Resumo

As our understanding of ovarian function in cattle has improved, our ability to control it has also increased. The development of Fixed-Time Artificial Insemination (FTAI) protocols at the end of the 20th century has increased exponentially the number of animals inseminated over the last 20 years. The main reasons for this growth were the possibility of obtaining acceptable pregnancy rates without heat detection and, above all, the induction of cyclicity in suckled cows in postpartum anestrus and prepubertal heifers at the beginning of the breeding season. Most FTAI treatments in South America have been based on the use of progesterone (P4) releasing devices and estradiol to synchronize both follicular wave emergence and ovulation, with pregnancy rates ranging from 40 to 60%. These protocols are implemented on a regular basis, allowing producers access to high-quality genetics, and increasing the overall pregnancy rates during the breeding season. In addition, it provided the professionals involved in these programs with a new source of income and the diversification of their practices into activities other than their usual clinical work. Many of these practices are now apparently at risk from restrictions on the use of estradiol by the European Union (EU) and other countries. However, the development of alternative protocols based on GnRH, with P4 devices and eCG and other new products that are not in the market yet will allow us to adapt to the new times that are coming. Logically, the challenge has already been raised and we must learn to use alternative protocols to try to continue increasing the use of this technology in beef and dairy herds. The objective of the present review is to describe the main aspects of banning estradiol in livestock production, the negative impacts on reproductive efficiency, and to present some alternative FTAI protocols for dairy and beef cattle.(AU)

Animals that exhibited estrus had greater pregnancy success compared to animals that did not exhibit estrus before fixed-time AI (FTAI). Estradiol is synthesized in bovine ovarian follicles under gonadotropin regulation and can directly and indirectly regulate the uterine receptivity and luteal function. Estradiol concentrations at FTAI impacted oviductal gene expression and has been reported to play an important role in establishing the timing of uterine receptivity. These changes have been reported to impact uterine pH and sperm transport to the site of fertilization. After fertilization, preovulatory estradiol has been reported to improve embryo survival likely by mediating changes in uterine blood flow, endometrial thickness and changes in histotroph. Cows with greater estradiol concentrations at the time of GnRH-induced ovulation also had a larger dominant follicle size and greater circulating progesterone concentrations on day 7. Therefore, it is impossible to accurately determine the individual benefit of greater estradiol concentrations prior to ovulation and greater progesterone concentrations following ovulation to pregnancy establishment, as these two measurements are confounded. Research has indicated an importance in the occurrence and timing of increasing preovulatory concentrations of estradiol, but increasing estradiol concentrations by supplementation may not be sufficient to increase fertility. Increased production of estradiol by the preovulatory follicle may be required to enhance fertility through the regulation of sperm transport, fertilization, oviductal secretions, the uterine environment, and embryo survival.(AU)

Resumo

This study aimed to observe the effects of 17 ß-estradiol replacements on the fecal microbiota in spayed cats. Individual samples of fresh feces were collected and stored at -80° C. Sequencing of the V3/V4 regions of the 16S rRNA gene was used, and bioinformatic analysis was performed. Firmicutes/Bacteriodetes ratio was lower in the group receiving estrogen replacement compared to the SHAM group (P = 0,005). Jaccard index (P = 0.123) and Yue & Clayton index (P = 0.094) did not reveal alpha and beta diversity differences. The linear discriminant analysis effect size (LefSe) identified Firmicutes and MegasPhaera as the biomarkers for the SHAM group, and Burkholderiales, Betaproteobacteria, Sutterellaceae, Suterella, Proteobacteria, Proteobacteria unclassified and Collinsella for the group receiving estrogen replacement.(AU)


O objetivo deste estudo foi observar os efeitos da reposição de 17 ß-estradiol na microbiota fecal de gatas castradas. Amostras individuais de fezes frescas foram colhidas e armazenadas a -80°C. Foi realizado o sequenciamento das regiões V3/V4 do gene 16S rRNA e a análise bioinformática. A razão Firmicutes/Bacteriodetes foi menor no grupo que recebeu reposição estrogênica em comparação ao grupo SHAM (P = 0,005). O índice de Jaccard (P = 0,123) e o índice de Yue & Clayton (P = 0,094) não revelaram diferenças na alfa e beta diversidade. A análise discriminatória linear de tamanho do efeito (LefSe) identificou Firmicutes e Megasphaera como biomarcadores para o grupo SHAM, e Burkholderiales, Betaproteobacteria, Sutterellaceae, Suterella, Proteobacteria, Proteobacteria não classificada e Collinsella para o grupo que recebeu reposição estrogênica.(AU)

Resumo

Este texto discute as estratégias farmacológicas para a manipulação do ciclo estral de fêmeas taurinas de corte, com foco na inseminação artificial em tempo fixo (IATF). Os zebuínos, rebanho predominante no Brasil, apresentam características de ciclo estral diferentes das raças taurinas, o que justifica a busca por estratégias hormonais adaptadas para o controle do ciclo estral nas subespécies. O estradiol combinado com a progesterona (P4) e prostaglandina F2 alfa (PGF) é o esquema hormonal mais comumente utilizado para a manipulação do ciclo estral em protocolos de IATF. Porém, o uso de GnRH combinado ou em substituição aos ésteres de estradiol vem sendo considerado. Coletivamente, os dados do nosso grupo reforçam a necessidade de customizar as abordagens para o controle do ciclo estral de acordo com a composição genética das fêmeas bovinas.(AU)


This text discusses pharmacological strategies to manipulate the estrous cycle of taurine and synthetic females, with a focus on timed artificial insemination (TAI). Zebu cattle, the predominant herd in Brazil, have different estrous cycle characteristics than taurine breeds, requiring different synchronization hormonal strategies for each subspecies. Estradiol combined with progesterone (P4) and prostaglandin F2 alpha (PGF) is the most used hormonal scheme for estrous cycle manipulation in TAI protocols. But the use of GnRH instead of estradiol esters is being considered. Collectively, our group's data reinforce the need to customize approaches to estrous cycle control according to the genetic composition of bovine females.(AU)

Estradiol cypionate (EC) or GnRH have been widely used for ovulation induction in timed embryo transfer (TET). EC administration increases the proportion of cows that show estrus, whereas GnRH promotes more synchronized ovulations. The aim of the present study was to evaluate the potential beneficial effects of combining EC and GnRH in TET. In experiment 1, no difference was observed on serum progesterone concentrations on Day 6 and 13 after GnRH treatment between GnRH and EC+GnRH groups. In experiment 2, pregnancy per embryo transfer (P/ET) did not differ (p = 0.69) between GnRH (62.8%) and EC+GnRH (58.7%) groups. In conclusion, combining EC and GnRH for ovulation induction does not increase progesterone secretion and pregnancy rate after TET in cattle.(AU)

Resumo

The aim of this study was to verify the efficiency and ovulation time after the administration of different inducers for synchronization of ovulation in beef cows. One hundred and eight non-lactating cows were distributed into the control group (CG; untreated; n=28), estradiol benzoate (EB) group (EBG; n=28); 17 beta-estradiol (17ßE) group (17ßEG; n=28), and deslorelin (DES) group (DESG; n=24). On day minus 11 (D-11) of the protocol, the CG underwent application of cloprostenol and ultrasound examination (US); on D0, progesterone (P4) was inserted plus EB; on D7, cloprostenol was applied; on D9, P4 was removed and cloprostenol plus 400 IU of equine chorionic gonadotropin (eCG) was injected. The EBG was subjected to treatment identical to that of the CG, except on D10, when the cows received EB. The 17ßE was subjected to the same protocol used in the CG except for the administration of 17ßE on D10. And, the DESG was subjected to the same treatment as the CG, except on D10, when the group received DES acetate. Twelve hours after the administration of EB, 17ßE and DES, ovarian US were performed every 6 hours. The preovulatory follicle (POF) diameters measured before ovulation were 19.5; 14.7; 18.7 and 19.8 mm respectively for CG, EBG, 17ßEG and DESG; and the time intervals between inducer application and ovulation were 20.2; 18.9; 21.0 and 22.5 hours respectively. In conclusion, all ovulation inducers were efficient in promoting ovulation; the inducers caused ovulation between 18.9 and 22.5 hours; EB promoted ovulation in a shorter time (P<0.05); 17ßE and DES showed greater variation in application/ovulation time between groups.


O objetivo do estudo foi verificar a eficiência e a ovulação após a administração de diferentes indutores para a sincronização da ovulação em vacas de corte. Cento e oito vacas não-lactantes foram distribuídas em grupo controle (GC; não tratadas; n=28); grupo benzoato de estradiol (BE) (GBE; n=28); grupo 17 beta-estradiol (17ßE) (G17ßE; n=28) e grupo deslorelina (DES) (GDES; n=24). No dia menos 11 (D-11) do protocolo, o GC recebeu cloprostenol e exame ultrassonográfico (US); ao D0, dispositivo de progesterona (P4) foi inserido mais BE; ao D7, cloprostenol foi aplicado; ao D9, a P4 foi removida e cloprostenol mais 400 UI de gonadotrofina coriônica equina (eCG) foi injetada. O GBE foi submetido a tratamento idêntico ao do GC, exceto ao D10, quando as vacas receberam BE. o G17ßE foi submetido ao mesmo protocolo usado no CG exceto pela administração de 17ßE ao D10. E, o GDES foi submetido ao mesmo tratamento que o CG, exceto ao D10, quando o grupo recebeu o acetato de DES. Doze horas após a administração de BE, 17ßE e DES, US ovarianos foram realizados a cada 6 horas. O diâmetro do folículo pré-ovulatório (FPO) medido antes da ovulação foi de 19,5; 14,7; 18,7 e 19,8 mm respectivamente para GC, GBE, G17ßE e GDES; e o intervalo de tempo entre a aplicação do indutor e ovulação foi 20,2; 18,9; 21;0 e 22,5 horas respectivamente. Em conclusão, todos os indutores da ovulação foram eficientes em promover a ovulação; os indutores acarretaram ovulação entre 18,9 e 22,5 horas; o BE promoveu a ovulação em menor espaço de tempo (P<0,05); 17ßE e DES demonstraram maior variação em aplicação/tempo de ovulação entre os grupos.

Resumo

O maior entrave para o êxito de um programa de transferência de embrião (TE) está relacionado às receptoras, tanto a qualidade de características reprodutivas, quanto a sincronização entre doadoras e receptoras. Éguas doadoras, geralmente, apresentam melhor escore corporal e disponibilidade alimentar nos períodos de seca, fazendo com que iniciem o seu ciclo estral precocemente enquanto éguas receptoras se encontram ainda em anestro ou período transicional, o que dificulta a TE. Vários estudos foram realizados com o objetivo de utilizar receptoras acíclicas como receptoras de embrião. Assim, podem ser utilizadas nos programas de TE quando submetidas a protocolos hormonais que mimetizam o ciclo estral natural. Uma diversidade de protocolos é encontrada na literatura, usando diferentes doses, posologias e intervalos do uso de estradiol associado a progestágenos. Portanto, objetivou-se analisar resultados de pesquisas com protocolos hormonais que poderiam ajudar a solucionar a escassez de receptoras cíclicas e maximizar os resultados da biotécnica de transferência de embriões.(AU)


There are obstacles to the success of equine embryo transfer programs, concomitantly there has been an increase in the popularity of the technique. One of the most urgent issues to overcome is related to the recipient mare availability and the donor-recipient synchrony. Usually, the donor mares have a higher body condition score and are fed a higher quality diet than recipient mares during the non-breeding season. Therefore, the donor mares start cycling earlier than recipient mares, which makes embryo transfer challenging. Several studies have been carried on finding hormonal protocols that mimic the natural oestrous cycle to allow the use of noncycling recipient mares for embryo transfer programs. Many protocols can be found in the literature, using different doses and intervals. Among the main difference is the use of progestogens, which can be used alone or in combination with oestradiol. The aim of this review was to analyse results that could help solve the lack of recipient mares and improve the results of the embryo transfer technique.(AU)

Elevadas taxas de descarte de vacas leiteiras relacionadas a baixos índices reprodutivos são observadas nos rebanhos comerciais. A utilização terapêutica de hormônios no intuito de induzir a lactação em animais hígidos é uma realidade. As doses hormonais utilizadas são elevadas e há a necessidade de experimentos que comprovem a possibilidade de redução. Nesse contexto, o objetivo desse projeto piloto foi reduzir as doses hormonais em vacas leiteiras submetidas a um protocolo de indução de lactação de 0,71mg/kg de peso vivo (PV) de benzoato de estradiol para 0,005mg/kg PV. Quatro vacas leiteiras receberam tratamento hormonal por 18 dias e iniciaram a lactação induzida após esse período. As vacas apresentaram produção de 32±4,3kg no pico lactacional induzido. A resposta lactacional das vacas foi considerada satisfatória e motivadora para experimentação de maior escala. Entretanto, são necessários estudos futuros que comprovem cientificamente o que foi encontrado nesse projeto piloto.


High culling rates of dairy cows related to low reproductive indices are found in commercial herds. The therapeutic use of hormones to induce lactation in healthy animals is a reality. The hormonal doses used are high and there is a need for experiments to prove the possibility of reduction. In this context, this study aimed to reduce the hormonal doses in dairy cows submitted to a lactation induction protocol from 0.71mg/kg of body weight (BW) of estradiol benzoate to 0.005mg/kg BW. Four dairy cows received hormone treatment for 18 days and started the induced lactation after this period. The cows had a production of 32±4.3kg at the induced lactational peak. The lactational response of the cows was considered satisfactory and motivating for larger scale experimentation. However, further studies are needed to scientifically prove what was observed in this pilot project.

This study aimed to determine the effect of antral follicle count (AFC), and pubertal status on the fertility of beef heifers. In this study, 230 Nelore heifers, 20±2 months of age, were subjected to an estradiol progesterone-based timed artificial insemination (TAI) program. On Day 0 of the TAI protocol, the heifers were examined by transrectal ultrasound to record videos of the ovaries. Later, in the darkroom of the laboratory of images, the videos were analyzed for AFC (≥ 3 mm) of each ovary. Females who failed the first TAI were resynchronized with the same hormonal protocol. The pregnancy status was evaluated by ultrasonography 30 days after each FTAI. The general mean of the AFC was 22.0 follicles. Thus, the heifers were divided into 2 groups according to AFC: Low AFC (˂ 22 follicles, n = 114), and High AFC (≥ 22 follicles, n = 116). No differences (P > 0.05) in the pregnancy per AI (P/AI) were observed between the Low and High AFC groups, and between pubertal and prepubertal categories. The P/AI was not different between heifers that displayed or did not estrus (P = 0.2). However, considering the estrus response of each AFC group, High AFC heifers that displayed estrus had greater P/AI (P = 0.01) than High AFC heifers that did not display estrus. In summary, AFC and pubertal status did not affect the fertility of Nelore heifers. In contrast, the P/AI of heifers that did not display estrus was lower than heifers observed in estrus only in the High AFC group.(AU)


Esse estudo teve como objetivo determinar o efeito da contagem de folículos antrais (CFA) e da maturidade sexual na fertilidade de novilhas de corte. Neste estudo, 230 novilhas Nelore, com 20 ± 2 meses de idade, foram submetidas a um protocolo de inseminação em tempo-fixo (IATF) a base de estradiol e progesterona. No Dia 0 do protocolo de IATF as novilhas foram examinadas por ultrassonografia transretal e vídeos dos ovários foram gravados para posterior CFA (≥ 3 mm) realizada na sala escura do laboratório de imagens. Trinta dias após a ultrassonografia, as fêmeas que falharam na primeira IATF foram ressincronizadas com o mesmo protocolo hormonal. A prenhez foi avaliada por ultrassonografia 30 dias após cada IATF. A média geral da CFA foi de 22 folículos; assim, as novilhas foram divididas em 2 grupos de acordo com a CFA: CFA baixa (˂ 22 folículos, n=114) e CFA alta (≥ 22 folículos, n=116). A prenhez por IA (P/IA) foi semelhante (P > 0,05) entre os grupos CFA baixa e alta e entre novilhas púberes e pré-púberes. A P/IA não foi diferente entre as novilhas que apresentaram ou não cio (P = 0,2). No entanto, novilhas com CFA alta que apresentaram cio tiveram maior P/IA (P = 0,01) do que novilhas com CFA alta que não apresentaram cio. Em conclusão, a CFA e a maturidade sexual não afetaram a fertilidade de novilhas. Por outro lado, a P/IA das novilhas que apresentaram cio foi maior do que das novilhas não observadas em cio apenas no grupo CFA alta.(AU)

The aim of the current study is to evaluate gene expression patterns of LH (lhr) and estrogen (er) receptors and plasma steroid levels during testicular development in Genyatremus luteus. Males were histologically classified as immature (n=7), maturing (n=7) and mature (n=7), based on the cellular structure of their testes. Plasma 11-KT concentration recorded peak at the final maturation stage. The highest plasma 17α-OHP concentrations were observed at the immature stage; they decreased at the maturation and mature stages. On the other hand, 17ß-estradiol (E2) recorded higher concentrations at the maturation stage. Er expression has significantly increased along the maturational development of animals' testes. The mRNA observed for the LH receptor has decreased from immature to maturing stage; it presented expression peak at the mature stage. There was high association between receptor gene expression and plasma steroid levels, mainly E2. The current study was the first to feature different reproductive maturation stages in male G. luteus specimens, based on cellular, endocrine and molecular aspects. In addition, it has shown that the gene expression profile for er and lhr receptors, as well as plasma 11-KT and E2 concentrations, are directly linked to testicular maturation, although they are not necessarily associated with the gonadosomatic index.


O objetivo deste estudo foi avaliar os padrões de expressão gênica dos receptores de LH (lhr) e de estrogênio (er) e dos níveis de esteróides plasmáticos durante o desenvolvimento testicular de Genyatremus luteus. Os machos foram classificados histologicamente em imaturos, em maturação e maduros, de acordo com a estrutura celular dos testículos. A concentração plasmática de 11-KT apresentou um pico na fase de maturação final (P<0.05). As maiores concentrações plasmáticas de 17α-OHP foram encontradas no estádio imaturo (P<0.05), com consequente diminuição nos estádios em maturação e maturo. O 17ß-estradiol (E2) apresentou maiores níveis de concentração no estádio em maturação (P<0.05). A expressão de er aumentou significativamente ao longo do desenvolvimento maturacional dos testículos (P<0.05). O mRNA para o receptor de LH diminuiu do estádio imaturo para o estádio em maturação (P<0.05) com consequente pico de expressão no estádio maduro. Houve alta relação entre a expressão gênica dos receptores e os níveis de esteróides plasmáticos, especialmente com E2. Em conclusão, este estudo caracterizou pela primeira vez, sob os aspectos celular, endócrino e molecular, os diferentes estádios de maturação reprodutiva em machos de G. luteus, demonstrando que o perfil da expressão gênica para os receptores er e lhr, bem como as concentrações plasmáticas de 11-KT e E2 foram diretamente relacionados à maturação testicular, apesar de não se relacionarem necessariamente com o índice gonadossomático.

In the present study, we investigate the effect of the presence or absence of corpus luteum (CL) at the beginning of a fixed-time artificial insemination (FTAI) protocol and to evaluate the impact of one-time use of intravaginal progesterone device (P4 device) in cows with or without CL. A total of 776 primiparous Nellore cows were subjected to FTAI approximately 45 days postpartum. In Experiment 1, 476 cows were divided into two experimental groups: with (CL-present, n=113) or without (CL-absent, n=363) CL, after ultrasound evaluation. On day 0 (D0), all cows received a new P4 device (1.0 g) and 2.0 mg estradiol benzoate (EB). Eight days later (D8), the P4 devices were withdrawn, and prostaglandin (15 mg), estradiol cypionate (0.5 mg), and eCG (300 IU) were administered i.m. All cows were inseminated 48 h after P4 device withdrawal (D10). In Experiment 2, the cows (n= 300) received (at D0) P4 devices that were previously used once in other cows with (n=109) or without CL (n=191) and 2 mg of EB. The same protocol as that used in Experiment 1 was performed from D8 onwards. In Experiment 1, the overall conception rate after FTAI was 55% (262/476). No difference was found in the conception rate between CL-present and CL-absent cows (52.2 vs. 55.5%). In Experiment 2, the conception rate obtained with P4 devices previously used in cows with CL (58.7%) was greater (P<0.05) than that obtained with P4 devices previously used in cows without CL (42.9%). Thus, this strategy resulted in a 15.8% increase in conception rate. In conclusion, the presence or absence of CL at the beginning of the FTAI protocol did not affect the conception rate in cows synchronized with the new P4 device, but the insertion of P4 devices previously used in cows with CL enhanced the conception rates in cows without CL.


No presente estudo, investigamos o efeito da presença ou ausência de corpo lúteo (CL) no início de um protocolo de inseminação artificial em tempo fixo (IATF) e avaliamos o impacto do uso único de dispositivo intravaginal de progesterona (dispositivo P4) em vacas com ou sem CL. Um total de 776 vacas Nelore primíparas, aproximadamente 45 dias pós-parto foram submetidas à IATF. No Experimento 1, após avaliação ultrassonográfica, 476 vacas foram divididas em dois grupos experimentais: com (CL-presente, n=113) ou sem (CL-ausente, n=363) CL. No dia 0 (D0), todas as vacas receberam um novo dispositivo de P4 (1,0 g) e 2.0 mg de benzoato de estradiol (BE). Após 8 dias (D8), os dispositivos P4 foram retirados e prostaglandina (15 mg), cipionato de estradiol (0,5 mg) e eCG (300 UI) foram administrados i.m. Todas as vacas foram inseminadas 48 horas após a retirada do dispositivo de P4 (D10). No Experimento 2, as vacas (n= 300) receberam (no D0) um dispositivo de P4 previamente utilizado uma única vez em outras vacas com (n=109) ou sem CL (n=191) e 2 mg de BE. O mesmo protocolo utilizado no Experimento 1 foi realizado a partir do D8. No experimento 1, a taxa geral de concepção após IATF foi de 55% (262/476). Não foi encontrada diferença na taxa de concepção entre as vacas com CL presente e CL ausente (52,2 vs. 55,5%). No Experimento 2, a taxa de concepção obtida com dispositivos P4 previamente utilizados em vacas com CL-presente (58,7%) foi maior (P<0,05) quando comparada aos dispositivos P4 previamente utilizados em vacas com CL-ausente (42,9%). Essa estratégia resultou em aumento de 15,8% na taxa de concepção. Em conclusão, a presença ou ausência de CL no início do protocolo de IATF não afetou a taxa de concepção em vacas sincronizadas com dispositivo novo de P4; e a eficácia dos dispositivos de P4 previamente utilizados em vacas com CL é maior durante seu segundo uso em vacas sem CL.

A IATF é uma biotecnologia da reprodução que permite o melhoramento genético dos rebanhos de corte, pois possibilita a sincronização da ovulação e realização da inseminação artificial sem necessidade de detecção do cio. O objetivo deste estudo, foi avaliar a eficiência da pré-sincronização com a utilização prévia de progesterona (P4) injetável, sua influência e efeitos na taxa de prenhez, por meio da realização de um protocolo de IATF em vacas de corte multíparas, em quatro fazendaslocalizadas no Mato Grosso do Sul-SP. Para o estudo foram utilizadas 396 vacas nelores, os animais de cada fazenda foram alocados em grupos distintos experimental e controle. Foram avaliadas 196 vacas pertencentes ao grupo controle, e 200 vacas tratadas com progesterona no total, sendo as fazendas descritas neste estudo como fazenda A, B, C e D. Dez dias antes do início do protocolo (D-10), foi aplicado progesterona injetável, via intramuscular em animais do mesmo lote, no D0 foi aplicado benzoato de estradiol e colocado o dispositivo intravaginal com liberação de progesterona, além da classificação dos animais que apresentavam ou não o corpo lúteo. No D8 o dispositivo foi retirado e aplicado uma dose de gonadotrofina coriônica equina, e cipionato de estradiol e de prostaglandina. A inseminação artificial em tempo fixo foi realizada no D10, com a utilização de sêmen de touros provados. O diagnóstico de gestação foi feito no D40, comparando os resultados das taxas de prenhez das vacas dos diferentes grupos, que foram tratadas ou não com progesterona de forma prévia ao protocolo. Sobre as taxas de prenhez dos animais tratados com progesterona, foram encontrados os resultados da fazenda A= 6,60%, fazenda B=10,80%, fazenda C= 3,90% e fazenda D= 3,70%, assim ficando evidente que essa biotecnologia da reprodução é lucrativa e contribui positivamente para a cadeia de produção de bovinos de corte. Baseado nos resultados, pode-se concluir que o uso da P4 (injetável) obteve êxito no índice de prenhez.(AU)


The IATF is a reproduction biotechnology that allows the genetic improvement of beef herds,as it allows the synchronization of ovulation and artificial insemination without the need for heat detection. The objective of this study was to evaluate the efficiency of pre-synchronization with the previous use of injectable progesterone (P4), its influence and effects on the pregnancy rate, through the performance of an IATF protocol in multiparous beef cows, in four farms located in Mato Grosso do Sul-SP. For the study, 396 Nelore cows were used, the animals from each farm were allocated into different experimental and control groups. A total of 196 cows belonging to the control group were evaluated, and 200 cows treated with progesterone in total, being the farms described in this study as farms A, B, C and D. Ten days before the beginning of the protocol (D-10), injectable progesterone was administered , intramuscularly in animals of the same batch, on D0 estradiol benzoate was applied and the intravaginal device was placed with progesterone release, in addition to the classification of animals that had or did not have the corpus luteum. On D8, the device was removed and a dose of equine chorionic gonadotropin, estradiol and prostaglandin cypionate was applied.Fixed-time artificial insemination was performed on D10, using semen from proven bulls. The pregnancy diagnosis was made on D40, comparing the results of the pregnancy rates of the cows of the different groups, which were treated or not with progesterone prior to the protocol. From the pregnancy rates of animals treated with progesterone, the results were found for farm A= 6.60%, farm B= 10.80%, farm C= 3.90% and farm D= 3.70%, thus becoming evident that this reproductive biotechnology is profitable and contributes positively to the beef cattle production chain. Based on the results, it can be concluded that the use of P4 (injectable) was successful in the pregnancy rate.(AU)


La IATF es una biotecnología de la reproducción que permite la mejora genética de rebaños de carne, ya que permite sincronizar la ovulación y la inseminación artificial sin necesidad de detección de celo. El objetivo de este estudio fue evaluar la eficiencia de la presincronización con el uso previo de progesterona inyectable (P4), su influencia y efectos en la tasa de preñez, mediante la realización de un protocoloIATF en vacas de carne multíparas, en cuatro fincas ubicadas en Mato Grosso do Sul-SP. Para el estudio se utilizaron 396 vacas Nelore, los animales de cada finca se distribuyeron en diferentes grupos experimentales y de control. Se evaluaron un total de 196 vacas pertenecientes al grupo control, y 200 vacas tratadas con progesterona en total, siendo las fincas descritas en este estudio como fincas A, B, C y D. Diez días antes del inicio del protocolo (D-10), se administró progesterona inyectable, por vía intramuscular en animales del mismo lote, sobre D0 se aplicó benzoato de estradiol y se colocó el dispositivo intravaginal con liberación de progesterona, además de la clasificación de animales que tenían o no cuerpo lúteo. El D8 se retiró el dispositivo y se aplicó una dosis de gonadotropina coriónica equina, estradiol y cipionato de prostaglandina. Se realizó inseminación artificial a tiempo fijo en D10, utilizando semen de toros probados. El diagnóstico de preñez se realizó el D40, comparando los resultados de las tasas de preñez de las vacas de los diferentes grupos, que fueron tratadas o no con progesterona previo al protocolo. A partir de las tasas de preñez de los animales tratados con progesterona se encontraron los resultados para la finca A= 6.60%, finca B= 10.80%, finca C= 3.90% y finca D= 3.70%, evidenciándose así que esta biotecnología reproductiva es rentable y contribuye positivamente a la cadena productiva del ganado vacuno de carne. Con base en los resultados, se puede concluir que el uso de P4 (inyectable) fue exitoso en la tasa de embarazo.(AU)

Prepubertal Nelore (G-N = 15) and crossbred Nelore x Aberdeen Angus heifers (G-NA = 15) were used for this study. AFC, live weight, body condition score (BCS), ovary and dominant follicle (DF) diameters were determined in each animal. Puberty induction was performed by insertion of a 4th use progesterone device (D0) which was removed on D12. Also, 1 mg estradiol benzoate was administered, and estrus intensity was classified (D12). At D21, the presence and diameter of the corpus luteum (CL) were registered. AFC was highly repeatable, regardless of hormone induction in both G-N (r=0.79) and G-NA (r=0.90). The mean AFC was greater in G-N compared to G-NA (24.2±8.5 vs. 17.7±9.0 follicles). A variation in BCS throughout the study occurred in G-NA, but not in G-N. The average weight gain (AWG) was greater in G-NA compared to G-N (0.69±0.33 vs. 0.40±0.29kg/day). The G-NA resulted in a larger diameter of DF at D12 than G-N (11.6±2.7 vs. 9.3±1.5mm). In conclusion, AFC was greater in Nelore heifers, although in both breeds this count was highly repeatable during puberty induction. Crossbred heifers had greater BCS and AWG with greater diameter of DF, indicating higher precocity when compared to Nelore heifers.


Novilhas pré-púberes Nelore (G-N=15) e mestiças Nelore x Aberdeen Angus (G-NA=15) foram utilizadas neste estudo. CFA, peso vivo, escore de condição corporal (ECC), diâmetros do ovário e folículo dominante (FD) foram determinados em cada animal. A indução da puberdade foi realizada pela inserção de um dispositivo de progesterona de quarto uso (D0), que foi retirado no D12. Além disso, 1mg de benzoato de estradiol foi administrado e a intensidade do estro foi classificada (D12). No D21, foram registrados a presença e o diâmetro do corpo lúteo (CL). A CFA foi altamente repetível, independentemente da indução hormonal em G-N (r=0,79) e G-NA (r=0,90). A CFA média foi maior em G-N em comparação com G-NA (24,2±8,5 vs. 17,7±9,0 folículos). Uma variação no ECC ao longo do estudo ocorreu em G-NA, mas não em G-N. O ganho de peso médio (GPM) foi maior em G-NA em comparação com G-N (0,69±0,33 vs. 0,40±0,29kg/dia). O G-NA resultou em um diâmetro maior de FD em D12 do que o G-N (11,6±2,7 vs. 9,3±1,5mm). Em conclusão, a CFA foi maior em novilhas Nelore, embora em ambas as raças essa contagem tenha sido altamente repetível durante a indução da puberdade. Novilhas mestiças apresentaram maior ECC e GPM com maior diâmetro de FD, indicando maior precocidade quando comparadas às novilhas Nelore.

The objective of this study was to investigate the effect of estrus expression, body condition score (BCS), different sires and semen batches on the conception rate of suckled Bos taurus beef cows submitted to timed artificial insemination (TAI). Data from 7,737 TAI were obtained from five farms in South Brazil. Cows received an estradiol plus progesterone-based estrous synchronization protocol. Only sires (n = 13) with at least two semen batches and 50 AI in two different lots were included in the study. Conception rate was higher for cows in estrus (66.8% vs. 26.3%) or with BCS ≥ 3 (57.2% vs. 51.6%) than in cows without estrus or BCS < 3, respectively (P≤0.001). Conception rate per sire ranged from 42.6% (Sire L) to 74.3% (Sire K). Conception rate in cows with BCS ≥ 3 and estrus occurrence varied from 47.4% (Sire L) to 86.1% (Sire A) among sires. Conception rate differed among semen batches of the same sire (B1 = 58.5%, B2 = 62.5%, and B3 = 83.2%). In conclusion, there was individual variation in conception rate among AI sires and semen batches within sires. The conception rate was increased by estrus occurrence and greater BCS.


O objetivo deste estudo foi investigar o efeito da manifestação de estro, o escore de condição corporal (ECC), os diferentes touros e partidas de sêmen na taxa de concepção de vacas Bos taurus lactantes de corte submetidas à inseminação artificial em tempo fixo (IATF). Dados de 7.737 IATF foram obtidos em cinco fazendas do sul do Brasil. As vacas receberam um protocolo de sincronização de estro à base de estradiol e progesterona. Somente touros (n=13) com no mínimo duas partidas e 50 inseminações em dois lotes diferentes foram incluídos no estudo. A taxa de concepção foi maior nas vacas em estro (66,8% vs. 26,3%) ou com ECC ≥ 3 (57,2% vs. 51,6%) do que em vacas sem estro ou com menor ECC, respectivamente (P≤0,001). A taxa de concepção por touro variou de 42,6% (Touro L) a 74,3% (Touro K). A taxa de concepção em vacas que demonstraram estro e com ECC ≥3 variou de 47,4% (Touro L) a 86,1% (Touro A) entre os touros. A taxa de concepção diferiu entre partidas de um mesmo touro (B1=58,5%, B2=62,5% e B3=83,2%). Em conclusão, houve variação individual na taxa de concepção entre touros e entre as partidas de sêmen de um mesmo touro. A taxa de concepção foi aumentada pela ocorrência de estro e maior ECC.

Background: Fixed Time Artificial Insemination (FTAI) has achieved a significant evolution in the last 18 years, however, despite the progress achieved by modern FTAI programs, the conception rates obtained are still low. Therefore, this study aimed to evaluate the interrelation between progesterone levels in the periovulatory period and reproductive parameters of Nellore cows submitted to an FTAI protocol. Materials, Methods & Results: On a random day, called day 0 (D0), 57 cows received a P4 device associated with the intramuscular (IM) application of 2.0 mg of estradiol benzoate. On D9, the P4 devices were removed and then were administered 500 µg of cloprostenol sodium IM; 0.6 mg of estradiol cypionate IM and 300 IUI of Equine Chorionic Gonadotrophin IM. Blood samples were collected for the determination of serum P4 concentrations on D9 and D11 of the protocol. The evaluations of follicular diameter (DFOL), follicular wall area (AFOL) and the vascularization area of the follicle wall (VFOL) were carried out on D11 using B-mode ultrasonography examination and colour Doppler, and then the artificial inseminations were performed. The evaluation of the corpus luteum diameter (CLD), of the total corpus luteum area (CLA), of the area of corpus luteum vascularization (CLV) and blood sampling for determination of postovulatory P4 levels (Post-P4) were performed on D24. For the analysis of the P4 concentration the chemiluminescence method was used, with a sensitivity of 0.1 ng/mL. According to the P4 concentrations on D11, cows were divided into 2 groups, LOW LEVELS OF P4 and HIGH LEVELS OF P4. The diagnosis of pregnancy was performed using transrectal ultrasonography on D45, at this point the cows were divided into 2 groups, PREGNANT and NON-PREGNANT. The correlation between DFOL and P4 dosage on D11 was moderate, negative and significant and between the AFOL and the serum P4 levels on D9, was moderate, negative and significant. As for the other correlations between follicular and luteal parameters and serum P4 levels, these were low to moderate, negative and not significant. Cows in the LOW LEVELS OF P4 group had significantly larger diameter and follicular areas than the cows in the HIGH LEVELS OF P4 group, the other follicular and luteal parameters showed no statistical difference. Of the total 57 cows that were inseminated, 30 cows became pregnant. Cows in the PREGNANT group had serum P4 levels on D9 equivalent to that obtained by the NON-PREGNANT group. However, at D11 the cows that became pregnant presented significantly lower serum P4 levels than cows that did not become pregnant. Discussion: The results of the interrelation between follicular parameters and P4 levels obtained in the present study, pointed out that the lower the levels of P4, the higher the follicular parameters, corroborating with other authors. Thus, larger preovulatory follicles provided high ovulation rates. Periovulatory serum P4 levels did not significantly affect the morphofunctional parameters of the CL. Such findings may be justified by high periovulatory P4 levels resulting from less efficient luteolysis, exert a negative effect on the results of FTAI protocols, because progesterone inhibits the release of LH pulses. It is concluded that lower periovulatory P4 levels established a favourable condition for follicular development and fertility, however, morphofunctional parameters of the corpus luteum were not affected.

A puberdade corresponde ao momento da primeira ovulação, que culmina com o desenvolvimento final dos órgãos e estruturas envolvidas no controle neuro-endócrino reprodutivo. É um fenômeno multifatorial com grande impacto na vida produtiva das futuras matrizes e em sua eficiência econômica, ocorrendo muitas interações entre os fatores internos e externos. A idade da puberdade está relacionada ao peso corporal, que possui grandes efeitos da genética do animal e sua interação com o ambiente, especialmente a nutrição e interação social com machos. Algumas espécies, como os bubalinos, sofrem efeito de outros elementos ambientais, como a luminosidade e o fotoperíodo. A puberdade novilhas búfalas ou bovinas pode ser adiantada por meio do emprego de biotécnicas: indução da puberdade ou inseminação artificial em tempo fixo (IATF). Em ambas, os protocolos baseiam-se, principalmente, em aplicação de progestágeno associado com fonte de estradiol; mas a indução também pode ser realizada utilizando outros hormônios, como hormônio liberador de gonadotrofinas, prostaglandinas e gonadotrofina coriônica equina. No emprego da IATF, novilhas pré-púberes podem ser induzidas e inseminadas com outro protocolo de sincronização ou podem ser inseminadas com o mesmo protocolo de IATF, podendo aumentar as taxas de prenhez e diminuir os custos. Adiantando a puberdade, ocorre incremento na produção, melhorando a eficiência econômica da atividade pecuária.(AU)


Puberty corresponds to the moment of the first ovulation, which culminates in the final development of the organs and structures involved in the neuro-endocrine control of reproduction. It is a multifactorial phenomenon with a great impact on the productive life of future matrices and their economic efficiency, with many interactions between internal and external factors. The age of puberty is related to body weight, which has great effects on the animal's genetics and its interaction with the environment, especially nutrition and social interaction with males. Some species, such as buffalo, are affected by other environmental elements, such as luminosity and photoperiod. Puberty in buffaloes and bovine heifers can be advanced through the use of biotechniques: induction of puberty or timed artificial insemination (TAI). In both, the protocols are mainly based on the application of progestogen associated with a source of estradiol; but induction can also be performed using other hormones, such as gonadotropin-releasing hormone, prostaglandins and equine chorionic gonadotropin. In the employment of the TAI, prepubertal heifers can be induced and inseminated with another synchronization protocol or they can be inseminated with the same TAI protocol, which can increase pregnancy rates and decrease costs. Advancing puberty, there is an increase in production, improving the economic efficiency of livestock activity.(AU)

Background: Buffalo breeding is common in many countries. Buffalo's milk is used in the production of mozzarella, yoghurt, ice cream, and various dairy desserts; meat is preferred in sausage production. The female buffaloes are bred to benefit from their milk and to obtain offspring. These animals, which are not suitable for feeding in barns, generally live in pastures, especially on wet land, and are very difficult to follow. Therefore, diseases occur randomly in slaughterhouses. Studies on genital system problems are very limited. Water buffaloes (Bubalus bubalis) are animals with economic value as many buffalo products are provided. In this respect, inspection and control of female genital disorders is fundamental to ensure good reproductive performance of female buffaloes. The aim of the study was to investigate pathomorphological lesions occurring in the ovaries of water buffaloes which were sent to slaughterhouses. Materials, Methods & Results: A total of 198 ovaries of water buffaloes were collected from various slaughterhouses located in Adapazari, Istanbul, Kocaeli, Samsun and Trabzon cities of Turkey. After macroscopic examination; tissue samples were fixed in 10 % buffered formalin, processed routinely and were stained with hematoxylin-eosin (HE). As a histochemical staining, Masson's trichrome staining was applied to characterize the lesions. Immunohistochemistry (IHC) was performed on 10 % neutral formalin fixed, paraffin-embedded, 4-6-µm-thick sections from ovaries using progesterone receptor (PR) with streptavidin-biotin complex peroxidase (StrepABC-P) method. Histopathologically, follicular cysts (n:147) and luteal cysts (n:22) were seen. While the cyst lumens were sometimes surrounded by granulosa and/or luteal cells, most of them were limited by the connective tissue capsule structure. This capsule structure was shown in blue by Masson's trichrome staining. Hemorrhage was observed in some cystic corpus luteum, diagnosed as corpus hemorrhagicum. In addition, cysts giving papillary extension into the lumen and inflammation of some ovaries were observed. Immunohistochemically, the staining with PR antibody in ovarian cysts showed no immunolabelling around the follicular cyst, while the nuclei of some of the luteal cells forming the luteal cyst had strongly nuclear positivity and slightly cytoplasmic positivity. In the biochemical examination of the fluids obtained from cystic ovaries (n: 37), the average of estradiol was 2.84 ng/mL (min: 0.01 ng/mL, max: 4.30 ng/mL) and progesterone average is 49.09 ng/mL (min: 1.88 ng/mL, max: 254.2 ng/mL). Discussion: Ovarian cysts in buffaloes seem to be among the serious fertility problems as in cattle. Although the exact cause of ovarian cyts has not been determined yet, it is known that the hypothalamic-pituitary-gonadal (HPG) axis is disrupted in cyst formation in general. In this study, the mean estradiol value was within the standard range but close to the lower limit; progesterone value was above the limit. The increase in progesterone level was compatible with the pathogenesis of cyst genesis. Beside this result, staining with PR was positive in the luteal cells that formed the luteal cyst immunohistochemically. In addition, although the hemorrhages observed in the corpus luteum are considered physiological, it should not be forgotten that they can be vital if they rupture. All these results show us the animals sent for slaughtering mostly have serious genital problems threatening their fertility.

The aim of this study was to assess in vitro sperm characteristics and pregnancies/AI (P/AI) of conventional and sex-sorted semen at timed-AI of suckled, multiparous Nelore cows. All cows (n=348) were submitted to a traditional estradiol/progesterone (P4)-based protocol. At 48h after P4-device removal, the estrous behavior was recorded, and AI was performed with conventional or sex-sorted semen from two bulls. The following sperm assessments were performed: CASA, Hyposmotic Test, sperm morphometry and chromatin structure by TB staining. P/AI were reduced (P<0.001) for sex-sorted compared to conventional semen in cows expressing estrus (27vs47%) or not (11vs.37%). Membrane integrity (Bull1: 30.3±9.6 vs. 52.3±12.4%, P=0.01; Bull2: 24.5±3.0 vs. 48.7±1.6%, P=0.006) and sperm concentration (Bulll: 23.2±0.6 vs. 43.0±0.8x10 sperm/mL, P<0.001; Bull2: 25.1+2.8 VS. 42.1±0.7x10 sperm/mL; P<0.001) were reduced in sex-sorted compared to conventional semen, for both bulls. Total and progressive motility were reduced in sex-sorted semen for Bull1 (TM: 49.7±15.9 vs. 94.9±1.9%, P=0.007; PM: 16.7±3.4 vs. 44.1±13.2%, P=0.009) and no differences were detected for Bull2 (TM: 45.0±17.5 vs. 68.2±19.1%, P=0.098; PM: 12.8±4.7 vs. 30.0±13.0%, P=0.065). Sperm ellipticity from sex-sorted was lower than conventional semen for Bull2 (0.306±0.01 vs. 0.342±0.02, P=0.02) and no difference was detected for Bull1 (0.332±0.01 vs. 0.330±0.01, P=0.55). Reduced in vivo fertility was observed for sex-sorted semen, regardless of estrous behavior. In vitro sperm quality of sex-sorted semen was compromised for both bulls, but differently affected for each sire.


O estudo teve como objetivo avaliar características espermáticas in vitro e a taxa de concepção (TC) de sêmen convencional e sexado em um programa de IATF tradicional de vacas Nelore pós-parto. Todas as vacas (n=348) foram submetidas ao mesmo protocolo de IATF à base de estradiol e de progesterona. Após 48 horas da retirada do implante, foi determinada a expressão de estro dos animais e a IA foi realizada com sêmen convencional e sexado de dois touros Angus. As seguintes características espermáticas foram avaliadas: análise computadorizada do sêmen, teste hiposmótico, morfometria espermática e estrutura cromatinica por meio da coloração com azul de toluidina. A TC foi menor (P<0,001) para sêmen sexado comparado ao convencional, em vacas que expressaram estro (27 vs. 47%) e que não apresentaram estro (11 vs. 37%). A integridade da membrana plasmática (Touro 1: 30,319,6 vs. 52,3+12,4%, P=0,010; Touro 2: 24,5+3,0 vs. 48,7±1,6%, P=0,006) e a concentração espermática (Touro 1: 23,2±0,6 vs. 43,0±0,8x10 sperm/mL, P<0,001; Touro 2: 25,1+2,8 vs. 42,1 +0,7x10'sperm/mL, P<0,001) foram menores no sêmen sexado comparado ao convencional, para ambos os touros. Motilidades total e progressiva foram menores no sêmen sexado comparado ao convencional para o Touro 1 (MT: 49,7±15,9 vs. 94,9±1,9%, P=0,007; MP: 16,7+3,4 vs. 44,1+13,2%, P=0,009), enquanto diferenças não foram detectadas no Touro 2 (MT: 45,0±17,5 vs. 68,2±19,1%, P=0,098; MP: 12,8±4,7 vs. 30,0±13,0%, P=0,065). Elipticidade espermática do sêmen sexado foi menor do que do sêmen convencional no Touro 2 (0,306±0,01 vs. 0,342±0,02, P=0,020), mas não houve diferença no Touro 1 (0,332±0,01 vs. 0,330±0,01, P=0,552). Reduzida fertilidade in vivo foi observada para o sêmen sexado em relação ao convencional, independentemente da expressão de cio das vacas. A qualidade seminal in vitro do sêmen sexado foi comprometida para ambos os touros, mas diferentemente afetada para cada reprodutor.

Resumo

Although studies have shown positive effects of gonadotropin releasing hormone (GnRH) or prostaglandin F2α (PGF2α) at the moment of fixed-time artificial insemination (FTAI) in the conception rate (CR) of cattle, its effects on treatments based on progesterone (P4) and estradiol benzoate (EB) is still not conclusive. The objective of this study was (1) to evaluate the effect of a PGF2α analogue at FTAI in the CR of crossbred beef cows submitted to a 11d FTAI protocol based on P4 and EB; and (2) to describe the CR between PGF2α-treated and control cows in different body condition scores (BCS) and parity categories. Crossbred (½ Nellore and ½ Angus) beef cows were submitted to a synchronization protocol and randomly assigned into 2 groups: Control (n = 163), at FTAI cows received 2 mL of saline solution as a placebo, and PGF2α (n = 163), at FTAI cows were treated with PGF2α analogue (10 mg of dinoprost tromethamine). Pregnancy diagnosis was performed 33d post-FTAI. Binary logistic regression model was used to analyze the effect of PGF2α treatment on CR. There was no difference in CR between PGF2α and control groups (P > 0.05; odds ratio (OR) = 0.92; confidence interval (CI) = 0.59-1.4). A greater CR was found in heifers (P = 0.0006, OR = 2.65, CI = 1.61 - 4.38) and multiparous (P = 0.0006, OR = 2.12, CI = 1.04 - 4.3) when compared to primiparous cows. Cows with low BCS (4; 9-point scale) showed lower CR when compared with moderate BCS (5-6; 9-point scale) (P < 0.05; OR = 0.10; CI = 0.06 - 0.18). There was no numerical difference on CR between PGF2α-treated and control cows in different BCS and parity categories. The results suggested that the CR in this study was not influenced by 10 mg PGF2α analogue at FTAI.(AU)

Resumo

The ovulation mechanism is one of the fascinating physiological processes in reproductive biology in mammals. From the reproductive point of view, the species have been classified as spontaneous or induced ovulators. Although the release of GnRH followed by the preovulatory LH surge is shared between both types of ovulation, the stimulus to initiate GnRH release varies between both categories. In spontaneous ovulators, ovulation depends on the systemic concentration of ovarian steroids, however, in induced ovulators, different stimuli such as copulation, environmental, and social cues can facilitate or induce ovulation regardless of the increases in systemic estradiol concentration. In this review, we document evidence that a male-derived protein is the main factor responsible for inducing ovulation and also modulating the ovarian function in the domestic South American camelid, the llama. The neurotrophin beta-Nerve Growth Factor (β-NGF) is the principal factor present in the semen of llamas responsible for inducing ovulation in this species. After the intrauterine deposit of semen during mating, β-NGF is absorbed through the endometrium to reach the circulatory system, where it reaches the hypothalamus and stimulates GnRH release. The potential site of action of this neurotrophin at the brain has not been elucidated, however, hypotheses are raised that the factor may cross the blood-brain barrier and stimulate upstream neuronal networks that lead to the stimulation of GnRH-secreting neurons. It is possible that β-NGF could be sensed at the median eminence without crossing the blood-brain barrier. Finally, it has been observed that this factor is not only a powerful stimulator of ovulation but also has a luteotrophic effect, resulting in the development of a corpus luteum capable of secreting more progesterone when compared to other ovulation-stimulating analogues.(AU)
