Resumo

ABSTRACT: Toxoplasma gondii can be eliminated in bovine semen. Cryopreserved semen is often used due to the fact that artificial insemination in dairy and beef cattle provides benefits in terms of production. However, little is known regarding the viability and infectivity of T. gondii tachyzoites in cryopreserved bovine semen. In the present study, cattle semen negative for T. gondii were contaminated with 1 x 106 tachyzoites (RH strain) and cryopreserved with and without different cryoprotectants, such as DMSO (concentrations of 2.5%, 5.0%, 7.5%, 8.0% and 10.0%) and glycerol (2.25%, 2.5%, 3.0%, 5.0%, 7.5% and 10.0%), followed by freezing in liquid nitrogen (-196°C). After 24 hours, the samples were thawed and inoculated in 10 mice per cryoprotectant concentration. The mice were evaluated for clinical signs of toxoplasmosis (rough coat, diarrhea, hypoactivity and sudden death) as well as serum titers of IgM and IgG and the presence of tachyzoites in the peritoneal lavage. The results revealed that T. gondii remained infective in all samples. Clinical signs of toxoplasmosis were observed in the mice beginning with the 6th day post-inoculation (DPI) and 100% lethality was found between the 7th and 9th DPI. Viable tachyzoites were recovered from peritoneal exudate of dead mice (except for the control group), with higher mean of tachyzoite counts in the intraperitoneal lavage for 5% DMSO (±3.32 x 106), 8% DMSO (±3.53 x 106), 3% glycerol (±4.75 x 106), 7.5% glycerol (±6.26 x 106) and the absence of cryoprotectant (±3.11 x 106). Seroconversion occurred in the treated groups, with titers of IgG from 1:16 to 1:128 and IgM from 1:16 to 1:512. T. gondii viability and infectivity were maintained in cattle semen during 24 hours of cryopreservation at -196°C with and without cryoprotectant. However, further studies are necessary to determine whether cryopreserved semen contributes to the spread of toxoplasmosis through artificial insemination.


RESUMO: Sabe-se que Toxoplasma gondii pode ser eliminado no sêmen bovino. A inseminação artificial em bovinos leiteiros e de corte proporcionou avanços e benefícios nas produções e para isso o sêmen criopreservado é frequentemente utilizado. No entanto, pouco se sabe sobre a viabilidade e infectividade dos taquizoítos de T. gondii em sêmen bovino criopreservado. Para isso o sêmen bovino, negativo para T. gondii, foi contaminado com 1x106 taquizoítos (cepa RH), criopreservados com ou sem diferentes crioprotetores como DMSO (2.5%, 5.0%, 7.5%, 8.0% e 10.0%) e Glicerol (2.25%, 2.5%, 3.0%, 5.0%, 7.5% e 10.0%) e congelados em nitrogênio líquido (-196°C). Após 24 horas, essas amostras foram descongeladas e inoculadas em 10 camundongos por diluente de concentração de crioprotetor. Os camundongos foram avaliados quanto a sinais clínicos de toxoplasmose (pele áspera, diarreia, hipoatividade e morte súbita), títulos séricos de IgM e IgG e presença de taquizoítos no lavado peritoneal. Os resultados mostraram que T. gondii se manteve infectante em todas as amostras, inclusive naquelas sem crioprotetor. Sinais clínicos de toxoplasmose foram observados nos camundongos a partir do 6º dia pós-inoculação (DPI) e 100% de letalidade foi verificada entre o 7º ao 9º DPI. Nos camundongos mortos, exceto no grupo controle, taquizoítos viáveis foram recuperados do exsudato peritoneal, com maior média de taquizoítos quantificados na lavagem intraperitoneal para DMSO a 5% (±3.32x106), 8% (±3.53x106) e glicerol 3% (±4.75x106), 7,5% (±6.26x106) e livre de crioprotetor (±3.11x106). A soroconversão ocorreu nos grupos tratados com títulos de IgG (1:16 a 1:128) e IgM (1:16 a 1:512). A viabilidade e infectividade do T. gondii no sêmen bovino durante as 24 horas de criopreservação a -196°C foram mantidas com ou sem crioprotetor. No entanto, mais estudos são necessários para verificar se o sêmen criopreservado contribui para a disseminação da toxoplasmose na inseminação artificial.

Resumo

Although cryopreservation is an efficient method for maintaining the biological and genetic resources of sperm, the sperm damage during the cryopreservation process cannot be ignored. It should be possible to obtain the most effective cryopreservation performance by accurately grasping the effects of various factors on the cryopreservation of sperm. The previous study demonstrated that a suitable standard protocol for cryopreservation of Korean native brindled cattle (Chikso) does not exist, based on the methods for semen cryopreservation of Chikso differ in each research center. The most obvious difference between most of protocols is the addition of glycerol before and after cooling during the Chikso cryopreserved semen process. Therefore we focused on the effects of glycerol addition time on the quality of cryopreserved Chikso sperm. In the present study, 27 individual Chikso samples were collected by transrectal massage and divided into two parts: the "cryopreservation method A" group (adding glycerol before cooling) and the "cryopreservation method B" group (adding glycerol after cooling). Meanwhile, the values of various sperm parameters were derived from each group, including sperm motility, kinematics, capacitation status, cell viability, and intracellular ATP levels, which we used to compare and evaluate sperm function. The results of this study indicated that during the semen cryopreservation process of the Chikso, the addition of glycerol after cooling yielded superior results in a variety of sperm parameters, such as sperm motility, progressive motility, rapid motility, VCL, VSL, VAP, ALH, capacitation status, viability, and intracellular ATP level after freezing and thawing. Our study is suggested that the glycerol addition time during the cryopreservation process for Chikso should be considered. In addition, our results may be provided reference to develop suitable the cryopreservation procedure of the Chikso sperm.(AU)

ABSTRACT Purpose: The development of cutting surface leakage and postoperative peritoneal adhesions (PPA) after sleeve gastrectomy (SG) are the most serious operative complications. We investigated the effectiveness of the newly developed glycerol and sodium pentaborate containing formulation on the prevention of these complications. Methods: Sixteen Sprague Dawley rats (mean weight 310 ± 50 g, mean age 3 months old) were divided into two groups, consisting of eight rats in each. SG and a double-layer suture technique were performed for each group. In study group, there was the mixture of 2 mL 3% glycerol plus 3% sodium pentaborate formulation, and in the control group 2 mL 0.9% NaCl was injected into the peritoneal cavity. Rats were sacrificed after 30 days, then macroscopic adhesion grade scoring and histopathological evaluations were assessed. Results: Macroscopic PPA scores in the control and study groups were 2.75 ± 0.16 and 1.50 ± 0.327, respectively (p = 0.004). Histopatologic fibrosis scores in the control and study groups were 0.87 ± 0.125 and 2.00 ± 0.26, respectively (p = 0.002). Conclusions: In SG operation model, glycerol plus sodium pentaborate compound decreased PPA formation and also increased stomach cut surface line fibrosis. This new formulation is hopeful for more safe SG operations.

ABSTRACT: Effects of plasticizer types and whey protein concentrate (WPC) as coatings were evaluated on internal quality and shelf life of eggs stored for 42 days. Eggs were coated with WPC at 8 % solution combined with the plasticizers glycerol (GLY), sorbitol (SOR), and propylene glycol (PRO). The eggs were stored at 20 °C. Weight loss, Haugh Unit (HU), albumen and yolk pH, and yolk index (YI) were evaluated weekly from day 0 to 42 days. After, electron microscopy of the eggshell structure was performed. The data was submitted to the analysis of variance and the effects of treatment, storage time, and the interaction between these factors were evaluated. There was significant interaction between factors (treatment and periods) for weight loss (p 0.0001), HU (p 0.0001), albumen (p 0.0001), and yolk pH (p 0.0001), and YI (p 0.0001). After 42 days, uncoated eggs showed greater weight loss (5.4 %), compared to WPC+GLY, (3.8 %), WPC+SOR (3.3 %) and WPC+PRO (3.9 %). Similar results were verified for HU at 42 days of storage. Uncoated eggs showed HU of 58.46 (B), while coated eggs showed higher values: WPC+GLY 66.58 (A), WPC+SOR 68.79 (A), and WPC + PRO 71.53 (A). The plasticizers GLY, SOR and PRO, associated with WPC, demonstrated effectiveness in maintaining the quality of eggs throughout the 42 days of storage. However, WPC+SOR showed superiority in preserving quality integrity of eggs. This result can be related to the chemical structure of SOR, making the combination more efficient for storage.

This study was conducted to evaluate the effects of the inclusion of low-purity crude glycerin (CG, 63.1% of glycerol) in the diets of feedlot-finished goats on the weights and yields of the non-carcass components, with are frequently used in regionals dishes (buchada and panelada). Forty castrated male goats with the initial mean body weight of 19.70 ± 2.30 kg were used. Experimental treatments consisted of inclusion of CG at levels of 0, 6, 12, and 18%, based on dry matter. The weights of the lungs, spleen, total organs (TWO), and TWO yield, as a function of the slaughter body weight, decreased linearly. It was shown that the rumen portion of the weight showed a quadratic effect, at the level of 10.71% of CG, with a minimum point of 0.37 kg. Similar behavior was observed for the reticulum, with a minimum point of 0.08 kg, at the level of 7.5% of CG. The weights and yields of the buchada and panelada were negatively influenced by the experimental diets. Low-purity crude glycerin may partially replace corn and be included in up to 6% of the dry matter of the finished goats' feedlot diet without adversely affecting weights and yields of organs, viscera, and regional dishes, such as buchada and panelada.(AU)

A nutrição animal tem evoluído ao longo dos anos, principalmente após a evolução nas técnicas de processamento dos alimentos. A partir do conhecimento da composição dos alimentos foi possível determinar o valor energético dos mesmos, que contribuiu para melhorias no sistema de produção, uma vez que as informações obtidas auxiliam nas formulações de rações, visando a produção animal e economia mais eficiente. Contudo, os grãos (milho e soja) tradicionalmente utilizados para compor as dietas sofrem com as variações de preço, por isso busca-se ampliar e diversificar através de pesquisas novas fontes energéticas que do ponto de vista nutricional possam contribuir positivamente para o desempenho dos animais e econômico com a redução dos custos com a alimentação. A glicerina e o glicerol, surge como um ingrediente alternativo por possuir alto valor energético, podendo ser utilizado como fonte de energia na alimentação de aves, sem prejudicar o desempenho e características de carcaça. No entanto, para que a sua utilização seja realizada, conhecer a sua composição físico-químicos se torna essencial, uma vez que este ingrediente se encontra disponível no mercado e não apresenta valores nutricionais padronizados, podendo influenciar nos resultados zootécnicos. Diante do exposto, objetivou-se com este trabalho dissertar através de revisão de literatura sobre o uso de coprodutos do biodiesel como fonte energética em dietas para aves sobre as características de desempenho e qualidade de carcaça.


Animal nutrition has evolved over the years, especially after the evolution of food processing techniques. With knowledge of the composition of the food it was possible to determine the energy value of the food, which contributed to improvements in the production system, since the information obtained helps feed formulations, aiming at more efficient animal production and economy. However, the grains (maize and soybean)traditionally used to compose diets suffer from price variations, so we have sought to expand and diversify through research new energy sources that from the nutritional point of view can contribute positively to the performance of animals and economically with the reduction of food costs. Glycerin and glycerol appear as an alternative ingredient because it has high energy value and can be used as a source of energy in poultry feed, without impairing performance and carcass characteristics. However, for its use to be carried out, knowing its physical-chemical composition becomes essential, since this ingredient is available on the market and does not present standardized nutritional values, which may influence the zootechnical results. Hence, the objective of this work is to present a literature review on the use of biodiesel co-products as an energy source in poultry diets about the performance and carcass quality characteristics.

O objetivo deste estudo foi avaliar o perfil de ácidos graxos (AG) do leite de vacas alimentadas com dietas à base de silagem de milho contendo 0% e 8,9% de glicerina bruta (GB) na matéria seca (MS). Foram utilizadas 18 vacas Holandês x Gir com 48±18 dias em lactação, produzindo 19,8±4,9kg/dia de leite. Utilizou-se delineamento de blocos ao acaso, com nove vacas/tratamento e duas medidas repetidas no tempo, com os resultados analisados por modelos mistos. A inclusão de GB na dieta não alterou a ingestão de MS, mas reduziu os consumos dos AG oleico, linoleico e α-linolênico. Os teores dos AG mirístico, palmítico, rumênico, vacênico, esteárico, oleico, linoleico, α-linolênico e CLA trans-10 cis-12 na gordura do leite foram semelhantes entre dietas. A inclusão de GB reduziu os teores dos AG elaídico e C18:1 trans-10 e aumentou os teores dos AG de cadeia ímpar linear e do ácido láurico. Não houve efeito da inclusão da GB sobre os índices de aterogenicidade e trombogenicidade da gordura do leite. Concluiu-se que a inclusão de glicerina bruta em dieta à base de silagem de milho não afetou a qualidade nutricional da gordura do leite de vacas Holandês x Gir.(AU)


The objective of this study was to evaluate the milk fatty acid (FA) composition of cows fed corn silage-based diets containing 0% and 8.9% of crude glycerin (GB) on a dry matter (DM) basis. Eighteen Holstein x Gyr cows with 48±18 days in milk and producing 19.8±4.9kg milk/day were used in the study. The experimental design was a randomized block with nine cows per treatment and two repeated measures. Results were analyzed using mixed models. The milk fat contents of myristic, palmitic, rumenic, vaccenic, estearic, oleic, linoleic, α-linolenic, and trans-10 cis-12 CLA were similar between diets. Dietary inclusion of GB decreased elaidic and trans-10 C18:1, and increased lauric acid and odd linear-chain FA contents in milk fat. Both atherogenicity and trombogenicity indices were unaffected by GB inclusion. It was concluded that GB inclusion in corn silage-based diets had no effect on the nutritional quality of milk fat from Holstein x Gyr dairy cows.(AU)

Concluiu-se que a inclusão de glicerina bruta em dieta à base de silagem de milho não afetou a qualidade nutricional da gordura do leite de vacas Holandês x Gir.(AU)


The objective of this study was to evaluate the milk fatty acid (FA) composition of cows fed corn silage-based diets containing 0% and 8.9% of crude glycerin (GB) on a dry matter (DM) basis. Eighteen Holstein x Gyr cows with 48±18 days in milk and producing 19.8±4.9kg milk/day were used in the study. The experimental design was a randomized block with nine cows per treatment and two repeated measures. Results were analyzed using mixed models. The milk fat contents of myristic, palmitic, rumenic, vaccenic, estearic, oleic, linoleic, α-linolenic, and trans-10 cis-12 CLA were similar between diets. Dietary inclusion of GB decreased elaidic and trans-10 C18:1, and increased lauric acid and odd linear-chain FA contents in milk fat. Both atherogenicity and trombogenicity indices were unaffected by GB inclusion. It was concluded that GB inclusion in corn silage-based diets had no effect on the nutritional quality of milk fat from Holstein x Gyr dairy cows.(AU)

Resumo

The morphology of the ruminal and omasal mucosa and hepatic parenchyma of 44 young bulls submitted to four types of concentrates were evaluated by varying the inclusion level of crude Glycerin - CG (0, 6, 12 and 18%). The variables evaluated were number of ruminal papillae per cm² of wall; area of the papillae and total absorption surface per cm² of wall; Mitotic index - MI, height and area of ruminal and omasal papillae; and hepatic morphology. The samples processed for inclusion in paraffin and stained with Hematoxylin-Eosin. The different levels of inclusion of CG in the diet did not interfere either in the number of papillae per cm2 of rumen wall (P= 0,70) or in the ruminal absorptive surface area (P= 0,89). Animals that received diets with levels 12 and 18% had higher MI of the ruminal epithelium (P< 0.01), but higher cell proliferation wasn't reflected in height gain and papillary area (P= 0,82). The omasal mucosa wasn't influenced by the inclusion of CG (MI e height gain and papillary area, P= 0,43, P= 0,56 e P= 0,9, respectively). The inclusion of GB in up to 18% of diet dry matter didn't alter the morphology of the rumen and omaso mucosa; and the integrity hepatic parenchyma.(AU)

The aim of this study was to evaluate the effect of crude glycerin supplementation on the chemical composition and fatty acid profile of the muscle of cattle grazing on Urochloa decumbens. Thirty-five crossbreed bulls, with an average initial body weight of 428.0 ± 32.11 kg, were supplemented with dry matter corresponding to 1% body weight for 74 days on pasture and distributed to five treatments (0, 3, 6, 9 and 12% low purity glycerin in the total dry matter of the supplement) in a completely randomised design. After 74 days, the animals were slaughtered, and the longissimus dorsi muscle was sampled and frozen for meat composition analysis. The addition of glycerin did not affect the mean moisture (73.6 ± 0.3%), protein (22.8 ± 0.4%), total lipid (1.1 ± 0.1%) and ash (1.0 ± 0.04%) contents of the animal muscle. However, there was an increasing linear effect (P < 0.05) on the contents of trans-vaccenic acid (increase of 175% in relation to the control) and conjugated linoleic acid (CLA) (25% increase in relation to the control). We observed an increasing linear pattern in monounsaturated fatty acid levels (39.8 to 44.5%) and a decreasing linear pattern in polyunsaturated fatty acid content (13.5 to 9.5%). Objetivou-se avaliar o efeito da inclusão de glicerina bruta no suplemento sobre a composição química e o perfil de ácidos graxos do músculo de bovinos terminados em pastagem de Urochloa decumbens. Para isso foram utilizados 35 touros azebuados, com peso inicial médio de 428,0 ± 32,11kg, suplementados com 1% do peso corporal, por 74 dias em pastagem e distribuídos em cinco tratamentos (0, 3, 6, 9 e 12% de glicerina de baixa pureza na matéria seca total do suplemento) em delineamento inteiramente casualizado. Após 74 dias os animais foram abatidos, o músculo Longissimus dorsi foi amostrado e congelado para análise de composição da carne. A adição de glicerina no suplemento não afetou (P>0,05) os teores médios de umidade (73,6%±0,3), proteína (22,8%±0,4), lipídeos totais (1,1%±0,1) e cinzas (1,0%±0,04) do músculo dos animais. Todavia, observou-se efeito linear crescente (P<0,05) para os teores de ácido trans-vacênico (acréscimo de 175% em relação ao controle) e ácido linoleico conjugado (CLA) (acréscimo de 25% em relação ao controle). Observou-se comportamento linear crescente para os teores de ácidos graxos monoinsaturados (39,8 para 44,5%) e linear decrescente para os teores de ácidos graxos poli-insaturados (13,5% para 9,5%). Artificial insemination with cooled semen is the most common practice in rabbit farms and any improvement on it helps to increase the efficiency and productivity of rabbit meat farms. Therefore, the aim of this study was to assess whether different cryoprotectant agents (CPA) as glycerol, N, N-Dimethylformamide (DMF) and N-Methyl-2-Pyrrolidone (NMP) can improve cooled rabbit sperm quality stored at 4ºC and 16ºC. Sperm samples were diluted with INRA 96® (Extender A), INRA 96® with 6% glycerol (Extender B) or 6% DMF (Extender C) or 6% NMP (Extender D) respectively and stored at 4ºC and 16ºC. Samples were then analysed at 4, 24, 48 and 72 hours after refrigeration by integrated sperm analysis system (ISAS®), eosin-nigrosin stain (vitality), hypo-osmotic swelling test (HOS test) and acrosome integrity test. Extender C showed higher percentage of motility, vitality and HOS test than extender B and D (p<0.05). Whereas sperm quality decreased over time (p<0.05), data showed that the addition of DMF kept the motility and sperm plasma membrane integrity after 24 hours of storage better than other diluents. Crude glycerine is a residue generated from the production of biodiesel. As there was an increase in the demand for biodiesel observed in recent years, there was also a concomitant increase in the production of glycerin, and this by-product is not being used by industries. Thus, new alternatives can be viable, such as the use in dairy cows’ food. The use of glycerin in the animal diet has shown to be a viable alternative to nutritional and economic aspects. Our aim in this review was to demonstrate the use of glycerin in dairy cow diets and the possible effects on its milk production. In several studies it is possible to observe that the crude glycerin can be included in the diet of ruminant animals without causing negative effects on their metabolism, ruminal fermentation and milk production. Studies on the deleterious effects of the inclusion of crude glycerin with high methanol content in the dairy cow diet should be performed.


A glicerina bruta é um resíduo oriundo da produção de biodiesel. Com o aumento da demanda por biodiesel nos últimos anos, houve também um aumento na produção de glicerina, um co-produto que não é utilizado pelas indústrias. Assim, novas alternativas podem ser viáveis, como o uso na alimentação de vacas leiteiras. O uso da glicerina na dieta dos animais demonstra ser uma alternativa viável no que diz respeito aos aspectos nutricionais e econômicos. Nosso objetivo nesta revisão foi abordar a utilização da glicerina em dietas de vacas leiteiras e seus possíveis efeitos sobre a produção de leite. Em vários estudos, é possível observar que a glicerina bruta pode ser incluída na dieta de ruminantes sem causar efeitos negativos sobre o metabolismo, fermentação ruminal e produção de leite. Estudos sobre os efeitos deletérios da inclusão de glicerina em bruto com alto teor de metanol na dieta de vacas leiteiras devem ser realizados.

Background: Alphitobius diaperinus, known as the lesser mealworm, is recognized as a problem for the poultry production sector. Several chemical methods have been employed in an attempt to control it, without successful results. Thus, alternative methods should be considered as an interesting approach to control this type of infestation. A recent study showed that glycerol monolaurate (GML) possesses insecticidal effect against bees. Therefore, the aim of this study was to evaluate, for the first time, the effect of GLM against the larval and adult forms of the lesser mealworm in vitro, and to verify whether GLM treated poultry litter used as soil fertilizer exerts any negative effect on its fauna.Materials, Methods & Results: In vitro tests were performed using three concentrations of GLM: 0.2, 0.4 and 1 mg mL-1, and the number of dead larvae and adults of A. diaperinus was counted on days 0, 2, 5 and 8 post-treatment. Poultry litter was pulverized with 1 mg mL-1 of GML, maintained under controlled conditions for 20 days and revolved every two days. Two ecotoxicological tests were performed using Enchytraeus crypticus. In the first test, GML was applied directly into the soil at doses of 0.5, 1 and 2 mg kg-1 of soil, while the second test used poultry litter as soil fertilizer at concentrations of 0, 2, 4 and 8 tons per hectare. Insecticidal action of GML against larvae and adults of A. diaperinus where only the 1 mg mL-1 concentration showed positive effect. GML caused 100% mortality of larvae two days after the beginning of treatment. Similarly, GML showed high efficacy to control adult forms of A. diaperinus, causing 83 and 90% of mortality on days 5 and 8 post-treatment, respectively.[...]
