RESUMO

MicroRNAs (miRNAs) often display different expression in many cancers and other diseases in current research studies. miR-223 expression is upregulated in rheumatoid arthritis. Also, miR-223 expression has been demonstrated to be highly expressed in pancreatic cancer and gastric cancer in comparison with normal tissue. However, whether miR-223 displays different expression in ovarian cancer and what its underlying functions are in ovarian cancer have remained unclear. In this study, we demonstrated that miR-223-3p was upregulated in ovarian cancer tissue. Next, we explored the functional role of miR-223-3p in ovarian cancer using SKOV3 and OVCAR3 cell lines. Our results suggested that miR-223-3p mimic promoted ovarian cancer cell proliferation, migration, and invasion in vitro. However, miR-223-3p inhibitor displayed the opposite effects. In addition, we demonstrated that miR-223-3p mimic promoted tumor growth in vivo. Furthermore, we found SOX11 (sex determining region Y-box 11) was inversely expressed with miR-223-3p in ovarian cancer (OC) cell lines and tissue specimens. miR-223-3p mimic decreased SOX11 expression. Overexpressing SOX11 inhibited ovarian cancer cell proliferation and invasion, which indicated that miR-223-3p regulated OC cell proliferation and invasion through targeting SOX11 expression. In conclusion, the findings of the present study demonstrated that miR-223-3p could be a potential therapeutic for ovarian cancer.

A Bu3P-mediated cyclization reaction of 3-cinnamoyl-4-hydroxy-2H-chromen-2-ones though electrophilic addition of acyl chlorides towards the synthesis of highly functionalized furo[3,2-c]coumarins bearing a phosphorus ylide moiety is described. These unprecedented cyclization reaction proceeds under mild reaction conditions within short reaction times (1 min to 1 h), and can be further applied in the synthesis of alkenyl-substituted furo[3,2-c]coumarins by the treatment with carbonyl electrophiles under basic conditions.

Maintenance hemodialysis (MHD) is one of the most important renal replacement therapies for patients with end-stage renal disease. However, long-term and frequent treatment not only damages the physiological functions of patients but also leads to serious economic burdens and mental stress. This can easily cause a series of psychological disorders in patients, resulting in severe rejection and fear of MHD. To reduce patient resistance and improve the quality of life of MHD, this article built an intelligent nursing system based on big data and then used the constructed intelligent nursing system to research MHD. Through experiments, it has been found that compared to self-efficacy intervention, intelligent nursing systems can control the concurrent rate of MHD patients below 14 %, and self-efficacy intervention methods can control the concurrent rate of MHD patients above 13 %. Moreover, using intelligent nursing systems can improve the ability of MHD patients to self-care. At the same time, before the use of intelligent nursing systems, the nursing satisfaction of MHD patients also varied greatly, with the overall satisfaction rate after use being 70 % higher than before. Using intelligent nursing systems can improve the satisfaction of MHD patients with nursing outcomes.

OBJECTIVE: This study aimed to evaluate the effectiveness and safety of coils plus glue in slope embankment technology versus coils plus sclerosant in treating reflux-type pelvic venous disorders. METHODS: The analysis included patients diagnosed with reflux-type pelvic venous disorders who were treated with coils plus glue (CPG) or coils plus sclerosant (CPS) from 2019 to 2021. The inclusion criteria were non-cyclic pain lasting more than 6 months, atypical varicose, and transvaginal Doppler ultrasonography (TVDUS) and computed tomographic venography(CTV) confirming the diagnosis and excluding compression factors and other diseases. Propensity score matching was performed at a 1:1.1 ratio based on the following covariates: age, pregnancy, BMI, pretreatment VAS, dysmenorrhea, dyspareunia, urinary urgency, tenesmus, low back pain, vulvar varicosities, vaginal varicosities and lower limb varices. The pain was relieved by embolizing the target lesions with different embolic materials. The efficacy and safety of the different embolization materials were compared by visual analog scale (VAS) and TVDUS examinations at 1, 3, 6, 12, 24, and 36 months. RESULTS: From a total of 495 patients, 88 patients were selected from the CPG group and 77 patients from the CPS group by propensity score matching. The patients were followed up for 36 months. The preoperative VAS score of the Coils Plus Glue(CPG)group was 8 (range 6-8), and the Coils Plus Sclerosant(CPS) score was 8 (range 7-8),P=0.64. The post-embolization VAS score of the CPG group was 2.05±0.37, and the CPS score was 2.14±0.35(P=0.55). A total of 28 cases (16.9%) showed complications, most of which were transient pain after embolization. No serious complications such as coil embolization to the lungs occurred. In addition, the CPG group used fewer coils than the CPS group by using the Slope Embankment Technique(SET) . The mean coil length of the CPG group was 77.18±33.82 cm, and the CPS group was 105.29±71 cm(P=0.001). The CPG group had an average operation time of 44.49±5.72 min, while the CPS group took 43.45±4.18 min on average(P=0.19). The radiation dose of the CPG group was 398.40±76.16mGy, and the radiation dose of the CPS group was 388±44.23mGy(P=0.30). The median recurrence free survival(RFS) of the CPG group was 34.23 months (95% CI 33.2-35.2), and the median RFS of the CPS group was 30.39 months (95% CI 28.2-32.6), LogRank=0.018. CONCLUSIONS: Embolization therapy for refluxing PeVD was safe and effective, and proficient use of SET with coils plus glue increased efficacy and reduced complications.

Background: Neuroendocrine carcinoma of the cervix (NECC) is more prone to lymphatic infiltration, lymph node involvement, local recurrence, and distant metastasis. Using concurrent chemoradiotherapy (CCRT) with or without adjuvant chemotherapy as the standard treatment for locally advanced NECCs and CCRT for patients with early lesions confined to the cervix. However, the prognosis of NECC patients treated with definitive radiotherapy (RT) is unknown. Immune checkpoint inhibitors are a promising therapeutic strategy for locally advanced cervical cancer. Some reports suggest that the expression of PD-L1 in solid tumors correlates with prognosis. Aim: This study investigates prognostic factors for survival in patients with neuroendocrine cervical carcinoma (NECC) treated with definitive RT and the relationship between PD-L1 expression and prognosis in these patients. Methods: This retrospective study included 66 patients with histologically confirmed NECC who received RT with or without chemotherapy. From January 2015 to December 2020, patients received routine extended-field irradiation (EFI), and PD-L1 expression was assessed by immunohistochemistry. The most commonly used chemotherapy agents were etoposide-platinum and paclitaxel-platinum. Results: PD-L1 expression was positive in 17 of 45 (37.8%) patients. There were 52 cases of pure NECC and 14 cases of mixed carcinoma. Sixty stage IB-III patients received definitive RT. The 3- and 5-year progression-free survival (PFS) was 39.8% and 34.1%, and 3- and 5-year overall survival (OS) was 48.0% and 40.2%, respectively. There was no significant difference in 3 and 5-year PFS and 3 and 5-year OS between patients with pure and mixed carcinoma. Positive PD-L1 expression was associated with higher 3-year PFS in patients with mixed histology. Univariate analysis showed that lymph node metastasis (LNM) and the International Federation of Gynecology and Obstetrics stages predicted 3- and 5-year PFS in patients who received definitive RT. The median OS in patients receiving less than four cycles and at least four cycles of chemotherapy (CT) was 26.0 and 44.0 months, respectively (P = 0.038); moreover, 3- and 5-year PFS was 34.1% and 25.7% in the former and 46.4% and 40.4% in the latter. There were no significant differences in OS and PFS between pelvic irradiation and prophylactic EFI in patients treated with definitive RT. There were no significant differences in para-aortic failure rate after concurrent chemoradiotherapy between patients who underwent pelvic irradiation or prophylactic EFI (P = 0.147). Conclusion: In patients with mixed NECC, positive PD-L1 expression is correlated with higher 3-year PFS. Chemoradiotherapy was effective for NECCs. The LNM and stage predicted PFS. Four or more cycles of chemotherapy improve prognosis. Prophylactic EFI did not significantly improve PFS and OS. Relevance for Patients: This study is relevant to patients as it confirms that chemoradiotherapy is effective for both early and locally advanced NECC and that four or more cycles of chemotherapy improved prognosis. The regimen should be carefully evaluated to ensure that patients receive the most effective radiation therapy for the prophylactic of para-aortic LNM. Potential risk factors for the recurrence of radical radiotherapy should be fully understood to minimize these risks. This study observed that PD-L1 expression positive in patients with mixed NECC types is correlated with higher 3-year PFS.

A new and general strategy for highly functional furo[3,4-c]coumarins and related furyl coumarin derivatives has been developed, which is based on an extraordinarily facile intramolecular Wittig reaction, starting from α,ß-unsaturated ketones, tributylphosphine, and acyl chlorides. The phosphorus ylides were proposed to be the key intermediates for constructing the crucial furan ring, leading to a wide variety of substituted furyl coumarins in one step.

We observed previously that two carbohydrate epitopes, extended type 1 chain Le(a)-Le(a) and Le(b)-Le(a), are expressed strongly in human gastric or colorectal cancer and cell lines derived therefrom, but their expression in human normal colorectal cells is highly limited. A monoclonal antibody, termed GNX-8, was established through immunization of "KM mice" with colonic cancer cell line Colo205, and with purified Le(b)-Le(a) glycosphingolipid, followed by screening human IgG directed to this antigen. KM mice possess human chromosome fragments and are capable of producing human immunoglobulin. GNX-8 reacted specifically with extended type 1 chain epitope Le(b)-Le(a), bound to all five colonic cancer cell lines so far tested, and displayed strong complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC). The antigens defined by GNX-8, expressed in Colo205 cells, were: (i) glycosphingolipids with epitope Le(b)-Le(a), whose reactivity was abolished upon defucosylation; (ii) glycoproteins with molecular mass range from 32 to >175 kDa, which were depleted in cells cultured in the presence of benzyl-alpha-GalNAc, indicating that these epitopes are O-linked glycans.Immunohistological reactivity of GNX-8 at 1 mug/ml, applied on tissue sections from colorectal and various other types of cancer, was much stronger than that with various normal cells and tissues. GNX-8 reactivity with normal cells required a much higher concentration (150 mug/ml), and this reactivity was based on cross-reaction with non-extended, normal blood group Le(b) antigen. Growth of subcutaneous xenograft of human colonic cancer cells, Colo205 or DLD-1, in nude mice or SCID mice, was strongly inhibited by administration of GNX-8. These observations, taken together, indicate that antibody GNX-8, directed specifically to Le(b)-Le(a) antigen, provides a novel direction of immunotherapy for human colorectal cancer. This study provides a practical method for production of the antibodies against malachite green (MG) and its primary metabolite leucomalachite green (LMG). Two ELISA kits are constructed with the MG and LMG antibodies for detection of the residual MG and LMG in fish muscle and fishpond water. The detection limit is established at the level of 0.05 microg/L for both MG and LMG. Our ELISA kits show the advantages of good specificity, high sensitivity, and convenience in rapid screening of MG and LMG residues. The sample of fishpond water, without extraction or prior preparation, is directly assayed by the ELISA kit. More then 80 fish samples can be simultaneously tested in a kit. The toxic crystal violet and its metabolite leucocrystal violet of illegal use in aquaculture are detected by our prepared MG and LMG antibodies, whereas the antibodies do not cross-react with common antibiotics, sulfonamides, and benzene derivatives.

Radiation-induced lung injury (RILI) is a common complication of thoracic radiotherapy, but efficacious therapy for RILI is lacking. This study ascertained whether glycyrrhetinic acid (GA; a functional hydrolyzed product of glycyrrhizic acid, which is extracted from herb licorice) can protect against RILI and investigated its relationship to the transforming growth factor (TGF)-ß1/Smads signaling pathway. C57BL/6 mice were divided into four groups: a control group, a GA group and two irradiation (IR) groups. IR groups were exposed to a single fraction of X-rays (12 Gy) to the thorax and administered normal saline (IR + NS group) or GA (IR + GA group). Two days and 17 days after irradiation, histologic analyses were performed to assess the degree of lung injury, and the expression of TGF-ß1, Smad2, Smad3 and Smad7 was recorded. GA administration mitigated the histologic changes of lung injury 2 days and 17 days after irradiation. Protein and mRNA expression of TGF-ß1, Smad2 and Smad3, and the mRNA level of Smad7, in lung tissue were significantly elevated after irradiation. GA decreased expression of TGF-ß1, Smad2 and Smad3 in lung tissue, but did not increase Smad7 expression. GA can protect against early-stage RILI. This protective effect may be associated with inhibition of the TGF-ß1/Smads signaling pathway.

RESUMO

RAPD (Random Amolified Polymorphism DNA) was employed to detect molecular markers linked to sex in S. grosvenorii by BSA (Bulked Segregant Analysis). 18 RAPD markers linked to sex were selected in BSA by screening 90 primers. Only the marker amplified by S1431 was present in all 8 male individuals tested while absent from all 8 female individuals tested. It showed that S1431 was a male RAPD marker linked to sex.

RESUMO

On the medium MS added the right amount of 2,4-D and LFS (Angustmycin) and cultured under dark condition, the callus from stem segments of Panax notoginseng could induce a lot of embryoids. In 2-3 months, the ratio of embryoid formation reached about 90%. Then transplanted on MS + 2,4-D 1.5 mg/L + LFS 2 mg/L and cultured under light 20001x, near 30% embryoids could grow and develop as robust plantlets.

In order to explore quick, efficient inducement and proliferation of Panax notogingseng callus, the stem sections were taken as explants in the following 5 groups of medium: 1. MS + 2,4-D 2 mg/L (CK), 2. MS + 2,4-D 2 mg/L + N6-BA 2 mg/L, 3. MS + 2,4-D 2 mg/L + KT 2 mg/L, 4. MS + 2,4-D 2 mg/L + ZT 2 mg/L, 5. MS + 2,4-D 2 mg/L + LFS 2 mg/L. The results showed: 1. LFS (Lingfasu, a new kind of CTK) was able to promote the callus formation earlier 1-2 week than CK and to raise the rate of induced callus as high as 81%, higher 30% than that on other 4 mediums; 2. On medium 5, the fresh weight of callus increased 360.2% after being cultured 40 d, on other 4 mediums increased only 13.4%-21.8%. At the same time, 1 g callus cultured 40 d could obtain dry material 81.5 mg on medium 5, but on other 4 mediums only 21.5-25.9 mg; 3. The callus cultured on medium 5. continual generation to generation could live as long as more than 3 years, on other 4 mediums the callus hardened and aged quickly.

OBJECTIVE: To explore the mechanism of medicated thread moxibustion of Zhuang medicine (MTMZ) in treating perimenopausal period syndrome. METHODS: Thirty rabbits were randomly divided into a normal group, a model group, a medicated thread group, a no-medicated thread group and a sham operation group, 6 cases in each group. The model of perimenopausal period syndrome was established by ovariectomizing the ovary. The medicated thread group was treated with MTMZ at "Qizhou" acupoint (Extra), "Xiaguanyuan"(Extra), "Shenshu" (BL 23) and "Pishu" (BL 20), etc., once each day for 4 weeks. The no-medicated thread group was treated with no-medicated thread moxibustion at the same acupoints, and there is no treatment in the other groups. The changes of hormone level in each group before and after the treatment were observed. RESULTS: After ovariectomizing the ovary, the serum estradiol (E2) in the model group [(308.33 +/- 12.58) pmol/L], the medicated thread group [(304.96 +/- 13.85) pmol/L] and the no-medicated thread group [(303.43 +/- 10.57) pmol/L] were lower than that in the normal group [(478.09 +/- 12.23) pmol/L] and the sham operation group [(488.05 +/- 11.45) pmol/L] (all P < 0.01). After treatment, the E2 level in medicated thread group [(338.92 +/- 11.23) pmol/L] was higher than before (P < 0.01) and that in the model group [(300.53 +/- 13.68) pmol/L] and the no-medicated thread group [(309.74 +/- 13.59) pmol/L] (both P < 0.01), and the serum follicle stimulating hormone [FSH, (58.90 +/- 5.29) U/L] and luteinizing hormone [LH, (64.65 +/- 5.23) U/L] were lower than those in the model group [(65.41 +/- 5.19) U/L], [(71.85 +/- 5.30) U/L] (both P < 0.05). CONCLUSION: MTMZ can increase the serum E2 and reduce the serum FSH and LH in ovariectomized rabbits, and this may be one of the mechanisms of MTMZ for treatment of perimenopausal period syndrome.

A prototype chronoamperometric biosensor for the determination of total cholesterol was developed that consists of a homemade potentiostat and disposable strips immobilized with Fe(3)O(4), cholesterol oxidase (ChOx), and cholesterol esterase (ChE). The principle of sensing cholesterol is based on the detection of reduction signal of hydrogen peroxide generated in two enzymatic reactions. The co-immobilization of ChE and ChOx allows the sensor to detect both concentrations of esterified and free cholesterol. The effects of biosensor on catalyst, enzymes, applied potential, and buffer pH was investigated, and the operation conditions were optimized. The detection of cholesterol can be accomplished in one step, a 10 microL of sample was dropped onto the area of sensing strip and the reduction signal was obtained at an applied potential of -200 mV (vs. Ag/Ag(+)). The pre-reaction time was set at 15s before applying potential on the strip and the sampling time was 5s. The sensing device displays a linear response over the range of 100-400mg/dL (R(2)=0.999) for cholesteryl oleate. The coefficient variation was determined as 5.06% (N=20) for 100mg/dL cholesteryl oleate and the detection limit is 19.4 mg/dL (S/N=3). The probable interferences in bio-matrix were selected to test the selectivity and no significant response was observed in the biosensor.
