ABSTRACT

Objective:To compare the effect of intranasal dexmedetomidine and oral chloral hydrate in deep sedation of children.Methods:The Pubmed, EMBase, CENTRAL (Issue 4, 2018), Web of science, CBM, Wanfang Data, CNKI and VIP databases from the inception to January 2019 were searched. Randomized controlled trials (RCTs) with dexmedetomidine and chloral hydrate as interventions were included and the data were analyzed by RevMam 5.3 and Stata 12.0 software. The success rate of deep sedation, the indicator of sedation onset time, the recovery time, the incidence of vomiting and bradycardia were compared.Results:A total of 7 RCTs involving 1 007 patients were included for analysis. The results showed that the success rate of deep sedation ( OR=2.55, 95 %CI:1.46-4.44, P<0.01) and the incidence of bradycardia ( OR=4.42, 95 %CI:1.82-10.74, P<0.01) in the dexmedetomidine nasal group were significantly higher than those in the chloral hydrate oral group. The recovery time was significantly shorter ( MD=-16.41, 95 %CI:-21.54-11.28, P<0.01) and the incidence rate of vomiting ( OR=0.04, 95 %CI:0.01-0.17, P<0.01) in dexmedetomidine nasal group was significantly lower than those in the chloral hydrate oral group. There was no significant difference in the indicator of sedation onset time ( MD=-0.47, 95 %CI:-2.71-1.22, P=0.46). Conclusion:Compared with the traditional oral chloral hydrate, intranasal dexmedetomidine has a higher sedation success rate and shorter recovery time after sedation with a lower incidence of nausea and vomiting.

ABSTRACT

Objective@#To search new prognosis-related genes in gastric cancer by analyzing the high-throughput sequencing data of gastric cancer in TCGA database, and then provide data support for future studies. @*Methods@#The RNA-seq expression matrix data and patient-related clinical data from 375 gastric cancer tissues and 45 adjacent noncancerous tissues were downloaded from the TCGA database. The data were collated and standardized based on the R language. The difference of gene expression was analyzed by the edgeR and DEseq software packages. The survival analysis of obtained differential genes was performed by the univariate and multivariate COX regressions combined with clinical data of patients, and then the genes with clinical significance were screened out. @*Results@#A total of 364 differential genes were obtained by the edgeR and DEseq analysis. Subsequently, the functional enrichment analysis found that these genes were mainly involved in protein digestion and absorption, cytochrome P450 system of drug and exogenous substance metabolism, chemical carcinogenesis, gastric acid secretion and so on. The univariate COX regression analysis showed that FAP, FAT3, PDK4 and ZNF365 genes had significant influences on the prognosis of gastric cancer patients. The multivariate COX stepwise regression analysis showed that the risk model constructed by FAP and PDK4 could predict the prognosis of gastric cancer patients. @*Conclusion: @#FAP, FAT3, PDK4 and ZNF365 genes may be the prognostic markers of gastric cancer, which may provide data supports for future clinical and basic studies.

ABSTRACT

ABSTRACT: The Nitrogen content of rice leaves has a significant effect on growth quality and crop yield. We proposed and demonstrated a non-invasive method for the quantitative inversion of rice nitrogen content based on hyperspectral remote sensing data collected by an unmanned aerial vehicle (UAV). Rice canopy albedo images were acquired by a hyperspectral imager onboard an M600-UAV platform. The radiation calibration method was then used to process these data and the reflectance of canopy leaves was acquired. Experimental validation was conducted using the rice field of Shenyang Agricultural University, which was classified into 4 fertilizer levels: zero nitrogen, low nitrogen, normal nitrogen, and high nitrogen. Gaussian process regression (GPR) was then used to train the inversion algorithm to identify specific spectral bands with the highest contribution. This led to a reduction in noise and a higher inversion accuracy. Principal component analysis (PCA) was also used for dimensionality reduction, thereby reducing redundant information and significantly increasing efficiency. A comparison with ground truth measurements demonstrated that the proposed technique was successful in establishing a nitrogen inversion model, the accuracy of which was quantified using a linear fit (R2=0.8525) and the root mean square error (RMSE=0.9507). These results support the use of GPR and provide a theoretical basis for the inversion of rice nitrogen by UAV hyperspectral remote sensing.


RESUMO: O teor de nitrogênio das folhas de arroz tem um efeito significativo sobre a qualidade do crescimento e o rendimento das culturas. Propõe-se e demonstrou-se um método não invasivo para a inversão quantitativa do teor de nitrogênio do arroz com base em dados de detecção remota hiperespectral coletados por um veículo aéreo não tripulado (UAV). As imagens de albedo do dossel de arroz foram adquiridas por uma imagem de imagem hiperespectral a bordo de uma plataforma M600-UAV. O método de calibração da radiação foi então usado para processar esses dados e a reflectância das folhas do dossel foi adquirida. A validação experimental foi realizada utilizando o campo de arroz da Universidade Agrícola de Shenyang, que foi classificado em 4 níveis de fertilizantes: nitrogênio zero, baixo teor de nitrogênio, nitrogênio normal e alto teor de nitrogênio. A regressão do processo gaussiano (GPR) foi então usada para treinar o algoritmo de inversão para identificar bandas espectrais específicas com a maior contribuição. Isso levou a uma redução no ruído e uma maior precisão de inversão. A análise de componentes praincipais (PCA) também foi usada para redução de dimensionalidade, reduzindo assim a informação redundante e aumentando significativamente a eficiência. Uma comparação com as medidas de verdade no solo demonstrou que a técnica proposta foi bem sucedida no estabelecimento de um modelo de inversão de nitrogênio, cuja precisão foi quantificada usando um ajuste linear (R2 = 0,8525) e o erro quadrático médio quadrado (RMSE = 0,9507). Estes resultados suportam o uso do GPR e fornecem uma base teórica para a inversão do nitrogênio do arroz pela detecção remota hiperespectral do UAV.

ABSTRACT

Objective To investigate the effect of different shear stress on the expression of Interleukin-8 (IL-8) and endothelin-1 (ET-1) in vascular endothelial cells.Methods Umbilical vein endothelial cell line (ECV-304) was used as the research cell.The shear stress of 5 dyne/cm2 and 10 dyne/cm2 was regulated by the self-made shearing stress control device.The shear stress in were maintained at 0,5 dyne/cm2 and 10 dyne/cm2 respectively in the control group,the low shear stress group and the high shear stress group.The effects on the levels of IL-8 and ET-1 were analyzed between the three groups at five time points of 30 min,60 min,120 min,300 min,420 min.Pearson correlation analysis was used to analyze the correlation between IL-8 and ET-1.Results The level of IL-8 in the three groups has been on the rise.At the three time points of 30 min,60 min and 120 min,there was no statistically significant difference in IL-8 level between the three groups (P > 0.05).At the two time points of 300 min and 420 min,the difference of the IL-8 level in the three groups was statistically different (P < 0.05),At the two time points of 300 min and 420 min,the IL-8 level in the low shear stress group was higher than that in the control group and the high shear group,the IL-8 level in the high shear group was higher than that in the control group (P < 0.05).The level of ET-1 in the control group and the low shear group showed an upward trend,and the ET-1 level in the high shear group increased continuously in the former 120 min,but began to decrease continuously since then.At the three time points of 30 min,60 min and 120 min,the difference of the ET-1 level in the three groups was statistically different (P < 0.05).The ET-1 level of the high shear group at three time points of 30 min,60 min and 120 min was higher than that of the control group,the level of ET-1 at two time points of 30 min and 60 min was higher than that in low shear stress group (P < 0.05),the ET-1 level of the low shear group at two time points of 60 min and 120 min was higher than that of the control group (P < 0.05).At the two time points of 300 min and 420 min,there was no significant difference in the level of ET1 between the three groups (P > 0.05).Pearson correlation analysis showed that there was a positive correlation between the expression of IL-8 and ET-1 (r =0.384,P =0.031).Conclusions Shear stress can affect the expression of IL-8 and ET-1 in vascular endothelial cells,the shear stress of 5 dyne/em2 can increase the level of IL-8 more efficiently,the shear force of 10 dyne/cm2 can increase the level of ET-1 more obviously,but it may cause damage to the vascular endothelial cells.

ABSTRACT

Aim To investigate the skeletal effects of ginseng flower bud(GF)on osteopenia induced by D-galactose using histomorphometry and biomechanical properties.Methods Fifty three-month-old male Sprague-Dawley rats were randomly divided into five groups.Rats in NS group(NS)were treated with NS(5 mL·kg-1·d-1)by subcutaneous injection and daily oral gavage with vehicle as control.Rats in the other four groups were given D-galactose at the dose of 100 mg·kg-1·d-1 by subcutaneous injection.Solvent control was performed between NS and DG: gastric irrigation with distilled water of 10 mL·kg-1·d-1.Other groups were: CP was gastric irrigated with integrated medicine(stanozolol 0.54 mg·kg-1·d-1+piracetam 432 mg·kg-1·d-1),GF(L)with ginseng flower bud of 0.486 g·kg-1·d-1 and GF(H)with ginseng flower bud of 2.43 g·kg-1·d-1 for 14 weeks.The longitudinal proximal tibial metaphyseal(PTM),the fifth lumbar vertebral body(LVB)and tibial shaft(Tx)sections were performed undecalcifiedly and used for bone histomorphometric analysis.858 Mini Bionix materials testing system was used to analyze the biomechanic properties of right femur via three-point bending test.The left femur was dried and assimilated,whose bone calcium(Ca),phosphate(P)content and bone hydroxyproline content were tested.Results Compared with D-glagatose group,in PTM of D-galactose treated rats,the%Tb.Ar was increased both in GF(L)and GF(H)treated groups.While the Tb.Sp was decreased.%Oc.S.Pm and Oc.N/mm decreased in GF(L),and those in GF(H)were decreased as well.In Tx,%Ct.Ar was raised,while%Ma.Ar was decreased in GF(L)and GF(H).The elastic load of femur was increased.Conclusions Compared with DG group,there are significant differences in bone histomorphometry of Tx and PTM in all doses of GF,but no significant changes are detected in hydroxyproline,Ca,and P content of femur.

ABSTRACT

Objective To explore the effects of dexmedetomidine on recovery quality of hypertension patients under general anesthesia,so as to provide reference for its clinical application.Methods 31 patients who underwent abdominal surgery were chosen as study subjects.They were randomly divided into observation group(15 patients) and control group(16 patients) according to the digital table.At the time of 15 min before the end of surgery,the patients in the observation group were injected with dexmedetomidine of 0.5g/kg,and the patients in the control group were injected with the same volume of normal saline instead in 15 min.At the time of before anesthesia(T0),before injection(T1),3 min after injection (T2),just before extubation(T3),1 min(T4),5 min(T5),15 min(T6) after extubation,the hemodynamic parameters,the sedation scores at the time of T5 and T6,and the occurrence of cough during extubation were recorded and compared between the two groups.Results During operation and extubation,the levels of SBP,DBP,HR and their fluctuations in the observation group were lower than those in the control group.At the time of T5 and T6,the sedation scores in the observation were lower than those in the control group.The mean rank and scores of the two groups [at the time of T5 were [(12.2±24.6) and (19.5±24.6),respectively,Z=2.27,P=0.022].The mean rank and scores of the two groups at the time of T6 were T6[(12.3±24.7) and (19.5±24.7),respectively,Z=2.25,P=0.024].During the time of extubation,the occurrence of cough in the observation group was lower than that in the control group.The mean rank of the cough score in the observation group and the control group was[(12.2±24.1) and (19.5±24.1),Z=2.33,P=0.020].Conclusion Small dose of dexmedetomidine can reduce the incidences of intraoperative cerebral vascular accident risk,improve the quality of general anesthesia recovery in hypertension patients,and thus should be recommend in clinical application.

ABSTRACT

Objective To evaluate the cardioprotection induced by ischemic preconditioning ( IPC) and hypoxic preconditioning ( HPC) which were carried out by using the double cardiopulmonary bypass ( CPB) circuits in in vivo hearts of dogs. Methods Eighteen healthy male dogs, weighing 17?5-24?5 kg, aged 13-24 months, were divided into 3 groups ( n=6 each) using a random number table: my?ocardial ischemia?reperfusion group (group I∕R), IPC group and HPC group. The double CPB circuits were established as follows: systemic and coronary circulation, and independent systemic and coronary cir?culation was carried out. In group IPC, the aorta was clamped, and the coronary circulation pump was sus?pended for 5 min followed by 5 min opening, repeating for 3 cycles. In group HPC, the aorta was clamped, the coronary circulation was started, and pure nitrogen was insufflated for 5 min followed by 5 min of oxygen insufflation, repeating for 3 cycles. CPB was performed for 1 h starting from the time point immediately after IPC or HPC. Before splitting of sternum ( T1 ) , after establishment of double CPB circuits ( T2 ) , at the end of preconditioning ( T3 ) , and at 60 and 120 min after restoration of spontaneous heart beat ( T4,5 ) , heat rate, mean arterial pressure, central venous pressure, left ventricular end?systolic pres?sure, left ventricular end?diastolic pressure and the maximum rate of increase∕decrease of left ventricular pressure were recorded. Blood samples were collected from the right internal jugular vein at T1 and T4,5 for determination of serum cardiac troponin I concentrations. The animals were sacrificed after determination of the parameter or after blood sampling at T5 , myocardial specimens were obtained for examination of the ul?trastructure and for detection of apoptosis in cardiomyocytes, and apoptosis index was calculated. Before aortic clamping, immediately after aortic unclamping and at 30 min after aortic unclamping, myocardial specimens were obtained for determination of ATP contents in cardiomyocytes. Results Compared with I∕R group, left ventricular end?systolic pressure was significantly increased, and the serum cardiac troponin I concentrations were significantly decreased at T4,5 , the myocardial ATP contents were significantly in?creased immediately after aortic unclamping and at 30 min after aortic unclamping, apoptosis index was sig?nificantly decreased ( P<0?05 or 0?01) , and the pathological changes were significantly attenuated in IPC and HPC groups. Compared with group IPC, the myocardial ATP contents were significantly increased (P<0?05), and the pathological changes were attenuated in group HPC. Conclusion Both HPC and IPC can exert cardioprotection when carried out by using the double CPB circuits, and HPC provides better cardioprotection than IPC in in vivo hearts of dogs.

ABSTRACT

Objective To observe the clinical efficacy of TCM periodic therapy combined with estradiol progesterone tablets (Diane-35) in the treatment for polycystic ovary syndrome (PCOS), and its effects on serum sex hormone, glucose and lipid metabolism.Methods Totally 74 patients with PCOS were randomly divided into Western medicine (WM) group and Chinese and Western medicine (CWM) group, 37 cases in each group. WM group was given Diane-35, while CWM group was treated with TCM periodic therapy additionally, for three courses, 28 d of one course. Clinical symptoms, sex hormones and glucose and lipid metabolism of two groups were observed before and after treatment and withdrawal treatment for 3 months.Results Two patients in each group were lost to follow-up. Markedly effective rate and total efficiency rate of CWM group were better than WM group (P<0.05). After treatment, symptom integrals of CWM group decreased significantly (P<0.01). Serum E2 increased (P<0.01), while LH, T and LH/FSH decreased in two groups (P<0.01). After treatment and withdrawal treatment for 3 months, improvement of clinical symptoms and sex hormone levels in CWM group was superior to CM group (P<0.01), FPG, FINS, TG, TC and BMI of CWM group decreased in CWM group (P<0.05), and lower than that of CM group (P<0.05,P<0.01). Adverse reaction rates of WM group and CWM group were 17.1% and 5.71% (P<0.05).Conclusion Efficacy of TCM periodic therapy combined with Diane-35 in treatment of PCOS is significant, which can obviously improve patients’ clinical symptoms, and regulate hormone and lipid metabolism disorders, with fewer adverse reactions.

ABSTRACT

Objective To observe clinical efficacy of Tianma Gouteng Decoction combined with Western medicine for early onset severe pre-eclampsia. Methods Sixty patients were randomly divided into treatment group with 30 cases and control group with 30 cases. Two groups were both given the routine Western therapy, while the treatment group was added with Tianma Gouteng Decoction. Indicators were observed and compared between two groups including blood pressure, 24 h UAE, D-dimer, fibrinogen and their effects on pregnancy. Results Compared with the control group, the blood pressure, 24 h UAE, fibrinogen, D-dimer, hypoproteinemia, and liver and kidney function were reduced obviously in the treatment group (P<0.05);fibrinogen and D-dimer of control group were not changed significantly;gestational weeks of treatment group were extended (15.2 ± 4.8)d, and neonatal asphyxia was 7 cases;gestational weeks of control group were extended (9.3 ± 2.2)d, and neonatal asphyxia was 11 cases. There was no significant difference about cesarean section rate between the two groups. Conclusion Tianma Gouteng Decoction combined with Western medicine for treatment of early onset severe pre-eclampsia have better clinical efficacy.

ABSTRACT

Objective To study the expression and clinical significance of Notch intracellular domain (NICD) in cervical cancer and the effects of N-[N-(3,5-difluorophenyl)acetyl-L-alanyl]-S-phenyl glycine t-butyl ester (DAPT), a γ-secretase inhibitor on the proliferation and apoptosis of cervical cancer cell lines. Methods Western blot was used to detect the expression of NICD in the tissues of 40 cervical cancers and 21 normal cervix and its relationship with clinical features of cervical cancer was also analyzed. Proliferation of SiHa and HeLa cervical cells was determined by methyl thiazolyl tetrazolium (MTT) assay, cell cycles and apoptosis and index of proliferation were detected by flow cytometry method. The expression of NICD in SiHa and HeLa cells incubated with DAPT was detected by western blot. Results The expression level of NICD in cervical cancers was significantly higher than that of normal cervical tissues (1.237±0.353 vs 0.938±0.105, P<0.05). The NICD expression was higher in cervical cancers with high grade,lymph node involvement and parametrial invasion than that with low-middle grade (1.496±0.540 vs 1.150±0.216), without lymph node involvement (1.419±0.532 vs 1.159±0.210) and no parametrial invasion (1.718±0.710 vs 1.183±0.258), respectively (all P<0.05). The expression of NICD in cervical adenocarcinoma was higher than that of squamous cell cancer (1.463±0.395 vs 1.162±0.187, P<0.05). After SiHa and HeLa cells were incubated with DAPT, NICD expression was significantly lower than that in control (P<0.05). The effects of DAPT inhibited the proliferation and prompted the apoptosis of SiHa and HeLa cells was depended on its concentrations and times. Conclusions NICD may play a key role in the occurrence and progress of cervical cancer. The mechanism of DAPT inhibited the proliferation and prompted the apoptosis of SiHa and HeLa cells may be due to decreased the formation of NICD.

Objective To investigate the different expressions of various subtypes of human leukocyte antigen-G(HLA-G)mRNA in placentas among early and late-onset severe preeclamptic and normal pregnant women. Methods Eleven placental samples from early-onset severe preeclamptic pregnancies and fourteen from late-onset severe preeclamptic pregnancies were collected as study group;eight placental samples from normal pregnancies were collected as control group.The levels of HLA-G mRNA in the three groups were measured by nested RT-PCR. Results Compared with the counterparts in the control group,the leveI of HLA-G1 subtype was significantly decreased (median:Early-onset group:1.37,P<0.05;Late-onset group:24.90,P<0.05;Control group:46.67,P<0.05)and the level of HLA-G5 subtype increased(median:Early-onset group 19.23;Late-onset group:3.65;Control group:1.33)in both early and late-onset severe preeclamptic group.The level of HLA-G2 subtype increased(median:Early-onset group:0;Late-onset group:21.59,P<0.05;Control group:5.39)in the late-onset severe preeclamptic group. Conclusion The reduced expression of HLA-G membrane-bound subtype mRNA in the placenta and the increased expression of HLA-G soluble subtype mRNA in the placenta may be related tO the pathogenesis of preeclampsia.

ABSTRACT

Objective: To study the effects of estrogen, progestogen and mifepristone on endometrial carcinoma cell lines in vitro. Methods: The well-differentiated endometrial cancer cell line Ishikawa and moderate-differentiated endometrial cancer cell line Hec-1B were cultured in vitro. The cells were divided into four groups:control, estrogen , estrogen and progestogen , estrogen and progestogen and mifepristone, then we implanted cells in 96-well plates to search the response in distinct hormones. Results: When stimulated by estrogen for 72 hours, the growth of Ishikawa cells was significantly higher than the control, Hec-1B cells only grew higher than the control, but it was no significance in statistics. Progestogen inhibited Ishikawa cells significantly after being stimulating for 72 hours, there was the same effect on Hec-1B cells after being stimulating for 96 hours. On the base of estrogen and progestogen, we added mifepristone,cells developed after 96 hours in Ishikawa cells and cells developed after 48 hours in Hec-1B cells. Conclusion: Estrogen can cause endometrial carcinoma cell growth; progestogen inhibits the hyperplasia induced by estrogen; mifepristone antagonizes the effect of progestogen on cell growth.

Objective:To study the functional differences between the two progestrone receptor isoforms(PR-A and PR-B) in human endometrial cancer,using antisense oligodeoxynucleotide(AS-ODN) to downregulate isoform B of progestrone receptor in endometrial carcinoma cell lines, After transfection of the oligodeoxynucleotide, several kinds of hormones were added in the cells to observe the different response,whereh to study the functional differences between the two isoforms. Methods: The well-differentiated endometrial cancer cell line Ishikawa and moderate-differentiated endometrial cancer cell line Hec-1B were cultrued in vitro. The cells were transfected with antisense, sense, and scramble-ODN. After 48 hours, the expressions of two progesterone receptor isoforms were detected by Western blot using specific antibody. Then the cells were planted in 96-well plates, transfected with antisense, sense, scramble-ODN and added in several hormones to search for the response in distinct hormones and oligodeoxynucleotides. Results: After transfecting antisense-ODN, two cell lines were down-regulated in progesterone receptor isoform B,but progesterone receptor isoform A was not down-regulated,and the progesterone receptor isoform B of cells transfected with sense and scramble-ODN was not changed. When stimulated by 17?-estradiol(E2)for 72 hours,the growth of Ishikawa cells was significantly higher than that of the control, Hec-1B cells only grew higher than control,but it was to significant in statistics.R5020 inhibited Ishikawa cells significantly after stimulating for 72 hours.There was the same effect in Hec-1B cells after stimulating for 96 hours.On the bases of E2 and R5020, we added mifepristone(RU486) .The cells developed after 96 hours in Ishikawa cells and developed after 48 hours in Hec-1B cells. When PR-B was down-regulated,the stimulating effect of E2 was enhanced, but the inhibitory effect of R5020 was de-creased, RU486 antagonized R5020 weaklier than the control. Conclusion: AS-ODN directed against the human PR-B can inhibit the expression of PR-B effectively,through which the PR-A expresses predominantly. E2 can cause endometrial carcinoma cell growth, PR-B is associated with the stimulating effect of E2 in endometrial carcinoma cells. Progestin (R5020) inhibits the hyperplasia induced by E2,PR-B is involved in the inhibitory effect of R5020. RU486 antagonizes the effect of R5020,inhibiting cell growth, PR-B is involved in the antagonizing effect of RU486.

ABSTRACT

Objective To study the role of human leukocyte antigen-G (HLA-G) in the carcinogenesis and development of choriocarcinoma. Methods We designed and synthesized a double strand small interference RNA (siRNA) of HLA-G, then transfected it into a HLA-G overexpressed choriocarcinoma cell line, JEG-3 by lipofectine; HLA-G mRNA level was detected by real time RT-PCR; HLA-G protein level was detected by western blot. The living cells of JEG-3 were counted under microscope after transfection by siRNA. Results Double strand siRNA of HLA-G effectively downregulated the mRNA level and the protein level of HLA-G. The mRNA levels by Ct value at different concentration (1.0, 2.5, 5.0 ?g/L) were 20.67?0.02, 21.37?0.03,21.43?0.02, respectively. ABSTRACT

Objective To explore an efficient way to modulate the expression of estrogen receptor (ER) ? and ?, and to build up a model of endometrial cancer cell expressing predominantly one isoform of ER and to verify the roles of ER ? and ? in the tumorigenesis of endometrial cancer associated with estrogen and tamoxifen (TAM). Methods A series of oligodeoxyribonucleotides (ODN) against ? or ? regions of ER ? or ? were synthesized and tested in human endometrial cancer cell lines (Ishikawa) that express functional ER ? and ?. The expressions of two ER isoforms were detected by western blot using specific antibodies. Then we studied the change of Ishikawa proliferation in response to 17?-estradiol and TAM under the influence of antisense ODN. Results (1) Transfection with antisense ODN directed against the ER? and ER? could significantly inhibit target protein production. (2) 17?-estradiol could increase the proliferation of Ishikawa cells, but they lost the ability to proliferate in response to 17?-estradiol after transfected with ER? antisense ODN especially at hours 24, 48 and 72 ( P
