The optimal regimen of sheep brain rabies vaccine inactivation with UV rays has been developed. The immunogenic activity of 22 experimental lots of UV-inactivated rabies vaccine was found to be considerably higher than that of commercial Fermi vaccine. The antigenic activity of the inactivated vaccine in animals was also high.

Experimental lyophilized tissue culture rabies vaccine inactivated with gamma-rays was prepared from the Vnukovo-32 strain and showed a sufficiently high immunogenic activity. Complete inactivation of the virus and the immunogenic potency of the vaccine depended upon the dose of irradiation and temperature during irradiation. The virus lost infectivity at 18 degrees C when irradiated with 2.6 X 10(6) r and at --70 degrees C with 4.55 X 10(6) r. However, the immunogenic potency of the vaccine inactivated at minus temperature without thawing was significantly higher.

The production of virus-neutralizing antibodies in high titers has been found to occur in persons immunized with tissue-culture rabies vaccines, concentrated and non concentrated, intramuscularly in a dose of 1 ml in accordance with reduced immunization schedules.

The results of the study of concentrated, purified, UV-inactivated cell-culture rabies vaccine, obtained from strain Vnukovo-22, passage 33-40, in the primary culture of Syrian hamster kidney cells, demonstrated the pronounced antigenic potency of this vaccine: when introduced intramuscularly in 3-4 injections at certain intervals, it induced the production of virus-neutralizing antibodies in high titers. In tests on volunteers the vaccine proved to be nonreactogenic.

Animal experiments and trials on humans have shown that purified and concentrated chromatographic tissue-culture rabies vaccine is safe and essentially more potent than commercial tissue-culture rabies vaccine.

A study of 6 series of concentrated and purified -- by ultracentrifugation -- dry cultural rabies vaccine inactivated with ultra-voilet rays demonstrated that in case of a 31--316 increase of concentration immunogenicity of the preparation rose 3--13-fold. Purification of the vaccine took place with its concentration. Additional purification of the concentrated vaccine on aluminium oxide led to a partial loss of the viral protein and to reduction of immunogenicity.

Tests in volunteers showed that the reactogenicity of rabies vaccine prepared from sheep brain and inactivated with ultraviolet rays was not greater than the reactogenicity of Fermi vaccine. At the same time it was found to have a higher activity when injected both in the form of 5% suspension (in full and decreased doses) and with brain tissue content as low as 2.5%.

The possibility of the determination of rabies antibodies by the serological method has been demonstrated, involving the use of culture rabies virus, strain Vnukovo-32, concentrated and purified by high speed centrifugation, for the sensitization of anserine erythrocytes. The neutralization test in mice and the passive hemagglutination test showed a correlation in rabies antibody titers in the sera of animlas immunized with rabies vaccines. The advantages of the passive hemagglutination test consist in the rapidity of obtaining the results, in a simple and economic method of carrying out the test.

The results obtained in the study of the specific potency of rabies vaccine prepared from sheep brain tissue and inactivated by UV irradiation indicate that, even in the presence of the lowest immunogenicity index (0.5), 5-6 injections of the vaccine, made not daily, but at interval of 3 and 7 days, induced the production of antibodies in the titers not lower than those resulting from 14-20 daily injections of the same vaccine or Fermi vaccine. The preparation inactivated by UV irradiation should be introduced for therapy according to the shortened immunization schedule with intervals, taking into account the immunogenicity index.

In animal experiments the antigenic activity of adsorbed concentrated tissue-culture rabies vaccine was shown to be significantly higher in comparison with the nonadsorbed concentrated preparation when introduced in 2 intramuscular injections at an interval of 21 and 30 days, as well as in comparison with commercial tissue-culture vaccine when introduced subcutaneously in a 14-day course of daily injections.

The study of purified rabies vaccine (tissue culture origin), concentrated 100 fold, in animal experiments has shown that the preparation, if introduced intramuscularly in 2 or 3 injections, has high antigenic activity. The intramuscular injections of the vaccine at an interval of 21 days or at intervals of 21 and 30 days after the first injection has proved to be the optimum immunization schedule ensuring a sufficiently high accumulation of virus-neutralizing antibodies by days 30--45. To achieve higher antibody titers, the vaccine may be introduced intradermally after 2 indispensable intramuscular injections of the preparation.
