Resumo

This experiment conducted using 20 Rahmani ewes at the last third of pregnancy in two equal groups. One group served as control, while the other group received Spirulina platensis(SP) at the rate of 0.5 gm 10 kg-1live body weight. The objective was to find out the effect of adding Spirulina platensisalgae to small ruminant rations on reproductive and productive traits and blood components of sheep. The experiments lasted for 120 days for both dams and their lambs after weaning. The findings proved that adding SP in ewes' diets had no effect on the average of live body weight change. Average milk yield was significantly (p <0.01) higher in the treatment group than the control. Lamb's birth weight and daily body gain of the treated group were significantly (p <0.01) higher than the control. Blood and serum picture profile of ewes were significantly higher when fed SP additive than the control.It could be concluded that the addition of SP to the ration of sheep positively preserved their health, productive and reproductive status as well as their lambs' growth rate. Also the additive improved the economic efficiency of treated animals by about 53.13%.(AU)

Resumo

We aimed to evaluate the inclusion of expeller soybean meal on the quality and milk production of Holstein cows, as well to estimate and compare the inherent parameters of the in vitro ruminal kinetics. Data collection was carried out on a private property, in the interior of the municipality of Dois Vizinhos - Paraná, Brazil. Twenty Holstein cows were used, divided into two treatments: one with the inclusion of expeller soybean meal and the other with conventional soybean meal. The design used was completely randomized with parity. Milk production was evaluated through weighing. Milk samples were collected on days 0, 15, 30 and 45 to evaluate the milk protein, fat, urea nitrogen, lactose and total solids. The In vitro ruminal kinetics of convencional and expeller soybean meal was performed at the Universidade Tecnológica Federal do Paraná - Paraná, Dois Vizinhos. The gas pressure and volume measures were taken after 1, 2, 3, 6, 8, 10, 12, 16, 20, 24, 30, 36, 48 and 72 hours of sample incubation. The results obtained were applied in a two-compartmental mathematical model with no latency time in the first compartment for description of ruminal kinetics. Data were analyzed by variance (ANOVA) and compared by the F test. There was no significant difference in any of the variables evaluated from mil production and quality when soybean meal expeller was included in the animals' diet (P>0.05). The expeller soybean meal showed a lower degradation compared to the conventional meal. There were no significant differences for milk production as for concentration of solids in milk.


O objetivo foi avaliar a inclusão do farelo de soja expeller na qualidade e produção do leite de vacas holandesas, assim como estimar e comparar os parâmetros inerentes a cinética ruminal in vitro do farelo de soja convencional e expeller. A coleta de dados a campo foi realizada em propriedade particular, no interior do município de Dois Vizinhos - Paraná, Brasil. Foram utilizadas 20 vacas holandesas, divididas em dois tratamentos: um com inclusão de farelo de soja expeller e o outro com farelo de soja convencional. O delineamento utilizado foi inteiramente casualizado com paridade. Foi avaliada a produção do leite através de pesagem diária. Amostras de leite foram coletadas nos dias 0, 15 e 30 para determinação de proteína, gordura, nitrogênio ureico, lactose e sólidos totais. cinética ruminal in vitro do farelo de soja convencional e farelo de soja expeller foi realizada na Universidade Tecnológica Federal do Paraná, campus Dois Vizinhos. As leituras de pressão e volume foram realizadas após 1, 2, 3, 6, 8, 10, 12, 16, 20, 24, 30, 36, 48 e 72 horas de incubação. Os resultados obtidos foram aplicados em modelo matemático bicompartimental sem latência no primeiro compartimento para descrição da cinética ruminal. Os dados foram submetidos à análise de variância (ANOVA) e comparados pelo Teste F. Quanto a produção e qualidade do leite, não houve diferenças significativa para as variáveis avaliadas quando incluído o farelo de soja expeller na dieta dos animais (P>0,05). O farelo de soja expeller apresentou uma degradação inferior comparada ao farelo convencional. Não houve diferenças significativas para a produção de leite como para a concentração de sólidos no leite.

Maternal colostrum provides the neonate with immunoglobulins, essential for passive immunity. However, storage and handling of colostrum can alter its physical and nutritional characteristics. The aim of this study was to evaluate the effects of three conservation methods on colostrum density, pH, total antioxidant capacity (TAC), protein (PP) and lipid peroxidation (LP). Colostrum samples were collected from 15 Bos indicus cows, and divided into three aliquots for storage using three methods: refrigeration at 4°C, freezing at -20°C, and lyophilization. For each method, four evaluation times were performed: day (d) 5, 10, 15, and 20 for refrigeration and d 15, 30, 45, and 60 for freezing and lyophilization. pH and density analyses were performed at each evaluation time for each method. On d 0 and 15 of storage, TAC, LP, and PP analyses were performed. A generalized linear model, Tukey's test for means comparisons, and a Pearson correlation analysis were conducted. A decrease in refrigerated colostrum pH was observed on d 15. Density of the lyophilized colostrum decreased, as compared with that of frozen colostrum. Lyophilization exhibited the lower PP values of samples, whereas refrigeration presented the highest values of LP and PP. No differences in colostrum TAC were observed between storage methods. A positive correlation between PP and colostrum density and a negative correlation between colostrum density and TAC were found. It was concluded that both freezing and lyophilization are suitable storage methods for bovine colostrum, as they limit proteins and lipids oxidation, and maintain the TAC of fresh colostrum.


O colostro materno fornece ao recém-nascido imunoglobulinas que são essenciais para a imunidade passiva. No entanto, o armazenamento e o manuseio do colostro podem alterar suas características físicas e nutricionais. O objetivo deste estudo foi avaliar os efeitos de três métodos de conservação na densidade, pH, capacidade antioxidante total (CAT), peroxidação protéica (PP) e lipídica (LP) do colostro. Amostras de colostro foram coletadas de 15 vacas Bos indicus, e divididas em três alíquotas para armazenamento usando três métodos: refrigeração a 4 °C, congelamento a -20 °C e liofilização. Em cada método, foram realizados quatro tempos de avaliação: dia (d) 5, 10, 15 e 20 para refrigeração e dia 15, 30, 45 e 60 para congelamento e liofilização. Além disso, as análises de pH e densidade foram realizadas em cada momento de avaliação para cada método. No dia 0 e 15 de conservação foram realizadas análises da CAT, PL e PP. Foi realizado um modelo linear generalizado, teste de Tukey para comparação de médias e análise de correlação de Pearson. Uma diminuição no pH do colostro refrigerado foi encontrada no dia 15. A densidade do colostro liofilizado diminuiu em comparação com o colostro congelado. A liofilização apresentou menor PP das amostras, enquanto a refrigeração apresentou os maiores valores de PL e PP. Não foram observadas diferenças no TAC do colostro entre os métodos de armazenamento. Foi encontrada uma correlação positiva entre PP e densidade do colostro e uma correlação negativa entre densidade do colostro e CAT. Assim, concluiu-se que tanto o congelamento quanto a liofilização são métodos adequados de armazenamento do colostro bovino, pois limitam a oxidação de proteínas e lipídios e mantêm o TAC do colostro fresco.

Effects were assessed of the dilutants TRIS and ACP - 101c® with the addition of different guinea fowl (Numida meleagris) egg yolk concentrations. Fifteen ejaculates were collected from five goats of the Anglo Nubian breed. The ejaculates were pooled and then divided into 12 groups, two control groups (GC1 TRIS, with 2.5% Gallus gallus domesticus hen egg yolk GOGD), (GC2 Control Group ACP - 101c®, with the addition of 2.5% Gallus gallus domesticus hen egg yolk GOGD) and ten experimental groups (EG), containing TRIS and ACP added with different concentrations of egg yolk from guinea hen (Numida meleagris) (TRIS 2,5% GONM; TRIS 5% GONM; TRIS 10% GONM; TRIS 15% GONM; TRIS 20% GONM; ACP® 2,5% GONM; ACP® 5% GONM; ACP® 10% GONM; ACP® 15% GONM; ACP® 20% GONM). Then cryopreservation was carried out and the samples stored in liquid nitrogen (-196 °C). After seven days, the samples were thawed and assessed for spermatic kinetics, immunofluorescence and sperm morphology. Analysis of GOMN by the CASA system showed that the various parameters were similar to those of GOGD (P>0.05). The membrane integrity, mitochondrial potential and the acrosome were not influenced by the treatment (P>0.05) nor by the dilutant used for cryopreservation (P>0.05). The spermatic morphology was also preserved by the different GOGD and GONM concentrations in the ACP® and TRIS dilutants, with no statistically significant differences (P<0.05). It was concluded that Numida meleagris egg yolk, as external membrane cryoproctant added to the dilutants ACP-101c® and TRIS, improved goat semen quality.(AU)

This research aimed to develop a fermented dairy beverage using whey as a dairy base with chia seed (Salvia hispanica L.) and acerola syrup (Malpighia emarginata) and to evaluate physicochemical parameters (pH, water activity, total soluble solids, acidity, syneresis), proximate composition (moisture, ash, proteins, milk fat, carbohydrates, and energy value), ß-carotene, ascorbic acid, microbiological and sensory parameters. The acidity results ranged from 1% to 1.27%; pH from 3.86 to 4.11; soluble solids from 15.67 to 21.6; water activity from 0.93 to 0.99; syneresis from 46.67 to 68.08; they presented satisfactory conditions for thermotolerant coliforms. Salmonella spp. showed cell viability of L. acidophilus: E4 (2.9 x 107 to 9.7 x 107 ) and E8 (1.3 x 107 to 8.6 x 107), however Bifidobacterium spp. did not show any viability. The drinks had a good acceptance rate by the tasters (7.0). The humidity results were from 74.21% to 74.34%; ash from 0.42% to 0.55%; proteins from 2.93% to 2.99%, milk fat from 1.47% to 0.93%; carbohydrate from 20.97% to 21.19%; energy value from 108.83% to 105.09% and ß-carotene from 12.33% to 8.19%; and ascorbic acid ranged from 222.23 (mg/100g) to 418.10 (mg/100g). It is concluded that formulated dairy drinks are considered a viable alternative for the food industry, with the differential of including chia seed and acerola pulp, due to their good sensory acceptance, physical-chemical stability, satisfactory sensory analysis, microbiological standards suitable in the 21-day storage period.


O objetivo desta pesquisa foi desenvolver uma bebida láctea fermentada utilizando o soro de leite como base láctea com semente de chia (Salvia hispanica L.) e xarope de acerola (Malpighia emarginata), avaliar parâmetros físico-químicos (pH, atividade de água, sólidos solúveis totais, acidez, sinérese), composição centesimal (umidade, cinzas, proteínas, matéria gorda láctea, carboidratos e valor energético), ß-caroteno, ácido ascórbico, parâmetros microbiológicos e sensorial. Os resultados de acidez variaram de 1% a 1,27%; pH de 3,86 a 4,11; sólidos solúveis de 15,67 a 21,6; atividade de água de 0,93 a 0,99; sinérese de 46,67 a 68,08. Apresentaram condições satisfatórias para Coliformes termotolerantes e Salmonella spp., apresentou viabilidade celular de L. acidophilus: E4 (2,9 x 107 a 9,7 x 107) e E8 (1,3 x 107 a 8,6 x 107), porém Bifidobacterium spp. não apresentou viabilidade. As bebidas tiveram bom índice de aceitação pelos provadores (7,0). Os resultados de umidade foram de 74,21% a 74,34%; cinzas de 0,42% a 0,55%; proteínas de 2,93% a 2,99%, matéria gorda láctea de 1,47% a 0,93%; carboidrato de 20,97% a 21,19%; valor energético de 108,83% a 105, 09% e ß-caroteno de 12,33% a 8,19%; e o ácido ascórbico variou de 222,23 (mg/100g) a 418,10 (mg/100g). Conclui-se que as bebidas lácteas formuladas são consideradas uma alternativa viável para indústria de alimentos, com o diferencial da inclusão da semente de chia e polpa de acerola, devido a sua boa aceitação sensorial, estabilidade físico-química, análise sensorial satisfatória, padrões microbiológicos adequados durante o período de estocagem de 21 dias.

We examined the effect of dried distillers grains with solubles (DDGS) in diets for late gestating sows on sow and litter performance, colostrum chemical composition, and sow behavior. Sixty gestating sows of 3.77±1.65 parity were divided into three groups of 20 animals, in a randomized block design with 0 (corn-soybean meal diet), 150, or 300 g kg−1 DDGS from 84 days of gestation until farrowing. Sows fed 300 g kg−1 DDGS presented a lower frequency of standing position and eating activity. The lying lateral position was more frequent in the first 40 min post-feeding for sows fed the highest DDGS level. There was no effect of diets on sow and litter performance; however, DDGS inclusion tended to increase lactation feed intake. Inclusion of DDGS was inclined to produce more lactose and less protein in colostrum, but did not affect colostrum fat, total solids, and ash contents. Inclusion of up to 300 g kg−1 DDGS in the diet of late gestation sows does not change sow and litter performance and colostrum composition compared with corn-soybean meal diets, but it favored animal welfare.(AU)

O tratamento do leite em ultra alta temperatura (UAT) possibilita a eliminação de patógenos e aumento do tempo de prateleira, no entanto, bactérias psicrotróficas são associadas a efeitos negativos no produto, como lipólise, rancificação e a geleificação. Resultado da resistência de alguns esporos ao processamento, e à possível atividade residual de enzimas termorresistentes. Assim, o objetivo deste estudo foi enumerar bactérias psicrotróficas e realizar a prova de lactofermentação em leites UAT comercializados na região de Curitiba, Paraná. Foram adquiridas 60 amostras de duas marcas (30 da marca A e 30 da marca B) quinzenalmente em três diferentes momentos. Em cada momento 10 amostras de um mesmo lote eram separadas em dois grupos (D0 e D7), sendo D0: cinco amostras avaliadas no dia da compra, e D7: cinco amostras avaliadas após incubação a 37± 1ºC/7 dias e analisadas quanto à enumeração de bactérias psicrotróficas, coloração de Gram e prova da lactofermentação. Em D0, uma (3,4%) amostra (marca A) apresentou crescimento de psicrotróficos (2,50 x 105 UFC/mL), Gram negativos. As amostras da marca B não apresentaram crescimento microbiano. Vinte e cinco (83,3%) amostras não formaram coágulo, e cinco (16,7%) produziram coágulo esponjoso. Em D7, nenhuma amostra apresentou contagem bacteriana, 20 (66,7%) não produziram coágulo e 10 (33,3%) produziram coágulo esponjoso. O processo UAT utilizado pela marca A não garantiu efetividade na redução de bactérias psicrotróficas, e o resultado do teste de lactofermentação indica a possível contaminação por coliformes em ambas as marcas. Ainda, a ausência de coágulos em diversas amostras pode ser um indicativo do uso de conservantes, o que é considerado fraude.


The beneficial effects of ultra-pasteurization milk treatment include elimination of milk pathogens, besides increasing the product's shelf-life, however, psychrotrophic bacteria are associated with several negative effects in the final product such as lipolysis, rancidification and gelation. This results from heat-resistant spores and its possible residual extracellular heat-resistant enzymes activity. With that in mind, this paper aimed to quantify psychrotrophic bacteria and perform a milk clotting test in UHT milk sold in Curitiba, Paraná. 60 samples from two brands were acquired (30 from A brand and 30 from B brand) each 15 days in different moments. Each time, 10 samples from a same batch were separated in two groups (D0 and D7), being D0: five samples analyzed in the purchase day, and D7: five samples analyzed after incubation at 37± 1ºC/7 days and evaluated regarding enumeration of psychrotrophic bacteria, Gram staining and milk clotting test. The heat-treating process effectiveness was evaluated by analyzing 30 samples from each brand, A and B, totalizing 60, being 10 from each different production batches, through the surface inoculation method in PCA, milk clotting test and classification of the isolated colonies by the Gram method. In D0, one (3,4%) sample (from A brand) resulted in Gram negative psychrotrophic growing (2,50 x 105 UFC/mL) . Samples from B brand did not result in bacterial growth. 25 samples (83,3%) did not clot, and five (16,7%) produced sponge clots. In D7, none of the samples presented bacterial growth, 20 (66,7%) did not clot and 10 (33,3%) produced sponge clots. Mesophilic bacterial culture was not done. The A brand UHT process did not grant sterilization regarding psychrotrophic bacteria, and the milk clotting results indicate possible coliforms contamination in both brands. Furthermore, clot absence in several samples could indicate preservatives usage, which is considered fraud.

Effects of plasticizer types and whey protein concentrate (WPC) as coatings were evaluated on internal quality and shelf life of eggs stored for 42 days. Eggs were coated with WPC at 8 % solution combined with the plasticizers glycerol (GLY), sorbitol (SOR), and propylene glycol (PRO). The eggs were stored at 20 °C. Weight loss, Haugh Unit (HU), albumen and yolk pH, and yolk index (YI) were evaluated weekly from day 0 to 42 days. After, electron microscopy of the eggshell structure was performed. The data was submitted to the analysis of variance and the effects of treatment, storage time, and the interaction between these factors were evaluated. There was significant interaction between factors (treatment and periods) for weight loss (p < 0.0001), HU (p < 0.0001), albumen (p < 0.0001), and yolk pH (p < 0.0001), and YI (p < 0.0001). After 42 days, uncoated eggs showed greater weight loss (5.4 %), compared to WPC+GLY, (3.8 %), WPC+SOR (3.3 %) and WPC+PRO (3.9 %). Similar results were verified for HU at 42 days of storage. Uncoated eggs showed HU of 58.46 (B), while coated eggs showed higher values: WPC+GLY – 66.58 (A), WPC+SOR – 68.79 (A), and WPC + PRO – 71.53 (A). The plasticizers GLY, SOR and PRO, associated with WPC, demonstrated effectiveness in maintaining the quality of eggs throughout the 42 days of storage. However, WPC+SOR showed superiority in preserving quality integrity of eggs. This result can be related to the chemical structure of SOR, making the combination more efficient for storage.

Weight loss, Haugh Unit (HU), albumen and yolk pH, and yolk index (YI) were evaluated weekly from day 0 to 42 days. After, electron microscopy of the eggshell structure was performed. The data was submitted to the analysis of variance and the effects of treatment, storage time, and the interaction between these factors were evaluated. There was significant interaction between factors (treatment and periods) for weight loss (p < 0.0001), HU (p < 0.0001), albumen (p < 0.0001), and yolk pH (p < 0.0001), and YI (p < 0.0001). After 42 days, uncoated eggs showed greater weight loss (5.4 %), compared to WPC+GLY, (3.8 %), WPC+SOR (3.3 %) and WPC+PRO (3.9 %). Similar results were verified for HU at 42 days of storage. Uncoated eggs showed HU of 58.46 (B), while coated eggs showed higher values: WPC+GLY 66.58 (A), WPC+SOR 68.79 (A), and WPC + PRO 71.53 (A). The plasticizers GLY, SOR and PRO, associated with WPC, demonstrated effectiveness in maintaining the quality of eggs throughout the 42 days of storage. However, WPC+SOR showed superiority in preserving quality integrity of eggs. This result can be related to the chemical structure of SOR, making the combination more efficient for storage.(AU)

The aim of this study was to examine the behavioral, productive, reproductive and thermoregulatory performance of ¾ and ⅞ Holstein/Gir (HG) dairy cows to identify which genetic composition suffers the least heat stress. A completely randomized design was adopted involving 20 multiparous, lactating crossbred cows from a dairy farm located in the municipality of Turvânia - GO, Brazil. During the experimental period, data on behavioral, productive, reproductive and thermoregulatory traits were collected every 15 days. Bioestat (5.0) statistical software was used for statistical analysis. The mean temperature-humidity index, ambient temperature and relative humidity obtained throughout the experimental period were 74.45, 30.51 ºC and 63.64%, respectively. In terms of reproductive performance, there was a significant difference (p < 0.05) between the genetic compositions for the time from calving to first service, with a longer period shown by the animals with greater Holstein breed genetic composition. Service period and calving interval differed significantly (p < 0.05) during the experimental period between the genetic compositions. A significant difference (p < 0.05) was also detected between the compositions for respiratory rate, with higher values observed in the animals with higher European genetic composition. Benezra thermal comfort index also differed significantly (p < 0.05), with the ⅞ HG animals showing a higher value. Lactating cows with a ⅞ HG genetic composition achieved better results in milk production, whereas those with ¾ HG genetic composition showed superiority in milk fat composition and reproductive and thermoregulatory performance, indicating greater adaptation to the climatic conditions of the region.(AU)


Objetivou-se avaliar o desempenho comportamental, produtivo, reprodutivo e termorregulador de vacas leiteiras ¾ e ⅞ Holandês/Gir (HG), com intuito de identificar qual composição genética sofre menor estresse térmico pelo calor. Foi utilizado um delineamento inteiramente casualizados com 20 vacas mestiças, em lactação, multíparas, pertencentes à propriedade leiteira localizada no município de Turvânia, GO. Durante o período experimental, a cada 15 dias foram coletados dados das características comportamentais, produtivas, reprodutivas e termorreguladoras. Para a realização das análises estatísticas foi utilizado o programa estatístico Bioestat (5.0). Os valores médios obtidos durante todo o período experimental para índice de temperatura e umidade, temperatura ambiente e umidade relativa do ar foram 74,45, 30,51ºC e 63,64%, respectivamente. Quanto ao desempenho reprodutivo, houve diferença significativa (p < 0,05) entre as composições genéticas para o intervalo do parto ao primeiro serviço, observando valor maior para os animais com maior composição genética da raça Holandesa. Para as características período de serviço e intervalo de parto foram constatadas diferenças significativas (p < 0,05) durante o período experimental entre as composições genéticas. Foi encontrado diferença significativa (p < 0,05) entre as composições na característica de frequência respiratória, observando-se maior frequência para os animais com maior composição genética europeia. Foi observado diferença significativa (p < 0,05) na característica índice de conforto de Benezra, onde foi encontrado maior valor para a composição genética ⅞ HG. As vacas em lactação com composição genética ⅞ HG obtiveram melhores resultados na produção de leite, mas em contrapartida a composição genética ¾ HG mostraram superioridade na composição de gordura do leite e no desempenho reprodutivo e termorregulador, evidenciando maior adaptação as condições climáticas da região.(AU)

O objetivo deste estudo foi investigar a interferência do teor de gordura do leite no ponto de congelamento (PC) obtido pelo infravermelho com transformada de Fourier (FTIR). Uma fração de leite cru foi desnatada, obtendo-se creme e leite desnatado. O leite integral e o desnatado foram adicionados com água (0%, 2%, 5%, 10%, 15% e 20%). O leite desnatado remanescente foi adicionado com creme (5%, 10%, 15%, 20%) e água (0%, 2%, 5%, 10%, 15% e 20%). As amostras foram analisadas para PC (crioscópio eletrônico e FTIR) e composição (FTIR). O PC medido por FTIR foi altamente correlacionado com o método do crioscópio eletrônico (acima de 98,5%) e, como esperado, a adição de água foi significativa (P≤0,001) ao aumentar o PC do leite em ambos os métodos. No entanto, o alto teor de gordura no leite cru resultou na redução do PC ao se utilizar o FTIR, enquanto o baixo teor de gordura resultou em aumento do PC (P≤0,001). Uma vez que a adição de água causou o efeito inverso do alto teor de gordura no PC medido por FTIR, é importante considerar o teor de gordura do leite para evitar interpretações erradas do PC quando se utiliza o método FTIR.(AU)


The objective of this study was to investigate the interference of milk fat content upon the Freezing Point (FP) obtained by Fourier-transform infrared (FTIR). A fraction of raw milk was skimmed, obtaining cream and skim milk. Whole and skim milk were added with water (0, 2, 5, 10, 15, and 20%). The remaining skim milk was added with cream (5, 10, 15, 20%) and water (0, 2, 5, 10, 15, and 20%). Samples were analyzed for FP (thermistor cryoscope and FTIR) and composition (FTIR). FP measured by FTIR was highly correlated with the thermistor cryoscope method (above 98.5%) and, as expected, water addition was significant (P≤0.001) upon increasing milk FP in both methods. However, high fat content in raw milk resulted in decreasing FP when using FTIR, while low fat content resulted in increased FP (P≤0.001). Since water addition caused the inverse effect of high fat on FP measured by FTIR, it is important to regard the fat content of milk to avoid misinterpretation of FP when using the FTIR method.(AU)

We aimed to evaluate the impact of milking management practices on milk quality though somatic cell count (SCC) and total bacterial count (TBC). By means of a survey that included farmers associated to two cooperatives, namely Cosulati in the state of Rio Grande do Sul and Castrolanda in the state of Paraná, the hypothesis that milking handling practices impact SCC and TBC levels negatively, regardless of the technological development level of the farm, was analyzed. For such, the quantile regression method was used to estimate models for the 10th, 50th, and 90th quantiles. The results confirmed that SCC and TBC levels decrease as the number of practices increases. However, only three practices impact SCC and TBC levels significantly, and their impact is not related to the farm technological level, according to tests performed by quantile regression. Finally, the study showed that some milk quality and management practices affect somatic cell count and total bacterial count more than others, which consists of using the correct active principle during dry cattle, post-dip, and the water temperature control periods.(AU)

Considerando os diversos problemas tecnológicos e econômicos que os microrganismos deteriorantes acarretam aos laticínios, o objetivo deste trabalho foi quantificar aeróbios mesófilos, psicrotróficos, termodúricos e esporulados no leite cru produzido na região Norte do Paraná e verificar a capacidade proteolítica e lipolítica desses microrganismos. Foram analisadas 20 amostras de leite cru e determinadas contagens de aeróbios mesófilos, psicrotróficos, termodúricos mesófilos, termodúricos psicrotróficos e esporos aeróbios e o perfil proteolítico e lipolítico de cada grupo. A contagem média de aeróbios mesófilos foi de 1,7x106UFC/mL acima do limite estabelecido pela Instrução Normativa 76 (3,0x105UFC/mL); 55% das amostras apresentaram contagens dentro dos padrões. As contagens de psicrotróficos variaram de 1,0x102 a 9,9x106UFC/mL e em 30% das amostras esse valor foi superior ao de mesófilos. A média de termodúricos mesófilos foi de 2,4x104UFC/mL, de termodúricos psicrotróficos 1,7x102UFC/mL e de esporos aeróbios foi de 5,0x101UFC/mL. Quanto ao perfil deteriorante, todos os grupos apresentaram atividade lipolítica e proteolítica elevadas, com predomínio de bactérias lipolíticas. Os valores encontrados indicam que o manejo higiênico-sanitário inadequado durante a ordenha e o resfriamento prolongado favorece a proliferação da microbiota deteriorante, que pode promover alterações significativas na qualidade e no rendimento do leite e seus derivados, mesmo após tratamento térmico.


Considering that technological problems and economic losses are basically caused by spoilage microorganisms on dairy industry, therefore the aim of this study was to quantify the mesophilic aerobic, psychrotrophic, thermoduric and aerobic spore-forming in raw milk produced in northern Paraná and also verify the proteolytic and lipolytic capacity of these microorganisms. Twenty samples of raw milk were analyzed and were determined mesophilic aerobic, psychrotrophic, thermoduric mesophilic, psychrotrophic thermoduric and aerobicspore-forming counts and proteolytic and lipolytic capacity from each group. The average mesophilic thermoduric was 2,4x104CFU/mL, psychrotrophic thermoduric was 1,7x102CFU/mL and aerobic spore-forming was 5,0x101CFU/mL. As for the spoilage profile, all groups showed higher lipase and proteolytic activity, with a predominance of lipolytic bacteria. The values found indicates the inappropriate hygienic-sanitary management during milking and maintaining molk refrigerated for long periods until pasteurize, it can favorsthe proliferation of spoilage microbiota, which can promote significant changes in the quality and yield of milk and dairy products, even after the heat treatment.

The objectives of our present study included the screening of single nucleotide polymorphisms (SNP) that show significant differences in allelic frequencies between two buffalo populations (Egyptian and Chinese buffaloes), categorization of functional genes associated with these SNP by gene ontology, and pathway analyses to further understand their potential values as candidate genes closely associated with milk yield trait in buffaloes. In this study, double digest restrictionsite associated DNA sequencing was performed on Illumina HiSeq 2500 platform for 20 and 25 female buffaloes from Egypt and China, respectively. Approximately 118 Gb of sequencing data were obtained, and a total of 110,129 and 150,535 putative SNP were detected in Egyptian and Chinese buffaloes, respectively. Focused only on those SNP that differed significantly in allelic frequencies between the two populations, we found that genes associated with these SNP were significantly over-represented in the ionotropic glutamate receptor pathway, the endothelin signaling pathway, and the gonadotropin-releasing hormone receptor pathway, which contained a total of 29 genes. Of these, nine genes (ADCY5, CACNA1A, CREB1, INHBA, INHBB, PIK3R1, PLCB1, PRKCE, and SMAD2) participating in the hormonal regulation of lactation, were considered to be promising candidate genes worthy of further investigations for favorable alleles associated with milk yield. Our results provide useful information about genetic variations in Egyptian and Chinese buffaloes. The potential influences of nine candidate genes and their associated SNP on milk yield need to be validated in more buffalo populations.(AU)

Two experiments were carried out in a 2 x 2 factorial arrangement with the objective of evaluating two methods of concentrate feeding for Holstein x Gyr cows fed 600 g kg-1 chopped elephantgrass-based diets supplemented at 45 g kg-1 DM with two types of sunflower oil (SO). The types of SO differed in the levels of oleic and linoleic fatty acids (FAs): high oleic/low linoleic acid – HO (73 and 10 g 100 g-1 FA, respectively) and medium oleic/medium linoleic acid – MO (43 and 34 g 100 g-1 FA, respectively). The concentrates containing HO SO or MO SO were supplied separately from the forage twice a day after the two milkings (TAD) or as part of a total mixed ration (TMR). In Experiment 1, a 4 x 4 Latin square design was used to evaluate the ruminal fermentation and degradation parameters in four rumen-cannulated cows (430±39 kg; 79±20 days in milk; 16.4±3.1 kg day-1 of milk). In Experiment 2, a randomized block design was used to evaluate the nutrient intake, plasma contents of metabolites and FAs, milk yield and composition, and FA profile of milk fat in 32 cows (444±84 kg; 75±31 days in milk; 15.4±4.8 kg day-1 of milk). The results were analyzed by mixed models (P < 0.05). The TMR diets promoted higher nutrient intake and rumen fermentation (higher ammonia N, acetate, propionate and total volatile FA contents) without affecting milk, fat, protein and lactose yields. TAD-fed cows presented higher feed efficiency and produced milk fat with a more nutritionally desirable FA composition, with higher vaccenic and rumenic acid contents and lower trans-10 C18:1 and palmitic acid contents. The DM intake, parameters of rumen fermentation and milk, fat, protein and lactose yields were similar for the HO SO and MO SO diets. The most nutritionally positive characteristics for human health in the milk fat of HO SO-fed cows were the higher eicosapentaenoic (+34%) and oleic acid (+11%) contents and lower palmitic acid content (-10%). Higher contents of vaccenic (+71%) and rumenic (+74%) acids and lower trans-10 C18:1 (-10%), elaidic (-32%), lauric (-14%) and myristic (-11%) acid contents were the most positive aspects of the milk fat of MO SO-fed cows. Considering the magnitudes of the differences in the levels of these FAs, it is concluded that the milk fat of cows fed MO SO showed a healthier milk FA profile.


Dois experimentos foram realizados em esquema de arranjo fatorial 2 x 2, com o objetivo de avaliar dois métodos de fornecimento de concentrado para vacas Holandês x Gir recebendo dietas à base de 600 g kg-1 de capim-elefante picado, suplementadas com dois tipos de óleo de girassol (OG) na base de 45 g kg-1 da matéria seca (MS) da dieta. Os OG diferiram por apresentar teores contrastantes dos ácidos graxos (AG) oleico e linoleico: alto teor de ácido oleico/baixo teor de ácido linoleico – HO (respectivamente, 73 e 10 g 100 g-1 de AG) e teores médios dos ácidos oleico e linoleico – MO (respectivamente, 43 e 34 g 100 g-1 de AG). Os concentrados contendo os OG foram fornecidos separadamente da forragem, duas vezes ao dia, após as duas ordenhas (TAD), ou como parte de uma dieta completa (TMR). No experimento 1, utilizou-se delineamento quadrado latino 4 x 4 para avaliar a fermentação e degradação ruminal em quatro vacas canuladas no rúmen (430±39 kg; 79±20 dias em lactação; 16,4±3,1 kg dia-1 de leite). No experimento 2, foi utilizado delineamento em blocos casualizados para avaliar o consumo de nutrientes, as concentrações plasmáticas de metabólitos e AG, e a produção, composição e o perfil de AG do leite em 32 vacas (444±84 kg; 75±31 dias em lactação; 15,4±4,8 kg dia-1 de leite). Os resultados foram analisados por modelos mistos (P < 0,05). As dietas fornecidas como TMR promoveram maior ingestão de nutrientes e fermentação ruminal (maiores teores de N amoniacal, acetato, propionato e AG voláteis totais), mas sem efeito nas produções de leite, gordura, proteína e lactose. As vacas que receberam concentrado duas vezes ao dia apresentaram maior eficiência alimentar e produziram leite com perfil de AG mais saudável, com maiores teores dos ácidos vacênico e rumênico, e menores de C18:1 trans-10 e ácido palmítico. O tipo de OG não influenciou a ingestão de MS, a fermentação ruminal e as produções de leite, gordura, proteína e lactose. As características nutricionais mais positivas na gordura do leite das vacas que receberam OG HO foram os maiores teores dos ácidos eicosapentaenoico (+34%) e oleico (+11%) e o menor teor de ácido palmítico (-10%). Maiores teores dos ácidos vacênico (+71%) e rumênico (+74%) e menores dos ácidos elaídico (-32%), láurico (-14%), mirístico (-11%) e C18:1 trans-10 (-10%) foram os aspectos nutricionais mais positivos na gordura do leite das vacas alimentadas com OG MO. Considerando as magnitudes das diferenças nos teores desses AG, conclui-se que a gordura do leite das vacas que receberam o OG MO apresentou perfil de AG mais saudável.

Two experiments were carried out in a 2 x 2 factorial arrangement with the objective of evaluating two methods of concentrate feeding for Holstein x Gyr cows fed 600 g kg-1 chopped elephantgrass-based diets supplemented at 45 g kg-1 DM with two types of sunflower oil (SO). The types of SO differed in the levels of oleic and linoleic fatty acids (FAs): high oleic/low linoleic acid HO (73 and 10 g 100 g-1 FA, respectively) and medium oleic/medium linoleic acid MO (43 and 34 g 100 g-1 FA, respectively). The concentrates containing HO SO or MO SO were supplied separately from the forage twice a day after the two milkings (TAD) or as part of a total mixed ration (TMR). In Experiment 1, a 4 x 4 Latin square design was used to evaluate the ruminal fermentation and degradation parameters in four rumen-cannulated cows (430±39 kg; 79±20 days in milk; 16.4±3.1 kg day-1 of milk). In Experiment 2, a randomized block design was used to evaluate the nutrient intake, plasma contents of metabolites and FAs, milk yield and composition, and FA profile of milk fat in 32 cows (444±84 kg; 75±31 days in milk; 15.4±4.8 kg day-1 of milk). The results were analyzed by mixed models (P < 0.05). The TMR diets promoted higher nutrient intake and rumen fermentation (higher ammonia N, acetate, propionate and total volatile FA contents) without affecting milk, fat, protein and lactose yields. TAD-fed cows presented higher feed efficiency and produced milk fat with a more nutritionally desirable FA composition, with higher vaccenic and rumenic acid contents and lower trans-10 C18:1 and palmitic acid contents. The DM intake, parameters of rumen fermentation and milk, fat, protein and lactose yields were similar for the HO SO and MO SO diets. The most nutritionally positive characteristics for human health in the milk fat of HO SO-fed cows were the higher eicosapentaenoic (+34%) and oleic acid (+11%) contents and lower palmitic acid content (-10%). Higher contents of vaccenic (+71%) and rumenic (+74%) acids and lower trans-10 C18:1 (-10%), elaidic (-32%), lauric (-14%) and myristic (-11%) acid contents were the most positive aspects of the milk fat of MO SO-fed cows. Considering the magnitudes of the differences in the levels of these FAs, it is concluded that the milk fat of cows fed MO SO showed a healthier milk FA profile.(AU)


Dois experimentos foram realizados em esquema de arranjo fatorial 2 x 2, com o objetivo de avaliar dois métodos de fornecimento de concentrado para vacas Holandês x Gir recebendo dietas à base de 600 g kg-1 de capim-elefante picado, suplementadas com dois tipos de óleo de girassol (OG) na base de 45 g kg-1 da matéria seca (MS) da dieta. Os OG diferiram por apresentar teores contrastantes dos ácidos graxos (AG) oleico e linoleico: alto teor de ácido oleico/baixo teor de ácido linoleico HO (respectivamente, 73 e 10 g 100 g-1 de AG) e teores médios dos ácidos oleico e linoleico MO (respectivamente, 43 e 34 g 100 g-1 de AG). Os concentrados contendo os OG foram fornecidos separadamente da forragem, duas vezes ao dia, após as duas ordenhas (TAD), ou como parte de uma dieta completa (TMR). No experimento 1, utilizou-se delineamento quadrado latino 4 x 4 para avaliar a fermentação e degradação ruminal em quatro vacas canuladas no rúmen (430±39 kg; 79±20 dias em lactação; 16,4±3,1 kg dia-1 de leite). No experimento 2, foi utilizado delineamento em blocos casualizados para avaliar o consumo de nutrientes, as concentrações plasmáticas de metabólitos e AG, e a produção, composição e o perfil de AG do leite em 32 vacas (444±84 kg; 75±31 dias em lactação; 15,4±4,8 kg dia-1 de leite). Os resultados foram analisados por modelos mistos (P < 0,05). As dietas fornecidas como TMR promoveram maior ingestão de nutrientes e fermentação ruminal (maiores teores de N amoniacal, acetato, propionato e AG voláteis totais), mas sem efeito nas produções de leite, gordura, proteína e lactose. As vacas que receberam concentrado duas vezes ao dia apresentaram maior eficiência alimentar e produziram leite com perfil de AG mais saudável, com maiores teores dos ácidos vacênico e rumênico, e menores de C18:1 trans-10 e ácido palmítico. O tipo de OG não influenciou a ingestão de MS, a fermentação ruminal e as produções de leite, gordura, proteína e lactose. As características nutricionais mais positivas na gordura do leite das vacas que receberam OG HO foram os maiores teores dos ácidos eicosapentaenoico (+34%) e oleico (+11%) e o menor teor de ácido palmítico (-10%). Maiores teores dos ácidos vacênico (+71%) e rumênico (+74%) e menores dos ácidos elaídico (-32%), láurico (-14%), mirístico (-11%) e C18:1 trans-10 (-10%) foram os aspectos nutricionais mais positivos na gordura do leite das vacas alimentadas com OG MO. Considerando as magnitudes das diferenças nos teores desses AG, conclui-se que a gordura do leite das vacas que receberam o OG MO apresentou perfil de AG mais saudável.(AU)
