Resumo

Abstract Tuberculosis is a communicable disease with high morbidity and mortality rates in developing countries. The study's primary objective is to compare conventional methods such as acid-fast bacillus (AFB) culture and microscopy with rapid diagnostic methods. The secondary objective is to compare histopathological and microbiological findings in suspected patients with tubercular lymphadenitis. A total of 111 samples (August 2018 to September 2019) of lymph nodes were processed for AFB microscopy, AFB cultures, drug-susceptibility testing (DST), histopathology, and Xpert Mycobacterium Tuberculosis (MTB)/resistance to Rifampin (RIF) assays. Out of 111 lymph node samples, 6 (5.4%) were positive for AFB smear microscopy, 84 (75.6%) were positive for AFB culture, 80 (70.7%) were positive on Gene Xpert, and 102 (91.8%) were indicative of tuberculosis for histopathology studies. Mycobacteria growth indicator tube (MGIT) culture positivity was 84 (75.6%) higher than solid Lowenstein-Jensen (LJ) culture 74 (66.6%). Positive cultures underwent phenotypic DST. Two cases were Multidrug-resistant (MDR) on DST, while three cases were Rifampicin resistant on Gene Xpert. The sensitivity of Genexpert was (62%) against the conventional AFB culture method. The poor performance of conventional lymphadenitis diagnostic methods requires early and accurate diagnostic methodology. Xpert MTB/RIF test can help in the treatment of multidrug-resistant TB cases. Nonetheless, rapid and conventional methods should be used for complete isolation of Mycobacterium tuberculosis.

Abstract The present study describes the haematological profile, feeding preference, and comparison of morphometric characters of blue rock pigeon (Columba livia) breeding pairs. For this purpose, 25 pairs (25 samples per sex) were sampled through Mist nets from district Okara and Bahawalnagar, Punjab, Pakistan. Birds were then anaesthetized with a combination of ketamine HCL (10 mg/kg) and diazepam (0.2 mg/kg) and subjected to morphometric measurements. 5µL blood also was taken from the jugular vein of each anaesthetized bird for haematological analysis. Few pairs were also dissected to remove gastrointestinal tracts (GITs) for food preferences. Results revealed that there are no significant differences in the haematological parameters and feeding preference of breeding pairs of Columba livia. The gut analysis further revealed, the major portion of gut contents consisted of pea and corn in most of the pairs. Regarding the mensural measurements, significant differences were recorded in the body weight, length of the longest primary feather, and chest circumference, whereas the rest of the studied parameters remain nonsignificant between sexes. So, it is concluded that apart from 3 morphometric parameters (body weight, length of longest primary feather and chest circumference), both sexes are alike in term of morphometry, haematology and food preference.


Resumo O presente estudo descreve o perfil hematológico, a preferência alimentar e a comparação de caracteres morfométricos de casais reprodutores de pombo-rocha (Columba livia). Para tanto, 25 pares (25 amostras por sexo) foram amostrados por meio de redes de névoa do distrito de Okara e Bahawalnagar, Punjab, Paquistão. As aves foram então anestesiadas com uma combinação de cetamina HCL (10 mg/kg) e diazepam (0,2 mg/kg) e submetidas a medidas morfométricas; 5 µL de sangue também foram retirados da veia jugular de cada ave anestesiada para análise hematológica. Poucos pares também foram dissecados para remover o trato gastrointestinal (GITs) para preferências alimentares. Os resultados revelaram que não há diferenças significativas nos parâmetros hematológicos e na preferência alimentar dos casais reprodutores de Columba livia. A análise intestinal revelou ainda que a maior parte do conteúdo intestinal consistia em ervilha e milho na maioria dos pares. Em relação às medidas mensurais, foram registradas diferenças significativas no peso corporal, comprimento da pena primária mais longa e circunferência torácica, enquanto os demais parâmetros estudados permanecem não significativos entre os sexos. Assim, conclui-se que além de três parâmetros morfométricos (peso corporal, comprimento da pena primária mais longa e circunferência torácica), ambos os sexos são semelhantes em termos de morfometria, hematologia e preferência alimentar.

Resumo

The purpose of this manuscript is to describe the clinical-pathological aspects of cardiac hypertrophy related to the presence of Corynebacterium spp. in three Didelphis albiventris cubs. In necropsy, macroscopically, in the heart, cardiomegaly, concentric hypertrophy of the ventricles and the ventricular septum were observed, with consequent reduction of the chamber. Microscopically, the primary lesions found in the heart were cardiomyocyte hypertrophy and necrosis, myocytolysis, and the presence of myriad basophilic bacteria. Liver fragments and endocardial swabs were sent for bacterial culture, in which pleomorphic Gram-positive rods grew, forming small and hemolytic colonies. Chemical tests demonstrated characteristics compatible with Corynebacterium spp. Thus, this report represents the first description of cardiac hypertrophy associated with Corynebacterium spp. in white-eared opossums cubs, representing an essential contribution to studying diseases in wild animals.(AU)

Fish stocking is a common management strategy in Brazil, yet no assessment has examined its real extent and features. The present work investigated fish stocking practices in Brazil between 2010 and 2019 to characterize propagule pressure, species composition, the use of non-native species, geographical and temporal trends, environment types, and responsible agencies. Based on information disseminated on the internet, the study detected 1,155 stocking events (ca. 115 events/year). In total, ca. 56.4 million fish were stocked, with an average of ca. 90 thousand fish/event. We found events in all Brazilian regions involving 436 municipalities and 21 states. Most events occurred in the Northeast region (66.3%), which received alone ca. 41 million fish. Reservoirs were the primary target environment, and the public sector conducted most events. Fish stocking involved 63 taxa, including 14 non-native taxa and three hybrids. Considering the events that informed composition, 62.4% released non-native species, which summed 19.7 million fish. These results provide a broad overview of fish stocking practices in Brazil, and reveal that this action is widely disseminated. Its frequent use, associated with the lack of proper criteria and the illegal stocking of non-native species, raise concerns about technical aspects, outcomes, and potential environmental impacts.


A estocagem de peixes é uma estratégia de manejo comum no Brasil, mas nenhum estudo investigou sua real extensão e características. O presente trabalho investigou práticas de estocagem de peixes realizadas no Brasil entre 2010 e 2019, para caracterizar a pressão de propágulos, composição de espécies, participação de espécies não-nativas, variação espaço-temporal, ambientes e órgãos responsáveis. Com base em informações divulgadas na internet, o estudo detectou 1155 eventos de estocagem (ca. 115 eventos/ano). No total, ca. 56,4 milhões de peixes foram liberados, com média de ca. 90 mil peixes/evento. Encontramos eventos em todas as regiões brasileiras, envolvendo 436 municípios e 21 estados. A região nordeste somou a maioria dos casos (66,3%), e sozinha recebeu cerca de 41 milhões de peixes. Os reservatórios foram o principal ambiente alvo, sendo o setor público o principal responsável pelas ações. A estocagem de peixes envolveu 63 táxons, incluindo 14 táxons não-nativos e três híbridos. Considerando os eventos que informaram a composição das espécies, 62,4% liberaram espécies não-nativas, que somaram 19,7 milhões de peixes. Estes resultados fornecem um amplo panorama das práticas de estocagem de peixes no Brasil, e revelam que essa ação de manejo é amplamente disseminada. Seu uso frequente, associado com a falta de critérios apropriados e a soltura ilegal de espécies não-nativas, suscita preocupações quanto a aspectos técnicos, resultados, e potenciais impactos ambientais.

Background: Pseudomonas aeruginosa is a gram-negative aerobic bacterium and non-glucose fermenting, that usually causes opportunistic infections in animals, including humans. It is rarely involved in primary disease. The antibioticresistant bacterial strains are mainly developed due to the inappropriate use of antibiotics, however treating P. aeruginosa infections can be difficult owing to their natural resistance to antibiotics. Furthermorer resistant microorganisms such as P. aeruginosa grow by developing biofilms. Inaccurate diagnoses and absence of adequate microbiological tests can cause difficulties in resolving cases. This report describes a case of chronic superficial infection in a bitch caused by multidrugresistant Pseudomonas aeruginosa (MDR-PA). Case: A 6-year-old bitch Shih Tzu, initially presented with an exudative erythematous lesion in the snout region, which progressed to deep lesions, and spread to the back and limbs; furthermore, the animal always experienced a fever before new wounds emerged. Lesion samples, collected using a swab and processed at the Veterinary Microbiology Laboratory of the Federal University of Jatai (UFJ), revealed the presence of Pseudomonas aeruginosa. The isolate was multidrug-resistant and a carrier of TEM and ppyR genes. In the diffusion disk antibiogram, the isolate was found resistant to 14 different antibiotics belonging to 6 classes. Antimicrobial resistance was also tested using the minimum inhibitory concentration (MIC) test against imipenem, ceftazidime, ciprofloxacin, ticarcillin + clavulanic acid and aztreonam present in the MIC test strip. Treatment with amikacin and muporicin proved to be effective; however, owing to lesions extending to the face and palpebral involvement, the animal lost its eyeballs. Discussion: Pseudomonas aeruginosa is frequently associated with nosocomial infections mainly affecting immunosuppressed patients. Among the antibiotics tested, the group with the highest number of ineffective antibiotics was beta-lactams, where sensitivity was only observed for ticarcillin and ceftazidime. Recent studies have demonstrated that ceftazidime can reduce biofilm volume, inhibit motility, and repress the expression of genes associated with bacterial adhesion in P. aeruginosa. Therefore, the production of biofilm in P. aeruginosa is an important virulence factor as it facilitates a stable environment for the microorganism, which protects the bacteria from contact with antimicrobials. In addition, prolonged exposure to a wide variety of antimicrobials creates an environment of selective pressure between microorganisms, facilitating the emergence of multidrug-resistant strains. Furthermore, it is now well recognized that low doses of antibiotics, administered during continuous and fluctuating treatments, can stimulate biofilm establishment and are partly responsible for biofilm-specific antimicrobial tolerance. The resistance profile of P. aeruginosa isolated from dogs varies considerably, and the presence of isolates with a possible biofilm production capacity represents a challenge for the interpretation of the antimicrobial susceptibility profile. Culture and antibiogram is fundamentally important, both clinically and in environmental monitoring, in addition to the use of antibiogram data for decision making in clinical treatment.

Background: Aural or auricular hematoma is an important and prevalent surgical condition in small animals practice, and commonly reported in companion pets. The condition is characterized by blood accumulation between the pinnae's dermal surface and the underlying perichondrium. In farm animals, most cases present surgical treatment with drainage of serosanguineous fluid from acute cases with clinical evolution ranging from 1 to 5 days. Therefore, the present work reports the clinical features, surgical treatment and outcome of aural hematomas in 3 small ruminants with a chronic evolution (7 to 20 days), detailing the post-surgical complications, such as wound infection and recurrence, and final cosmetic appearance of the pinnae. Cases: Upon physical examination, all animals were alert and presenting a good body condition score. Clinical alteration was restricted to a bilateral (Case 1) or unilateral (Cases 2 & 3) painless, and fluid-filled swelling, presenting doughy consistency masses within the fluid during pinna manipulation. The fluid-filled swellings were located on the concave (Case 1 - left ear & Case 2) and convex (Case 1 - right ear & Case 3) surface of the pinna. No primary pruritic disorders of the pinna were detected, and a diagnosis of traumatic aural hematoma was proposed. Due to the chronicity of the cases, surgical approach was advisable. The small ruminants underwent general anesthesia, and a linear incision over the skin overlying the hematoma was performed (Cases 1 & 2). After removal of blood and fibrin clots, the cavity was flushed and captonated size 0 nylon mattress sutures were performed on either side of the incision, in order to obliterate the dead space. Due to recurrence after 14 days, Case 2 was submitted to a modified surgical approach using a S-shaped incision combined to multiple drainage holes (MDH) using a disposable 6 mm biopsy punch. The same approach has performed in Case 3. Then, size 0 nylon mattress sutures followed by a tight protective pressure and absorbent bandage combined with an Elizabethan collar was applied. Postoperatively, Case 1 presented surgical site infection and the antibiotic was changed after bacterial culture and antibiogram results. Hospital discharge varied from 14 to 19 days' post-surgery. All animals presented some degree of ear retraction and a linear or S-shaped scar on the affected ear, but final cosmetic appearance was satisfactory to all owners. Discussion: Aural or auricular hematomas are a frequent disorder in dogs and cats clinical practice. In sheep, the few reported cases have been associated with pruritic disorders of the pinna, such as ticks infestation and sarcoptic mange. Whilst in goats, traumatic injuries, such as ear tagging and trauma from others goats, are considered the main cause, especially in breeds with pendulous ear. In our cases with chronic evolution (> 7 days), the most important clinical feature was the presence of doughy consistency masses within the fluid, suggesting matured blood and fibrin clots. A linear incision with captonated size 0 nylon mattress sutures was associated to post-surgical complications, such as wound infection and aural hematoma recurrence. Therefore, a S-shaped incision associated to creation of MDH, in order to achieve better drainage, was the chosen approach in the second surgery (Case 2) and on the subsequent patient (Case 3). To the best of our knowledge, this is the first report using the MDH approach in farm animals, that appears to be an effective treatment for chronic aural hematomas in small ruminants.

Abstract Resveratrol, a natural polyphenol found in tempeh, has not been investigated especially in vitro as a neuroprotective agent against 2-methoxyethanol (2-ME)-induced beta-amyloid cytotoxicity. Beta amyloid peptides (A) could initiate neurotoxic events and neuron-inflammatory response via microglial activation. However, it remains unknown whether the neurotoxic effect of beta-amyloid and/or associated with the potential of 2-ME to induce neurotoxic effects on primary culture of nerve cells induced by 2-ME. This study investigated potential neuroprotective of trans-resveratrol a promising agent tempeh and soybean seed coats-derived against beta-amyloid cytotoxicity on primary culture of nerve cells induced by 2-methoxyethanol. Biotium and MTT assays were used to analyze neurons, which were isolated from the cerebral cortex of fetal mice at gestation day 19 (GD-19). A standard solution of 2-methoxyethanol was dosed at 10 L. The cultured cells were randomly divided into the following groups: (1) 2-ME group + resveratrol standard, (2) 2-ME group + resveratrol isolated from tempeh, (3) 2-ME group + resveratrol isolated from soybean seed coats, and (4) the control group, without the addition of either 2-ME or resveratrol. Exposure of the primary cortical neuron cells to beta-amyloid monoclonal antibody pre-incubated for 24 h with 10 µL of 4.2 µg/mL resveratrol and 7.5 mmol/l 2-methoxy-ethanol additions. Here, we report that the addition of 2-ME and resveratrol (standard and isolated from tempeh) of cell culture at concentrations of 1.4, 2.8 and 4.2 µg/mL showed that the majority of neurons grew well. In contrast, after exposure to 2-ME and Beta-amyloid, showed that glial activated. These findings demonstrate a role for resveratrol in neuroprotective-neurorescuing action.

Chronic Progressive Lymphoedema (CPL) is a disabling disease of draft horses that affects lymphatic system function typically in the distal limbs. Deformities of distal limbs, especially of the pastern, with fibrosis and skin nodular lesions are consequences of disease progression. A 15-year-old Friesian stallion presented for evaluation with history of forelimb enlargement and nodule formations distal to the carpus for four years. Simple radiographs showed soft tissue nodular lesions and venous contrasted radiography showed intense enlargement of II common digital palmar vein of both forelimbs. Tissue culture tests revealed Streptococcus equi and Proteus mirabilis isolation and skin scraping test identified Chorioptes bovis mites. Histological examination revealed perivasculitis and lymph vessels distention. History, clinical and histological findings, and complementary exams suggested CPL diagnosis. We were unable to find previously published cases describing this disease in Brazil, where the increasing number of draft horses requires attention to this problem. The correct and early diagnosis substantially delays disease progression. Therefore, we highlight the need for nationwide propagation of these data to ensure better diagnosis and early treatment of future CPL cases.


O linfedema crônico progressivo (LCP) é uma enfermidade incapacitante de cavalos de tração que afeta o sistema linfático e se manifesta especialmente na região distal dos membros. A progressão da doença resulta em deformações na região distal dos membros, especialmente na quartela, com fibrose e lesões nodulares na pele. Foi atendido um garanhão da raça Friesian, de 15 anos de idade, com histórico de aumento de volume, nodulações e deformações na região distal dos membros torácicos, com início há aproximadamente quatro anos. Ao exame radiográfico simples, foram identificadas nodulações na pele e no subcutâneo e, ao exame venoso contrastado, identificou-se marcante dilatação II veia digital palmar comum em ambos os membros torácicos. Foi isolado Streptococcus equi e Proteus mirabilis ao cultivo tecidual e identificou-se sarna do tipo corióptica ao raspado de pele. No exame histopatológico, foram identificadas perivasculite e dilatação de vasos linfáticos. Os achados clínicos, histórico, de raça do animal e os exames complementares direcionaram o diagnóstico para o LCP. A pesquisa da literatura nacional não revelou descrições clínicas dessa enfermidade, mas, devido ao aumento do número de animais de tração no país nos últimos anos, a descrição dessa enfermidade se torna importante. O diagnóstico correto e precoce é necessário para retardar a progressão da doença. Nesse sentido, ressalta-se a relevância da divulgação nacional desses achados, a fim de contribuir para o diagnóstico precoce e permitir o retardo da evolução da doença.

Resumo

The objective of the present work was to evaluate the performance of a compartmentalized anaerobic reactor (CAR) followed by anaerobic filter in real scale, effluent treatment from fish fridge processing. The work was developed in a fish fridge located in the south of the state of Minas Gerais, which has an effluent treatment plant composed of static screen, grease removal device, CAR reactor and anaerobic filter. The monitoring happened trough physical-chemical parameters of fluids and effluents from all sampling points of the plant biweekly. The parameters evaluated were temperature, pH, chemical oxygen demand (COD), biochemical oxygen demand (BOD), total suspended solids (TSS), volatile suspended solids (VSS), volatile fatty acids (VFA) and alkalinity. Operational problems found in the primary treatment were not limiting for the system performance, which managed to meet the parameters of environmental legislation in Minas Gerais according to the efficiency of removal of COD, BOD and VSS from wastewater from fish processing.


O presente trabalho teve como objetivo avaliar o desempenho de reator anaeróbio compartimentado (RAC) seguido por filtro anaeróbio em escala real tratando efluentes do processamento de frigorífico de pescado. O trabalho foi desenvolvido em um frigorífico de pescado localizado no sul do estado de Minas Gerais, que possui uma estação de tratamento de efluentes composta por peneira estática, caixa de gordura, reator ABR e filtro anaeróbio. O monitoramento do sistema consistiu em um conjunto de análises físico-químicas dos afluentes e efluentes de todos os pontos de coleta de amostras da estação de tratamento coletados quinzenalmente. Os parâmetros avaliados foram temperatura, pH, demanda química de oxigênio (DQO), demanda bioquímica de oxigênio (DBO), sólidos suspensos totais (SST), sólidos suspensos voláteis (SSV), ácidos voláteis totais (AVT) e alcalinidade. Problemas operacionais encontrados no tratamento primário não foram limitantes para o sistema, que conseguiu atender parâmetros da legislação ambiental de Minas Gerais quanto à eficiência de remoção de DQO, DBO e SSV das águas residuárias do processamento de pescado. O sistema de tratamento é uma alternativa promissora para o tratamento de efluentes do processamento de pescado.

Resumo

Respiratory problems due to tracheobronchial foreign bodies (FBs) are unusual in horses; although equines kept in pastures eventually inhale FBs, as conifer twigs of Araucaria angustifolia. A 1,5-year-old Criolle foal was presented with hemoptysis, dyspnea, restlessness and fever (40.9 ºC rectal temperature). Complete blood count showed intense neutropenia, monocytosis, thrombocytopenia and hypoproteinemia. Treatment was carried out but no clinical improvement was observed. At the post-mortem examination, marked amount of dark red liquid was observed in the thoracic cavity (hemothorax). The lung parenchyma was diffusely consolidated, predominantly in the cranioventral area, associated with mild pleural fibrin deposition. The right primary bronchus was obliterated by a Araucaria angustifolia pine branch measuring 18 cm in length, with adjacent darkened areas (lung consolidation). Microscopically, there was diffuse necrosis with severe hemorrhage in the lungs, associated with marked neutrophilic inflammatory infiltrate, numerous coccoid bacterial aggregates, and fibrinous pleuritis. Additionally, there was diffuse alveolar edema and multifocal thrombosis. Lung fragments were submitted for bacterial culture and mixed bacterial growth was observed with a predominance of Streptococcus equi subsp. zooepidemicus. Inhalation of branches is not commonly reported in horses, but it must be included in the differential diagnoses of pneumonia, and attention should be taken when allowing horses to graze in areas where the plant occurs.


Problemas respiratórios devido a corpos estranhos (CEs) traqueobrônquicos são incomuns em equinos, embora cavalos em pastagem possam eventualmente aspirar CEs, como galhos de Araucaria angustifolia. Um potro Crioulo, 1,5 anos, apresentou hemoptise, dispneia, inquietação e temperatura retal de 40,9 ºC. O hemograma revelou intensa neutropenia, monocitose, trombocitopenia e hipoproteinemia. O tratamento foi realizado, mas sem sucesso. Na cavidade torácica, foi observada grande quantidade de líquido avermelhado livre (hemotórax). Os pulmões estavam difusamente consolidados, predominantemente cranioventral e com discreta deposição de fibrina sobre a superfície pleural. O brônquio principal direito estava obliterado por um ramo de pinheiro de Araucaria angustifolia com 18 cm de comprimento. Microscopicamente, notou-se necrose de coagulação pulmonar difusa com hemorragia severa, infiltrado inflamatório neutrofílico marcado, numerosos agregados bacterianos cocoides e pleurite fibrinosa. Fragmentos de pulmão foram submetidos ao isolamento bacteriológico e abundante crescimento misto com predominância de Streptococcus equi subsp. zooepidemicus foi observado. A inalação de grimpas de pinheiro não é comumente relatada em equinos, mas deve ser incluída nos diagnósticos diferenciais de pneumonia e deve-se ter atenção ao introduzir cavalos no campo com a presença da planta.

Background: Urethral tumors are uncommon in canines, represented predominantly by transitional cell carcinoma, marked by aggressive behavior associated with short life expectancy. Definitive diagnosis is achieved by histopathological analysis. Surgery associated with chemotherapy is the main therapeutic alternative. The aim of this paper is to report a case of primary transitional cell carcinoma of the urethra in a bitch, submitted to surgical treatment associated with conventional adjuvant chemotherapy and metronomic chemotherapy, achieving survival of 21 months, to date. Case: A 12-year-old bitch mixed breed was admitted at the Veterinary Hospital of Federal Rural University of Rio de Janeiro (UFRRJ), manifesting hematuria and urinary incontinence for one month. Physical examination indicated that vital parameters were within normal limits. Laboratory tests of blood count and serum biochemistry, urinalysis, culture, urinary antibiogram, and abdominal ultrasound were performed. The ultrasound image suggested a neoplastic urethral process. Because of the suspicion of neoplasm, a thoracic X-ray was performed, showing no evidence of metastasis. Computed tomography (CT) of the abdominal region was performed, revealing an expansive lesion in the urethra with loss of definition of the walls and urethral lumen extending to the level of the pelvic floor, measuring about 2.9 x 1.4 x 1.2 cm. After preanesthetic exams, the animal underwent resection and surgical anastomosis of the affected urethral region. The sectioned tissue was sent for histopathological analysis, with diagnosis of transitional cell carcinoma. Adjuvant chemotherapy was performed using carboplatin at a dose of 250 mg/m², intravenously, every 21 days for 6 sessions. After completing the protocol, abdominal ultrasonography was performed again, which showed a cicatricial process in the urethral region of the surgical excision, with no sign of tumor recurrence. A metronomic chemotherapy protocol was then started with cyclophosphamide at a dose of 15 mg/m², daily for 6 months, with periodic oncological follow-up. At the end of the period, the animal remained under periodic follow-up with ultrasound exams performed at 2-month intervals, and has been free from recurrence for 21 months until now. Discussion: Urethral neoplasms account for 0.5 - 2% of all canine tumors, and are represented mostly by transitional cell carcinomas. Clinical signs of urinary obstruction and hematuria are the most common, as reported in the patient in question. The definitive diagnosis is by histopathological examination. However, imaging tests such as computed tomography are important in the initial investigation and surgical planning. The indicated treatment is surgical resection of the mass with subsequent adjuvant chemotherapy or radiotherapy. The chemotherapy protocol associating cisplatin and piroxicam can achieve remission rates of 70%, but this association has a high nephrotoxic effect. In the present case, cisplatin was replaced by carboplatin in order to reduce the nephrotoxic effects of chemotherapy. The metronomic protocol using cyclophosphamide was used as maintenance therapy, at the end of the conventional chemotherapy protocol. Survival achieved was longer than that described in the literature in cases of transitional cell carcinomas in the urethral region. Therefore, multimodal therapy using surgery associated with conventional and metronomic chemotherapy, is an option in canines with urethral carcinoma.

Background: Osteomyelitis is defined as a bone inflammation involving the cortical and medullary regions, usually caused by the local invasion of opportunistic microorganisms. The inflammatory reaction of bone may extend to the periosteum and soft tissues, compromising adjacent structures far from the initially infected foci. Different classifications of transmission routes, gravity levels, and tissues involved in animal and human osteomyelitis are available. In humans, the infection can reach bone tissue by exogenous or hematogenous pathways. This paper reports an atypical case of mandibular pyogranulomatous osteomyelitis in an ewe caused by concomitant Pseudomonas aeruginosa and Lactococcus raffinolactis infection. Case: The animal presented a 1-month history of progressive mandibular enlargement refractory to conventional therapy. In a physical examination, an increased volume located in the ventrolateral region of the right ramus of the mandible was observed. Fine-needle aspiration of the lesion enabled isolation in bacteriological culture of Pseudomonas aeruginosa and Lactococcus raffinolactis using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDITOF MS). Besides support care procedures and antimicrobial treatment approaches for the sheep based on in vitro tests, the animal died due to the severity of the clinical signs and the progressive worsening of the general health status. The radiographic image examination of the mandibular region revealed a severe and infiltrative periodontal reaction, with a predominance of a great number of neutrophils and macrophages, necrotic areas, and bone destruction, characterized histologically as a pyogranulomatous rection. At post mortem examination, a large pyogranuloma was observed in the entire horizontal branch of the mandible as well, showing a dark yellowish content of coarse consistency, caseous appearance, and bone fragmentation. Discussion: Ovine mandibular osteomyelitis is a well-established bone inflammation involving the cortical and medullary regions, characterized clinically by local enlargement, asymmetry, pain sensitivity, edema, hyperthermia, infiltrate caseous or suppurative material, and bone rarefaction. In the current report, 1-month history of progressive enlargement of the mandibular region, prostration, and weight loss in an ewe were referred. Where clinical and epidemiological features, bacteriological, cytological, histological, and mass spectrometry diagnostic approaches were assessed to diagnostic. Most reports involving the etiology of ovine mandibular osteomyelitis have been diagnosed based on classical phenotypic tests. Here, the concomitant identification of P. aeruginosa and L. raffinolactis infection was possible using mass spectrometry (MALDI-TOF), highlighting the importance of molecular methods in the diagnosis of animal diseases. In addition, the differentiation between Lactococcus and Enterococcus species is difficult, which could underestimate the diagnosis of Lactococcus species as a primary pathogen from animal diseases. We report, for the first time, a fatal case of mandibular pyogranulomatous osteomyelitis in a sheep caused by Pseudomonas aeruginosa and Lactococcus raffinolactis coinfection.

Abstract The access to sufficient numbers of spermatogonial stem cells (SSCs) is a prerequisite for the study of their regulation and further biomanipulation. Rho kinase (ROCK) belongs to a family of serine/threonine kinases and involves in a wide range of fundamental cellular functions. The aim of the present study was to study the effect of ROCK inhibitor, Y-27632 (0.1-40 µM), during the primary culture of ovine SSCs. SSCs were collected from 3-5-month-old's lamb testes. The viability of SSCs, the apoptosis assay of SSCs, the intracellular reactive oxygen species (ROS) analysis, and the SSCs markers and apoptosis-related gene expressions were detected by MTT reduction assay, Annexin V-FITC/ Propidium Iodide (PI) dual staining, flow cytometry and real-time-PCR studies, respectively. Morphological analyses indicated that the 5-10 µM Y-27632 had an optimal effect on the number of presumptive SSCs colonies and the area covered by them after a 10 days culture. The cell viability, apoptosis and necrosis of SSCs after 10 days' culture were not affected in comparison with the control group, and the 20 µM of Y-27632 resulted in significantly decreased cell viability (P<0.05) and an increased necrosis of cells. On day 10 after culture, the expression of P53 was decreased with an increase from 0 to 10 µM in the Y-27632 dose. In the 20 µM Y-27632 group, the expressions of P53 and Bax were higher and the Bcl-2 was lower than other groups and these values were significantly different from 5 and 10 µM Y-27632 groups (P<0.05). The level of intracellular ROS was decreased with an increase in the Y-27632 dose from 5 to 20 µM in comparison with the control group. In conclusion, the present study demonstrated that Y-27632 at a concentration of 5-10 µM provided optimal culture conditions for the primary culture of ovine SSCs.
