Abstract This study aimed to identify and characterize the antimicrobial susceptibility profile of bacteria found in primary endodontic infections in the teeth of patients treated at the Dental Clinic of the University of Ribeirão Preto, São Paulo, Brazil. From September to December 2019, samples were obtained from 21 patients with primary endodontic infections. The collections were carried out in triplicate using paper cones placed close to the total length of the root canal. Bacterial isolation was performed in Brain Heart Infusion agar, Blood agar, and other selective culture media cultured at 37°C for up to 48 h under aerobiosis and microaerophilic conditions. The bacterial species were identified using the Vitek 2 automated system. The disk diffusion method on agar Müeller-Hinton was used to assess antimicrobial susceptibility with the recommended antimicrobials for each identified bacterial species. A total of 49 antibiotics were evaluated. Fifteen of the 21 samples collected showed bacterial growth, and 17 bacterial isolates were found. There were 10 different bacterial species identified: Enterococcus faecalis (four isolates), Streptococcus mitis/oralis (three isolates), Streptococcus anginosus (three isolates) being the most common, followed by Staphylococcus epidermidis, Enterococcus faecium, Streptococcus constellatus, Streptococcus alactolyticus, Enterobacter cloacae, Klebsiella variicola, and Providencia rettgeri (one isolate of each species). The analysis demonstrated significant susceptibility to most of the tested antibiotics. However, some Enterococcus isolates resisted the antibiotic's erythromycin, ciprofloxacin, and tetracycline. A Staphylococcus epidermidis isolate was characterized as multidrug-resistant. Five Streptococcus isolates were non-susceptible to all antibiotics tested.

Abstract The survival of endodontically treated teeth depends on the remaining tooth structure. The aim of this study was to evaluate the impact of different access cavities on root canal preparation, restorative protocol, and fracture resistance of endodontically treated teeth. Fifty-six mandibular molars were divided into control (n=8) and experimental (n=16) groups according to access cavity: Traditional, Conservative, and Truss; and redistributed (n=8) according to instrumentation protocols: Reciproc Blue and R-motion. After, teeth were scanned in micro-CT and then filled and redistributed according to composite resin restoration (n=8): Filtek One BulkFill and Filtek Z350. A new micro-CT scan was performed to analyze the restorative material. Then, samples were submitted to fracture resistance testing and the failure pattern was determined. Data were analyzed using paired T-test, ANOVA, Tukey, and chi-square tests (α=0.05). In Truss, R-Motion promoted less transportation in different thirds of root canals. Higher percentages of voids (5.05%) and filling material (11.7%) were observed in Truss. Fracture resistance values were higher for the control group, followed by Truss, Conservative, and Traditional. The predominant failure pattern was type-II. In Truss, reciprocating instruments with smaller taper showed less canal transportation. Also, Truss provided higher values of fracture resistance, although it presented a higher percentage of voids and remaining filling material. Thus, in Truss, reciprocating files with smaller taper showed less canal transportation, and these cavities provided higher values of fracture resistance, although it presented a higher percentage of voids and remaining filling material.

Abstract The aim of this study was to evaluate the influence of different instruments and mechanisms of agitation of the irrigating solution on the penetration by confocal laser scanning microscopy (CLSM), and the bond strength (BS) of the filling material to the dentin by push-out test. Human premolars were distributed in two experimental groups (n=30), according to the instrumentation system: ProTaper Next (PN) or XP-endo Shaper (XS) and three irrigation protocol (n=10): NaviTip (Nv), XP-endo Finisher (XF), passive ultrasonic irrigation/ XP-endo Finisher (PUIXF). The 2.5% NaOCl and 17% EDTA were the irrigating solutions used. The filling was performed using a lateral condensation technique, with AH Plus added to 0.1% rodhamine B. The roots were sectioned, obtaining 2 slices for each third of the root. The first slice was submitted to the sealer penetration analysis and the push-out test. The second slice was submitted to qualitative analysis (SEM) of the union interface. For the penetration (%) Tukey showed that PNXF (82.38±13.48), PNPUIXF (82.48± 12.02), XSXF (82.24±11.28) and XSPUIXP (86.23±10.45) presented the highest values, different (p<0.05) from PNNv (68.29±15.12) and XSNv (71.41±16.50). The BS (MPa) test evidenced that in the cervical third, PNPF (4.92±1.04) and XPSPF (5.03±0.70) obtained the highest values differing from the others groups (p<0.05). CSLM and SEM showed greater penetration of the sealer on the entire length of the root canal when the irrigation solution was stirred. In conclusion that the association PUI and XP-endo Finisher favored the penetration and bond strength of the filling material to the root dentin.
Resumo O objetivo neste estudo foi avaliar a influência de diferentes instrumentos e mecanismos de agitação da solução irrigadora na penetração e resistência de união (RU) do AH Plus à dentina. Pré molares humanos foram distribuídos em dois grupos experimentais (n=30), de acordo com o sistema de instrumentação: ProTaper Next (PN) ou XP-endo Shaper (XS) e de acordo com três protocolos de irrigação (n=10): NaviTip (Nv), XP-endo Finisher (XF), passive ultrasonic irrigation/ XP-endo Finisher (PUIXF). A irrigação do canal radicular foi realizada com NaOCl 2,5% e EDTA 17%. A obturação foi realizada por meio da técnica da condensação lateral, com AH Plus acrescido de rodhamina B 0,1%. As raízes foram seccionadas, obtendo-se 2 slices de 2,0 mm para cada terço radicular. O primeiro slice foi submetido a análise da penetração do cimento e ao teste de push-out e o segundo slice submetido a MEV para análise da interface de união. Para penetração (%) Tukey evidenciou que PNXF (82,38±13,48), PNPUIXF (82,48±12,02), XSXF (82,24±11,28) e XSPUIXF (86,23±10,45) apresentaram os maiores valores, diferentes (p<0,05) de PNC (68,29±15,12) e XPSC (71,41±16,50). Na RU (MPa) foi evidenciado que PNPF (4,92±1,04) e XPSPF (5,03±0,70), no terço cervical, obtiveram os maiores valores diferentes dos demais (p<0,05). MCVL e MEV evidenciaram maior penetração do cimento em toda extensão da parede do canal radicular inclusive nas regiões polares, quando foi realizada a agitação da solução irrigadora. Concluiu-se que a associação PUI e XP-endo Finisher favoreceu a penetração e resistência de união do material obturador à dentina radicular.

Abstract This study evaluated by the efficacy of rotary and reciprocating systems to remove filling material by micro-CT. Sixty human canines were instrumented up to F3 by ProTaper Universal system and filled with AH Plus/ gutta-percha. Specimens were submitted to 1,200 thermal cycles, scanned by microCT SkyScan 1176 and randomly distributed in groups according to the filling material removal protocol (n = 10): ProTaper Universal, ProTaper Universal/xylol, ProTaper Next, ProTaper Next/xylol, Reciproc and Reciproc/xylol. After filling removal, specimens were analyzed by microCT to separately verify the presence of gutta-percha and sealer remnants. The data (mm3) were analyzed by two way ANOVA and Tukey´s test (5%). For the remaining gutta-percha, the use of xylol provided the highest values for Reciproc (7.60 ± 0.73), which was statistically different from the other groups (p < 0.05) that presented lower values and were statistically similar to each other (p > 0.05). For the remaining sealer, xylol provided the highest values (21.25 ± 6.94) different (p < 0.05) from the use of instruments alone (11.47 ± 9.45). ProTaper Next presented the lowest values (8.16 ± 1.37) for the remaining sealer, different (p < 0.05) from that of Reciproc (24.67 ± 6.32). The qualitative analysis revealed that all groups presented some remaining root filling material, with the highest volumes presented by Reciproc and Reciproc/xylol. In conclusion the rotary systems provided greater removal of filling material regardless of the use of xylol. The use of xylol negatively interfered with the action of the reciprocating system in the removal of the gutta-percha and sealer.

Abstract Green chemistry has been applied in different areas due to the growing demands for renewable processes and one of them is nanotechnology. The aim of this study was to characterize a formulation containing silver nanoparticles (AgNPs) produced by a green synthesis and to evaluate its antimicrobial activity. The formulation will be used as an intracanal dressing exploiting the AgNPs' antimicrobial properties, which are crucial to prevent infections and bacterial reinfections that can compromise endodontic treatments. In the green synthesis, silver nitrate was employed as the precursor salt, maltose as a reducing agent, and gelatin as a stabilizing agent. The formulation was prepared mixing 50 % of a liquid containing the AgNPs and 50 % of hydroxyethylcellulose gel at 1.5 % with proper evaluation of the process inherent parameters. Techniques such as molecular absorption spectrometry and dynamic light scattering were used in characterization step. The antimicrobial activity of the AgNPs against Escherichia coli ATCC 25922, Enterococcus faecalis NCTC 775, Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 25923 and Streptococcus mutans ATCC 25175 was verified according to National Comittee for Clinical Laboratory Standards (NCCLS) by determining minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). The obtained results indicated the formulation containing AgNPs produced by a green synthesis was properly characterized by the selected techniques. Furthermore, the formulation assessment proved that it is suitable for the proposal as well as it has potential to be used as an intracanal dressing since presented antimicrobial activity against all bacterial strains evaluated.
Resumo A química verde tem sido aplicada em diferentes áreas devido à crescente demanda por processos renováveis e uma delas é a nanotecnologia. O objetivo deste estudo foi caracterizar uma formulação contendo nanopartículas de prata (AgNPs) produzidas por meio de síntese verde e avaliar sua atividade antimicrobiana. A formulação será usada como curativo intracanal explorando as propriedades antimicrobianas das AgNPs que são cruciais para prevenir infecções e reinfecções bacterianas que podem comprometer os tratamentos endodônticos. Na síntese verde, nitrato de prata foi empregado como sal precursor, maltose como agente redutor e gelatina como agente estabilizador. A formulação foi preparada misturando-se 50% do líquido contendo as AgNPs e 50% de gel de hidroxietilcelulose a 1,5% com avaliação adequada dos parâmetros inerentes ao processo. Técnicas como espectrometria de absorção molecular e espalhamento dinâmico de luz foram usadas na etapa de caracterização. A atividade antimicrobiana das AgNPs contra Escherichia coli ATCC 25922, Enterococcus faecalis NCTC 775, Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 25923 e Streptococcus mutans ATCC 25175 foi verificada de acordo com o National Comittee for Clinical Laboratory Standards (NCCLS), determinando-se a concentração inibitória mínima (MIC) e a concentração bactericida mínima (MBC). Os resultados obtidos indicaram que a formulação contendo AgNPs produzidas por meio de síntese verde foi devidamente caracterizada pelas técnicas selecionadas. Além disso, a avaliação da formulação provou que ela é adequada para a proposta, bem como tem potencial para ser utilizada como curativo intracanal já que apresentou atividade antimicrobiana contra todas as cepas bacterianas avaliadas.

Abstract The aim of this study was to verify the effect of the implant volume loss, vertical misfit between abutment and prosthetic platform, prosthetic screw loosening torque, and screw stress distribution in titanium and zirconia abutments. Ten CAD/CAM system custom abutments of each material were milled and attached to the titanium implants. The implant volume loss was evaluated by microtomography, the vertical misfit with optical microscopy, and digital torque wrench measured the prosthetic screw loosening. All experimental analyses were performed before and after mechanical cycle (1,000,000 cycles, 100 N/2 Hz). Virtual models of the structures were created for finite element analysis, and the stress on the screw obtained with von Mises procedure. Data were analyzed using an independent t-test, two-way ANOVA for repeated measures, and Tukey's HSD test (a=0.05). There was no significant difference in the implant volume loss for the two abutment materials (p=0.662). Titanium abutments provided higher loosening torque values after mechanical cycling (p<0.001). Lesser marginal misfit was obtained with titanium abutments before and after mechanical cycling (p<0.001). The stress distribution on the screw was similar between abutment materials. In conclusion, CAD/CAM custom titanium abutment reduced the marginal misfit and increased the torque maintenance of prosthetic screws when compared to CAD/CAM custom zirconia abutment.
Resumo O objetivo neste estudo foi verificar o efeito da diminuição de volume do implante, desajuste vertical entre o pilar e plataforma protética, torque de afrouxamento do parafuso protético e distribuição da tensão no parafuso em pilares de titânio e zircônia. Dez pilares personalizados de cada material foram fresados e conectados aos implantes de titânio. A diminuição de volume do implante foi avaliada com microtomografia, o desajuste vertical com microscopia óptica e o torque de afrouxamento do parafuso protético com chave de torque digital. Todas as análises experimentais foram realizadas antes e após aplicação do ciclo mecânico (1.000.000 ciclos, 100 N/2 Hz). Modelos virtuais das estruturas foram criados para análise por elementos finitos e a tensão no parafuso obtida com valores de von Mises. Os dados foram analisados usando teste t independente, análise de varância dois fatores para medidas repetidas e teste de Tukey HSD (a=0,05). Não houve diferença significativa na diminuição de volume do implante para os dois materiais do pilar (p= 0,662). Os pilares de titânio proporcionaram maiores valores de torque de afrouxamento após o ciclo mecânico (p<0,001). O menor desajuste marginal foi obtido com os pilares de titânio antes e após o ciclo mecânico (p<0,001). A distribuição da tensão no parafuso foi similar entre os materiais. Em conclusão, os pilares personalizados de titânio reduziram o desajuste marginal e aumentaram a manutenção do torque dos parafusos protéticos quando comparados aos pilares de zircônia.

Abstract To compare the shaping ability of different single-file systems in the preparation of mesial curved canals of mandibular molars using micro-CT technology. Fifteen mesial roots of mandibular molars with two independent and curved canals (n = 30) were selected, scanned at a resolution of 26.7 μm anatomically matched, and distributed into three groups (n = 10), according to the preparation system: WaveOne 25.08, Reciproc 25.08, and OneShape 25.06. A final micro-CT scanning was performed, data sets were registered with their respective counterparts, and compared regarding the three-dimensional (volume, surface area, and structure model index - SMI) and two-dimensional (perimeter, area, roundness, major and minor diameters) parameters, as well as, canal transportation, using ANOVA and post hoc Tukey tests (α = 5%). Overall, no difference was observed between groups regarding area, perimeter, volume, surface area, and canal transportation (p > 0.05). Within group, no canal transportation was significantly higherobserved in the apical third (0.10 ± 0.05 mm) compared to coronal (0.08±0.040 mm) and middle (0.07 ± 0.03 mm) thirds (p < 0.05). Structure model index (SMI) was statistically higher after preparation with OneShape instrument (0.36 ± 0.26) compared to other systems (p < 0.05). Within the parameters of this study, similar shaping ability was observed in the preparation of mesial curved root canals of mandibular molars with Reciproc, OneShape and WaveOne systems.

Abstract Objective: To evaluate the effect of ultrasonic and sonic activation on physicochemical properties of AH Plus, MTA Fillapex, ADSeal, GuttaFlow Bioseal, and GuttaFlow 2 sealers. Methodology: Three experimental groups were formed: no activation (NA), ultrasonic activation (UA), and sonic activation (SA). The sealers were manipulated according to the manufacturers' instructions. A 3-mL syringe was adapted to receive 1 mL of sealer. Activation was performed with a 20/.01 ultrasonic insert (20 s/1W) in the UA group. A size 35.04 sonic tip was used (20 s/10,000 cycles/min-1) in the SA group. The molds for physicochemical analysis were filled and evaluated according to ANSI/ADA specification no. 57: setting time (ST), flow (FL), dimensional change (DC), solubility (SB), and radiopacity (RD). Statistical analysis was performed by Kruskal-Wallis, one-way ANOVA, and Tukey's tests (P<0.05). Results: Regarding ST, only AH Plus and GuttaFlow 2 in the NA group met the ANSI/ADA standards. All FL values were greater than 20 mm in diameter, as determined by ANSI/ADA. The tested sealers and protocols did not comply with the ANSI/ADA standards for DC. As for SB, only MTA Fillapex, regardless of the activation protocol, did not follow the ANSI/ADA standards. All of the investigated sealers, regardless of the activation protocol, presented radiographic density higher than 3 mm Al, as proposed by ANSI/ADA. Conclusions: UA and SA promoted changes in the physicochemical properties of the evaluated root canal sealers, mainly in ST and F. Thus, it is important to evaluate the physicochemical properties of endodontic sealers associated with activation techniques prior to clinical application in order to determine whether the properties follow the parameters set by ANSI/ADA, ensuring safety and quality of root canal filling.

Abstract: The aim of this study was to evaluate the bond strength of different triccalcium silicate cements to retrograde cavity using a push out test. Thirty maxillary central incisors were shaped using #80 hand files and sectioned transversally. Root slices were obtained from the apical 4 mm after eliminating the apical extremity. The specimens were embedded in acrylic resin and positioned at 45° to the horizontal plane for preparation of root-end cavities with a diamond ultrasonic retrotip. The samples were divided into three groups according to the root-end filling material (n = 10): MTA Angelus, ProRoot MTA and Biodentine. A gutta-percha cone (#80) was tugged-back at the limit between the canal and the root-end cavity. The root-end cavity was filled and the gutta-percha cone was removed after complete setting of the materials. The specimens were placed in an Instron machine with the root-end filling turned downwards. The push-out shaft was inserted in the space previously occupied by the gutta-percha cone and push out testing was performed at a crosshead speed of 1.0 mm/min. There was no statistically significant difference in resistance to push out by the materials tested (p > 0.01). MTA Angelus and ProRoot MTA showed predominantly mixed failure while Biodentine exhibited mixed and cohesive failures. The tricalcium silicate-based root-end filling materials showed similar bond strength retrograde cavity.

Abstract The aim of this study was to evaluate the effects of proanthocyanidin (PA) and chlorhexidine (CHX) on the bond strength (BS), failure pattern, and resin-dentin interface morphology of the endodontic sealers EndoREZ and AH Plus after 24 h and 6 months of water storage. A total of 120 prepared bovine roots were divided into six groups: AH Plus, CHX+AH Plus, PA+AH Plus, EndoREZ, CHX+EndoREZ, and PA+EndoREZ. Dentin was treated for 1 or 5 min with 2% CHX or 15% PA, respectively. Roots were filled and stored in water for 24 h or 6 months (n = 10). Root slices were subjected to push-out test and scanning electron microscopy (SEM). Data were compared using two-way ANOVA and student's t-test (α = 5%). BS decreased over time for AH Plus and untreated EndoREZ (p < 0.05). At 24 h, AH Plus had higher BS than EndoREZ (p < 0.001), with no differences among treatments for both sealers (p > 0.05). At 6 months, EndoREZ had higher BS values for CHX and PA than control (p < 0.05). AH Plus had higher BS than EndoREZ (p < 0.001), while with CHX or PA, similar BS was observed in both sealers (p > 0.05). Cohesive and mixed failures were observed in all groups. SEM revealed sealer tags in the root dentin. In conclusion, BS decreased with time and AH Plus had higher BS than EndoREZ in untreated dentin; however, CHX or PA enhanced long-term BS of EndoREZ. Overall, dentin treatment affected failure pattern and resin-dentin interface morphology, particularly for EndoREZ.

Abstract This study evaluated the stress distribution in endodontically treated teeth, weakened (W) or not weakened (NW), restored with different materials of prosthetic crown using 3D-FEA. Models of a maxillary canine were constructed based on micro-CT images and divided into the groups: G1 (control) - sound tooth; G2 to G7 - endodontically treated teeth restored with glass fiber post (GFP); which G2 to G4 simulated NW root and G5 to G7 simulated W root. For crown material the teeth were restored with: G2 and G5: metallic coping and ceramic veneering, G3 and G6: zirconia coping and ceramic veneering, G4 and G7: alumina coping and ceramic veneering. Load of 180 N was applied at the incisal third of lingual surface at 45º. Models were supported by the periodontal ligament (x=y=z=0). The von Mises stress (VMS) values were calculated. The W teeth presented higher VMS at coping when compared to NW teeth and group G1 showed lower VMS value. For crown material, for both W or NW teeth, increasing VMS was found at metallic, zirconia and alumina coping, respectively. Metallic coping showed a better performance despite its unfavorable esthetics, suggesting as an appropriate material for prosthetic restoration of endodontically treated teeth.
Resumo Este estudo avaliou a distribuição de tensão em dentes tratados endodonticamente, fragilizados (F) ou não fragilizados (NF), restaurados com diferentes materiais para a coroa protética utilizando 3D-FEA. Modelos de um canino maxilar foram construídos baseados em imagens de micro-CT e divididos em grupos: G1 (controle) - dente hígido; G2 a G7 - dentes tratados endodonticamente com pino de fibra de vidro (PFV), sendo que G2 a G4 simularam raízes NF e G5 a G7 simularam raízes F. Para o material das coroas os dentes foram restaurados com: G2 e G5: coping metálico e revestimento cerâmico, G3 e G6: coping de zirconia e revestimento cerâmico, G4 e G7: coping de alumina e revestimento cerâmico. Carregamento de 180 N foi aplicado na superfície lingual em seu terço incisal com 45 graus de inclinação. Os modelos foram suportados pelo ligamento periodontal (x=y=z=0). Os valores da tensão de von Mises (VMS) foram calculados. Os dentes F apresentaram maiores valores VMS para o coping quando comparados aos dentes NF, sendo que o G1 apresentou menores valores VMS. Para o material das coroas, ambos F ou NF aumentaram VMS no coping metálico, zirconia e alumina, respectivamente. Copings metálicos apresentaram melhor comportamento mecânico apesar de não favorecerem a estética, o que sugere ser um material apropriado para a restauração de dentes tratados endodonticamente.

Abstract The aim of the present study was to evaluate the cytotoxicity of root canal sealers under conditions closely resembling a clinical reality. A primary human fibroblast cell line was seeded in 24-well acrylic plates with Dulbecco’s modified Eagle’s medium supplemented with 10% serum fetal bovine (SFB) and incubated for 24 h. Root canals from premolars were filled and individually attached to nylon devices to be stabilized in the wells with the already seeded cells. Specimens were divided into groups as follows: Control: gutta-percha cones (GPC); AH Plus+GPC; Sealapex+GPC; MTA Fillapex+GPC and Endofill+GPC. After 24 and 48 h, cell viability and morphology were evaluated by MTT assay and scanning electron microscopy (SEM), respectively. Statistical analysis was performed by Mann-Whitney test, complemented by Kruskal Wallis test (p<0.05). Only Endofill presented cytotoxicity after 24 h. MTA Fillapex and Endofill reduced the production of succinic desidrogenase after 48 h. AH Plus was non-toxic at any time point. SEM showed that the AH Plus and MTA Fillapex groups presented fibroblasts with morphology close to the control group, while the Endofill group presented few cells with thin extensions cells. The present study showed that good results were present in AH Plus and Sealapex, but not the Endofill group after 48 h. The method used enabled evaluation of the cytotoxicity of the studied sealers that diffused through the root apex.
Resumo O objetivo do presente estudo foi avaliar a citotoxicidade dos cimentos dos canais radiculares em condições próximas à realidade clinica. Uma linhagem primária de fibrolastos humanos foi semeada em placas acrílicas de 24-poços com meio de cultura Dulbecco’s modified Eagle’s medium suplementado com 10% de soro fetal bovino e incubados por 24 h. Os canais radiculares de pré-molares foram obturados e individualmente adaptados aos dispositivos de nylon para serem estabilizados nos poços com as células já semeadas. Amostras foram dividas de acordo com os grupos: Controle: cones de gutta-percha (CGP); AH Plus+CGP; Sealapex+CGP; MTA Fillapex+CGP e Endofill+CGP. Após 24 e 48 h, a viabilidade e a morfologia celular foram avaliadas pelo ensaio de MTT e microscopia eletrônica de varredura (MEV), respectivamente. Análises estatísticas foram realizadas pelo teste de Mann-Whitney, complementadas por Kruskal Wallis (p<0,05). Apenas o Endofill apresentou citotoxicidade após 24 h. MTA Fillapex e Endofill reduziram a produção da enzima desidrogenase succinica após 48 h. AH Plus não apresentou toxicidade em nenhum momento. MEV mostrou que os grupos AH Plus e o MTA Fillapex apresentaram fibroblastos com morfologia próxima ao grupo controle, enquanto que o grupo do Endofill apresentou poucas células com finos prolongamentos. O presente estudo demonstrou que resultados satisfatórios foram apresentados nos grupos AH Plus e Sealapex, mas não para o Endofill após 48 h. O método utilizado permitiu avaliar a citotoxicidade dos cimentos que se difundem pelo ápice radicular.

Abstract This study evaluated the removal of filling material with ProTaper Universal Rotary Retreatment system (PTR) combined with solvents and the influence of solvents on the bond strength (PBS) of sealer to intraradicular dentin after canal reobturation. Roots were endodontically treated and distributed to five groups (n = 12). The control group was not retreated. In the four experimental groups, canals were retreated with PTR alone or in combination with xylol, orange oil, and eucalyptol. After filling material removal, two specimens of each group were analysed by SEM and µCT to verify the presence of filling remnants on root canal walls. The other roots were reobturated and sectioned in 1-mm-thick dentin slices that were subjected to the push-out test. Data were analysed by two-way ANOVA and Tukey’s test (α = 0.05). SEM and µCT analysis revealed that all retreatment techniques left filling remnants on canal walls. The control group (3.47 ± 1.21) presented significantly higher (p < 0.05) PBS than the experimental groups. The groups retreated with PTR alone (2.59 ± 0.99) or combined with xylol (2.54 ± 0.77) and orange oil (2.32 ± 0.93) presented similar bond strength (p > 0.05), and differed significantly from the group with eucalyptol (1.89 ± 0.63). The solvents reduced the PBS of the sealer to dentin and no retreatment technique promoted complete removal of filling material.

Abstract The aim of this study was to assess the influence of sealer and light-curing unit on regional bond strength of resin composite to the weakened roots. Ninety roots of incisors were experimentally weakened, subjected to biomechanical preparation and filled with either Endofill, AH Plus or MTA Fillapex The roots were desobturated e reinforced with resin composite and fiber post light-activated with one of the light sources: halogen at 600 mW/ cm2 (QTH-600), LED at 800 mW/ cm2 (LED-800) and LED at 1500 mW/ cm2 (LED-1500). The roots were sectioned in slices from cervical, middle and apical root-reinforcement regions and analyzed by push out test, scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). Bond strength data were analyzed using three-way ANOVA and Tukey´s test (α=0.05). Specimens filled with AH Plus had higher bond strength, followed by MTA Fillapex and Endofill (p<0.05). For light-curing unit, LED-1500 presented superior bond strength than LED-800, which was higher than QTH-600 (p<0.05). The cervical region had the greatest mean values (p<0.05) while apical part showed the lowest bond strength (p<0.05). CLMS revealed remaining filling material in the dentinal tubules for all groups. The eugenol-containing sealer (Endofill) compromised the push-out bond strength of composite resin to the root dentin. Bond strength was favored in the cervical region, and when LED-1500 was used.
Resumo O objetivo neste estudo foi avaliar a influência do cimento endodôntico e da fonte de luz fotoativadora na resistência de união (RU) regional da resina composta às raízes fragilizadas. Noventa raízes de incisivos foram experimentalmente fragilizados, submetidos ao preparo biomecânico e obturadas com Endofill, AH Plus ou MTA Fillapex. Os canais foram desobturados e reforçados com pinos de fibra de vidro e resina composta fotoativada com uma das fontes de luz: halógena a 600 mW/cm2 (QTH-600), diodo emissor de luz a 800 mW/cm2 (LED-800) e LED a 1500 mW/cm2 (LED-1500). As raízes foram seccionadas em slices provenientes dos terços cervical, médio e apical das regiões da raiz reforçada e analisadas por meio do teste de push out, microscopia eletrônica de varredura (MEV) e microscopia confocal de varredura a laser (MCVL). Os dados de resistência de união foram analisados por ANOVA a três critérios e teste de Tukey (α=0,05). Os espécimes obturados com AH Plus apresentaram maior resistência de união, seguido do MTA Fillapex e do Endofill (p<0,05). Para fonte de luz fotoativadora, LED-1500 apresentou resistência de união superior ao LED-800 que foi maior que QTH-600 (p<0,05). A região cervical obteve as maiores médias (p<0,05), enquanto que a região apical apresentou a menor resistência de união (p<0,05). MCVL revelou remanescente de material endodôntico nos túbulos dentinários para todos os grupos. O cimento contendo eugenol (Endofill) comprometeu a resistência de união da resina composta à dentina radicular. A resistência de união foi favorecida na região cervical, e quando o LED-1500 foi empregado.

Abstract Root canal irrigants are used to minimize the negative effects of smear layer on endodontic sealer retention. The aim of this study was to evaluate the efficacy of agitation of 17% ethylenediaminetetraacetic acid (EDTA) with ultrasonic, 1064-nm Nd:YAG and 980-nm diode laser on the retention of an epoxy-based sealer to the root canal walls. Forty single-rooted bovine teeth were instrumented with ProTaper rotary system and divided into four groups according to the final irrigation protocol (n = 10): (1) 17% EDTA (control); (2) 17% EDTA with 50-s ultrasonic agitation; (3) 17% EDTA with 50-s diode laser (2-W) agitation; and (4) 17% EDTA with 50-s Nd:YAG (1.5-W) laser agitation. After endodontic filling with gutta-percha F5 master cone and Sealer 26, the roots were sectioned at the cervical, middle, and apical root thirds to obtain 1.5-mm slices. Push-out tests were performed using a universal testing machine at a 1 mm/min crosshead speed. Data were analyzed using two-way ANOVA and Tukey's tests (α=0.05). Apical root thirds had significant higher retention values than cervical and middle thirds (p < 0.05). EDTA with 1064-nm Nd:YAG or 980-nm diode laser presented the highest retention values and was significantly different from EDTA with ultrasonic agitation and EDTA only (p < 0.05). Adhesive failures were predominant to EDTA only group. Mixed failures were predominant to all agitation groups. 1064-nm Nd:YAG and 980-nm diode laser EDTA agitation enhanced the retention of the epoxy-based sealer to the root canal walls compared with that due to EDTA only or EDTA with ultrasonic agitation.
Resumo Irrigantes para canais radiculares são usados para minimizar os efeitos negativos da camada de smear na retenção do cimento obturador. O objetivo deste estudo foi avaliar a eficácia da agitação do ácido etilenodiaminotetracético a 17% (EDTA) com ultrassom, Nd:YAG 1064-nm e laser diodo 980-nm na retenção de um cimento obturador à base de resina epóxica nas paredes do canal radicular. Quarenta dentes bovinos unirradiculares foram instrumentados com o sistema rotatório ProTaper e divididos em quatro grupos de acordo com o protocolo de irrigação final (n = 10): (1) EDTA 17% (controle); (2) EDTA 17% com agitação ultrassônica por 50-s; (3) EDTA 17% com agitação com laser diodo (2-W) por 50-s; e (4) EDTA 17% com agitação com laser Nd:YAG (1,5-W) por 50-s. Após obturação endodôntica com cone principal F5 e cimento Sealer 26, as raízes foram seccionadas nos terços radiculares cervical, médio e apical para obtenção de slices de 1,5-mm. Testes de push-out foram realizados utilizando uma máquina universal de ensaios com velocidade de carga de 1 mm/min. Os dados foram analisados utilizando os testes two-way ANOVA e Tukey's (α=0,05). Os terços radiculares apicais tiveram força de adesão significantemente maior que os terços cervical e médio (p<0,05). EDTA com Nd:YAG 1064-nm ou laser diodo 980-nm apresentaram os maiores valores de força de adesão e foram significantemente diferentes do EDTA com agitação ultrassônica e EDTA apenas (p < 0,05). Falhas adesivas foram predominantes apenas no grupo EDTA. Falhas mistas foram predominantes em todos os grupos de agitação. A agitação do EDTA com Nd:YAG 1064-nm e laser diodo 980-nm aumentou a força de adesão do cimento à base de resina epóxica às paredes do canal radicular comparado com a obtida com EDTA apenas ou EDTA com agitação ultrassônica.

Abstract This study aimed to evaluate the performance of the Wave One, Easy ProDesign Logic and One Shape systems in the preparation of long oval root canals. Forty-two mandibular incisors were randomized into three groups: Group I, Wave One Primary (WO) (#25.08); Group II, Easy ProDesign Logic (EPL) (#25.06) and Group III, One Shape (OS) (#25.06). Micro-computed tomography scans were obtained pre- and post-preparation of the samples. Instrument fractures or loss of working length did not occur in any of the groups. There was a statistically significant difference in total root canal volume between the WO and EPL (p<0.05) and OS systems (p<0.05) The mean percentages of instrumented canal area in the middle thirds were 76.9% in the WO, 62.3% in the EPL and 71.8% in the OS (p>0.05). The OS system had the strongest correlation between volume increase and instrumented area (R=0.63). The WO, EPL and OS systems presented mean preparation times of 2.13 min, 0.54 min, and 2.21 min (p<0.05), respectively. All three systems enabled the safe preparation of long oval root canals. The WO system most effectively increased root canal volume, but it did not affect the instrumented areas. The OS system had the strongest correlation between volume increase and instrumented area, while the EPL system was the fastest.
Resumo Este estudo teve como objetivo avaliar o desempenho dos sistemas Wave One, Easy ProDesign Logic e One Shape no preparo de canais radiculares ovais. Quarenta e dois incisivos inferiores foram randomizados em três grupos: Grupo I, Wave One primary (WO) (#25.08); Grupo II, Easy ProDesign Logic (EPL) (#25.06) e Grupo III, One Shape (OS) (#25.06). As amostras foram submetidas a microtomografia computadorizada antes e após o preparo. Não ocorreram fraturas de instrumento ou perda de comprimento de trabalho em qualquer dos três grupos. Houve diferença estatisticamente significante no volume total do canal radicular entre o WO e EPL. A média do percentual de área instrumentada do canal no terço médio foi 76,9% no grupo WO, 62,3% no grupo EPL e 71,8% no grupo OS (p>0,05). O sistema OS teve a correlação mais forte entre o aumento de volume e área instrumentada (R=0,63). Os sistemas WO, EPL e OS apresentaram tempos de preparo médios de 2,13 min, 0,54 min e 2,21 min (p<0,05). Todos os três sistemas foram seguros no preparo canais radiculares ovais. O sistema WO foi mais eficaz no aumento do volume de canal, entretanto, isso não afetou as áreas instrumentadas. O sistema OS teve a correlação mais forte entre o aumento de volume e área instrumentada, enquanto o sistema EPL foi o mais rápido.

This study evaluates the effect of two thermoplastic obturation systems (MicroSeal and Obtura II) on bond strength of different sealers to intraradicular dentin. Sixty root canals of human canines were prepared using ProTaper rotary files (crown-down technique) and irrigated with 2.5% sodium hypochlorite and 17% EDTA. The root canals were filled by MicroSeal, Obtura II, or lateral compaction techniques using AH Plus and Epiphany SE. 1.5 mm thick root slices were subjected to the push-out test. ANOVA and Tukey's test showed that the bond strength values (MPa) observed in the groups obturated with MicroSeal (2.96 ± 2.72) and Obtura II (2.68 ± 2.18) did not significantly differ from each other (p > 0.05) but were significantly higher than that observed in the group obturated with lateral condensation (2.01 ± 1.48; p < 0.05). There were no statistically significant differences in strength (p > 0.05) among the root canal thirds (cervical: 2.44 ± 2.03; middle: 2.50 ± 2.27; and apical: 2.70 ± 2.34). Adhesive failures were predominant (60%) in all groups. In conclusion, MicroSeal and Obtura II techniques, using AH plus sealer, increased the resistance to displacement of the filling material, when compared with lateral compaction. Moreover, when used with Epiphany SE, these obturation systems did not affect the bond strength of the material to root dentin.

Abstract The aim of this study was to evaluate the effects of different root canal filling techniques on the bond strength of epoxy resin-based sealers. Sixty single-rooted canines were prepared using ProTaper (F5) and divided into the following groups based on the root filling technique: Lateral Compaction (LC), Single Cone (SC), and Tagger Hybrid Technique (THT). The following subgroups (n = 10) were also created based on sealer material used: AH Plus and Sealer 26. Two-millimeter-thick slices were cut from all the root thirds and subjected to push-out test. Data (MPa) was analyzed using ANOVA and Tukey's test (α = 0.05). The push-out values were significantly affected by the sealer, filling technique, and root third (p < 0.05). AH Plus (1.37 ± 1.04) exhibited higher values than Sealer 26 (0.92 ± 0.51), while LC (1.80 ± 0.98) showed greater bond strength than THT (1.16 ± 0.50) and SC (0.92 ± 0.25). The cervical (1.45 ± 1.14) third exhibited higher bond strength, followed by the middle (1.20 ± 0.72) and apical (0.78 ± 0.33) thirds. AH Plus/LC (2.26 ± 1.15) exhibited the highest bond strength values, followed by AH Plus/THT (1.32 ± 0.61), Sealer 26/LC (1.34 ± 0.42), and Sealer 26/THT (1.00 ± 0.27). The lowest values were obtained with AH Plus/SC and Sealer 26/SC. Thus, it can be concluded that the filling technique affects the bond strength of sealers. LC was associated with higher bond strength between the material and intra-radicular dentine than THT and SC techniques.

The present study evaluated the reliability of FEA on the results of different mechanical properties (E and v) of materials. Two 3D models of a maxillary canine with endodontic treatment, intracanal post, composite resin core and restored with porcelain-fused-to-metal crown were generated according to micro-CT images. Two groups with different E and ν values for porcelain, metal coping alloy, resin cement and composite resin were established. The materials' properties for group GL were based on literature data, while for group GIE the impulse excitation technique was used. A load of 180 N was applied at 45° on the incisal third of the lingual surface of the canine tooth. All models were supported by the periodontal ligament (x=y=z=0). The von Mises stress (VMS) was calculated. The stress values revealed differences between the groups for both VMS distribution and value. The porcelain (GL: 5.966 MPa; GIE: 7.478 MPa), metal coping (GL: 3.811 MPa; GIE: 0.973 MPa) and core (GL: 4.771 MPa; GIE: 0.026 MPa) were significantly affected. In conclusion, this study showed that the determination of mechanical properties (E and ν) of materials is essential for the reliability on the results of FEA.
Resumo O presente estudo avaliou a confiabilidade do MEF nos resultados de diferentes propriedades mecânicas (E e v) dos materiais. Dois modelos 3D de um canino superior com tratamento endodôntico, pino intrarradicular, núcleo em resina composta e restaurado com coroa metalocerâmica foram gerados de acordo com imagens de micro-CT. Dois grupos com diferentes valores de E e v para a cerâmica, coping metálico, cimento resinoso e resina composta foram criados. As propriedades dos materiais para o grupo GL foram baseadas em dados obtidos da literatura, enquanto para o grupo GIE a técnica de excitação por impulso foi utilizada. Carga de 180 N foi aplicada a 45° na superfície lingual do terço incisal do canino. Todos os modelos foram suportados pelo ligamento periodontal (x=y=z=0). Tensões equivalentes de von Mises (VMS) foram calculadas. Os valores de tensões revelaram diferença entre os grupos, ambos para a distribuição e valor das VMS. A cerâmica (GL: 5,966 MPa; GIE: 7,478 MPa), coping metálico (GL: 3,811 MPa; GIE: 0,973 MPa) e núcleo (GL: 4,771 MPa; GIE: 0,026 MPa) foram significativamente afetados. Em conclusão, este estudo mostrou que a determinação das propriedades mecânicas (E and v) dos materiais é essencial para a confiabilidade dos resultados do MEF.

This study evaluated the bond strength of root-end filling materials to root-end cavities using a new methodology. Twenty maxillary central incisors were subjected to biomechanical preparation (#80 hand file) and sectioned transversally 2 mm short of the apex and 4 mm coronally to this point. The root cylinders were embedded in acrylic resin and positioned at 45° to the horizontal plane for preparation of root-end cavities with a diamond ultrasonic retrotip. Two groups (n=10) were formed according to the root-end filling material: MTA and Super EBA. A gutta-percha cone (#80) was tug-backed at the limit between the canal and the root-end cavity. The cavity was filled and the gutta-percha cone was removed after complete setting of the sealer. The specimens were placed in an Instron machine with the root-end filling turned downwards. The push-out shaft was inserted in the space previously occupied by the gutta-percha cone and run at a crosshead speed of 1.0 mm/min for pushing out the root-end filling material. Data were analyzed by ANOVA (α=5%). Super EBA (6.03±1.31) presented higher bond strength (MPa) than MTA (1.81±0.45) (p>0.05). There was a predominance of cohesive failures for Super EBA and mixed for MTA. The protocol of specimen preparation is effective and introduces a specific methodology for assessing bond strength of root-end filling materials to dentin. Among the materials, Super EBA presented the highest bond strength.
Este estudo avaliou a resistência de união de materiais retrobturadores a cavidades retrógradas, usando uma nova metodologia. Vinte incisivos centrais superiores foram submetidos ao preparo biomecânico (lima manual #80) e seccionadas transversalmente a 2 mm do ápice e a 4 mm do primeiro corte. Os cilindros de raízes foram incluídos em resina acrílica e posicionado a 45° em relação ao plano horizontal para confecção das retrocavidades com ponta ultrassônica diamantada. Dois grupos (n = 10) foram formados de acordo com os cimentos utilizados: MTA e Super EBA. Um cone de guta-percha (#80) foi travado no limite da retrocavidade com o canal radicular. A retrocavidade foi preenchida e, após o tempo de endurecimento do cimento utilizado, o cone de guta-percha foi removido. Os espécimes foram fixados na máquina universal de ensaios Instron com a retrobturação voltada para baixo. A haste empregada para pressionar o cimento retrobturador no teste de push-out foi inserida no espaço anteriormente ocupado pela guta-percha e acionada com velocidade de cruzeta de 1,0 mm/min, para extrusão do material retroburador. Os dados foram analisados por meio do teste ANOVA (α=5%). O cimento Super EBA (6,03 ± 1,31) apresentou maior resistência de união (MPa) comparado ao cimento MTA (1,81 ± 0,45) (p> 0,05). Houve predomínio de falhas coesivas para o cimento Super EBA e falhas mistas para o MTA. O protocolo de preparação das amostras é eficaz e apresenta uma metodologia específica para avaliação da resistência de união de materiais retrobturadores à dentina. Dentre os materiais testados, o cimento Super EBA apresentou a maior resistência de união.
