RESUMEN

High-finesse microcavities offer a platform for compact, high-precision sensing by employing high-reflectivity, low-loss mirrors to create effective optical path lengths that are orders of magnitude larger than the device geometry. Here, we investigate the radiation hardness of Fabry-Pérot microcavities formed from dielectric mirrors deposited on the tips of optical fibers. The microcavities are irradiated under both conventional (∼ 0.1 Gy/s) and ultrahigh (FLASH, ∼ 20 Gy/s) radiotherapy dose rates. Within our measurement sensitivity of ∼ 40 ppm loss, we observe no degradation in the mirror absorption after irradiation with over 300 Gy accumulated dose. This result highlights the excellent radiation hardness of the dielectric mirrors forming the cavities, enabling new optics-based, real-time, in-vivo, tissue-equivalent radiation dosimeters with ∼ 10 micron spatial resolution (our motivation), as well as other applications in high-radiation environments.

The aim of the study was to evaluate the effect of total mixed ration particle size (length) and breed of cow on intake dynamics, animal performance and CH4 emissions, comparing high yielding Holstein and low yielding Girolando cows. The experimental design was 2 × 2 Latin Square arranged as a crossover factorial scheme with two diets (short particle size, SPS and long particle size, LPS) and the two breed compositions. The design comprised two periods of 26 d each, where all data collection was performed at cow level. No influence of the particle size occurred for the passage rate, neutral detergent fiber digestibility, performance and milk composition, methane emissions or ruminal fermentation parameters. Girolando cows had greater dry matter intake (DMI) when fed SPS, while Holsteins had the same (P < 0.05). Girolando cows had lower dry matter digestibility when fed LPS compared to SPS, while Holsteins had the opposite effect (P < 0.05). Also, the digestibility of crude protein and non-fibrous carbohydrates decreased in Girolando cows fed LPS, but not in Holsteins (P < 0.05). Girolando cows reduced DMI by 10.6% when fed LPS diet (P < 0.05). Girolando had an increased eating rate (+24 g of DM/min; P < 0.05) compared to Holstein cows, but Holstein cows had a lower CH4 intensity (by 29.7%: P < 0.05). Girolando cows increased the dry matter intake when fed a diet with short particle size, while the same did not happen in Holsteins. Dry matter digestibility increased in Holsteins when fed long particle size, while the opposite was observed in Girolando cows. Nutrient digestibility was reduced in Girolando cows when fed short particle size. Particle size did not influence eating time, eating rate, feed trough visits, visits with intake, milk yield and composition regardless of the breed. Reducing particle size increased CH4 intensity in both breeds.

This study aimed to evaluate intake, energy and nitrogen balance as well as methane emission in Holstein and ½ Holstein ½ Gyr (Girolando-F1) cows during the transition period. Twenty-four cows (12 Holstein and 12 Girolando-F1) were used to evaluate feed intake, apparent digestibility, heat production and methane emission, carried out in two periods: from 28 to 19 days pre-calving and from 15 to 23 days post-calving. A completely randomised design was used and data were analysed by ANOVA within periods (pre- and post-calving) considering the main effect of genetic groups. Girolando-F1cows presented greater body condition score (BCS) compared with Holstein. During pre-calving, there were no differences between genetic groups, except for highest heat production per kilogram of metabolic body weight for Holstein cows. After calving, Holstein cows had greater intake of DM, nitrogen, NDF per kg of BW and produced more heat per kg of metabolic body weight. Holstein cows yielded more milk and fat-corrected milk (FCM4%) compared with Girolando-F1 cows. Holstein cows presented higher methane emission per unit of BW and of metabolic weight. Emissions of enteric methane per kilogram of milk and per kilogram of FCM4% tended to be lower for Holstein compared with Girolando-F1 cows. Nitrogen and energy retention were similar for both Holstein and Girolando-F1 at pre- and post-calving. Despite differences in BCS, DMI, and milk yield, Girolando-F1 and Holstein cows present overall similar energy efficiency, albeit Holstein cows tended to present less methane emission per kg of eligible product (milk).

We aimed to evaluate the effects of different drying methods for banana residues on the energy metabolism and respirometry of growing lambs. Twenty Santa Inês x Dorper lambs were fed five experimental diets: Tifton 85 grass hay (Control), shade-dried banana leaf hay (LH Shade), shade-dried pseudostem banana hay (PH Shade), sun-dried banana leaf hay (LH Sun), and sun-dried banana pseudostem hay (PH Sun). Nutrient intake and digestibility were assessed in metabolic cages, whereas O2 consumption and CO2, methane, and heat production were measured in a respirometry chamber with animals fed at maintenance and ad libitum levels. Nutrient and energy intake was not influenced by diet. Pseudostem hay had higher apparent digestibility of dry matter (71.5%), organic matter (72.4%), and neutral detergent fiber (58.0%). However, this led to greater energy loss in the form of methane (12.1%). The banana residue hays and drying methods did not alter oxygen consumption, CO2 production, or heat production of animals fed ad libitum or during maintenance. On the other hand, the use of leaf hay resulted in a reduction of 24.7% in enteric methane production of animals fed ad libitum. The inclusion of pseudostem hay is recommended in sheep feedlot diet. This residue provided greater use of DM, however promoted a greater loss of energy in the form of methane, resulting in similar energy consumption. The drying methods did not reduce the availability of nutrients and the sun drying method is recommended, since it is a faster drying method.

The knowledge of the relationship between the chemical structure of food components with their mechanisms of action is crucial for the understanding of diet health benefits. This review relates the chemical variability present in coffee beverages with the mechanisms involved in key physiological events, supporting coffee as a polyvalent functional food. Coffee intake has been related with several health-promoting properties such as neuroprotective (caffeine, chlorogenic acids and melanoidins), anti-inflammatory (caffeine, chlorogenic acids, melanoidins, diterpenes), microbiota modulation (polysaccharides, melanoidins, chlorogenic acids), immunostimulatory (polysaccharides), antidiabetic (trigonelline, chlorogenic acids), antihypertensive (chlorogenic acids) and hypocholesterolemic (polysaccharides, chlorogenic acids, lipids). Nevertheless, caffeine and diterpenes are coffee components with ambivalent effects on health. Additionally, a large range of potentially harmful compounds, including acrylamide, hydroxymethylfurfural, furan, and advanced glycation end products, are formed during the roasting of coffee and are present in the beverages. However, coffee beverages are part of the daily human dietary healthy habits, configuring a coffee paradox.


The multi-targeted bioactive features of coffee compounds reinforce coffee as a functional food beverage.Polysaccharides and melanoidins positively modulate gut microbiota.Caffeine and phenolics are neuroprotective, anti-inflammatory, antidiabetic and antihypertensive.The balance between potential health and harmful compounds configures a coffee paradox.Harmful compounds are present in trace levels in coffee, not conferring toxicity.

Biosimilars are biological medicines highly similar to a previously licensed reference product and their licensing is expected to improve access to biological therapies. This study aims to present an overview of biosimilars approval by thirteen regulatory authorities (RA). The study is a cross-national comparison of regulatory decisions involving biosimilars in Argentina, Australia, Brazil, Chile, Canada, Colombia, Europe, Hungary, Guatemala, Italy, Mexico, Peru and United States. We examined publicly available documents containing information regarding the approval of biosimilars and investigated the publication of public assessment reports for registration applications, guidelines for biosimilars licensing, and products approved. Data extraction was conducted by a network of researchers and regulatory experts. All the RA had issued guidance documents establishing the requirements for the licensing of biosimilars. However, only three RA had published public assessment reports for registration applications. In total, the investigated jurisdictions had from 19 to 78 biosimilars approved, most of them licensed from 2018 to 2020. In spite of the advance in the number of products in recent years, some challenges still persist. Limited access to information regarding the assessment of biosimilars by RA can affect confidence, which may ultimately impact adoption of these products in practice.

Infrared thermography (IRT) is a non-invasive and efficient tool to detect changes in the animal's body surface temperature, which is directly associated with the animal's energy loss. Methane emission represents a significant energy loss, particularly in ruminants, as well as heat production. Therefore, the aim of this study was to correlate skin temperature measured using IRT with heat production (HP) and methane emission in lactating Holstein and crossbred ½ Holstein x ½ Gyr (Gyrolando-F1) cows. Six Gyrolando-F1 and four Holstein cows, all primiparous, at mid lactation were used to evaluate daily HP and methane emission using indirect calorimetry in respiratory chambers. Thermographic images were taken at anus, vulva, ribs (on the right side), left flank, right flank, right front foot, upper lip, masseter and eye; IRT was performed every hour during 8 h after the morning feeding. Cows received the same diet ad libitum. Daily methane emission was positively correlated with IRT taken at the right front foot 1 h after feeding in Gyrolando-F1 cows (r = 0.85, P < 0.05) and with IRT taken at the eye 5 h after feeding in Holstein cows (r = 0.88, P < 0.05). HP was positively correlated with IRT taken at the eye 6 h after feeding in Gyrolando-F1 cows (r = 0.85, P < 0.05) and with IRT taken at the eye 5 h after feeding in Holstein cows (r = 0.90, P < 0.05). Infrared thermography was positively correlated with HP and methane emission in both Holstein and Gyrolando-F1 lactating cows, although the anatomical points and times of image acquisition yielding the greatest correlation coefficients varies among breeds.

OBJECTIVE: To validate the level of agreement between self-report and clinical examination for oral conditions and evaluate the effect of sociodemographic conditions on the validity of self-report among women aged 60 and older. METHODS: A cross-sectional study was conducted in a social community center for seniors in Southern Brazil. Sociodemographic data (age, level of education, and income) were measured. Participants were interviewed and clinically examined for the number of teeth (DMF-T index) and the use of dental prostheses. The self-reported number of teeth in each arch and the use of dental prostheses were gathered through interviews. The level of agreement was estimated using the observed agreement, Kappa statistics, sensitivity/specificity (edentulism/prostheses) and Lin's concordance correlation coefficient, and related tests (number of teeth). The validity of the oral conditions was estimated according to sociodemographic information. RESULTS: Ninety-nine women participated in the study. High levels of agreement were observed for edentulism (97.8%; 95%CI 92.8;99.7; Kappa 0.947) and the use of dental prostheses (97.0%; 95%CI 91.3;99.4; Kappa 0.922). In both conditions, despite achieving similar concordance correlation coefficients (ranging from weak to moderate), the mean number of upper teeth was lower in clinical examination (7.1 ± 5.2) compared with self-reported (8.6 ± 3.6), while the opposite was observed for lower teeth (clinical examination: 9.1 ± 3.4; self-reported: 6.6 ± 5.3). Larger differences were found among women of low income and educational levels. CONCLUSIONS: Our findings suggest that the participants' socio-economic position might influence their self-reported number of teeth.

We present a simple, continuous, cavity-enhanced optical absorption measurement technique based on high-bandwidth Pound-Drever-Hall (PDH) sideband locking. The technique provides a resonant amplitude quadrature readout that can be mapped onto the cavity's internal loss rate and is naturally compatible with weak probe beams. With a proof-of-concept 5-cm-long Fabry-Perot cavity, we measure an absorption sensitivity ∼10-10cm-1/Hz from 30 kHz to 1 MHz, and a minimum value of 6.6×10-11cm-1/Hz at 100 kHz, with 38 µW collected from the cavity's circulating power.

BACKGROUND: Ionophore antibiotics improve the efficiency of energy metabolism, which has driven their use as a feed additive in ruminants for decades. Currently, they have not been approved in many countries, generating a challenge for the immediate search for plant extracts with a similar mode of action on rumen metabolism. This study evaluated the effects of enriched Prosopis juliflora (mesquite) piperidine alkaloid extract (MPA) levels as an alternative phytoadditive to sodium monensin (MON) in sheep. RESULTS: The MPA diet did not differ from MON with regard to nutrient intake. A quadratic effect (P < 0.05) was observed for organic matter and neutral detergent fibre digestibility, with respective maximum point at 25.40 and minimum point at 0.95 mg kg-1 MPA. The MPA levels linearly decreased (P < 0.05) faecal nitrogen loss. MPA did not differ from MON with regard to nutrient digestibility, and MPA levels increased (P < 0.05) the proportion of digestible energy and metabolizability from dietary gross energy. The MPA levels linearly decreased (P < 0.05) enteric CH4 production, the yield showing lower (P < 0.05) energy loss as CH4 than MON. CONCLUSION: The results show that MPA levels of 17.3 and 27.8 mg kg-1 are enteric CH4 inhibitors and enhance energy and protein utilization, indicating a promising alternative to MON for ruminants. © 2022 Society of Chemical Industry.

Both memory and choice are influenced by context: Memory is enhanced when encoding and retrieval contexts match, and choice is swayed by available options. Here, we assessed how context influences risky choice in an experience-based task in two main experiments (119 and 98 participants retained, respectively) and two additional experiments reported in the Supplemental Material available online (152 and 106 participants retained, respectively). Within a single session, we created two separate contexts by presenting blocks of trials in distinct backgrounds. Risky choices were context dependent; given the same choice, people chose differently depending on other outcomes experienced in that context. Choices reflected an overweighting of the most extreme outcomes within each local context rather than the global context of all outcomes. When tested in the nontrained context, people chose according to the context at encoding and not retrieval. In subsequent memory tests, people displayed biases specific to distinct contexts: Extreme outcomes from each context were more accessible and judged as more frequent. These results pose a challenge for theories of choice that rely on retrieval as guiding choice.

RESUMEN

Several classes of polysaccharides have been described to have hypocholesterolemic potential, namely cholesterol bioaccessibility and bioavailability. This review will highlight the main mechanisms by which polysaccharides are known to affect cholesterol homeostasis at the intestine, namely the effect (i) of polysaccharide viscosity and its influence on cholesterol bioaccessibility; (ii) on bile salt sequestration and its dependence on the structural diversity of polysaccharides; (iii) of bio-transformations of polysaccharides and bile salts by the gut microbiota. Different quantitative structure-hypocholesterolemic activity relationships have been explored depending on the mechanism involved, and these were based on polysaccharide physicochemical properties, such as sugar composition and ramification degree, linkage type, size/molecular weight, and charge. The information gathered will support the rationalization of polysaccharides' effect on cholesterol homeostasis and highlight predictive rules towards the development of customized hypocholesterolemic functional food.

The objective of this study was to evaluate the metabolic profile of Santa Inês ewe lambs fed diets for early or late-maturing diets with or without nutrient restrictions. The experiment consisted of a 2 × 2 completely randomized factorial experiment with either early- or late-maturity feed formulation according to "Nutrient Requirements of Small Ruminants" with or without 15% crude protein (CP) and total digestible nutrients (TDN) restrictions in diets formulated, five replications, and 20 ewe lambs averaging 15.1±2.6 kg. Lambs on early-maturity diets consumed greater (P<0.05) dietary ether extract (EE), non-fibrous carbohydrates, and TDN than those on late-maturity diets. Lambs on early-maturity diets had 7.11% greater dry matter digestibility (DMD) compared to lambs fed late-maturity diets. Lambs fed late-maturity diets, in general, had greater intake (IN), excreted (EN), and retained (RN) N as well as greater RN/IN and EN/IN ratios. There were no differences in blood total protein or albumin among lambs fed for different finishing maturity targets. Diets designed for late-maturing lambs resulted in greater microbial N and CP as well as rumen and metabolizable, degradable, and undegradable rumen and metabolizable CP. The selection of diets for early or late maturity carcasses depends on the production system goals. Diets without restrictions are recommended for early-maturity carcass finishing while diets with 15% CP and TDN restriction are recommend for late-maturity finishing.

This study aimed to evaluate the effects of plane of nutrition and advancing days of pregnancy (DP) on maternal body composition and fetal development. Differing planes of nutrition were established by 2 feeding regimens (FR): ad libitum (AL) or maintenance (MA). Sixty-two nonlactating multiparous Holstein × Gyr cows with average body weight of 480 ± 10.1 kg and an age of 5 ± 0.5 yr were used. Cows were divided into 3 groups: pregnant (n = 44), nonpregnant (n = 12), and baseline reference cows (n = 6). The 56 pregnant and nonpregnant cows were randomly allocated into 2 different FR: AL or MA. Cows fed at MA received 1.15% of their body weight on a dry matter (DM) basis, receiving corn silage and a concentrate-based diet at a ratio of 93:7 on a DM basis. Reference group cows were slaughtered at the beginning of the experimental period to estimate body composition and empty body weight. To evaluate the effects of DP, pregnant and nonpregnant animals were slaughtered at d 140, 200, 240, and 270 of gestation. Feeding regimen affected maternal tissue composition. Days of pregnancy affected fresh weight (FW), DM, and energy content, but no differences were observed for crude protein (CP) and ether extract (EE) because of DP. Feeding regimen affected mammary gland components (CP, EE, and energy content), but not fresh or dry weights. Days of pregnancy influenced almost all mammary gland components except energy content. Regarding the uterus, FR affected only fresh and dry weights; however, DP affected every uterus component measured. The only interaction between FR and DP in this study was observed for placental FW. Cows fed AL on d 270 presented the same placental FW as cows at MA and AL on d 200 and 240. Further, pregnant cows fed at MA on d 270 had greater placental FW than cows fed AL at this day. Days of pregnancy, but not FR, influenced the composition of fetal fluids in pregnant cows. Finally, cows fed at MA had greater FW for the fetus than cows fed AL; however, fetus composition changed over DP. The FW, DM, EE, and energy content increased until d 270, but CP decreased. In conclusion, the novelty of our data presents how changes due to FR and DP occur in maternal tissues and the conceptus.

OBJECTIVE: To describe the clinical and epidemiological profile of cases with confirmed microcephaly or central nervous system (CNS) findings associated with congenital Zika virus infection and other infectious etiologies in the state of Rio de Janeiro, Brazil, from November 2015 to July 2017. METHOD: A cross-sectional study was performed with 298 cases (as defined by the Ministry of Health) communicated to the Rio de Janeiro State Department of Health in the study period. Demographic, epidemiological, clinical, radiological, and laboratory variables were assessed. Descriptive bivariate and multivariable logistic regression analysis was used to determine the association between specific factors and death outcome. RESULTS: The median age of mothers was 24 years; 30.9% reported fever and 64.8% reported a rash during pregnancy. The median head circumference at birth was 29 cm, and median birth weight was 2 635 g. An etiological diagnosis of congenital Zika was made in 46.0%, whereas 13.8% were diagnosed with syphilis, toxoplasmosis, rubella, cytomegalovirus, and herpes simplex infections (STORCH), with predominance of syphilis, and 40.3% had an unspecified infectious agent. CNS findings other than microcephaly were observed in 88.3%, especially intracranial calcifications, ventriculomegaly, and brain atrophy. Overall lethality was 7.0% - 19.0% in laboratory confirmed Zika cases and 22.2% in toxoplasmosis cases. Multivariable analysis revealed birth weight as the main predictor of death. CONCLUSIONS: Despite the Zika epidemic, 13.8% of the studied cases were diagnosed with STORCH. The lethality and high frequency of neurological findings beyond microcephaly reflect severe infection, with impact on families and health care system.


OBJETIVO: Describir el perfil clínico-epidemiológico de los casos confirmados de microcefalia y de alteraciones del sistema nervioso central (SNC) relacionados con la infección congénita por el virus del Zika y otras etiologías infecciosas en el Estado de Río de Janeiro en el período comprendido entre noviembre del 2015 y julio del 2017. MÉTODOS: Se realizó un estudio transversal de 298 casos (según la definición del Ministerio de Salud) notificados a la Secretaría de Estado de Salud de Río de Janeiro en el período objeto de estudio. Se analizaron variables demográficas, epidemiológicas, clínicas, radiológicas y de laboratorio, con un análisis estadístico descriptivo bivariado y de regresión logística múltiple para estudio de los factores relacionados con la defunción. RESULTADOS: La edad mediana de las madres fue de 24 años; un 30,9% informó fiebre y un 64,8%, exantema durante la gestación. La mediana del perímetro cefálico al nacer fue de 29 cm y la del peso, de 2635 g. El diagnóstico etiológico fue de infección congénita por el virus del Zika en un 46,0%; sífilis, toxoplasmosis, rubéola, infección por citomegalovirus e infección por el virus del herpes simple (STORCH) en un 13,8%, con predominio de sífilis; e infección por un agente infeccioso no definido en un 40,3%. Se describieron alteraciones del SNC diferentes de microcefalia en un 88,3%, con predominio de calcificaciones cerebrales, ventriculomegalia y atrofia cerebral. La letalidad total alcanzó 7,0%; se confirmaron en el laboratorio 19,0% de los casos de infección por el virus del Zika y 22,2% de los casos de toxoplasmosis. En el análisis de regresión logística múltiple, el peso al nacer fue el principal pronóstico de defunción. CONCLUSIONES: A pesar de la epidemia de la infección por el virus del Zika, 13,8% de los casos fueron causados por STORCH. La letalidad y la elevada presencia de malformaciones neurológicas, además de microcefalia, muestran la gravedad de la infección y sus repercusiones para las familias y para el sistema de salud.

The effect of exogenous amylase on the in vitro rumen digestion kinetics of whole-crop maize silage made from dent (RB9004) or flint grain type (RB9308) was evaluated at different phenological stages: soft dough (SOD), early dent (EAD), ½ milkline (½M) and ¾ milkline (¾M). Forage was harvested from 70 to 110 days after sowing. Two rumen-cannulated cows receiving or not exogenous amylase (0.7 g/kg dry matter-DM, provided to achieve 396 kilo Novo units of amylase activity/kg of TMR DM) were used as donor of ruminal fluid. The in vitro gas production kinetics was evaluated according to a dual-pool logistic model. The chemical composition and gas production kinetics were affected by the hybrid and phenological stages. The flint hybrid had lower range for chemical analysis among physiological stages. Harvesting at ½M and ¾M improved DM content, bromatological composition and silage quality parameters compared to dent or flint types. Amylase (i) increased methane (CH4 ) production and in vitro dry matter digestibility (IVDMD) in ½M stage, (ii) improved digestion kinetics by reducing lag time and increasing total gas production and fermentation rates of non-fibrous carbohydrates (NFC) and fibrous carbohydrates (FC), and (iii) increased extent and fermentation rate of NFC and increased fermentation rate of FC fraction in whole-crop maize silages produced from dent or flint types in all phenological stages. Harvesting between ½M and ¾M is the best phenological stage to improve chemical composition and silage quality parameters. Exogenous amylase showed improvements on fibre digestion of silages at ½M and ¾M phenological stages in both grain types of corn.

The objective was to evaluate reproductive characteristics of crossbred Girolando (Gyr x Holstein) bulls from two breed compositions and correlate these results with infrared thermography data. Evaluations were performed considering sperm motility, vigor and morphology; scrotal circumference; body morphology and temperament. Infrared thermography was performed to determine surface temperatures of ocular and scrotal areas. Thermoregulation capacity was assessed by differences between air and rectal temperatures, air and maximum temperatures in ocular and scrotal areas, and dorsal and ventral lines of the scrotum. Data analysis was performed using a linear mixed model (breed composition as fixed effect and year of evaluation as random effect). Spearman correlation coefficient was used to associate thermography and reproductive data. Girolando 3/4 Holstein bulls had higher (P ≤ 0.001) scrotal circumference and higher average body morphology and temperament, whereas 5/8 Holstein bulls had a higher (P < 0.001) percentage of major-type and total sperm defects. Girolando 3/4 Holstein bulls had scrotal temperatures 0.8 °C higher (P < 0.001) and 5/8 Holstein bulls had 9.8% and 10.6% higher differences on "rectal - scrotal area" and "ocular area - scrotal area" temperatures, respectively. Ocular area temperatures had negative correlations (P ≤ 0.1) with sperm motility and vigor, and positive correlations (P < 0.001) with minor-type and total sperm defects. Ventral line scrotal temperatures had positive correlations (P < 0.001) with minor-type and total sperm defects. Girolando 3/4 Holstein bulls were found to be superior to 5/8 Holstein bulls for reproductive characteristics. Under non-stressing climatic conditions, semen characteristics of Girolando bulls were more influenced by breed composition than by the capacity for scrotal thermoregulation. Correlations between semen quality and scrotal temperatures can aid in the identification of bulls for breeding, particularly when a large number of animals are in the tests.

The objective of this study was to evaluate the production, consumption, and energy balance parameters of primiparous 3/4 and 7/8 Holstein × Gir (HG) dairy cows fed two diets of differing energy levels during the postpartum period. At the beginning of the study, 28 days prepartum, the average weight of both genetic groups was 498 ± 12 kg and body condition score (BCS) was 3.5 ± 0.05. At the end of the study, 61 days postpartum, the 3/4 HG cows had higher weight and body condition scores than the 7/8 HG (456 ± 8 and 429 ± 8 kg and 3.13 ± 0.03 and 2.94 ± 0.03 BCS for 3/4 HG and 7/8 HG, respectively). Milk from cows fed the high-energy diet had higher percentages of fat, protein, lactose, and total dry extract than cows fed the low-energy diet. Cows fed the high-energy diet had higher net energy intake (95.3 ± 1.9 vs. 88.1 ± 2.1 MJ/day) and higher energy balance (3.64 ± 2.13 vs - 6.02 ± 2.30 MJ/day). The 3/4 HG cows displayed higher energy for maintenance (33.1 ± 0.4 MJ/day) than the 7/8 HG (31.5 ± 0.5 MJ /day). In conclusion, although the primiparous 3/4 HG were heavier than the 7/8 HG and had a higher body condition score, no differences in milk produced up to 60 days postpartum were observed. The higher energy diet during the postpartum period increased energy balance, resulting in higher production of milk fat, protein, and lactose.

Microbial communities play critical roles in the gastrointestinal tracts (GIT) of preruminant calves by influencing performance and health. However, little is known about the establishment of microbial communities in the calf GIT or their dynamics during development. In this study, next-generation sequencing was used to assess changes in the bacterial communities of the rumen, jejunum, cecum, and colon in 26 crossbred calves at four developmental stages (7, 28, 49, and 63 days old). Alpha diversity differed among GIT regions with the lowest diversity and evenness in the jejunum, whereas no changes in alpha diversity were observed across developmental stage. Beta diversity analysis showed both region and age effects, with low numbers of operational taxonomic units (OTUs) shared between regions within a given age group or between ages in a given region. Taxonomic analysis revealed that several taxa coexisted in the rumen, jejunum, cecum, and colon but that their abundances differed considerably by GIT region and age. As calves aged, we observed lower abundances of taxa such as Bacteroides, Parabacteroides, and Paraprevotella with higher abundances of Bulleidia and Succiniclasticum in the rumen. The jejunum also displayed taxonomic changes with increases in Clostridiaceae and Turicibacter taxa in older calves. In the lower gut, taxa such as Lactobacillus, Blautia, and Faecalibacterium decreased and S24-7, Paraprevotella, and Prevotella increased as calves aged. These data support a model whereby early and successive colonization by bacteria occurs across the GIT of calves and provides insights into the temporal dynamics of the GIT microbiota of dairy calves during preweaning development.IMPORTANCE The gastrointestinal tracts (GIT) of ruminants, such as dairy cows, house complex microbial communities that contribute to their overall health and support their ability to produce milk. For example, the rumen microbiota converts feed into usable nutrients, while the jejunal microbiota provides access to protein. Thus, establishing a properly functioning GIT microbiota in dairy calves is critical to their productivity as adult cows. However, little is known about the establishment, maintenance, and dynamics of the calf GIT microbiota in early life. In this study, we evaluated the bacterial communities in the rumen, jejunum, cecum, and colon in dairy calves across preweaning development and show that they are highly variable early on in life before transitioning to a stable community. Understanding the dairy calf GIT microbiota has implications for ensuring proper health during early life and will aid in efforts to develop strategies for improving downstream production.

Genetic, social, and environmental conditions contribute to the development of depression, but the pathophysiological mechanisms are still unclear. Data accumulated in recent years provide significant evidence for a direct role of galanin (GAL). This study aimed to investigate the relation between SNPs in the galaninergic system and depressive symptoms in adolescents. A total of112 adolescents aged 10-18years participated in this study. The Children Depression Inventory (CDI) was used to evaluate depressive symptoms. The effects of rs948854 and rs4432027 SNPs, both located within the promoter region of the GAL gene, rs11665337 in the GALR1 receptor, and rs8836 in the GALR2 receptor on depressive symptoms were examined. The results indicated that 30.4% of the participants had depression. We found that girls were significantly more likely to be depressive than boys. Furthermore, rs948854 minor (G) allele was associated with depressive symptoms. Adolescents carrying the GG and AG genotype for the A/G (rs948854) SNP showed higher CDI scores than those carrying homozygous AA. The binomial logistic regression analysis revealed that adolescents carrying the GG genotype at SNP rs948854 had a higher likelihood of being depressive than adolescents carrying the AA or AG genotypes (P=0.033). Moreover, individuals whose mothers had a positive history for depression and who were sedentary were more likely to display depressive symptoms (P=0.013 and P=0.032, respectively). In conclusion, the SNP rs948854 in the GAL gene seems to be involved in the modulation of depressive state, especially in individuals with GG genotype.
