Resumo

The purpose of this research was to confirm the changes occurring in the foot system of the heifers challenged with lipopolysaccharides (LPS), at the clinical, serum and histological levels. We studied 16 clinically healthy heifers, 14 months of age, placed in a confinement system. All the animals were provided with an accelerometer collar to establish their activity. They were categorized into two groups: the LPS group (n=8), or those which were administered two intravenous applications of 2 mL containing 0.5 g/kg of body weight of LPS, with a 24-hour interval and the Control group (n=8) which were given two infusions of 2 mL of saline solution in the same time interval. General clinical examination and blood collection were done at 0, 4 and 8 hours post the LPS challenges and analyses of the hemograms and paroxonese-1 were performed. The animals were then slaughtered on day 4 and the laminar tissue was collected for histological analysis. The LPS group revealed a lower total leukocyte count with heart rate and greater activity. None of the animals revealed any abnormal signs symptomatic of foot pathology after histological analysis. Hence, the challenge with LPS failed to induce any clinical and histological changes in the foot tissue compatible with laminitis.(AU)


O objetivo deste trabalho foi verificar alterações do sistema podal á nível clínico, hematológico e histológico de animais desafiados com lipopolissacarídeo (LPS). Foram utilizadas 16 novilhas de corte com 14 meses de idade, clinicamente saudáveis, em sistema de confinamento. Todos os animais continham uma coleira com acelerômetro para verificar atividade, sendo divididos em 2 grupos: LPS (n= 8), que recebeu duas aplicações de 2 mL contendo 0,5 g/kg de peso corporal de LPS via intravenosa, com intervalo de 24 horas e Controle (n= 8) que recebeu duas aplicações de 2 mL de solução salina com o mesmo intervalo de tempo. Foram realizados exame clinico geral e coletas de sangue às 0, 4 e 8 horas após os desafios com LPS para análise de hemogramas e paroxonase-1. Os animais foram abatidos aos 5 dias sendo executada a coleta de tecido laminar para análise histológica. A análise estatística foi executada com a utilização do programa NCSS(2004) através de ANOVA-medidas repetidas, considerando o efeito do tratamento do período e sua interação (tratamento*horas), sendo considerados significativos valores de P 0,05. O grupo LPS apresentou superior contagem total de leucócitos com maior frequência cardíaca e atividade. Nenhum animal apresentou sinal de anormalidade compatível com laminite através da análise histológica. Entretanto, o desafio com LPS foi capaz de gerar alterações a nível clínico, constatado pelas alterações nos parâmetros de frequência cardíaca, respiratória e temperatura corporal, além de mudanças no leucograma.(AU)

Resumo

The objective of this study was to evaluate the effect of exogenous insulin administration on follicular growth, estrus presentation and conception rate during a protocol of ovulation synchronization. Dairy cows were subjected to the Heatsynch protocol, with the insertion of an intravaginal device containing 1.9 g of progesterone (CIDR) and an intramuscular injection containing 2.5 mg of GnRH on day 0. On day 7, the CIDR was removed and subjects were given 12.5 mg of dinoprost. Also on day 7, Insulin Group (IG, n = 21) animals received a subcutaneous injection containing 0.25 IU / kg of recombinant human insulin and the control group (CG, n = 25) received a 0.9% NaCl injection. On day 8, an injection of 0.5 mg of estradiol cypionate was administered to all cows. Animals were inseminated 12 hours after estrus presentation or at day 10 at fixed time. Follicular development was evaluated on days 7, 9, and 10 using transrectal ultrasonography, estrus presentation was observed between days 8 and 10, and conception rate was evaluated 30 days after AI. There were no differences in growth rate, follicular diameter, estrus presentation, and conception rate. Therefore, application of a single dose of insulin does not promoted an increase in follicular size, estrus presentation and conception rate in dairy cows.(AU)


O objetivo deste estudo foi avaliar o efeito da administração exógena de insulina sobre crescimento folicular, apresentação de cio e taxa de concepção durante um protocolo de sincronização da ovulação. As vacas holandês lactantes foram submetidas ao protocolo Heatsynch, com inserção do dispositivo intravaginal contendo 1,9 g de progesterona (CIDR) no dia 0 e uma injeção intramuscular de 2,5 mg de GnRH. No dia 7, o CIDR foi removido e foi aplicado 12,5 mg de dinoprost. O desenvolvimento folicular foi avaliado nos dias 7, 9 e 10 por ultrassonografia transretal, a apresentação de cio foi observada entre os dias 8 e 10 e a taxa de prenhez/IA foi avaliada 30 dias após a IA. Não houve diferença quanto a taxa de crescimento e diâmetro folicular, apresentação de cio e taxa de concepção. A aplicação de uma dose única de insulina não promove o incremento no tamanho folicular, apresentação de cio e taxa de prenhez/IA em vacas de leite.(AU)

The aim of this study was to evaluate the economic viability of the use of a lactation induction protocol in Holstein heifers. Were collected data of nutritional, reproductive and sanitary management pre and post-lactation period from 30 induction heifers (IG) and 30 pregnant heifers (CG). All animals were inseminated around 14 months of age and started lactation at 23±4 months. The animals of IG were heifers do not pregnant after two inseminations and natural service e were induced to lactation according to protocol: from the 1st to the 8th day, 30 mg of estradiol benzoate were administered daily, together with 300 mg of progesterone. From the 9th until the 14th, animals only received daily doses of 20 mg estradiol benzoate. On 16th day, 0.56 mg of sodium cloprostenol was administered and injections of 40 mg of dexamethasone sodium phosphate were administered daily from the 19th to the 21st day. On the 1st, 8th, 15th and 22nd day, the animals received a dose of sometribove zinc. The animals of CG were managed according to the farm routine. The costs of the CG during pre and post lactation (nutrition, sanitary, reproduction management) e were higher than the IG, however, when evaluating the economic viability due to lower milk production, the induced group did not leave profit in the first year of use. When comparing the lactation induction with the sale of animals that did not conceive and purchase of animals for replacement, this last scenario showed itself as a better alternative.


O objetivo deste estudo foi avaliar a viabilidade econômica do uso de um protocolo de indução a lactação em novilhas da raça Holandês. Foram coletados dados econômicos do manejo nutricional, reprodutivo e sanitário pré e pós lactação de 30 novilhas induzidas à lactação (IG) e 30 novilhas prenhes (CG). Todos os animais foram inseminados em torno de 14 meses de idade e iniciaram a lactação com aproximadamente 23±4 meses. Os animais do IG foram novilhas que não conceberam após duas inseminações e monta natural, e foram induzidas à lactação com o protocolo: do 1° ao 8° dia receberam aplicação de 30 mg de benzoato de estradiol juntamente com 300 mg de progesterona. A partir do 9° dia até o 14° receberam 20 mg de benzoato de estradiol. No dia 16 ocorreu uma aplicação de 0.56 mg cloprostenol sódico e nos dias 19 ao 21, de 40 mg de dexametasona. Nos dias 1, 8, 15, e 22, os animais receberam 500 mg somatotropina recombinante bovina. Os animais do CG foram manejados conforme a rotina da propriedade. Os custos do CG durante o pré e lactação (nutrição, sanidade, reprodução) foram superiores ao grupo induzido. No entanto, ao avaliar a viabilidade econômica, devido a menor produção leiteira, o grupo induzido não deixou lucro na propriedade no primeiro ano de utilização. Ao comparar a indução de lactação com a venda de animais que não conceberam e a compra de animais para reposição, este último cenário mostrou-se como uma melhor alternativa.

Background: Low reproductive efficiency has been one of the main factors that lead to dairy herd culling in the reproductive age. In multiparous animals, such inefficiency and culling can occur because of various factors and may be associated with occasional endocrine failures. To avoid revenue losses that incur due to lack of lactation, lactation is artificiallyinduced in the animals that are not pregnant, using a defined protocol. The aim of this study was, therefore, to evaluateand compare the reproductive performance of heifers submitted to the induction protocol with that of pregnant heifersduring the transition period.Materials, Methods & Results: Sixty Holstein heifers, 32 ± 0.6 months of age, were divided into two groups: ControlGroup (Control, n = 30) comprising pregnant heifers that were accompanied since 21 days before the expected calvingdate until 224 days in milk (DIM) and an Induction Group (Induction, n = 30) comprising non-pregnant heifers submittedto a lactation induction protocol, accompanied from the beginning of the protocol until 224 DIM. For evaluation of theendocrine profile (progesterone and estradiol concentrations) of these animals, blood samples were collected at two periods: the pre-lactation (weeks -3, -2 and -1) and post-lactation (weeks 1 and 4). Heifers from both groups were submittedto weekly reproductive evaluations, from the beginning of lactation until 35 DIM. Uterine examinations were performedusing ultrasonography and vaginoscopy to evaluate uterine content, cervical opening, and mucosal aspect. Females ingood reproductive health were subjected to a hormonal protocol for timed artificial insemination (TAI). The goal of this study was to evaluate the effects of strategic insulin administration on follicular growth, progesterone production, and pregnancy rate in beef cattle. Two experiments were conducted, in experiment 1, 215 cows crossbreed cows (Bos indicus × Bos taurus) (Control Group, n = 109; Insulin Group, n = 106) were submitted to fixed-time artificial insemination (TAI) with application of 2 mg of estradiol benzoate and a vaginal device containing progesterone on day 0. On day 9, the device was removed and 0.5 mg of estradiol cypionate was administered with 12.5 mg of dinoprost and 0.25 UI kg-1 of insulin for the treated animals. On day 11, artificial insemination (AI) was performed on all animals. On days 9 and 11 according to the protocol, was evaluated follicular growth and estrus manifestation, and 30 days after AI the pregnancy rate. In experiment 2, 85 cows were utilized, including Aberdeen Angus, Hereford, and crossbreeds (Control Group, n = 49 and Insulin Group, n = 36), and were submitted to the protocol for fixed-time embryo transfer (FTET), and progesterone concentrations were evaluated. On day 0, the cows received 2 mg of estradiol benzoate and a vaginal device containing progesterone. On day 5 of the protocol, 0.150 mg of cloprostenol was administered. On day 8, the vaginal device was removed and 20 mg of purified porcine pituitary extract and 0.25 UI kg-1 of insulin was given to the animals in the treatment group, and on day 9, 1 mg of estradiol benzoate was administered. On day 17, an embryo transfer was performed and blood collection for evaluation of progesterone levels in animals submitted to FTET were collected. In experiment 1, the follicular growth rate and the pregnancy rate were similar between groups (P > 0.05) and in experiment 2, the production of P4 was not different between the animals that received the insulin application and the control group...

Background: Reproductive efficiency is one of the most important factors affecting the performance dairy cattle, and fixedtime artificial insemination (TAI) is an important biotechnology factor employed in bovine livestock. The achievement of satisfactory results in TAI protocols depends of several factors, such as health and body condition, milk yield, environmental temperatures, ovulation time, semen manipulation, as well as semen quality. The goal of this study was to evaluate the conception rate of heifers and multiparous cows based on different Holstein bulls used for artificial insemination (AI). The effects of the type of management AI, animal category, and season of the year were also examined.Materials, Methods & Results: A three-year database on the reproductive management of a dairy farm was formed. All the cows and heifers were of the Holstein breed, in semi-intensive nutritional management, milked twice a day, and received a total mix ration. After 45 days in milk, when cows were observed twice daily with estrus signs, posterior artificial insemination occurred 12 h after estrus visualization was made. However, cows that were not observed in estrus were submitted to a timed artificial insemination (TAI). In this study, these fixed factors were considered: bulls utilized for the AI (totalizing 10 animals with at least 100 inseminations in each season, denoted by the letter B and a number - B1, B2, B3, B4, B5, B6, B7, B8, B9, B10); the season of the insemination (spring/summer being from November to April and autumn/winter from May to October), category animal (heifers, animals that never have been calved, or multiparous, animals that have already given birth more than once), and the type of AI (which included TAI using a protocol for synchronization, but cows having not necessarily shown estrus signs at the AI; ETAI: using protocol of synchronization, cows showing estrus signs at the AI;[...]

Background: In order to reduce the effects of a negative energy balance, some measures have been taken into account in nutritional management during the transition period. The use of yeast, has been a good alternative used to improve the rumen metabolism and helping the adjustment of the microbiotato the new diet. The aim of the study was to evaluate the effects of supplementing a combination of yeast culture and hydrolyzed yeast on the metabolism of dairy cows during the transition period.Materials, Methods & Results:The experiment was conducted in a semi-extensive system, using 20 Holstein cows, divided equally into a control group (CG) and a supplemented group (SG). The SG received 28 g/animal/day of a combination of yeast culture and hydrolyzed yeast from 20 ± 2 days pre-calving until early lactation (18 ± 3 days). Serum concentrations of non-esterified fatty acids (NEFA), albumin and urea were determined at calving, and for three time points during the early postpartum period and three time points during the early lactation period. Regarding energy metabolism, prepartum concentrations of NEFA were higher than the physiological standard in both groups. However, NEFA, albumin and urea decreased during the early postpartum period in the supplemented animals and could be attributed to the yeast in enhancing ruminal microorganisms’ cellulolytic capacity, increasing fibre digestibility and starch utilization.Discussion: The increased concentration of non-esterified fatty acids (NEFA) due to the mobilization of fat deposits that happens in the transition period, especially in the postpartum period reflects the cow’s adaptation to the negative energy balance (NEB). The lower concentrations of NEFA observed in the present study could be attributed to the effect of the yeast in enhancing the ruminal microorganisms’ cellulolytic capacity.[...]

Resumo

Background: Diarrhea generates economic impacts due high morbidity and mortality losses, and results in delayed physical development and treatment costs. The therapy consists in using broad-spectrum antibiotics, such as drugs belongingto the quinolones group, as the enrofloxacin. It is currently available on the market a single dose enrofloxacin, whichreaches plasma peak concentration five hours after the injection, and in 30 minutes reaches the therapeutic concentrationsin plasma. The aim of this study was to evaluate the clinical, haematological and biochemical parameters in calves withdiarrhea treated with a single dose of a fast action response enrofloxacin.Materials, Methods & Results: For this study, 10 heifers with 1 day of life were classified according appearance of diarrhea clinical symptoms in two experimental groups: Sickness Group (SG) n = 5 and Healthy Group (HG) n = 5. For theSG, the adopted therapeutic protocol was a single dose (7.5 mg / kg / IM) of a fast action enrofloxacin and the GS animalswere not treated. During the first five weeks of life, the animals were clinically monitored (heart rate, respiratory rate, rectaltemperature and capillary refill time) and blood samples were collected for hemogram and biochemical analysis of albumin(ALB), total protein (TP) and globulin (GLO), and determination of the relations ALB:GLO and TP:fibrinogen. Besides,the growth measures were used to obtain the average daily gain (ADG) of weight, height, chest girth and rump width.From the diagnosis of diarrhea, the animals were monitored daily for 3 consecutive days by clinical examination and bloodcollection for hemogram. The statistical analyzes were performed by using version 9.0 SAS software. The ADG of weight,height, chest girth, rump width and concentrations of blood metabolites (ALB, TP, GLO, ALB:GLO, TP:Fibrinogen) didnot differ between the groups during the first five weeks of life.
