RESUMO

The subfamily Phlebotominae comprises important insects for public health. The use of complementary tools such as molecular taxonomy is necessary for interspecific delimitation and/or discovery of cryptic species. Here, we evaluated the DNA barcoding tool to identify different species in the southwestern Brazilian Amazon. For this, we collected sand flies in forest fragments along the highway BR-317, in the municipality of Brasiléia, state of Acre, Brazil. The specimens were DNA-barcoded using a fragment of the cytochrome c oxidase subunit I (COI) gene. The sequences were analyzed to generate K2P pairwise genetic distances and a Neighbour-joining tree. The sand fly barcodes were also clustered into Molecular Operation Taxonomic Units (MOTU) using Automatic Barcode Gap Discovery (ABGD) approach. A total of 59 COI sequences comprising 22 nominal species and ten genera were generated. Of these, 11 species had not been sequenced before, thus being new COI sequences to science. Intraspecific genetic distances ranged between 0 and 4.9%, with Pintomyia serrana presenting the highest values of genetic distance, in addition to having been partitioned into three MOTUs. Regarding the distances to the nearest neighbour, all species present higher values in relation to the maximum intraspecific distance, in addition to forming well supported clusters in the neighbour-joining analysis. The DNA barcoding approach is useful for the molecular identification of sand flies from Brasiléia, state of Acre, and was efficient in detecting cryptic diversity of five species which can be confirmed in future studies using an integrative approach. We also generated new COI barcodes for Trichophoromyia auraensis, Nyssomyia shawi, and Psychodopygus paraensis, which may play a role in the transmission of Leishmania spp. in the Brazilian Amazon.

Evidence of sylvatic yellow fever was first reported in Atlantic Forest areas in Espírito Santo, Brazil, during a yellow fever virus (YFV) outbreak in 1931. An entomological survey was conducted in six forest sites during and after an outbreak reported ~80 years after the last case in the area. Among 10,658 mosquitoes of 78 species, Haemagogus leucocelaenus, and Hg. janthinomys/capricornii were considered the main vectors as they had a relatively high abundance, co-occurred in essentially all areas, and showed high YFV infection rates. Sabethes chloropterus, Sa. soperi, Sa. identicus, Aedes aureolineatus, and Shannoniana fluviatilis may have a secondary role in transmission. This is the first report of Sa. identicus, Ae. aureolineatus, and Sh. fluviatilis infected with YFV. Our study emphasizes the importance of entomological monitoring and maintenance of high vaccination coverage in receptive areas to YFV transmission.

OBJECTIVES: Here, data concerning the sandfly fauna of Brasiléia municipality, Leishmania DNA-detection rates and the identification of blood meal sources of insects captured in 2013-2015 are presented. METHODS: Parasite detection in female sandflies was performed individually by multiplex polymerase chain reaction (PCR) (Leishmania kDNA/sandfly cacophony-gene), with the identification of Leishmania spp. by hsp70-PCR and sequencing. The identification of blood gut-content from fed females was performed by cyt b-PCR and sequencing. FINDINGS: A total of 4,473 sandflies were captured. A subgroup of 864 non-blood-fed females evaluated for the presence of Leishmania DNA showed 2.9% positivity for Leishmania (Viannia) braziliensis and L. (V.) guyanensis. The identification of blood meal sources was performed in 96 blood-fed females, allowing the identification of 13 vertebrate species. In nine/96 fed females, DNA from L. (V.) shawi, L. (V.) guyanensis, L. (V.) braziliensis and Endotrypanum sp. was detected. MAIN CONCLUSIONS: In Brumptomyia sp. and Evandromyia termitophila, the first report of Leishmania DNA-detection is provided in Acre; Nyssomyia shawi is implicated as potential vector of L. (V.) braziliensis and L. (V.) guyanensis for the first time in Brazil.

Sand fly (Diptera: Psychodidae) taxonomy is complex and time-consuming, which hampers epidemiological efforts directed toward controlling leishmaniasis in endemic regions such as northeastern Brazil. Here, we used a fragment of the mitochondrial cytochrome c oxidase I (COI) gene to identify sand fly species in Maranhão State (northeastern Brazil) and to assess cryptic diversity occurring at different spatial scales. For this, we obtained 148 COI sequences of 15 sand fly species (10 genera) from Maranhão (fine spatial scale), and joined them to COI sequences from other Brazilian localities (distant about 2,000 km from Maranhão, broad spatial scale) available in GenBank. We revealed cases of cryptic diversity in sand flies both at fine (Lutzomyia longipalpis (Lutz and Neiva) and Evandromyia termitophila (Martins, Falcão and Silva)) and broad spatial scales (Migonemyia migonei (França), Pressatia choti (Floch and Abonnenc), Psychodopygus davisi (Root), Sciopemyia sordellii (Shannon and Del Ponte), and Bichromomyia flaviscutellata (Mangabeira)). We argue that in the case of Bi. flaviscutellata, the cryptic diversity is associated with a putative new species. Cases in which DNA taxonomy was not as effective as morphological identification possibly involved recent speciation and/or introgressive hybridization, highlighting the need for integrative approaches to identify some sand fly species. Finally, we provide the first barcode sequences for four species (Brumptomyia avellari (Costa Lima), Evandromyia infraspinosa (Mangabeira), Evandromyia evandroi (Costa Lima and Antunes), and Psychodopygus complexus (Mangabeira)), which will be useful for further molecular identification of neotropical species.

BACKGROUND: Because domestic dogs are reservoir hosts for visceral leishmaniasis (VL) in Brazil, one of the approaches used to reduce human disease incidence is to cull infected dogs. However, the results of controlled intervention trials based on serological screening of dogs and killing of seropositive animals are equivocal. A prophylactic vaccine to protect dogs from being infectious to the sand fly vector could be an effective strategy to provide sustained control. Here, we investigated whether a currently licensed commercial subunit rA2 protein-saponin vaccine (Leish-tec®) had an additional effect to dog culling on reducing the canine infectious populations. METHODOLOGY/PRINCIPAL FINDINGS: This prospective study was conducted in an L. infantum highly endemic area of southeast Brazil. At the onset of the intervention, all of the eligible dogs received through subcutaneous route a three-dose vaccine course at 21-day intervals and a booster on month 12. For the purpose of comparison, newly recruited healthy dogs were included as the exposed control group. To ascertain vaccine-induced protection, dogs were screened on clinical and serological criteria every 6 months for a 2-year follow-up period. Antibody-based tests and histopathological examination of post-mortem tissue specimens from euthanized animals were used as a marker of infection. The standardized vaccine regime, apart from being safe, was immunogenic as immunized animals responded with a pronounced production of anti-A2-specific IgG antibodies. It should be noted the mean seroconversion time for infection obtained among immunized exposed dogs (~ 18 months), which was twice as high as that for unvaccinated ones (~ 9 months). After two transmission cycles completed, the cumulative incidence of infection did differ significantly (P = 0.016) between the vaccinated (27%) and unvaccinated (42%) dogs. However, the expected efficacy for the vaccine in inducing clinical protection was not evident since 43% of vaccine recipients developed disease over time. Our estimates also indicated that immunoprophylaxis by Leish-tec® vaccine in addition to dog culling might not have an impact on bringing down the incidence of canine infection with L. infantum in areas of high transmission rates. CONCLUSIONS/SIGNIFICANCE: Leish-tec® as a prophylactic vaccine showed promise but needs to be further optimized to be effective in dogs under field conditions, and thereby positively impacts human incidence.

DNA barcoding has been an effective tool for species identification in several animal groups. Here, we used DNA barcoding to discriminate between 47 morphologically distinct species of Brazilian sand flies. DNA barcodes correctly identified approximately 90% of the sampled taxa (42 morphologically distinct species) using clustering based on neighbor-joining distance, of which four species showed comparatively higher maximum values of divergence (range 4.23-19.04%), indicating cryptic diversity. The DNA barcodes also corroborated the resurrection of two species within the shannoni complex and provided an efficient tool to differentiate between morphologically indistinguishable females of closely related species. Taken together, our results validate the effectiveness of DNA barcoding for species identification and the discovery of cryptic diversity in sand flies from Brazil.

The aim of this study was to evaluate the sand fly fauna of American visceral leishmaniasis (AVL) endemic areas within the Central Atlantic Forest Biodiversity Corridor, State of Espírito Santo, southeastern Brazil. The sand fly captures were performed between January, 1989 and December, 2003 in localities where autochthonous cases of AVL were recorded, as well as in their boundary areas. Sand flies were collected from surrounding houses and domestic animal shelters using two to five CDC automatic light traps, and manual captures were also performed using mouth aspirators in one illuminated Shannon trap during the first four hours of the night. We used cladistic analysis to determine the geographic relationships among the collected sand fly species as well as the index species for the occurrence of other sand flies. A total of 62,469 sand flies belonging to 17 species and eight genera was collected in 164 localities from nine municipalities with AVL records. The richness (S=17) and diversity (H=0.971) of sand flies were lower than in conservation areas and similar to modified environments in the Atlantic Forest of Espírito Santo. Lutzomyia longipalpis was identified in 79 localities. The cladistic analysis identified Evandromyia lenti as the index species for Lutzomyia longipalpis. The latter seems to be the main vector of AVL in the Central Atlantic Forest Biodiversity Corridor due to its high abundance and distribution matching the disease occurrence. Therefore, Evandromyia lenti may be used as an index species for the occurrence of Lutzomyia longipalpis.

Canine visceral leishmaniasis (CVL) is the major source of human visceral leishmaniasis (VL) and is transmitted from dogs to sand flies to humans. To control the spread of this disease, early and accurate detection of infected dogs is critical but challenging. Here we demonstrate the potential of the Dual-Path Platform (DPP(®)) CVL rapid test for detecting K26/K39-reactive antibodies in sera from clinically symptomatic (n=60) and asymptomatic (n=60) Leishmania infantum-infected dogs. For the specificity evaluation, assays were performed using known negative diagnostic serum samples (n=59) and cross-reaction control sera (n=11) from animals born in a VL-free area of Brazil. The diagnostic kit displayed high specificity (96%) but low sensitivity (47%) in identifying parasite-positive dogs without signs of CVL. However, the test sensitivity was significantly higher (98%) in diseased cases, indicating that this convenient test may be useful to identify the most infectious dogs. Efforts should be pursued to obtain a more sensitive DPP-multiplexed test parameter (i.e. based on simultaneous yet separate antibody detection of carefully selected multiple antigens of diagnostic utility) for effective serodiagnosis of early-infected dogs, as this will likely allow more efficient canine removal regimens than those used in practice by public health services.

Entre os anos de 2004 e 2008 foram realizadas coletas de Simuliidae em várias localidades no Estado do Espírito Santo, Brasil. No total, 66 córregos e rios das doze principais bacias hidrográficas do estado foram amostrados. Dezessete espécies foram coletadas, sendo que nove delas representam ocorrências novas para o estado (Simulium pertinax, S. jujuyense, S. rubrithorax, S. subnigrum, S. travassosi, S. spinibranchium, S. hirtipupa, S. lutzianum e S. anamariae), duplicando o número de espécies conhecidas no estado.


We conducted a freshwater survey from 2004 to 2008 in several localities of Espírito Santo, Brazil. In total, 66 streams and rivers were sampled in the 12 hydrographic basins of the state. We collected and identified 17 species of which nine represent new records (Simulium pertinax, S. jujuyense, S. rubrithorax, S. subnigrum, S. travassosi, S. spinibranchium, S. hirtipupa, S. lutzianum e S. anamariae). These findings doubled the number of records of black fly species to the State of Espírito Santo, Brazil.

Uma busca por angiostrongilídeos foi realizada em cortes histológicos de vísceras provenientes de 129 pequenos mamíferos não voadores capturados na Reserva Biológica de Duas Bocas, município de Cariacica, Espírito Santo, Brasil. Não foram encontrados vermes ntra-arteriais. Dentro da árvore brônquica de dois indivíduos, identificados como Metachirus nudicaudatus e Didelphis aurita foram encontrados vermes cilíndricos, provavelmente filarídeos. Infecção humana, tanto por Angiostrongylus costaricensis quanto por Angiostrongylus cantonensis, já foi registrada no Espírito Santo inclusive nesta região de Cariacica, porém não há dados sobre a infecção natural em outros mamíferos. Este relato trata de uma tentativa pioneira de identificar angiostrongilídeos em animais coletados em uma Reserva Biológica no Estado do Espírito Santo.

A ocorrência de leishmaniose visceral americana no Estado do Espírito Santo, Brasil, sempre foi associada à presença do vetor Lutzomyia longipalpis. A distribuição geográfica desse vetor no estado está relacionada à ocorrência de características geoclimáticas específicas, tais como clima seco, baixas altitudes, relevo acidentado e afloramentos rochosos A ocorrência de casos humanos autóctones de leishmaniose visceral americana em municípios sem essas características geoclimáticas justificou o presente estudo, com objetivo de testar a associação entre a ocorrência de leishmaniose visceral americana e a presença de Lu. longipalpis. Flebotomíneos foram capturados mensalmente de julho de 2006 a agosto de 2007 com uso de armadilhas de Shannon e CDC em dois municípios com registros de leishmaniose visceral americana autóctone e um sem registros. Foram capturados 13.112 flebotomíneos, mas nenhum Lu. longipalpis foi encontrado. A ausência de Lu. longipalpis e a possível participação de outro vetor de leishmaniose visceral americana nessas localidades foram discutidas.

In Brazil, introduced malaria occurs from the flat to the sloping hot areas, predominantly outside the Amazon Region, where endemic malaria has occurred in the past. This is a consequence of human migrations to other Brazilian states, including the state of Espírito Santo (ES). The objective of this study was to use geoprocessing to define the areas at risk of introduced malaria transmission and evaluate the vectorial importance of species of anophelines in ES. Anophelines were sampled from 1997-2005 in 297 rural localities identified or not identified as foci of malaria during the last 20 years. The geoclimatic variables temperature, relief and marine influence were obtained from a database of the ES Natural Units. The 14,663 anophelines captured belonged to 22 species. A significant association was found between the occurrence of malaria foci and the presence of hot, low-lying areas or gently undulating to undulating relief. The occurrence of the disease was associated with the presence of Anopheles darlingi and Anopheles aquasalis. Geoprocessing was determined to be a useful tool for defining areas at risk for malaria and vectors in ES.

Phlebotomine sand flies present great species diversity, especially in Brazil, where there are about 300 known species. This work describes a new species of sand fly found in the Brazilian state of Espírito Santo, in the Reserva Biológica de Duas Bocas, municipality of Cariacica. Spermathecae with superficial striations and the common duct longer than the genital fork permit the inclusion of the new species in the subgenus Evandromyia s. str., series rupicola. The new species resemble Ev. rupicola from which it may be distinguished by the aspects of the spermatheca and the length of the genital filament of the male, longer in the new species.

RESUMO

Nós inventariamos os pequenos mamíferos não-voadores em Viana, Espírito Santo, sudeste do Brasil, em 1981-1982 e 2006-2007. Foram capturados 439 pequenos mamíferos não-voadores pertencentes a três ordens (Didelphimorphia, Rodentia e Lagomorpha) e seis famílias (Didelphidae, Sciuridade, Cricetidae, Muridae, Echimyidae e Leporidae). As espécies mais abundantes foram os roedores Akodon cursor e Nectomys squamipes e o marsupial Metachirus nudicaudatus. Registramos pela primeira vez a ocorrência do roedor equimídeo Euryzygomatomys spinosus no estado do Espírito Santo. A riqueza específica (S = 21) e o índice de diversidade de Shannon (H = 2,23) estão entre os maiores registrados para pequenos mamíferos da Mata Atlântica no estado, mesmo quando comparados aos valores obtidos para unidades de conservação. Esses maiores valores de riqueza e diversidade encontrados estão provavelmente associados à heterogeneidade de hábitat e evidenciam a necessidade de conservação dos fragmentos florestais de Viana, os quais são importantes depositários da biodiversidade da Mata Atlântica.


We surveyed non-flying small mammals at Viana, Espírito Santo, southeastern Brazil, in 1981-1982 and 2006-2007. We trapped 439 non-flying small mammals belonging to three orders (Didelphimorphia, Rodentia, and Lagomorpha) and six families (Didelphidae, Sciuridade, Cricetidae, Muridae, Echimyidae, and Leporidae). The most abundant species were the rodents Akodon cursor and Nectomys squamipes and the marsupial Metachirus nudicaudatus. We recorded the echimyid rodent Euryzygomatomys spinosus for the first time in the state of Espírito Santo. The species richness (S = 21) and Shannon diversity index (H = 2.23) are among the highest recorded for Atlantic Forest small mammals in the state, even when compared to values from protected areas. These higher richness and diversity values are probably related to habitat heterogeneity and highlight the need for conservation of Viana's forest fragments, which are an important source of Atlantic Forest biodiversity.

In Brazil, introduced malaria occurs from the flat to the sloping hot areas, predominantly outside the Amazon Region, where endemic malaria has occurred in the past. This is a consequence of human migrations to other Brazilian states, including the state of Espírito Santo (ES). The objective of this study was to use geoprocessing to define the areas at risk of introduced malaria transmission and evaluate the vectorial importance of species of anophelines in ES. Anophelines were sampled from 1997-2005 in 297 rural localities identified or not identified as foci of malaria during the last 20 years. The geoclimatic variables temperature, relief and marine influence were obtained from a database of the ES Natural Units. The 14,663 anophelines captured belonged to 22 species. A significant association was found between the occurrence of malaria foci and the presence of hot, low-lying areas or gently undulating to undulating relief. The occurrence of the disease was associated with the presence of Anopheles darlingi and Anopheles aquasalis. Geoprocessing was determined to be a useful tool for defining areas at risk for malaria and vectors in ES.

RESUMO

Phlebotomine sand flies present great species diversity, especially in Brazil, where there are about 300 known species. This work describes a new species of sand fly found in the Brazilian state of Espírito Santo, in the Reserva Biológica de Duas Bocas, municipality of Cariacica. Spermathecae with superficial striations and the common duct longer than the genital fork permit the inclusion of the new species in the subgenus Evandromyia s. str., series rupicola. The new species resemble Ev. rupicola from which it may be distinguished by the aspects of the spermatheca and the length of the genital filament of the male, longer in the new species.

Esse é o primeiro registro de Evandromyia (Aldamyia) sericea para o Sudeste do Brasil e, até então, o registro mais ao sul da espécie no mundo. Os espécimes foram coletados na Reserva Biológica de Duas Bocas, município de Cariacica, estado do Espírito Santo. As capturas de flebotomíneos foram feitas mensalmente de abril a junho de 2008 durante quatro noites consecutivas usando duas armadilhas automáticas do tipo CDC e uma armadilha de Shannon modificada para cada noite. O encontro dessa espécie é consistente com a hipótese de proximidade filogeográfica entre a Floresta Amazônica e a Mata Atlântica.


This is the first record of Evandromyia (Aldamyia) sericea in southeastern Brazil and report the southernmost record ever of this species in the world. Specimens were collected in the Reserva Biológica de Duas Bocas, municipality of Cariacica, state of the Espírito Santo. Captures of the sand flies were carried out monthly from April to June 2008 during four consecutives nights using two automatic light traps (CDC model) and a modified Shannon trap for each night. The finding of this species corroborates the hypothesis of the phylogeographical proximity of the Amazon Rainforest and the Atlantic Forest.
