RESUMEN

A new chromone derivative, aspergione A (1), along with seven known metabolites, was isolated from a mangrove endophytic fungus, Aspergillus sp. GXNU-B1, which was collected from mangrove Acanthus ilicifolius L. Their structures and the absolute configuration of 1 were elucidated based on the analysis of HR-ESI-MS, NMR, and ECD calculation. Compounds 1-8 were evaluated for their anti-inflammatory effects on the production of nitricoxide (NO). Compounds 1 and 8 have potent inhibitory effects against NO production in activated macrophages with IC50 values of 38.26 and 44.30 µM, respectively.

Chemical investigation of the fermentation extract of the mangrove endophytic fungus Aspergillus sp. GXNU-A1, isolated from Acanthus ilicifolius L., discovered an undescribed pair of enantiomers (asperphenyltones A and B (±1)), together with four previously described metabolites: nodulisporol (2), isosclerone (3), 2,3,4-trihydroxy-6-(hydroxymethyl)-5-methylbenzyl alcohol (4), and 4,6-dihydroxy-5-methoxy-7-methyl-1,3-dihydroisobenzofuran (5). Analyses of the 1D and 2D NMR spectroscopic data of the compounds supported their structural assignments. The presence of the asperphenyltones A and B, which are a pair of enantiomers, was established by HR-ESI-MS, 1D and 2D NMR data and confirmed by single-crystal X-ray diffraction analysis. Metabolites 1-5 were evaluated for their anti-inflammatory effects on the production of nitric oxide (NO), and 1, 3, and 4 showed significant potential inhibitory activities against NO production in activated macrophages with IC50 values of 26-40 µM, respectively.

RESUMEN

Transcatheter arterial chemoembolization (TACE) is the common choice of non-open surgery for hepatocellular carcinoma (HCC) now. In this study, a simple TACE robotic system of 4-degree-of-freedom is proposed to get higher accuracy and stability of the surgery operation and reduce X-ray exposure time of the surgeons. The master-slave control strategy is adopted in the robotic system and a customized sigmoid function is designed to optimize the joystick control of the master-slave robotic control system. A force-sensing module is developed to sense the resistance of the guide wire in linear delivery motion and an auxiliary bending feedback method based on constraint pipe with a film sensor is proposed. With two force-sensing methods, the safety strategy of robotic motion with 9 different motion constraint coefficients is given and a human-computer interface is developed. The TACE robot would monitor the value of the force sensor and the analog voltage of the film sensor to adopt the corresponding motion constraint coefficient in every 10 ms. Vascular model experiments were performed to validate the robotic system, and the results showed that the safety strategy could improve the reliability of the operation with immediate speed constraint and avoid potential aggressive delivery.

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous environmental contaminants with a number of them being carcinogenic. One of the approaches to assess human exposure to PAHs is to measure their urinary metabolites, monohydroxyl polycyclic aromatic hydrocarbons (OH-PAHs), with a method allowing for high throughput and short turn-around time. We developed a method to quantify nine urinary OH-PAHs by using supported liquid phase extraction (SLE) and isotope dilution gas chromatography tandem mass spectrometry (GC-MS/MS). SLE demonstrated advantages over the traditionally used liquid-liquid extraction techniques. The target analytes with spiked deuterated and 13C-labeled internal standards were extracted from urine by SLE after enzymatic cleavage of the glucuronide and sulfate conjugates. The extracted analytes were then derivatized with N-Methyl-N-(trimethylsilyl) trifluoroacetamide (MSTFA), and analyzed by GC-MS/MS. Six solvent mixtures were evaluated as the SLE extraction solvent, and pentane:chloroform (7:3, v/v) was selected due to its best overall analytical performance. Method detection limits for the 9 analytes ranged from 2.3 to 13.8 pg/mL. Precision and accuracy were satisfactory. SLE and internal isotope labeled standard combination reduced matrix effect effectively. This new method using SLE sample preparation techniques coupled with GC-MS/MS proves applicable to urinary measurements for PAH exposure studies for general population.

Essentially all women are exposed to polycyclic aromatic hydrocarbons (PAHs), formed during incomplete combustion of organic materials, including fossil fuels, wood, foods, and tobacco. PAHs are ovarian toxicants in rodents, and cigarette smoking is associated with reproductive abnormalities in women. Biomonitoring of hydroxylated PAH (OH-PAH) metabolites in urine provides an integrated measure of exposure to PAHs via multiple routes and has been used to characterize exposure to PAHs in humans. We hypothesized that concentrations of OH-PAHs in urine are associated with reproductive function in women. We recruited women 18-44years old, living in Orange County, California to conduct daily measurement of urinary luteinizing hormone (LH) and estrone 3-glucuronide (E13G) using a microelectronic fertility monitor for multiple menstrual cycles; these data were used to calculate endocrine endpoints. Participants also collected urine samples on cycle day 10 for measurement of nine OH-PAHs. Models were constructed for eight endpoints using a Bayesian mixed modeling approach with subject-specific random effects allowing each participant to act as a baseline for her set of measurements. We observed associations between individual OH-PAH concentrations and follicular phase length, follicular phase LH and E13G concentrations, preovulatory LH surge concentrations, and periovulatory E13G slope and concentration. We have demonstrated the feasibility of using urinary reproductive hormone data obtained via fertility monitors to calculate endocrine endpoints for epidemiological studies of ovarian function during multiple menstrual cycles. The results show that environmental exposure to PAHs is associated with changes in endocrine markers of ovarian function in women in a PAH-specific manner.

An optoelectronic oscillator (OEO) with wideband frequency tunability and stable output based on a bandpass microwave photonic filter (MPF) has been proposed and experimentally demonstrated. Realized by cascading a finite impulse response (FIR) filter and an infinite impulse response (IIR) filter together, the tunable bandpass MPF successfully replaces the narrowband electrical bandpass filter in a conventional single-loop OEO and serves as the oscillating frequency selector. The FIR filter is based on a tunable multi-wavelength laser and dispersion compensation fiber (DCF) while the IIR filter is simply based on an optical loop. Utilizing a long length of DCF as the dispersion medium for the FIR filter also provides a long delay line for the OEO feedback cavity and as a result, optical tuning over a wide frequency range can be achieved without sacrificing the quality of the generated signal. By tuning the wavelength spacing of the multi-wavelength laser, the oscillation frequency can be tuned from 6.88 GHz to 12.79 GHz with an average step-size of 0.128 GHz. The maximum frequency drift of the generated 10 GHz signal is observed to be 1.923 kHz over 1 hour and its phase noise reaches the -112 dBc/Hz limit of our measuring equipment at 10 kHz offset frequency.

The aim of this study is to develop and validate an analytical method for the quantitation of ten urinary monohydroxylated polycyclic aromatic hydrocarbons (OH-PAHs) through high pressure liquid chromatography/tandem mass spectrometry (HPLC/MS/MS). After enzymatic deconjugation, urine samples were extracted by liquid-liquid extraction (LLE) and OH-PAHs were analyzed by HPLC/MS/MS operated in negative electrospray ionization (ESI) and multiple reaction monitoring (MRM) mode. LLE was conducted with the solvent mixture of pentane and toluene, which reduced the matrix interferences and enhanced the method sensitivity significantly. Deuterated and (13)C-labeled analogs are used as internal standards. Calibration curves of all target analytes shows favorable linearity within the concentration range of 5.9-15,000.0 ng/L for different OH-PAHs with the regression coefficients above 0.993. The limits of detection (LODs) in pooled urine ranged from 1.72 to 17.47 ng/L, which were much lower than those obtained by a gas chromatography/high resolution mass spectrometry (GC/HRMS) method. The method shows satisfactory accuracy and precision when analyzing three different levels of OH-PAHs spiked in pooled urine. Except for 1-hydroxynaphthalene, recoveries of other OH-PAHs were in the range of 100 ± 20% with a variation coefficient of less than 13%. The measurement of OH-PAHs from a QC sample of the Centers for Disease Control and Prevention (CDC) generated results close to the values measured by CDC. This method has been successfully employed in the California Biomonitoring Program.

RESUMEN

A photonic approach for microwave frequency measurement is proposed. In this approach, an optical carrier is modulated by an unknown microwave signal through a phase modulator. The modulated optical signal is then split into two parts; one part passes through a spool of polarization maintaining fiber (PMF) and the other one, through a dispersion compensation fiber (DCF), to introduce different microwave power penalties. After the microwave powers of the two parts are measured by two photodetectors, a fixed frequency-to-power mapping is established by obtaining an amplitude comparison function (ACF). A proof-of-concept experiment demonstrates frequency measurement over a range of 10.5 GHz, with measurement error less than +/-0.07 GHz.

RESUMEN

We develop new expressions for power fading and average power fading that are induced by first-order polarization mode dispersion, polarization-dependent chromatic dispersion, chromatic dispersion, and chirp under both (i) carrier suppressed modulation and (ii) odd-order optical sideband and carrier suppression. Experimental results show the effectiveness of the theoretical analysis. Further, based on the expressions, we propose a technique for optically compensating the polarization mode dispersion-induced power fading in carrier suppressed modulation.
