Resumo

We describe here hematological and biochemical findings in a cow with metastatic cholangiocarcinoma. On physical examination, apathy with congested mucous membranes, tachycardia and enlarged abdomen; hypoperistalsis in rumen, abomasum and intestines. In hematological examination, icteric plasma with leukocytosis by neutrophilia with regenerative left shift and inversion of the proportion between lymphocytes and segmented neutrophils, and hypofibrinemia. In serum biochemistry: hyperproteinemia, hypoalbuminemia, hyperglobulinemia, low albumin/globulin ratio and high levels of aspartate aminotransferase, gamma-glutamyltransferase. The cow died and in necropsy was observed a severe and diffuse increase in volume predominantly on the hepatic right lobe and moderate increase of volume in mesenteric lymph nodes. Which was diagnosed microscopically as a cholangiocarcinoma, characterized by arrangements in irregular and coalescent ducts. In conclusion, cholangiocarcinoma may cause important hematological and biochemical changes in cattle.(AU)

Resumo

Background: Babesiosis is endemic in Pakistan and is one of the most important bovine diseases that causes huge economic losses and high mortality in young animals. A hematobiochemical study was conducted to unveil the difference between diseased and healthy animals in selected districts i.e., Faisalabad (31° 25' 7.3740'' N and 73° 4' 44.7924'' E), Toba Tek Singh (30° 58' 9.7392'' N and 72° 27' 40.7484'' E) and Jhang (31° 16' 40.9656'' N and 72° 18' 42.3360'' E) of Punjab, Pakistan. Materials, Methods & Results: A total of 518 (Cattle = 360, Buffalo = 158) blood samples were collected. The samples were analyzed by polymerase chain reaction (PCR) targeting apocytochrome b-gene (Babesia bovis-gene) (CYTb) followed by haemato-biochemical analysis. Chi-square test for univariate analysis was used to analyze the data. In summer the PCR-based prevalence was 29.4 (53/180) and 24.05% (19/79) in cows and buffaloes, respectively. On the other hand, in winter results showed that 12.7 (23/180), 13.92 % (11/79) samples positive for Babesia genus from cows and buffaloes, respectively. The positive samples were further investigated for hematological and biochemical analysis. The results revealed that, the mean value of hematological parameters like RBCs, Hb, PCV, MCV and MCHC was significantly (P < 0.05) decreased in infected animals (cows and buffaloes) as compared to the non-infected ones. While the biochemical parameters like Alanine aminotransferase (ALT), Aspartate aminotransferase (AST), cholesterol and Lactate dehydrogenase were significantly (P < 0.05) increased in infected animals as compared to healthy animals. This study is the first molecular and hematobiochemical evidence of Babesia bovis in dairy herds of Punjab province, Pakistan. Discussion: Bovine babesiosis is one of the important tick-borne diseases (TBD) affecting dairy industry. In bovines, among 3 Babesia species that cause the disease B. bovis is more pathogenic with high mortality and morbidity. Pakistan is situated in tropical and sub-tropical region where the humidity is high in some part of countries. This high humidity mostly favors the reproduction of the ticks thus higher prevalence of TBDs in this region. Initially the babesiosis was diagnosed by light microscopy using thin blood smear stained with Giemsa stain. Many studies verified that PCR is a more specific and sensitive tool than conventional techniques for the detection of carrier / asymptomatic ruminants. The haemato-biochemical profile is another valuable footprint to track the disease. Keeping in view the above-mentioned fact the present project has been planned to evaluate the haemato-biochemical alteration between health and Babesia infected cattle along with the molecular detection of Babesia species involved in bovine babesiosis. The mean values of haematobiochemical parameters in clinically ill and healthy animals revealed that the mean values of hematological parameters like RBCs, Hb, PCV, and HCT were significantly decreased in diseased animals as compared to the healthy ones. All these might be due the fact that the parasite is intra-erythrocytic in nature and destruction of red blood cells results in significant (P < 0.05) decrease level of all the hematological parameters. The mean value of ALT in babesiosis infected cattle was significantly higher as compared to healthy cattle. The mean values of AST and LDH in babesiosis infected cows was significantly higher as compared to that in healthy cows. The elevation in liver enzymes in babesiosis may be due to the hepatic damage and lesions induced by the parasite during its multiplication in the blood followed by disturbed liver function. These enzymes are present in high concentrations in the muscles and liver. High level of these enzymes in the blood is indicator of organ necrosis or damage.

Background: Hormonal and physiological changes during pregnancy period have significant effects on animals' metabolisms. The purpose of the present study is to evaluate the variations of aspartate aminotransferase (AST) and gamma-glutamyl transferase (GGT) levels according to different gestation trimesters of the pregnant mares to assess changes of embryonic losses and abortions. Materials, Methods & Results: Blood samples from 49 thoroughbred mares were analyzed during this study. Age ratio of the mares is 5-18 years and their body condition score (BCS; score 1 to 9) varies between 5 and 6. A total of 28 mares had given birth to healthy foals at the end of a healthy gestation period (Group C), whereas 11 mares had embryonic losses (Group E) and 10 mares had late period abortions (Group A). Following the confirmation of pregnancy, one blood sample per gestation trimester were taken (14-16 days of pregnancy for 1st trimester; 180 days for 2nd trimester, 270 days for 3rd trimester). Early embryonic losses (loss of a 16-25 days embryo) were observed in 6 of 11 mares in Group E and the 5 of 11 mares had late period embryonic losses (loss of a 35-40 days embryo). In Group A, 6 mares had the abortion within 7th month and the remaining 4 mares had the abortion within the 8th month of pregnancy. Repeated Measures ANOVA, Kruskal-Wallis test and Mann-Whitney test were performed for statistical analysis, the mean AST level in Group C (n = 28) was higher during the 1st trimester in comparison to 2 following trimesters (respectively: P = 0.011 ; P = 0.01). Besides, no statistical difference was observed between 2nd and 3rd trimester regarding AST activation (P > 0.05). The mean GGT level in Group C was significantly decreased lower in the 3rd trimester compared to 1st and 2nd trimester (respectively: P = 0.007 ; P = 0.009). No statistical difference was observed between 2nd and 3rd trimester regarding GGT activation (P > 0.05). Among all groups (C, E, A) no significant difference was observed on AST levels (P > 0.05), nonetheless GGT levels had a significant rise (P = 0.039) in Group A in comparison to the 1st trimester levels of Group C. In Group A, there was a statistical decrease of AST during the 2nd trimester in comparison to the 1st trimester (P = 0.001), accompanied by a decrease of GGT activation during the 2nd trimester compared to the 1st trimester (P = 0.009). Discussion: The aminotransferases are catalisors that play an important role on the metabolism of amino acids and carbohydrates. Although serum AST levels were within the reference ranges during 3 trimesters in this study, serum AST levels were determined to be decreased in the 2nd and 3rd trimesters in healthy pregnant mares compared to the 1st trimester. It is thought that these results can be obtained due to the increase in metabolic needs during pregnancy. In this study, serum GGT levels remained within physiological limits with a tendency to decrease as the pregnancy advances, in accordance with previous study results. Serum GGT levels that are used as an indicator to liver damage may vary during the pregnancy as a result of increased metabolic load. It is thought that the increase of serum GGT levels in Group A might be related to the fetal (chromosome-related) issues that may result to pregnancy losses. As a result, it is considered that serum AST and GGT levels in mares might be valuable parameters that predict embryonic loss or abortus cases.

ABSTRACT Purpose: To investigate experimentally the effects of Tropifexor, a farnesoid X receptor agonist, on liver injury in rats with obstructive jaundice. Methods: Forty healthy Wistar albino female rats were divided randomly in selected groups. These groups were the sham group, control group, vehicle solution group, Ursodeoxycholic acid group and Tropifexor group. Experimental obstructive jaundice was created in all groups, except the sham one. In the blood samples obtained, aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), total bilirubin and direct bilirubin levels were established and recorded. Additionally, liver malondialdehyde, myeloperoxidase and catalase enzyme activity in the tissue samples were studied. Histopathological analysis was also performed. Results: No statistical difference was found between the control group and the Tropifexor group when AST, ALT and ALP values were compared. However, it was found that the Tropifexor group had statistically significant decreases in the values of GGT, total bilirubin and direct bilirubin (p < 0.05). Additionally, Tropifexor decreased the median values of malondialdehyde and myeloperoxidase, but this difference was not statistically significant compared to the control group. Finally, the Tropifexor group was statistically significant in recurring histopathological liver damage indicators (p < 0.05). Conclusions: Tropifexor reduced liver damage due to obstructive jaundice.

ABSTRACT Purpose: To delve into the influence of paeoniflorin (PA) on abating primary biliary cholangitis (PBC)-induced liver fibrosis and its causative role. Methods: Our team allocated the mice to control group, PA group, PBC group and PBC+PA group. We recorded the weight change of mice in each group. We used Masson staining for determining liver fibrosis, immunofluorescence staining for measuring tumor necrosis factor-α (TNF-α) expression, quantitative real-time polymerase chain reaction (qRT-PCR) for assaying related gene expression, as well as Western blot for testing related protein expression. Results: The weight of PBC model mice declined. Twenty-four weeks after modeling, the positive rate of anti-mitochondrial antibody-M2 (AMA-M2) in PBC mice reached 100%. Alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), hydroxyproline (HYP), laminin (LN), procollagen type III (PC III), and malondialdehyde (MDA) contents saliently waxed (p<0.01). Meanwhile, superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) activity patently waned (p<0.01). Liver fibrosis levels were flagrantly higher (p<0.01), and TNF-α, NOD-like receptor protein 3 (NLRP3), caspase-1, interleukin-18 (IL-18), and interleukin-1β (IL-1β) protein or gene expression were manifestly up-regulated (p<0.01). PA could restore the weight of PBC mice, strikingly restrain the positive expression of AMA-M2, and down-regulate serum ALP, ALT, AST, HYP, LN, PC III, MDA in PBC mice (p<0.01). PA could also significantly up-regulate SOD and GSH-px levels (p<0.01), down-regulate IL-1β, IL-18, caspase-1, NLRP3, and TNF-α protein or gene expression in PBC mice (p<0.01) and inhibit liver fibrosis levels (p<0.01). Conclusions: PA can reduce PBC-induced liver fibrosis in mice and may function by curbing the formation of NLRP3.

Durante o periparto, as vacas leiteiras são submetidas a uma grande demanda de energia, ao mesmo tempo em que reduzem sua ingestão de matéria seca. O balanço energético negativo, resultante dessa equação, acarreta severos transtornos metabólicos, à produção e, principalmente, à reprodução. O objetivo do presente estudo foi avaliar o efeito da colina protegida sobre os parâmetros metabólicos, o intervalo entre parto e concepção e a produção de leite em vacas no período de transição. Cinquenta e quatro vacas leiteiras foram divididas em três grupos: controle, suplementação com colina por 10 dias pré-parto (T10) e suplementação com colina por 20 dias pré-parto (T20). Após o parto, foram mensurados os teores de frutosamina, colesterol, ácidos graxos não esterificados (AGNE), beta-hidroxibutirato (BHB), aspartato aminotransferase (AST), gamaglutamiltransferase (GGT) e total de oxidantes (TOS), nos dias 10, 20 e 30. Ainda foram avaliadas produção de leite e intervalo entre parto e concepção. Não houve efeito da suplementação com colina sobre os parâmetros sanguíneos e a produção. O intervalo entre parto e concepção foi menor no grupo T20. A colina suplementada por 20 dias durante o pré-parto melhorou a performance reprodutiva de vacas leiteiras(AU)


During the periparturient dairy cows undergo a large energy demand, at the same time reducing their intake of dry matter. The negative energy balance resulting from this equation leads to severe metabolic disorders in production, and mainly in reproduction. The aim of this study was to evaluate the effect of protected choline on metabolic parameters, reproductive performance, and milk production in cows during the transition period. Fifty-four dairy cows were divided into three groups: control, supplementation with choline for 10 days prepartum (T10) and supplementation with choline for 20 days prepartum (T20). After delivery we measured fructosamine levels, cholesterol, non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), aspartate aminotransferase (AST), gamma glutamyltransferase (GGT), and total oxidant (TOS) on days 10, 20 and 30. We also evaluated milk production and interval between calving and conception. There was no effect of supplementation with choline on blood and production parameters. The interval between calving and conception was lower in the T20 group. Choline supplemented by 20 during the antepartum improved reproductive performance of dairy cows, although it did not change the metabolic profile.(AU)

The current study was intended to investigate the responses of freshwater fish Carassius gibelio as an in vivo model by measuring the activity of the enzyme of the liver. Fish exposed to sub-lethal toxicities due to zinc metal and crude oil. Fish were treated to 0.6 and 1.2 mg/l for zinc ions as well as 500 and 2500 mg/l for crude oil during 96h, and the alterations in serum enzyme activities were determined aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Zinc toxicity elevated activity levelin hepatic enzymes in a significant way (P < 0.05). The present study appeared the slightly augmented levels of AST in the serum of C. gibelio when exposed to sub-lethal Zinc concentration while the serum ALT activity increased in response to the high level of Zn exposure when compared to control after (96 h). In addition, acute concentrations of crude oil after 96 h exposure can cause harmful effects on Carassius gibelio including the induction of (AST). However, not effecting found (ALT). We are concluded that changes in the activity of the enzyme can be considered as a pointer of the pollution impact in fish. (AU)

Background: Bovine mastitis causes major economic losses for milk producers by reducing the quantity and the quality of the milk or even leading to the complete loss of the mammary gland secretory capacity. During the transition period, dairy cows are susceptible to infectious diseases; therefore, markers that allow early identification of cows in higher risk of developing diseases are especially useful at this time. Therefore, the aim of this study was to evaluate serum markers in the pre and postpartum of multiparous dairy cows with clinical mastitis and with health condition in the postpartum period in a semi-extensive management system.Materials, Methods & Results: Thirty-Six Holstein cows were monitored daily during milking until 59 days postpartum and were categorized according to the pre-milking strip cup test into clinical mastitis (mastitis group (MG)) and absence of symptoms (control group (CG)) that were negative to the test, representing the health cows. All cows were reared as one group and maintained in a semi-extensive pasture-based system. Blood samples were collected weekly after morning milking via venipuncture of the coccinea vein into tubes without anticoagulant and grouped for prepartum (-21 to 0 days from calving), early postpartum (0 to 30 days from calving), and late postpartum (30 to 59 days from calving) periods. Milk production was recorded daily. The serum markers albumin, aspartate aminotransferase (AST), phosphorus, gammaglutamyltransferase (GGT) and non-esterified fatty acids (NEFA) were measured. Statistical analyses were performed using SAS®. The cases of clinical mastitis occurred on average at 37.2 ± 4.9 days postpartum. Health cows (CG) had higher milk production compared to the mastitis group (MG) only in the late postpartum period (P 0.05).[...]

Background: One of the valuable tests for diagnosis of cardiovascular and liver diseases is measuring of AST activity. One of the main enzymes of transaminases group is aspartate aminotransferase. Previous Studies have shown that some alteration may occur in mitochondria function as the result of different disease or taking different medication; these changes in mitochondrial and cytosolic AST isozymes can be the sign of disorders. According to the role of steroid hormone in induction of its effects on protein synthesis genes, this study is conducted to shed some light on mechanisms and the interference of steroid hormones and antibiotics.Materials, Methods & Results: In this study, male Wistar rats were injected intramuscularly with Testosterone, progesterone and estradiol; while tetracycline and streptomycin injections were intraperitoneal. Testosterone, progesterone and estradiol injections were carried out in a short-term (15 days) and long-term (45 days) periods. Steroid hormones were dissolved in sesame in a way that by each injection, 0.2 mL sesame oil (containing specific amount of hormone) was injected to the rat. Control group was kept in the same condition except that their sesame oil injection contained no hormone. Tetracycline and Streptomycin injection was carried out for 5 days at 7 am and pm, at 50 mg/kg dosage intraperitoneally. In short- and long-term periods, rats were divided into four groups of 6-member. The concentrations were the same in the periods and 0.2 mL sesame oil was injected intramuscularly. 1 mg testosterone, 12 mg progesterone and 0.2 mg estradiol were intramuscularly injected to rats in group 2, 3 and 4, respectively [10]. Resumo

Background: One of the valuable tests for diagnosis of cardiovascular and liver diseases is measuring of AST activity. One of the main enzymes of transaminases group is aspartate aminotransferase. Previous Studies have shown that some alteration may occur in mitochondria function as the result of different disease or taking different medication; these changes in mitochondrial and cytosolic AST isozymes can be the sign of disorders. According to the role of steroid hormone in induction of its effects on protein synthesis genes, this study is conducted to shed some light on mechanisms and the interference of steroid hormones and antibiotics.Materials, Methods & Results: In this study, male Wistar rats were injected intramuscularly with Testosterone, progesterone and estradiol; while tetracycline and streptomycin injections were intraperitoneal. Testosterone, progesterone and estradiol injections were carried out in a short-term (15 days) and long-term (45 days) periods. Steroid hormones were dissolved in sesame in a way that by each injection, 0.2 mL sesame oil (containing specific amount of hormone) was injected to the rat. Control group was kept in the same condition except that their sesame oil injection contained no hormone. Tetracycline and Streptomycin injection was carried out for 5 days at 7 am and pm, at 50 mg/kg dosage intraperitoneally. In short- and long-term periods, rats were divided into four groups of 6-member. Background: Bovine mastitis causes major economic losses for milk producers by reducing the quantity and the quality of the milk or even leading to the complete loss of the mammary gland secretory capacity. During the transition period, dairy cows are susceptible to infectious diseases; therefore, markers that allow early identification of cows in higher risk of developing diseases are especially useful at this time. Therefore, the aim of this study was to evaluate serum markers in the pre and postpartum of multiparous dairy cows with clinical mastitis and with health condition in the postpartum period in a semi-extensive management system.Materials, Methods & Results: Thirty-Six Holstein cows were monitored daily during milking until 59 days postpartum and were categorized according to the pre-milking strip cup test into clinical mastitis (mastitis group (MG)) and absence of symptoms (control group (CG)) that were negative to the test, representing the health cows. All cows were reared as one group and maintained in a semi-extensive pasture-based system. Blood samples were collected weekly after morning milking via venipuncture of the coccinea vein into tubes without anticoagulant and grouped for prepartum (-21 to 0 days from calving), early postpartum (0 to 30 days from calving), and late postpartum (30 to 59 days from calving) periods. Milk production was recorded daily. The serum markers albumin, aspartate aminotransferase (AST), phosphorus, gammaglutamyltransferase (GGT) and non-esterified fatty acids (NEFA) were measured. Statistical analyses were performed using SAS®. The cases of clinical mastitis occurred on average at 37.2 ± 4.9 days postpartum. Health cows (CG) had higher milk production compared to the mastitis group (MG) only in the late postpartum period (P < 0.05). Background: The malignant neoplasms, in general, constitute one of the main causes of death in dogs and cats. The multiple myeloma is a neoplasm characterized by the monoclonal proliferation of plasma cells in the bone marrow, causing several systemic symptoms and it is rare in pets, representing less than 1% of the malignant neoplasms in these animals. It usually occurs in older dogs. Thus, the purpose of this report was to describe a case of multiple myeloma in a 10-year-old poodle dog, occurred in the Veterinary Hospital Unit of the State University of Ceará. Case: A 10-year-old Poodle bitch was admitted in the Veterinary Hospital Unit of the State University of Ceará presenting lump erythematosus, alopecic, soft and ulcers in the region of the digits of the left front leg, pain and lameness. The blood count evidenced normocytic and normochromic anemia, presence of rouleaux, lymphopenia and monocytopenia, and the biochemical exams have demonstrated an elevated value of alanine aminotransferase (ALT). The cytology of the nodule indicated probable diagnosis of plasma cell neoplasm and radiographic examination showed the presence of bone lysis, and therefore the patient underwent amputation of the affected limb surgery. A biopsy of the nodule indicated neoplastic proliferation, with an arrange compatible to plasmacytoma of cleaved type. A follow-up consultation was realized 30 days after surgery, and 2 new nodules appeared in the superior lip region and close to the surgery site. A new blood count indicated anemia with presence of rouleaux, thrombocytopenia, leucopenia, lymphopenia and monocytopenia, and the biochemical results showed a discrete diminution of aspartate aminotransferase(AST) and a considerable increase on the alkaline phosphatase levels. The cytology of the new nodules indicated plasmacytoma, and was realized a myelogram, that showed dysplastic marrow plasma cells, with more than 20% of plasma cells...

There are several causes of carcass condemnation in poultry processing plants, including dorsal cranial myopathy (DCM), in which the anterior latissimus dorsi (ALD) muscle is affected. DCM etiology has not been elucidated yet, but this lesion impairs the visual quality of carcasses and causes economic losses due to downgrading and condemnation. The effects of this lesion on the systemic health of broilers are still unknown. In this context, the objective of this study was to evaluate muscle injury and systemic health indicators in broilers presenting or not DCM. The following parameters were evaluated: complete blood count (CBC), total protein, albumin, globulin, cholesterol, lactate, and glucose serum levels, aspartate aminotransferase (AST) and creatine kinase (CK) serum activities, and breast muscle pH. Blood samples were collected from 800 42-day-old broilers before feed withdrawal and transportation. In the processing plant, 28 carcasses presented DCM during inspection, and 28 carcasses with no DCM were used as controls. Blood biochemical parameters were not significantly different between broilers with DCM and the controls, except for AST and CK, which activities were higher in the DCM group than in the control group, suggesting that the DCM does not affect the systemic health of the broilers.(AU)

Purpose: To investigate the effects of lycopene and resveratrol pretreatment on hepatic hyperplasia in partially hepatectomized rats. Methods: The lycopene group and the resveratrol group received 40 mg/kg/day of lycopene or resveratrol, respectively (dissolved in olive oil or in saline solution, respectively) and administered via a gastric tube for 30 days. The partially hepatectomzed (PH) control groups received saline or olive oil via a gastric tube for 30 days, respectively, and the normal control group received no treatment. Liver tissue and intracardiac blood samples were obtained 24, 36 or 48 h after PH. Results: No areas of fibrosis were detected. No significant changes in mitotic index, in the number of apoptosis events or in aspartate aminotransferase and alanine aminotransferase levels were observed. Conclusions: Lycopene and resveratrol pretreatment did not interfere on hepatic hyperplasia in partially hepatectomized rats.(AU)

O uso de bST é uma ferramenta que propicia um aumento considerável na produção diária de leite. Entretanto, esta suplementação pode ter influencia na função hepática e no lipidograma. Sendo assim, o objetivo deste estudo foi avaliar o perfil lipídico e hepático e também a produção láctea de vacas suplementadas com este hormônio. Para isso, foram utilizadas 10 vacas em produção, 90 dias pós-parto, com média de 30 litros/dia. Realizaram-se cinco aplicações com intervalo de 14 dias entre elas. As coletas de sangue para análise de colesterol, triglicerídeos, AST, GGT e albumina foram feitas nos dias zero, 20, 40 e 60 do experimento. Já a produção de leite foi avaliada em três momentos dentro do período de 70 dias. Não houve diferença entre os grupos em nenhum dos parâmetros avaliados tanto para perfil lipídico e função hepática, entretanto, promoveu uma maior produção de leite no período estudado. Sendo assim, conclui-se que a suplementação de bST não exerce influência negativa sobre o lipidograma de vacas leiteiras em produção, bem como não causa dano hepático durante a suplementação deste hormônio por um período de 70 dias e, também possibilita uma maior produção de leite neste período.(AU)

There was no difference between groups in any of the parameters evaluated for both serum lipid profile and liver function, but milk production was increased in control group. Therefore, it is concluded that supplementation of bST does not result negative influence on the serum lipid profile of dairy cows in production, and does not cause liver damage in supplementation for a period of 70 days, and increases milk production in this period.(AU)

Sin embargo, esta suplementación puede tener influencia en la función hepática y perfil lipídico. Por lo tanto, el objetivo de este estudio fue evaluar el perfil lipídico y hepatico y también la producción de leche de vacas alimentadas con esta hormona. Para este fin, se utilizaron 10 vacas en producción, 90 días después del parto, con un promedio de 30 litros/día. Se realizó cinco veces con un intervalo de 14 días entre ellos.
