RESUMO

The timely and accurate diagnosis of candidemia, a severe bloodstream infection caused by Candida spp., remains challenging in clinical practice. Blood culture, the current gold standard technique, suffers from lengthy turnaround times and limited sensitivity. To address these limitations, we propose a novel approach utilizing an Electronic Nose (E-nose) combined with Time Series-based classification techniques to analyze and identify Candida spp. rapidly, using culture species of C. albicans, C.kodamaea ohmeri, C. glabrara, C. haemulonii, C. parapsilosis and C. krusei as control samples. This innovative method not only enhances diagnostic accuracy and reduces decision time for healthcare professionals in selecting appropriate treatments but also offers the potential for expanded usage and cost reduction due to the E-nose's low production costs. Our proof-of-concept experimental results, carried out with culture samples, demonstrate promising outcomes, with the Inception Time classifier achieving an impressive average accuracy of 97.46% during the test phase. This paper presents a groundbreaking advancement in the field, empowering medical practitioners with an efficient and reliable tool for early and precise identification of candidemia, ultimately leading to improved patient outcomes.

This study aimed to resveratrol supplementation (at 5 or 10 mg/kg) and a hydroethanolic extract of canjiqueira fruits (150 mg/kg) on female SWISS mice. Total cholesterol, high-density lipoprotein (HDL), triglyceride levels, gestation rates, and embryonic implantation rates in their female Offspring was evaluated. In conclusion, the consumption of canjiqueira fruit extract altered the lipid profile of their female offspring, and did not impact their reproductive performance. Supplementing female SWISS mice with 10 mg/kg of resveratrol increased total cholesterol, triglycerides, and HDL levels, thereby enhancing the reproductive efficiency of their offspring.

Coralsnakes of the genus Micrurus are a diverse group of venomous snakes ranging from the southern United States to southern South America. Much uncertainty remains over the genus diversity, and understanding Micrurus systematics is of medical importance. In particular, the widespread Micrurus nigrocinctus spans from Mexico throughout Central America and into Colombia, with a number of described subspecies. This study provides new insights into the phylogenetic relationships within M. nigrocinctus by examining sequence data from a broad sampling of specimens from Mexico, Guatemala, Honduras, Nicaragua, Costa Rica, and Panama. The recovered phylogenetic relationships suggest that M. nigrocinctus is a species complex originating in the Pliocene and composed of at least three distinct species-level lineages. In addition, recovery of highly divergent clades supports the elevation of some currently recognized subspecies to the full species rank while others may require synonymization.

BACKGROUND: Two experiments were conducted to establish an optimal NE challenge model and evaluate the efficacy of stimbiotic (STB) supplementation in necrotic enteritis (NE) challenged broilers. In Exp. 1, a total of 120 Arbor Acres (AA) broilers (45.0 ± 0.21 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Vaccine treatments included non-challenge (0), × 10 the recommended dose (× 10) or × 20 the recommended dose (× 20) by the manufacturer. Clostridium perfringens (CP) treatments were non-challenge (No) or 3 mL of 2.2 × 107 CFU CP challenge (Yes). In Exp. 2, a total of 72 AA broilers (40.17 ± 0.27 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Dietary treatments included non-additive (CON), 100 mg/kg STB (STB) and 100 mg/kg STB on top of a typical commercial blend including an essential oil, probiotics, and enzyme (CB). Challenge treatments included non-NE challenge (No) and NE challenge (Yes) as established in Exp. 1. RESULTS: In Exp. 1, CP and vaccine challenge decreased (P < 0.05) body weight (BW), body weight gain (BWG) and feed intake (FI), and increased (P < 0.05) the number of broilers with diarrhea and intestinal lesions. The oral administration of × 20 recommended dose of vaccines coupled with 3 mL of 2.2 × 107 CFU CP resulted in (P < 0.01) a significantly increased incidence of wet litter and intestinal lesions. Thus, this treatment was chosen as the challenge model for the successful inducement of NE in Exp. 2. In Exp. 2, the NE challenge negatively affected (P < 0.01) growth performance, ileal morphology, immunoglobulin contents in blood, caecal microbiota in the caecum, footpad dermatitis, intestinal lesion scores, tumour necrosis factor (TNF-α) and endotoxin in the serum compared with the non-NE challenged birds. The supplementation of STB and CB in diets enhanced (P < 0.05) growth performance, intestinal microbiota, and blood profiles by stimulating ileal morphology (VH and VH:CD) and propionate production in the cecum, and there were no differences in measured variables between STB and CB supplemented birds. CONCLUSION: Overall, these results indicate that STB supplementation was able to reduce the inflammatory response and improve the performance of NE challenged birds, and the supplementation of STB alone was as effective as a typical commercial blend containing a number of other additives.

Comprehensive assessments of species' extinction risks have documented the extinction crisis1 and underpinned strategies for reducing those risks2. Global assessments reveal that, among tetrapods, 40.7% of amphibians, 25.4% of mammals and 13.6% of birds are threatened with extinction3. Because global assessments have been lacking, reptiles have been omitted from conservation-prioritization analyses that encompass other tetrapods4-7. Reptiles are unusually diverse in arid regions, suggesting that they may have different conservation needs6. Here we provide a comprehensive extinction-risk assessment of reptiles and show that at least 1,829 out of 10,196 species (21.1%) are threatened-confirming a previous extrapolation8 and representing 15.6 billion years of phylogenetic diversity. Reptiles are threatened by the same major factors that threaten other tetrapods-agriculture, logging, urban development and invasive species-although the threat posed by climate change remains uncertain. Reptiles inhabiting forests, where these threats are strongest, are more threatened than those in arid habitats, contrary to our prediction. Birds, mammals and amphibians are unexpectedly good surrogates for the conservation of reptiles, although threatened reptiles with the smallest ranges tend to be isolated from other threatened tetrapods. Although some reptiles-including most species of crocodiles and turtles-require urgent, targeted action to prevent extinctions, efforts to protect other tetrapods, such as habitat preservation and control of trade and invasive species, will probably also benefit many reptiles.

Analysis of digital images by smartphone was used for copper quantification in sugarcane spirit (cachaça) samples through the formation of blue complex between copper and cuprizone. An experimental design was carried out to evaluate the best complexation reaction conditions. Moreover, different image acquisition procedures (external camera coupled to a smartphone or the smartphone camera) with different regions of interest sizes, distances in image acquisition, and concentration ranges of the calibration curve and the influence of processing the curve in univariate and multivariate modes, by PLS, were evaluated. The results obtained in three real samples and two spikes were compared with those of UV-Vis spectrophotometry, used as a reference method, and they show the potential of the proposed method for the accurate determination of copper. When compared to traditional techniques, the proposed method has the advantages of portability and low cost in addition to requiring a smaller amount of reagents.

RESUMO

We describe a new species of gymnophthalmid lizard, Oreosaurus bisbali sp. nov., from the Turimiquire Massif in northeastern Venezuela. It is one of three reptile species known to be endemic to the region and the fifth species of Oreosaurus from Venezuela. It can be distinguished from all congeners by a combination of rectangular dorsal scales at midbody, having two scales separating femoral pores medially, and lacking a nasoloreal suture. The new species is likely to be restricted in distribution to the highlands of the Turimiquire Massif, which have been significantly degraded by agriculture.

We examine, for the first time, biogeographic patterns in a series of tropical montane coastal systems in northern South America. We use amphibians and reptiles, which constitute the most critical communities based upon the prevalence of endemic taxa, to assess the region's biodiversity. The montane coastal system spans an east-west distance of 925 km. It includes peaks ranging from 549 m to 2765 m above sea level and encompasses the montane complexes of northern Venezuela (including Isla de Margarita), an outlier at Santa Marta (Colombia), and ranges on the islands Trinidad and Tobago. The area supports 14 family level amphibian clades and 23 family level reptile clades. Fieldwork, museum specimen surveys, and a literature review suggest that biodiversity decreases at higher elevations. Here we examine the biogeographic patterns in the region to assess the role of the montane systems as possible refugia. We also look at the possible island and sky island effects using data from altitudes >200 m. At lower elevations, we tabulated 294 species, comprising 112 amphibians and 182 reptiles. About 45% of these taxa are endemic or exclusive to different sub-regions. At mid-elevation montane cloud forests, we find a much-reduced biodiversity with a total of 125 species (66 amphibians and 59 reptiles) exclusive or restricted to the region, and few species shared between systems. We find that biogeographical patterns follow a natural topographic disposition above 200 m in elevations. At the lower elevation cut off, there are 118 species (26 amphibians and 92 reptiles) shared among two or more of the studied mountain systems, suggesting a common origin and dispersal events, despite what seem to be topographic barriers. Biogeographical relationships support a topographic disposition of the region with close associations between the islands of Trinidad and Tobago, the Paria Range and the Turimiquire Massif, and close associations between the Sierra Nevada de Santa Marta and the Sierra de San Luis. Overall, the biogeographic relationships between amphibians and reptiles are similar. Species diversity in the eastern Caribbean region is less rich than in the west. This study includes the first herpetological surveys at the two easternmost mountains (Cerro La Cerbatana and Campeare) belonging to the Paria Range biogeographic unit, and aims to contribute to a better understanding of the rich biodiversity of the region.

Periapical Radiographs are commonly used to detect several anomalies, like caries, periodontal, and periapical diseases. Even considering that digital imaging systems used nowadays tend to provide high-quality images, external factors, or even system limitations can result in a vast amount of radiographic images with low quality and resolution. Commercial solutions offer tools based on interpolation methods to increase image resolution. However, previous literature shows that these methods may create undesirable effects in the images affecting the diagnosis accuracy. One alternative is using deep learning-based super-resolution methods to achieve better high-resolution images. Nevertheless, the amount of data for training such models is limited, demanding transfer learning approaches. In this work, we propose the use of super-resolution generative adversarial network (SRGAN) models and transfer learning to achieve periapical images with higher quality and resolution. Moreover, we evaluate the influence of using the transfer learning approach and the datasets selected for it in the final generated images. For that, we performed an experiment comparing the performance of the SRGAN models (with and without transfer learning) with other super-resolution methods. Considering Mean Square Error (MSE), Peak Signal to Noise Ratio (PSNR), Structural Similarity Index (SSIM), and Mean Opinion Score (MOS), the results of SRGAN models using transfer learning were better on average. This superiority was also verified statistically using the Wilcoxon paired test. In the visual analysis, the high quality achieved by the SRGAN models, in general, is visible, resulting in more defined edges details and fewer blur effects.

This work proposes a simple device for thermometric titration using a contactless sensor. A low-cost infrared sensor (around $5) was adapted in a disposable, polystyrene vessel for monitoring the temperature to determine the endpoint of neutralization, redox, and complexation titrations, using a homemade syringe pump to titration control. Open-source software was developed to control the device using the Raspberry Pi platform and to perform automatic endpoint determination. The influence of sample volume, the flow rate of titrant, and sensor distance from the solution were evaluated to lower the variation among measurements. The parameters chosen in this regard were 10 mL of sample, 0.8 mL min-1 of titrant solution flow rate, and 3 cm of distance from the sensor to the solution. Results for the determination of the acidity of sauces and the ascorbic acid and calcium content of pharmaceutical products were compared with those from official compendia. The performance of the sensor also was compared with an infrared camera. The proposed method agreed with conventional ones to an extent ranging from 93% to 106%. The robust analytical performance and low cost of the system are essential features that could broaden the use of enthalpimetric analysis in several laboratories.

The use of the solar photo-Fenton process for water treatment requires monitoring of the main conditions, especially the total dissolved iron concentration and the consumption of hydrogen peroxide. In this study, a new methodology using the PhotoMetrixPRO application was validated for rapid monitoring of total dissolved iron and hydrogen peroxide concentrations, and was tested in the solar photo-Fenton process. A comparison was made between the results obtained using a reference spectrophotometric method and the PhotoMetrixPRO application employing a portable device. Both methods were validated in terms of linearity, sensitivity, precision, robustness, and matrix effects. The degree of dispersion between the series of measurements obtained using UV-vis and portable device tool was low and was in compliance with the established Brazilian and ICH validation criteria. Additionally, PhotoMetrixPRO enabled the use of a smaller sample volume. The total volume generated of each sample is 1 mL, reducing 6 and 10 times the wastes produced in different validated methods. These results evidencing that the miniaturization can provide positive advantages in terms of simplicity, cost effectiveness, and less environmental impact. PhotoMetrixPRO offers significant advantages including rapid analysis, smaller sample volumes, and greater portability and accessibility.

Tobago is a small island on the southeast edge of the Caribbean Plate with a continental flora and fauna. Using DNA sequences from Genbank, new sequences, and morphological data from the snakes Erythrolamprusepinephalus, E.melanotus, E.reginae, and E.zweifeli, the species status of specimens of a Tobago snake previously considered to be Erythrolamprusreginae was assessed. Erythrolampruszweifeli, long considered a subspecies of E.reginae, was found to be a northern Venezuela-Trinidad endemic and the sister to E.reginae. The trans-Andean species E.epinephalus is shown to be non-monophyletic while the Costa Rican lineage of E.epinephalus is weakly supported as the sister to the Tobago population. The Tobago Erythrolamprus is described as a distinct taxon based upon five specimens from four localities in lower montane rainforest. Much of the new species range includes the Main Ridge Forest Reserve of Tobago, the oldest protected forest in the Western Hemisphere. All known locations fall within a 400-ha area, and its total geographic distribution is likely to be less than 4,566 ha. The restricted distribution of this new snake makes it a likely candidate for threatened status. The new species also becomes another biogeographic link between northern Venezuela and Tobago.

Reproductive diseases, mainly endometritis, are important hurdles in cattle raising, In the current study we evaluated gross, bacteriological, cytological, and histological findings from selected sites of the genital from 23 slaughtered cows and tested whether there is an association between these findings and the probability of reaching a reliable diagnosis. The results from the examinations of macroscopic aspects of uterine secretions, the cytological, bacteriological, and histopathological findings were then correlated. There was no significant correlation (P>0.05) of the statistical data from different parts of the genital tract. Trueperella pyogenes and Escherichia coli were isolated from the vagina in 3/23 cases. In only 2/23 samples Enterococcus faecalis and a gram-negative, oxidase-positive bacteria were isolated from the cervix uteri. Only Staphylococcus epidermidis, in 1/23 case, was isolated from the uterus. Histopathological findings in uterus from samples of Groups II (moderate lesions) and III (severe lesions) did not translated in grossly visible changes. Samples from reproductive tracts with secretion in the vagina and cervix uteri had no detectable changes in the other parameters analyzed from this portion. Uterus with positive bacterial culture had evidence of ascendant inflammation judging by the high granulocyte count in the three analyzed portions. This study reinforces that vaginitis and cervicitis in the cow diagnosed only by clinical examination does not reflect the real status of the uterine health. For this reason, treatment of uterine disorders should be conducted based on reliable tests to determine the appropriate therapy for each situation.(AU)


Doenças reprodutivas causam altas perdas econômicas nos rebanhos bovinos, sendo a endometrite uma das alterações mais relevantes. Os objetivos desta pesquisa foram avaliar a associação dos achados fisiopatológicos em exames macroscópicos, bacteriológicos, citológicos e histopatológicos nas porções do trato genital de 23 vacas abatidas, bem como avaliar a necessidade de associação destes exames para efetuar diagnóstico fidedigno. A avaliação macroscópica da secreção, os exames histopatológico, citológico e as bactérias identificadas foram correlacionados. Não houve associação (P>0,05) dos resultados nas diferentes porções do trato genital. Na vagina foram isoladas as bactérias Trueperella pyogenes e Escherichia coli. Na cérvix, em apenas 2/23 (8,6%) amostras isolou-se Enterococcus faecalis e gram negativo oxidase positiva. No útero houve isolamento apenas da bactéria Staphylococcus epidermidis. As amostras histopatológicas classificadas em grupo II e III não apresentaram alterações detectadas no exame macroscópico. As amostras com secreção não fisiológica na vagina e cérvix não apresentaram alterações nos outros exames na porção uterina. A amostra com cultura bacteriológica positiva no útero demonstrou uma infecção ascendente através da alta contagem de granulócitos nas três porções analisadas. O presente estudo reforça o conceito de que a vaginite e cervicite diagnosticadas pelo exame clinico na vaca não retratam o real status da saúde uterina e por esta razão, o tratamento do útero deve ser conduzido com critério, alicerçado nos exames complementares para definir a terapia adequada para cada situação.(AU)

Riama is the most speciose genus of the Neotropical lizard family Gymnophthalmidae. Its more than 30 montane species occur throughout the northern Andes, the Cordillera de la Costa (CC) in Venezuela, and Trinidad. We present the most comprehensive phylogenetic analysis of Riama to date based on a total evidence (TE) approach and direct optimization of molecular and morphological evidence. Analyses use DNA sequences from four loci and 35 phenotypic characters. The dataset consists of 55 ingroup terminals representing 25 of the 30 currently recognized species of Riama plus five undescribed taxa, including an endemic species from the Sierra Nevada de Santa Marta (SNSM) in Colombia, and 66 outgroup terminals of 47 species. Analysis results in a well-supported hypothesis in which Riama is polyphyletic, with its species falling into three clades. The Tepuian Anadia mcdiarmidi nests within one clade of Riama, and the recently resurrected Pantodactylus nests within Cercosaura. Accordingly, we propose a monophyletic taxonomy that reflects historical relationships. Analysis of character evolution indicates that the presence/absence of prefrontals-a cornerstone of the early genus-level taxonomy of cercosaurines-is optimally explained as having been plesiomorphically present in the most recent common ancestor of Cercosaurinae and lost in that of the immediately less inclusive clade. Multiple independent reversals to present and subsequent returns to absent occur within this clade. To evaluate the impact of phenotypic evidence on our results, we compare our TE results with results obtained from analyses using only molecular data. Although phenotypic evidence comprises only 1.2% of the TE matrix, its inclusion alters both the topology and support values of the clades that do not differ. Finally, current phylogenetic evidence reveals a SNSM-CC-Trinidad-tepuis biogeographical link. We hypothesize that an ancient connection facilitated the exchange of species between the SNSM and the CC.

OBJECTIVE: Hydroxyapatite is used as a drug-delivery system for bone therapy applications because of its biocompatibility, bioactivity, and osteoconductive properties. In addition, hydroxyapatite nanoparticles (HApN) might be used as a theranostic probe. The aim of this study was to prepare and characterize hydroxyapatite mesoporous nanoparticles, and radiolabel these nanoparticles with technetium-99m (Tc). Moreover, biodistribution studies were carried out in healthy mice. MATERIALS AND METHODS: HApN were synthesized and characterized. Tc-HApN was prepared by adding Tc-pertechnetate to a dispersion of HApN in the presence of stannous chloride. Radiochemical purity and in-vitro stability were determined. The circulation time of Tc-HApN was determined by measuring blood radioactivity in healthy mice. In addition, biodistribution studies were carried out in healthy mice at 1 and 4 h after injection. RESULTS: Tc-HApN showed high radiochemical purity (98.7±0.2%) and in-vitro stability until 24 h. Tc-HApN levels in blood decreased in a biphasic manner, with an α half-life of 1.8 min and a ß half-life of 126.9 min. High uptake was achieved in the liver and spleen because of the macrophage uptake. Furthermore, bone uptake was higher than that of the surrounding muscle, resulting in high bone-to-muscle ratios. CONCLUSION: HApN were synthesized successfully with suitable characteristics for in-vivo applications. Tc-HApN was prepared and showed high stability. Tc-HApN presented increasing bone uptake over time, showing a higher affinity to bone tissues in contrast to surrounding muscle. The present results, together with further studies, may indicate a potential application of HApN as a nanocarrier for bone diseases.

The developing world often lacks the resources to effectively treat the most serious injuries including osteomyelitis following open fractures or surgical fracture treatment. Antibiotic cement beads are a widely accepted method of delivering antibiotics locally to the infected area following trauma. This study is based in Cambodia, a low income country struggling to recover from a recent genocide. The study aims to test the effectiveness of locally made antibiotic beads and analyse their effectiveness after being gas sterilised, packaged and kept in storage Different antibiotic beads were manufactured locally using bone cement and tested against MRSA bacteria grown from a case of osteomyelitis. Each antibiotic was tested before and after a process of gas sterilisation as well as later being tested after storage in packaging up to 42 days. The gentamicin, vancomycin, amikacin and ceftriaxone beads all inhibited growth of the MRSA on the TSB and agar plates, both before and after gas sterilisation. All four antibiotics continued to show similar zones of inhibition after 42 days of storage. The results show significant promise to produce beads with locally obtainable ingredients in an austere environment and improve cost effectiveness by storing them in a sterilised condition.

INTRODUÇÃO: A área luminal mínima (ALM), obtida pelo ultrassom intracoronário (USIC), é preditora de eventos em pacientes com lesões de moderada gravidade, identificadas à angiografia coronária. Por outro lado, a presença de cálcio é preditora de estabilidade das lesões. O objetivo deste estudo é avaliar qual a influência da quantidade total de cálcio coronário, obtido pela tomografia computadorizada cardíaca (TCC), na gravidade de lesões moderadas, avaliadas por meio de USIC. MÉTODOS: Realizamos USIC de 27 lesões moderadas, em uma série consecutiva de 22 pacientes com indicação para o procedimento, e que tinham TCC prévia. Foram obtidas medidas da ALM, carga de placa e porcentual de área de estenose. RESULTADOS: Os pacientes eram, em sua maioria, do sexo masculino (85%), com idade de 60 ± 9 anos e 41% eram diabéticos. Observamos correlação significativa entre ALM e escore de cálcio total (r = 0,67; P = 0,002), mas não houve correlação entre escore de cálcio total e carga de placa (r = 0,02; P = 0,93) ou porcentual de área de estenose (r = 0,3; P = 0,32). Os preditores independentes da ALM na análise multivariada foram idade (P = 0,02), índice de massa corporal (P = 0,01), diabetes (P = 0,02), dislipidemia (P = 0,02), proteína C-reativa ultrassensível (P = 0,02), glicemia (P = 0,02), HDL-colesterol (P = 0,02), triglicerídeos (P = 0,02), fração de ejeção do ventrículo esquerdo < 50% (P = 0,02) e escore de cálcio total (P = 0,03). CONCLUSÕES: O escore de cálcio total apresenta correlação positiva com a ALM em artérias coronárias, sugerindo que maior quantidade de cálcio na árvore coronária implica lesões ateroscleróticas moderadas com maiores áreas luminais ao USIC.


BACKGROUND: The minimal luminal area (MLA) measured by intravascular ultrasound (IVUS) is a predictor of cardiovascular events in patients with angiographically moderate lesions. On the other hand, the presence of calcium is a predictor of lesion stability. The objective of this study is to evaluate the influence of the total amount of coronary calcium, obtained by multislice CT scanner (MDCT), on the severity of moderate lesions as assessed by IVUS. METHODS: IVUS was performed in 27 moderate lesions in a consecutive series of 22 patients with indication for the procedure who had a previous MDCT. Measurements of the minimal luminal area (MLA), plaque burden and percentage area stenosis were obtained. RESULTS: Most patients were male (85%), with mean age of 60 ± 9 years and 41% were diabetic. A significant correlation was observed between the MLA and total calcium score (r = 0.67; P = 0.002), but there was no correlation between the total calcium score and plaque burden (r = 0.02; P = 0.93) or percentage area stenosis (r = 0.3; P = 0.32). Independent predictors of MLA in the multivariate analysis were age (P = 0.02), body mass index (P = 0.01), diabetes (P = 0.02), dyslipidemia (P = 0.02), high-sensitive Creactive protein (P = 0.02), glucose (P = 0.02), HDL-cholesterol (P = 0.02), triglycerides (P = 0.02), left ventricular ejection fraction < 50% (P = 0.02) and total calcium score (P = 0.03). CONCLUSIONS: The total calcium score correlates positively with MLA in coronary arteries, suggesting that a large calcium burden in the coronary tree is related to angiographically moderate lesions with larger luminal areas at IVUS.
