Resumo

ABSTRACT: The aquatic oomycete Pythium insidiosum is an emerging pathogen highly relevant in human and veterinary medicine and an etiologic agent of pythiosis, a disease of worldwide distribution mainly affecting horses, dogs, and humans, presenting cutaneous, subcutaneous, ocular, gastrointestinal, and systemic forms. The available therapeutic methods to treat this disease and its forms are not entirely effective, thus highlighting the need to investigate the forms of treatments with better efficacy, such as compounds from different pharmacological classes, compounds of natural origin, and new technological alternatives, including nanotechnology. Therefore, this study evaluated scientific publications regarding the use of nanotechnology in P. insidiosum treatment. For this, a systematic literature review, was carried out on articles published from 2010 to 2022 on the LILACS, MEDLINE, Google Scholar, PubMed, and SciELO databases using the descriptors 'Pythium insidiosum,' 'pythiosis,' 'nanotechnology,' 'nanoparticles,' 'nanoemulsion,' and 'treatment.' We reported 162 articles for the researched theme; although, only four studies were included because they met the criteria established herein. A meta-analysis was used for the statistical analysis of the data obtained in vitro studies, and we reported the use of nanotechnology can be a promising alternative in developing antimicrobial compounds with anti-P. insidiosum activity. Nevertheless, additional research is needed to verify the potential use of this technology in clinical therapy against P. insidiosum infections.


RESUMO: O oomiceto aquático Pythium insidiosum é um patógeno emergente de relevância em medicina humana e veterinária. É o agente etiológico da pitiose, uma enfermidade de distribuição mundial, que acomete principalmente em equinos, caninos e seres humanos, podendo apresentar-se nas formas cutâneas, subcutâneas, oculares, gastrointestinais e sistêmicas. Considerando que os métodos terapêuticos disponíveis para o tratamento da doença não são completamente efetivos, há uma necessidade de investigar formas de tratamentos com melhor eficácia, como os compostos de diferentes classes farmacológicas, compostos de origem natural, bem como, novas alternativas tecnológicas, incluindo a nanotecnologia. Deste modo, este trabalho objetivou avaliar publicações científicas referentes a utilização de nanotecnologia em P. insidiosum. Para isso, realizou-se uma revisão sistemática da literatura, buscando artigos no período de 2010 a 2022, nas bases de dados LILACS, MEDLINE, Google Scholar, PubMed e SciELO, utilizando-se os descritores Pythium insidiosum, pitiose, nanotecnologia, nanopartículas, nanoemulsão e tratamento. Encontrou-se 162 artigos com familiaridade a temática pesquisada; no entanto, apenas quatro estudos foram incluídos, pois atendiam os critérios estabelecidos na pesquisa. Para análise estatística dos dados obtidos nos estudos in vitro, utilizou-se meta-análise. Demonstrou-se o promissor uso de nanotecnologia como alternativa no desenvolvimento de compostos antimicrobianos com atividade anti-P. insidiosum. Entretanto, constata-se que estudos adicionais se fazem necessários para verificar o potencial uso desta tecnologia na terapêutica clínica contra infecções por P. insidiosum.

Resumo

The goat milk industry has unquestionably grown in recent years due to the high demand for dairy products, which are considered nutritious and hypoallergenic. As a result, security measures are required in the production chain to provide consumers with safe products, although the concept of biosecurity is still incipient in Brazilian goat farming. Therefore, this study aimed to review the main biosecurity measures applied to dairy goat farms and suggest a program that contemplates these measures to promote animal health and welfare, given that biosecurity bolsters environmental sustainability and improves this agricultural sector. Biosecurity programs (BP) are composed of a set of measures and procedures aimed at herd health and applied in all stages of animal husbandry, interacting with different sectors that make up the production system and providing a set of policies and operational norms to protect herds against potentially pathogenic agents. Thus, BP require planning, execution, monitoring, audits, updates, understanding of the procedures, and awareness of those involved in the production chain. In addition, it includes continuing education programs and the development of contingency plans for specific emergencies. The information included in this study provides input to dairy goat farms to implement practices that improve the productivity of this agricultural sector.


A caprinocultura leiteira é um setor produtivo em crescimento, devido à alta demanda dos produtos lácteos considerados nutritivos e hipoalergênicos. Em consequência disso, são exigidas medidas de segurança na cadeia produtiva a fim de fornecer produtos inócuos ao consumidor. No entanto, os conceitos de biosseguridade ainda são incipientes na caprinocultura brasileira. Desta forma, este estudo tem a finalidade de revisar as principais medidas de biosseguridade aplicadas à caprinocultura leiteira e sugerir um programa que contemple estas medidas visando promover a saúde e o bem-estar animal. Além disso, a biosseguridade propicia a sustentabilidade do meio ambiente e potencializa melhorias neste setor agropecuário. Salienta-se que um programa de biosseguridade (PB) é composto por um conjunto de medidas e procedimentos de atenção à saúde do plantel, aplicados em todas as etapas da criação animal, interagindo com diversos setores que compõe o sistema produtivo, além de propiciar a implantação de um conjunto de políticas e normas operacionais, com o objetivo de proteger os rebanhos contra a introdução de qualquer agente infeccioso potencialmente patogênico. O PB necessita de planejamento, execução, monitoramento, auditorias, atualizações, bem como demanda de compreensão dos procedimentos e da sensibilização de todos os envolvidos nos processos da cadeia produtiva. Adicionalmente, inclui programas de educação continuada de todos os indivíduos e a elaboração de planos de contingência para situações emergenciais específicas. As informações incluídas neste estudo fornecerão aporte às propriedades de caprinocultura de leite para a implementação de ações que promovam melhorias na produtividade deste setor agropecuário.

Resumo

The purpose of this review was to address the applicability of nanotechnology in veterinary medicine, with an emphasis on research in Brazil from 2013 to 2020. Firstly, we introduced to the general aspects of applicability of nanotechnology in veterinary medicine, and lately we pointed the research involving nanoscience performed in Brazil, in the studied period. Nanotechnology is the field of science that has the capacity to organize matter in nanoscale structures (1 to 100 nm), enabling innovations in different areas including biotechnology, agriculture, disease diagnosis, food and clothing industry, electronics, and pharmacological therapies. In veterinary medicine, several studies are being carried out in the world, mainly in the areas that involve search of new treatment options and the development of immunotherapy, as well as in the diagnosis of diseases. In Brazil, it is clear that the use of nanotechnology in veterinary medicine is still incipient, but it can be considered a growing area. In addition, several points have to be reflected and researched, including some adverse effects and implications to validate the safe use of nanotechnology in veterinary medicine. Therefore, this review highlighted the nanotechnology as a promise alternative in the current context of Brazilian technological innovation involving animal health, as well as a possible diagnostic tool and highlighting its potential therapeutic use in disease control in veterinary medicine. Regarding future perspectives, we believed that greater investment in science and technology could contribute to the advancement and strengthening of nanotechnology in Brazil.


O objetivo desta revisão foi abordar a aplicabilidade da nanotecnologia na medicina veterinária, com ênfase nas pesquisas no Brasil de 2013 a 2020. Primeiramente, apresentam-se os aspectos gerais da aplicabilidade da nanotecnologia em medicina veterinária e, posteriormente, apontam-se as pesquisas envolvendo nanociência realizadas no Brasil, no período estudado. A nanotecnologia é o campo da ciência capaz de organizar a matéria em estruturas de nanoescala (1 a 100 nm), permitindo inovações em diferentes áreas, incluindo biotecnologia, agricultura, diagnóstico de doenças, indústria de alimentos e vestuário, eletrônica e terapias farmacológicas. Na medicina veterinária, diversos estudos estão sendo realizados no mundo, principalmente nas áreas que envolvem a busca de novas opções de tratamento e o desenvolvimento de imunoterapia, bem como no diagnóstico de doenças. No Brasil, está claro que o uso da nanotecnologia na medicina veterinária ainda é incipiente, mas pode ser considerada uma área em crescimento. Além disso, vários pontos devem ser refletidos e pesquisados, incluindo alguns efeitos adversos e implicações para validar o uso seguro da nanotecnologia na medicina veterinária. Nesta revisão, destaca-se a nanotecnologia como uma alternativa promissora no atual contexto de inovação tecnológica brasileira, envolvendo saúde animal, o diagnóstico e o potencial uso terapêutico no controle de doenças em medicina veterinária. Em relação às perspectivas futuras, acredita-se que maiores investimentos em ciência e tecnologia são fatores fundamentais a contribuir para o avanço e fortalecimento da nanotecnologia no Brasil.

Bacteriophages have been investigated as alternative to the treatment of bacterial infections, including bovine mastitis, in production animals. In this meta-analysis, we evaluated in vitro efficiency of phages of Staphylococcus aureus against S. aureus, which is involved in the etiology of bovine mastitis. Seventeen studies were included and the bacterial lytic activity was extracted using proportion analysis. The lytic efficiency of phages was obtained in this meta-analysis using a random-effects model [significant difference (P<0.05)]. Forest plots were used to graphically represent the efficiency of phages on bacterial isolates. Most phages (e.g., CS1, DW2, ΦSA011, ΦSA012, ΦSA022, ΦSA023, ΦSA024, ΦSA025, ΦSA037, ΦSA038, ΦSA039, ΦSA041, ΦSA042, ΦSA043, ΦSA044, MSA6, Ufv-aur2 to Ufv-aur11, SAH-1, SPW, vB_SauM_JS25, SaPh1 to SaPh6, SA, SANF, SA2, ΦSA012, ΦSA039, phi11, phiIPLA88, phiIPLA35, phiIPLA-RODI, phiIPLA-C1C, SAJK-IND, vBSP-A1, vBSP-A2, STA1.ST29, EB1.ST11, EB1.ST27, Remus, and ISP) were efficiently lytics or infected most S. aureus isolates, demonstrating 80% (P<0.05) lytic efficiency. The phages SA, SANF and SA2, also demonstrated lytic activity or infected the non-Staphylococcus aureus and Macrococcus caseolyticus isolates. In this meta-analysis, we compared and demonstrated the in vitro efficiency and host range of S. aureus phages. Additionally, the phages represent an alternative to be researched to treat bovine mastitis in dairy cattle caused by the prevalent microorganism, S. aureus.

Background: Pythiosis is a chronic inflammatory disease that is caused by oomycete Pythium insidiosum. This illness affects several species including humans and horses. Equine is the most affected species, having no predisposition for breed, gender, or age. It is usually shown in cutaneous and subcutaneous forms, and the lesions, which grow quickly and are hard to treat, are located mainly in the extremities. The diagnosis is made via epidemiology, clinical signs, and macroscopic and microscopic aspects of the lesion. This study describes a case of cranioesophageal pythiosis in a horse, examining the epidemiological, clinical and pathological characteristics. Case: A 12-year-old male quarter horse, weighing 515 kg was taken to the Veterinary Hospital at the University Center of Espírito Santo (UNESC). The horse had an increase in volume in the cranioesophageal region, coughing, difficulty breathing, and a runny nose. On clinical examination, the horse showed an enlargement in the submandibular and retropharyngeal lymph nodes, subcutaneous edema in the larynx region, and a temperature of 38.2ºC. According to the owner, cough was recurrent and had lasted about 12 months even after treatment with different kinds of antimicrobials. On radiographic exam, there was a marked decrease in the tracheal lumen and increased soft tissue radiopacity in the region adjacent to the narrowing. The animal was taken to surgery to remove the mass, but he died because of complications during surgery. The animal’s owner did not allow necropsy, but a fragment of the mass in the cranioesophageal was removed and sent for histological examination. The fragment was fixed in 10% formalin and processed using routine histological analysis. Macroscopically, the mass was light yellowish and ulcerated, and it measured 7.0 × 5.0 × 5.0 cm. Resumo

The study evaluated the ovicidal activity of enzymatic extracts of Purpureocillium lilacinum and Trichoderma virens against trichostrongylid eggs from sheep. Filtered extract (FE) and macerated crude extract (MCE) were prepared from fungal cultures in minimal broth. In the experiment, 100 trichostrongylid eggs, obtained from the feces of naturally infected sheep, were exposed to fungal extracts for 24 and 48 hours/25°C. In the control group, eggs were incubated in minimal broth. The number of L1 larvae was ascertained. Each treatment consisted of four repetitions and the experiment was repeated five times. It was observed that the effect of FE and MCE of P. lilacinum and T. virens on egg hatchability differed from that of the control group. MCE of T. virens and P. lilacinum showed higher ovicidal activity than FE over both periods and at 48 hours of exposure, respectively. From the percentage reductions in hatchability of the eggs, MCE was shown to be superior to FE for both fungi. This study demonstrated the ovicidal potential of these fungi against trichostrongylid eggs. However, further studies are needed in order to identify the molecules responsible for the ovicidal effects, and to evaluate the behavior of fungal extracts in biotic and abiotic interactions.(AU)


O estudo avaliou a atividade ovicida de extratos enzimáticos de Purpureocillium lilacinum e Trichoderma virens sobre ovos de tricostrongilídeos de ovinos. Extrato filtrado (EF) e extrato macerado bruto (EMB) foram preparados a partir de culturas fúngicas em caldo mínimo. No ensaio experimental, 100 ovos de tricostrongilídeos, obtidos de fezes de ovinos naturalmente infectados, foram expostos durante 24 e 48 horas/25ºC aos extratos dos fungos. No grupo controle, os ovos foram incubados em caldo mínimo. O número de larvas L1 foi determinado. Cada tratamento consistiu em quatro repetições e o experimento foi repetido cinco vezes. Observou-se que o efeito ovicida do EF e EMB de P. lilacinum e T. virens diferiu do grupo controle. O EMB de T. virens e P. lilacinum evidenciou atividade ovicida superior ao EF em ambos os períodos avaliados e em 48 horas de exposição, respectivamente. O percentual de redução de eclodibilidade evidenciou que o EMB foi superior ao EF em ambos os fungos. Este estudo demonstra o potencial ovicida desses fungos sobre ovos de tricostrongilídeos. No entanto, estudos adicionais são necessários para identificar as moléculas responsáveis pelo efeito ovicida, bem como avaliar o comportamento dos extratos fúngicos em interações bióticas e abióticas.(AU)

Resumo

Purpureocillium lilacinum is a nematophagous fungus used in biological control against some parasites, including Toxocara canis. This study researched the infectivity of embryonated T. canis eggs after exposure to the fungus P. lilacinum. T. canis eggs were exposed to P. lilacinum for 15 or 30 days and subsequently administered to Swiss mice (n=20). Control group consisted of mice who received T. canis embryonated eggs without fungal exposure. Forty-eight hours after infection, heart, lung, and liver from animals of each group were collected to assess larval recovery. The organs of mice that received embryonated eggs exposed to the fungus showed a lower average larval recovery (P 0.05) suggesting that exposure of T. canis eggs to P. lilacinum was able to reduce experimental infection. Under the evaluated conditions, the interaction time between the fungus and the parasite eggs was not a significant factor in larvae recovery. P. lilacinum may be considered a promising T. canis biological control agent. However, further studies are needed to determine a protocol for the use of this fungus as a biological control agent.(AU)


Purpureocillium lilacinum é um fungo nematófago com potencial para uso no controle biológico de parasitos, incluindo Toxocara canis. Este estudo pesquisou a infectividade de ovos de T. canis embrionados após exposição ao fungo P. lilacinum . Ovos de T. canis foram expostos ao fungo por 15 ou 30 dias e subsequentemente administrados a camundongos Swiss (n=20). O grupo controle consistiu de camundongos que receberam ovos embrionados do parasita sem exposição ao fungo. Quarenta e oito horas após a infecção, coração, pulmão e fígado dos camundongos foram coletados para avaliar a recuperação larval. Os órgãos dos animais que receberam ovos embrionados expostos ao fungo apresentaram menor média de recuperação larval (P 0,05) do que os infectados com ovos sem exposição ao fungo, sugerindo que a exposição dos ovos de T. canis a P. lilacinum foi capaz de reduzir a infecção experimental. Nas condições avaliadas, o tempo de interação entre o fungo e os ovos do parasito não foi um fator significativo na recuperação das larvas. P. lilacinum pode ser considerado um promissor agente de controle biológico de T. canis, no entanto, mais estudos são necessários para avaliar o emprego deste fungo como um agente de controle biológico.(AU)

We aimed to genotype the South American clinical isolates of Pythium insidiosum using the single nucleotide polymorphisms (SNP) of the ribosomal DNA sequences (rDNA). Previously, an SNP-based multiplex-PCR was able to distinguish three different clades of P. insidiosum isolates. Thus, we used this assay to evaluate South American clinical isolates of P. insidiosum (n=32), standard strains from Costa Rica (n=4), Thailand (n=3), Japan (n=1), and India (n=1), a standard strain of Pythium aphanidermatum, and Brazilian environmental isolates of Pythium torulosum, Pythium rhizo-oryzae and Pythium pachycaule voucher (n=3). It was possible to allocate each American P. insidiosum isolate to clade I, the isolates of India, Japan, and Thailand to clade II, and the Thai isolate to clade III. P. aphanidermatum, P.torulosum, P.rhizo-oryzae and P.pachycaule voucher isolates were not amplified. For the first time, a P. insidiosum isolate from Uruguay, South America, was included in molecular analyzes. By SNP-based multiplex-PCR, it was possible to perform the identification and genotyping of the South American isolates of P. insidiosum, demonstrating similar genetic characteristics of these isolates.(AU)


O objetivo deste estudo foi genotipar isolados clínicos de Pythium insidiosum da América do Sul utilizando polimorfismos de nucleotídeo único (SNP) de sequências de rDNA. Anteriormente, um multiplex-PCR baseado em SNP foi capaz de distinguir P. insidiosum em três diferentes clados. Dessa forma, utilizamos este método para avaliar isolados clínicos de P. insidiosum da América do Sul (n=32), cepas padrão da Costa Rica (n=4), Tailândia (n=3), Japão (n=1) e Índia (n=1), uma cepa padrão de Pythium aphanidermatum e isolados ambientais brasileiros de Pythium torulosum; Pythium rhizo-oryzae e Pythium pachycaule voucher (n=3). Os isolados analisados foram alocados aos clados: I (americanos), II (isolados da Índia, Japão e Tailândia), e III (um isolado tailandês). P. aphanidermatum, P.torulosum, P.rhizo-oryzae e P.pachycaule voucher não foram amplificados. Pela primeira vez, um isolado de P. insidiosum do Uruguai foi incluído em análises moleculares. Através da multiplex-PCR baseada em SNP, foi possível realizar a identificação e genotipagem dos isolados sul-americanos de P. insidiosum, demonstrando características genéticas semelhantes entre esses isolados.(AU)

Successful disease treatment depends on molecular studies under indoor conditions with experimental infection protocols that facilitate understanding the disease and the drug`s efficacy. The internal transcribed spacer (ITS) region was sequenced from three isolates, which were identified as Saprolegnia aenigmatica. Subsequently, healthy fish were immunosuppressed with dexamethasone (1.2 mg kg-1) and descaled to the skin using a sharp scalpel. These individuals were isolated in individual aquariums maintained at 22°C. Individuals in one group were subcutaneously inoculated with 9,000 zoospores (DDZ treatment), a second group was exposed to oomycetes in water with three colonized baits (DDB), a third group was maintained in water without zoospores (DD), and a control group (C) consisted of healthy animals. After 48 and 96 hours, two animals from each group were euthanized for fungal reisolation. The fish from groups DD and C did not show clinical signs, and no oomycetes were isolated. The animals from the DDZ and DDB groups showed cotton-wool-like masses on the skin, and S. aenigmatica was re-isolated. Thus, for infection using zoospores or baits parasitized by S. aenigmatica, an immunosuppressor (dexamethasone) and a sharp scalpel can be used effectively to establish an experimental infection in P. mesopotamicus.


O sucesso do tratamento de uma enfermidade depende do estudo de moléculas em condições de laboratório por meio de protocolos de infecção experimental que viabilizam a compreensão da doença e da eficácia dos fármacos. Pela sequência ITS foram identificadas três cepas de Saprolegnia aenigmatica. Dessa forma, pacus sadios foram submetidos à imunossupressão com dexametasona (1,2 mg kg-1), esfoliados com auxílio de bisturi e distribuídos em aquários a 22ºC. Após este procedimento, um grupo de animais foi inoculado com 9.000 zoósporos/peixe subcutâneo (DEZ), a outro foram adicionadas três iscas colonizadas com o oomiceto na água (DEI), um terceiro grupo foi mantido sem contato com o oomiceto (DE) e um quarto grupo, de animais sadios, representaram o controle (C). Após 48 e 96h deste procedimento, foram eutanaziados animais de cada grupo para reisolamento. Os animais do grupo DE e C não apresentaram sinais clínicos e não foi reisolado o oomiceto. Porém, tanto os animais do grupo DEZ quanto como os animais do grupo DEI apresentaram micélio branco na pele e foi reisolado Saprolegnia aenigmatica. Assim, a infecção com zoósporos ou com iscas colonizadas por S. aenigmatica, com o uso de dexametasona e abrasivo epitelial são formas eficazes de infecção experimental em P. mesopotamicus.

This study investigated the frequency of oocysts of Cryptosporidium spp. in feces from dogs and cats in five municipalities in the southern region of the state of Rio Grande do Sul. The risk factors associated with infection were also investigated. Feces samples from 110 dogs and 18 cats were stained using the auramine method. At the time of feces sampling, a questionnaire with semi-open-ended questions was applied to the animal guardians and all data obtained underwent statistical analysis. The real frequency of oocysts of Cryptosporidium spp. was 24.63% (27 dogs and two cats). Only four samples of dog feces were diarrheic and no presence of oocysts was observed in any of them. Variables that represented risk factors for infection were: homemade food, untreated water, circulation of animals on grassy terrain and living in the same environment as other animals (cattle). The results made it possible to inferring that within the population studied, the frequency of parasitism due to Cryptosporidium spp. in dogs was relevant and emphasize the asymptomatic nature of this infection. The adopting control measures are highlighted, particularly in relation to variables that represent risk factors for this infection.(AU)

No momento da coleta de fezes aplicou-se um questionário aos tutores dos animais com questões semiabertas e os dados foram submetidos à análise estatística. A frequência verdadeira de oocistos de Cryptosporidium spp. foi de 24,63% (27 cães e 2 gatos). Apenas quatro amostras de fezes caninas eram diarreicas e todas sem oocistos. As variáveis que representaram fatores de risco para a infecção foram: alimentos de preparo caseiro, água não tratada, circulação dos animais em terreno gramíneo e convivência com outros animais, principalmente bovinos. Os resultados sugerem que a frequência de cães parasitados por Cryptosporidium spp. é relevante, reforçando o caráter assintomático da infeção. Destaca-se a importância da adoção de medidas de controle, particularmente das variáveis que representaram fatores de risco à infecção.(AU)

Background: Balantidium coli is a commensal protozoan that infects several animals, but it has pigs as its natural reservoir. In the presence of predisposing factors, B. coli can become pathogenic for swine, causing enteric lesions. Infections determined by this protozoan may be a risk to public health, due to dysentery in animal keepers and veterinarians. This study aimed to determine the occurrence of infection by B. coli in pigs of distinct husbandry categories, as well as unlike physiological state, kept in farms with different hygienic-sanitary standards, located in the central region of Rio Grande do Sul State in Brazil. Materials, Methods & Results: Stool samples were collected from 12 different farms with different hygienic-sanitary standards being four farms of finisher pigs (G1; n = 287), four farms with pregnant females (G2; n = 60) and four farms with lactation sows (G3; n = 40), and all samples were submitted to coproparasitological analyses to verify the presence of cysts or trophozoites of B. coli. The infection levels were considered mild (1-100 cyst/trophozoite), moderate (101- 300 cyst/trophozoite) and high (>300 cysts/trophozoite). In addition, information about hygienic-sanitary conditions of each farm was collected. The occurrence of B. coli infection in all swine stool samples analyzed was 60.9% (236/387); however, in G1, G2 and G3 was 54.7% (157/287), 91.7% (55/60) and 65% (26/40) respectively. There was significant difference in the occurrence of G1 (P < 0.05) except between farms B and C (67.9% and 56.6% respectively). There was also a significant difference (P < 0.05) between the occurrence of B. coli found in G2 and G3. The infection levels were considered predominantly mild in G1; and mild to moderate in G2 and G3. Background: Escherichia coli (E. coli) is an enteropathogen that commonly causes diarrhea in calves. However, not all E. coli isolates are pathogenic. The aim of this study was to identify E. coli virulence factors derived from fecal samples collected in the southern region of Rio Grande do Sul state (RS) from calves with and without diarrhea, as well as investigate the antimicrobial susceptibility of E. coli isolates from calves with diarrhea. Materials, Methods & Results: Forty stool samples were collected in 12 farms, each one from calves having one day to six months of age, with and without diarrhea. The total DNA of from these isolates was extracted and a PCR using primers specific for the virulence factors Stx1, Eae, F41, F5 and STa was conducted. The susceptibility testing used the disk diffusion method and the susceptibility profile was evaluated against the following antimicrobials: ampicillin, penicillin, chloramphenicol, enrofloxacin, gentamicin, trimethoprim, sulfonamide, tetracycline and streptomycin. From all calves, 15 (15/40, 37.5%) had diarrheal stools and 25 (25/40, 62.5%) had normal or semi-liquid stools. Twelve (12/40; 30%) E. coli isolates showed at least one virulence factor. These factors were found in four isolates (4/15; 26.6%) from diarrheal stools and eight isolates (8/25; 28.5%) from normal stool. The Stx1 factor was identified in five isolates (5/40; 12.5%), and the Eae and the Sta factors in one (1/40; 0.2%) and in atypical associations between Stx1 and Eae and also between Eae and F41 in two isolates (2/40; 0.5%). Also, the Eae and Sta factors were identified in one isolate (1/40; 0.2%). The susceptibility test showed resistance to penicillin and tetracycline in 93% and 80% of the tested isolates, respectively. The aim of this study was to identify E. coli virulence factors derived from fecal samples collected in the southern region of Rio Grande do Sul state (RS) from calves with and without diarrhea, as well as investigate the antimicrobial susceptibility of E. coli isolates from calves with diarrhea. Materials, Methods & Results: Forty stool samples were collected in 12 farms, each one from calves having one day to six months of age, with and without diarrhea. The total DNA of from these isolates was extracted and a PCR using primers specific for the virulence factors Stx1, Eae, F41, F5 and STa was conducted. The susceptibility testing used the disk diffusion method and the susceptibility profile was evaluated against the following antimicrobials: ampicillin, penicillin, chloramphenicol, enrofloxacin, gentamicin, trimethoprim, sulfonamide, tetracycline and streptomycin. From all calves, 15 (15/40, 37.5%) had diarrheal stools and 25 (25/40, 62.5%) had normal or semi-liquid stools. Twelve (12/40; 30%) E. coli isolates showed at least one virulence factor. These factors were found in four isolates (4/15; 26.6%) from diarrheal stools and eight isolates (8/25; 28.5%) from normal stool. The Stx1 factor was identified in five isolates (5/40; 12.5%), and the Eae and the Sta factors in one (1/40; 0.2%) and in atypical associations between Stx1 and Eae and also between Eae and F41 in two isolates (2/40; 0.5%). Also, the Eae and Sta factors were identified in one isolate (1/40; 0.2%). The susceptibility test showed resistance to penicillin and tetracycline in 93% and 80% of the tested isolates, respectively. Resumo

Efficient handling programs that provide high quality colostrum in adequate amounts to dairy farm calves are needed to assure their health and survival. Replacers (or milk substitutes) often become necessary when colostrum presents inadequate quality, or in order to break the cycle of infectious disease transmission. In this study we aimed to assess the effect of anaerobic fermentation processing (colostrum silage) on bacterial that represent interest to animal health. Colostrum samples were inoculated with cultures of Brucella abortus , Escherichia coli , Leptospira interrogans serovar Copenhageni , Mycobacterium bovis , Salmonella Enteritidis , Salmonella Typhimurium , Staphylococcus aureus , and Bacillus cereus and then subjected to anaerobic fermentation. On the first day, and every seven days until 30th days after fermentation, the samples were cultured and colony forming units counted. At seven days of fermentation, B. abortus , L. interrogans , and M. bovis were not detected. At 14th days of fermentation, E. coli , S. aureus , S. Enteritidis and S. Typhimurium were no longer detected. However, we were able to detect both lactic acid bacteria and B. cereus until 30th days of fermentation. From this study we suggested that anaerobic fermentation processing can inhibit important bacteria that cause economical losses for the cattle industry. The observations suggested that colostrum silage is a promising form to conserve bovine colostrum.(AU)


Eficientes programas de manejo que permitem o fornecimento de colostro de alta qualidade e em quantidades adequadas para bezerros são necessários para garantir a sua saúde e sobrevivência. Substitutos do leite tornam-se frequentemente necessários quando o colostro apresenta qualidade inadequada ou a fim de quebrar o ciclo de transmissão de doenças infecciosas. Este estudo teve como objetivo avaliar o efeito da fermentação anaeróbica do colostro bovino (silagem de colostro) sobre bactérias de interesse para a saúde animal. Alíquotas de colostro foram inoculadas com culturas de Brucella abortus , Escherichia coli , Leptospira interrogans serovar Copenhageni , Mycobacterium bovis , Salmonella Enteritidis , Salmonella Typhimurium , Staphylococcus aureus e Bacillus cereus e submetidas ao processo de fermentação anaeróbia. No primeiro dia e a cada sete dias até o 30o dia de fermentação, as alíquotas foram cultivadas e analisadas pela contagem de unidades formadoras de colônias. Aos sete dias de fermentação, unidades formadoras de colônias de B. abortus, L. interrogans e M. bovis não foram detectadas e unidades formadoras de colônia de E. coli , S. aureus , S. Enteritidis e S. Typhimurium não foram observadas no 14o dia de fermentação. No entanto, B. cereus e bactérias ácido lácticas foram detectadas até o 30o dia de fermentação. Este estudo sugere que o processo de fermentação anaeróbia do colostro bovino proporciona efeitos inibitórios sobre as bactérias avaliadas, constituindo-se num promissor método de conservação do colostro bovino.(AU)
