BACKGROUND: Muscle quality is a strong independent predictor of physical function. Body mass and fatness, muscle mass and cardiorespiratory fitness are known to influence muscle quality. OBJECTIVE: To identify the contributors of muscle quality in young and postmenopausal women and whether hormone replacement therapy (HT) could influence this relationship at the age of menopause. METHODS: Fifty-four postmenopausal women, 27 not on HT (PMW) and 27 on HT (PMW-HT), and 33 young women (YW) were evaluated for (1) body composition (body mass index, BMI), total fat mass (FM, %), appendicular lean mass (ALM, in kg and %), and appendicular skeletal mass index (ASMI = ALM/height2 in kg/m2); (2) absolute peak oxygen uptake (VO2peak, in ml/min) and relative peak oxygen uptake (VO2peakRel in ml/kg/min); and (3) absolute isometric knee extension strength (iKES in kg) and relative isometric knee extension strength (iKES/BMI and iKES/ALM). RESULTS: YW, PMW and PMW-HT had similar BMI (32.1 ± 10.2, 27.3 ± 4.7 and 26.7 ± 4 kg/m2) and FM (39.8 ± 10.0, 39.8 ± 7.3 and 39.9 ± 7.1%), respectively. Correlations were found between iKES/BMI index and FM (r = -0.52), ALM (r = 0.32) and VO2peak (r = 0.31). Regression analysis demonstrated that, in YW, total amount of variance in iKES/BMI was mostly explained by ALM (42%), whereas, in PMW and PMW-HT, it was cumulatively explained by FM along with VO2peakRel (34 and 46%, respectively). CONCLUSION: The main contributors of muscle quality differ between young and postmenopausal women and HT does not seem to influence this relationship.

There is very little information available about Culicoides species (Diptera: Ceratopogonidae) in the western Brazilian Amazon. However, studies of the fauna of this region are essential to knowledge of the species and potential vectors within it. Thus, the present study aims to evaluate the abundance, richness and composition of Culicoides species in rural areas in the state of Rondônia, Brazil. Culicoides specimens were collected in forest and pasture environments in the municipality of Porto Velho, using light traps. A total of 1708 individuals (1136 females and 572 males) belonging to 33 species were collected; 28 of these samples represent new records for the state of Rondônia and include the first record of Culicoides contubernalis in Brazil. Culicoides insignis was the most abundant species (86.1%). Species richness was greater in forest areas (32 species, 96.96%), whereas pastures presented the greatest number of Culicoides captured (n = 1540, 90.1%). This study shows that Culicoides populations differ between forest and pasture environments and indicates that the abundance of C. insignis is an important factor in epidemiological vigilance studies in the region.

Studying genetic diversity among a group of genotypes is important in genetic breeding because identifying hybrid combinations of greater heterotic effect also increases the chance of obtaining plants with favorable allele combinations in an intra-population selection program. The objective of this study was to compare different types of long and extra-long staple cotton and their genetic diversity in relation to the fiber traits and some agronomic traits in order to grant breeding programs. Diversity analysis among 29 cotton accessions based on qualitative and quantitative traits and joint including qualitative and quantitative traits was performed. Analysis based on qualitative and quantitative traits and joint met the accessions in three, two, and three groups, respectively. The cross between genotypes Giza 59 and Pima unknown was the most promising to generate segregating populations, comprising simultaneously resistance (based on molecular markers) to blue disease and bacterial blight, partial resistance to root-knot nematode, smaller size, in addition to good fiber characteristics. These populations can be used in recurrent selection programs as donors of alleles for development of long-staple cotton genotypes.

Cotton produces one of the most important textile fibers of the world and has great relevance in the world economy. It is an economically important crop in Brazil, which is the world's fifth largest producer. However, studies evaluating the genotype x environment (G x E) interactions in cotton are scarce in this country. Therefore, the goal of this study was to evaluate the G x E interactions in two important traits in cotton (fiber yield and fiber length) using the method proposed by Eberhart and Russell (simple linear regression) and reaction norm models (random regression). Eight trials with sixteen upland cotton genotypes, conducted in a randomized block design, were used. It was possible to identify a genotype with wide adaptability and stability for both traits. Reaction norm models have excellent theoretical and practical properties and led to more informative and accurate results than the method proposed by Eberhart and Russell and should, therefore, be preferred. Curves of genotypic values as a function of the environmental gradient, which predict the behavior of the genotypes along the environmental gradient, were generated. These curves make possible the recommendation to untested environmental levels.

Final cotton quality is of great importance, and it depends on intrinsic and extrinsic fiber characteristics. The objective of this study was to estimate general (GCA) and specific (SCA) combining abilities for technological fiber traits among six upland cotton genotypes and their fifteen hybrid combinations, as well as to determine the effective genetic effects in controlling the traits evaluated. In 2015, six cotton genotypes: FM 993, CNPA 04-2080, PSC 355, TAM B 139-17, IAC 26, and TAMCOT-CAMD-E and fifteen hybrid combinations were evaluated at the Experimental Station of Embrapa Algodão, located in Patos, PB, Brazil. The experimental design was a randomized block with three replications. Technological fiber traits evaluated were: length (mm); strength (gf/tex); fineness (Micronaire index); uniformity (%); short fiber index (%), and spinning index. The diallel analysis was carried out according to the methodology proposed by Griffing, using method II and model I. Significant differences were detected between the treatments and combining abilities (GCA and SCA), indicating the variability of the study material. There was a predominance of additive effects for the genetic control of all traits. TAM B 139-17 presented the best GCA estimates for all traits. The best combinations were: FM 993 x TAM B 139-17, CNPA 04-2080 x PSC 355, FM 993 x TAMCOT-CAMD-E, PSC 355 x TAM B 139-17, and TAM B 139-17 x TAMCOT-CAMD-E, by obtaining the best estimates of SCA, with one of the parents having favorable estimates for GCA.

Using commercial cultivars to compose crossing blocks in cotton is a promising strategy, because these materials have desirable agronomic and technological characteristics. The objective of this study was to evaluate the genetic diversity among 16 cotton cultivars cultivated in two environments in the State of Mato Grosso, the largest national producer, using agronomical and technological traits. There was significant effect to cultivars for all traits, while genotype x environment interaction was significant only for average boll weight, short fiber index, and maturity of fibers. Therefore, because of the presence of genotype x environment interaction for three traits, we chose to study genetic diversity among cotton cultivars separately in each environment and investigate the interaction impact on the diversity among genotype pairs. Based on agronomical and technological performance and genetic diversity among cultivars in both environments, the most promising cross involves FM 910 and LD CV 02. We also observed that lint percentage and average boll weight presented a higher discrimination capacity in both environments.

Seed cotton yield is a trait governed by multiple genes that cause changes in the performance of genotypes depending on the cultivation environment. Breeding programs examine the genotype x environment interaction (GE) using precise statistical methods, such as AMMI (additive main effects and multiplicative interaction) and GGE biplot (genotype main effects + genotype x environment interaction). The AMMI method combines the analysis of variance and principal components, to adjust the main effects (genotypes and environments) and the effects of GE interaction, respectively. The GGE biplot groups the genotype additive effect together with the multiplicative effect of the GE interaction, and submits both of these to the principal components analysis. The aim of this study was to investigate the association between the AMMI and GGE biplot methods and select cotton genotypes that simultaneously showed high productivity of seed cotton and stability in Mato Grosso environments. Trials were conducted with cotton cultivars in eight environments across Mato Grosso State in the 2008/2009 crop season. The experiment used a randomized block design with 16 genotypes and four replicates per genotype x environment combination. Data for seeds cotton productivity were analyzed by AMMI and GGE biplot methods. Both methods were concordant in the discrimination of environments and genotypes for phenotypic stability. The genotypes BRS ARAÇÁ and LD 05 CV had high seed cotton productivity and phenotypic stability, and could be grown in all environments across Mato Grosso State.

The harmonic mean of the relative performance of genotypic predicted value (HMRPGV) method has been used to measure the genotypic stability and adaptability of various crops. However, its use in cotton is still restricted. This study aimed to use mixed models to select cotton genotypes that simultaneously result in longer fiber length, higher fiber yield, and phenotypic stability in both of these traits. Eight trials with 16 cotton genotypes were conducted in the 2008/2009 harvest in Mato Grosso State. The experimental design was randomized complete blocks with four replicates of each of the 16 genotypes. In each trial, we evaluated fiber yield and fiber length. The genetic parameters were estimated using the restricted maximum likelihood/best linear unbiased predictor method. Joint selection considering, simultaneously, fiber length, fiber yield, stability, and adaptability is possible with the HMRPGV method. Our results suggested that genotypes CNPA MT 04 2080 and BRS CEDRO may be grown in environments similar to those tested here and may be predicted to result in greater fiber length, fiber yield, adaptability, and phenotypic stability. These genotypes may constitute a promising population base in breeding programs aimed at increasing these trait values.

With the changes in spinning technology, technological cotton traits, such as fiber length, fiber uniformity, fiber strength, fineness, fiber maturity, percentage of fibers, and short fiber index, are of great importance for selecting cotton genotypes. However, for accurate discrimination of genotypes, it is important that these traits are evaluated with the best possible accuracy. The aim of this study was to determine the number of measurements (repetitions) needed to accurately assess technological traits of cotton genotypes. Seven experiments were conducted in four Brazilian States (Ceará, Rio Grande do Norte, Goiás, and Mato Grosso do Sul). We used nine brown and two white colored fiber lines in a randomized block design with four replications. After verifying the assumptions of residual normality and homogeneity of variances, analysis of variance was performed to estimate the repeatability coefficient and calculating the number of repetitions. Trials with four replications were found to be sufficient to identify superior cotton genotypes for all measured traits except short fiber index with a selective accuracy >90% and at least 81% accuracy in predicting their actual value. These results allow more accurate and reliable results in future researches with evaluating technological traits in cotton genotypes.

To date, path analysis has been used with the aim of breeding different cultures. However, for cotton, there have been few studies using this analysis, and all of these have used fiber productivity as the primary dependent variable. Therefore, the aim of the present study was to identify agronomic and technological properties that can be used as criteria for direct and indirect phenotypes in selecting cotton genotypes with better fibers. We evaluated 16 upland cotton genotypes in eight trials conducted during the harvest 2008/2009 in the State of Mato Grosso, using a randomized block design with four replicates. The evaluated traits were: plant height, average boll weight, percentage of fiber, cotton seed yield, fiber length, uniformity of fiber, short fiber index, fiber strength, elongation, maturity of the fibers, micronaire, reflectance, and the degree of yellowing. Phenotypic correlations between the traits and cotton fiber yield (main dependent variable) were unfolded in direct and indirect effects through path analysis. Fiber strength, uniformity of fiber, and reflectance were found to influence fiber length, and therefore, these traits are recommended for both direct and indirect selection of cotton genotypes.

Brazil is among the five largest producers of cotton in the world, cultivating the species Gossypium hirsutum L. r. latifolium Hutch. The cultivars should have good fiber quality as well as yield. Genetic improvement of fiber traits requires the study of the genetic structure of the populations under improvement, leading to the identification of promising parent plants. To this end, it is important to acquire some information, such as estimates of genetic variance components and heritability coefficients, which will support the appropriate choice of the breeding strategy to be employed as well as enable the estimation of gains from selection. This study aimed to evaluate some agronomic characteristics, such as fiber quality and yield, estimating genetic parameters for the purpose of predicting earnings. Twelve cultivars of cotton, including four male progenitors (CNPA 01-42, BRS Verde, Glandless, and Okra leaf) and eight female progenitors (Delta opal, CNPA 7H, Aroeira, Antares, Sucupira, Facual, Precoce 3, and CNPA 8H), were used in performing crosses according to design I, proposed by Comstock and Robinson (1948). The experimental design was a randomized block with four replications. We observed genetic variability among all traits as well as higher efficiency of selection for the gains related to traits. Our results showed that the combined selection presented the highest genetic gains for all traits. For fiber length, the female/male selection and the combined selection resulted in the highest genetic gain.

Toxoplasma gondii is a parasite of great medical and veterinary importance that has worldwide distribution and causes toxoplasmosis. There are few treatments available for toxoplasmosis and the search for plant extracts and compounds with anti-Toxoplasma activity is of utmost importance for the discovery of new active drugs. The objective of this study was to investigate the action of a protein extract and a protease inhibitor enriched fraction from J. curcas seed cake on developing tachyzoites of T. gondii-infected Vero cells. The protein extract (JcCE) was obtained after solubilization of the J. curcas seed cake with 100 mM sodium borate buffer, pH 10, centrifugation and dialysis of the resulting supernatant with the extracting buffer. JcCE was used for the in vitro assays of anti-Toxoplasma activity at 0.01, 0.1, 0.5, 1.5, 3.0 and 5.0 mg/ml concentration for 24 h. The results showed that JcCE reduced the percentage of infection and the number of intracellular parasites, but had no effect on the morphology of Vero cells up to 3.0 mg/mL. The cysteine protease inhibitor enriched fraction, which was obtained after chromatography of JcCE on Sephadex G-75 and presented a unique protein band following SDS-PAGE, reduced both the number of T. gondii infected cells and intracellular parasites. These results suggest that both JcCE and the cysteine protease inhibitor enriched fraction interfere with the intracellular growth of T. gondii.

Culicoides biting midges (Diptera: Ceratopogonidae) are of great medical and veterinary importance because the haematophagous females of some species can transmit diseases to humans and animals. In order to determine the presence and seasonal abundance of the bluetongue virus (BTV) vector Culicoides insignis Lutz at domestic animal sheds in northeastern Brazil, insects were collected once a month between January and December 2010. Light traps were set from 18.00 to 06.00 hours at a pigsty, chicken coop and bovine corral. Culicoides insignis accounted for 81% of the 22,316 specimens collected. Other well-represented species were: Culicoides paucienfuscatus Barbosa (3246 individuals), Culicoides diabolicus Hoffman (308), Culicoides leopoldoi Ortiz (224) and Culicoides duartei Tavares and Luna Dias (221). The remainder accounted for 4% of the total sample. Culicoides insignis occurred mostly at the cattle corral, 98.2% in the rainy season. This study confirms the presence and close association of C. insignis with cattle in Maranhão state, northeastern Brazil and emphasizes the risk of bluetongue infections spreading in the area.

OBJECTIVE: To assess the adverse effects of the chronic use of zidovudine/lopinavir/ritonavir in a rat pregnancy model.Type of article: Original paper. DESIGN: A prospective experimental study. SETTING: Department of Obstetrics, São Paulo Federal University (UNIFESP). METHODS: 40 pregnant EPM-1 albino rats were randomly allocated into four groups of 10 animals each: control (Ctrl) group (untreated) and three experimental groups (Exp1, Exp2 and Exp3), which received zidovudine/lopinavir/ritonavir in the corresponding doses of 10/13.3/3.3; 30/39.9/9.9 and 90/119.7/29.7 mg/Kg/day from the first up to the 20th day of pregnancy, respectively. The rats were treated by gavage daily. Body weights were recorded on days 0, 7, 14 and 20. At term, the rats were sacrificed and the implantation sites, number of live and dead fetuses and placentas, resorptions and fetal and placental weights were recorded. The fetuses were evaluated for external abnormalities under a stereomicroscope. The chi-square test was used to compare death rates between groups. RESULTS: Weight gain during pregnancy no showed significant differences between groups. Average weight gains between the 7th and 20th day were 45.70 ± 5.27 g for Ctrl; 48.49 ± 3.64 g for Exp1; 45.39 ± 6.22 g for Exp2 and 44.19 ± 6.78 g for Exp3. However, the percentage weight gain in the 7th was lower in groups Exp2 and Exp3 and in the 14th in the Group Exp2. All other parameters assessed did not differ significantly between groups. Exp2 and Exp3 in relation of the others. CONCLUSIONS: The chronic exposure of pregnant rats to high doses of zidovudine/lopinavir/ritonavir in association resulted in a significant reduction in maternal body weight gain but was not associated with significant adverse fetal parameters.

PURPOSE: To evaluate the morphological aspects in rats subjected to an association of the antiretroviral drugs zidovudine/lopinavir/ritonavir in different doses administered throughout the gestational period. MATERIALS AND METHODS: Forty pregnant rats were randomly allocated into four groups: control (Ctrl) and experimental (Exp1, Exp2, and Exp3), which received zidovudine/lopinavir/ritonavir in the doses of 10/13.3/3.3, 30/39.9/9.9, and 90/119.7/29.7 mg/kg per day from the first to the 20th day of pregnancy, respectively. At term, the animals were euthanized and maternal and fetal organ samples were removed for morphological analysis. RESULTS: No major changes were identified in the group treated with the lowest dosing compared with the control. In group Exp2, the authors found hepatocytes with eosinophilic cytoplasm, pyknotic nuclei, and vasodilation. The proximal convoluted tubules of maternal kidneys showed eosinophilic areas and hyperchromatic nuclei, as well as signs of vasodilation. In the group treated with the highest dose (Exp3); the morphological changes in the maternal kidneys and livers were similar and more pronounced than those found in Exp2. The maternal pancreas of groups Exp2 and Exp3 evidenced moderate and progressive signs of tissue damage. The morphological features of all fetal livers, kidneys, and pancreases were normal. CONCLUSION: High doses of zidovudine/lopinavir/ritonavir association during the entire rat pregnancy period can cause definite morphological changes in maternal liver, kidneys, and pancreas. On the other hand, the corresponding fetal organs were not affected.

METHODS: Three groups (n = 10 each) of adult pregnant rats were treated with an oral solution of HAAR (Exp 1 = 10/5/20 mg/kg b.w.; Exp 2 = 30/15/60 mg/kg b.w.; Exp 3 = 90/45/180 mg/kg b.w.) from day "0" up to the 20th day of pregnancy. A fourth group served as a control. At term (20th day) the rats were killed under deep anesthesia and the number of implantations, resorptions, living fetuses, placentae and intrauterine deaths were recorded. RESULTS: The highest HAAR doses caused lower maternal weight gain, lower litter weights, and lower placental weights compared to the control group. CONCLUSIONS: HAAR during the entire period of rat pregnancy can reduce maternal body weight gain and lower term placental weight.

Inhibitory glycine receptors (GlyRs) are mainly expressed in the spinal cord and in the midbrain, where they control motor and sensory pathways. We describe here a fast potentiation of GlyR by intracellular Ca2+. This phenomenon was observed in rat spinal cord neurons and in transfected human cell lines. Potentiation develops in <100 ms, is proportional to Ca2+ influx, and is characterized by an increase in GlyR apparent affinity for glycine. Phosphorylation and G protein pathways appear not to be involved in the potentiation mechanism. Single-channel recordings in cell-attached and excised patches, as well as whole-cell data suggest the presence of a diffusible cytoplasmic factor that modulates the GlyR channel gating properties. Ca2+-induced potentiation may be important for rapid modulation of glycinergic synapses.

Castor cake is a by-product of the extraction of oil from from seeds of castor plants (Ricinus communis). This by-product contains high levels of proteins, but a toxic protein, ricin, limits its use as an animal feed. Ricin can be efficiently inactivated by treatment with calcium oxide (CaO), which can be evaluated by a cytotoxicity assay using LLC-MK2 cells. The mechanism by which the CaO treatment inactivates ricin, however, is unclear. We report the structural changes responsible for ricin inactivation. Purified ricin was treated with 0.6% CaO and then analyzed by mass spectrometry. This treatment degraded the ricin at preferential sites. The aqueous CaO solution had a pH >12, which preferentially cleaved asparagine residues, followed by glutamine, serine and glycine residues. The alkaline pH affected the tertiary structure of the ricin, cleaving its polypeptide chains and thereby eliminating its cytotoxic activity.

The effects of increasing levels of solvent-extracted palm kernel meal (SPKM) and corn distillers dried grains (CDG) in corn silage-based diets on feed intake and milk production were examined in 2 experiments. In Experiment 1, 20 Holstein cows averaging 100 d in milk (DIM) (SD = 61.5) at the start of the experiment were utilized in an 11-wk randomized complete block design with 4 treatments in 5 blocks to study effects of increasing levels of SPKM in the diet. During a 3-wk preliminary period, cows were fed a standard diet. At the end of the preliminary period, cows were blocked by 4% fat-corrected milk yield, parity number (primiparous and multiparous), and DIM, and were assigned randomly to 1 of 4 experimental diets. The total mixed ration (TMR) consisted of (dry matter basis) 40% corn silage, 5% coarsely chopped wheat straw, and 55% concentrate. The increasing dietary levels of SPKM were achieved by replacing protein sources and citrus pulp with SPKM and urea (0, 5, 10, and 15% SPKM and 0.06, 0.22, 0.38, and 0.55% urea for SPKM0, SPKM5, SPKM10, and SPKM15, respectively). In Experiment 2, 18 Holstein cows averaging 93 DIM (SD = 49.1) at the start of the experiment were utilized in an 11-wk randomized complete block design with 3 treatments in 6 blocks to study effects of increasing levels of CDG in the diet. The preliminary period lasted for 2 wk. Assignment of cows to treatments was the same as in Experiment 1. The TMR consisted of (dry matter basis) 40% corn silage, 5% coarsely chopped wheat straw, and 55% concentrate. The increasing dietary levels of CDG were achieved by replacing soybean meal and citrus pulp with CDG and urea (0, 7, and 14% CDG and 0, 0.22, and 0.49% urea for CDG0, CDG7, and CDG14, respectively). There were no significant treatment effects on dry matter intake, milk yield, or milk composition in Experiment 1. Inclusion of SPKM tended to increase protein and lactose contents of milk. The SPKM0 diet promoted body weight loss. There were no treatment effects in Experiment 2, except for milk protein content, which was decreased by CDG. Plasma Lys concentration tended to be affected by SPKM and CDG inclusions. Plasma concentrations of 3-methylhistidine and Leu seemed to be related to body protein degradation/synthesis. The feeding of SPKM up to 15% in the diet decreased feed costs without detrimental effects on productive responses and tended to increase milk protein content. The inclusion of CDG in diets based on corn silage and corn byproducts might decrease milk protein content due to an unbalanced supply of AA, particularly Lys.

BACKGROUND: Bleeding remains a major limitation of standard anticoagulant drugs that target the extrinsic and common coagulation pathways. Recently, intrinsic coagulation factors are increasingly being investigated as alternative targets for developing anticoagulant drugs with lower bleeding risk. OBJECTIVES: Goals were to (i) identify novel anticoagulants selectively targeting intrinsic coagulation pathway and (ii) characterize and further improve the properties of the identified anticoagulants. METHODS AND RESULTS: We have isolated and sequenced a specific factor XIa (FXIa) inhibitor, henceforth named Fasxiator, from the venom of the banded krait snake, Bungarus fasciatus. It is a Kunitz-type protease inhibitor that prolonged activated partial thromboplastin time without significant effects on prothrombin time. Fasxiator was recombinantly expressed (rFasxiator), purified, and characterized to be a slow-type inhibitor of FXIa that exerts its anticoagulant activities (doubled activated partial thromboplastin time at ~ 3 µmol L(-1) ) by selectively inhibiting human FXIa in in vitro assays. A series of mutants were subsequently generated to improve the potency and selectivity of recombinant rFasxiator. rFasxiatorN17R,L19E showed the best balance between potency (IC50 ~ 1 nmol L(-1) ) and selectivity (> 100 times). rFasxiatorN17R,L19E is a competitive slow-type inhibitor of FXIa (Ki  = 0.86 nmol L(-1) ), possesses anticoagulant activity that is ~ 10 times stronger in human plasma than in murine plasma, and prolonged the occlusion time of mice carotid artery in FeCl3 -induced thrombosis models. CONCLUSION: We have isolated an exogenous FXIa specific inhibitor, engineered it to improve its potency by ~ 1000 times and demonstrated its in vitro and in vivo efficacy. These proof-of-principle data supported the further development of Fasxiator as a novel anticoagulant candidate.
