Resumo

Scuticociliatosis, caused by an opportunistic ciliate protozoan, is responsible for significant economic losses in marine ornamental fish. This study reports the occurrence of Uronema spp., parasitizing ten species of marine reef fish at an ornamental fish wholesaler: Blue green damselfish (Chromis viridis), Vanderbilts Chromis (Chromis vanderbilti), Pennant coralfish (Heniochus acuminatus), Threespot angelfish (Apolemichthys trimaculatus), Goldspotted angelfish (Apolemichthys xanthopunctatus), Sea goldie (Pseudanthias squamipinnis), Orchid dottyback (Pseudochromis fridmani), Threadfin butterflyfish (Chaetodon auriga), Vagabond butterflyfish (Chaetodon vagabundus), and Bluecheek butterflyfish (Chaetodon semilarvatus). Diseased fish showed disorders such as hemorrhages and ulcerative lesions on the body surface. Histopathological analysis of the muscle, liver, gut, kidney, spleen, gills, and stomach revealed hemorrhages and degeneration of muscle fiber, vacuolar degeneration of hepatocyte, inflammatory process and granuloma in the liver, atrophy of intestinal villi, inflammatory process and granuloma in the kidney, melanomacrophage centers, as well as inflammatory process in the spleen, epithelial cells hyperplasia and granuloma formation in the gills, and vacuolar degeneration and eosinophils in the stomach. Due to the severity of the disease, it is necessary to implement biosecurity measures with rapid and accurate diagnosis to minimize the risk of economic losses caused by Uronema spp.(AU)


Scuticociliatose, causada pelo protozoário ciliado oportunista, é responsável por significativas perdas econômicas em peixes marinhos ornamentais. O estudo relata a ocorrência de Uronema spp., parasitando dez espécies de peixes de recife em um distribuidor de peixes ornamentais: Blue green damselfish (Chromis viridis), Vanderbilts Chromis (Chromis vanderbilti), Pennant coralfish (Heniochus acuminatus), Threespot angelfish (Apolemichthys trimaculatus), Goldspotted angelfish (Apolemichthys xanthopunctatus), Sea goldie (Pseudanthias squamipinnis), Orchid dottyback (Pseudochromis fridmani), Threadfin butterflyfish (Chaetodon auriga), Vagabond butterflyfish (Chaetodon vagabundus), e Bluecheek butterflyfish (Chaetodon semilarvatus). Peixes doentes apresentaram distúrbios como hemorragias e lesões ulcerativas na superfície do corpo. A análise histopatológica do músculo, fígado, intestino, rim, baço, brânquias e estômago revelou hemorragias e degeneração das fibras musculares, degeneração vacuolar de hepatócitos, processo inflamatório e granuloma no fígado, atrofia das vilosidades intestinais, processo inflamatório e granuloma no rim, centros de melanomacrófagos e processo inflamatório no baço, hiperplasia das células epiteliais, bem como formação de granuloma nas brânquias e degeneração vacuolar e eosinófilos no estômago. Devido à gravidade da doença, é necessário implementar medidas de biossegurança com diagnóstico rápido e preciso para minimizar o risco de perdas econômicas causadas por Uronema spp.(AU)

Resumo

This study evaluated the parasitic fauna of hybrids tambacu (Colossoma macropomum × Piaractus mesopotamicus) and patinga (P. mesopotamicus × Piaractus brachypomus) and their host-parasite-environment interaction during the warm and cold seasons in two fish farms located in the State of Mato Grosso do Sul, Brazil, South America. A total of 120 fish, being 60 fish for species (30 in the warm season and 30 in the cold season) were examined. Water quality was measured weekly to evaluate the interaction between environmental conditions and parasitism. Fifteen species of parasites were found: Ichthyophthirius multifiliis, Chilodonella hexasticha e Trichodina sp. (Protozoa), Henneguya piaractus, Myxobolus colossomatis and Myxobolus cuneus (Myxozoa); Anacanthorus penilabiatus, Mymarothecium boegeri, Mymarothecium viatorum, and Notozothecium janauachensis (Monogenoidea), Goezia spinulosa and Goezia sp. (Nematoda), Echinorhynchus jucundus (Acanthocephala), and Dolops carvalhoi, Lernaea cyprinacea (Crustacea). The ciliate protozoan I. multifiliis and monogenoids were the most prevalent parasites in the cold and warm seasons for both hybrids in the different culture systems intensive production and sportive fishing. It was observed that the serrasalmid hybrids are more susceptible to parasites and harbor high diversity of parasites in relation to the parental species C. macropomum, P. mesopotamicus and P. brachypomus, with a predominance of ectoparasites.(AU)


Este estudo avaliou a fauna parasitária de híbridos tambacu (Colossoma macropomum × Piaractus mesopotamicus) e patinga (P. mesopotamicus × Piaractus brachypomus) e a interação hospedeiro-parasito-ambiente durante as estações quente e fria em duas pisciculturas localizadas no estado de Mato Grosso do Sul, Brasil, América do Sul. Foram examinados um total de 120 peixes, sendo 60 peixes por espécie. A qualidade da água foi mensurada semanalmente para avaliar a interação entre condições ambientais e parasitismo. Foram encontradas quinze espécies de parasitos: Ichthyophthirius multifiliis, Chilodonella hexasticha e Trichodina sp. (Protozoa); Henneguya piaractus, Myxobolus colossomatis e Myxobolus cuneus (Myxozoa), Anacanthorus penilabiatus, Mymarothecium boegeri, Mymarothecium viatorum e Notozothecium janauachensis (Monogenoidea), Goezia spinulosa e Goezia sp. (Nematoda), Echinorhynchus jucundus (Acanthocephala), Dolops carvalhoi eLernaea cyprinacea (Crustacea). O protozoário ciliado I. multifiliis e helmintos monogenoides foram os parasitos mais prevalentes nas estações fria e quente para ambos os híbridos nos diferentes sistemas de cultivo, produção intensiva comercial e pesca esportiva. Observou-se que os híbridos de serrasalmídeos são mais suscetíveis aos parasitos e abrigam alta diversidade parasitária em relação às espécies parentais, C. macropomum, P. mesopotamicus e P. brachypomus em ambiente de cultivo com predominância de ectoparasitos.(AU)

Resumo

Cryptocaryon irritans is an obligate parasitic ciliate protozoan of wild and cultured marine fish. It causes white spot disease, and infections with this pathogen can cause significant losses for aquarists and commercial marine cultures worldwide. This study reports the occurrence of C. irritans parasitizing the ornamental reef fish, yellowtail tang, Zebrasoma xanthurum. Six days after being introduced to a new environment, 11 yellowtail tangs had white spots scattered across their bodies and fins. Suspicion of infection with C. irritans was evaluated by scraping the skin to confirm clinical diagnosis. After confirmation, the yellowtail tangs were transferred to a hospital aquarium and treated with the therapeutic agent Seachem Cupramine® for 15 days. During the treatment period, the copper concentration was monitored daily. At the end of the treatment, none of the yellowtail tangs showed clinical signs of white spots on their bodies, and skin scraping confirmed the yellowtail tangs were no longer infected. Subsequently, the yellowtail tangs were released for sale.(AU)


Cryptocaryon irritans é um protozoário ciliado parasita obrigatório de peixes marinhos selvagens e cultivados. Causa a doença dos pontos brancos. Infecções por esse patógeno pode causar perdas significativas para aquaristas e para marinocultura comercial em todo o mundo. Este estudo relata a ocorrência de C. irritans parasitando a espécie de peixe ornamental de recife Zebrasoma xanthurum. Seis dias após serem introduzidos no novo ambiente, onze peixes apresentaram pontos brancos espalhados pelo corpo e nadadeiras. Suspeitando de infecção por C. irritans foi realizado raspado de pele para confirmar o diagnóstico clínico. Após a confirmação, os peixes foram transferidos para um aquário hospital e tratados com o agente terapêutico Seachem Cupramine® por 15 dias. Durante o período de tratamento o monitoramento da concentração de cobre foi diário através da utilização de um fotômetro. Após o final do tratamento, todos os peixes já não apresentavam mais sinais clínicos de pontos brancos pelo corpo, e foram negativos no raspado de pele, sendo foram encaminhados para comercialização.(AU)

Resumo

The phylum Ciliophora is organized in 14 classes with ~8,000 described species ( Gao et al., 2016 ). Among unicellular microeukaryotes, ciliates are the most specialized, diversified and with the highest complexity in terms of cellular organization ( Puytorac, 1994 ). They typically occupy basal trophic levels and display a wide geographical distribution occurring in almost all environments such as marine, fresh and brackish waters, and also in edaphic systems like soils, mosses and lichens ( Lynn, 2008 ). Their diversity in lotic systems have been extensively studied in the northern hemisphere because of their potential use as water quality bioindicators ( Wiackowski, 1981 ; Primc, 1988 ; Groliére et al., 1990; Madoni, 2005 ). However, scarce information is available in literature about their diversity in aquatic ecosystems in Brazil, even though its privileged hydrological condition. The access to ciliate diversity and biogeography is a challenge tasks because they are diminute organisms, difficult to identify ( Finlay and Fenchel, 1999 ), there is a lack of experienced specialists ( Foissner, 2006 ) and a number of species stay encysted for most of their life cycles ( Foissner, 2004 ). In Brazil, significant works were performed with this emphasis such as conducted in Rio Grande do Sul (Safi et al., 2014), Paraná ( Pauleto et al., 2009 ; Buosi et al., 2011 ; Velho et al., 2005 , 2013 ), São Paulo ( Bagantini et al., 2013 ; Regali-Seleghim et al., 2011 ), Pará ( Castro et al., 2014 ), Minas Gerais ( Dias et al., 2008 , 2010 ) and Rio de Janeiro ( Paiva and Silva-Neto, 2004a , b ) states. Inventory studies constitute the first step for development of applied biotechnological usage of ciliates. Regali-Seleghim et al. (2011) highlight the importance of more works surveying the diversity of ciliates in less studied regions of Brazil given their ecological importance. Moreover, establishment of in vitrocultures will contribute with information to biomonitoring programs ( Madoni and Romeo, 2006 ; Shi et al., 2012 ) and for better evaluation of the biotechnological potential of these organisms ( Mansano et al., 2016 ). This present work aimed to survey the diversity of the species of ciliated protists in a neotropical lotic urban system located in Southern region of Minas Gerais state, Brazil.(AU)

ABSTRACT Physiological reorganization is described for the first time for the hypotrich ciliate genus Apoamphisiella Foissner, 1997, based on a population of A. vernalis (Stokes, 1887) Berger, 2006 collected from an eutrophic pond in the state of Minas Gerais, Brazil. Stomatogenesis is epiapokinetal, and the cirral pattern reorganizes from six typical fronto-ventral-transverse primordia, plus at least one short primordium located between IV and V. Primordia I and II originate from disaggregating undulating membranes and buccal cirrus, respectively; primordium III originates from left frontoventral cirrus plus a streak of basal bodies extending from the anterior end of oral primordium; primordium IV and the short extra primordia are formed from basal bodies perhaps associated to the posterior end of III plus the disaggregation of anteriormost cirri of the left ventral row. Primordia V and VI arise within the right ventral row as a single streak, splitting to form the two primordia. Marginal primordia develop initially within the pre-existent marginal rows, continuing to reorganize outside, pushing the old marginal cirri leftwards. Two dorsomarginal kineties develop associated to the right marginal row primordium. Dorsal ciliature also reorganizes from within the pre-existent dorsal kineties. The two macronuclear nodules approach each in middle reorganizers, briefly touching each other. At least one micronucleus undergo division. The process of physiological reorganization in A. vernalis resembles that of the North American Paraurostyla weissei complex representative, and along with features of the interphase morphology, indicate that Apoamphisiella belongs or is related to the Cyrtohymena-Paraurostyla group, within the Dorsomarginalia.

Resumo

This study reports the occurrence of Uronema sp., which was found to be parasitizing three species of marine reef fish imported into Brazil and maintained in quarantine: Vanderbilts Chromis (Chromis vanderbilti), blue-green damselfish (Chromis viridis), and sea goldie (Pseudanthias squamipinnis). During the quarantine period, some fish presented with behavioral disorders and hemorrhages and ulcerative lesions on the body surface. Histopathological analysis showed hemorrhages, inflammation comprising mononuclear and granular cells in the skeletal muscle, and necrosis of the skin and the secondary lamellae of the gills, and parasites were also observed in the renal capsule. The absence of transboundary measures available to prevent the occurrence of ornamental fish diseases is also discussed.(AU)

O presente estudo relata a ocorrência de Uronema sp., parasitando três espécies de peixes ornamentais de recife importados para o Brasil e mantidos em quarentena: Vanderbilts Chromis (Chromis vanderbilti), blue-green damselfish (Chromis viridis), e sea goldie (Pseudanthias squamipinnis). Durante o período de quarentena, alguns peixes apresentaram alterações comportamentais tais com hemorragias e lesões ulcerativas na superfície da pele. A histopatologia mostrou hemorragias, inflamação com células mononucleares e granulares na musculatura, e necrose da pele e lamelas secundárias das brânquias, e parasitas também foram observados na cápsula renal. A ausência de medidas transfronteiriças disponíveis para prevenir a ocorrência de doenças de peixes ornamentais também é discutida.(AU)

Resumo

The effects of cassava wastewater on nutrient digestibility and rumen protozoa were evaluated in 32 Santa Inês sheep fed a diet with Tifton hay, ground corn cob, and concentrate, containing 0, 500, 1000, or 1500 mL cassava wastewater. Uncastrated sheep at an average age of 167 days and average weight of 25.3 kg were housed in individual stalls and distributed in a randomized complete design with four treatments and eight replicates. The experimental period was 70 days: 15 days for adaptation to the diets and 60 days to collect the ingredients and diets supplied. Prior to this stage, the animals were adapted to the consumption of cassava wastewater, in collective cages, for seven days. Apparent digestibility was determined by partial feces collection and by using the LIPE® external marker in 250-mg capsules. The apparent digestibility of nutrients did not differ (P>0.05) in relation to the cassava wastewater levels tested. Eleven genera of rumen protozoa were identified, and they formed a community that, except for Ophryoscolex, did not differ among the cassava wastewater treatments with respect to composition. However, Entodinium species were observed at a higher density (P<0.05) in sheep that received 1500 mL of cassava wastewater. The use of cassava wastewater in the sheep diet does not affect the digestibility of nutrients or composition of rumen protozoa; however, it causes a significant increase in Entodinium density.(AU)


Os efeitos da manipueira sobre a digestibilidade de nutrientes e protozoários ruminais foram avaliados utilizando-se 32 ovinos Santa Inês submetidos a dietas compostas por feno de Tifton, rolão de milho e concentrado, acrescidas de 0, 500, 1000 ou 1500 mL de manipueira. Foram utilizados ovinos não castrados com idade media de 167 dias e com peso médio e aproximadamente 25,3 kg. Foram alojados em gaiolas individuais e distribuídos em delineamento inteiramente casualizado com quatro tratamentos e oito repetições. O período experimental foi de 70 dias: 15 dias para adaptação às dietas e 60 dias para coleta dos ingredientes e rações. Antes desta fase, os animais foram adaptados ao consumo de manipueira em gaiolas coletivas durante um período de 7 dias. A digestibilidade aparente foi determinada pela coleta parcial de fezes e pelo uso do marcador externo LIPE® em cápsulas de 250 mg. A digestibilidade aparente dos nutrientes avaliados não diferiu (P>0,05) em relação aos níveis de manipueira estudados. Onze gêneros de protozoários foram identificados, compondo uma comunidade que à exceção de Ophryoscolex, não diferiu entre os tratamentos em sua composição. Entretanto, as espécies de Entodiniumapresentaram maior densidade (P<0,05) em ovinos que receberam 1500 mL de manipueira. The effects of cassava wastewater on nutrient digestibility and rumen protozoa were evaluated in 32 Santa Inês sheep fed a diet with Tifton hay, ground corn cob, and concentrate, containing 0, 500, 1000, or 1500 mL cassava wastewater. Uncastrated sheep at an average age of 167 days and average weight of 25.3 kg were housed in individual stalls and distributed in a randomized complete design with four treatments and eight replicates. The experimental period was 70 days: 15 days for adaptation to the diets and 60 days to collect the ingredients and diets supplied. Prior to this stage, the animals were adapted to the consumption of cassava wastewater, in collective cages, for seven days. Apparent digestibility was determined by partial feces collection and by using the LIPE® external marker in 250-mg capsules. The apparent digestibility of nutrients did not differ (P>0.05) in relation to the cassava wastewater levels tested. Resumo

This study proposes improvements to the pyridinated silver carbonate impregnation technique in rumen ciliate protozoa in order to provide a standardized impregnation protocol for usable for the largest possible number of species in domestic ruminants. The proposed improvements are based on results obtained from impregnation of oral infraciliature and nuclear apparatus of 36 rumen ciliate species, which are symbionts of domestic ruminants. Compared to established protocols for morphology of rumen ciliates, impregnation of a wider range of genera and species was observed with the proposed protocol. The impregnation time varied according to size, ciliate taxon, or both, varying from shorter (5 minutes) for small entodiniomorphid ciliates ( 80 m) and the genus Dasytricha, to longer (30 minutes) for large entodiniomorphid ciliates (>80 m) and the genus Isotricha. The proposed protocol is simple and easily reproducible. It is also advantageous for taxonomic, animal science, and ecological studies that aim to inventory the ruminal biota as well as understand the population structure of rumen ciliates and their relationship with the host.

Resumo

Abstract A new species of Tripartiella is described from the gills of the wild saddle cichlid Aequidens tetramerus in north Brazil. Wet smears of skin and gills of examined fish were air-dried at room temperature and impregnated with Kleins dry silver method for examination of the adhesive discs structures and denticles. Total prevalence of parasitism was 65%. This ciliate is characterized as a small-sized trichodinid, body diameter 37.03 ± 4.9 m, adhesive disc 30.50 ± 2.71 m, denticulate ring 13.28 ± 0.8 m and 24 ± 2.0 denticles. Taxonomic and morphometric data for the new species are discussed.


Resumo Um nova espécie de Tripartiella encontrada nas brânquias de Aequidens tetramerus da região Norte do Brasil é descrita. Esfregaços de pele e brânquias de peixes examinados foram secos à temperatura ambiente e impregnados pelo método de Klein para observação das estruturas do disco adesivo e dentículos. A prevalência do parasitismo foi de 65%. Este ciliado é caracterizado como pequeno trichodinídeo, com corpo medindo 37,03 ± 4,9 m de diâmetro, disco adesivo com 30,50 ± 2,71 m, anel denticulado com 13,28 ± 0,8 m e 24 ± 2,0 dentículos. Dados taxonômicos e morfométricos para a nova espécie são discutidos.

Ciliates are an essential component of microbial food webs, connecting biomass production to higher trophic levels and providing substrates for bacterial growth. Bacteria are widespread in sea sediment and the first to metabolize organic matter. Heavy metals are toxic and bind to particulate organic matter. This work aims to study the influence of heavy metals (Zn and Cu) on D. appendiculata and their naturally associated bacteria, both from Guanabara Bay during 96 h bioassay with 0, 0.001, 0.009, 0.05, 0.1 mg L-1 and maximum concentration stipulated by CONAMA 357 (Zn 5.0; Cu 1.0 mg L-1). It was analyzed for tolerance, resistance and biomass production. In Zn control, ciliate produced 1.24x102-2.47x103 μg C cm-3 (0-96 h), with 0.009 mg Zn L-1 reaches 2.49x102-1.93x103 μg C cm-3 (0-96 h). Their naturally associated bacteria, in Zn control, produced 5.0x10-2-5.67x10-2 μg C cm-3 (0-96 h), with 0.1 mg Zn L-1 4.87x10-2-8.38x10-2 μg C cm-3 (0-96 h). In Cu control, D. appendiculata produced 1.04x102-3.12x102 μg C cm-3 (0-96 h), with 0.009 mg Cu L-1 biomass was 8.31x101-7.27x10-2 μg C cm-3 (0-96 h) and with CONAMA concentration was below detection level. Their naturally associated bacteria, in Cu control, produced 1.20x10-2-5.14x10-2 μg C cm-3 (0-96 h) and with 0.1 mg Cu L-1 7.40x10-4-3.81x10-2 μg C cm-3 (0-96 h), with CONAMA concentration 5.14x10-2 μg C cm-3 (96 h). D. appendiculata are tolerant to 0.09 mg L-1 and resistant to 0.1 mg L-1 after 24 h to Zn-Cu and LD50 stipulated was 1.17 (Zn) 0.90 (Cu) mg L-1. Microbial loop was functional in low concentration of Zn and Cu, but their diversity in nature was affected.(AU)

Protozoans are microbial eukaryotes known to be ideal for aquatic ecotoxicological testing. Therefore, aiming to evaluate their possible use in standardized ecotoxicity assays we determined the best cultivation conditions and growth curves for two strains of the ciliated protozoan Paramecium caudatum Ehenberg 1833. One strain (PC1) was isolated from the Monjolinho reservoir-SP (22° 01 S and 47° 53 W) where the mean dissolved O2 is 5.205 mgL-1, pH 6.43 and electrical conductivity 34.75 μS cm-1. The other strain (PC2) from the Óleo lagoon in Jataí Ecological Station- SP (21° 36 S and 47° 49 W), with mean dissolved of 4,42 mgL-1, pH 4.90 and electrical conductivity 10.96 μS cm-1. The higher mean cellular biovolume and density obtained were used to evaluate different cultivation parameters such as temperature, pH, light exposition, stirring regimen and cultivation flask size. Considering protozoan densities and cell biovolume, the best culture conditions for PC1 strain was pH 9.0 at 30 °C and for PC2 was pH 7.0 at 27.5 °C. For both strains and evaluated parameters (biovolume and density), the bottle size did not interfered in the results and absence of light resulted in better ones. Concerning agitation, for PC2, there was no significant difference for this parameter and for PC1 in the absence of shaking we obtained better results. Therefore we decided to make the tests in the absence of light and without shaking. Regarding cell biovolume, the best condition was pH 9.0 at 25 ° C for the PC1 strain and pH 8.0 at 20 ° C for PC2 strain. The generation time calculated in a 96 hours growth curve was 8.35 hours for PC1 and 7.6 hours for PC2. (AU)
