RESUMEN

The liver is an essential multifunctional organ, which constantly communicates with nearly all tissues. It has raised the concern that microgravity exposure can lead to liver dysfunction and metabolic syndromes. However, molecular mechanisms and intervention measures of the adverse effects of microgravity on hepatocytes are limited. In this study, we utilized the random positioning machine culture system to investigate the adverse effects on hepatocytes under simulated microgravity (SMG). Our results showed that SMG impaired hepatocyte viability, causing cell cycle arrest and apoptosis. Compared to normal gravity, it also triggered lipid accumulation, elevated triglyceride (TG) and ROS levels, and impaired mitochondria function in hepatocytes. Furthermore, RNA sequencing results showed that SMG upregulated genes implicated in lipid metabolisms, including PPARγ, PLIN2, CD36, FABPs, etc. Importantly, all these defects can be suppressed by melatonin, a potent antioxidant secreted by the pineal gland, suggesting its potential use of therapeutic intervention.

RESUMEN

Utilizing the periodicity of the rotating machinery, dynamic clearance measurement can be achieved with a single swept light source without any additional auxiliary devices, which has the advantages of simplicity and reliability. However, there is a shortcoming that previous algorithm is not fast enough to achieve real-time measurement when the machinery rotates at high speed. Aiming at this shortcoming, utilizing the correlation between mimic signal and measurement signal, combined with information for multiple periods, the fast algorithms and dynamic clearance corrected model were proposed. And the relationship between demodulation speed and cycle numbers was also discussed. Simulation was carried out to discuss the influence of different algorithm on the demodulation speed and accuracy. A test system was set up in the simulated environment for clearance measurement, and the results show that, the demodulation time of the proposed algorithm costs decreased dramatically, the speed has increased by about ten times, and the dynamic clearance measurement error is less than 2 µm.

The wavelength of microwave is longer than that of lightwave, causing the interferometric optical path difference induced by the measurand changes to be relatively smaller than that of microwave, which results in the lower sensitivity of sapphire fiber Fabry-Perot interferometer (FPI) sensor in microwave band. To improve sensitivity, a parallel FPI sensing system is constructed, in which a section of sapphire fiber connected to a single-mode transmission fiber is used as a sensing FPI, and a single-mode fiber (SMF) with a slightly different optical path from a sensing FPI is utilized as a reference FPI. By connecting two FPIs in parallel, Vernier effect is formed to improve sensitivity. The influence of relationship between the optical path difference of the reference FPI and the sensing FPI on the sensitivity amplification factor is analyzed based on the microwave interference spectrum of the parallel FPI. A section of sapphire fiber with the length of 8 cm is used as temperature sensor to construct high-temperature sensing system. The results demonstrate that the temperature sensitivity reaches about 2338.68kHz/°C, which is approximately 130 times higher than that of the sensing FPI alone. Furthermore, when the difference of optical path between the sensing FPI and the reference FPI is kept constant while the sensing FPI is unchanged, the amplification factor of the temperature sensitivity is approximately 2.64 times higher with longer length of the reference FPI compared to the situation with shorter length of the reference FPI.

BACKGROUND: Acute pancreatitis in pregnancy (APIP) with a high risk of death is extremely harmful to mother and fetus. There are few models specifically designed to assess the severity of APIP. Our study aimed to establish a clinical model for early prediction of severity of APIP. METHODS: A retrospective study in a total of 188 patients with APIP was enrolled. The hematological indicators, IAP (intra-abdominal pressure) and clinical data were obtained for statistical analysis and prediction model construction. RESULTS: According to univariate and multivariate logistic regression analysis, we found that red cell distribution width (RDW), neutrophil-lymphocyte ratio (NLR) and Intra-abdominal pressure (IAP) are prediction indexes of the severity in APIP (p-value < 0.05). Our novel clinical prediction model was created by based on the above three risk factors and showed superior predictive power in primary cohort (AUC = 0.895) and validation cohort (AUC = 0.863). A nomogram for severe acute pancreatitis in pregnancy (SAPIP) was created based on the three indicators. The nomogram was well-calibrated. CONCLUSION: RDW, NLR and IAP were the independent risk factors of APIP. Our clinical prediction model of severity in APIP based on RDW, NLR and IAP with predictive evaluation is accurate and effective.

BACKGROUND: This study aims to identify the relationship between iris -ciliary angle (ICA) and the vault. Additionally, we also seek to investigate the chain mediating effects of the ICL haptic related factors on this relationship. METHODS: The participants were categorized into three groups according to the ICA value as follows: low ICA group (< 35°); moderate ICA group (35°-70°); high ICA group (> 70°). We compared the preoperative ocular characteristics and postoperative examinations among the three groups. Multiple variable stepwise regression was performed to establish the vault prediction formula. The Process V4.0 in SPSS and Hayes's PROCESS model 6 was conducted to further elucidate the mediating effects of the final tip point of ICL haptic and the ICL arc-lens arc on the relationship between the ICA and vault. RESULTS: There was a significant difference in the positions of the ICL haptic among three ICA groups. The regression vault equation was Vault = 679.42-7.26*TCA + 192.30*ACD-196.37*CLR + 73.21* STS(horizontal).A significant negative correlation was found between the ICA and vault (P < 0.01).The chain mediation model revealed that the final tip point of ICL haptic and the ICL arc-Lens arc were sequential mediators between ICA and vault (effect = -1.63, 95% CI = -2.72--0.73). CONCLUSION: The ICA was associated with vault via the mediation effect of the final tip point of the ICL haptic and the ICL arc -lens arc. Assessment of ICL haptic related parameters adds significant information to interpret the vault after surgery.

Characterized by compensatory hyperplasia dependent on hepatocyte proliferation, the liver will initiate regeneration after partial hepatectomy (PH) and acute or chronic injuries. A variety of genes and noncoding RNAs play pivotal roles in these cell proliferation and growth processes. However, it is still unclear how competition endogenous RNAs (ceRNAs) modulate cellular activities during each phase of liver regeneration, and the specific mechanisms of posttranscriptional gene expression regulation in hepatocyte proliferation remain to be elucidated. To investigate the mechanism of liver regeneration through RNA-seq profiling and to determine the role of miR-34b-5p/PDK1 on hepatocyte proliferation, we established a 2/3 PH mouse model for whole transcriptome profiling based on high-throughput sequencing techniques. We subsequently constructed a lncRNA-miRNA-mRNA ceRNA regulatory network through integrative analyses of RNA interactions. Finally, plasmid transfection in NCTC 1469 cells, dual luciferase reporter gene assay, quantitative real-time PCR, western blotting, Cell Counting Kit-8, and EdU-DNA synthesis cell proliferation assay were used to demonstrate the role of the miR-34b-5p/PDK1 axis in hepatocyte proliferation in vitro. A total of 1443 mRNAs (962 up, 481 down), 48 miRNAs (35 up, 13 down), and 1955 lncRNAs (986 up, 969 down) were identified as significantly differentially expressed. We then successfully constructed a ceRNA regulatory network consisting of 7 lncRNAs, 15 miRNAs, and 347 mRNAs based on the predicted inverse interactions among ceRNAs. Additionally, miR-34b-5p/PDK1 was predicted to be closely related to hepatocyte proliferation. We further demonstrated that miR-34b-5p could bind specifically to the 3'-untranslated region (3'-UTR) of PDK1 using the dual luciferase reporter assay. Ectopic overexpression of miR-34b-5p significantly reduced the mRNA and protein expression of PDK1, while it markedly inhibited the proliferation of mouse NCTC 1469 cells in vitro. In contrast, knocking down miR-34b-5p exhibited the inverse effects on PDK1 expression and hepatocyte proliferation. Through analyzing the ceRNA network during mouse liver regeneration, this study reveals that miR-34b-5p can inhibit hepatocyte proliferation through negatively regulating PDK1 and may be a potential pharmacological intervention target.

BACKGROUND: Hepatocellular carcinoma (HCC) is the third most common cause of cancer death worldwide, with an overall 5-year survival rate of less than 18%, which may be related to tumor microvascular invasion (MVI). This study aimed to compare the clinical prognosis of HCC patients with or without MVI after radical surgical treatment, and further analyze the preoperative risk factors related to MVI to promote the development of a new treatment strategy for HCC. METHODS: According to the postoperative pathological diagnosis of MVI, 160 study patients undergoing radical hepatectomy were divided into an MVI-negative group (n = 68) and an MVI-positive group (n = 92). The clinical outcomes and prognosis were compared between the two groups, and then the parameters were analyzed by multivariate logistic regression to construct an MVI prediction model. Then, the practicability and validity of the model were evaluated, and the clinical prognosis of different MVI risk groups was subsequently compared. RESULT: There were no significant differences between the MVI-negative and MVI-positive groups in clinical baseline, hematological, or imaging data. Additionally, the clinical outcome comparison between the two groups presented no significant differences except for the pathological grading (P = 0.002) and survival and recurrence rates after surgery (P < 0.001). The MVI prediction model, based on preoperative AFP, tumor diameter, and TNM stage, presented superior predictive efficacy (AUC = 0.7997) and good practicability (high H-L goodness of fit, P = 0.231). Compared with the MVI high-risk group, the patients in the MVI low-risk group had a higher survival rate (P = 0.002) and a lower recurrence rate (P = 0.004). CONCLUSION: MVI is an independent risk factor for a poor prognosis after radical resection of HCC. The MVI prediction model, consisting of AFP, tumor diameter, and TNM stage, exhibits superior predictive efficacy and strong clinical practicability for MVI prediction and prognostication, which provides a new therapeutic strategy for the standardized treatment of HCC patients.

To improve the fringe contrast and the sensitivity of Fabry-Perot (FP) pressure sensors, a silicone rubber FP pressure sensor based on a spherical optical fiber end face is proposed. The ratio of silicone rubber ingredients and the diameter and thickness of silicone rubber diaphragm were optimized by a simulation based on experimental tests that analyzed elastic parameters, and the influence of the radius of a spherical optical fiber and the initial cavity length of the sensor on the fringe contrast was investigated and optimized. Pressure sensor samples were fabricated for pressure test and temperature cross-influence test. Gas pressure experimental results within a pressure range of 0~40 kPa show the average sensitivity of the sensor is -154.56 nm/kPa and repeatability error is less than 0.71%. Long-term pressure experimental results show it has good repeatability and stability. Temperature experimental results show its temperature cross-sensitivity is 0.143 kPa/°C. The good performance of the proposed FP pressure sensor will expand its applications in biochemical applications, especially in human body pressure monitoring.

To improve the precision of dynamic distance measurement based on the frequency-swept interferometry (FSI) system, a Doppler-induced error compensation model based on a scheme increasing the frequency sweeping rate is proposed. A distance demodulation method based on a Fourier transformation is investigated when the defined quasi-stationary coefficient approaches a constant. Simulations and experiments based on dynamic distance with a sinusoidal change demonstrate that the proposed method has a standard deviation of 0.09 µm within a distance range of 4 µm at a sweeping rate of 60 KHz.

Transient receptor potential vanilloid 3 (TRPV3) is a member of the TRP superfamily. Previous studies have demonstrated that TRPV3 is associated with myocardial fibrosis. However, the role of TRPV3 in hepatic fibrosis and its underlying mechanisms are still unclear. This study aimed to elucidate the underlying effects of TRPV3 on hepatic fibrosis at multiple biological levels. First, immunohistochemical staining was performed to examine TRPV3 expression in human hepatic cirrhosis tissues. Then, we established a CCl4-induced hepatic fibrosis mouse model. The TRPV3 selective agonist drofenine and its inhibitor, forsythoside B, were intraperitoneally injected to investigate the relationship between TRPV3 and liver fibrosis progression. Finally, in vitro studies were performed using hepatic stellate cells (HSCs) to discover the potential molecular biological mechanisms. Immunohistochemistry revealed TRPV3 overexpression in liver cirrhosis. In the liver fibrosis groups, TRPV3 inhibitor treatment significantly reduced liver fibrosis, while TRPV3 agonist exacerbated its progression. In HSCs, knocking down TRPV3 with siRNA impaired DNA synthesis and cell proliferation and increased cell apoptosis. Furthermore, we found that knockdown of TRPV3 could reduce the lectin like oxidized lowdensity lipoprotein receptor-1 (LOX-1) protein levels. Our research suggests that lower expression or functional levels of TRPV3 can ameliorate the inflammatory response and fibrotic tissue proliferation.

When the signal-to-noise ratio (SNR) of an optical fiber sensor is lower than a specific value, jump errors will appear in the maximum-likelihood-estimation (MLE) algorithm. This research proposes a quantitative calculation method based on the MLE algorithm. This method can calculate the probability of jump errors by different SNRs. The SNR threshold can also be obtained in the MLE algorithm for the low-finesse interference spectrum. From the simulation, we find that when the SNR varies from 0 dB to 25 dB, the possibility of jump errors decreases exponentially. Subsequent experiments also verify the correctness of the simulation results.

A fast real-time demodulation method based on the coarsely sampled spectrum is proposed for transient signals of fiber optic extrinsic Fabry-Perot interferometers (EFPI) sensors. The feasibility of phase demodulation using a coarse spectrum is theoretically analyzed. Based on the coarse spectrum, fast Fourier transform (FFT) algorithm is used to roughly estimate the cavity length. According to the rough estimation, the maximum likelihood estimation (MLE) algorithm is applied to calculate the cavity length accurately. The dense wavelength division multiplexer (DWDM) is used to split the broadband spectrum into the coarse spectrum, and the high-speed synchronous ADC collects the spectrum. The experimental results show that the system can achieve a real-time dynamic demodulation speed of 50 kHz, a static measurement root mean square error (RMSE) of 0.184 nm, and a maximum absolute and relative error distribution of 15 nm and 0.005% of the measurement cavity length compared with optical spectrum analyzers (OSA).

BACKGROUND: This is a case of spontaneous toric implantable collamer lens (TICL) rotation that occurred twice in the left eye of a patient. CASE PRESENTATION: A 24-year-old gentleman received TICL implantation for treatment of myopic astigmatism encountered with spontaneous rotation of approximately 90° from its original position. TICL reposition procedure was performed with visual outcome of 20/16. Surprisingly, a re-rotation of TICL occurred. The patient underwent a replacement of non-toric ICL with a larger size after careful re-evaluation with final visual outcome of 20/20. A short and small ciliary process with shallow ciliary sulcus and posteriorly positioned ciliary body was found by Ultrasound Biomicroscopy (UBM). CONCLUSIONS: The unique morphology of the ciliary body may have a potential association with the vault and instability of implanted ICL. Careful examination of the ciliary body morphology is essential in preoperative evaluation.

High-temperature blackbody radiation can provide the light source for broadband fiber sensing systems. To improve coupling efficiency, a coupling model of light intensity from the blackbody cavity into a cone-shaped optical fiber was proposed. Influence of parameters of cone-shaped fiber and the blackbody cavity on coupling efficiency of light intensity were analyzed. Results show that coupling efficiency first decreases and then increases with the increase of the angle of cone-shaped fiber. When the angle of the cone is around 30°, coupling efficiency reaches the maximum. Cone-shaped fibers with angles around 8.8°, 19.4°, and 27.4° were fabricated by chemical etching. Experiments were carried out with cone-shaped fibers and a fiber with a flat end face. Results show that compared with the fiber with a flat end face, coupling efficiency of cone-shaped fiber with an angle around 8.8° is lower, cone-shaped fiber with an angle around 19.4° is almost the same, cone-shaped fiber with angle 27.4° is much stronger, and efficiency is improved by approximately 38%. The experimental results are consistent with the theoretical analysis.

PURPOSE: To assess corneal power measurements obtained by the OPD SCAN III Topographer in eyes with prior myopic small incision lenticule extraction (SMILE) surgery. METHODS: Sixty untreated myopic eyes of sixty subjects and forty previous myopic SMILE surgery eyes of forty subjects were consecutively enrolled in the present study. Manifest refraction, OPD SCAN III and Pentacam HR were performed. Keratometric measurements assessed by OPD SCAN III-simulated keratometry, average pupil power and effective central corneal power (ECCP) were compared with mean keratometry (Km) obtained by Pentacam HR in the untreated group and the clinical history method (CHM) in the treated group. RESULTS: In the untreated group, no statistically significant differences were revealed between all corneal power measurements obtained with OPD SCAN III and Km. In the treated group, all the corneal power measurements were statistically different from the CHM except for the Haigis method and the Shammas method, while ECCP had a statistically but not clinically significant overestimation of 0.42 D with 95% limit of agreement (LOA) of - 0.81 D to 1.64 D. The three modified ECCP had better prediction performance with narrower 95% of LOA lying in (- 1.20, 1.20 D) (- 1.22, 1.23 D) and (- 0.90, 1.00 D), respectively. CONCLUSIONS: The ECCP provided with OPD SCAN III could be used as an alternative option for the CHM after specific modifications in eyes with previous myopic SMILE surgery when the preoperative data are unavailable considering the narrowest agreement between the modified ECCP and the CHM. Otherwise, caution must be raised considering the wide LOA.

BACKGROUND/AIMS: Genomic adaptations to high altitudes have been well studied in the last several years; however, the roles of microRNAs (miRNAs), which are essential modulators of a variety of genes and key cellular processes, have rarely been explored. Here, we explored the interactions between miRNAs and their target genes as an adaptation to high altitude in an avian species, the great tit (Parus major), which is widely distributed across the Eurasian continent at altitudes between 4500 m and sea level. Because the MAPK signaling pathway plays a crucial role in the hypoxia response in the great tit, we chose MAPK1 as a target candidate gene. METHODS: We established a great tit embryonic fibroblast line and subsequently studied the relationship between miRNA-19b-3p and MAPK1 in normoxia and hypoxia groups. Meanwhile, the great tit embryonic fibroblasts (GEFs) were treated or transfected with miR-19b-3p mimics, inhibitors, or si-MAPK1, and their proliferation was subsequently assessed using the MTT assay. The expression of the miRNAs and MAPK1 was measured by real-time PCR and Western blotting. RESULTS: We identified 14 miRNAs in the cardiac tissues of great tits that are related to hypoxia adaptation. MAPK1 binds only to miR-19b-3p of the 14 miRNAs predicted by both TargetScan and miRanda software. Specifically, we validated the computational prediction of miR-19b-3p binding to the 3'UTR of MAPK1 using a luciferase reporter assay. Our results show that miR-19b-3p promotes GEFs proliferation and up-regulates MAPK1 expression. Moreover, miR-19b-3p mimics and MAPK1 knockdown induce GEFs apoptosis and regulate the cell cycle under hypoxic conditions. CONCLUSIONS: Our study is the first to describe an important miRNA-mediated regulatory mechanism of high altitude adaptation in a non-model wild songbird and highlights the importance of studies on miRNA-mediated mechanisms of hypoxic adaptations in other animals.

Fiber Bragg grating (FBG) sensors are prone to spectral distortions in practical applications, which may cause large demodulation errors. There are many algorithms demodulating FBG spectra, but no approach is able to evaluate demodulation results under spectral distortions. To address this problem, four spectral indices are comparatively utilized to quantify distortions of spectra. Seven representative demodulation algorithms are adopted to analyze impacts of various simulated distorted spectra. Simulation results show that the side-mode suppression ratio (SMSR) of the spectrum is the most suitable spectral index for accuracy evaluation of demodulation results. In particular, when SMSR is larger than 6.3 dB, demodulation results will be accurate, and they are inaccurate when SMSR is smaller than 2.1 dB. The effectiveness of the proposed evaluation approach is validated with fatigue experiments of 9 FBG sensors.

Multipotent skin-derived precursors (SKPs) are dermal stem cells with the capacity to reconstitute the dermis and other tissues, such as muscles and the nervous system. Thus, the easily available human SKPs (hSKPs) hold great promises in regenerative medicine. However, long-term expansion is difficult for hSKPs in vitro. We previously demonstrated that hSKPs senesced quickly under routine culture conditions. To identify the underlying mechanisms so as to find an effective way to expand hSKPs, time-dependent microarray analysis of gene expression in hSKPs during in vitro culture was performed. We found that the senescence of hSKPs had a unique gene expression pattern that differs from reported typical senescence. Subsequent investigation ruled out the role of DNA damage and classical p53 and p16(INK4a) signaling in hSKP senescence. Examination of cyclin-dependent kinase inhibitors revealed the involvement of p15(INK4b) and p27(KIP1). Further exploration about upstream signals indicated the contribution of Akt hypo-activity and FOXO3 to hSKP senescence. Forced activation of Akt and knockdown of FOXO3, p15(INK4b) and p27(KIP1) effectively inhibited hSKP senescence and promoted hSKP proliferation. The unique senescent phenotype of human dermal stem cells and the role of Akt-FOXO3-p27(KIP1)/p15(INK4b) signaling in regulating hSKP senescence provide novel insights into the senescence and self-renewal regulation of adult stem cells. The present study also points out a way to propagate hSKPs in vitro so as to fulfill their promises in regenerative medicine.

R-spondin proteins are novel Wnt/ß-catenin agonists, which signal through their receptors leucine-rich repeat-containing G-protein coupled receptor (LGR) 4/5/6 and substantially enhance Wnt/ß-catenin activity. R-spondins are reported to function in embryonic development. They also play important roles in stem cell functions in adult tissues, such as the intestine and mammary glands, which largely rely on Wnt/ß-catenin signaling. However, in the skin epithelium and hair follicles, the information about R-spondins is deficient, although the expressions and functions of their receptors, LGR4/5/6, have already been studied in detail. In the present study, highly-enriched expression of the R-spondin family genes (Rspo1/2/3/4) in the hair follicle dermal papilla is revealed. Expression of Rspo1 in the dermal papilla is specifically and prominently upregulated before anagen entry, and exogenous recombinant R-spondin1 protein injection in mid-telogen leads to precocious anagen entry. Moreover, R-spondin1 activates Wnt/ß-catenin signaling in cultured bulge stem cells in vitro, changing their fate determination without altering the cell proliferation. Our pioneering study uncovers a role of R-spondin1 in the activation of cultured hair follicle stem cells and the regulation of hair cycle progression, shedding new light on the governance of Wnt/ß-catenin signaling in skin biology and providing helpful clues for future treatment of hair follicle disorders.

In scientific research and engineering application, improving the power of fiber device is an important topic, which leads to observably rise of temperature in fiber core at the same time. In this paper, Thermal effect and its influence on absorption spectrum and lifetime of Erbium-doped fiber are studied with numerical modeling. Lorentz broadening of sub-levels is used to build the mathematical relationship between temperature and absorption spectrum. The McCumber Theory is applied to deduce the lifetime of Erbium-doped fiber in different temperature. Temperature experiments of absorption and emission spectrum from 25 to 900 ℃ are carried out, which show that the wavelength of absorption peak near 980nm increase at rate of 0.625 nm/100 ℃, the ratio of absorption peak near 1 530 nm declines at a rate of 0.001 9 dB·(m℃)-1 and the broadband of absorption spectrum near 1 530 nm increase with rising temperature. The linear variation of lifetime and peak absorption in experiment proves that the theoretical model is reasonable when the temperature is below 600 ℃.
