Resumo

This study examines the in vitro growth and ex vitro establishment of Brassavola tuberculata in relation to the micropropagation system and sucrose concentration employed in the in vitro culture. A completely randomized experimental design was utilized, employing a 2 x 5 factorial arrangement. The experimental period began with seedlings cultivated in vitro for 180 days, which were subsequently transferred to Murashige and Skoog culture media containing sucrose concentrations of 0, 15, 30, 45, or 60 g L-1. The cultures were subjected to two micropropagation systems: conventional and gas exchange. After 90 days of in vitro cultivation, the plants were evaluated, transplanted into a substrate, and placed in a screened nursery for ex vitro cultivation. After 300 days of ex vitro cultivation, the survival and initial characteristics of the plants were assessed. The micropropagation system allowing gas exchange and sucrose concentrations up to 30 g L-1 enhanced the shoot and root growth of in vitro propagated plants. No noticeable anatomical differences were observed after 90 days of in vitro culture among the different sucrose concentrations and micropropagation systems used. In the ex vitro establishment, irrespective of sucrose concentration, the micropropagation system facilitating gas exchange positively influenced all evaluated characteristics.


Objetivou-se com este trabalho avaliar o crescimento in vitro e estabelecimento ex vitro de Brassavola tuberculata em função do sistema de micropropagação e da concentração de sacarose utilizados no cultivo in vitro. Foi utilizado o delineamento inteiramente casualizado e os tratamentos arranjados em esquema fatorial 2 x 5. Para o início do período experimental, foram utilizadas plântulas cultivadas in vitro por 180 dias, sendo transferidas para meios de cultivo Murashige e Skoog contendo 0, 15, 30, 45 ou 60 g L-1 de sacarose, e as culturas submetidas a dois sistemas de micropropagação: convencional ou com troca gasosa. Após 90 dias de cultivo in vitro, as plantas foram avaliadas e na sequência plantadas em substrato e acondicionadas em viveiro telado para o cultivo ex vitro. Após 300 dias de cultivo ex vitro, as plantas foram avaliadas quanto à sobrevivência e às mesmas características iniciais. A utilização do sistema de micropropagação que permite trocas gasosas, em conjunto com concentrações de sacarose de até 30 g L-1, proporcionou aumento no crescimento de parte aérea e do sistema radicular das plantas propagadas in vitro. As diferentes concentrações de sacarose e os sistemas de micropropagação utilizados não apresentaram diferenças anatômicas perceptíveis aos 90 dias de cultivo in vitro. Já no estabelecimento ex vitro, independente da utilização de sacarose, o sistema de micropropagação que permite trocas gasosas influenciou positivamente todas as características avaliadas.

Resumo

Anaerobic soil disinfestation (ASD) is an ecological alternative to chemical soil fumigation. However, little is known about the potential of this technique for the management of Meloidogyne javanica and Stromatinia cepivora. To test the hypothesis that ASD reduces the viability of these two pathogens, we assessed ethanol (5%, v:v) and sucrose (5%, m:v) as carbon sources for ASD, for an incubation period of three weeks. Twenty kilograms of soil naturally infested with M. javanica (82 ± 43 J2 100 cm-3 soil) were placed into a plastic container. Polyester traps, each with 15 S. cepivora sclerotia, were buried at 10 and 20 cm depth per container. ASD with diluted ethanol or sucrose (5% v:v or m:v) was compared to the untreated control (UTC), chemical fumigant metam sodium (MS), and soil saturation with water. In comparison to the UTC, ASD using ethanol reduced the numbers of J2 in soil and the galls in tomato roots by more than 93%, a degree of suppression similar to that achieved when using MS. The viability of sclerotia of S. cepivora was reduced by ASD using ethanol or sucrose from 38.12 to 58.1% compared to the UTC. ASD for three weeks using ethanol or sucrose (5%) reduces the viability of M. javanica and S. cepivora in the microcosm.(AU)

Consequently, several literatures are rife with ancient uses of honey as complementary and alternative medicine, with relevance to modern day health care, supported by evidence-based clinical data, with little attention given to honeys nutritional functions. The moisture contents of honey extracted from University of Veterinary and Animal Sciences, Lahore honey bee farm was 12.19% while that of natural source was 9.03 ± 1.63%. Similarly, ash and protein contents of farmed honey recorded were 0.37% and 5.22%, respectively. Whereas ash and protein contents of natural honey were 1.70 ± 1.98% and 6.10 ± 0.79%. Likewise fat, dietary fiber and carbohydrates contents of farmed source documented were 0.14%, 1.99% and 62.26% respectively. Although fat, dietary fiber and carbohydrates contents of honey taken from natural resource were 0.54 ± 0.28%, 2.76 ± 1.07% and 55.32 ± 2.91% respectively. Glucose and fructose contents of honey taken out from honeybee farm were 27% and 34% but natural source were 22.50 ± 2.12% and 28.50 ± 3.54%. Glucose and fructose contents of honey taken out from honeybee farm were 27% and 34% but natural source were 22.50 ± 2.12% and 28.50 ± 3.54%. Similarly, sucrose and maltose contents of farmed honey were 2.5% and 12% while in natural honey were 1.35 ± 0.49% and 8.00 ± 1.41% respectively. The present study indicates that such as moisture, carbohydrates, sucrose and maltose contents were higher farmed honey as compared to the natural honey. In our recommendation natural honey is better than farmed honey.

Consequentemente, várias literaturas estão repletas de usos antigos do mel como medicina complementar e alternativa, com relevância para os cuidados de saúde modernos, apoiados por dados clínicos baseados em evidências, com pouca atenção dada às funções nutricionais do mel. O teor de umidade do mel extraído da Universidade de Veterinária e Ciências Animais, fazenda de abelhas de Lahore, foi de 12,19%, enquanto o de fonte natural foi de 9,03 ± 1,63%. Da mesma forma, os teores de cinzas e proteínas do mel cultivado foram de 0,37% e 5,22%, respectivamente. Já os teores de cinzas e proteínas do mel natural foram de 1,70 ± 1,98% e 6,10 ± 0,79%. Resumo

This study was aimed to assess the efficiency of coconut water extender with addition of soy lecithin and sucrose as nonpermeable cryoprotectants for canine semen vitrification, using a simple method that yields a high survival rate of spermatozoa for clinical use. Twelve ejaculates from 12 adult normozoospermic dogs were collected separately by digital manipulation and only the second semen fraction was used in this study. After evaluation of volume, concentration, viability, total and progressive motility, velocity parameters and morphology, semen was diluted with a coconut water extender (50% (v/v(volume per volume)) coconut water, 25% (v/v) distilled water and 25% (v/v) 5% anhydrous monosodium citrate solution) with addition of soy lecithin and fructose at 1% and 0.25M sucrose until final concentration of 100x106 spermatozoa/ml. After equilibration at 5ºC for 60 minutes, semen was vitrified by "direct dropping method" into liquid nitrogen in spheres with a volume of 30 µl. After a week of storage the spheres were devitrified as three of them were dropped into 0.5 mL of CaniPlus AI medium (Minitüb, Germany), which was previously warmed in a water bath at 42ºC for 2 minutes and evaluated about the above mentioned parameters. It was found that vitrification resulted in a lower percentage of viable sperms, normal morphology, total and progressive motilities (p0.05) compared to fresh semen samples. In conclusion, our results demonstrate that vitrification with coconut water extender with addition of 1% soy lecithin and 0.25M sucrose as cryoprotectants, has an excellent potential for routine canine sperm cryopreservation.(AU)

Resumo

The identification of promising commercial varieties for irrigated crops in semiarid conditions based on characters of interest to sugarcane agro-industries is important, considering the search for high sugarcane yields to obtain sustainable crops. Thus, the objective of this work was to evaluate production components of agroindustrial interest of different sugarcane varieties grown under subsurface drip irrigation in the Semiarid region of Brazil, during two crop cycles. A randomized block experimental design was used, in an 8×2 factorial arrangement (varieties and cycles) with three replications. The sugarcane juice quality, sugar yield, and culm yield were evaluated in each cycle. Gross sugar percentage, sugarcane culm yield, juice apparent sucrose content, and juice purity were affected by the crop cycles. Soluble solids content presented strong significant correlation with most variables of agroindustrial interest. The path coefficient analysis showed that juice apparent sucrose content was the explanatory character with higher direct effect on soluble solids and that this variable presented high indirect effects on most explanatory variables. The sugarcane varieties RB72454 and Q124 are recommended for production managements with drip irrigation under semiarid conditions. Soluble solids content can be used as an alternative for direct selection of varieties with higher sugar yields in different cycles.(AU)

Microbial contamination of the wort during the fermentation process causes significant losses in ethanol production worldwide and creates a dependence of the industry on chemicals and antibiotics to control contamination. Therefore, this study used electron beam (e-beam) to disinfect wort from sugarcane (Saccharum officinarum L.) molasses and investigate the bioethanol fermentation. Four treatments (T0 - T3) were carried out using ionizing doses of radiation through the electron accelerator: 0 (control), 10, 20, and 40 kGy. Total mesophiles, total bacteria, sucrose, glucose, fructose, phenolics, flavonoids, hydroxymethylfurfural (5-HMF), and Furfural were measured. An alcoholic fermentation assay was performed after the irradiation process. The irradiated treatments showed no inversion of sugars and formation of the inhibitory by-products flavonoids, furfural and 5-HMF, except for the phenolic compounds. The lower dose tested (10 kGy) reduced more than 99.9 % of the total mesophiles and more than 99.99 % of the total bacteria in the substrate. In the fermentation, the irradiated worts presented similar (p > 0.05) yields (92, 93, and 94 %) and ethanol productivity levels (0.89, 0.88, and 0.87 g L-1 h-1, for T1, T2, and T3 respectively). However, all treatments presented higher yields and productivity (p < 0.05) when compared to the control (88 % and 0.85 g L-1 h-1), highlighting the possible use of e-beam in wort fermentation at a lower dose (10 kGy). This allows reduction in losses caused by microbial contamination, besides increasing fermentation yield and productivity with lower energy consumption.(AU)

Meat products represent an important component of the human diet and are a good source of nutrients. Food-borne microorganisms are the main pathogens that cause human diseases as a result of food consumption, especially products of animal origin. The objective of the present research was to verify the antibacterial activity of the essential oil of Thymus vulgaris against strains of Klebsiella pneumoniae, Pseudomonas aeruginosa and Staphylococcus saprophyticus isolated from meat products. For this, the analyses of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were performed in microdilution plates. The association of the product with antimicrobials was also studied using disk diffusion. And the anti-adherent activity, which was determined in the presence of sucrose, in glass tubes. Thyme oil showed a strong inhibitory activity against K. pneumoniae, P. aeruginosa and S. saprophyticus, with the MIC values ranging from 64 to 512 µg/mL, and bactericidal effect for most strains, with MBC values ranging from 256 to 1,024 µg/mL. T. vulgaris oil exhibited varied interactions in association with the antimicrobials, with synergistic (41.67%), indifferent (50%) and antagonistic (8.33%) effects. Regarding the anti-adherent activity, the test product was effective in inhibiting the adherence of all bacterial strains under study. Therefore, thyme oil presents itself as an antibacterial and anti-adherent agent against K. pneumoniae, P. aeruginosa and S. saprophyticus, being a natural product that can represent an interesting alternative in the efforts to combat foodborne diseases.


Os produtos cárneos representam um importante componente da dieta humana e constituem uma boa fonte de nutrientes. Microrganismos de origem alimentar são os principais patógenos que causam doenças humanas como resultado do consumo de alimentos, principalmente, produtos de origem animal. O objetivo da presente pesquisa foi verificar a atividade antibacteriana do óleo essencial de Thymus vulgaris frente às cepas de Klebsiella pneumoniae, Pseudomonas aeruginosa e Staphylococcus saprophyticus isoladas de produtos cárneos. Para isso, foram realizadas as análises de Concentração Inibitória Mínima (CIM) e a Concentração Bactericida Mínima (CBM) em placas de microdiluição. Assim como, o estudo de associação do produto com antimicrobianos, realizado por difusão em disco. E a atividade antiaderente, que foi determinada na presença de sacarose, em tubos de vidro. O óleo de tomilho apresentou uma forte atividade inibitória contra K. pneumoniae, P. aeruginosa e S. saprophyticus, com os valores de CIM variando entre 64 a 512 µg/mL, e efeito bactericida para a maioria das cepas, com valores de CBM entre 256 a 1.024 µg/mL. O óleo de T. vulgaris exibiu interações variadas na associação com os antimicrobianos, com efeitos sinérgicos (41,67%), indiferente (50%) e antagonista (8,33%). Em relação a atividade antiaderente, o produto teste foi eficaz na inibição a aderência de todas cepas bacterianas em estudo. Portanto, o óleo de tomilho apresenta-se como agente antibacteriano e antiaderente frente a K. pneumoniae, a P. aeruginosa e a S. saprophyticus, sendo um produto natural que pode representar uma alternativa interessante nos esforços para combater doenças transmitidas por alimentos.

The objectives of this work were to investigate the occurrence of parasites in feces of cats, compare different coproparasitological techniques for their diagnosis and determine associations with parasitism. The samples were processed using three different coproparasitological techniques: centrifugal flotation in sucrose, centrifugal flotation in ZnSO4 and simple sedimentation. The parasitic association between parasitism and variables such as age, sex and fecal consistency was performed using the chi-square test or the G test with a significance level of 5%. A total of 237 samples were analyzed, of which 93 (39.2%) were positive, being Ancylostoma spp. (17.3%), Giardia intestinalis (12.2%), Platynosomum illiciens (8.0%), Cystoisospora spp. (6.3%), Toxoplasma gondii/ Hammondia hammondi (3.4%), Diphyllobothriidae (2.1%), Toxocara spp. (1.7%), Dipylidium caninum (1.3%) and Mesocestoides spp. (0.8%). In the parasitism association analysis, it was possible to verify a statistical difference in the age category for Cystoisospora spp. (p=0.001) observing a strong relationship between parasitism and young animals, the association with sex proved to be important for P. illiciens (p<0.001) with a higher frequency of parasitized females and fecal consistency revealed to be related to the parasites G. intestinalis (p=0.007) and P. illiciens (p=0.033) showing a higher number of positive animals for these parasites with normal fecal consistency. In conclusion, we observed a higher occurrence of Ancylostoma spp. and G. intestinalis in fecal samples from domestic cats received in routine diagnoses and the presence of other parasites with zoonotic potential, as well as the relationship of these diagnosed parasites with the categories sex, age and fecal consistency.(AU)


Os objetivos deste trabalho foram investigar a ocorrência de parasitos em fezes de gatos, comparar diferentes técnicas coproparasitológicas para o seu diagnóstico e determinar as associações com o parasitismo. As amostras foram processadas usando três diferentes técnicas coproparasitológicas: centrífugo-flutuação em sacarose, centrífugo-flutuação em ZnSO4 e sedimentação simples. A associação parasitária entre o parasitismo e variáveis, como idade, sexo e consistência fecal, foi realizada pelo teste de qui-quadrado ou teste de G com nível de significância de 5%. No total, foram analisadas 237 amostras, das quais 93 (39,2%) foram positivas, sendo diagnosticados os parasitos Ancylostoma spp. (17,3%), Giardia intestinalis (12,2%), Platynosomum illiciens (8,0%), Cystoisospora spp. (6,3%), Toxoplasma gondii/ Hammondia hammondi (3,4%), Diphyllobothriidae (2,1%), Toxocara spp. (1,7%), Dipylidium caninum (1,3%) e Mesocestoides spp. (0,8%). Na análise de associação entre o parasitismo e as outras variáveis, foi possível verificar diferença estatística na categoria idade para Cystoisospora spp. (p=0,001), observando forte relação entre o parasitismo e animais jovens. A associação com o sexo demonstrou ser importante para P. illiciens (p<0,001), com maior frequência de fêmeas parasitadas. E a consistência fecal revelou ter relação com os parasitos G. intestinalis (p=0,007) e P. illiciens (p=0,033), com maior frequência de fezes com consistência normal nos animais positivos para esses parasitos. Em conclusão, foi observada uma maior ocorrência de Ancylostoma spp. e G. intestinalis em amostras fecais de gatos domésticos, recebidas em diagnósticos de rotina e a presença de outros parasitos com potencial zoonótico, como também, a relação desses parasitos diagnosticados com as categorias sexo, idade e consistência fecal.(AU)

In a randomized controlled trial, we evaluated the effects of five oral electrolyte solutions (OESS) with different compositions of water, electrolyte, and acid-base balances of diarrheal neonatal calves. Osmotic diarrhea and dehydration were induced with sucrose in milk, spironolactone, and hydrochlorothiazide for 48 h in thirty 10-day old Holstein healthy calves with 43.5 ± 3.80 kg BW who were fed with natural milk. They were allocated to five treatment groups (n=6) based on the administered OES (commercial: OES A, B, C, D; and non-commercial: OES UEL). On the day of treatment, the calves received 6L of OES in two doses apart from milk intakes. Venous blood samples were collected at -48h (start of induction), -24h, Oh (start of the treatment day), 8h, 16h, 24h, 48h, and 72h. TPP, glucose, D-lactate, L-lactate, pH, pCO2, HCO3, BE, Na, K, CI, SID3, SIG, AG, Atot and percentage change in plasma volume (%PV) were measured or calculated and analyzed by two-way repeated measures ANOVA. All the calves developed osmotic diarrhea, mild to moderate dehydration, hyponatremia, relative hyperchloremia, and moderate to severe metabolic acidosis. The tested OESS were well accepted by the calves and effective in reversing dehydration, electrolyte imbalances, and metabolic acidosis. OES D did not completely correct hyponatremia, and SEO B caused transient hyperglycemia. It has been concluded that all the tested OESS are safe and effective for the treatment of diarrhea in calves with moderate degrees of dehydration and metabolic acidosis, which indicates that they have proper compositions for this purpose.


Em um ensaio clínico controlado e aleatório, foram comparados os efeitos de cinco soluções eletrolíticas orais (SEOs) sobre os equilíbrios hídrico, eletrolítico e ácido-base de bezerros neonatos diarreicos. Diarreia osmótica e desidratação foram induzidas com sacarose no leite, espironolactona e hidroclorotiazida, por 48h, em 30 bezerros Holandeses com 10 dias de idade, 43,5 ± 3,8kg de peso, e alimentados com leite natural. Eles foram distribuídos em cinco grupos de tratamento (n=6) de acordo com a SEO administrada (comercial: SEO A, B, C, D; e não comercial: SEO UEL). No dia do tratamento, os bezerros receberam 6L de SEO em duas doses além da ingestão de leite. Amostras de sangue venoso foram coletadas em -48h (início da indução), - 24h, Oh (início do dia de tratamento), 8h, 16h, 24h, 48h e 72h. PPT, glicose, lactatos Le D, pH, pCO2, HCO3, BE, Na, K, CI, SID3, SIG, AG, Atot e variação percentual no volume plasmático (%VP) foram mensurados ou calculados, e analisados por meio de ANOVA de duas vias de medidas repetidas. Todos os bezerros desenvolveram diarreia osmótica, desidratação leve a moderada, hiponatremia, hipercloremia relativa e acidose metabólica moderada a grave. As SEOs testadas foram bem aceitas pelos bezerros e eficazes para a reversão da desidratação, dos desequilíbrios eletrolíticos e da acidose metabólica. A SEO D não corrigiu completamente a hiponatremia, e a SEO B causou hiperglicemia transitória. Conclui-se que todas as SEOs são eficazes para o tratamento da diarreia em bezerros com graus moderados de desidratação e de acidose metabólica, o que indica que possuem composições adequadas para esse fim.

Abstract The action of substances with non-permeable cryoprotectant potential, besides glucose, has not yet been studied for the species Prochilodus brevis. The objective of this work was to evaluate the action of four non-permeable cryoprotectants on this species sperm cryopreservation. Five pools were cryopreserved in a solution of 5% glucose and 10% dimethyl sulfoxide (Me2SO) associated or not (control) with cryoprotectants egg yolk (5, 10 or 12%), soy lecithin (2.5, 7.5 or 10%), sucrose (5, 10 or 20%) and lactose (5, 8 or 15%). After thawing, samples were evaluated for sperm kinetics (total motility, motility duration, velocities, and wobble - WOB), morphology and membrane and DNA integrity. The treatments containing egg yolk improved significantly (P<0.05) results when compared the control for the membrane integrity parameter. When compared to other treatments, egg yolk, at any concentration, presented higher results (P<0.05) for membrane integrity, total motility, curvilinear velocity (VCL) and average path velocity (VAP) parameters. Egg yolk also showed the best results for WOB, but it did not differ from 5% and 8% lactose and 5% and 20% sucrose. Soy lecithin had the lowest percentages of morphologically normal sperm (P<0.05), while the other treatments did not differ from each other. There was no difference regarding DNA integrity data. Thus, 5% egg yolk is indicated as a non-permeable cryoprotectant for P. brevis, in association with 5% glucose and 10% Me2SO.

Brazil is a large country with high biodiversity in its different regions. However, species of native fruits widely reported in the southern Brazil have not been properly explored so far, remaining underutilized by the food industry. This study evaluated the polyphenolic profile, as well as the composition of organic acids and sugars of the pulps of the feijoa, and the uvaia from southern brazilian highlands. The uvaia pulp showed the highest total polyphenol content and the highest antioxidant capacity by the methods used. The polyphenol (+)-catechin (6.54 mg 100g-1) was the major phenolic compound in uvaia pulp, which has not yet been reported in the literature for fruits of other regions. In addition, the feijoa pulp stood out for the presence of (-)-epicatechin (18.29 mg 100g-1). The pulps of native fruits in this study only quantified values for citric and malic acids. Malic acid (553.00 mg 100g-1) was the main acid in the uvaia pulp, and citric acid (455.60 mg 100g-1) was the main acid in the feijoa pulp. It is possible to note that the feijoa pulp showed the highest total sugar content (11.14 g 100g-1) and was the only pulp that contained sucrose. The uvaia pulp, conversely, showed fructose (3.10 g 100g-1) as the main sugar. The results obtained in this study contributed to the valuation and conservation of the species investigated, representing a promising alternative for the use of these native fruits in the development of new products.


O Brasil é um grande país e com elevada biodiversidade em suas diferentes regiões. Entretanto, espécies de frutas nativas amplamente encontradas no sul do Brasil ainda não foram devidamente exploradas, permanecendo desconhecidas e subutilizadas pela indústria de alimentos. Nesse sentido, o objetivo deste estudo foi avaliar o perfil fenólico, bem como a composição de ácidos orgânicos e de açúcares de polpas de feijoa (Acca sellowiana) e de uvaia (Eugenia pyriformis) provenientes do planalto sul brasileiro. A polpa de uvaia apresentou o maior teor de polifenóis totais e a maior capacidade antioxidante pelos métodos utilizados. A (+)-catequina (6.54 mg 100g-1) foi o principal composto fenólico da polpa da uvaia, o que ainda não foi relatado na literatura para frutas de outras regiões. Além disso, a polpa da feijoa se destacou pela presença de (-)-epicatequina (18.29 mg 100g-1). Dentre os ácidos orgânicos avaliados, as polpas de frutas nativas apresentaram valores para os ácidos cítrico e málico, sendo o ácido málico (553.00 mg 100g-1) o principal na polpa de uvaia, e o cítrico (455.60 mg 100g-1) na polpa de feijoa. Em relação aos açúcares, a polpa da feijoa apresentou o maior teor de açúcar total (11.14 g 100g-1) e foi a única polpa que continha sacarose. Já a polpa da uvaia apresentou a frutose (3.10 g 100g-1) como principal açúcar. Os resultados obtidos neste estudo contribuem para a valorização e conservação das espécies investigadas, representando uma alternativa promissora para a utilização dessas frutas nativas no desenvolvimento de novos produtos.

The "sempre-viva" species Comanthera mucugensis is endemic to the municipality of Mucugê, Bahia, Brazil, where it was widely exploited through extractivism to be commercialized as an ornamental, causing a drastic reduction of its population, so that it is now classified as endangered. Although its main area of occurrence is now protected for being within Chapada Diamantina National Park, the continued risks of inclement conditions and anthropic actions make it necessary to develop alternative methods for ex situ conservation of the species, such as in vitro conservation. Therefore, the objective of this study was to test the effect of different concentrations of salts and sucrose on the in vitro conservation of Comanthera mucugensis. Two salts concentrations of the medium MS (½ and ») and two sucrose levels (7.5 and 15.0 g L-1) were tested, and the experimental design was completely randomized with four treatments. After 365 days, the survival, growth and regeneration of the conserved plants were analyzed, achieving up to 100% survival, reduced shoot growth and maintenance of regenerative capacity. Reduction of the concentration of salts and sucrose in the culture medium is indicated to conserve the plants in vitro for a period of one year.(AU)

Sucrose- and diuretics-based protocols are widely used to induce osmotic diarrhea and dehydration in calves, but they fail to cause metabolic acidosis. In previous studies, calves were fed milk replacers and deprived of water. In this study, we assessed the water, electrolyte, and acid-base imbalances in calves that were fed whole milk and were not completely deprived of water during the induction period. Healthy, male Holstein calves aged 10-12 days were assigned to two groups: free access to water (FWG; n=17) and water deprivation at night (DWG; n=21); and osmotic diarrhea was induced with sucrose added to milk, spironolactone (2mg kg-1) and hydrochlorothiazide (2mg kg-1) orally every 8h for 48h. pH, pCO2, HCO3-, BE, Na+, K+, Cl-, SID3, TPP, AG, Atot, glucose, L-lactate, D-lactate, SIG, and percentage change in plasma volume were measured in venous blood samples taken at 0, 24, and 48h. Data were analyzed using two-way repeated measures ANOVA. Calves showed diarrhea, mild (FWG) to moderate (DWG) dehydration, hyponatremia, and moderate (FWG) to severe (DWG) metabolic acidosis. AG and D-lactate levels were higher and SIG was lower in the DWG, and there was no hyper-L- or D-lactatemia. The magnitude of metabolic acidosis was similar to that observed in natural cases of diarrhea. The protocol for inducing osmotic diarrhea and dehydration should be applied to calves that are fed whole milk and are not completely deprived of water.


Protocolos baseados em sacarose e diuréticos são amplamente usados para induzir diarreia osmótica e desidratação em bezerros, porém não provocam acidose metabólica. Em estudos anteriores, os bezerros foram alimentados com substitutos de leite e privados de água. Neste estudo, avaliaram-se os desequilíbrios hídrico, eletrolítico e ácido base em bezerros que foram alimentados com leite integral e não foram completamente privados de água durante o período de indução. Bezerros HPB machos sadios com 10 a 12 dias de idade foram distribuídos por dois grupos: acesso livre à água (FWG; n=17) e privação de água durante a noite (DWG; n=21); e a diarreia osmótica foi induzida com sacarose adicionada ao leite, espironolactona (2mg kg-1) e hidroclorotiazida (2mg kg-1) por via oral a cada 8h durante 48h. pH, pCO2, HCO3-, BE, Na+, K+, Cl-, SID3, PPT, AG, Atot, glicose, lactato L, lactato D, SIG e alteração percentual no volume plasmático foram medidos em amostras de sangue venoso colhidas em 0, 24 e 48h. Os dados foram analisados usando ANOVA de medidas repetidas bifatorial. Os bezerros apresentaram diarreia, desidratação leve (FWG) a moderada (DWG), hiponatremia e acidose metabólica moderada (FWG) a grave (DWG). Os valores de AG e de lactato D foram maiores e o de SIG foi menor no DWG, e não houve hiperlactatemia L ou D. A magnitude da acidose metabólica foi semelhante à observada em casos naturais de diarreia. O protocolo para induzir diarreia osmótica e desidratação deve ser aplicado a bezerros alimentados com leite integral e não totalmente privados de água.

The establishment of minimum growth conditions is essential for in vitro germplasm conservation. Changes to the basic medium and carbon source concentrations are important factors for reducing plant growth in vitro. This study adjusted a protocol for the in vitro conservation of 'Florida Rough' lemon plants. Microcuttings (approximately 1 cm) from plants that were previously cultivated in vitro were inoculated into test tubes with 10 mL of woody plant medium (WPM) at different concentrations (1/1, 1/2 and 1/4) and supplemented with 0, 12.5, 25 and 50 g.L-1 of sucrose, solidified with 7 g.L-1 agar and adjusted to pH 5.8. The experiment was completely randomized in a 3 x 4 factorial design with 15 replications and was maintained under controlled conditions for 360 days. After this period the plant height in cm (PH), the plant dry mass in g (PDM) and the 21 numbers of green leaves (NGL), senescent leaves (NSL) and microcuttings (NM) were evaluated. The variables that best explained the observed behavior of the 'Florida Rough' lemon plants were NGL and PH, with values of 61.63 and 35.08%, respectively. The original concentration of the WPM with the addition of 25 g. L-1 of sucrose yielded the best 'Florida Rough' lemon plant growth reduction in vitro while maintaining the physiologically health of the plants.


O estabelecimento de condições de crescimento mínimo é fundamental para a conservação in vitro de germoplasma. Alterações nas concentrações do meio básico, na fonte de carbono, são fatores importantes na desaceleração do crescimento de plantas in vitro. Este trabalho teve como objetivo ajustar um protocolo de conservação in vitro de plantas do limoeiro 'Rugoso da Flórida'. Microestacas de plantas previamente cultivadas in vitro, com aproximadamente 1 cm, foram inoculadas em tubos de ensaio contendo 10 mL do meio de cultura WPM em diferentes concentrações (1/1, 1/2 e 1/4), suplementado com 0; 12,5; 25 e 50 g.L-1 de sacarose, solidificado com 7 g.L-1 de ágar e o pH ajustado em 5,8. O experimento foi instalado em delineamento inteiramente casualizado com 15 repetições, em esquema fatorial 3 x 4, e mantido sob condições controladas durante 360 dias. Após este período, foram avaliadas altura de planta em cm (AP), número de folhas verdes (NFV), número de folhas senescentes (NFS), número de microestacas (NM) e massa da planta seca em g (MPS). As variáveis que mais contribuíram para explicar a diferença existente no comportamento das plantas do limoeiro 'Rugoso da Flórida' no meio de cultura WPM foram NFV e AP com 61,63 e 35,08%, respectivamente. O meio de cultura WPM na sua concentração original com a adição de 25 g.L-1 de sacarose promoveu os melhores resultados na redução do crescimento das plantas in vitro do limoeiro 'Rugoso da Flórida', mantendo-as fisiologicamente sadias.

Background: Equine colic syndrome comprises numerous conditions associated with abdominal pain in horses. Impaction, a common cause of this manifestation, is strongly related to these animals' diet. Highly fibrous diets such as sugarcane can predispose horses to colic. The clinical condition can be worsened by fermentative processes, which lead to dysbiosis, circulatory disorders and even endotoxemia. The aim of this study was to report 4 cases of colic syndrome among 8 horses that underwent an experiment to adapt them to a sugarcane-based diet, and to correlate the animals' clinical conditions to the forage they ingested. Cases: Eight male castrated Mangalarga Marchador horses, between 5.5 and 7 years old, were subjected to an experiment to test the feasibility of sugarcane as forage. Four of these horses were taken to the Large Animal Veterinary Hospital (HVGA) of the Federal Rural University of Rio de Janeiro for treatment of abdominal signs of discomfort a few days after the exclusive consumption of sugarcane, in a proportion of 1.75% of live weight in dry matter. The animals' symptoms ranged from behavioral signs indicative of pain to changes in vital parameters and structure of the feces, as well as changes revealed by transrectal palpation. Three of the 4 cases presented impaction in the small colon, and 1 of the horses also presented impaction in the right dorsal colon and rostral displacement of the pelvic flexure, with accumulation of contents in the right ventral colon and sternal flexure. Two of the cases were treated medically, while the other 2 required surgical intervention. The clinical condition of all the patients evolved favorably and they were discharged between 2 and 18 days. Discussion: Colic originating in the digestive system is a syndrome strongly associated with management, especially with respect to confinement, nutrition, and parasite control. During the experiment, 4 of the 8 horses fed with sugarcane presented with colic syndrome. The low quality of sugarcane fiber is due to the high degree of lignification of the plant cell wall, which favors accumulation of ingesta. The poor digestibility and sweet taste of this roughage favor increased consumption. Furthermore, its high sucrose content, associated with an increased rate of passage in the small intestine, alters the intestinal microbiome, and hence, the fermentation byproducts and pH of the ingesta. High intestinal content, allied to longer retention times in the colon and activation of the renin-angiotensin-aldosterone system, promote greater dryness of the ingesta, predisposing the occurrence of impactions in the most distal portion of the large intestine. Intestinal distension and mesenteric traction caused by the accumulation of contents and gases trigger pain, which can worsen due to displacement of the large colon. Small colon impaction, which is easily identified by transrectal palpation, evolves gradually and its treatment, both clinical and surgical, tends to have a favorable prognosis. The need for alternative food sources for horses is a growing demand; however, sugarcane as an exclusive roughage has been shown to be unsafe for horses. The low quality of the fiber and the high sucrose content of this forage can alter the digestive physiology of horses through changes in the passage rate, microbiome and motility of digesta, predisposing them to intestinal dysfunction, ingesta compaction and displacement of the large colon.

Cattleya tigrina A. Rich has been suffering heavy losses in its natural habitat and it is now included in the list of plants that are vulnerable to extinction. The development of in vitro propagation and conservation methodologies, as well as acclimatization, are considered important for species at the risk of extinction, as they promote the multiplication and conservation of the species, hence avoiding the loss of their genetic variability. The present study established the protocol of micropropagation and the in vitro conservation of C. tigrina. For the in vitro propagation, the study tested two volumes of the MS medium and two medium consistencies (stationary liquid and semi-solid). For acclimatization, the substrate mixtures containing pine bark, charcoal, vermiculite, and coconut coir were analyzed. For the in vitro conservation, different concentrations of the salts were tested in the MS medium, together with the osmotic regulators (sucrose, mannitol, and sorbitol), and at two temperatures (18 and 25 °C). The results obtained inferred that the semi-solid medium was superior to the stationary liquid medium in the variables of survival and the presence of roots, while the liquid medium was superior to the semi-solid medium in the number of shoots. For acclimatization, pine bark was the substrate where the plants developed an improved height, with sprouting, and rooting. The conservation was satisfactory and the plants remained viable for a period of 730 days, with the MS medium with 25% of the salts, and at temperatures of 18 ºC or 25 ºC. The plants were propagated in the stationary liquid MS medium (10 mL) and the semi-solid medium (25 mL), while they were acclimatized in pine bark and preserved in the MS medium with 25% of the salts (18 ºC or 25 ºC).


A Cattleya tigrina A. Rich vem sofrendo grandes perdas no seu habitat natural, sendo assim foi inclusa na lista de vulneráveis a extinção. O desenvolvimento de metodologias de propagação e conservação in vitro, bem como de aclimatização, são consideradas importantes para espécies em risco de extinção, por promover a multiplicação e conservação da espécie, evitando a perda do seu material genético. Desta forma, o presente trabalho visou estabelecer protocolo de micropropagação e conservação in vitro de C. tigrina. Para propagação in vitro testou volumes de meio de cultura e duas consistências do meio MS (líquido estacionário e semissólido). Para aclimatização, analisou misturas de substratos contendo casca de pinus, carvão vegetal, vermiculita e pó de coco. Para conservação in vitro, foram testados diferentes concentrações de sais no meio MS, reguladores osmóticos (sacarose, manitol e sorbitol), e duas temperaturas (18 e 25 °C). Os resultados obtidos inferem que, o meio de cultura semissólido foi superior ao líquido nas variáveis sobrevivência e presença de raízes, enquanto que o meio líquido foi superior ao meio semissólido em números de brotos. Na aclimatização a casca de pinus foi o substrato em que as plantas se desenvolveram melhor em altura, brotação e enraizamento. A conservação foi satisfatória e as plantas permaneceram viáveis por um período de 730 dias, usando 25% dos sais MS e temperatura de 18 ºC ou 25 ºC. As plantas podem ser propagadas em meio MS líquido estacionário (10 mL) ou semissólido (25 mL), aclimatizadas em casca de pinus e conservadas em 25% dos sais MS (18 ºC ou 25 ºC).
