RESUMO

Serum from partially hepatectomized rats promoted DNA synthesis in primary adult rat hepatocyte cultures. If the rats had been exposed to sub-lethal gamma-irradiation immediately following operation or if their serum, collected at 3 h, was exposed to irradiation in vitro, the growth-promoting activity was destroyed. Prostaglandin E2 also stimulated DNA synthesis in the cultures; if PGE2 was irradiated in serum from intact or partially hepatectomized rats its growth-promoting activity was markedly diminished.

RESUMO

Rats given an LD50 dose of Be2+ showed reduced activities of ornithine decarboxylase and tyrosine aminotransferase in liver in response to dexamethasone induction. Control fed animals showed 'superinduction'. Be2+ also inhibited the uptake of [3H]orotic acid into rapidly labelled RNA of ribonucleoprotein particles extracted from liver nuclei in isomolar solutions at pH 8.0. Consistent with inhibition of cytoplasmic protein kinase reported previously (Kaser et al., 1980), the uptake of [32P]Pi into proteins in the ribonucleoprotein particles was also diminished.

RESUMO

Selective substrates and inhibitors have been used to measure kinases phosphorylating endogenous proteins in rat liver nuclei during growth and regeneration after partial hepatectomy. Peaks in activity were found at 5, 22, and 29 hours after partial hepatectomy. Administration of alpha 1 and beta adrenergic blockers suggested that the Be2+ sensitive and cyclic AMP-dependent protein kinases were interdependently regulated by Ca2+ and cyclic AMP.

Initiation sites were enumerated in rat liver nucleosomes with Echerichia coli RNA polymerase. One site was present in approx. 3.5 mononucleosomes and in a 1200-base-pair length of polynucleosomes. S-phase nuclei or normal nuclei phosphorylated in vitro showed increased numbers of sites; the elongation rates were unchanged.

1. Differences in washing procedures and conditions of dialysis affected the type of histone f3 obtained from rat liver and thymus nuclei. 2. Sulphur-rich preparations of f3 contained phosphate and had SH+SS: phosphorus ratio 1:2. 3. Sulphur-rich f3 from livers of [(14)C]adenine-labelled rats gave counts indicating a phosphorus: adenine ratio 1:1. 4. Evidence was presented that histones f3 and f1 exist in nuclei as families of proteins, differing with respect to their sulphur or phosphorus content or both. 5. The significance of the different states of phosphorylation and oxidation of histones f1 and f3 was discussed.

1. The incorporation of (14)C-labelled amino acids into acid-extractable proteins from rat-liver and -thymus nuclei confirmed the existence of a protein component with a higher uptake than that into the major histone components. 2. This rapidly labelled component appeared to contain the thiol groups detectable in the acid extracts. 3. Histone f1 contained 1mol. of serine phosphate/mol. of mol.wt. of 42000-43000. 4. Phosphate was present in other components of the 50mm-hydrochloric acid extract from liver and thymus nuclei, and was probably associated with the thiol-containing component. 5. The difference in amino acid uptake into the histones of diffuse and dense chromatin was confirmed. Dense chromatin was found to have a higher proportion of disulphide than did diffuse chromatin.

1. The proportion of thiol groups in the total thiol+disulphide of histone extracts from fertilized eggs from Echinus and Psammechinus was increased during periods of structural alterations in the nucleus. 2. The probable start of DNA synthesis in the fertilized eggs coincided with periods of maximum thiol content. 3. Histone extracts from rat liver and regenerating liver were predominantly in the thiol form and no significant variations could be detected during the first 30hr. after partial hepatectomy. 4. An assay system was developed to follow the phosphorylation believed to be associated with the arginine-rich histone F3. Phosphorylation increased by 50% at 1 and 2hr. after partial hepatectomy. The phosphate content also increased during the period of DNA synthesis. 5. The increased phosphorylation found 1hr. after partial hepatectomy was not prevented by actinomycin or prior irradiation. 6. The phosphate content of histone F1 was very high in livers from foetal rats and declined in neonatal rats similarly to the decline in DNA synthesis.

1. The capacity of the histone-DNA complex to act as a template for RNA synthesis is increased by phosphorylation of histone F1. 2. In regenerating liver, DNA synthesis is preceded by phosphorylation of histone F1. 3. Exposure to ionizing radiation in vivo decreases and delays the phosphorylation of histone F1 in regenerating liver, and decreases it in the hyperplastic kidney. 4. Histone F1 is phosphorylated to a greater extent in dividing than in resting tissues.

1. In regenerating rat liver the phosphate content of the lysine-rich histone F1, but not that of the more arginine-rich histone F3-1, increases during the period of DNA synthesis. 2. The phosphorylation of histone F1 in this ;S period' is decreased by gamma-irradiation, but, like phosphate uptake into DNA, is affected to an even greater extent if the irradiation is given in the presynthetic period. 3. Histones from three species of sea-urchin eggs show similarities to the F2 and F3 groups of histones from mammalian thymus gland. 4. The proportion of thiol to total thiol plus disulphide in acid extracts from sea-urchin eggs varies from less than 20% in mature unfertilized eggs to 59% just before cleavage. 5. The phosphorylated forms of histones F1 and F3 are less effective in decreasing DNA synthesis by DNA polymerase than the non-phosphorylated forms. 6. Oxidation of thiol groups on histone F3-1 does not affect its capacity to decrease DNA synthesis in vitro.

Nuclei were isolated from Echinus eggs through the first cleavage cycle by modification of existing techniques. When these nuclei were extracted with 2 M NaCl and the supernatant diluted to 0.15 M, large amounts of non-histone proteins remained in solution. The precipitated nucleoprotein contained expected amounts of DNA and a protein analogous to mammalian histone H1. Extrachromosomal histone H1 was eliminated by the modified isolation procedure. Amounts of nuclear proteins soluble in 0.15 M NaCl reached a peak in G2. Histones and non-histone proteins were phosphorylated postfertilization, in early prophase and in telophase.

Micrococcal-nuclease digestion of rat liver nuclei selectively released mononucleosomes associated with ADP-ribosylated [Caplan, Ord & Stocken (1978) Biochem. J.174, 475-483] histone H1. Two classes of mononucleosome were detected, those that leaked out during digestion and those that were subsequently released by 5mm-sodium phosphate buffer (pH6.8)/0.2mm-NaEDTA. The former, from which histone H1 had been dissociated, contained 140-base-pair-length DNA and core histones;the latter contained core particles and mononucleosomes with histone H1 and 200-base-pair-length DNA. When normal liver nuclei were phosphorylated with [gamma-(32)P]ATP, dissociated histone H1, which could be separated from core particles with Sephadex G-200, showed (32)P uptake. (32)P uptake into histones H2A and poly(ADP-ribosyl)ated H3 was appreciable in core particles, but was less evident in nucleosomes still containing histone H1. When [(3)H]-thymidine was given to partially hepatectomized rats in S-phase, 5-10min pulses in animals of over 300g body wt. showed the presence of high-specific-radioactivity DNA in released core particles and mononucleosomes compared with DNA retained in the nuclear pellets. Mononucleosomes from rat livers in S-phase with new, [(3)H]lysine-containing histones, had higher (32)P incorporation in histones H1 and their core histones, than for di- or tri-nucleosomes. Thermal-denaturation properties of control and phosphorylated mononucleosomes and core particles were very similar; removal of histone H1 and non-histone chromosomal proteins in 0.5m-NaCl markedly increased the proportion of DNA ;melting' below 70 degrees C.

1. ADP-ribose is found in rat liver nuclei covalently bound to histone F1, to a non-histone protein, and to a small peptide. 2. A single unit of ADP-ribose, covalently bound to phosphoserine, was isolated from an enzymic hydrolysate of histone F1. ADP-ribose-bearing peptides were isolated from a tryptic digest of the histone. 3. It is proposed that the 1'-hydroxyl group of ADP-ribose is linked to the phosphate group of phosphoserine in histone F1. 4. The incorporation of 32P into ADP-ribose on histone F1 a parallels the DNA content through the cell cycle. An increased incorporation of the nucleotide into the other derivatives is observed during S phase. 5. It is suggested that the ADP-ribose derivative of histone F1 has a role in maintaining the G0 state and that one or both of the other derivatives is concerned with control of DNA synthesis.

1. DNA synthesis in Echinus esculentus eggs kept at 10 degrees C takes place just after fusion, 0.75-1.5h after fertilization, and at telophase at about 2.67-3.33h after fertilization. 2. An increase in the thiol/thiol+disulphide ratio in acid extracts from washed nuclear fractions of the eggs is found at fusion, at early stages of mitosis and at telophase. When DNA is being synthesized, the relative amount of thiol in the extracts increases. 3. There are at least five thiol-containing histones in the acid extract together with a diffusible thiol peptide containing methyl-lysine and 3-methylhistidine and a thiol-containing acidic protein.

1. Two minor protein fractions (B and C) were separated from histone F1 by chromatography on DEAE-cellulose. Fraction B was acidic. 2. Uptake of (32)P in vivo into histone F1 but not into fractions B and C was stimulated by partial hepatectomy. 3. It is suggested that partial hepatectomy causes an increase in the number of histone F1 molecules phosphorylated.

Four subfractions of histone F1 from rat thymus were obtained by preparative polyacrylamide-gel electrophoresis. These subfractions are closely related in primary structure but show marked differences in radioactivity when derived from rat thymus nuclei labelled in vitro with [(32)P]phosphate.

RESUMO

A non-histone protein was obtained by extraction of nuclei derived from rat liver or thymus or ascites-hepatoma cells with 5% (w/v) HClO(4). Separation from histone F1 was achieved by chromatography on DEAE-cellulose. The purified component P1 was characterized and the formation of complexes with histone F1 and polylysine was studied.
