Abstract Desvenlafaxine succinate (DVS) inhibits serotonin reuptake selectively and is approved for major depressive disorders. This research investigated influence of DVS on modulating brain monoamine and oxidative stress in mice. The antiepileptic potential of DVS (10, 20, or 30 mg/kg/i.p.) in pentylenetetrazole (PTZ; 85 mg/kg) with i.p. route of administration, strychnine (STR; 75 mg/kg) with i.p. route, pilocarpine (400 mg/kg) with s.c. route and maximal electroshock MES-induced convulsion in mouse models. The activities of oxidative stress, i.e. superoxide dismutase (SOD), glutathione (GSH) and lipid peroxidation (LPO) as well as gamma-aminobutyric acid (GABA) in the brains of PTZ-induced convulsive mice. Treatment with DVS increased the latency to develop siezures and declined mortalities in rodents against PTZ, STR and pilocarpine-induced convulsions. Results of MES-leaded siezures revealed that DVS reduced tonic hind limb extension duration and mortalities significantly. Brain, SOD, GSH and GABA level were significantly (P 0.01) increased and LPO reduced significantly (P 0.01) after DVS treatment. Furthermore, the DVS did not show any motor coordination signs in the rotarod test. We demonstrated that the role of DVS in convulsion genesis in mice under control condition and attenuate the PTZ-induced oxidative damage.

Aluminum is a neurotoxicant and one of the most harmful metals in the environment; it is producing tissue inflammation and oxidative stress. Curcumin is an effective antioxidant and neuroprotective compound with medicinal potential. Curcumin's effect on AL toxicity was investigated in this study. Two groups of 70 mature adult albino rats were used, each of which was subdivided into five groups: Control, Vehicle, Curcumin, Aluminum, and Curcumin + Aluminum group. For two periods of 20 and 40 days, animal models were administered orally AlCl3 (20 mg kg-1 bw) and/or Curcumin (100 mg kg-1 bw). In the cerebral cortex, aluminum caused a significant rise (p < 0.05) in lipid peroxidation and DNA fragmentation, as well as a significant decrease (p < 0.05) in antioxidant enzymes such as catalase, superoxide dismutase, and glutathione peroxidase. In the brain hippocampus, aluminum caused a major reduction (p < 0.05) in neurotransmitters (dopamine and serotonin), while Acetylcholine esterase activity increased sharply (p < 0.05). Aluminum also triggered histological analyses in the hippocampus of the brain. Curcumin co-administration considerably reduced the increase in lipid peroxidation and DNA fragmentation, but also enhanced the depletion of antioxidant enzymes. Curcumin also reversed the decline in neurotransmitters, the increase in Acetylcholine esterase, and the distortion in the brain hippocampus.(AU)

Two hundred and forty Japanese quail aged 125 days were randomly allocated to five treatment groups based on laying (%) and quail's weight (90.71 ± 1.8% egg/day × 100 and 178.05 ± 9.38 g, respectively), each of which included six replicates of eight quails. The diets were formulated based on corn, soybean meal, and industrial amino acids. An optimal diet achieves 100% of amino acids required by the quail requirements, except for threonine. Five treatments were made: 20% less amino acid; 10% less amino acid; optimal diet; 10% more amino acid; and 20% more amino acids than those in the optimal diet. The increase in amino acid levels in a fixed Lys: amino acid ratio led to histological alterations in the liver and uterine epithelium, reduction in blood lipid peroxidation, lower hepatic HSP70 gene expression, and the performance of laying Japanese quail. The optimal diet based on the NRC with an adjusted Thr: Lys 78 ratio (Lys 1.0%) improved the performance and efficiency of Japanese quail from 125 to 230 days of age.(AU)

Growth-promoting bacteria Azospirillum brasilense, used as an inoculant in corn culture, can be severely affected by the exposure temperature, with a lethal effect above 35°C, when cultivated alone under laboratory conditions. Such effects may limit the associative interaction between plant-bacteria, with reduced inoculation efficiency, resulting in a lower growth rate of the plant and an increase in oxidative stress. Thus, the objective of the research was to evaluate the efficiency of the inoculation process with A. brasilense in seeds and in the initial growth of seedlings of two corn cultivars submitted to different temperatures. Were utilized corn hybrids seed Syn 488 and Syn 505. The experimental design was completely randomized in a 2 x 4 factorial scheme (with and without inoculation of A. brasilense x 4 sowing temperatures: 20, 25, 30, and 35ºC), with four replications. The inoculation efficiency in corn seedlings submitted to different temperatures was evaluated through the following tests: germination, first count, seedling length and dry weight. In addition, responses at the biochemical level of the interaction (temperatures x inoculation) for the content of photosynthetic pigments and hydrogen peroxide (H2O2), antioxidant enzymes and lipid peroxidation were evaluated. The inoculation with A. brasilense changed the morphological and biochemical responses of corn seedlings, and the inoculation process was efficient for all temperatures tested. The best results were under temperatures 25, 30, and 35°C. Plant and bacteria when exposed to temperature conditions (30°C inoculated) showed a significant increase in plant biomass and activity of antioxidant enzymes.(AU)

Maternal colostrum provides the neonate with immunoglobulins, essential for passive immunity. However, storage and handling of colostrum can alter its physical and nutritional characteristics. The aim of this study was to evaluate the effects of three conservation methods on colostrum density, pH, total antioxidant capacity (TAC), protein (PP) and lipid peroxidation (LP). Colostrum samples were collected from 15 Bos indicus cows, and divided into three aliquots for storage using three methods: refrigeration at 4°C, freezing at -20°C, and lyophilization. For each method, four evaluation times were performed: day (d) 5, 10, 15, and 20 for refrigeration and d 15, 30, 45, and 60 for freezing and lyophilization. pH and density analyses were performed at each evaluation time for each method. On d 0 and 15 of storage, TAC, LP, and PP analyses were performed. A generalized linear model, Tukey's test for means comparisons, and a Pearson correlation analysis were conducted. A decrease in refrigerated colostrum pH was observed on d 15. Density of the lyophilized colostrum decreased, as compared with that of frozen colostrum. Lyophilization exhibited the lower PP values of samples, whereas refrigeration presented the highest values of LP and PP. No differences in colostrum TAC were observed between storage methods. A positive correlation between PP and colostrum density and a negative correlation between colostrum density and TAC were found. It was concluded that both freezing and lyophilization are suitable storage methods for bovine colostrum, as they limit proteins and lipids oxidation, and maintain the TAC of fresh colostrum.

Background: Oocytes and embryos produce energy through mitochondrial oxidative phosphorylation by using oxygen. The membrane structure of the embryo is mostly composed of unsaturated fatty acids, for this reason DNA fragmentation, apoptosis, and abnormal gene expression are shaped as a result of the lipid peroxidation during culture. Oxidative stress (OS) is one of the most important problems affecting the in vitro embryo development. Antioxidant supplementation to the culture medium has been an alternative way to reduce cell damage caused by oxidative stress in in vitro embryo production systems. In this study, it was aimed to determine the effect of L-ergothioneine on blastocyst development when added to the culture medium. Materials, Methods & Results: The material of the study consisted of oocytes aspirated from the ovaries of Holstein cows which were collected from the local slaughterhouse. The ovaries were delivered to the laboratory within 2-3 h in a thermos which provided a constant temperature of 25-30o C with physiological saline (0.9%) containing antibiotics. All follicles in the 3-8 mm range on the ovaries were aspirated using 20 G needle. The collected follicle fluid was filtered through filters with a pore diameter of 70 micrometers. Cells remaining in the filter were washed with OPU medium and transferred to the petri dishes. Fluids were examined under a stereomicroscope. The cumulus-oocyte complexes were classified, and A and B quality oocytes were included to the study (A, B, C, and D quality COC). Oocytes aspirated from the ovaries and collected later on were incubated in IVM medium for 22 h. After maturation, it was taken into IVF medium, semen was added and incubated for 20-22 h. Possible zygotes to be taken to the culture stage were transferred to culture (IVC) drops with (L-ergothioneine 100 µL/mL (n:121) added and without antioxidant (control (n:124)), and kept in the incubator for 6-7 days. Evaluated on the 7th day differences in in vitro embryo production stages were evaluated with the Chi-square test. The study was run in 5 replicates each time, with at least 20 possible zygotes for per group being cultured. It was determined that 262 (87.33%) of a total of 300 oocytes undergoing in vitro maturation were matured. It was determined that 245 of the mature oocytes were fertilized (93.51%). The cleavage rates of the groups were determined as 87.60% and 86.29%, respectively. Eighty-two (33.47%) blastocysts were obtained from 245 zygotes taken into the culture stage, and the blastocyst rates in the groups were found to be 40.50% and 26.61%, respectively. After the study, it was determined that the statistical difference between L-ergothioneine and control in cleavage rates was insignificant (P > 0.05) and blastocyst rates was significant (P < 0.05) Discussion: Oxygen content above normal ratios can increase the formation of reactive oxygen species (ROS), particularly hydrogen peroxide (H2 O2 ), hydroxyl radical (HO·), and peroxyl radicals (ROO·). The increased rate of ROS negatively affects the success of IVP in mammalian embryos. It was observed that L-ergothioneine, which has high antioxidant activity, improved blastocyst development rates, and higher blastocyst rates could be achieved compared to the control group. By investigating the use of L-ergothioneine in different doses, it was thought that the dose with the highest antioxidant activity could be added to the culture medium in in vitro embryo production and more blastocysts could be produced.

Biochemical and physiological parameters, growth, and yield of field crops especially salt sensitive crops like chickpea are affected adversely by salinity in arid to semi-arid regions. To investigate the effect of different salinity levels on growth, biochemical and physiological parameters of chickpea genotypes, a pot experiment following CRD, two factor factorial design, was conducted in the glasshouse at the Institute of Biotechnology and Genetic Engineering, The University of Agriculture, Peshawar, Pakistan. Ten (10) kg of soil was filled in each pot and salinity levels were maintained @ S0= 0 mM NaCl, S1= 50 mM NaCl, S2= 100 mM NaCl and S3= 150 mM by applying NaCl and 5 genotypes of chickpea (KK-2, Bhakkar-2011, Bittle-98, Punjab-2008, and CM-98) were used. At crop maturity, growth parameters, physiological, biochemical, and ionic parameters were measured using standard analysis procedures. Salinity reduced the growth and yield of all genotypes, but the rate of decrease was different among the genotypes tested. From the results, a decrease in K concentration, K/Na ratio, transpiration rate, stomatal conductance, N, and P was observed in all genotypes with the increase in salinity. An increase in salinity level increased the proline content (35.45%), crude protein (42%), H2O2 (19%), lipid peroxidation (62%), carbohydrates (23.22%), and Na+ concentration (137%). The highest level of salinity, 150 mM NaCl has exhibited the highest salinity stress in all parameters. Genotype KK-2 and Bhakkar-11 showed a lower rate of relative decrease in yield (4.5 and 12%), K+/Na+ ratio (23.34 and 11.47%), and K+ concentration (7.9 and 11%), respectively, and the lowest relative increase in Na+ accumulation (20.3 and 0.48%), @ 50 mM salinity compared to control. Genotype KK-2 and Bhakkar-11 proved better @ 50mM salinity. The findings suggest that the critical level of the salinity must be kept in mind and the salt-tolerant genotypes should be cultivated in salt affected soils.

Abstract The objective of this study was to investigate the effect of Spirulina platensis extract (SPE) addition to the freezing extender on freezability, lipid peroxidation, ultrastructure alterations and fertilizing potentials of frozen-thawed buffalo bull spermatozoa. Semen samples were collected with artificial vagina from five adult fertile bulls and diluted with Tris-base extender containing SPE (1, 5, 10 and 20 μg/mL) or without SPE (control). Diluted semen was cooled to 4 °C throughout one hour and frozen in 0.25 mL straws: prior to being stored in liquid nitrogen. Cryopresreved spermatozoa were assessed for post-thawing sperm motility, viability, acrosomal integrity, ultrastructure changes, antioxidant activities, lipid peroxidation and fertility rate. The current results clearly indicated that adding 10μg/mL SPE to the freezing extender significantly improved (P< 0.05) post-thawing motility and decrease the percentage of acrosomal damage (51.67±6.02% and 16.33±1.46%, respectively) compared with the control (28.33±4.41% and 26.33±1.77%, respectively). Moreover, addition of 10 μg/mL SPE to the semen extender significantly diminished (P< 0.05) MDA concentration (10.66±2.40 nmol/109) compared with the control (22.66±4.26 nmol/109). Therefore, the present results revealed that addition of 10μgl/mL SPE to the freezing extender might improve semen quality and reduce cryodamage of the buffalo bull spermatozoa.

Therefore, the present results revealed that addition of 10μgl/mL SPE to the freezing extender might improve semen quality and reduce cryodamage of the buffalo bull spermatozoa.(AU)

The inoculation with A. brasilense changed the morphological and biochemical responses of corn see

The black oat (Avena strigosa Schreb.) stands out as a forage of great importance in Brazilian agriculture. However, the productivity and quality of this forage can be affected by abiotic factors, such as temperature and water availability, which affect the physiological processes and facilitate the accumulation of free radicals (reactive oxygen species - ROS). Thus, the objective of this study was to understand the biochemical changes in black oat plants subjected to water deficit at different temperatures. Experiments were conducted in a greenhouse in two experimental periods, which presented an average temperature of 20 °C and 24 °C, respectively. Black oat seeds, of the variety IAPAR 61, were sown in pots and the plants were irrigated for 60 days. After which, the pots were covered with plastic bags and the irrigation was suspended. The analyses were carried out in five periods of evaluation - M1: plants before the suspensionof irrigation, M2: plants at the first wilting point, M3: three days after plastic removal and irrigation return, M4: four days after M3 and before the second suspension of irrigation, and M5: the second wilting point. The levels of total protein and malondialdehyde (MDA), and the activity of the enzymes catalase (CAT) and ascorbate peroxidase (APX), were analyzed. The experimental design was completely randomized, with six replications, in a factorial scheme of average temperature × water management × periods of evaluation, and the means were compared by Tukey's test at 5%. In response to water deficiency and temperature increase, black oat plants increased their levels of total soluble proteins, and there was greater lipid peroxidation due to the increase in malondialdehyde content. There was no change in the activity of the enzymes catalase and ascorbate peroxidase under water deficit, and these activities decreased with increasing temperature.

This study aimed to evaluate the antioxidant potential of citric acid in plant protein based-diets offered to juvenile tambaqui, Colossoma macropomum. Four isonitrogenous and isoenergetic diets containing different levels of citric acid (0, 10, 20, and 30 g.kg−1) were formulated. Fish (n = 160; 27.56±1.03 g) were randomly distributed in 310-L water tanks (n = 16), with four replicates per treatment. After the experimental period (80 days), animals were anesthetized, and blood was collected for cell count. They were euthanized and dissected to remove livers for subsequent oxidative stress analyses. No significant differences were observed in the levels of thrombocytes, total leukocytes, lymphocytes, positive granular leukocytes, and immature leukocytes among the experimental treatments. However, there were fewer monocytes and neutrophils in fish fed the diet containing 30 g.kg−1 of citric acid. All citric acid diets increased glutathione concentration and reduced lipid peroxidation levels in livers. Therefore, citric acid acts as a potent antioxidant in juvenile tambaqui fed plant protein-based diets and, thus, can improve their maintenance conditions in production systems.

Canine sperm is a very delicate cell that is quite susceptible to oxidative stress since the cytoplasm is restricted and features little antioxidant reserves. Furthermore, the sperm membrane has some polyunsaturated fatty acids sensitive to lipid peroxidation, which makes it important to addition antioxidant substances to the diluter aiming at decreasing such stress to the sperm cell, particularly during seminal cryopreservation. Several antioxidants have been used in this process in some domestic animal's species, however, the use of palmitic acid has been little reported in works on cryopreservation of semen of the canine species. Hence, this study aimed to assess the effect of addition antioxidants palmitic acid and vitamin E to the Tris-egg yolk diluter on the semen quality of dogs after thawing. Samples were collected from the ejaculates of 4 adult dogs, apparently healthy, of the American Pit Bull Terrier breed of kennels in the city of Teresina, PI, places where the pre-freezing procedures of the dog's semen were performed. The samples were diluted in Tris citric acid fructose (3.28 g Tris-hydroxymethyl-aminomethane, 1.78 g citric acid monohydrate and 1.25 g D-fructose), dissolved in 100 mL distilled water, and added 20% egg yolk and 6% glycerol, at the concentration of 100x106 sptz/mL. The semen samples were divided into 3 mL aliquots to form 3 experimental groups: G1 - Only Tris-egg yolk (Control group); G2 - Tris-egg yolk + 100 µM palmitic acid; and G3 - Tris-egg yolk + 116 µM vitamin E. Semen was collected weekly over a period of little over 2 months. After thawing, thermorresistance test (TTR) was carried out at 0, 30, 60, and 90 min to assess spermatics motility and vigor, in addition to analysis of integrity of plasma membrane, acrosomal membrane and mitochondrial activity of the sperm, using fluorescent probes. These assessments were performed out at the Animal Reproduction Biotechnology Laboratory (LBRA/UFPI). In the TTR, G2 and G3 didn't exhibit significant results for spermatics motility or vigor when compared with the control group. The palmitic acid and vitamin E also had no significant effects on the parameters of acrosomal membrane integrity or mitochondrial activity. However, sperm cryopreserved with the addition of palmitic acid exhibited significant differences for plasma membrane integrity, providing greater protection to the sperm cells in G2. The palmitic acid is one of the most saturated fatty acids in human semen, with reports of great proportions also in the seminal plasma of dogs. Its main role is to protect the plasma membrane from external damage, improving viability and fertility of the sperm after cryopreservation. Data is scarce in the literature on the composition of fatty acids in canine semen and regarding the use of palmitic acid as a seminal antioxidant in that species, which grants further studies aiming to investigate such valuable information for canine reproduction. It is concluded that addition palmitic acid at 100µM concentration to the Tris-egg yolk diluter was able to preserve the integrity of the plasma membrane during the process of cryopreservation of canine semen.(AU)

Boar spermatozoa are very susceptible to cryopreservation injuries and, for this reason, pig remains one of the few species in which fresh semen is still preferred to thawed one for routine artificial insemination (AI). The present work evaluated the effect of supplementing boar sperm thawing medium with Silvafeed SP (SSP), a mixture of Chestnut and Quebracho wood extracts (60/40 w/w) rich in polyphenols (92.4% tannin content) on in vitro fertilization (IVF) and on the following sperm parameters: sperm motility (assessed by CASA), viability, acrosome integrity, mitochondrial function and lipid peroxidation (assessed by flow cytometry) and capacitation status (immunolocalization of tyrosine phosphorylated proteins). Thawed spermatozoa were incubated 1 h at 37°C in BTS without (CTR) or with (5, 10, 20 µg/mL) SSP. After incubation sperm suspension was divided in three aliquots: one was used for IVF trials, one for sperm analysis, and the last one was capacitated for 1 h at 39°C 5% CO2 in IVF medium. Sperm motility parameters, viability, acrosome integrity, mitochondrial functionality, lipid peroxidation and tyrosine phosphorylated protein immunolocalization, used as capacitation parameter, were not influenced by SSP. However, oocytes inseminated with thawed spermatozoa pretreated with all the different SSP concentrations presented a significant (P < 0.01) increase in penetration rate compared to CTR. In addition, 5 µg/mL SSP exerted a positive effect (P<0.05) on the total efficiency of fertilization. These results encourage the use of SSP in the thawing medium since post-thawing fertility is a limit for the large-scale use of boar frozen semen.

Root system architecture plays a fundamental role in the adaptation of rice to environments subjected to abiotic stresses. This study aimed to characterize the root architecture and morphology of two Brazilian upland rice varieties which display contrasting behavior towards drought tolerance: the tolerant Catetão and susceptible Mira varieties. Two experiments were carried out under greenhouse and growth chamber conditions. The experimental design was completely randomized in a 2 × 2 factorial scheme (varieties × drought stress conditions) with four replicates. Rice varieties were subjected to control and drought conditions for 14 days. Drought stress was applied either by withholding water until the desirable soil tension was achieved or by addition of 20% polyethylene glycol 6000 (PEG) to the nutritive solution. After harvest, the dry weight, root architecture, as well as physiological and root features were assessed. Under drought stress, a higher root biomass was recorded in Catetão compared to Mira. A higher ratio of deep rooting (RDR) was observed for Catetão, while Mira remained stable in response to drought stress. The evaluated physiological features showed that the Catetão variety is less likely to compromise membrane integrity and lipid peroxidation during drought stress. Moreover, analyzed root features exhibited a significant increase in lateral root emission and root density for Catetão in response to drought treatment, which may be considered an important feature when selecting for superior genotypes.(AU)

The purpose of this study was to evaluate different lipid sources in diets for silver catfish juveniles regarding aspects of productive performance, centesimal composition, hepatic histology, biochemical responses, and oxidative stress. A total of 300 juveniles with an initial mean weight of 18.45 ± 1.22g were distributed in 20 net-pens of 1m3, arranged in a 200 m2 masonry tank, for a period of 90 days. The experimental diets were isoprotein (29.05% digestible protein) and isoenergetic (3,250 kcal.kg-1), containing different sources of oils (soybean, sunflower, fish, canola and olive) at a concentration of 3.0%. The animals were distributed in a completely randomized design, with five treatments and four replicates. The data were submitted to ANOVA, and the means, when significant, were compared by the Tukey test at 5% significance level. Visceral fat index was higher for fish fed diets containing sunflower oil (P0.05) were observed for the variables of productive performance, centesimal composition, and blood biochemistry. A difference (P<0.05) was observed in hepatic histology, where the treatment with soybean oil had a higher number of hepatocytes. The animals fed with fish oil had a higher TBARS lipid peroxidation and a higher GST enzyme activity (P<0.05). Therefore, the different lipid sources can be used to feed this species without any damage to productive performance. It is not advisable to include sunflower oil, as it provides greater deposition of visceral fat.
