Resumo

Background: Caudal vena cava thrombosis (CVCT) is a serious disease that affects cattle. Due to being commonly a fatal pathology, it causes economic losses for producers and national livestock. Thus, the present study describes the epidemiological, clinical, ultrasonographic imaging and pathological findings in 4 cattle with CVCT attended at the Veterinary Hospital (HV) of the Institute of Veterinary Medicine (IMV) of the Federal University of Pará (UFPA). Cases: The animals were crossbreds of the Gir x Holstein and Jersey x Holstein breeds, aged between 4 and 8 years old, raised in semi-extensive and intensive systems. The main clinical signs were pale mucous membranes, reluctance to move, markedly positive venous pulse, engorged jugular with positive stasis test, and serous to mucopurulent nasal exudation. The auscultation of the lung fields revealed tachypnea, silent areas, wheezing, and pleural friction, in addition to coughing, expiratory dyspnea, mouth breathing, and expiratory grunts. One animal had severe hemoptysis. The ultrasound examination performed on a bovine revealed a circular and dilated caudal vena cava in cross-section. Laboratory tests in 3 cattle revealed anemia, leukocytosis with neutrophil left shift, and increased liver enzymes. At necropsy, all cattle had thrombi in the hepatic segment of the caudal vena cava. In the lung, multiple abscesses and areas of parenchymal consolidation, crateriform areas, as well as thrombi in the arteries were observed. Pleural effusion and ascites were seen in all cattle. Clotted blood was seen in the trachea, bronchi, and on rumen contents of an animal. Histopathological alterations seen in the liver were centrilobular hepatocytes with frequent intensely eosinophilic cytoplasm, and pyknotic, karyorrhexic, or absent nuclei and cell borders barely distinguishable. In the lung were nodular and random formations, with a thick wall of mature connective tissue and a central area full of cellular debris, necrotic cells, and intact and degenerated neutrophils (abscesses). Discussion: The set of diagnostic tools that include epidemiology, clinical signs and clinical examinations, ultrasound, necropsy, and histopathology were efficient in the diagnosis of CVCT. The possible causes that led the animals to develop CVCT were diffuse septic pododermatitis in the medial nail of the right pelvic limb associated with traumatic reticuloabomasitis and liver abscesses. In 1 cow, it was not possible to establish the probable cause of CVCT, but for the other cattle in the present study, the probable causes are in agreement with studies that have shown that this disease can occur as a sequel to several septic conditions such as jugular phlebitis, mastitis, hoof rot, enteritis, pneumonia, traumatic reticulopericarditis, acidosis and rumen laminitis, as well as omphalophlebitis in calves. The tachypnea, serous to purulent nasal exudation, pulmonary wheezing, pleural friction, coughing, and expiratory dyspnea, usually with open mouth breathing and expiratory grunts evidenced in the animals of this study, occurred as a result of embolic abscess pneumonia. The presence of multiple lung abscesses, areas of parenchymal consolidation, crateriform foci, and thrombi in the pulmonary arteries and chronic suppurative pneumonia, found at necropsy of the animals in the present study, are related to the development of a thrombus in the caudal vena cava that detaches and embolizes and lodges in the pulmonary arteries. The histopathological findings in 1 cow are compatible with lesions found at necropsy and draw attention to embolic pneumonia and liver lesions, which, are related to thrombi in pulmonary arteries and abscesses formed from CVCT, as well as venous stasis exerted in the return circulation.

Background: The peroneus tertius muscle is responsible for tarsal flexion and coordinate extension of the stifle joint. The most common causes of rupture are excessive effort when trying to rise on a slippery surface, mounting, or being mounted. Rupture of the peroneus tertius muscle is characterized by an inability to flex the hock. Most cases are not curable, presenting an unfavorable to poor prognosis, and the animal has to be euthanized. This study aimed to report the clinical picture and treatment of a bilateral rupture of the peroneus tertius muscle in a cow from the municipality of Castanhal, state of Pará (Amazonian biome). Case: We report the clinical picture and treatment of a bilateral rupture of the peroneus tertius muscle in a 4-year-old cow, mixed breed, weighing approximately 400 kg, calved 3 months ago, belonging to a batch of 40 cattle kept on extensive breeding system in a pasture of Urochloa (Brachiaria) brizantha. The clinical signs appeared after the cow was mounted by a 1,100 kg bull and consisted of lameness, falling into sternal decubitus with hind limbs extended backward, hyperextension of the hocks with flexed stifle, with the tibia and metatarsus in a straight line, which lead to a 90-degree position of the femur and knee. Abrasions in the dorsal region of the fetlocks reinforce evidence of dragging hooves. Diagnosis of bilateral rupture of the peroneus tertius muscle was performed by a clinical examination and semiological test. Nonsteroidal anti-inflammatory drugs (flunixin meglumine, 2,2 mg/kg, every 24 h for 4 days) were prescribed for treatment, in addition to resting in a paddock with good availability of pasture, water, and mineral salt. After 3 months, we observed an improvement of the clinical signs, and after 6 months, an almost complete recovery. Only a slight difficulty in flexing the hock when moving remained. Discussion: In the present report, bull mounting was the probable cause of the hyperextension of both hocks and the bilateral rupture of the peroneus tertius muscle. According to the handler, these clinical signs were only evidenced after the animal was mounted. As stated in the literature, accidents such as falls, slips, mounting or being mounted, as well as excessive traction and over-elevation of the limb are considered predisposing factors for the onset of the disease. Rupture of the peroneus tertius muscle is unusual in production animals; therefore, the low occurrence of reports on this kind of rupture in the literature makes this case even more unique. The bilateral injury differs from cases described in the literature, in which the reported injuries were always unilateral. Our findings reinforce the hypothesis that the act of mounting was the triggering factor for the bilateral rupture of the animal peroneus tertius muscle. The breeding system also differs from the cases diagnosed by the same authors, since they describe the disease in bovines kept on intensive or semi-intensive breeding. In the present case, the cattle were kept on an extensive breeding system. The extension and flexion tests used in the clinical examination allowed for the diagnosis of rupture of the peroneus tertius muscle in both limbs, similarly to what has been reported by other authors. Anamnesis, clinical findings, and exclusion of different diagnoses were decisive for establishing the diagnosis. The treatment chosen was efficient, even though the case was severe, as the injury occurred in both hind limbs.

Background: Toxic agents are chemical substances or physical agents that, when interacting with living organisms, cause harmful effects. For animals, toxic products include those intended to combat endo and ectoparasites, rodenticide products, and heavy metals. Minerals and dietary additives, even if essential to the animal, can become toxic agents, among which sodium chloride, copper, urea, and ionophore antibiotics stand out. This study aimed to survey the diagnoses of accidental poisoning in ruminants over 65 years as recorded in the files of the Pathological Anatomy Sector of the Federal Rural University of Rio de Janeiro. Materials, Methods & Results: The diagnoses of poisoning by ticks, rodenticides, heavy metals, macro and micronutrients, and dietary additives in ruminants were made based on the association of epidemiological, clinical, and anatomopathological findings and, in some cases, by histochemical (rubeanic acid and Masson trichrome) and toxicological examinations. A review of data recorded over 65 years identified 372 poisonings in ruminants. Of these, 85.5% (318/372) were in bovine species and 14.5% (54/372) in goats, sheep, and buffaloes. The outbreak of poisoning by organophosphates resulted in the death of 16 cattle that ingested contaminated broken rice and corn. The spraying of this product on the bags was intended to control insects. Intoxication due to excessive administration of abamectin resulted in the intoxication of 16 calves, in two distinct farms. Toxicosis caused by coumarin derivatives resulted in the death of six cattle after accidental ingestion of these rodenticides near the pens. Arsenic poisoning occurred due to ingestion of ant poison, available in bone meal and a mineral salt, through baths with arsenic-based ticks, or in cases later confirmed by toxicological analysis, resulting in the death of 109 cattle. Resumo

Although, serological assays for detecting immune response against Toxoplasma gondii in goats and sheep are frequent in Brazil, studies showing lesions and confirmation of infectious causes of abortion in goats in the state of Rio de Janeiro have not been reported. This study aimed to describe an abortion outbreak caused by T. gondii in a goat herd in the state of Rio de Janeiro. At histological examination, necrotizing encephalitis and lymphoplasmacytic myositis were observed in goat fetuses submitted to necropsy. The diagnoses were confirmed by immunohistochemistry, which revealed positive marking structures compatible with T. gondii bradyzoites in the brain and skeletal muscle cysts. T. gondii should be included among the causes of abortion in goats in the Southeast region of Brazil.(AU)


Embora sejam frequentes as pesquisas sorológicas da toxoplasmose em caprinos e ovinos no Brasil, os estudos que apresentam as lesões e a confirmação das causas infecciosas de aborto em caprinos no estado do Rio de Janeiro não foram encontrados. Neste trabalho descreve-se Toxoplasma gondii como causa de aborto em uma criação de caprinos no estado do Rio de Janeiro. Ao exame histológico foi observada encefalite necrosante e miosite linfoplasmocítica nos fetos caprinos submetidos à necropsia. A confirmação do diagnóstico foi realizada com o exame de imuno-histoquímica, que revelou marcação positiva de cistos com bradizoítos de T. gondii no encéfalo e em músculo esquelético. T. gondii deve ser incluído entre as causas de aborto em cabras na região sudeste do Brasil.(AU)

Background: Ionophore antibiotics are food additives with coccidiostatic or antimicrobial action; they are also used as growth promoters, ruminal pH regulators, volatile fatty acid molar modifiers, and methanogenesis reducers. However, these compounds have the potential to cause microbial resistance, in addition to the risk of intoxication. Ionophore poisoning may be caused by excessive intake, sensitivity of certain animal species, and concomitant use with other drugs. In Brazil, cases of ionophore poisoning in buffalos are rare. This study aims to describe the epidemiological, clinical, and pathological findings of lasalocid poisoning in buffalo calves. Case: A visit was made to a farm in the municipality of Mojú, Pará state to care for Murrah buffalo calves. After weaning, the buffalos were grazed in paddocks with Panicum spp., and received a supplement of mineral, protein, and vitamin. This supplement contained, per kg, 250 g PB, 50 g Ca, 20 g P, 8 g S, 39 g Na, 20 mg Co, 557 mg Cu, 200 mg Fe, 12.4 mg Se, 2040 mg Zn, 0.19 mg biotin, 26750 IU of vitamin A, 4175 IU of vitamin D, 155 IU of vitamin E and 300 mg/kg of lasalocid. The product was made available to all calves, at 1-2 g/kg body weight (BW), according to the manufacturer's recommendations. Older calves were raised together with those less than 30 days old; as a result, the older calves tended to eat more, which could lead to a supplementation consumption of more than 1 kg body weight per animal per day. It was reported that between 40 and 60 days after the introduction of this supplement, 16 calves fell ill and died due to apathy, motor instability, tremors, and distended neck. The herd had a mortality rate of 33.3%. Two calves underwent a necroscopic examination at the Pathology Section of the Veterinary Institute of the Federal University of Pará. Macroscopic examination revealed extensive pale areas in the skeletal muscles, myocardium, and tongue. Fragments of these muscles and various organs were collected, fixed in 10% buffered formalin, processed according to the routine histological technique, and stained with hematoxylin-eosin and Masson's trichrome stain. Microscopic examination of the histologic samples revealed foci of muscle atrophy and necrosis characterized by an increase in cytoplasmic eosinophilia associated with the loss of stretch marks, and hyperchromatic nuclei that were displaced to the periphery. The necrosis of the muscle fibers was highlighted by Masson's trichrome staining. Discussion: The diagnosis of lasalocid poisoning in buffalo calves was based on epidemiological data, clinical findings, results of macroscopic and histopathological examination, and based on the estimated ionophore intake, obtained directly from the supplement label and by the calf's handler. Based on the absence of stratification of the calves by similarity of age and because the buffalo calves older than 30 days could eat more than 1 kg of the supplement (containing 300 mg/kg of lasalocid), it was possible to estimate the intake of lasalocid per kg CP (body weight). Therefore, the intake of lasalocid by a 70-kg buffalo calf in approximately 90 days and daily supplement consumption between 1 and 1.5 kg would be between 4.2 and 6.4 mg/kg of body weight. This report reinforces that notion that buffalo calves should never ingest ionophores; however, if necessary, strict protocols must be followed to avoid poisoning in these animals. This study highlighted the fact that stratification of buffaloes by different age groups during feeding became a risk factor that allowed greater consumption by older animals; this led to the estimated consumption of 4.2-6.4 mg/kg of lasalocid.

Bitches and intact males seem predisposed. Thismesenchyme tumor is commonly mistaken with lipoma due to its soft texture and subcutaneous site and often requireshistopathology to confirm its diagnosis. Microscopic examination also enables the evaluation of surgical margins and ruleout possible infiltrative sites. Complete surgical excision is usually curative. This study reports a case of non-infiltratingangiolipoma in a dog.Case: A 14-year-old mixed-breed dog was presented to a veterinary clinic in the city of Rio de Janeiro, RJ, Brazil. Onpalpation, a painless mass was noted, with high mobility and covered by intact hirsute skin in the right subcutaneousventrolateral region. Computed tomography of the chest showed an expansive mass of uptake only from the edges of thesoft tissues of the right subcutaneous ventrolateral region. The mass was homogeneous and well delimited, suggesting aneoplastic process. Subsequently, the mass was surgically removed, fixed in 10% buffered formalin, and sent for histopathological analysis. On macroscopic examination, the mass was well delimited, without skin coverage, and measured2.3×1.9×0.6 cm. The consistency was smooth and unctuous in appearance with a compact cream-colored surface withblackish multifocal spots. Under microscopy, the histological sections showed neoplasm of mature adipocytes and of endothelial cells of blood vessels benign were filled with a marked amount of red blood cells...

Background: Pituitary pars intermedia (PI) adenoma is a benign adenohypophysis neoplasm, rare in Brazil, which maycompress adjacent structures and lead to dysfunctions of the endocrine organs. The most affected species are equines,particularly aged animals. This neoplastic disease is often associated with Cushing’s syndrome, when the pituitaryadrenocortical axis is affected. However, this neoplasm is seldom associated with clinical blindness. This paper describesa case of pituitary pars intermedia (PI) adenoma which caused blindness in a mare.Case: An emaciated, blind mare that had difficulty finding water and food was referred to “Hospital Veterinário de GrandesAnimais” of “Universidade Federal Rural do Rio de Janeiro”, in Seropédica, state of Rio de Janeiro, Brazil, and later tothe “Setor de Anatomia Patológica” (SAP/UFRRJ) for necropsy. During necropsy it was found that the animal’s pituitarygland consisted of a circumscribed globous mass measuring 3.5x2.5x2.5 cm, symmetrical and well delimited at the sellaturcica. Several organs were collected, fixed in 10% formalin and then processed for histological examination. The brainwas subjected to serial cleavage along the path of the vision organs. All collected tissues were stained with hematoxylinand eosin; the pituitary was stained with Schiff Periodic Acid (PAS) to differentiate neoplastic cells and with Luxol FastBlue to evidence demyelination. Histopathology found that the pars intermedia (PI) was thickened and compressed theneurohypophysis and adenohypophysis. Cells were arranged in a disorganized manner or formed follicles or cysts containing eosinophilic material (colloid), with granules strongly positive after staining with PAS. Proliferated cells werelarge, polyhedral to ovoid, and contained clear basophilic granular material. The nuclei were spherical to ovoid and therewere mild anisocytosis and anisocariasis. Cysts were frequently found in the pars intermedia (PI)...

Background: Hemangiosarcoma is a malignant mesenchymal neoplasm of endothelial cells that mainly affects dogs and is less common in horses, cattle, goats, pigs, and sheep. In ruminants, however, few cases of hemangiosarcoma have been reported. Although this neoplasm may primarily occur in any tissue, it most often originates in the spleen, liver, heart, and skin. This study reports the clinical and pathological findings of a bovine hemangiosarcoma.Case: A 10-year-old lactating Girolando cow from the municipality of Vassouras/RJ was examinated because of uncontrolled cough and bilateral hemoptysis, loss of appetite, fever, weight loss, severe decline in milk production (a decrease of 28 L), and respiratory wheezing during chest auscultation. We performed a necropsy and collected fragments of various organs which were fixed using 10% buffered formalin and sent to the Pathology Anatomy Sector (SAP) of the Federal Rural University of Rio de Janeiro (UFRRJ) for routine histological assessment. Macroscopically, large hemorrhagic areas were observed in the lungs, mainly in the cranioventral portion of the pulmonary lobes, multifocal areas of hemorrhages in the liver, and a 10x8x5 cm soft red mass was observed in the spleen. Microscopically, was observed lung proliferation of endothelial cells arranged in tapes, supported by a collagenous stroma associated with severe multifocal lobular hemorrhage with a large number of siderophages and diffuse and marked edema, and congestion. In the mediastinal lymph node, liver, and spleen, the proliferation of neoplastic endothelial cells equivalent to those described in the lungs was observed. The histological sections of liver and tumor spleen were subjected to immunohistochemical analysis for von Willebrand factor, in which marking was observed in neoplastic endothelial cells.[...]

This study reports the clinical and pathological findings of a bovine hemangiosarcoma.Case: A 10-year-old lactating Girolando cow from the municipality of Vassouras/RJ was examinated because of uncontrolled cough and bilateral hemoptysis, loss of appetite, fever, weight loss, severe decline in milk production (a decrease of 28 L), and respiratory wheezing during chest auscultation. We performed a necropsy and collected fragments of various organs which were fixed using 10% buffered formalin and sent to the Pathology Anatomy Sector (SAP) of the Federal Rural University of Rio de Janeiro (UFRRJ) for routine histological assessment. Macroscopically, large hemorrhagic areas were observed in the lungs, mainly in the cranioventral portion of the pulmonary lobes, multifocal areas of hemorrhages in the liver, and a 10x8x5 cm soft red mass was observed in the spleen. Microscopically, was observed lung proliferation of endothelial cells arranged in tapes, supported by a collagenous stroma associated with severe multifocal lobular hemorrhage with a large number of siderophages and diffuse and marked edema, and congestion. In the mediastinal lymph node, liver, and spleen, the proliferation of neoplastic endothelial cells equivalent to those described in the lungs was observed. The histological sections of liver and tumor spleen were subjected to immunohistochemical analysis for von Willebrand factor, in which marking was observed in neoplastic endothelial cells.[...](AU)

The steers were then slaughtered in order to evaluate macroscopic changes in the gastrointestinal system. Rumen and abomasum of each steer was emptied, washed with tap water and grossly examined. One steer presented ruminal lesion, while the other forty-nine did not show any ruminal injury. Throughout the experiment the affected steer showed reduction of daily weight gain; but during clinical examination no additional clinical sign was seen. At gross inspection, a reduction in the amount of muscular and adipose tissue was seen. Bovine leyomiosarcomas have been described in several sites, but smooth muscle tumors that arise from the skin are rare in domestic animals. These neoplasms may show variable histologic features depending on their grade of differentiation; therefore, diagnostic tools as special stain, electron microscopy and immunohistochemistry are essential for the definitive diagnosis. Due to the lack of reports of skin leyomiosarcomas in cattle, we described the occurrence of a leyomiosarcoma in the skin and subcutaneous tissue of a Nellore cow. Case: A 10-year-old Zebu Nellore cow was admitted at the Veterinary Diagnosis Center (CEDIVET) of Pará Federal University (UFPA), Brazil, to investigate a tumor on the hip. At clinical examination the cow showed a decreased appetite and weight loss. Due to the bad prognosis the veterinarian recommended euthanasia. Representative tissue samples were submitted for histopathological and immunohistochemistry evaluation at the Department of Pathology of the Federal Rural University of Rio de Janeiro (UFRRJ). The slides were stained by Hematoxylin and Eosin (HE), Masson’s trichrome and Van Gieson stains and submitted for immunohistochemistry with primary antibodies vimentin, troponin C, smooth muscle alpha actin and desmin. Grossly, the tumor appeared as a multilobular, ulcerated and firm 30 cm diameter mass with black and hemorrhagic areas on the surface. The mass was scattered throughout the subcutaneous tissue and infiltrated the adjacent muscle. On light microscopy, the neoplasm consisted of a non-encapsulated proliferation into the derma, composed of highly pleomorphic spindle to round cells arranged in broad interlacing fascicles and small islands. Metastatic lesions were found in the lungs and local lymph nodes. Masson’s trichrome stain revealed collagen production and Van Gieson stains was inconsistent...

Background: Dermatosparaxis is an autosomal recessive genetic disease that affects the connective tissue of animals. Collagen proteins form fibrillar structures that provide strength and structure to the extracellular matrix of tissues and organs in the body. Therefore, changes in collagen synthesis result in hyperextensibility and skin fragility. Similar to dermatosparaxis in animals, some cases of Ehlers-Danlos syndrome type VIIC have been reported in humans. The objective of this study was to describe the clinical, anatomopathological, and molecular findings of a case of dermatosparaxis in a sheep bred by crossing the Santa Inês and White Dorper breeds. Case: A case of dermatosparaxis was reported in a 20-day-old female sheep, a cross between the Santa Inês and White Dorper breeds. The sheep exhibited hyperextensibility of the skin with minimal tension and increased joint volume shortly after birth. Because of clinical worsening and the poor prognosis of the disease, the owners opted for euthanasia. The necropsy revealed large ulcerated areas in several parts of the body, including the cervical, sternal, scapular, and thoracic regions, and the inner and outer thighs. The skin was extremely easy to be removed during the necropsy and the use of a scalpel was not necessary. Extensive areas of hemorrhage were observed in the subcutaneous tissue and small intestine. The histopathological findings of the skin lesions evidenced the presence of dermatosparaxis, particularly regarding the disorganization of collagen fibers. The tests involving Masson’s trichrome staining, picrosirius red staining, and Gomori’s trichrome staining under polarized light evidence of collagen fiber dysplasia.Discussion: The diagnosis of dermatosparaxis in this study was based on clinical, anatomopathological, and molecular analysis. O presente estudo teve como objetivo descrever a ocorrência de otite parasitária causada por Rhabditis blumi em bovinos leiteiros de raça Gir de uma fazenda da região Norte do Brasil. Foram coletadas amostras de 42 bovinos por swab e lavado dos condutos auditivos externos (CAE). Ao exame clínico, 9,5% (4/42) dos bovinos apresentavam sintomatologia nervosa, como leve a moderada rotação da cabeça, apatia, flacidez dos lábios e ptose palpebral unilateral, alopecia das regiões da cabeça e cupim, causados pelo desconforto e prurido da região auricular, alteração na mastigação e acúmulo de alimento na cavidade oral. Adicionalmente, 71,4% (30/42) dos abovinos mostraram a presença do parasita no cerúmen dos condutos auditivos. À análise microscópica do material do saco conjuntival foi observado presença do parasita em 90% (9/10) dos bovinos avaliados. Os 30 bovinos positivos para Rhabditis spp. foram distribuídos aleatoriamente em três grupos de 10 animais: (G1) Bovinos controle, (G2) Bovinos tratados com ivermectina 1% pour on e (G3) Bovinos submetidos a lavado dos condutos auditivos externos (CAE). Cada tratamento foi repetido três vezes com intervalo de sete dias. No G1 os 10 bovinos mantiveram-se infectados durante todo o estudo. No G2 20% dos bovinos foram negativos após os dois primeiros tratamentos, porém, mostraram-se positivos na terceira avaliação. No G3 todos os bovinos mantiveram-se positivos, sendo observada apenas diminuição da carga parasitária. A identificação por análise molecular por meio de fragmentos amplificados da expansão D2/D3 do 28S rDNA confirmou a presença apenas da espécie Rhabditis blumi nos animais. Baseado nas observações clínicas, morfológicas e moleculares pode-se relatar o primeiro caso de R. blumi em bovinos da raça Gir no Estado do Pará, através da compra de animais oriundos de áreas onde a otite parasitária tem sido diagnosticada, principalmente de Minas Gerais, para formar animais mestiços (Gir x Holandês). Desta forma ressalta-se a importância do exame clínico prévio dos animais a serem transferidos para outras propriedades ou regiões. Este relato também parece ser o primeiro sobre a presença de R. blumi no saco conjuntival de bovinos. O tratamento com ivermectina no G2 não surtiu melhora clínica dos bovinos.(AU)

Forty-two samples were collected from cattle by swab washed from the external auditory canal (EAC). On clinical examination, in 71.4% (30/42) of the cattle the parasite was found in the cerumen of the ear canal, along with alopecia of head and hump caused by discomfort and itching of the auricular region. At microscopic analysis of material from the conjunctival sac the parasite was found in 90% (9/10) of the evaluated cattle. In addition, 9.5% (4/42) of the cattle showed nervous symptoms, such as mild to moderate rotation of the head, apathy, flaccid lips and unilateral ptosis, change in chewing and food accumulation in the oral cavity. Thirty cattle positive for Rhabditis spp. were randomly divided into three groups of 10 animals each: (G1) Cattle Control, (G2) Cattle treated with ivermectin 1% pour-on, and (G3) Cattle undergoing wash of the external auditory canal (EAC). Each treatment was repeated three times with intervals of seven days. In G1, 10 cattle remained infected throughout the study. In G2, 20% of the cattle were negative after the first two treatments, however were positive at the third evaluation. In G3, all cattle remained positive, but with decrease in parasite load. Identification by molecular analysis of amplified fragments through the expansion D2/D3 28S rDNA confirmed the presence of only Rhabditis blumi. Based on clinical, morphologic and molecular examination, it appears to be the first report of the occurrence of R. blumi infection in Gir cattle in the State of Pará, due to the purchase of cattle from areas where parasitic otitis has been diagnosed, as from Minas Gerais, to produce crossbred animals (Gir x Holstein). This emphasizes the importance of prior clinical examination by the veterinary service in order to transfer only healthy animals to other properties or regions. This appears also to be the first report on R. blumi infection of the conjunctival sac in cattle. Treatment with ivermectin in G2 did not produce clinical improvement.(AU)

Resumo

O diagnóstico precoce e específico da paratuberculose ainda é um desafio. Isto pode estar associado à baixa sensibilidade dos testes laboratoriais e ou à variação da resposta imunológica frente à infecção por Mycobacterium avium subsp. paratuberculosis. Mundialmente, é uma enfermidade que causa importantes prejuízos econômicos, em especial, à bovinocultura leiteira, devido ao caráter crônico da infecção. No Brasil, a paratuberculose já foi descrita em diversas espécies de ruminantes domésticos e em vários estados, o que demonstra que a enfermidade está presente em território nacional e há a necessidade de elaboração de técnicas de diagnóstico para a confirmação da infecção. O objetivo deste trabalho foi caracterizar os achados anátomo-histopatológicos e imuno-histoquímicos em intestino e linfonodos mesentéricos de bovinos assintomáticos, provenientes de rebanhos positivos para paratuberculose localizados no estado do Rio de Janeiro, Brasil. O estudo macroscópico revelou alterações inespecíficas tais como áreas avermelhadas na mucosa do intestino, aumento do volume das placas de Peyer e dos linfonodos mesentéricos, além disso, observou-se que vasos linfáticos mesentéricos estavam dilatados e esbranquiçados. Do total de 52 vacas leiteiras avaliadas, a histopatologia revelou infiltração granulomatosa, por vezes com formação de células gigantes multinucleadas, em mucosa e ou submucosa de jejuno, íleo e em linfonodos mesentéricos, principalmente na região cortical, em 32 animais. Estes bovinos foram submetidos à coloração de Ziehl-Neelsen cujo teste não demonstrou reação positiva, no entanto, quando analisados pelo teste imunohistoquímico para Mycobacterium spp. observou-se imunorreação em 6 animais. Desta forma, a histopatologia e imunohistoquímica pode ser uma importante ferramenta para diagnóstico da paratuberculose subclínica. (AU)


The early and specific diagnosis of paratuberculosis remains a challenge due to the low sensitivity of the currently available laboratory tests and also because of variations in the immune response towards infection with Mycobacterium avium subsp. paratuberculosis. Globally this disease causes significant economic losses, primarily in dairy cattle, owing to the chronic nature of the infection. Paratuberculosis has been described in a number of Brazilian states and from a diversity of domestic ruminant species clearly demonstrating that the disease is present in the country and highlighting the requirement for the development of diagnostic techniques for confirmation of infection and for epidemiological analyses. The aim of this study was to characterize the anatomo-histopathological and immunohistochemical findings in the bowel and mesenteric lymph nodes of assymptomatic cattle, derived from paratuberculosis positive herds located in state of Rio de Janeiro, Brazil. Macroscopic examination during necropsy revealed nonspecific changes including reddening of the gut mucosa, increased volumes for the Peyer's patches and mesenteric lymph nodes and in some case dilation and whitening of the mesenteric lymphatic vessel. Histopathology revealed granulomatous infiltration, occasionally with the formation of giant cells in the jejunal and ileal mucosa or sub-mucosa, and/or in the cortical region of the mesenteric lymph nodes, in 32 of the 52 cattle examined. Tissue sections from these animals were subjected to Ziehl-Neelsen staining, but the presence of acid-fast bacilli was not observed. Subsequent analysis, employing genus specific immunohistochemisty for Mycobacterium, revealed areas of immunoreactivity in sections prepared from a total of six animals. The results of this investigation highlighted the value of histopathology and particularly immunohistochemistry as tools for the diagnosis of subclinical paratuberculosis. (AU)
