Resumo

Folic acid is closely linked to cobalamin, which is a carrier of hydroxymethyl and ant groups. The objective of this work was to evaluate the effect of adding folic acid to the TRIS-yolk diluter and possible effect on the membrane, mitochondria and acrosome of sheep sperm after thawing the semen. There were six sheep and seven collections of each animal in sessions between 48 and 72 hours. After collection and analysis, the semen samples were mixed and submitted to the formation of a pool. Diluted in TRIS-yolk medium, and divided into 3 groups; Group control; Group 2: 10,000 µM of folic acid and in Group 3: 5000 µM of folic acid. Subsequently, the semen samples were packaged in 0.25 mL straws and processed in a semen freezing machine. After a few days, the semen was thawed and evaluated: plasma membrane integrity, acrosome function and integrity. They were analyzed using an analysis of variance (ANOVA) followed by the Newman-Keuls test, using SAS. The additionof a micronutrient with potential antioxidant action in the semen indicates that it can reduce the action of free radicals that alter the plasma membrane and sperm DNA. The evaluation of the plasma membrane integrity, mitochondrial activity and the acrosome integrity of post-thawed sperm were not significantly different between the groups evaluated. Thus, it is concluded that the addition of folic acid in concentrations of 5000 µM and 10000 µM to the TRIS-yolk seminal diluter does not significantly influence the variables evaluated in this experiment.

The research aimed to evaluate the antioxidant effect of supplementation of 0.5μM, 5μM and 50μM of oleic acid to the TRIS-yolk extender on the mitochondrial potential (MIT) during the cryopreservation of goat sperm. For that, four Anglo-nubian goats were used, in which five samples / animal were collected, using artificial vagina. After evaluating the swirling and motility of the ejaculates, the pool was made, then diluted in TRIS-Gem and divided according to the treatments. After processing, the samples were packaged in 0.25mL straws and cryopreserved using the TK 3000® machine. After a minimum of 5 days of storage in a cryogenic cylinder, thawing was performed to assess the MIT of goat sperm after cryopreservation, using the lipophilic cationic fluorochrome JC-1. The data were submitted to analysis of variance (ANOVA), using the general linear models procedure (Proc GLM), and the Duncan test was used to compare the averages, with a 5% probability. The analyzes were performed using the Statistical Analysis System program (SAS Institute Inc, 2013). Thus, it was observed that the concentrations of 0.5μM and 5μM of oleic acid maintained the mitochondrial potential similar to the control, differing (p<0.05) only the concentration of 50μM. It can be concluded that 0.5μM and 5μM oleic acid are able to maintain the mitochondrial potential, prolonging the viability of cryopreserved goat sperm.

The analyzes were performed using the Statistical Analysis System program (SAS Institute Inc, 2013). After analysis, it was observed that the control group had the best percentage, and differed significantly (p<0.05) from the treatment with 50μM of arachidonic acid. It was concluded that the 50μM arachidonic acid concentration is not effective to maintain the integrity of the plasma membrane, and to minimize the oxidative stress of cryopreservation.

Objetivou-se verificar os efeitos, nos parâmetros espermáticos, na integridade mitocondrial, acrossomal e de membrana em células espermáticas, desencadeados pelo uso do Tris (Tris hidroximetil aminometano) suplementado com óleo de Mauritia flexuoxacomo diluente para criopreservação de sêmen caprino. Quatro caprinos clinicamente saudáveis foram utilizados. Os animais eram alimentados diariamente com volumoso (Pennisetum purpureum, Schum.), concentrado (ração peletizada com teor de 20% proteína, 300 g/animal/dia) e sal mineral específico para Caprinos (Caprinofós®), à vontade. Dois ensaios foram realizados: I ­ Teste de toxicidade; II ­ Criopreservação do sêmen com concentrações ideais. No teste de toxicidade as concentrações avaliadas foram: 5%, 10%, 15% e 20% de diluente a base de óleo de Mauritia flexuoxa. Após o teste de toxicidade, foi escolhido a concentraçãoque apresentou o melhor resultado (5%). Logo após, foram realizadas mais 32 coletas, que foram diluídas em Tris-gema-glicerol (grupo controle) ou diluente contendo óleo vegetal (Mauritia flexuoxa). As amostras foram criopreservadas com auxílio do aparelho Tk3000®. Após o período mínimo de uma semana as palhetas foram descongeladas em banho-maria a 37 °C por 30 segundos, acondicionadas em microtubos de centrifugação e homogeneizadas para a análise imediata de motilidade, vigor espermático e morfologia. Em seguida, por meio de sondas fluorescentes foram avaliadas a integridade de acrossomo, membrana plasmática (Diacetato de Carboxifluresceína e Iodeto de Propídeo) e função mitocondrial sob microscopia de epifluorescência. Quanto a motilidade e vigor, integridade mitocondrial e acrossomal, o grupo buriti foi inferior ao grupo controle. O Tris suplementado com óleo de Mauritia flexuoxa na concentração de 5% não influenciou significativamente a qualidade espermática, porém, observou-se morfologia e integridade de membrana favoráveis. Dessa forma, sendo uma alternativa para substituição de diluentes a base de produtos de origem animal.

As for motility and vigor, mitochondrial and acrosomal integrity, the buriti group was inferior to the control group. Tris supplemented with Mauritia flexuoxa oil at a concentration of 5% did not significantly influence sperm quality, however, favorable motility, morphology and membrane integrity were observed. Thus, being an alternative to replace diluents based on products of animal origin.

Soon after, a further 32 samples were obtained, which were diluted in Tris-glycerol (control group) or diluent containing vegetable oil (Mauritia flexuoxa). The samples were cryopreserved using the Tk3000® machine. After a minimum of one week, the samples were thawed in a 37 ° C water bath for 30 seconds, packed in centrifugation microtubes and homogenized for immediate analysis of motility, sperm vigor and morphology. Then, by means of fluorescent probes, the integrity of the acrosome, plasma membrane (Carboxyflurescein diacetate and Propidium Iodide) and mitochondrial function under epifluorescence microscopy were evaluated. As for motility and vigor, mitochondrial and acrosomal integrity, the buriti group was inferior to the control group. Tris supplemented with Mauritia flexuoxa oil at a concentration of 5% did not significantly influence sperm quality, however, favorable motility, morphology and membrane integrity were observed. Thus, being an alternative to replace diluents based on products of animal origin.

Resumo

This study aimed to evaluate physically and structurally ejaculates from locally adapted goats in therainy season. Semen pool formed by the four goats of each breed was evaluated for physical, diluted in Tris-eggyolk and frozen. After thawing, it was diluted in Tris and incubated with carboxyfluorescein diacetate andpropidium iodide, to evaluate the integrity of the plasma membrane and JC1 to assess mitochondrial activity.The percentage of cells with damaged membrane and mitochondrial activity in spermatozoa between breed werecompared, every hour for three hours. There was no significant difference (P > 0,05) in relation to thepercentage of cells with damaged plasma membrane, as well as between the incubation times. The Azul breedhad lower mitochondrial activity (P < 0,05). The evaluated semen of all breeds showed no significant damage tothe plasma membrane in the rainy season, but the Azul breed had lower mitochondrial activity in sperm cells.(AU)

Resumo

We evaluated the effect of Recombinant Bovine Somatotropin (rbST) seven days before the start of thesynchronization protocol on pregnancy rate in goats. Were used 101 females without defined breed in semiintensivesystem, randomly divided into two groups. Seven days before the start of estrus synchronization, G1 (n= 49) received 125 mg of rbST, IM, and G2 (n = 52) physiological saline, subcutaneously. In D0, the groupsreceived 1 mL of BE, IM, and were synchronized with intravaginal sponges impregnated with 60 mg ofmedroxyprogesterone for 11 days. In D9, they received 300 IU of eCG and 75 µg of PGF2α. On D11, thesponges were removed, we performed IATF 36 and 48 hours later, transcervically. It was used pool of AngloNubians semen. Gestational diagnosis was performed 30 days later. A single dose administration of rbST, sevendays before estrus synchronization, did not increase the pregnancy rate.(AU)

Resumo

The semen cryopreservation process causes damages in spermatozoa characteristics. This study aimedto evaluate the effects of tea of Copaíba extract (Copaifera luetzelburgii), in different concentrations, added tothe Tris-egg yolk extender, in the quality of post-cryopreserved spermatozoa from locally adapted breeds ofgoats. Were used four (4) goats Breeds (Azul, Canindé, Moxotó e Repartido) adults, male, six of each breed,aged between 1 and 6 years, a pool of ejaculates were formed to analyze. The animals were randomly dividedinto four experimental treatments: TI (Control), TII (0,1mg), TIII (0,2mg) and TIV (0,5mg). The samples werefrozen and stored, after 30 days they were thawed for analysis by term-resistance test (TTR). The addition of teaof copaíba extract showed no significant difference between treatments by TTR, therefore the use of tea copaíbaextract in seminal diluting not increment the sperm longevity post-thawing.(AU)

Resumo

The aim of this research was to evaluate the effects of supplementation of maturation medium withcaptopril in oocyte in vitro maturation (IVM). 470 CCOs were recovered and classified into grades I and II,being divided into four groups: G1 (n = 56), that was control group; G2 (n = 152) 20 mM of captopril; G3 (n =126) 40 uM of captopril; and G4 (n = 136) 80 uM of captopril, then these were subsequently submitted to IVMprocess for 24 hours at 38.5°C in 5% CO2. After 24 hours of maturation, oocytes were denuded and evaluatedfor the first polar body extrusion and were, therefore, considered matured. The addition of the Captopril inmedium of oocytes maturation has not improved the IVM rate.(AU)

Resumo

This study evaluated the quality of ejaculated from adapted breeds of goats as the morphologic aspects,functional and thermoresistance. An aliquot of the pool of each breed was used for morphological evaluation bywet preparation, quantifying sperm defects and dividing them into classes. The samples were cryopreservedusing TK3000 machine and evaluated by thermoresistance test by checking motility and sperm vigor every hour.For plasma membrane functionality analysis, was used hiposmotic test. There was no difference (P > 0.05)between the breeds in all sperm defects before and after cryopreservation, as well as for the thermoresistancetimes for motility and sperm vigor (P > 0.05). There was a significant reduction in motility and sperm vigor 120minutes after thawing of the samples. There was no difference (P > 0.05) between the percentage of sperm withfunctional membrane between the evaluated breeds. Therefore, the evaluated breeds have morphology,functionality and similar sperm thermoresistance.(AU)

This study aims to evaluate the effects of tea of copaíba extract (Copaifera luetzelburgii), in differentconcentrations, added to the Tris-egg yolk extender, the functionality of the plasma membrane sperm ofcryopreserved goats. Were used four goats breeds, adults, male, six of each breed, aged between 1 and 6 years,a pool of ejaculates were formed to analyze. The animals were randomly divided into four experimentaltreatments: TI without adding of tea of copaíba extract (control); TII, added 0,1 mg of tea of copaíba extract;TIII, 0,2 mg; and TIV, 0,5 mg. The aim of this study was to evaluate the effects of supplementation of maturation medium withcaptopril, and its consequences in the bovine maturation, fertilization and embryo development in vitro. 326bovine ovaries from a local slaughterhouse were used. 1101 CCOs were recovered and distributed in fourgroups: G1 (n = 112), that was control group; G2 (n = 322) 20μM of captopril; G3 (n = 367) 40μM ofcaptopril; and G4 (n = 300) captopril 80μM and later submitted to IVM. The matured CCOs were fertilized, andco-incubated. After fertilization, 676 presumptive zygotes were cultured and maintained in the greenhouse for 7days. The total number of viable embryos was 12; 39; 32 and 31, respectively in the experimental groups.Considering the experimental conditions adopted, it was concluded that the addition of the Captopril in mediumof oocytes IVM positively doesnt influence embryonic development, as evidenced by the similar percentages ofembryo production.(AU)

This study aims to evaluate the fertility rate of sheep semen diluted and cooled to 4ºC in powdercoconut water (ACP-102®) with and without egg yolk in artificial insemination by cervical route in sheep. Tworam lambs and 72 ewe lambs were used, divided into 3 groups: ACP-102® without egg yolk and cooled to 4ºCfor 12 hours, ACP-102® with 2,5% egg yolk also cooled as the first group and a control group with fresh semendiluted in ACP-102® without egg yolk. Pregnancy diagnosis was performed 30 days later. Pregnancy rates weresignificantly different (P < 0.05) between ACP-SGF (58,3%) and ACP-SG4 (20,8%) groups. The ACP-CG4(37,5%) group did not differ in relation to the ACP-SGF. Therefore, the addition of egg yolk to ACP-102®extender favors the viability of sheep semen, cooled to 4ºC for 12 hours and fertility of artificially inseminatedsheep by cervical route.(AU)

Resumo

The aim of this study was to investigate the effects of Tris use supplemented with Mauritia Flexuoxa oilas diluent for goat semen cryopreservation. four goats, clinically healthy were used. The animals were fed dailywith roughage, concentrate and mineral salt will. Initially, we conducted a toxicity test total of 32 collections.After the toxicity test, the concentration was chosen that showed the best result. Soon after, there were 32 morecollections, which were diluted in Tris-egg yolk-glycerol (control group) or GC diluent containing vegetable oil(Mauritia flexuoxa) GB. Samples were cryopreserved with the aid of Tk3000® device. Subsequently sampleswere thawed and analyzed for motility and sperm morphology force. It was observed obtaining post thawmotility in the group plus oil burity which shows a possible alternative to replacement products derived fromanimal origin.(AU)
