MSCs) in healthy stallions, and its outcome on seminal parameters and fertility. In Experiment 1, 24 stallions were divided into treatment group (TG) and control group (CG). In the TG, an intratesticular application of MSC was performed, and in the CG, only PBS was used. Measurements of testicular volume, surface temperature and Doppler ultrasonography were performed 24 and 48 hr after treatments. Fifteen days after application, the testicles were removed and submitted to histological analysis. In Experiment 2, 3 fertile stallions received similarly treatment with MSCs. Physical examination and sperm analysis were performed weekly during 60 days after treatment, and at the end, semen from one of them was used for artificial inseminations of 6 healthy mares. In Experiment 1, clinical examinations showed no signals of acute inflammation on both groups according to the analysed variables (p > .05). Also, no signal of chronic inflammation was observed on histological evaluation. In Experiment 2, stallions presented no physical alterations or changes in sperm parameters, and a satisfactory fertility rate (83%; 5/6) was observed after AI. The results support the hypothesis that intratesticular application of bone marrow MSCs is a safe procedure, and this could be a promising alternative to treat testicular degenerative conditions.

Boar semen cannot be immediately cryopreserved, it need be hold at 17 °C prior to cryopreservation, holding time has been used to improve cryopreserved boar semen, since holding time allows a prolonged interaction between spermatozoa and seminal plasma components. However, until now only few periods of holding time have been studied, and boar semen had been held at 17 °C for 24 h to facilitate its manufacture. Thus, this experiment aims to study the effect several holding time (0, 4, 8, 12, 24, 28 and 32 h) on boar spermatozoa post-thawed (PT) characteristics. Fifteen sperm-rich fractions of ejaculate were extended in Beltsville Thawing Solution and storage at 17 °C. After each holding time (0, 4, 8, 12, 24, 28 and 32 h), a sample was centrifuged, and sperm pellet was diluted in an extender composed of sugars, amino acids, buffers, 20% egg yolk (v/v), antibiotics, 2% glycerol as a cryoprotectant, and 2% methylformamide (v/v). Cryopreservation was performed with an automatic cryopreservation system. Cryopreserved boar semen was evaluated to spermatozoa kinetics, plasma and acrosomal membranes integrity, mitochondrial membrane potential, detection of superoxide anion, plasma membrane fluidity, and peroxidation. Twenty-four hours of holding increase total and progressive motility, rapid spermatozoa, and integrity of plasma and acrosome membranes. To mitochondrial membrane potential, 32 h is needed. However, holding time was not able to control the superoxide anion amount neither membrane lipid peroxidation, and had no effects on membrane fluidity. Thus, to reach the best results of PT boar semen the ideal holding time is 24 h.

RESUMO

The use of stallion frozen semen minimizes the spread of disease, eliminates geographic barriers, and preserves the genetic material of the animal for an unlimited time. Significant progress on the frozen thawed stallion semen process and consequently fertility has been achieved over the last decade. These improvements not only increased fertility rates but also allowed cryopreservation of semen from "poor freezers." This article reviews traditional steps and new strategies for stallion semen handling and processing that are performed to overcome the deleterious effects of semen preservation and consequently improve frozen semen quality and fertility.

The Web-based Executive Functioning Questionnaire (Webexec) is a brief scale developed to assess executive functions via online format. It has been used in different contexts, but its adaptation to other cultures is still restricted. This study aimed to perform a cross-cultural adaptation of the Webexec for a Brazilian sample considering the psychometric properties of the scale. This study used a sample of 295 Brazilian participants, with a mean age equal to 20.69 (SD = 6.030). This is a longitudinal study with reapplication of the scale six weeks after the test phase. Classical and contemporary methods were applied to analyze the psychometric properties of the Webexec. The results showed that the scale presented excellent psychometric properties for the Brazilian version, considering validity evidence based on the content and internal structure of Webexec, as well as reliability and precision. However, it is considered that other relational and experimental studies should be carried out with a larger sample size and for different population groups.

The two-step protocol (2 S) is currently used for boar semen cryopreservation. In this method, the cryoprotectant penetrant agents (CPAs) are added at 5 °C to reduce the toxicity of CPAs. An alternative is the one-step protocol (1 S), which is easier, cheaper, and reduces the necessity of equipment, but could increase the toxicity of CPAs. Currently, there are no studies that compared both protocols for boar semen cryopreservation. This experiment aimed to study the effect of cryopreservation protocol (1 S vs 2 S) on boar spermatozoa. In the one-step protocol, after centrifugation, the spermatozoa pellet was resuspended at 17 °C in the extender containing CPAs to achieve a concentration of 1 × 109 spermatozoa/mL and then submitted to cryopreservation. For the two-step protocol, the sperm pellet was resuspended in fraction A at 17 °C to achieve a concentration of 1.5 × 109 spermatozoa/ mL, and then allowed to cool to 5º C before fraction B with CPA was added to the sample to achieve a final concentration of 1 × 109 spermatozoa/mL and followed by freezing. The cryopreservation protocol did not impact total motility at 5 °C (1 S: 78.5 % vs 2 S: 79 %, p > 0.05). After thawing, the two-step protocol improved (p < 0.05) total (1 S: 18.2 % vs 2 S: 29.5 %) and progressive motility (1 S: 9 % vs 2 S: 15%). Further, the 2 S protocol increased (p < 0.05) the percentage of rapid spermatozoa (1 S: 8.7 % vs 2 S: 14.6 %) and spermatozoa with intact plasma and acrosomal membrane (IAIP) (1 S: 40.5 % vs 2 S: 61.5 %), and increased (p < 0.05) live sperm cells with high mitochondrial potential (MHIP) (1 S: 42.9 % vs 2 S: 60 %). The boar semen cryopreservation method (TRT) did not (p > 0.05) alter membrane lipid disorder, lipid peroxidation, and superoxide anion. Thus, the best method for boar semen cryopreservation is the two-step protocol.

This study aimed to evaluate the effects of mesenchymal stem cell-conditioned medium (MSC-CM) on sperm parameters, intrauterine polymorphonuclear neutrophils (PMN), intrauterine fluid accumulation (IUF), and fertility in mares. In experiment 1, two ejaculates from ten stallions were extended to 50 million sperm/mL using a milk-based extender. Thereafter, 20 mL of extended semen was added of MSC-CM as follows: 0, 5, 10, 15, and 20 mL. Sperm kinetics and plasma membrane integrity were evaluated immediately after dilution (T0) and 2 h post-incubation at 37 °C (T2). In experiment 2, mares characterized as resistant (n = 13) or susceptible (n = 7) to endometritis were inseminated with fresh semen 24 h post-induction of ovulation in two (Control and CM-1) and three (Control, CM-1, and CM-2) cycles in a crossover, as follows: control, no pharmacological interference; CM-1, supplementation of semen insemination dose at 3:4 (v:v, MSC-CM:semen); CM-2, 30 mL of MSC-CM was infused into the uterus 24 h before insemination. Endometrial cytology and uterine fluid were collected 6 and 24 h after insemination to evaluate the number of PMNs and concentrations of interleukins IL6, IL10, and TNFα. IUF was determined by ultrasonography 24 and 48 h after insemination. Pregnancy status was diagnosed 14 days after ovulation. The addition of MSC-CM to semen did not influence sperm parameters at T0 and T2 (P > 0.05) and reduced (CM-1; P < 0.05) the number of PMNs at 6 h post-insemination in resistant mares. In susceptible mares, PMNs at 6 and 24 h post-insemination, as well as IUF were reduced (P < 0.05) in both treated cycles (CM-1 and CM-2). In addition, MSC-CM downregulated IL6 and upregulated IL10 concentrations in the uterus of susceptible mares after insemination. There were no differences in fertility rates among groups both in resistant (Control, 77%, 10/13; CM-1, 62%, 8/13) and susceptible mares (Control, 42.8%, 3/7; CM-1, 57.1%, 4/7; CM-2, 85.7%. 6/7). In conclusion, MSC-CM did not affect sperm parameters when mixed with diluted semen, and reduced post-insemination inflammatory responses in mares.

The present study aimed to compare semen parameters and fertility of cooled donkey semen extended in a commercially available skim milk (SKM) based extender and the same extender with cholesterol-loaded cyclodextrin (SKM-CLC). In Experiment 1, thirty-five ejaculates from seven jacks were split in SKM and SKM-CLC, extended at 50 million sperm/mL and stored at 5°C for 48 hours. Total motility (TM), progressive motility (PM), percentage of sperm with rapid motility (RAP) were assessed with CASA. Plasma membrane stability (PMS), and high mitochondrial membrane potential (HMP) were assessed with the combination of Yo-Pro and MitoStatusRed with flow cytometry. Semen was assessed before (0), 24 and 48h after cooling. In Experiment 2, two estrous cycles of 15 mares were used for fertility assessment. Mares were examined every other day by transrectal ultrasonography and had ovulation induced with 250 µg of histrelin acetate when a ≥35 mm follicle was first detected. Mares were randomly inseminated with semen obtained from one jack. Semen was extended in either SKM or SKM-CLC and cooled-stored for 24 hours. Pregnancy diagnosis was carried out 15-day post-ovulation. Data were analyzed with a mix model and Tukey's as posthoc and logistic regression model. Significance was set at P ≤ .05. There were no differences in TM, PM, RAP, PMS, and HMP for semen extended in either extender immediately before cooling (P > .05). There was a reduction in TM, PM, RAP, PMS, and HMP overtime across groups (P < .05); however, semen extended with SKM-CLC had superior TM, PM, RAP, PMS, and HMP than semen extended in SKM at 24- and 48-hours post-cooling (P < .05). Mares bred with semen extended in SKM had a lower conception rate (13%, 2/15 cycles) than cycles bred with SKM-CLC (47%, 7/15 cycles; P < .05). In conclusion, incorporating CLC into SKM extender improved cooling ability and fertility of donkey semen in horse mares. It remains to be determined if similar results can be obtained in clinical practice with mares and jennies.

The aim of this study was to evaluate the effect of coconut water as a component of extender in different formulations for cooling equine sperm. One ejaculate of fourteen stallions was collected. Sperm was diluted to 50 × 106 sperm/mL using five different extenders: ACP-105: powdered coconut water extender (ACP-105, ACP Biotecnologia, Brazil); ACP-Milk: ACP-105 + 20 g/L of skimmed milk; ACP-EY 2.5%: ACP-105 + 2.5% of egg yolk; ACP-EY 5%: ACP-105 + 5% of egg yolk; and BotuSêmen (Botupharma, Botucatu, Brazil) and cooled in passive cooling device (BotuFlex, Botupharma, Botucatu, Brazil) at 5 and 15°C for 24 hours. Sperm kinetics and plasma membrane integrity (PMI) were evaluated by computer-assisted sperm analysis and fluorescence staining, respectively, at T0 (before cooling) and T24 (24 hours after cooling). Sperm kinetics did not differ at T0 among groups (P > .05); however, at T24, these parameters were significantly lower in ACP-105 (5°C, total motility [TM]: 9.2 ± 3.6%; progressive motility [PM]: 2.7 ± 1.6%; percentage of fast-moving spermatozoa [RAP]: 4.8 ± 3.0%; 15°C, TM: 10.6 ± 3.0%; PM: 1.1 ± 0.5%; RAP: 4.8 ± 1.9%) and ACP-EY 5% (5°C, TM: 28.0 ± 6.3%; PM: 5.7 ± 1.8%; RAP: 15.9 ± 6.0%; 15°C, TM: 30.0 ± 6.0%; PM: 6.9 ± 2.1%; RAP: 17.6 ± 5.3%) compared with BotuSêmen (5°C, TM: 66.2 ± 5.6%; PM: 21.1 ± 2.8%; RAP: 53.9 ± 6.1%; 15°C, TM: 63.4 ± 5.4%; PM: 17.2 ± 2.8%; RAP: 51.4 ± 6.3%) (P < .05). All groups exhibited similar PMI at tested moments and cooling temperatures (5°C: 83%; 15°C: 84%) (P > .05). Further studies are necessary to evaluate powdered coconut water in different compositions of sperm extender; however, coconut-based extender as used in this study was not an alternative to preserve sperm parameters of cooled equine sperm.

Non-steroidal anti-inflammatory drugs (NSAIDs) are a therapeutic option for the treatment of inflammation. However, negative effects of non-selective NSAIDs for treatment of mares with endometritis have been described, including delayed uterine clearance and impairment of ovulations. Firocoxib is a specific cyclooxygenase-2 (COX-2) inhibitor and has the ability to act in the uterus of mares. We investigated the effects of firocoxib on ovulation rate, numbers of polymorphonuclear neutrophils (PMNs), and COX-2 protein levels in the endometrial tissue of susceptible mares after insemination. Two experiments were conducted. In experiment 1, twenty mares were evaluated in two consecutive estrous cycles broken into the following groups: Control - no pharmacological interference; Treatment - mares were treated with 0.2 mg/kg of firocoxib orally. The treatment began on the day of ovulation induction, and firocoxib was administered until one day after artificial insemination (AI). Ovulation was induced with 1 mg of deslorelin acetate and the mares were inseminated 24 h after the injection. Ovulation was confirmed 48 h after induction, and embryos were collected eight days after ovulation. Experiment 2: Nine mares susceptible to persistent mating-induced endometritis (PMIE) were artificially inseminated. The mares were examined with ultrasound and inseminated with fresh semen in two consecutive cycles, control and treated, in a cross-over study design. The amount of intrauterine fluid was measured, and endometrial samples were collected 24 h after AI. The number of PMNs was determined by endometrial cytology and biopsy, and COX-2 labeling in endometrial samples was evaluated by immunohistochemistry. Firocoxib treatment did not induce ovulatory failure or affect embryo recovery rate in Experiment 1. In Experiment 2, firocoxib treatment reduced inflammation after AI in mares as evidenced with results regarding PMN numbers/percentage and endometrial COX-2 staining. In conclusion, the proposed treatment with firocoxib reduced endometrial inflammation in mares susceptible to PMIE after breeding, with no adverse effects.

This study presents the Brazilian version of the Child-Adolescent Perfectionism Scale (CAPS), one of the most widely used instruments for assessing perfectionism in young people. The objective of this study was to present the adaptation and evidence of validity for the CAPS in Brazilian Portuguese. The study was conducted on 599 adolescents (Mean age = 15.46 SD = 1.15), 66% girls. We investigated CAPS's internal structure, measurement properties according to age and sex, and its relationship with other questionnaires. We found a good fit for a two-factor structure for the CAPS controlling for an acquiescent responding style. We found evidence of invariance across participants' sex and lack of bias (using a MIMIC approach) due to participant age. The CAPS scores were associated with personality traits and a measure of mental health. The CAPS can be used as a questionnaire to assess perfectionism in Brazilian adolescents.


O presente estudo apresenta a versão brasileira da Child-Adolescent Perfectionism Scale (Caps), um dos instrumentos mais utilizados para avaliação do perfeccionismo em adolescentes. O objetivo do presente estudo foi apresentar a adaptação e evidências de validade da Caps para o português brasileiro. O estudo foi realizado com uma amostra de 599 adolescentes (média de idade = 15,46 DP = 1,15), 66% meninas. Foi investigada a estrutura interna da Caps, e a relação da medida com idade e sexo, bem como sua associação com outros questionários. Foi encontrado um ajuste aceitável para uma estrutura de dois fatores para a Caps, controlando o estilo de resposta aquiescente. Foi encontrada evidência de invariância de medida entre o sexo dos participantes e ausência de viés (usando uma abordagem MIMIC) devido à idade dos participantes. Houve associação dos escores da Caps com traços de personalidade e uma medida de saúde mental. A Caps pode ser usada como um questionário para avaliar o perfeccionismo em adolescentes brasileiros


Este estudio presenta la versión brasileña de la Child-Adolescent Perfectionism Scale (CAPS), uno de los instrumentos más utilizados para la evaluación del perfeccionismo en jóvenes. El objetivo de este estudio fue presentar la adaptación y las pruebas de validez del CAPS en el portugués de Brasil. El estudio se realizó sobre una muestra de 599 adolescentes (Edad media = 15,46 DE = 1,15), 66% niñas. Se investigó la estruc-tura interna del CAPS, las propiedades de medida según edad y sexo, y su relación con otros cuestionarios. Encontramos un buen ajuste para una estructura de dos factores para el CAPS controlador el estilo de respuesta aquiescente. Encontramos evidencia de invariancia entre el sexo de los participantes y falta de sesgo (usando un enfoque MIMIC) debido a la edad de los participantes. Hubo una asociación de las pun-tuaciones CAPS con rasgos de personalidad y una medida de salud mental. El CAPS podría ser utilizado como cuestionario para evaluar el perfeccionismo en adolescentes brasileños

O Transtorno do Estresse Pós-traumático (TEPT) é caracterizado por apresentar sintomas de desconforto e medo diante de situações diretas e/ou imaginadas em relação a experiências que ameaçam a integridade pessoal. As Funções Executivas (FE) são processos responsáveis pela análise de uma situação, tomada de decisão e ações voltadas à resolução de problemas. Sabe-se que o TEPT está associado a prejuízos em diferentes dimensões das FE. Deste modo, o presente estudo investigou a produção científica sobre o tema feita nos últimos 12 anos. Duzentos e três registros foram identificados e 18 deles cumpriram os critérios de inclusão. A avaliação das pesquisas incluídas nesta revisão apontou o comprometimento das funções executivas e a reabilitação neuropsicológica mostrou-se eficiente como estratégia de intervenção para adultos diagnosticados com TEPT. A ausência de informações sobre a intensidade dos sintomas presentes no TEPT foi uma limitação comum aos estudos encontrados nesta pesquisa.(AU)


Posttraumatic Stress Disorder (PTSD) is characterized by symptoms of uneasiness and fear when faced with direct and/or imagined situations related to experiences that threaten personal integrity. Executive Functions (EF) are processes defined by situational analysis, making decisions and taking actions to solve problems. It is known that PTSD is associated with dysfunctions in different dimensions of EF. Accordingly, the present study investigated the scientific production on the subject considering the last 12 years. A total of203 records were identified, with 18 of them meeting the inclusion criteria. The evaluation of the studies included in this review showed the impairment of EF and that neuropsychological rehabilitation has proven to be an efficient intervention strategy for adults diagnosed with PTSD. The absence of information about the intensity of symptoms present in PTSD was a common limitation of the studies found in this review.(AU)


El Trastorno de Estrés Postraumático (TEPT) se caracteriza por presentar síntomas de malestar y miedo ante situaciones directas y/o imaginadas en relación con experiencias que amenazan la integridad personal. Las Funciones Ejecutivas (FE) son procesos definidos por el análisis de una situación, toma de decisiones y acciones encaminadas a la resolución de problemas. Se sabe que el TEPT está asociado con disfunciones en diferentes dimensiones de la FE. De esta manera, el presente estudio investigó la producción científica sobre el tema en los últimos 12 años. Se identificaron 203 registros y 18 de ellos cumplieron los criterios de inclusión. La evaluación de los estudios incluidos en esta revisión mostró que el compromiso de las funciones ejecutivas, y la rehabilitación neuropsicológica, demostró ser eficiente como estrategia de intervención para adultos diagnosticados con TEPT. La ausencia de información sobre la intensidad de los síntomas presentes en el TEPT fue una limitación común de los estudios encontrados en esta investigación.(AU)

Persistent mating-induced endometritis (PMIE) results in decreased fertility in horses, thereby causing a significant impact in the horse market. Platelet-rich plasma (PRP), a modulator of the inflammatory response, has been largely used in veterinary medicine. Here, we investigated the effects of PRP on uterine inflammation, conception rate, endometrial polymorphonuclear neutrophil (PMN) migration, and COX-2 protein levels in the endometrial tissue. Thirteen PMIE-susceptible mares were used for artificial insemination (AI). The mares were inseminated with fresh semen in three consecutive cycles in a cross-over study design. The following cycle classifications were used: control cycle, no pharmacological interference; pre-AI, 20 mL of PRP was infused 24 h before AI; and post-AI, 20 mL of PRP was infused four h after AI. Follicular dynamics were monitored daily by transrectal ultrasound. When a follicle larger than 35 mm was detected, ovulation was induced with deslorelin acetate (1 mg, im). AI was performed 24 h after ovulation induction. Intrauterine fluid (FLU) was evaluated by ultrasonography before and 24 h after AI. PMNs in uterine cytology (CYT) and biopsy (HIS) were also observed before and 24 h after AI. Pregnancy was determined within 14 days after ovulation. Number of COX-2 positive cells was evaluated by immunohistochemistry. Both PRP treatments resulted in a decrease of PMNs in the CYT after breeding when compared to controls. FLU did not differ between cycles; however, the conception rates were significantly higher in the PRP mares. Mares positive for endometritis decreased in both treatment groups, and a more intense positive COX-2 labeling was observed in the control group when compared to the two treatment groups. In conclusion, PRP beneficially reduces inflammatory response in PMIE mares independent of when treatments were administered, thus increasing chances of successful pregnancy.

O insight cognitivo ou clínico refere-se à capacidade de atribuição de sentido aos sintomas presentes em transtornos psicóticos, especialmente naqueles relacionados ao espectro da esquizofrenia. A Beck Cognitive Insight Scale (BCIS) foi desenvolvida com a finalidade de auxiliar o tratamento de pessoas com o insight comprometido. No entanto, este instrumento ainda não foi devidamente adaptado ao contexto brasileiro. Este estudo teve como objetivo fazer a adaptação transcultural da BCIS para o português do Brasil por meio da evidência de validade de conteúdo. Seis juízes avaliaram os itens desta escala quanto à clareza da linguagem, pertinência prática, relevância teórica e relação item-dimensão. A BCIS apresentou concordância satisfatória nos índices de validade de conteúdo e homogeneidade das respostas referente à análise da clareza da linguagem, pertinência prática e relevância teórica e concordância entre item-dimensão teórica, com reformulação de alguns itens. A retrotradução da escala recebeu aval positiva de uma das autoras originais. No entanto, ainda é necessária a verificação das propriedades psicométricas desta versão da BCIS.


Cognitive or clinical insight refers to the ability to assign meaning to symptoms present in psychotic disorders, especially those related to the schizophrenia spectrum. The Beck Cognitive Insight Scale (BCIS) was developed to support the treatment of people with impaired insight. However, this instrument has not yet been adapted to the Brazilian context. This study aimed to make the cross-cultural adaptation of the BCIS into Brazilian Portuguese through evidence of content validity. Six judges evaluated the items of this scale regarding clarity of language, practical relevance, theoretical relevance and item-dimension relatedness. The BCIS presented satisfactory agreement in the indices of content validity and homogeneity of responses regarding the analysis of clarity of language, practical relevance and theoretical relevance and agreement between item-theoretical dimension, reformulating some items. The back-translation of the scale received a positive endorsement from one of the original authors. However, it is still necessary to verify the psychometric properties for this version of the BCIS.


El insight cognitivo o clínico se refiere a la capacidad de asignar un significado a los síntomas presentes en los trastornos psicóticos, especialmente aquellos relacionados con el espectro de la esquizofrenia. La Escala de Insight Cognitivo de Beck (BCIS) fue desarrollada para apoyar el tratamiento de personas con insight comprometido. Sin embargo, este instrumento todavía no ha sido adaptado al contexto brasileño. Este estudio tuvo como objetivo realizar la adaptación transcultural de la BCIS al portugués brasileño mediante pruebas de validez de contenido. Seis jueces evaluaron los ítems de esta escala en cuanto a la claridad del lenguaje, la relevancia práctica, la relevancia teórica y la relación ítem-dimensión. La BCIS presentó una concordancia satisfactoria en los índices de validez de contenido y homogeneidad de respuestas en cuanto al análisis de la claridad del lenguaje, relevancia práctica y relevancia teórica, y concordancia entre ítem-dimensión teórica, reformulando algunos de los ítems. La retraducción de la escala recibió una aprobación positiva de una de las autoras. Todavía es necesario verificar las propiedades psicométricas de esta versión de la BCIS.

RESUMO

Cognitive therapies comprise a widely recognized group of psychological interventions with considerable empirical support for different mental health conditions. These interventions can be used both in a traditional clinical setting and in crisis situations, especially those involving self-perception of overestimated risk, high vulnerability, and low coping capacity. The world is currently under a crisis, specifically, the COVID-19 pandemic, which can bring substantial impact on people's routine and cause significant psychological distress. In this context, telehealth interventions are becoming a standard practice and, thus, cognitive therapists could also support patients and people in the general population using this tool for the emerging issues. This text provides a brief overview of the main issues that may arise in the current pandemic situation as possible targets for the practice of cognitive therapists and the main evidence-based strategies that could be used in this context.(AU)


As terapias cognitivas compreendem um grupo amplamente reconhecido de intervenções psicológicas, com considerável suporte empírico para diferentes condições de saúde mental. Essas intervenções podem ser utilizadas tanto em ambiente clínico tradicional quanto em situações de crise, principalmente aquelas envolvendo autopercepção de risco superestimado, alta vulnerabilidade e baixa capacidade de enfrentamento. O mundo vive atualmente uma situação de crise, especificamente, a pandemia de COVID-19, que pode trazer um impacto substancial na rotina das pessoas e causar sofrimento psicológico significativo. Nesse contexto, as intervenções de cuidado online estão se tornando uma prática padrão e, assim, os terapeutas cognitivos também poderiam apoiar seus pacientes e pessoas da população em geral usando esta ferramenta para as questões emergentes. Este texto fornece uma breve visão geral das principais questões que podem surgir na atual situação de pandemia como possíveis alvos para a prática de terapeutas cognitivos e as principais estratégias baseadas em evidências que podem ser utilizadas por eles neste contexto.(AU)


Las terapias cognitivas comprenden un grupo ampliamente reconocido de intervenciones psicológicas, con considerable apoyo empírico para diferentes condiciones de salud mental. Estas intervenciones pueden utilizarse tanto en un entorno clínico tradicional como en situaciones de crisis, especialmente aquellas que implican un riesgo auto percibido sobrestimado, alta vulnerabilidad y baja capacidad de afrontamiento. El mundo atraviesa actualmente una situación de crisis, específicamente la pandemia de COVID-19, que puede tener un impacto sustancial en la rutina de las personas y causar una angustia psicológica significativa. En este contexto, las intervenciones de atención en línea se están convirtiendo en una práctica estándar y, por lo tanto, los terapeutas cognitivos también podrían ayudar a sus pacientes y personas en la población general utilizando esta herramienta para problemas emergentes. Este texto ofrece una breve descripción de los principales problemas que pueden surgir en la situación pandémica actual como posibles objetivos para la práctica de los terapeutas cognitivos y las principales estrategias basadas en la evidencia que pueden ser utilizadas por ellos en este contexto.(AU)

Degenerative changes of the endometrium are directly related to age and fertility in mares. Chronic degenerative endometritis (CDE) is correlated with uterine fluid retention and reduced ability to clear uterine inflammation. Recent research in the areas of equine surgery and sports medicine has shown that platelet-rich plasma (PRP) treatment acts as an immunomodulator of the inflammatory response. Therefore, the aim of this study was to determine if the uterine infusion of PRP could modulate the local inflammatory response and modify the intrauterine NO concentrations after artificial insemination (AI) in both normal mares and those with CDE. Thirteen mares with endometrium classified as grade III on the histology (mares with CDE) and eight mares with endometrial histological classification I or II-a normal mares were selected to investigate the effect of PRP therapy. The mares were inseminated with fresh semen in two consecutive cycles in a crossover study design. Thereby, each mare served as its own control and the treatment was performed with intrauterine PRP infusion four hours after AI. The percentage of neutrophils in uterine cytology (CIT, %), uterine fluid accumulation observed on ultrasonography (FLU, mm) and nitric oxide concentration of uterine fluid (NO, µM) were analyzed before and 24 hours after AI. The results reported that mares with CDE (CIT, 68.3 ± 3.27, FLU, 10.7 ± 1.61) have a higher (P < 0.05) intrauterine inflammatory response after AI than normal mares (CIT, 24.4 ± 3.56, FLU, 0), but NO concentrations did not differ (P > 0.05) between categories of mares. In treated cycles with PRP, the intrauterine inflammatory response decrease (P < 0.05) in CDE mares (CDE: CIT, 31.4 ± 6.48, FLU, 5.5 ± 1.28; normal mares: CIT, 13.5 ± 4.31, FLU, 0) when compared with nontreated cycle (CDE: CIT, 68.3 ± 3.27, FLU, 10.7 ± 1.61; NM: CIT, 24.4 ± 3.56, FLU, 0), but did not modify NO concentrations in uterine fluid. Thus, we can conclude that PRP was effective in modulating the exacerbated uterine inflammatory response to semen in mares with CDE but did not reduce NO concentrations in intrauterine fluid.

Fertility rates of donkey semen in jennies are lower compared to mares. The aims of this study were to evaluate different sperm cryopreservation methods and insemination strategies to improve the fertility of donkey semen in jennies. Three experiments were performed: (1) the comparison of two freezing methods of donkey semen (conventional method and automated method); (2) the determination of a suitable insemination dose of fresh donkey semen for jennies and mares; and (3) the influence of the semen deposition site on fertility of jennies inseminated with frozen donkey semen. For experiment 1, no differences were observed in total motility, angular velocity, curvilinear velocity, straight-line velocity, and plasma membrane integrity between samples frozen with the conventional (Styrofoam box) and the automated method (TK 4000C). However, the automated method provided higher values of progressive motility and rapid cells in frozen-thawed samples in comparison with the conventional method (P < 0.05). For experiment 2, mares were bred using 500 × 10(6) fresh sperm (M); and jennies using 1 × 10(9) (J1) or 500 × 10(6) fresh sperm (J5). Pregnancy rates in M, J1, and J5 were 93% (14/15), 73% (11/15), and 40% (6/15), respectively. When using different insemination doses, 500 × 10(6) or 1 × 10(9) sperm, no significant difference was observed in pregnancy rates of mares (M, 14/15) and jennies (J1, 11/15). Furthermore, there was no significant difference between the two insemination doses in jennies. However, with an insemination dose of 500 × 10(6) fresh sperm, the pregnancy rates were significantly higher in mares (M, 14/15) than in jennies (J5, 6/15; P < 0.05). For experiment 3, the inseminations were carried out in the uterine body (UB) or in the uterine horn of jennies with frozen-thawed donkey semen. No pregnancies were achieved with inseminations performed in the UB (0/12). The pregnancy rate for uterine horn group was 28.26% (13/46) and thus significantly higher than the UB group (0%; 0/12; P < 0.05). In conclusion, the automated method showed higher values on progressive motility and rapid cells parameters compared to the conventional method and can be used as an alternative for freezing donkey semen. The increase in the number of sperm cells per insemination dose using fresh donkey semen improved the fertility rates in jennies. The deep horn inseminations using frozen-thawed donkey semen increased the pregnancy rate in jennies, and the multiple inseminations may be an option to improve the fertility rates of donkey semen in jennies.

O estudo verificou a associação e a influência entre variáveis sociodemográficas, laborais, impactos da pandemia (desesperança, contaminação, óbito na família), traços de personalidade e de saúde mental em profissionais de saúde brasileiros em dois tempos distintos da pandemia de COVID-19. Foram incluídos 155 profissionais que responderam um questionário online. Foi utilizado o modelo de redes para a análise dos dados. Os sintomas de depressão, ansiedade e estresse foram os mais influentes no modelo testado e apresentaram associações com a desesperança nos dois tempos. No tempo 1, o estigma foi uma das variáveis mais influentes. No tempo 2, o estigma e a ansiedade reduziram sua influência, enquanto o estresse e a desesperança tornaram-se mais influentes. Os alvos das intervenções para os profissionais de saúde podem ser diferenciados no início e no avanço do contexto pandêmico, mas cabe contínua focalização do estresse e da desesperança


The study verified the association and influence between sociodemographic and labor variables, pandemic impacts (hopelessness, contamination, death in the family), personality and mental health traits in Brazilian health professionals at two different times of the COVID-19 pandemic. There were 155 professionals included who replied to an online questionnaire. The network model was used for data analysis. Symptoms of depression, anxiety, and stress were the most influential variables in the model tested and showed connections with hopelessness at both times. At time 1, stigma was one of the most influential variables. At time 2, stigma and anxiety reduced their influence, while stress and hopelessness became more prominent. The targets of interventions for health professionals can be differentiated in relation to the onset and progression of the pandemic context, but with a continuous focus on the level of stress and hopelessness for intervention


El estudio verificó la asociación e influencia entre variables sociodemográficas y laborales, impactos de la pandemia (desesperanza, contaminación, muerte en la familia), rasgos de personalidad y salud mental en trabajadores sanitarios brasileños en dos momentos de la pandemia del COVID-19. Se incluyeron 155 profesionales que respondieron a un cuestionario en línea. Se utilizó el modelo de red para el análisis. Los síntomas de depresión, ansiedad y estrés fueron las variables más influyentes y mostraron conexiones con la desesperanza en ambos momentos. En el momento 1, el estigma fue una de las variables más influyentes. En el momento 2, el estigma y la ansiedad redujeron su influencia y el estrés y la desesperanza se hicieron más prominentes. Los objetivos de las intervenciones para los trabajadores sanitarios se pueden diferenciar en el inicio y en el avance de la pandemia, pero es necesario un enfoque continuo en el estrés y desesperanza

Boar semen cryopreservation remains a challenge due to the extension of cold shock damage. Thus, many alternatives have emerged to improve the quality of frozen-thawed boar sperm. Although the use of seminal plasma arising from boar sperm-rich fraction (SP-SRF) has shown good efficacy; however, the majority of actual sperm evaluation techniques include a single or dual sperm parameter analysis, which overrates the real sperm viability. Within this context, this work was performed to introduce a sperm flow cytometry fourfold stain technique for simultaneous evaluation of plasma and acrosomal membrane integrity and mitochondrial membrane potential. We then used the sperm flow cytometry fourfold stain technique to study the effect of SP-SRF on frozen-thawed boar sperm and further evaluated the effect of this treatment on sperm movement, tyrosine phosphorylation and fertility rate (FR). The sperm fourfold stain technique is accurate (R2 = 0.9356, p > 0.01) for simultaneous evaluation of plasma and acrosomal membrane integrity and mitochondrial membrane potential (IPIAH cells). Centrifugation pre-cryopreservation was not deleterious (p > 0.05) for any analyzed variables. Addition of SP-SRF after cryopreservation was able to improve total and progressive motility (p < 0.05) when boar semen was cryopreserved without SP-SRF; however, it was not able to decrease tyrosine phosphorylation (p > 0.05) or improve IPIAH cells (p > 0.05). FR was not (p > 0.05) statistically increased by the addition of seminal plasma, though females inseminated with frozen-thawed boar semen plus SP-SRF did perform better than those inseminated with sperm lacking seminal plasma. Thus, we conclude that sperm fourfold stain can be used to simultaneously evaluate plasma and acrosomal membrane integrity and mitochondrial membrane potential, and the addition of SP-SRF at thawed boar semen cryopreserved in absence of SP-SRF improve its total and progressive motility.

The purpose of the present study was to compare two protocols for equine frozen semen programs using either postovulation insemination or fixed-time insemination (FT), evaluating both pregnancy rates and intrauterine fluid (IUF) accumulation after artificial insemination with semen obtained from either highly or poorly fertile stallions. Six ejaculates from two stallions (n = 12) were processed. After thawing, semen samples were evaluated by computerized semen analysis. Fifteen mares (30 cycles) were inseminated with frozen semen from highly fertile stallion A, and 14 mares (28 cycles) were inseminated with frozen semen from poorly fertile stallion B. Ovulations were induced with 1 mg (intramuscular) of deslorelin acetate after the observation of a greater than 35 mm follicle and uterine edema. In postovulation insemination group, mares were inseminated once with 800 × 10(6) total sperm in a maximum 6-hour interval after ovulation. In FT group, mares were inseminated twice with 400 × 10(6) total sperm, 24 and 40 hours after induction. Mares were ultrasonographically examined for IUF accumulation 24 hours and for pregnancy diagnosis 14 days after the last insemination. Although IUF accumulation was more evident in mares inseminated once postovulation, pregnancy rates were similar for both protocols, regardless of the stallion, although a significant effect of the stallion was observed. These results indicated that FTs may be used for both highly and poorly fertile stallions as a practical tool to help spreading the use of frozen semen in equine reproduction programs.

The objective of this study was to determine whether replacing the egg yolk with soybean lecithin in the Botu-Crio® cryodiluent would maintain the fertility of cryopreserved stallion sperm. Two experiments were performed to evaluate cell freezability. In experiment 1, sperm from 15 stallions were frozen in Botu-Crio® (BC) or Botu-Crio® which contained 45g/L soybean lecithin (BCLS45) in place of the egg yolk. In experiment 2, we compared different concentrations of soybean lecithin: 0, 10.0, 12.5, 15.0, 17.5 and 20.0g/L (BC, BCLS10, BCLS12.5, BCLS17.5 and BCLS20, respectively). In experiment 1, sperm frozen in BC and BCLS45 exhibited similar (P>0.05) percentages of total motile sperm (61% and 61%, respectively); progressively motile sperm (27% and 27%, respectively) and sperm with intact plasma membranes (IMP; 53% and 57%, respectively). Similarly, sperm frozen in BC or BC containing any concentration of soybean lecithin maintained similar (P>0.05) percentages of total motile sperm (61-68%) and progressively motile sperm (27-31%). In the first fertility trial, we used cryopreserved semen from a single stallion was inseminated into mares. The semen from the sperm that were frozen in BC diluent resulted in a higher fertility rate (66%, 16/24) compared to the sperm that were frozen in BCLS45 diluent (17%, 5/29; P<0.01). Similarly, in a second fertility trial, the mares that were inseminated with the sperm that were frozen in BC diluent exhibited a higher fertility rate (66%, 16/24) compared to the mares that were inseminated with the sperm that were frozen in BCLS20 (40%, 10/25; P<0.05). Finally, in a third trial, the sperm that were frozen in BC resulted in a higher fertility rate in mares (75%, 18/24) compared to the sperm that were frozen in BCLS10 (41%, 10/24; P<0.05). Although replacing the egg yolk in the BC cryodiluent with soybean lecithin provided similar laboratory results for stallion sperm, after cryopreservation, the sperm that was frozen with soybean lecithin in the diluent correlated with lower fertility rates. Based on these results, we concluded that the use of BCLS can be used as an alternative diluent for cryopreserving stallion sperm. However, the resulting reduced fertility rate is a matter of concern. Further studies are necessary to clarify the reasons for this decrease in fertility and to determine the optimal lecithin concentration for diluents to freeze stallion sperm.
