ntrodução: As enteroparasitoses são causadas por protozoários e helmintos e representam um importante proble-ma de saúde pública. As crianças em idade pré-escolar e escolar apresentam maior vulnerabilidade à contaminação por enteroparasitas pelo fato de estarem mais expostas a fatores de risco, tais como hábitos precários de higiene e aglomeração em locais fechados, e por não apresentarem o sistema imunológico totalmente desenvolvido. Os para-sitas mais comumente encontrados nessa faixa etária compreendem Giardia lamblia, Ascaris lumbricoides, Trichuris trichiura e ancilostomídeos. Objetivos: Avaliar a prevalência de enteroparasitos em crianças pré-escolares e escola-res e em caixas de areia de instituições públicas de ensino no município de Sinop-MT. Métodos: O estudo realizado foi transversal, quantitativo com delineamento experimental. Foi realizado no período de julho de 2015 a setembro de 2016. Foram avaliadas crianças pré-escolares e escolares de 3 a 12 anos escolhidas por amostragem de conveniência de quatro instituições da rede municipal de ensino localizadas na região central de Sinop ­ MT e sete afastadas do centro da cidade e que atendem novos bairros e a zona rural. As amostras de fezes foram coletadas pelos responsá-veis das crianças que encaminharam para as instituições de ensino e, posteriormente, foram transportadas ao labo-ratório para realização do exame parasitológico. As amostras de areia foram coletadas a partir de caixas das áreas de recreações de todas as instituições de ensino, sendo coletada 100 gramas de areia da raspagem superficial e pro-funda dos quatro quadrantes e do centro, totalizando 10 amostras/caixa. Os métodos parasitológicos utilizados para análises das amostras de fezes e areia foram Hoffmann, migração ativa, centrífugo-flutuação espontânea por Faust e flutuação espontânea por Willis. Resultados: Foram avaliadas 646 amostras de fezes de crianças pré-escolares e escolares de 3 a 12 anos, apresentando 21,05% de positividade para parasitos intestinais. Giardia lamblia apresentou maior prevalência, seguida de Entamoeba coli, Endolimax nana,Ascaris lumbricoides, Enterobius vermicularis, Hy-menolepis nana, ancilostomídeo e Entamoeba hartmanni. As 80 amostras de areia das caixas das áreas de recreação infantil avaliadas, apresentaram 100% de positividade para G. lamblia, seguida de 66,66% para Toxocara sp. nas areias das áreas centrais e 100% nas periféricas, 66,66% para Toxoplasma gondii nas centrais e 80% nas periféricas, 33,33% para A. lumbricoides nas centrais e 60% nas periféricas e 33,33% de Strongyloides stercoralis somente nas areias das áreas centrais. Conclusão: A faixa etária mais acometida foi verificada entre as crianças de 6 a 12 anos e pode estar relacionada aos precários hábitos de higiene e assim, maior risco de contaminação. Pode-se concluir que os resultados estão de acordo com a maioria dos estudos epidemiológicos realizados no Brasil, demonstrando elevada prevalência de enteroparasitoses em crianças em idade escolar (AU)


Introduction: Protozoa and helminths cause enteroparasitoses, which constitute an important public health issue. Preschoolers and school children, who do not have a fully developed immune system, are more vulnerable to con-tamination by intestinal parasites because they are more exposed to risk factors, such as poor hygiene habits and crowding in closed places. The most common parasites affecting this age group include Giardia lamblia, Ascaris lumbricoides, Trichuris trichiura, and hookworms. Objectives: To evaluate the prevalence of intestinal parasites in preschoolers, school children, and sandboxes of government schools in the city of Sinop ­ state of Mato Gros-so (MT). Methods: This cross-sectional, quantitative study with an experimental design was carried out from July 2015 to September 2016 and included preschoolers and school children aged 3 to 12 years, chosen by convenience sampling from government schools in the school district of the city of Sinop ­ MT; four schools were in the central region of Sinop, whereas seven schools, attended by children living in the suburban and rural areas, were far from the city center. Fecal samples were collected by the children's family and sent to the schools, from where they were transported to the laboratory for parasitological examination. Sand samples were collected from sandboxes in the play areas of the schools. More specifically,100 g of sand was collected by scraping one of the four quadrants or the center of the sandbox surface or deeper into the sandbox, totaling 10 samples/box. The parasitological methods spontaneous sedimentation described by Hoffmann, active migration described by Rugai, centrifugal-spontaneous flotation described by Faust, and spontaneous flotation described by Willis were used to analyze the fecal and sand samples. Results: A total of 646 fecal samples obtained from preschoolers and school children aged 3 to 12 years were evaluated; 21.05% of the samples tested positive for intestinal parasites. As for the sand samples, 100% tested positive for intestinal parasites. In all the fecal and sand samples that tested positive for intestinal parasites, G. lamblia predominated, followed by Toxacara sp, Toxoplasma gondii, A. lumbricoides, Enterobius vermicularis, Hymenolepis nana, Strongyloides stercoralis, hookworm, Entamoeba coli, Endolimax nana, and Entamoeba hart-manni, commensal amoebae of the large intestine, were also identified. Conclusion: Children aged 6 to 12 years were the most affected by enteroparasitoses. This may be related to poor hygiene habits, which puts these children at greater risk of contamination. The results agree with most epidemiological studies conducted in Brazil and show a high prevalence of enteroparasitoses in children (AU)

Studies demonstrate that obesity can increase tumor development. Botryosphaeran, a fungal (1→3)(1→6)-ß-D-glucan, presents antimutagenic, antiproliferative and pro-apoptotic activities. This study evaluated the effects of botryosphaeran on tumor development and metabolic and hematological parameters in tumor-bearing obese and non-obese rats. Obesity was induced by a high-fat and high-sugar diet, while control rats received standard diet and water without sugar for 10 weeks. On 8th-week, Walker-256 tumor cells were inoculated in the rats, and treatment with botryosphaeran (12 mg/Kg b.w.) started. Groups:control tumor-CT, control tumor botryosphaeran-CTB, obese tumor-OT and obese tumor botryosphaeran-OTB. On 10th-week, tumor development, cachexia, metabolic and hematological parameters were analyzed. Tumor development and cachexia were significantly higher in the OT group compared to the CT group, and botryosphaeran attenuated these parameters. OT rats presented accumulation of adipose tissue, reduced muscle mass, glucose intolerance, insulin resistance, hyperglycemia, anemia, leukocytosis, and thrombocytopenia. Botryosphaeran corrected insulin resistance and hyperglycemia, modulated cholesterol levels, and increased leukocyte and lymphocytes in obese rats, which can be attributable to an inflammatory response against the Walker-256 tumor, contributing to a lower tumor development. Our data demonstrated that botryosphaeran was effective in attenuating tumor growth and in improving the metabolic and hematological profiles of the tumor-bearing rats, demonstrating its potential role in the cancer's management.

Obesity is an important risk factor in tumor development. Botryosphaeran, a (1 â†’ 3)(1 â†’ 6)-ß-D-glucan, produced by the fungus Botryosphaeria rhodina (MAMB-05), is a high molecular mass, water-soluble exopolysaccharide. It consists of a main chain of (1 â†’ 3)-linked ß-d-glucose units, with a degree of branching of ~22% at carbon-6 with glucose and gentiobiose residues linked through ß-(1 â†’ 6)-bonds, and presents a triple helix conformation. Botryosphaeran presents anticlastogenic, antiproliferative, pro-apoptotic and anti-obesogenic activities. This study evaluated the effects of botryosphaeran on tumor development in obesity and analyzed its mechanism of action. Obesity was induced in male Wistar rats by a high-fat/high-sugar diet. After 9 weeks, rats were divided into two groups: Obese Tumor (OT) and Obese Tumor Botryosphaeran (OTB), and inoculated with 1 × 107 Walker-256 tumor cells, and treatment with botryosphaeran (30 mg/kg b.w./day via gavage for 15 days) commenced. On the 11th week, biological parameters, tumor development, metabolic profile, erythrogram and protein expression were evaluated. Botryosphaeran significantly reduced tumor growth, body-weight loss and cachexia. Furthermore, botryosphaeran decreased mesenteric fat and insulin resistance, corrected macrocytic anemia, and increased Forkhead transcription factor-3a (FOXO3a) activity. Our study demonstrated the potential role of botryosphaeran in the management of cancer in tumor-bearing obese rats by increasing insulin sensitivity and FOXO3a activity.

AIMS: Cancer is a multifactorial disease characterized by an uncontrolled growth of cells that can lead to cachexia-anorexia syndrome. Botryosphaeran, a fungal (1 â†’ 3)(1 â†’ 6)-ß-D-glucan produced by Botryosphaeria rhodina MAMB-05, has presented antimutagenic, antiproliferative, pro-apoptotic, hypoglycemic and hypocholesterolemic effects. This study evaluated the effects of botryosphaeran (30 mg/kg b.w./day) on tumor development and cachexia syndrome in Walker-256 tumor-bearing rats, and also the metabolic and hematological profiles of these animals. MATERIALS AND METHODS: Male Wistar rats were divided into 3 groups: control (C), control tumor (CT) and control tumor botryosphaeran (CTB). On the first day, 1 × 107 Walker-256 tumor cells were inoculated subcutaneously into the right flank of the CT and CTB rats, and concomitantly treatment with botryosphaeran (30 mg/kg b.w./day) started. After the 15th day of treatment, biological parameters, tumor development, cachexia, glucose and lipid profiles, hemogram and protein expression were analyzed. KEY FINDINGS: Botryosphaeran significantly reduced tumor development (p = 0.0024) and cancer cachexia, modulated the levels of glucose, triglycerides and HDL-cholesterol, and corrected macrocytic anemia. Botryosphaeran also increased significantly the bax expression in the tumor tissue (p = 0.038) demonstrating that this (1 â†’ 3)(1 â†’ 6)-ß-D-glucan is increasing the apoptosis of tumor cells. p53, p27, bcl-2, caspase-3 and Forkhead transcription factor 3a (FOXO3a) protein expression were similar among the groups. SIGNIFICANCE: This study demonstrated that botryosphaeran was effective in decreasing tumor development and cachexia by direct and indirect mechanisms increasing apoptosis and improving the metabolic and hematological profiles.

Sporotrichosis is a chronic infection caused by the dimorphic fungus Sporothrix schenckii, involving all layers of skin and the subcutaneous tissue. The role of innate immune toll-like receptors 2 and 4 in the defense against this fungus has been reported, but so far, there were no studies on the effect of cell wall major components over the cytosolic oligo-merization domain (NOD)-like receptors, important regulators of inflammation and responsible for the maturation of IL-1ß and IL-18, whose functions are dependents of the caspase-1 activation, that can participate of inflammasome. It was evaluated the percentage of activation of caspase-1, the production of IL-1ß, IL-18, IL-17, IFN-γ and nitric oxide in a Balb/c model of S. schenckii infection. It was observed a decreased activity of caspase-1 during the fourth and sixth weeks of infection accompanied by reduced secretion of the cytokines IL-1ß, IL-18 and IL-17 and high production of nitric oxide. IFN-γ levels were elevated during the entire time course of infection. This temporal reduction in caspase-1 activity coincides exactly with the reported period of fungal burden associated with a transitory immunosuppression induced by this fungus and detected in similar infection models. These results indicate the importance of interaction between caspase-1, cytokines IL-1ß and IL-18 in the host defense against S. schenckii infection, suggesting a participation the inflammasome in this response.

Toll-like receptors (TLRs) play an important role in immunity, since they bind to pathogen surface antigens and initiate the immune response. However, little is known about the role of TLR-2 in the recognition of S. schenckii and in the subsequent immune response. Therefore, the aim of this study was to evaluate the involvement of TLR-2 in the immune response induced by S. schenckii. C57BL/6 mice (WT) and C57BL/6 TLR-2 knockout (TLR-2-/-) were used to evaluate, over a period of 10 weeks of sporotrichotic infection, the influence of TLR-2 over macrophages production of IL-1ß, IL-12 and TNF-α, their stimulation level by NO release and the production of IFN -γ, IL-6, IL-17 and TGF-ß by spleen cells. The results showed that the production of pro-inflammatory mediators and NO, TLR-2 interference is striking, since its absence completely inhibited it. IL-17 production was independent of TLR-2. The absence of Th1 response in TLR2-/- animals was concomitant with IL-17 production. Therefore, it can be suggested that TLR-2 absence interferes with the course of the infection induced by the fungus S. schenckii.

Sporotrichosis is a subcutaneous mycosis that is caused by the dimorphic fungus Sporothrix schenckii. This disease generally occurs within the skin and subcutaneous tissues, causing lesions that can spread through adjacent lymphatic vessels and sometimes leading to systemic diseases in immunocompromised patients. Macrophages are crucial for proper immune responses against a variety of pathogens. Furthermore, macrophages can play different roles in response to different microorganisms and forms of activation, and they can be divided into "classic" or "alternatively" activated populations, as also known as M1 and M2 macrophages. M1 cells can lead to tissue injury and contribute to pathogenesis, whereas M2 cells promote angiogenesis, tissue remodeling, and repair. The aim of this study was to investigate the roles of M1 and M2 macrophages in a sporotrichosis model. Toward this end, we performed phenotyping of peritoneal exudate cells and evaluated the concomitant production of several immunomediators, including IL-12, IL-10, TGF-ß, nitric oxide, and arginase-I activity, which were stimulated ex vivo with cell wall peptide-polysaccharide. Our results showed the predominance of the M2 macrophage population, indicated by peaks of arginase-I activity as well as IL-10 and TGF-ß production during the 6th and 8th weeks after infection. These results were consistent with cellular phenotyping that revealed increases in CD206-positive cells over this period. This is the first report of the participation of M2 macrophages in sporotrichosis infections.

Sporotrichosis is an infection caused by the dimorphic fungus Sporothrix schenckii. Toll-like receptors (TLRs) play an important role in immunity, since they bind to pathogen surface antigens and initiate the immune response. However, little is known about the role of TLR-2 and fungal surface antigens in the recognition of S. schenckii and in the subsequent immune response. This study aimed to evaluate the involvement of TLR-2 and fungal surface soluble (SolAg) and lipidic (LipAg) antigens in phagocytosis of S. schenckii and production of immune mediators by macrophages obtained from WT and TLR-2(-/-) animals. The results showed that TLR-2(-/-) animals had had statistical lower percentage of macrophages with internalized yeasts compared to WT. SolAg and LipAg impaired phagocytosis and immunological mediator production for both WT and TLR-2(-/-). The absence of TLR-2 led to lower production of the cytokines TNF-α, IL-1ß, IL-12 and IL-10 compared to WT animals. These results suggest a new insight in relation to how the immune system, through TLR-2, recognizes and induces the production of mediators in response to the fungus S. schenckii.
