RESUMEN

Similarity evaluation of complicated chromatographic profiles is a potential protocol for the identification and quality control of herbal medicinal products to ensure their biological activity. In this work, a high-performance liquid chromatography method was established for controlling the batch quality of the extract from Portulaca oleracea L. Using this method, the coefficients of correlation of the similarity of 10 batches extract of P. oleracea L. were ≥ 0.97. The 10 batch extracts from P. oleracea L. possessed stable antiproliferative activity in Aspergillus flavus. The antiproliferative activity stability is correlated with the stability quality of the of the extract from P. oleracea L. Therefore, the present study successfully set up a sensitive and efficient method which might be used to guarantee stable biological activity of the extract from P. oleracea L.

Oxidative stress is a phenomenon caused by an imbalance between the production and accumulation of reactive oxygen species in cells and tissues that eventually leads to the production of various diseases. Here, we investigated the antioxidant effects of the extract from Sonchus brachyotus DC. (SBE) based on the 0.2% oxazolone-induced intestinal oxidative stress model of zebrafish. Compared to the model group, the treatment group alleviated oxazolone-induced intestinal tissue damage and reduced the contents of malondialdehyde, reactive oxygen species, IL-1ß, and TNF-α and then increased the contents of superoxide dismutase, glutathione peroxidase, and IL-10. The 16s rDNA gene sequencing findings demonstrated that SBE could increase the relative abundance of Fusobacteriota, Actinobacteriota, and Firmicutes and decrease the relative abundance of Proteobacteria. Based on the correlation analysis between the oxidative stress biomarkers and intestinal flora, we found that the trends of oxidative stress biomarkers were significantly correlated with intestinal microorganisms, especially at the genus level. The correlations of MDA, IL-1ß, and TNF-α were significantly negative with Shewanella, while SOD, GSH-Px, and IL-10 were significantly positive with Cetobacterium, Gemmobacter, and Flavobacterium. Consequently, we concluded that the antioxidant effect of SBE was realized through the interaction between oxidative stress biomarkers and gut microbiota.

RESUMEN

An unprecedented strategy to access highly enantioenriched dihydropyrazoles is described. It involves formal [4+1] cycloadditions of in situ-derived azoalkenes and sulfur ylides catalyzed by a chiral copper/Tol-BINAP complex. A variety of synthetically and biologically important dihydropyrazoles have been obtained with high enantioselectivities (up to 97:3 er) in good yields (83-97%).

Transition metal-mediated templating and self-assembly have shown great potential to construct mechanically interlocked molecules. Herein, we describe the formation of the bimetallic [3]catenane and [4]catenane based on neutral organometallic scaffolds via the orthogonality of platinum-to-oxygen coordination-driven self-assembly and copper(I) template-directed strategy of a [2]pseudorotaxane. The structures of these bimetallic [3]catenane and [4]catenane were characterized by multinuclear NMR {1H and 31P} spectroscopy, electrospray ionization time-of-flight mass spectrometry (ESI-TOF-MS), and PM6 semiempirical molecular orbital theoretical calculations. In addition, single-crystal X-ray analyses of the [3]catenane revealed two asymmetric [2]pseudorotaxane units inside the metallacycle. It was discovered that tubular structures were formed through the stacking of individual [3]catenane molecules driven by the strong π-π interactions.

To investigate the physiological response of IAA (indoleacetic acid) to Trichosanthes kirilowii under aluminum stress and the mitigation of DNA damage, the effects of IAA (0, 10, 25, 50, 75 µmol·L-1 denoted by I0, I10, I25, I50 and I75, respectively) on antioxidant enzyme activity, malondialdehyde (MDA), photosynthetic characteristics, root activity, chlorophyll content and DNA damage of two varieties of T. kirilowii under 300 and 800 µmol·L-1 aluminum environment (denoted by Al300 and Al800, respectively) were examined in hydroponic culture experiments with Hebei Anguo (aluminium tolerant variety) and Zhejiang Puyang Trichosanthes kirilowii (aluminum sensitive variety). The results showed that the activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), photosynthesis ability, and root activity of both varieties were inhibited to different degrees by aluminum stress, MDA content was significantly increased, and DNA damage was aggravated. The maximum increase of SOD, CAT and POD activities in Anguo and Puyang T. kirilowii under aluminum stress by IAA application were 15.0%, 31.2%, 72.3% and 13.8%, 26.9%, 26.4%, respectively, chlorophyll content increased by 49.9% and 17.9%, MDA accumulation decreased by 39.2% and 22.4% and fluorescence parameters were significantly improved. The treatment of 25 µmol·L-1 IAA significantly increased root activity by 159.1% and 90.9%, while 50 µmol·L-1 IAA effectively slowed the DNA tailing phenomenon in roots, with the product (OTM) value of tail DNA percentage content and tail moment being decreased by 27.6%. 10-50 µmol·L-1 IAA could effectively improve the physiological activity of T. kirilowii under aluminum stress and slow the degree of DNA damage. The tolerance of Anguo variety to aluminum stress was stronger than that of Puyang variety.

In the title complex, {[HgI(2)(C(17)H(13)N(5)O(2))]·C(3)H(7)NO}(n), the Hg atom is coordinated by two I atoms and two N atoms from two different ligands in a distorted tetra-hedral environment. Hg atoms are bridged by N,N'-di-3-pyridylpyridine-2,6-dicarboxamide ligands, forming a helical chain running along the a axis.

Up till now, studies have not been conducted on how the combination of Quercetin (Q), Aconitine (A) and apoptosis induction affects human cervical carcinoma HeLa cells. The result of our findings shows that the combination of Q and A (QA) is capable of synergistically inhibiting the proliferation of HeLa cells in a number of concentrations. QA synergistically inhibits the proliferation of MDR1 gene in the HeLa cells. It is concluded based on our result that QA induces apoptosis and ER stress just as QA-induced ER stress pathway may mediate apoptosis by upregulating mRNA expression levels of eIF2α, ATF4, IRE1, XBP1, ATF6, PERK and CHOP in the HeLa cells. The up-regulating of mRNA expression level of GRP78 and activation of UPR are a molecular basis of QA-induced ER stress.

The research efforts on oligoacene systems are still relatively limited mainly due to the synthetic challenge and the extreme instability of longer acenes. Herein, these two issues have been overcome through elaborative modification and the stable pentacene species has been successfully synthesized. Additionally, a series of bis(diarylamino) compounds linked by variable-length oligoacene bridges ranging from one to five fused rings (benzene (1a), naphthalene (1b), anthracene (1c), tetracene (1d) and pentacene (1e)) have been prepared to probe the effect of the extent of π-conjugation on the electron transfer properties. Compound 1c exhibits a high planarity between the anthracyl bridge and the two nitrogen cores and the molecular packing shows a two-dimensional herringbone characteristic. Combined studies based on electrochemistry and spectroelectrochemistry demonstrate that (i) the electronic coupling across the oligoacene linkers between two diarylamine termini exponentially decrease with a moderate attenuation constant (ß) of 0.14 Å-1 in these length-modulated systems and (ii) the associated radical cations [1a]+-[1e]+ are classified as the class II Robin-Day mixed-valence systems. Furthermore, density functional theory (DFT) calculations have been conducted to gain insight into the nature of electron transfer processes in these oligoacene systems.

Anion receptors based on an 8-thiourea substituted quinoline with pentafluorinated (1a) or nonfluorinated (1b) biarylamide groups in the 2-position show similar binding of halide anions with somewhat higher association constants for the more acidic fluorinated derivative. Surprisingly, binding affinities for the halides in the case of the nonfluorinated 1b are similar in nonpolar chloroform or polar DMSO as solvent. Thorough thermodynamic investigations based on NMR van't Hoff analysis show that anion binding in chloroform is mainly enthalpically driven. In DMSO, entropy is the driving force for the binding of the ions with replacement of attached solvent.
