ABSTRACT: This study presented relevant aspects about the Amazonian environment and how it impacts the thermal comfort of domestic buffaloes (Bubalus bubalis) raised in the Eastern Amazon. Furthermore, strategies for monitoring and mitigating animal heat stress are presented, based on research results with the species. Although domestic buffaloes are considered adaptable animals, exposure to intense solar radiation causes thermal discomfort. This condition is expressed in biophysical indicators, in metabolic, endocrine, behavioral responses, and in body thermographic patterns. Therefore, the biometeorological monitoring of production is crucial to support decision-making regarding environmental management strategies, genetic selection of thermotolerant individuals, and increase in animal welfare. Lastly, the use of silvopastoral systems can help to provide higher thermal comfort, which is a condition that directly impacts the productivity of milk and meat buffaloes when they are raised in tropical regions, such as in the Eastern Amazon.

Water buffalo (Bubalus bubalis) production in Mexico has generated particular interest in understanding the bases of the development of this species. As a result, alliances have emerged between scientific, governmental, industrial, and productive sectors and other organisms involved with this species nationally and internationally. We must recognize that thanks to specific anatomical characteristics, water buffaloes present a low incidence of complications during calving, though they are not exempt from obstetric and perinatological problems that can end in morbidity and mortality for mother and/or offspring, with losses for both branches of dual-purpose production systems: calves-milk and meat, and draft animals. This article analyzes scientific advances related to water buffalo production in Mexico, including productivity, thermoregulation, behavior, and physiology, and key subthemes like obstetric and perinatological handling, imprinting, milking, and dual-purpose production systems. It highlights features inherent to milk production and the neurophysiological and metabolic processes involved in obtaining meat and milk. Finally, it presents a brief overview of current legislation and summarizes key areas of opportunity.(AU)

Leptospirosis is a zoonosis that can infect buffaloes and other mammalians, and it's caused by Gram-negative bacteria of the genus Leptospira spp. The infection occurs through contact with urine or a contaminated environment with the mucous membranes of the animals, and the disease has two phases: leptospiremia and leptospiruria. The objective of this study was to determine the frequency of leptospirosis in buffaloes in the state of Paraná, Brazil, as well as the main serovars present in this area. A total of 276 blood samples were collected from buffaloes from 18 properties located in different cities and mesoregions of Paraná state. A microscopic agglutination test (MAT) was performed to detect the presence of anti-Leptospira spp. antibodies and to identify its serovars. The prevalence was 77.17% (213/276) of buffaloes seropositive for at least one serovar. The most frequent serovar was Icterohaemorrhagiae, followed by the Pomona serovar and serovar Hardjo. This study seems to be the first to determine the frequency of buffaloes with leptospirosis in the state of Paraná. Prevention and control measures are important to control the disease in herds.

This study aimed to evaluate phosphorus (P) concentration in serum and bone, the percentage of ash and the specific bone density of buffaloes on Ilha de Marajó before and after mineral supplementation. For this study, 14 crossbred buffaloes of Murrah and Mediterranean descent aged between 18 and 36 months were used. The average values of P before supplementation in serum and bone, the percentage of bone ash and the specific bone density were 5.68mg/dL±1.18, 16.53%±0.53, 59.95%±1.96 and 1.52g/cm3±0.32, respectively, which demonstrated P deficiency in animals raised on Ilha de Marajó. After supplementation with P for a period of seven months, the values were 6.61mg/dL±0.87, 16.90%±0.56 and 60.30%±0.95 and 1.71g/cm3±0.21, respectively. These results showed a significant increase in P concentration in blood serum, specific bone density and percentage of P in ash (P<0.05), but there was no significant increase in the percentage of ash. The average increase in P in the serum and ash did not reach normal levels in all animals; however, 28.6% of the animals had normal values of P in serum and 50% in the ash, and 64.3% had normal specific bone density values. The nonre-establishment, in some of the animals, of the variables of P serum and bone after supplementation for seven months may have occurred as a result of the low intake of the mineral mixture and by the low concentration of P in the Brachiaria brizantha cv. Marandu used for feeding animals during the experiment.

Buffalo milk production is an activity that has grown in recent years, mainly due to the physicochemical characteristics found in this type of milk, which is the second most produced in the world. The objective of this study was to implement a milking system and monitor the identity and quality standards of buffalo milk produced at the Experimental Agronomic Station of the Federal University of Rio Grande do Sul. Samples were collected fortnightly between June and November 2021, totaling ten collections. The means obtained at the end of the monitoring were 4.84 g/100 g for fat, 4.64 g/100 g for protein, 5.06 g/100 g for lactose, 15.26 g/100 g for total solids, 10.42 g/100g for non-fat solids, 0.18 g lactic acid/100 mL of milk for acidity, 1.037 g/cm3 for density, -0.533°C for cryoscopic index, 3.5 x 105 cells/mL for somatic cell count, and 8.0 x103 CFU/ml for standard plate count. Residues of antibiotics and antiparasitics were not found. An increase in the concentration and frequency of some fatty acids was observed, after an increment in the nutritional management of the animals, in addition to predominance of palmitic acid (C16:0) and oleic acid (C18:1- cis (n9)). The results indicate a significant improvement in the quality of the milk in the assessment period, due to the corrective actions that were established, and the standard plate count showed good agricultural practices as to milk sourcing and the hygiene of the materials, being below the limits required by NI 76. The buffalo milk produced by the herd at the Experimental Station showed physicochemical and microbiological results in accordance with the legislation, being a raw material to be explored in the prospection of derivative products.

Transport is considered a stressful period for livestock that can cause economic and biological losses if inadequate handling procedures exist. Several manuals and guides include recommendations for common domestic species based on scientific evidence. Still, the legislative freedom of each country means that they are not mandatory worldwide, nor do they cover all species, such as the water buffalo (Bubalus bubalis). This review aims to analyze the legal strictures currently applicable to the transport of this species. Critical aspects of the legislation that address the pre-transport, transportation, and post-transport stages are discussed, focusing on Asian and Latin American countries and the European Union. Areas of opportunity to improve the norms and laws that impact public policies, civil aspects, and water buffalo welfare in production systems are elucidated.

Domestic buffalo production plays an economically important role in the Brazilian Amazon, but they are susceptible to many diseases favored by the tropical climate and annually flooded habitats, including ocular diseases. In this context, it is important to select genotypes that maximize innate ocular immunity in Amazonian herds. We aimed to characterise, for the first time, gene expression profiles of the innate immune system in the conjunctival membrane of buffalo. Ocular conjunctival tissue samples were collected from 60 clinically healthy slaughtered animals in the northern Brazilian state of Amapá. The samples were histologically processed for classification into three groups according to the quantitative degree of lymphoid tissue associated with the conjunctiva (discrete, G1; slight, G2; and moderate, G3 presence of lymphoid tissue). RT-PCR was used to quantify gene expression of inflammatory cytokine (IL6, IL10, TNFA, IFNG), Toll-like receptor 4 (TLR4), and Defensin beta 110 (DEFB110), relative to the endogenous GAPDH gene. G1 animals presented low expression for IL6, IL10, TNFA, and DEFB110, while G2 exhibited high expression for IL6, IL10, IFNG, and TLR4. All G3 animals showed high expression for all tested genes. These results suggest a greater resistance to pathogenic microorganisms of buffalos in the G3 group, and the proportion of lymphoid tissue associated with the conjunctiva may be related to the immune resistance of individuals.(AU)

Bovine viral diarrhea virus (BVDV) is a highly infectious pathogen that affects bovines worldwide leading to great economic impact. Although Brazil has the largest commercial cattle population throughout the world and an increasing buffalo breeding industry, the country has no control or eradication program for BVDV. In this perspective, the aim of this study was to evaluate the occurrence of BVDV in cattle and buffaloes from two Brazilian states. Four different ELISA tests were performed and confirmed by virus neutralization testing (VNT). The presence of BVDV antibodies in the serum or plasma from 77 cattle from six herds (ELISA-1 and ELISA-4) and from 89 buffaloes from three herds (ELISA-1 through ELISA-4) was detected. Extraction of viral RNA was performed from the serum or plasma samples for the detection of BVDV by RT-PCR analysis. Amplified nucleotide sequences were used to construct a phylogenetic tree. In cattle, ELISA-1 detected 49.4% of seropositive animals, while ELISA-4 detected 37.7%. In buffaloes, ELISA-1 failed to detect any seropositive animals, while ELISA-2 and ELISA-3 detected 20.2% of seropositive animals, and ELISA-4 detected 21.3%. Eight of the nine herds tested had seropositive animals. The rate of PCR positive animals was 6.5% in cattle and 9% in buffaloes. Subtype 1d was found in cattle, and subtypes 1d and 1f were found in buffaloes. This is the first-time subtype 1f has been reported in Brazil. The absence of a control and eradication program seems to be favoring the spread of BVDV in the Brazilian herds. In addition, the improvement of diagnostic strategies for BVDV in buffaloes are required.

It is well known that transporting livestock is stressful for the animals and a primary cause of skin lesions. The effects of transport on the water buffalo (Bubalus bubalis) have not been studied extensively despite reports of high incidences of trauma during and after these mobilizations. The present review article analyzes the frequency of skin injuries suffered by water buffaloes during transport and several key contributing factors. It also discusses the consequences of injuries on the quality of carcasses. Important aspects identified include inappropriate vehicle design that does not comply with the dimensions recommended by international institutions (e.g., height, type of flooring, characteristics of the ramp), handling methods, and the ability of stock people to manage water buffaloes, all of which impact the incidence of injuries. Our analysis of these elements will contribute to identifying critical control points and areas of opportunity while also suggesting strategies to ensure the welfare of these animals during transport and the quality of water buffalo products and by-products.

Background: Buffalo breeding is common in many countries. Buffalo's milk is used in the production of mozzarella, yoghurt, ice cream, and various dairy desserts; meat is preferred in sausage production. The female buffaloes are bred to benefit from their milk and to obtain offspring. These animals, which are not suitable for feeding in barns, generally live in pastures, especially on wet land, and are very difficult to follow. Therefore, diseases occur randomly in slaughterhouses. Studies on genital system problems are very limited. Water buffaloes (Bubalus bubalis) are animals with economic value as many buffalo products are provided. In this respect, inspection and control of female genital disorders is fundamental to ensure good reproductive performance of female buffaloes. The aim of the study was to investigate pathomorphological lesions occurring in the ovaries of water buffaloes which were sent to slaughterhouses. Materials, Methods & Results: A total of 198 ovaries of water buffaloes were collected from various slaughterhouses located in Adapazari, Istanbul, Kocaeli, Samsun and Trabzon cities of Turkey. After macroscopic examination; tissue samples were fixed in 10 % buffered formalin, processed routinely and were stained with hematoxylin-eosin (HE). As a histochemical staining, Masson's trichrome staining was applied to characterize the lesions. Immunohistochemistry (IHC) was performed on 10 % neutral formalin fixed, paraffin-embedded, 4-6-µm-thick sections from ovaries using progesterone receptor (PR) with streptavidin-biotin complex peroxidase (StrepABC-P) method. Histopathologically, follicular cysts (n:147) and luteal cysts (n:22) were seen. While the cyst lumens were sometimes surrounded by granulosa and/or luteal cells, most of them were limited by the connective tissue capsule structure. This capsule structure was shown in blue by Masson's trichrome staining. Hemorrhage was observed in some cystic corpus luteum, diagnosed as corpus hemorrhagicum. In addition, cysts giving papillary extension into the lumen and inflammation of some ovaries were observed. Immunohistochemically, the staining with PR antibody in ovarian cysts showed no immunolabelling around the follicular cyst, while the nuclei of some of the luteal cells forming the luteal cyst had strongly nuclear positivity and slightly cytoplasmic positivity. In the biochemical examination of the fluids obtained from cystic ovaries (n: 37), the average of estradiol was 2.84 ng/mL (min: 0.01 ng/mL, max: 4.30 ng/mL) and progesterone average is 49.09 ng/mL (min: 1.88 ng/mL, max: 254.2 ng/mL). Discussion: Ovarian cysts in buffaloes seem to be among the serious fertility problems as in cattle. Although the exact cause of ovarian cyts has not been determined yet, it is known that the hypothalamic-pituitary-gonadal (HPG) axis is disrupted in cyst formation in general. In this study, the mean estradiol value was within the standard range but close to the lower limit; progesterone value was above the limit. The increase in progesterone level was compatible with the pathogenesis of cyst genesis. Beside this result, staining with PR was positive in the luteal cells that formed the luteal cyst immunohistochemically. In addition, although the hemorrhages observed in the corpus luteum are considered physiological, it should not be forgotten that they can be vital if they rupture. All these results show us the animals sent for slaughtering mostly have serious genital problems threatening their fertility.

ABSTRACT: The fluorescence polarization assay (FPA), two variants (V) of the indirect enzyme-linked immunosorbent assay (I-ELISA) and the competitive enzyme-linked immunosorbent assay (C-ELISA) were evaluated in buffaloes to detect antibodies against Brucella spp. The V1 of I-ELISA identifies them through the monoclonal (M23) anti-bovine IgG (I-ELISAM23) and the V2 through the ProteinA / G (I-ELISA-A/G). Serum samples of 862 buffaloes (Bubalus bubalis) from the Northeast of Argentina (NEA) were analyzed using the complement fixation test (CFT) as the reference. Receiving Operator Characteristic (ROC) analysis defined for the area under the curve (AUC) determined the cutoff points, sensitivity (Se) and specificity (Sp) for each test. CFT identified 107 positive and 755 negative sera. The best AUC (0.986), Concordance with CFT (96.3%) and kappa value (0.843) was obtained by I-ELISA A/G test. This assay showed the highest Se (95.33%) and C-ELISA the highest Sp (97%). FPA failed to measure the antibodies in 23 (2.65%) serum samples due to unsuccessful reading. I-ELISA M23 proved to be ineffective to diagnose brucellosis in bubaline sera. The four serological tests showed cutoff points lower than those standardized for bovines. As conclusion, I-ELISA A/G, C-ELISA and FPA with its limitations would be effective techniques for the diagnosis of brucellosis in buffaloes in the NEA, requiring an appropriate cut-off point to guarantee their maximum performance in this species.

A CFT identificou 107 soros positivos e 755 soros negativos.

Results on appropriate, inappropriate, and painful slaughtering methods of river buffalo (Bubalus bubalis) are discussed. Undesirable consequences of poor desensitization and/or slaughtering procedures, causing pain and suffering to animals, are also approached. These considerations are relevant as unsuitable methods can reduce consumer acceptance and commercial value of the meat obtained from animals undergoing deficient slaughtering procedures due to a lack of concern regarding their welfare. Relevant databases –Scopus, Science Direct, and PubMedwere searched for this review, including keywords such as pain, slaughter, stunning, Bubalus bubalis, and animal welfare. A detailed description of river buffalo's encephalic anatomy was compared to cattle concerning correct stunning and slaughter methods. The neurobiological processes of pain felt by animals when inadequately desensitized was also examined. Finally, recent findings on stunning methods for buffaloes were discussed, including encephalography. It was concluded that different animal species may require different stunning methods; at the same time, the optimal anatomical site also varies between species. Therefore, it is of the utmost importance to train the personnel performing these procedures (transport, stunning, slaughtering) to avoid mishandling that jeopardizes animal welfare by generating stimuli that trigger neurophysiological responses and, in turn, translate into pain during slaughter and reduced meat quality.(AU)

The fluorescence polarization assay (FPA), two variants (V) of the indirect enzyme-linked immunosorbent assay (I-ELISA) and the competitive enzyme-linked immunosorbent assay (C-ELISA) were evaluated in buffaloes to detect antibodies against Brucella spp.

Buffaloes are one of the important farm animals in the south of Iraq and play an essential economical role mainly acting as dairy, meat, and draft animals. This study intended to diagnose buffalo mycotic eye infections in Thi-Qar province/Iraq. Some 250 buffaloes in the herd of 3,700 animals suffered from eye infections from December 2017 to November 2018. Eye swabs were collected from each infected eye of the affected buffaloes of both sexes before treatment. The animals were in different age groups. All samples were transferred to the laboratory in transfer media, and cultured on Sabouraud dextrose (SDA) agar with and without 0.05 g/mL and 0.4 g/mL chloramphenicol and cycloheximide, respectively. Later, the agars were incubated at 25oC and 37oC. The total percentage of eye infection was (6.75%), constituting (49.2%) mycotic infections. The predominant clinical manifestations that appeared on the infected buffaloes were eye inflammation represented by congestion, lacrimation, the opacity of cornea and edema, and reduced productivity of the infected animals. Different fungal isolates were identified from the samples including Aspergillus fumigates, Aspergillus flavus, Aspergillus niger, Penicillium spp., Alternaria spp., Fusarium spp., Candida spp., Cladosporium spp., Rhodotorula spp., Mucor spp. and Rhizopus spp. Calves buffaloes below one-year-old were more prone to mycotic infection than one-year-old or more. Additionally, male buffaloes were more susceptible to infection than females. In conclusion, this study isolated various types of fungus from the inflamed eyes of buffaloes. Fungal eye infection and the potential risk factors for fungal keratitis in buffaloes were also investigated. The study also approved the rapid diagnosis of fungi by direct microscopic detection and culture. The author recommends future studies including large numbers of the buffalo herd in Iraq to determine the epidemiology of this condition in the country.(AU)

The skin is the largest organ in the body and can be affected by a series of pathological processes. Dermatological diseases in general are a frustrating aspect in the clinic of large animals. Thus, the objective of this study was to report skin lesions diagnosed in buffaloes in the region of Baixo Amazonas, state of Pará. 156 buffaloes of the Murrah, Mediterranean and crossbred breeds were taken; Adults and young people, with ages varying from eight months to nine years. On physical examination, it was found that 36.5% (57/156) of the animals had abscesses at the vaccination site. Of these, 12.2% (19/156) were abscesses with the presence of alopecia and 1.3% (2/156) with fistulation. In addition, 1.3% (2/156) of the animals separated at the vaccination site, ulcerated wounds with the presence of purulent secretion. It was identified that 3.8% (6/156) of the buffaloes had the location of the fire mark, with burns, with a crusted, erythematous and exudative aspect, in the croup region. The majority 64.1% (100/156) of the animals separation of the ears, 1.9% (3/156) dissipation by penetration of the horn and in 1.2% (2/156) a skin lesion was observed in any limb posterior right. It was concluded that it was possible to diagnose several cutaneous lesions in buffaloes in the region of Baixo Amazonas, with the error in handling animals being the main causes of the diagnosed injuries. In addition, there was a correlation between two groups of acquired animals (with and without injury).

Background: The bovine alphaherpesvirus type 1 (BoHV-1) belonging to the order Herpesvirales, family Herpesviridae, subfamily Alphaherpesvirinae and genus Varicellovirus. Bovine alphaherpesvirus 1 (BoHV-1) infections have a great importance due to the high rates of dissemination in cattles worldwide. Although, the BoHV-1 was largely related in cattle, buffaloes were also classified as host for the virus. However, studies that determine seroepidemiological data in this species are scarce and necessary. The aim of the present study was to determine the presence of antibodies against BoHV-1in healthy beef buffaloes using the virus neutralization (VN) technique.Cases: This work portrays an epidemiological survey, based on a sectional study characterized by blood samples collectedfrom 54 Murrah buffalo, aged 6 to 24 months, from the municipalities of Guaraqueçaba, Ponta Grossa, Antonina andDoutor Ulysses, located in Paraná state, being 20, 14, 10 and 10 samples from each location, respectively. Thirty-sevenpercent (20/54) of the samples were collected at slaughterhouse with registration at the Federal Inspection Service (SIF)of the Ministry of Agriculture, Livestock and Food Supply (MAPA), and 63% (34/54) at the buffalo’s farms. The serumsamples were collected in sterile tubes without anticoagulant in stored isothermal boxes, with serum separation and storedat -20ºC. The samples were sent to the Animal Virology Laboratory of Universidade Estadual de Londrina, UEL for serological analysis. Serological diagnosis using the virus neutralization (VN) technique was performed according the OIEmanual of diagnostic tests and vaccines for terrestrial animals, using the BoHV-1 Los Angeles strain. Forth-five of the 54samples (83.3%) evaluated generated titers of antibodies against BoHV-1 present in all evaluated herds, and the samplescollected in herd from Antonina city were 100% positive, following by 80% in Guaraqueçaba...
