RESUMO

Crop yield must increase to keep pace with growing global demand. Past increases in crop production have rarely been attributable to an individual innovation but have occurred when technologies and practices combine to form improved farming systems. Inevitably this has involved synergy between genotypic and management improvements. We argue that research focused on developing synergistic systems that overcome clear production constraints will accelerate increases in yield. This offers the opportunity to better focus and multiply the impact of discipline-focused research. Here we use the rainfed grain production systems of south-eastern Australia as a case study of how transformational change in water productivity can be achieved with research focused on genotype × management synergies. In this region, rainfall is low and variable and has declined since 1990. Despite this, growers have maintained yields by implementing synergistic systems combining innovations in (i) soil water conservation, (ii) crop diversity, (iii) earlier sowing, and (iv) matching nitrogen fertilizer to water-limited demand. Further increases are emerging from synergies between genetic improvements to deliver flowering time stability, adjusted sowing times, and potential dual-purpose use. Collaboration between agronomists, physiologists, and crop breeders has led to development of commercial genotypes with stable flowering time that are in early phases of testing and adoption.

RESUMO

Wheat is a major food crop, with around 765 million tonnes produced globally. The largest wheat producers include the European Union, China, India, Russia, United States, Canada, Pakistan, Australia, Ukraine and Argentina. Cultivation of wheat across such diverse global environments with variation in climate, biotic and abiotic stresses, requires cultivars adapted to a range of growing conditions. One intrinsic way that wheat achieves adaptation is through variation in phenology (seasonal timing of the lifecycle) and related traits (e.g., those affecting plant architecture). It is important to understand the genes that underlie this variation, and how they interact with each other, other traits and the growing environment. This review summarises the current understanding of phenology and developmental traits that adapt wheat to different environments. Examples are provided to illustrate how different combinations of alleles can facilitate breeding of wheat varieties with optimal crop performance for different growing regions or farming systems.

BACKGROUND: Good establishment is important for rapid leaf area development in wheat crops. Poor establishment results in fewer, later-emerging plants, reduced leaf area and tiller number. In addition, poorly established crops suffer from increased soil moisture loss through evaporation and greater competition from weeds while fewer spikes are produced which can reduce grain yield. By protecting the emerging first leaf, the coleoptile is critical for achieving good establishment, and its length and interaction with soil physical properties determine the ability of a cultivar to emerge from depth. RESULTS: Here we characterise a locus on chromosome 1AS, that increases coleoptile length in wheat, which we designate as Lcol-A1. We identified Lcol-A1 by bulked-segregant analysis and used a Halberd-derived population to fine map the gene to a 2 cM region, equivalent to 7 Mb on the IWGSC genome reference sequence of Chinese Spring (RefSeqv1.0). By sowing recently released cultivars and near-isogenic lines in the field at both conventional and deep sowing depths, we confirmed that Locl-A1 was associated with increased emergence from depth in the presence and absence of conventional dwarfing genes. Flanking markers IWB58229 and IWA710 were developed to assist breeders to select for long coleoptile wheats. CONCLUSIONS: Increased coleoptile length is sought in many global wheat production areas to improve crop emergence. The identification of the gene Lcol-A1, together with tools to allow wheat breeders to track the gene, will enable improvements to be made for this important trait.

Many upcoming experiments in antimatter research require low-energy antiproton beams. With a kinetic energy in the order of 100 keV, the standard magnetic components to control and focus the beams become less effective. Therefore, electrostatic components are being developed and installed in transfer lines and storage rings. However, there is no equipment available to precisely map and check the electric field generated by these elements. Instead, one has to trust in simulations and, therefore, depend on tight fabrication tolerances. Here we present, for the first time, a noninvasive way to experimentally probe the electrostatic field in a 3D volume with a microsensor. Using the example of an electrostatic quadrupole focusing component, we find excellent agreement between a simulated and real field. Furthermore, it is shown that the spatial resolution of the probe is limited by the electric field curvature which is almost zero for the quadrupole. With a sensor resolution of 61 V/m/sqrt[Hz], the field deviation due to a noncompliance with the tolerances can be resolved. We anticipate that this compact and practical field strength probe will be relevant also for other scientific and technological disciplines such as atmospheric electricity or safeguarding near power infrastructure.

OBJECTIVE: To describe clinically relevant, physiological measurements collected during a 3 hour duration of alfaxalone total intravenous anaesthesia. STUDY DESIGN: Case series. ANIMALS: A total of 112 client-owned middle-aged or older dogs. METHODS: Dogs were premedicated with intramuscular acepromazine (0.03 mg kg-1). Anaesthesia was induced and subsequently maintained for up to 3 hours with alfaxalone administered intravenously. Dogs breathed 100% oxygen via an endotracheal tube. Heart rate, respiratory rate and blood pressure were evaluated 30 minutes after administration of acepromazine and used as baseline values for comparisons of intra-anaesthetic data. Blood glucose was measured 1 week prior to anaesthesia and every hour during alfaxalone anaesthesia. Quality and duration of recovery were recorded. Mean data for physiological variables were compared over three time points-before induction of anaesthesia, for the first hour of anaesthesia and from 60 minutes to discontinuation of anaesthesia. RESULTS: Mean induction dose of alfaxalone was 1.4 mg kg-1 [95% confidence interval (CI) 1.3-1.5). Post induction apnoea for >60 seconds occurred in 13 (11.6%) dogs. Mean alfaxalone infusion rate during the first 60 minutes of anaesthesia was 0.099 mg kg-1 minute-1; mean infusion rate was 0.092 mg kg-1 minute-1 from 60 minutes until discontinuation of anaesthesia. Heart rate was well maintained; hypotension (mean arterial blood pressure < 60 mmHg) was encountered in 23 (21%) dogs. Blood glucose levels did not alter during anaesthesia. Median time between discontinuation of alfaxalone infusion and extubation was 17 (7-35 minutes), time to assuming sternal recumbency was 75 (58-110 minutes), and time to standing was 109 (88-140 minutes). CONCLUSIONS AND CLINICAL RELEVANCE: Alfaxalone infusion provided effective anaesthesia in this population. In a minority of cases, respiratory and haemodynamic support of the patient was required.

Identifying and reducing impacts from mercury sources in the environment remains a considerable challenge and requires process based models to quantify mercury stocks and flows. The stable isotope composition of mercury in environmental samples can help address this challenge by serving as a tracer of specific sources and processes. Mercury isotope variations are small and result only from isotope fractionation during transport, equilibrium, and transformation processes. Because these processes occur in both industrial and environmental settings, knowledge of their associated isotope effects is required to interpret mercury isotope data. To improve the mechanistic modeling of mercury isotope effects during gas phase diffusion, an experimental program tested the applicability of kinetic gas theory. Gas-phase elemental mercury diffusion through small bore needles from finite sources demonstrated mass dependent diffusivities leading to isotope fractionation described by a Rayleigh distillation model. The measured relative atomic diffusivities among mercury isotopes in air are large and in agreement with kinetic gas theory. Mercury diffusion in air offers a reasonable explanation of recent field results reported in the literature.

OBJECTIVE: To evaluate the sedative and analgesic effects of intramuscular buprenorphine with either dexmedetomidine or acepromazine, administered as premedication to cats and dogs undergoing elective surgery. STUDY DESIGN: Prospective, randomized, blinded clinical study. ANIMALS: Forty dogs and 48 cats. METHODS: Animals were assigned to one of four groups, according to anaesthetic premedication and induction agent: buprenorphine 20 µg kg(-1) with either dexmedetomidine (dex) 250 µg m(-2) or acepromazine (acp) 0.03 mg kg(-1), followed by alfaxalone (ALF) or propofol (PRO). Meloxicam was administered preoperatively to all animals and anaesthesia was always maintained using isoflurane. Physiological measures and assessments of pain, sedation and mechanical nociceptive threshold (MNT) were made before and after premedication, intraoperatively, and for up to 24 hours after premedication. Data were analyzed with one-way, two-way and mixed between-within subjects anova, Kruskall-Wallis analyses and Chi squared tests. Results were deemed significant if p ≤ 0.05, except where multiple comparisons were performed (p ≤ 0.005). RESULTS: Cats premedicated with dex were more sedated than cats premedicated with acp (p < 0.001) and ALF doses were lower in dex cats (1.2 ± 1.0 mg kg(-1) ) than acp cats (2.5 ± 1.9 mg kg(-1)) (p = 0.041). There were no differences in sedation in dogs however PRO doses were lower in dex dogs (1.5 ± 0.8 mg kg(-1) ) compared to acp dogs (3.3 ± 1.1 mg kg(-1) ) (p < 0.001). There were no differences between groups with respect to pain scores or MNT for cats or dogs. CONCLUSION: Choice of dex or acp, when given with buprenorphine, caused minor, clinically detectable, differences in various characteristics of anaesthesia, but not in the level of analgesia. CLINICAL RELEVANCE: A combination of buprenorphine with either acp or dex, followed by either PRO or ALF, and then isoflurane, accompanied by an NSAID, was suitable for anaesthesia in dogs and cats undergoing elective surgery. Choice of sedative agent may influence dose of anaesthetic induction agent.

Objective. The objective of this study was to separately quantify the stability of the megavoltage imager (MVI) and radiation head of an Elekta Unity MRL, throughout full gantry rotation.Approach. A ball-bearing (BB) phantom was attached to the radiation head of the Unity, while a single BB was placed at isocentre. Images were acquired during rotation, using the MVI. These images were processed using an in-house developed MATLAB program to reduce the errors resulted by noise, and the positions of the BBs in the images were analysed to extract MVI and radiation head sag data.Main results. The results returned by this method showed reproducibility, with a mean standard deviation of 7µm for the position of BBs across all gantry angles. The radiation head was found to sag throughout rotation, with a maximum course of movement of 0.59 mm. The sag pattern was stable over a period greater than a year but showed some dependence on gantry rotation direction.Significance. As MRL is a relatively new system, it is promising to have data supporting the high level of precision on one Elekta Unity machine. Isolating and quantifying the sources of uncertainty in radiation delivery may allow more sophisticated analysis of how the system performance may be improved.

Wheat (Triticum aestivum L.) is the most widely-grown crop in the Mediterranean semi-arid (150-400 mm) cropping zones of both southern Australia and the inland Pacific Northwest (PNW) of the United States of America (United States). Low precipitation, low winter temperatures and heat and drought conditions during late spring and summer limit wheat yields in both regions. Due to rising temperatures, reduced autumn rainfall and increased frost risk in southern Australia since 1990, cropping conditions in these two environments have grown increasingly similar. This presents the opportunity for southern Australian growers to learn from the experiences of their PNW counterparts. Wheat cultivars with an obligate vernalization requirement (winter wheat), are an integral part of semi-arid PNW cropping systems, but in Australia are most frequently grown in cool or cold temperate cropping zones that receive high rainfall (>500 mm p.a.). It has recently been shown that early-sown winter wheat cultivars can increase water-limited potential yield in semi-arid southern Australia, in the face of decreasing autumn rainfall. Despite this research, there has to date been little breeding effort invested in winter wheat for growers in semi-arid southern Australia, and agronomic research into the management of early-sown winter wheat has only occurred in recent years. This paper explores the current and emerging environmental constraints of cropping in semi-arid southern Australia and, using the genotype × management strategies developed over 120 years of winter wheat agronomy in the PNW, highlights the potential advantages early-sown winter wheat offers growers in low-rainfall environments. The increased biomass, stable flowering time and late-summer establishment opportunities offered by winter wheat genotypes ensure they achieve higher yields in the PNW compared to later-sown spring wheat. Traits that make winter wheat advantageous in the PNW may also contribute to increased yield when grown in semi-arid southern Australia. This paper investigates which specific traits present in winter wheat genotypes give them an advantage in semi-arid cropping environments, which management practices best exploit this advantage, and what potential improvements can be made to cultivars for semi-arid southern Australia based on the history of winter wheat crop growth in the semi-arid Pacific Northwest.

Wheat production in southern Australia is reliant on autumn (April-May) rainfall to germinate seeds and allow timely establishment. Reliance on autumn rainfall can be removed by sowing earlier than currently practiced and using late summer and early autumn rainfall to establish crops, but this requires slower developing cultivars to match life-cycle to seasonal conditions. While slow-developing wheat cultivars sown early in the sowing window (long-cycle), have in some cases increased yield in comparison to the more commonly grown fast-developing cultivars sown later (short-cycle), the yield response is variable between environments. In irrigated wheat in the sub-tropics, the variable response has been linked to ability to withstand water stress, but the mechanism behind this is unknown. We compared short- vs. long-cycle cultivars × time of sowing combinations over four seasons (2011, 2012, 2015, and 2016) at Temora, NSW, Australia. Two seasons (2011 and 2012) had above average summer fallow (December-March) rain, and two seasons had below average summer fallow rain (2015 and 2016). Initial plant available water in each season was 104, 91, 28, and 27 mm, respectively. Rainfall in the 30 days prior to flowering (approximating the critical period for yield determination) in each year was 8, 6, 14, and 190 mm, respectively. We only observed a yield benefit in long-cycle treatments in 2011 and 2012 seasons where there was (i) soil water stored at depth (ii) little rain during the critical period. The higher yield of long-cycle treatments could be attributed to greater deep soil water extraction (<1.0 m), dry-matter production and grain number. In 2015, there was little rain during the critical period, no water stored at depth and no difference between treatments. In 2016, high in-crop rainfall filled the soil profile, but high rainfall during the critical period removed crop reliance on deep water, and yields were equivalent. A simulation study extended our findings to demonstrate a median yield benefit in long-cycle treatments when the volume of starting soil water was increased. This work reveals environmental conditions that can be used to quantify the frequency of circumstances where long-cycle wheat will provide a yield advantage over current practice.

The Wheat Initiative (WI) and the WI Expert Working Group (EWG) for Agronomy (www.wheatinitiative.org) were formed with a collective goal to "coordinate global wheat research efforts to increase wheat production, quality, and sustainability to advance food security and safety under changing climate conditions." The Agronomy EWG is responsive to the WI's research need, "A knowledge exchange strategy to ensure uptake of innovations on farm and to update scientists on changing field realities." The Agronomy EWG aims to consolidate global expertise for agronomy with a focus on wheat production systems. The overarching approach is to develop and adopt a systems-agronomy framework relevant to any wheat production system. It first establishes the scale of current yield gaps, identifies defensible benchmarks, and takes a holistic approach to understand and overcome exploitable yield gaps to complement genetic increases in potential yield. New opportunities to increase productivity will be sought by exploiting future Genotype × Environment × Management synergies in different wheat systems. To identify research gaps and opportunities for collaboration among different wheat producing regions, the EWG compiled a comprehensive database of currently funded wheat agronomy research (n = 782) in countries representing a large proportion of the wheat grown in the world. The yield gap analysis and research database positions the EWG to influence priorities for wheat agronomy research in member countries that would facilitate collaborations, minimize duplication, and maximize the global impact on wheat production systems. This paper outlines a vision for a global WI agronomic research strategy and discusses activities to date. The focus of the WI-EWG is to transform the agronomic research approach in wheat cropping systems, which will be applicable to other crop species.

Fine particles or sediments have various effects on water quality and aquatic ecosystems. Thus, understanding the dynamics of these fine particles between water body and stream bed is an important issue in sediment research. Previous studies and analysis of empirical data suggest that fine particles are stored in the sediment bed in the low flow regime, where flow rate is smaller than the critical flow rate that mobilizes the sediment bed. These fine particles are re-suspended during flood events when the flow rate becomes larger than the critical flow rate that mobilizes bed material. The transition from pattern recognition to process analysis required incorporation of the dominant processes controlling fine particle dynamics within gravel-bedded streams into a model. The process analysis was performed using continuous flow and turbidity data at two locations on the Russian River in California to test process descriptions and then calibrate a quantitative model to represent those processes. The resulting process model coupled fine particle retention within the sediment bed by filtration and sedimentation with the release of accumulated fine particles in response to flood events. Model parameters, such as the critical flow rate required for initiating sediment bed fluidization, the maximum fine particle storage capacity within the sediment bed, and background particle concentration for the watershed, were estimated from the monitoring data. Model calibration optimized the filtration and the sediment bed fluidization parameters over two or three years of data. Overall, the difference between modeled and observed fine particle mass released from the sediment bed was within 20% of the measured mass.

In man, central sensitisation (CS) contributes to the pain of osteoarthritis (OA). Dogs with spontaneous OA may also exhibit CS. Electrophysiological reflex measurements are more objective than behavioural assessments and can be used to evaluate CS in preclinical and clinical studies. It was hypothesised that dogs suffering from OA would exhibit electrophysiological characteristics indicative of CS, associated with reduced diffuse noxious inhibitory controls (DNICs). One hundred and seventeen client-owned dogs were recruited to the study. Hind limb nociceptive withdrawal reflex thresholds, stimulus response, and temporal summation characteristics were recorded, during alfaxalone anaesthesia, from 46 OA dogs, 29 OA dogs receiving nonsteroidal anti-inflammatory drugs (OANSAIDs), and 27 breed- and weight-matched control dogs. Efficacy of DNIC was evaluated in 12 control and 11 of the OA dogs, by application of a mechanical conditioning stimulus to the contralateral forelimb. Nociceptive withdrawal reflex thresholds were higher in OA compared with control dogs (P = 0.02). Stimulus response characteristics demonstrated an augmented response in OANSAID dogs compared with OA (P < 0.001) and control (P < 0.001) dogs. Temporal summation demonstrated exaggerated C-fibre-mediated responses in both OA (P < 0.001) and OANSAID (P = 0.005) groups, compared with control animals. Conditioning stimulus application resulted in inhibition of test reflex responses in both OA and control animals (P < 0.001); control animals demonstrated greater inhibition compared with OA (P = 0.0499). These data provide evidence of neurophysiological changes consistent with CS in dogs with spontaneous OA and demonstrate that canine OA is associated with reduced DNIC.

The transport of fluids miscible with water arises in groundwater contamination and during remediation of the subsurface environment. For concentrated salt solutions, i.e., brines, the increased density and viscosity determine mixing processes between these fluids and ambient groundwater. Under downward flow conditions, gravitational and viscous forces work against each other to determine the interfacial mixing processes. Historically, mixing has been modeled as a dispersive process, as viscous fingering, and as a combination of both using approaches that were both analytical and numerical. A compilation of previously reported experimental data on vertical miscible displacements by fluids with significant density and viscosity contrasts reveals some agreement with a stability analysis presented by Hill (1952). Additional experimental data on one-dimensional dispersion during downward displacement of concentrated salt solutions by freshwater and freshwater displacement by brines support the stability analysis and provides an empirical representation for dispersion coefficients as functions of a gravity number and a mobility ratio.

This study aimed to analyse UK pharmacovigilance data to quantify adverse events (AEs) associated with the non-steroidal anti-inflammatory drug (NSAID) molecules found in veterinary medicines authorised for use in dogs and cats. It was hypothesised that the frequency of AEs would be lower when associated with cyclo-oxygenase-2 selective (coxib), compared to non-selective (non-coxib) NSAIDs. The UK Veterinary Medicines Directorate (VMD) supplied frequencies of AEs derived from Periodic Safety Update Reports subdivided by formulation and species for each NSAID molecule. Frequencies of AEs were similar between species. The five most reported AEs were emesis, death, anorexia, lethargy, and diarrhoea. Reported frequency of emesis, renal insufficiency and death was higher with injectable compared to oral NSAIDs (P = 0.043). Reported frequency of emesis, lethargy and death was higher with coxib, compared to non-coxib NSAIDs (P = 0.029). Median (range) interval since authorisation was shorter for coxibs at 5 (2.5-9) years compared to non-coxibs at 15 (12-25) years. A negative correlation between time elapsed since authorisation and the frequency of AEs was identified (rs = -0.11 to -0.94). Higher frequency of reported AEs with injectable NSAIDs may be related to perioperative administration. The AE frequency associated with coxib and non-coxib NSAIDs may be confounded by changes in reporting habits over time. This study highlights the value of interrogating passive surveillance data to identify low frequency AEs and the need to facilitate improvement in recording and collecting AEs in small animal practice.

Wind resuspension (or entrainment) provides a source of dust and contaminants for the atmosphere. Conventional wind erosion models parameterize dust resuspension flux with a threshold velocity or with a horizontal abrasion flux; in the absence of abrasion the models assume dust flux is transient only. Our experiments with an uncrusted, fine material at relative humidities exceeding 40% show a long-term steady dust flux in the absence of abrasion, which fits the approximate form: F(d) = 3.6(u*)(3), where F(d) is the dust flux (in mug/m(2) s), and u* is the friction velocity (in m/s). These fluxes are generally too small to be significant sources of dust in most models of dust emission. However, they provide a potential route to transport contaminants into the atmosphere. In addition, dust release is substantial during the initial transient phase. Comparison with field data suggests that the particle friction Reynolds number may prove a better parameter than u* for correlating fluxes and understanding the potential for abrasion.

Many non-aqueous phase liquids (NAPLs) are expected to spread at the air-water interface, particularly under non-equilibrium conditions. In the vadose zone, this spreading should increase the surface area for mass transfer and the efficiency of volatile NAPL recovery by soil vapor extraction (SVE). Observations of spreading on water wet surfaces led to a conceptual model of oil spreading vertically above a NAPL pool in the vadose zone. Analysis of this model predicts that spreading can enhance the SVE contaminant recovery compared to conditions where the liquid does not spread. Experiments were conducted with spreading volatile oils hexane and heptane in wet porous media and capillary tubes, where spreading was observed at the scale of centimeters. Within porous medium columns up to a meter in height containing stagnant gas, spreading was less than ten centimeters and did not contribute significantly to hexane volatilization. Water film thinning and oil film pinning may have prevented significant oil film spreading, and thus did not enhance SVE at the scale of a meter. The experiments performed indicate that volatile oil spreading at the field scale is unlikely to contribute significantly to the efficiency of SVE.

Horizontal infiltration experiments were performed to validate a plug flow model that minimizes the number of parameters that must be measured. Water and silicone oil at three different viscosities were infiltrated into glass beads, desert alluvium, and silica powder. Experiments were also performed with negative inlet heads on air-dried silica powder, and with water and oil infiltrating into initially water moist silica powder. Comparisons between the data and model were favorable in most cases, with predictions usually within 40% of the measured data. The model is extended to a line source and small areal source at the ground surface to analytically predict the shape of two-dimensional wetting fronts. Furthermore, a plug flow model for constant flux infiltration agrees well with field data and suggests that the proposed model for a constant-head boundary condition can be effectively used to predict wetting front movement at heterogeneous field sites if averaged parameter values are used.

In unsaturated soil, methane and volatile organic compounds can significantly alter the density of soil gas and induce buoyant gas flow. A series of laboratory experiments was conducted in a two-dimensional, homogeneous sand pack with gas permeabilities ranging from 110 to 3,000 darcy. Pure methane gas was injected horizontally into the sand and steady-state methane profiles were measured. Experimental results are in close agreement with a numerical model that represents the advective and diffusive components of methane transport. Comparison of simulations with and without gravitational acceleration permits identification of conditions where buoyancy dominates methane transport. Significant buoyant flow requires a Rayleigh number greater than 10 and an injected gas velocity sufficient to overcome dilution by molecular diffusion near the source. These criteria allow the extension of laboratory results to idealized field conditions for methane as well as denser-than-air vapors produced by volatilizing nonaqueous phase liquids trapped in unsaturated soil.
